Abstract:Grapes, particularly the species Vitis vinifera L., are one of the most widely grown crops in the world. Winemaking processes generate a high amount of residues, which currently pose an environmental and economic sustainability problem for companies in the sector. For this reason, solutions are being explored for the development of new products with high-added value derived from the valorization of these residues. One of the wastes produced by winemaking processes is grape pomace, which chemical composition is promising because it is rich in compounds with high antioxidant activity, such as polyphenols (anthocyanins, flavonols, flavan-3-ols, procyanidins), phenolic acids, resveratrol, and fiber. Commonly grape pomace is used to produce distillates and to extract tartaric acid and coloring substances such as enocyanin. Recently, alternative uses of grape pomace have been adopted, such as the production of extracts with antioxidant properties, fermentation substrates, composting and biomass for energy production, and fiber extraction for the development of high-value-added products enriched with bioactive molecules from grape pomace. Here, we discuss how bioactive molecules from grape pomace are involved in various human biological functions and their applications in the agri-food sector.

Valorization of Bioactive Compounds from Lingonberry Pomace and Grape Pomace with Antidiabetic Potential

Abstract:In recent years, increased attention has been paid to the recovery of bioactive compounds from waste and by-products resulting from the agro-industrial sector and their valorization into new products, which can be used in the health, food, or agricultural industry, as innovative and sustainable approaches to waste management. In this work, two of these by-products resulting from the fruit-processing industry were used for the recovery of bioactive compounds (polyphenols), namely lingonberry pomace (Vaccinium vitis-idaea) and grape pomace (Vitis vinifera). Two green extraction techniques were employed to obtain hydroalcoholic extracts (solvent: 50% EtOH, 10% mass): ultrasound-assisted extraction (UAE) and accelerated solvent extraction (ASE). The extracts were subjected to micro- and ultrafiltration processes, and further analyzed to determine the bioactive compound content through spectrophotometric (UV-Vis) and chromatographic (HPLC-PDA) methods. Additionally, the extracts exhibited significant enzyme inhibition, particularly against α-amylase and β-glucosidase, suggesting potential anti-diabetic properties. The extracts characteristics, polyphenolic content, antioxidant capacity and enzyme inhibitory ability, were statistically compared, and significant differences were found between the two extraction methods. The grape pomace concentrated extracts showed a pronounced inhibitory activity on both analyzed enzymes compared to the lingonberry pomace concentrated extracts, closer to the standard used; e.g., IC50 α-amylase = 0.30 ± 0.01 μg/mL (IC50 acarbose = 0.3 ± 0.01 μg/mL), IC50 α-glucosidase = 0.60 ± 0.01 μg/mL (IC50 acarbose = 0.57 ± 0.02 μg/mL). These findings highlight the potential of agro-industrial residues as bioactive compound resources, with their valorization through application in food, nutraceutical, or pharmaceutical industries therefore contributing to the sustainable development and promotion of circular economy principles with the recovery of valuable inputs from plant by-products.

Abstract:From a circular economy perspective, the appropriate management and valorization of winery wastes and by-products are crucial for sustainable development. Nowadays, grape pomace (GP) has attracted increasing interest within the food field due to its valuable content, comprising nutritional and bioactive compounds (e.g., polyphenols, organic and fatty acids, vitamins, etc.). Particularly, GP polyphenols have been recognized as exhibiting technological and health-promoting effects in different food and biological systems. Hence, GP valorization is a step toward offering new functional foods and contributing to solving waste management problems in the wine industry. On this basis, the use of GP as a food additive/ingredient in the development of novel products with technological and functional advantages has recently been proposed. In this review, we summarize the current knowledge on the bioactivity and health-promoting effects of polyphenolic-rich extracts from GP samples. Advances in GP incorporation into food formulations (enhancement of physicochemical, sensory, and nutritional quality) and information supporting the intellectual property related to GP potential applications in the food industry are also discussed.

Abstract:Grape pomace is the main by-product of vine-winery chains. It requires adequate treatment and disposal but is also an economically underused source of bioactive plant secondary metabolites. This study aimed to investigate the antibacterial effects of polyphenolic extracts from Aglianico (Vitis vinifera L.) grape pomace. In particular, hydroethanolic extracts obtained via an ultrasonic-assisted extraction technique were selected for antimicrobial tests. The extracts were screened for their antibacterial effects against foodborne pathogens that were both Gram-positive, in the case of Staphylococcus aureus and Bacillus cereus, and Gram-negative, in the case of Escherichia coli and Salmonella enterica subsp. enterica serovar Typhimurium, showing variable bacteriostatic and bactericidal effects. In addition, our results demonstrated that the tested grape pomace extracts can reduce the inhibitory concentration of standard antibiotics. Interestingly, selected extracts inhibited biofilm development by S. aureus and B. cereus. Overall, these new insights into the antibacterial properties of grape pomace extracts may represent a relevant step in the design of novel therapeutic tools to tackle foodborne diseases, and in the management of resistant biofilm-related infections.

