Abstract:Although Aurantii Fructus (AF) and Aurantii Fructus Immaturus (AFI) are both the fruits of the same rutaceae plant at different stages of growth, they exert similar yet distinct clinical effects. The chemical composition is crucial for quality control as well as therapeutic application. To address this concern, it is significant to evaluate the similarities and differences of the constituents in both AF and AFI. The extract of AF and AFI were comprehensively analyzed by ultra fast liquid chromatography-photodiode array detector-triple-time of flight-tandem mass spectrometry (UFLC-DAD-Triple TOF-MS/MS). Among the 40 compounds detected, 19 metabolites were detected in both the AF and AFI; whereas 13 compounds were only detected in AF and five constituents were exclusively detected in AFI. In particular, even in AFI, three compounds were only identified in AFI (Citrus aurantium' L. and its cultivar). Among the 18 compounds confirmed by standard database, 13 compounds were reported in AF and AFI for the first time. Furthermore, the distinction was also revealed by the content of naringin, hesperidin, neohesperidin, and synephrine. The study directly contributed to the similarities and differences of AF and AFI. Herein, similarities and the differences in chemical profiles of AF and AFI could explain the current clinical applications.

Abstract:To establish a comprehensive quality control method for total flavonoid of Fructus Aurantii.RP-HPLC and spectrophotometry were applied for the quantitative and fingerprint analysis of total flavonoid of Fructus Aurantii. The contents of naringin and neohesperidin were determined on an Agilent SB-C₁₈column (4.6 mm × 250 mm, 5 μm). The mobile phase was composed of 0.02 % H₃PO₄ and CH₃CN (80:20). The flow rate was 1 ml/min with DAD detected at 280 nm. The column temperature was maintained at 35°C. The fingerprints were developed on an Agilent SB-C₁₈ column (4.6 mm × 250 mm, 5 μm). The mobile phase was composed of 0.5 % HAc and CH₃OH with a linear gradient elution. The ratio of 0.5 % HAc and CH₃OH was: 0 min, 80:20; 10 min, 60:40; 35 min, 30:70; 50 min, 0:100. The flow rate was 1 ml/min with DAD detected at 320 nm. The column temperature was maintained at 30 degree. Meanwhile, the contents of total flavonoid were determined at 283 nm.The contents range of naringin, neohesperidin and total flavonoid were 38.3 %- 47.2%, 21.0 %- 28.5% and 79.9%-88.6 %, respectively. The fingerprints of the effective fractions showed 12 common peaks and the fingerprint similarity was all above 98.0 % compared with the standard chromatogram.The method reported in this paper can be used effectively for the quality control of total flavonoid of Fructus Aurantii.

Abstract:Quzhou Aurantii Fructus (QAF), the dried immature fruit of Citrus changshan-huyou Y.B. Chang, is similar to Aurantii Fructus (AF), the dried immature fruit of Citrus aurantium L. or its cultivars, in terms of composition, pharmacological action, and appearance. However, potential chemical markers to distinguish QAF from AF remain unknown owing to the lack of a comprehensive systematic chemical comparison aligned with discriminant analysis. To achieve a better understanding of the differences in their composition, this study aimed to identify the basic chemical compounds in QAF (n = 42) and AF (n = 8) using ultra-performance liquid chromatography coupled with electron spray ionization and quadrupole time-of-flight mass spectrometry (UPLC−QTOF/MS) and gas chromatography coupled with mass spectrometry (GC−MS). Principal component analysis (PCA), orthogonal partial least squares-discriminant analysis (OPLS−DA), and hierarchical clustering analysis (HCA) were used to further analyze, screen, and verify potential chemical markers; the antioxidant capacity was assayed in vitro. A total of 108 compounds were found in QAF and AF, including 25 flavonoids, 8 limonoids, 2 coumarins, and 73 volatile components. The chemometric analysis indicated that the main components in QAF and AF were very similar. Trace differential components, including 9 flavonoids, 2 coumarins, 5 limonoids, and 26 volatile compounds, were screened as potential chemical markers to distinguish between QAF and AF. Additionally, the antioxidant capacity of QAF was found to be greater than that of AF. This research provides insights into the quality control and clinical application of QAF.

