Abstract:The development of an environmental methodology for polyethylene terephthalate (PET) degradation and separation from cotton is significant for the full utilization of waste polyester/cotton blends (PCBs). Therefore, it is crucial to find a catalyst that can efficiently catalyze PET degradation and completely separate from cotton. In this work, betaine based deep eutectic solvents (DESs) were first synthesized and innovatively used for selective degradation of PET fibers from PCBs. As a result, PET fiber was degraded completely, a purified monomer bis (2-hydroxyethyl terephthalate) (BHET) with a yield of 85% was obtained that could be repolymerized to produce PET, and 95% of the cotton was recycled and maintained its original structural integrity. Meanwhile, the synergistic mechanism was also proposed by in-situ IR, 1H‐NMR and DFT calculations. This work provides a new approach to recycle full components of waste PCBs.

Abstract:Waste plastics are serious environmental threats due to their low degradability and low recycling rate. Rapid and efficient waste plastics recycling technologies are urgently demanded for a sustainable future. Herein, we report a rapid, closed-loop, and streamlined process to convert polyesters such as poly(ethylene terephthalate) (PET) back to its purified monomers. Using trifluoromethanesulfonic acid or metal triflates as the recyclable catalyst, polyesters such as PET can be completely depolymerized by simple carboxylic acids within 1 h. By coupling this acidolysis with a subsequent hydrogenolysis process, the consumed carboxylic acid was recovered and the closed-loop of PET depolymerization could be established. All catalysts and depolymerization agents are fully recycled while only PET and hydrogen are consumed.

Abstract:The immune system relies on cellular communication and often on the recognition of carbohydrates by mammalian lectins. Galactose (Gal)-containing structures are involved in both the innate and adaptive immune systems. Gal is a ligand for Gal/N-acetylgalactosmine (GalNAc) receptors and galectins, and is part of the scaffold structure that synthesizes oligosaccharide ligands for selectins, siglecs and other lectins of the immune system. Gal residues are added to glycoproteins and glycolipids by members of a large family of galactosyltransferases. The expression of many of these enzymes is regulated by the action of cytokines, and becomes altered in various disease states. Specific galactosyltransferases have been shown to control cell adhesion and leukocyte functions. Antibodies need to be galactosylated for normal function, and undergalactosylated immunoglobulin (Ig) is associated with rheumatoid arthritis, while Gal is lacking in the IgA of patients with IgA nephropathy. Interactions involving Gal play important roles in host defenses; they can also result in serious pathophysiology. Galactosyltransferases represent potential targets for the control of cell growth and apoptosis, inflammation and infections.
Selective depolymerization of PET to monomers from its waste blends and composites at ambient temperature

Abstract:Recycling of polyethylene terephthalate (PET), one of the largest varieties of synthetic polymers and the primary textile feedstock, presents a significant challenge when waste PET blends or composites are involved. Despite numerous efforts to address waste PET, it is difficult to achieve energy-efficient and selective depolymerization of PET through physical sorting systems or existing chemical means. Herein, we report a dichloromethane (DCM) and ethanol (EtOH) solvent system to selectively depolymerize PET into monomers while fully recover other mixture components. The complete depolymerization of PET and recovery of more than 99% of terephthalic acid (TPA) monomer can be achieved at room temperature within 30 min. After the reaction, TPA can be separated by simple filtration due to its insolubility in the solution. The filtrate can be reused for the recovery of PET. Molecular dynamics simulation shows that the high activity and selectivity is attributed to activating effect of DCM to PET ester groups, as well as pore-forming effect to promote PET interfacial mass transfer. Environmental energy impact and carbon emissions were evaluated and the results demonstrate the environmental friendliness of this method. This approach enables direct and efficient recycling of PET monomers from waste blends and composites, providing a viable solution to plastic and textile pollution.

Integrated oncology course for first-year medical students.