Abstract:Grape pomace is one of the most abundant solid by-products generated during winemaking. A lot of products, such as ethanol, tartrates, citric acid, grape seed oil, hydrocolloids, bioactive compounds and dietary fiber are recovered from grape pomace. Grape pomace represents a major interest in the field of fiber extraction, especially pectin, as an alternative source to conventional ones, such as apple pomace and citrus peels, from which pectin is obtained by acid extraction and precipitation using alcohols. Understanding the structural and functional components of grape pomace will significantly aid in developing efficient extraction of pectin from unconventional sources. In recent years, natural biodegradable polymers, like pectin has invoked a big interest due to versatile properties and diverse applications in food industry and other fields. Thus, pectin extraction from grape pomace could afford a new reason for the decrease of environmental pollution and waste generation. This paper briefly describes the structure and composition of grape pomace of different varieties for the utilization of grape pomace as a source of pectin in food industry.

Abstract:Grape pomace (GP), a major byproduct obtained from the winemaking process, is characterized by a high amount of phenolic compounds and secondary plant metabolites, with potential beneficial effects on human health. Therefore, GP is a source of bioactive compounds with antioxidant, antimicrobial, and anti-inflammatory activity. As people are paying more attention to sustainability, in this work, we evaluate two different extractions (aqueous and hydroalcoholic) of GP bioactive compounds. In vitro simulated gastrointestinal digestion of the GP extracts was performed to improve the bioavailability and bioaccessibility of polyphenols. The antioxidant activity (ABTS and DPPH assays) and the phenolic characterization of the extracts by UHPLC-DAD were evaluated. The antimicrobial effects of GP antioxidants in combination with a probiotic (Lactiplantibacillus plantarum) on the growth of pathogenic microorganisms (Escherichia coli, Bacillus megaterium, and Listeria monocytogenes) were evaluated. As a result, an increase of antioxidant activity of aqueous GP extracts during the gastrointestinal digestion, and a contextual decrease of hydroalcoholic extracts, were detected. The main compounds assessed by UHPLC-DAD were anthocyanins, phenolic acids, flavonoids, and stilbenes. Despite lower antioxidant activity, due to the presence of antimicrobial active compounds, the aqueous extracts inhibited the growth of pathogens.

Abstract:Grape pomace is commonly considered a waste product of monovarietal red wine production. Methods: HPLC-DAD analysis was performed to determine the polyphenol and flavonoid contents of all the extracts obtained from Montepulciano d’Abruzzo red wine and grape skins whereas, GC-MS was applied to the determination of fatty acid composition in grape seeds oil. Biological characterization involves antioxidant and antimicrobial assays for all the extracts and seeds oil; Their ability to inhibit α-glucosidase, α-amylase, α-tyrosinase, and ChE enzymes was also detected, together with anti-inflammatory activity on wine, grape skin extracts, and seeds oil by lipoxygenase (5-LOX) and LPS-stimulated macrophage release assays. Data indicate significative polyphenols content (199.31 ± 7.21 mgGAE/g), antioxidant (CUPRAC assay (1036.98 mgTE/g)), enzymatic inhibition (α-tyrosinase: 151.30 ± 1.20 mgKAE/g) and anti-inflammatory activities for wine-organic extract 2, while the antimicrobial activity of grape skin decoction is higher than those reported by wine extracts on three bacterial strains. Interestingly only dealcoholized wine and wine-aqueous extract exerts inhibitory effects on α-glucosidase (20.62 ± 0.23 mmolACAE/g and 19.81 ± 0.03 mmolACAE/g, respectively), while seeds oil is rich in oleic and linoleic acids. These results confirm the strong antioxidant properties of Montepulciano d’Abruzzo grape pomace, suggesting the potential use of this waste product as functional food supplements in the human diet and in cosmeceutics.

Abstract:Grape is one of the most well-known fruits worldwide, being transformed into distinct food products, one of which is wine. A significantly large portion of grape production is exclusively destined for winemaking which, as an industrial process, produces significant amounts of by-products. Grape pomace is considered the major by-product of winemaking and consists of skins, seeds and stems that are left behind after the stage of grapes' pressing. According to the Waste Framework Directive (Directive 2008/98/EC) of the European Parliament, "waste prevention should be the first priority of waste management and re-use and material recycling should be preferred to energy recovery from waste". This review aims to approach the recent advances over the valorization of grape pomace as a value-adding material, exploring its potential uses in the industry. The main methods for this purpose are discussed, including traditional methods (such as distillates production, animal feed, and soil fertilizer), its use as a technological aid in different industrial processes (e.g., adsorption, immobilization, food packaging and cosmetics), its addition in food products, highlighting its potential effects on physicochemical, functional and sensory attributes of these products, and as a raw material for bioenergy production. The large number of strategies that have been employed for the possible use of grape pomace show its high revalorization potential, with studies being conducted on several different food products of different categories, such as dairy, meat, fish and bakery products or even wine itself. The main positive effect of this addition consists the increase of the antioxidant activity of the medium, contributing towards enhanced shelf-life, both microbiologically and physicochemically. Given the fact that a linear production model is not satisfactory for sustainable development and focus has been shifted towards circular models, it appears that grape pomace consists a major value-adding component that can promote such economy schemes, as it can be reutilized in numerous ways across a number of industrial categories, including the winery that it came from. However, this study highlighted also the absence of a comprehensive legislative framework covering the addition of grape pomace in foods, of studies analyzing completely the cost of grape pomace revalorization, and of specific research on green/food-grade extraction methods of valuable compounds from grape pomace.