Abstract:In traditional Chinese medicine, Aurantii Fructus Immatures (AFIs) have been utilized for more than 2000 years. The proportions of different fruit parts are crucial for evaluating AFI quality in China. However, the basis for this statement's substance is unclear. Differences in quality are intimately correlated with a plant's metabolite composition. On the basis of a widely targeted metabolome, this study intended to investigate the metabolite composition and evaluate the antioxidant capacity of the peel and pulp of an AFI. Metabolites were identified and quantified by UHPLC-QqQ-MS. To assess their antioxidant ability, DPPH and ABTS assays were carried out. There were 1327 chemical compounds identified by UHPLC-QqQ-MS. After screening the differential metabolites using a multivariate statistical analysis, it was found that there were 695 significant differences in the metabolites between the peel and the pulp. Among them, it was discovered that the content of active ingredients in the peel group was higher than that in the pulp group. Furthermore, the aqueous extracts from the peel showed stronger antioxidant capacities than those from the pulp. The metabolites and antioxidant capacities were significantly different between the peel and the pulp. This study of different fruit parts might provide a guide for AFI quality assessments.

Abstract:Hemodiafiltration (HDF) was created in the late 1970s to combine the best of hemodialysis (HD) and hemofiltration, i.e. superior removal of both small and large solutes. Thus, optimal conditions for both diffusion and convection are required. The need for large volumes of sterile fluid has shaped the evolution of HDF, and regulatory issues around systems for on-line fluid preparation have delayed the clinical development of the therapy. Not until such systems became generally available in the mid-1990s was it possible to increase the efficiency of HDF therapy to match and exceed the upgrade that had taken place for HD. Results from recent studies indicate that the potential for improving the outcome of dialysis lies in increasing the convective clearance of HDF above that of high-flux HD.

Abstract:Fructus Aurantii is a traditional medicated diet in East Asia. To determine the underlying chemical markers responsible for the quality and efficacy of Fructus Aurantii, a sensitive metabolomic method was applied to distinguish Fructus Aurantii in Jiangxi Province from other two geographical locations (Hunan Province and Chongqi City) in China. In the present study, multivariate analyses were adopted to compare chemical compositions in 21 batches of Fructus Aurantii samples. Among three geographical origins, 23 differential compounds were structurally identified. Serum pharmacochemistry exhibited that 22 components could be detected in rat serum. Six differential and absorbed components were selected as six potential markers. Statistical analysis of the effects of six potential markers on the quality of Fructus Aurantii revealed that the content of markers varied widely in three origins. Six differential and absorbed components were evaluated further biological activity. Neohesperidin, naringin and meranzin showed inhibitory effect on acetylcholinesterase that regulates gastrointestinal motility in vitro and in silico, suggesting that these three components may be determined as the active biomarkers of Fructus Aurantii. Latent Transforming Growth Factor-/ l Associates to Fibroblast Extracellular Matrix via Latent ' II 3 FB Binding Protein

Abstract:The role of latent transforming growth factor-/~ (TGF-/3) binding protein (LTBP) in the association of TGF-~ 1 to the extracellular matrix of cultured fibroblasts and HT-1080 fibrosarcoma cells was studied by immunochemical methods. The matrices were isolated from the cells, and the levels of LTBP and TGF/~1 were estimated by immunoblotting and immunoprecipitation. LTBP, TGF-/31, and its propeptide (latencyassociated peptide, LAP) were found to associate to the extracellular matrix. Immunoblotting analysis indicated that treatment of the cells with plasmin resulted in a concomitant time and dose dependent release of both LTBP and TGF-~I from the extracellular matrix to the supernatant. Comparison of molecular weights suggested that plasmin treatment resulted in the cleavage of LTBP from the high molecular weight fibroblast form to a form resembling the low molecular weight LTBP found in platelets. Pulse-chase and immunoprecipitation analysis indicated that both the free form of LTBP and LTBP complexed to latent TGF-/$ were efficiently incorporated in the extracellular matrix, from where both complexes were slowly released to the culture medium. Addition of plasmin to the chase solution resulted, however, in a rapid release of LTBP from the matrix. Fibroblast derived LTBP was found to associate to the matrix of HT-1080 cells in a plasmin sensitive manner as shown by immunoprecipitation analysis. These results suggest that the latent form of TGF-/31 associates with the extracellular matrix via LTBP, and that the release of latent TGF-/~ 1 from the matrix is a consequence of proteolytic cleavage(s) of