The course is characterized by a high level of integration of the involved basic and clinical sciences and is designed according to the principles of problem-based learning. A limited number of lectures provides a frame of reference. A series of case histories introduces the students to various aspects of neoplastic disease and after group discussions leads the students to individual learning activities. During the course only general principles are touched upon. Students, as well as teaching staff, appear to be highly appreciative of the integrated approach.
From Polyester Plastics to Diverse Monomers via Low-Energy Upcycling

Abstract:Polyester plastics, constituting over 10% of the total plastic production, are widely used in packaging, fiber, single-use beverage bottles, etc. However, their current depolymerization processes face challenges such as non-broad spectrum recyclability, lack of diversified high-value-added depolymerization products, and crucially high energy consumption. Herein, an efficient strategy is developed for dismantling the compact structure of polyester plastics to achieve diverse monomer recovery. Polyester plastics undergo swelling and decrystallization with a low depolymerization energy barrier via synergistic effects of polyfluorine/hydrogen bonding, which is further demonstrated via density functional theory calculations. The swelling process is elucidated through scanning electron microscopy analysis. Obvious destruction of the crystalline region is demonstrated through X-ray crystal diffractometry curves. PET undergoes different aminolysis efficiently, yielding nine corresponding high-value-added monomers via low-energy upcycling. Furthermore, four types of polyester plastics and five types of blended polyester plastics are closed-loop recycled, affording diverse monomers with exceeding 90% yields. Kilogram-scale depolymerization of real polyethylene terephthalate (PET) waste plastics is successfully achieved with a 96% yield.
Efficient Alcoholysis of Poly(ethylene terephthalate) by Using Supercritical Carbon Dioxide as a Green Solvent

Abstract:In order to reduce the environmental impact of poly(ethylene terephthalate) (PET) plastic waste, supercritical fluids were used to facilitate effective recovery via improved solvent effects. This work focuses on the mechanisms of supercritical CO2 (ScCO2) during the alcoholysis processing of PET using systematic experiments and molecular dynamics (MD) simulations. The results of the alcoholysis experiment indicated that PET chips can be completely depolymerized within only an hour at 473 K assisted with ScCO2 at an optimal molar ratio of CO2/ethanol of 0.2. Random scission of PET dominates the early stage of the depolymerization reaction process, while specific scission dominates the following stage. Correspondingly, molecular dynamics (MD) simulations revealed that the solubilization and self-diffusion properties of ScCO2 facilitate the transportation of alcohol molecules into the bulk phase of PET, which leads to an accelerated diffusion of both oligomers and small molecules in the system. However, the presence of excessive CO2 has a negative impact on depolymerization by weakening the hydrogen bonding between polyester chain segments and ethanol, as well as decreasing the swelling degree of PET. These data provide a deep understanding of PET degradation by alcohols and the enhancement of ScCO2. It should be expected to achieve an efficient and high-yield depolymerization process of wasted polyesters assisted with ScCO2 at a relatively low temperature.

Abstract:Abstract Less than 9% of the plastic produced is recycled after use, contributing to the global plastic pollution problem. While polyethylene terephthalate (PET) is one of the most common plastics, its thermomechanical recycling generates a material of lesser quality. Enzymes are highly selective, renewable catalysts active at mild temperatures; however, the current consensus is that they lack activity towards the more crystalline forms of PET. We report here that when used in moist-solid reaction mixtures instead of the typical dilute aqueous solutions, enzymes can directly depolymerize high crystallinity PET in 13-fold higher space-time yield and a 15-fold higher enzyme efficiency than prior reports. Further, this process shows a 26-fold selectivity for terephthalic acid over other hydrolysis products, which allows the direct synthesis of UiO-66 metal-organic framework.

Abstract:Efficient chemical upcycling of polyethylene terephthalate (PET) waste into value-added chemicals has attracted considerable attention in a broad range of environmental and energy fields. The mechanochemical depolymerization of PET waste emerges as a potential approach for large-scale industrial applications, enabling the conversion of PET waste into reusable monomers. Herein, we conducted a pilot-scale test by depolymerization of 1 kg of PET waste in a planetary ball mill. The incremental optimization of the depolymerization process was investigated systematically by adjusting parameters such as the type of ball mill, the ratio of liquid volume to solid mass, the grinding jars, and the milling frequency. Remarkably, monomers with a yield of more than 97 % when converting kilogram-scale PET waste, including used textiles and plastic bottles, were achieved in a planetary ball mill at 400 rpm for 80 min. This notable enhancement will substantially benefit future potential industrial-scale applications.

Abstract:The application of deep eutectic solvents (DESs) in the catalytic degradation of polyethylene terephthalate (PET) has attracted increasing attention due to the high efficiency, biodegradability, and low toxicity of DESs. How-ever, the relationship between the structure and activity of DESs is still ambiguous. Herein, we report an efficient 1,5,7-triazabicyclo[4.4.0]decium acetate/zinc acetate (HTBD-OAc/Zn(OAc)(2)) DES catalyst for recycling waste PET based on a combination of experimental and theoretical studies. First, the HTBD-OAc/Zn(OAc)(2) DES structure, in which the HTBD-OAc moiety and Zn(OAc)(2) were connected through a N-H-O hydrogen bonding interaction, was clearly determined through theoretical calculations and experimental verification. Then, the HTBD-OAc/Zn(OAc)(2) was found to be effective for waste PET glycolysis, where the PET conversion reached 100%, and the bis(hydroxyalkyl) terephthalate (BHET) yield reached 88.5% at 190 C within 40 min in atmosphere. Additionally, the catalyst had high reuse performance; BHET yield remained 84.6% after the catalyst was used for over seven cycles. Density functional theory (DFT) calculations demonstrated that the PET glycolysis reaction mediated by the HTBD-OAc/Zn(OAc)(2) DES included two stages, and the 1,5,7-triazabicyclo[4.4.0]decium cation (HTBD) played a crucial role. In the first stage, the active species [Zn]-OCH2CH2OH anion was generated through proton transfer from HTBD to OAc-, which was followed by HOCH2CH2O-H bond activation through the cooperation of HTBD and Zn. In the second stage, catalytic depolymerization of PET occurred through the cooperative functions of the Zn center and HOCH2CH2O- anion ligand.

Abstract:Alcoholysis of poly(ethylene terephthalate) (PET) waste to produce monomers, including methanolysis to yield dimethyl terephthalate (DMT) and glycolysis to generate bis-2-hydroxyethyl terephthalate (BHET), is a promising strategy in PET waste management. Here, we introduce an efficient PET-alcoholysis approach utilizing an oxygen-vacancy ( V o )-rich catalyst under air, achieving space time yield (STY) of 505.2 g DMT ·g cat −1 ·h −1 and 957.1 g BHET ·g cat −1 ·h −1 , these results represent 51-fold and 28-fold performance enhancements compared to reactions conducted under N 2 . In situ spectroscopy, in combination with density functional theory calculations, elucidates the reaction pathways of PET depolymerization. The process involves O 2 -assisted activation of CH 3 OH to form CH 3 OH * and OOH * species at V o -Zn 2+ –O–Fe 3+ sites, highlighting the critical role of V o -Zn 2+ –O–Fe 3+ sites in ester bond activation and C–O bond cleavage. Moreover, a life cycle assessment demonstrates the viability of our approach in closed-loop recycling, achieving 56.0% energy savings and 44.5% reduction in greenhouse-gas emissions. Notably, utilizing PET textile scrap further leads to 58.4% reduction in initial total operating costs. This research offers a sustainable solution to the challenge of PET waste accumulation.