Abstract:Objective:To study on method for evaluating the quality of Chinese herb Fructus Aurantii. Methods: Chemical fingerprint and quantitative analysis of Chinese herb Fructus Aurantii were established by HPLC method. The fingerprints of Fructus Aurantii were developed on an Agilent Hypersil ODS column (4. 0 mm - 250 mm, 5 (μm). The mobile phase was composed of 0. 5% HAc and CH3OH with a linear gradient elution. The similarity was calculated with the similarity calculation software of Fingerprint Chromatography of traditional Chinese medicine that the pharmacopoeia committee recommended. Meanwhile, the contents of naringin in Fructus Aurantii were determined on an Agilent Hypersil ODS column(4. 0 mm x 150 mm,5 μm). The mobile phase was composed of 0. 3% HAc and CH3OH(35:65). The flow rate was 1 mL ?min-1 with DAD detected at 280 nm. The column temperature was maintained at 30 ℃. The two approaches were synchronized to evaluate the quality of the herb. Results: The content of naringin in Fructus Aurantii came from different sources changed from 3. 19% to 5. 50%. From the results of fingerprints analysis, it could be found that the samples from different patches showed little difference, while,from different geography origins showed a significant variation. Conclusion: The method reported in this paper can be used scientifically and effectively to evaluate the quality of Chinese herb Fructus Aurantiμi.

Abstract:This study presents an efficient strategy based on microwave-assisted extraction (MAE), HPLC-DAD-MS/MS and high-speed counter-current chromatography (HSCCC) for the rapid extraction, identification, separation and purification of active components from the traditional Chinese medicine Fructus Aurantii Immaturus. An LC-DAD-MS/MS method was applied for the screening and structural identification of main components in crude extract, and five components were preliminarily identified as neoeriocitrin, narirutin, naringin, hesperidin and neohesperidin according to their UV and mass spectra. An efficient MAE method for the extraction of the three most abundant components (narirutin, naringin and neohesperidin) was optimized by the combination of univariate and multivariate approaches. The crude extract was then separated and purified by HSCCC and a total of 61.6 mg of narirutin, 207.3 mg of naringin and 159.5 mg of neohesperidin at high purities of 98.1, 97.2 and 99.5%, respectively, were obtained from 1.42 g of crude extract. The recoveries of these compounds were 86, 93 and 89%, respectively.

Abstract:A method for quantitative determination of the main active ingredient in traditional Chinese medicine was developed and validated, using liquid-liquid chromatography combined with high performance liquid chromatography, which was successfully applied to synephrine in Fructus Aurantii Immaturus. A biphasic solvent system composed of ethyl acetate - n-butanol - water - trifluoroacetic acid (1:2:3:0.003, v/v) was used to purify the crude extract of the samples by liquid-liquid chromatography. Then, quantitative analysis of synephrine in Fructus Aurantii Immaturus could be well determined by high performance liquid chromatography. Moreover, the collection time and retention time of synephrine could be well predicted using a continuous-stirred tank reactors model. Compared with previous studies, the present method showed advantages of good recovery, good repeatability, high selectivity to target components and simple operation.

Abstract:Objective: To compare the differences of the chemical constitutions of Frutus Aurantii Immaturus of different origin Places.Method: By one-dimensional FT-IR spectroscopy,second derivative spectra and two-dimensional related spectral analysis of IR spectra,the features of these spectrum peaks of Frutus Aurantii Immaturus were identified.Result: In one-dimensional map,the raw material of Frutus Aurantii Immaturus from 5 different origins had the similar peak shape among the rang of 400-4 000 cm-1,and 7 common peaks.Frutus Aurantii Immaturus from each origin place had the same chemical content.In the second derivative spectra the characteristic peaks of Frutus Aurantii Immaturus from Yunnan were less and blunter,the more peaks of Frutus Aurantii Immaturus from Jiangsu and Jiangxi were found,and the shapes of peaks were sharp.The two-dimensional related spectral map showed there were obvious differences between the peaks of Frutus Aurantii Immaturus from 5 different origin places.Conclusion: This IR spectra technique can identify Frutus Aurantii Immaturusof different origin places.
Distinguishing Foeniculum vulgare fruit from two adulterants by combination of microscopy and GC-MS analysis

Abstract:Foeniculum vulgare fruit (FVF) is a widely used traditional medicine. However, two adulterants, namely Anethum graveolens fruit (AGF) and Cuminum cyminum fruit (CCF), have been found in use as FVF in China owing to similar appearance and odor. For the purpose of accurate differentiation of the three herbal medicines, extensive anatomical examination and chemical profiling were conducted. Using light microscopy and fluorescence microscopy, the macroscopic and microscopic features of the three species were compared. It was found that some microscopic characteristics, including transverse shape of mericarp, presence or absence of reticulate cells and non-glandular hairs, as well as fluorescence of endocarp, were of diagnostic significance. Moreover, essential oils were qualitatively and semi-quantitatively analyzed using GC-MS. The analytical results indicated significant chemical variations in different species: in FVF, trans-anethole (83.20%) was the predominant volatile compound followed by estragole (5.03%) and limonene (3.45%), while in AGF, the first, second and third compounds with highest content were carvone (42.58%), apiol (20.76%) and limonene (20.32%), and in CCF were cuminlaldehyde (36.00%), 2-caren-10-al (23.25%) and γ-terpinene (9.65%), respectively. In conclusion, the proposed light microscopy coupled with fluorescence microscopy and/or GC-MS analysis allowed successful distinguishing FVF from AGF and CCF.

Abstract:The lateral roots of Aconitum carmichaelii, known in Chinese as fuzi, are officially recognized as a materia medica in the Chinese Pharmacopoeia and used culinarily to prepare herbal soups. A strategy combining UPLC-qToF-MS analysis of A. carmichaelii and its intraspecies and interspecies chemometrics study was developed to examine the distribution of Aconitum marker metabolites. Four diterpenoid alkaloids were recognized to be important markers in fuzi, and another 15 markers were identified to differentiate A. carmichaelii from adulterant species. The detected fuzi markers, mesaconitine (47) and hypaconitine (51), are known to be the principal toxins in this herb, while fuziline (6) and benzoylmesaconine (25) are associated with its medicinal properties. Additional marker compounds have been detected in other Aconitum species that are useful for identifying adulteration. High-Performance Liquid Chromatography Fingerprint Analysis of Smilacis Glabrae Rhizoma and Its Adulterants