Abstract:As the demand of polyethylene terephthalate (PET) increases worldwide along with the waste generated from its use, it is urgent to develop cost-efficient and sustainable recycling processes, such as depolymerization that yields monomers with virgin-like qualities. However, most of these processes require harsh conditions and their true mechanisms are poorly understood, leading to marginal gains in energy efficiency and yield. In spite of the lack of solubility of PET, we demonstrate that the swelling and plastification of PET in a good solvent allows a better mass transport of ethylene glycol and the catalyst into the polymer during glycolysis. Based on these insights, we report a process in which PET is depolymerized into bis(2-hydroxyethyl)terephthalate (BHET) with a yield of 88 % at 65 °C within 1 h in the presence of 1,3-Dioxolane as green cosolvent carrier. The improved mass transport allows to perform the depolymerization of PET even below the T g of the polymer. Indeed, by kinetic modelling we demonstrated that a heterogeneous depolymerization process could be easily transformed into an homogeneous process by an appropriate solvent selection. The environmental impact of the proposed process, including solvent recovery, is compared to the solvent-free counterpart and the results demonstrates that by using 1,3-Dioxolane, the carbon footprint of the newly developed glycolysis process can be reduced up to 20 % due to the increased energy efficiency. This process enables a viable recycling strategy of PET into its repetitive unit, contributing to the development of competitive chemical recycling solutions to reduce PET-derived pollution.

Abstract:Significance Although polyethylene terephthalate (PET) is the most recycled plastic, its recycling typically involves thermomechanical means, producing a plastic of lower quality that cannot be recycled again. In contrast, breaking PET down to its building blocks allows for repolymerization into high-quality PET. Chemical depolymerization of PET requires hazardous chemicals and conditions. Enzymes are environmentally benign, renewable catalysts that work under mild conditions. Several enzymes capable of depolymerizing low-crystallinity PET have been reported; however, efficient depolymerization of highly crystalline forms of PET typically found in consumer products has remained elusive. Here, we report that, in moist-solid reaction mixtures, the cutinase from Humicola insolens can efficiently hydrolyze highly crystalline PET to terephthalic acid with a 50% yield, without any amorphization or pretreatment of PET.

Abstract:Poly(ethylene terephthalate) (PET) is the most abundant polyester plastic and is causing serious environmental pollution. Rapid biological depolymerization of PET waste at a large scale requires powerful engineered enzymes with excellent performance. Here, we designed a computational strategy to analyze the ligand affinity energy of enzymes to PET chains by molecular docking with the dynamic protein conformations, named affinity analysis based on dynamic docking (ADD). After three rounds of protein engineering assisted by ADD, we drastically enhanced the PET-depolymerizing activity of leaf-branch-compost cutinase (LCC). The best variant LCC-A2 depolymerized >90% of the pretreated, postconsumer PET waste into corresponding monomers within 3.3 h at 78 °C, and over 99% of the products was terminal depolymerization products (terephthalic acid and ethylene glycol), representing the fastest PET depolymerization rate reported to date in the bioreactor under optimal condition. Structural analysis revealed interesting features that improved the ligand affinity and catalytic performance. In conclusion, the proposed strategy and engineered variants represent a substantial advancement of the biological circular economy for PET.

Abstract:Low-impact, closed-loop recycling of plastics is crucial to sustainably managing these pervasive and resource-intense materials. Our approach aimed to reinvent chemical recycling by integrating it with electrochemical processes to generate reactants electrochemically and depolymerize plastic in situ, with the objective of reducing both costs and environmental impacts. We investigated electrochemically mediated alkaline hydrolysis and methanolysis of poly(ethylene terephthalate) (PET) to achieve the following advantages over conventional methods: access to more extreme reactivity from applying an electrochemical driving force, process intensification, and application of more moderate operating conditions. Total PET conversion and product yields were measured to systematically investigate the performance effects of catholyte methanol content, anolyte buffering, and temperature. Leveraging these insights to improve experimental conditions, we achieved 45 mol% PET conversion in 5 hours at ambient pressure and relatively moderate temperature (50 °C) in 0.1 M NaClO4 (100 mol% methanol) catholyte and 0.1 M Na3PO4 anolyte.