Abstract:OBJECTIVE To investigate the chemical differences between Smilacis Glabrae Rhizoma and its adulterants,and provide a basis for the identification and quality evaluation of Tufuling samples purchased from the markets.METHODS The method of HPLC fingerprinting was used to analyze Smilacis Glabrae Rhizoma,Heterosmilacis Chinensis Rhizoma,Heterosmilacis Yunnanensis Rhizoma,and 102 Tufuling samples which were collected around China.The fingerprints were analyzed by the methods of similarity and principal component analysis (PCA).RESULTS The analytical method of HPLC fingerprinting was established.Eleven,fifteen and eight common peaks were selected in the fingerprints of Smilacis Glabrae Rhizoma,Heterosmilacis Chinensis Rhizoma,and Heterosmilacis Yunnanensis Rhizoma,respectively.Only five common peaks were found in the fingerprints of the three species,which were No.1,2,3,13,and 14 peaks.A total of twelve peaks were characterized in the three fingerprints.Nine peaks were characterized in the fingerprint of Smilacis Glabrae Rhizoma,among which,four constituents were characterized for the first time.Six and two constituents were for the first time characterized in the fingerprints of Heterosmilacis Chinensis Rhizoma and Heterosmilacis Yunnanensis Rhizoma,respectively.The result of PCA analysis showed that the chemical differences between the three species were quite obvious and they could be distinguished from each other.The established method was used for the analysis of Tufuling samples purchased from the markets.Sixty-five samples were identified as Smilacis Glabrae Rhizoma,seventeen samples were identified as Heterosmilacis Yunnanensis Rhizoma,and twenty samples were identified as Heterosmilacis Chinensis Rhizoma.CONCLUSION The established method is simple and reliable,and the development of fingerprint and its chemical pattern recognition provide the way and basis for identification of Smilacis Glabrae Rhizoma and its adulterants.

Abstract:A reliable method based on high performance liquid chromatography–diode array detector–electrospray ionization–ion trap–time of flight–mass spectrometry (HPLC–DAD–ESI–IT–TOF–MS) was developed for the identification of phenolic acids and flavonoids in Apocynum venetum L. leaves and its adulterant, Pocynum hendersonii (Hook. f.) Woodson leaves. A total of 21 compounds were identified or tentatively identified, including 4 phenolic acids and 17 flavonoids. 3-O-caffeoylquinic acid (3-CQA) and caffeic acid were detected for the first time in A. venetum leaves; 4-O-caffeoylquinic acid (4-CQA), 3-CQA, caffeic acid, quercetin-3-O-(6″-O-malonyl)-galactoside, quercetin-3-O-(6″-O-malonyl)-glucoside, kaempferol-3-O-(6″-O-malonyl)-glucoside, kaempferol-3-O-(6″-O-acetyl)-glucoside, and kaempferol-3-O-dihexoside were detected for the first time in P. hendersonii leaves. Cluster analysis was employed to analyze 24 batches of A. venetum leaves and 5 batches of P. hendersonii leaves collected from various regions in China. The analysis, which was based on the 21 compounds, indicated that profiles of these compounds were distinct between the two species, and among A. venetum leaf samples from different origins. 18 of these 21 compounds were selected as the markers and simultaneously analyzed by HPLC–DAD for the first time. The quantitative analytical method was validated and subsequently applied to the comprehensive quality evaluation of 24 batches of A. venetum leaves.

Abstract:Fructus Gardeniae (FG) is the desiccative and ripe fruits of Gardenia jasminoides Ellis in the Rubiaceae family, which is a commonly used in traditional Chinese medicine (TCM) for clearing away heat, detoxification, relieving restlessness, and eliminating blood stasis. At the same time, it has also been announced as the first batch of TCM with homology of medicine and food. Fructus Gardeniae Grandiflorae (FGG), the fruit of Gardenia jasminoides Ellis var. grandiflora Nakai (Rubiaceae), is a common counterfeit herbal medicine of FG, which still appears in the TCM market, and causes a certain degree of confusion. In order to effectively distinguish FG and its adulterant, the compounds in these two species were thoroughly characterized firstly by ultrahigh-performance liquid chromatography/quadrupole–orbitrap mass spectrometry (UHPLC/Q-Orbitrap MS). Furthermore, a pseudo-targeted metabonomics method with 60 targeted ion pairs was established based on UHPLC-triple quadrupole-linear ion trap mass spectrometry (UHPLC-QTRAP-MS) for discrimination. Multivariate statistical analysis showed that FG and FGG were clustered obviously, and 13 significantly differential markers were screened out by variable importance for projection (VIP) > 1 and p < 0.05 for the construction of the partial least squares (PLS) regression prediction model. The validation of the model proved that its prediction ability was quite satisfactory. Moreover, based on the absolute quantitative analysis of these 13 characteristics, the quality control standards of FG and FFG were established. In summary, an integral method of pseudo-targeted metabonomics combined with chemometrics analysis and a PLS regression model was proposed to provide an effective identification strategy for discrimination FG and FGG.

Abstract:C ontractions of skeletal muscles provide the stability and power for all body movements. Consequently, any impairment in skeletal muscle function results in some degree of instability or immobility. Factors that influence skeletal muscle structure and function are therefore of great interest both scientifically and clinically. Injury, disease, and old age are among the factors that commonly contribute to impairment in skeletal muscle function. The goal of this article is to update current concepts of skeletal muscle physiology. Particular emphasis is placed on mechanisms of injury, repair, and adaptation in skeletal muscle as well as mechanisms underlying the declining skeletal muscle structure and function associated with aging. A Preliminary Study on the Molecular and Chemical Characteristics of Authenticity (daodixing)of a Traditional Chinese Medicinal Plant-Forsythia Suspense Vahl

Abstract:To address the authenticity (daodixing, a concept used to judge the quality of Traditional Chinese Medicines (TCM) mainly based on the growing environments as well as the varieties of medicinal plants) of a Traditional Chinese Medicinal plant, Forsythia suspensa, the genetic relationship of 11 cultivars from sixdifferent regions of Shanxi province (Licheng,Changzhi, Tunliu, Huguan, Pingshun, Anze) was analyzed using the randomly amplified polymorphic DNAs (RAPD) technique. The contents of forsythin, an indicative component of Forsythia suspensa, in these cultivars were analyzed using high performance liquid chromatography(HPLC). The RAPD analysis showed that there was a significant genetic diversity among the 11 cultivars. These cultivars were arranged into three genetic branches (A, B and C) using the Philip3.6 software and Wagner′s method. The branch A containing the only cultivar from Huguan was genetically distant from the other two branches, B and C, which were relatively close. The HPLC analysis showed that there was also a great variation in the contents of forsythin among these cultivars. Most dramatic variations were found between the branchA (8.7 mg/g) and the branchB or the branchC. However no significant variations were found within the three branches, nor between the branchB (the mean 0.53mg/g)and the branchC (the mean 0.67mg/g). These results suggest that the formation and accumulation of forsythin could be affected substantially by the genetic basis of Forsythia suspense. It is particularly true in this case since these cultivars were collected from relatively less diverse environments. This work has shown some promise that the authenticity of Traditional Chinese Medicinal plants can be addressed using RAPD and HPLC techniques.
Development of an HPLC Fingerprint for Quality Control and Species Differentiation Offructus Schisandrae

Abstract:The ripe fruits of Schisandrae chinensis have a long history of use in traditional Chinese medicine to treat diseases and improve health. There is substantial evidence that lignan constituents are mainly responsible for the beneficial effects of this plant medicine. The amounts of the major bioactive lignans in this plant vary widely with species, habitat, and the collecting time, and as such, establishment of an HPLC fingerprint for quality control of this herbal medicine is of particular importance. To achieve this, ten batches of Fructus schisandrae chinensis were collected from Tieli, in China, and their chemical components were analyzed under optimized HPLC conditions. On the basis of the chromatographic data, a consistent HPLC fingerprint pattern containing 20 common peaks was obtained. Among these common peaks, six were identified as schizandrin, schizandrol B, schisantherin, deoxyschiandrin, gamma-schizandrin B, and schizandrin C. On the basis of this HPLC fingerprint and principal-components analysis, the quality of fifteen samples from different producing areas of China was objectively assessed, and the species difference between Fructus schisandrae sphenantherae and Fructus schisandrae chinensis was clearly differentiated. To summarize, the data described in this study offer valuable information for quality control and proper use of Fructus schisandrae chinensis.