Abstract:Developing efficient and cost-effective methodologies for high value-added conversion of waste plastic delivers substantial environmental and economic benefits. Herein, we develop a novel approach utilizing boric acid in the methanolysis of waste polyethylene terephthalate (PET) to derive pure dimethyl terephthalate (DMT) and boronic acid esters through in-situ capture of ethylene glycol (EG). It not only upcycles waste PET but also eliminates intricate EG purification processes. Catalyzed by magnalium-aluminum-layered double oxides (Mg 4 Al 1 -LDO), this method achieved 100% conversions of PET with 96% and 100% yields of arylboronic esters and DMT, respectively. Kinetic studies and in-situ Fourier-transform infrared spectroscopy (FT-IR) demonstrated the pivotal role of the monodentate methoxy species, generated through the interaction of medium basic Mg–O ion pairs and methanol. This method demonstrates applicability for the upcycling of assorted discarded PET wastes, polyesters, and polycarbonates with EG units, highlighting its potential as a comprehensive solution for waste plastic management.
Computational redesign of a hydrolase for nearly complete PET depolymerization at industrially relevant high-solids loading

Abstract:Abstract Biotechnological plastic recycling has emerged as a suitable option for addressing the pollution crisis. A major breakthrough in the biodegradation of poly(ethylene terephthalate) (PET) is achieved by using a LCC variant, which permits 90% conversion at an industrial level. Despite the achievements, its applications have been hampered by the remaining 10% of nonbiodegradable PET. Herein, we address current challenges by employing a computational strategy to engineer a hydrolase from the bacterium HR29. The redesigned variant, TurboPETase, outperforms other well-known PET hydrolases. Nearly complete depolymerization is accomplished in 8 h at a solids loading of 200 g kg −1 . Kinetic and structural analysis suggest that the improved performance may be attributed to a more flexible PET-binding groove that facilitates the targeting of more specific attack sites. Collectively, our results constitute a significant advance in understanding and engineering of industrially applicable polyester hydrolases, and provide guidance for further efforts on other polymer types.

Abstract:Plastic pollution presents a global challenge, impacting ecosystems, wildlife, and economies. Polyethylene terephthalate (PET), widely used in products like bottles, significantly contributes to this issue due to poor waste collection. In recent years, there has been increasing interest in plant biomass-degrading enzymes for plastic breakdown, due to the structural and physicochemical similarities between natural and synthetic polymers. Filamentous fungi involved in hemicellulose degradation have developed a complex mode of action that includes not only enzymes but also biosurfactants; surface-active molecules that facilitate enzyme-substrate interactions. For this reason, this study aimed to mimic the mechanism of biomass degradation by repurposing plant cell wall degrading enzymes including a cutinase and three esterases to cooperatively contribute to PET degradation. Surfactants of different charge were also introduced in the reactions, as their role is similar to biosurfactants, altering the surface tension of the polymers and thus improving enzymes' accessibility. Notably, Fusarium oxysporum cutinase combined with anionic surfactant exhibited a 2.3- and 1.6-fold higher efficacy in hydrolyzing amorphous and semi-crystalline PET, respectively. When cutinase was combined with either of two ferulic acid esterases, it resulted in complete conversion of PET intermediate products to TPA, increasing the overall product release up to 1.9- fold in presence of surfactant. The combination of cutinase with a glucuronoyl esterase demonstrated significant potential in plastic depolymerization, increasing degradation yields in semi-crystalline PET by up to 1.4-fold. The approach of incorporating enzyme cocktails and surfactants emerge as an efficient solution for PET degradation in mild reaction conditions, with potential applications in eco-friendly plastic waste management.

Abstract:The efficient depolymerization of polyesters under mild conditions remains a significant challenge. Herein, we demonstrate a highly efficient strategy for the degradation of a diverse array of waste polyesters as low to 80 °C, 1 bar H2 . The key to the success of this transformation relied on the initial transesterification of macromolecular polyester into more degradable oligomeric fragments in the presence of CH3 OH and the subsequent hydrogenation by the use of the rationally designed quinaldine-based Ru complex. Controlled experiments and preliminary mechanistic studies disclosed the quinaldine-based catalysts could be hydrogenated to the eventually active species, which has been confirmed by X-ray diffraction analysis and directly used as a catalyst in the hydrogenolysis of polyester. The strong viability and high activity of this new species in protic solvent were explained in detail. Besides, the crucial role of CH3 OH in promoting reaction efficiency during the whole process was also elucidated. The synthetic utility of this method was further illustrated by preparing 1,4-cyclohexanedimethanol (CHDM) from waste polyethylene terephthalate (PET).