Abstract:Grape pomace (GP) is a major byproduct worldwide, and it is well known for its bioactive compounds, such as fibers and phenolic compounds, that are popular for their impact upon human health, including gastrointestinal health. The objective of this work was to evaluate the chemical composition and biological activities of an enzymatic GP extract, as well as to investigate how gastrointestinal digestion (GID) modulates these properties. GP extract was previously produced using an enzymatic cocktail with xylanase activity and was then exposed to simulated conditions of GID, characterized for its chemical composition, and screened for antimicrobial, prebiotic, and antioxidant activities. The safety of this ingredient after GID was also assessed. GP extract presented high contents of dietary fiber and other carbohydrates, including xylooligosaccharides, in addition to minerals and phenolic compounds. In vitro simulated GID revealed that xylobiose was resistant to gastric conditions, unlike phenolic compounds. The use of 2% (w/v) of this ingredient proved to be a potential carbon source that could be fermented by Lactobacillus and Bifidobacterium spp, even after digestion. The extract also exhibited strong antioxidant and antimicrobial activities against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa; however, after GID, the antioxidant capacity decreased, and the antimicrobial capacity was strongly reduced or lost. Furthermore, the extract safety was also guaranteed on Caco-2 intestinal cells. This novel and green GP extract proved to be composed of relevant bioactive molecules, including xylooligosaccharides, polyphenols, organic acids, and minerals, which provided different biological properties; it has potential applications in the food industry such that it can be used as an ingredient in the development of new functional foods.

Abstract:The wine industry produces large amounts of grape pomace (GP), a waste that needs to be disposed of properly. Bioactive compounds with high added value can be recovered from GP as an interesting strategy to reduce the environmental impact. Here, two different technologies were employed to recover polyphenol compounds from GP: microwave hydrodiffusion and gravity (MHG) and ultrasound-assisted extraction (UAE). The further purification of UAE and MHG extracts was carried out through solid-phase extraction (SPE) to obtain three fractions, F1, F2 and F3. ATR-FTIR analysis confirmed the presence of sugar and polysaccharide components in F1, as well as non-anthocyanin and anthocyanin compounds in F2 and F3, respectively. Also, the chemical profile was determined by HPLC-UV-DAD, identifying the presence of catechin in F2, and malvidin-3-O-glucoside chloride and cyanidin chloride derivative as the main anthocyanin compounds in F3. The fractions and their parental extracts were characterized for total phenolic content (TPC) and scavenger activity by in vitro assays. We found that F2-MHG and F3-MHG contained phenol contents 6.5 and 8.5 times higher than those of the parental non-fractionated extracts. Finally, F3-MHG (100 μg/mL, w/v) was shown to reduce the proliferation of HT-29 cells.

Abstract:Raspberry fruit pomace, a byproduct of juice production, was studied as a potential source of antioxidant compounds. Target high-performance liquid chromatography analysis of important polyphenolic compounds (gallic, p-coumaric, caffeic, quercitrin, chlorogenic, and ellagic acid) was performed together with analysis of total phenolic content (TPC), total flavonoid content (TFC), total anthocyanins content (TAC), and antioxidant capacity (via ferric reducing antioxidant power (FRAP) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays). The differences in polyphenolic content of Rubus idaeus L. pomace were evaluated following ultrasound-assisted extraction and conventional maceration with different organic solvents. Additionally, the yield of free phenolics was measured in hydrolyzed pomace extracts. The results obtained show that the ultrasound method maximizes the quantity of antioxidant compounds in terms of TPC (27.79 mg/L gallic acid equivalent (GAE)), TFC (8.02 mg/g quercetin equivalent (QE)), TAC (7.13 mg/L cyanidine-3-glucoside equivalent (C3G Eq)), caffeic (19.17 μg/g), chlorogenic (3.56 μg/g), ellagic (105.52 μg/g), and gallic acids (8.75 μg/g), as well as FRAP (1002.72 μmol/L) and DPPH assays (969.71 μmol/mL vitamin C equivalent (vit C Eq); 567.00 μmol/100 g Trolox equivalent (TE)). On the other hand, conventional maceration maximizes the yield of quercetin and p-coumaric acid. In terms of biowaste valorization, raspberry fruit pomace has a promising industrial potential and may prove to be useful in the development of antioxidant dietary supplements.

Abstract:In the last few years, literature data have reported that health status is related to the consumption of foods rich in polyphenols, bioactive compounds found in the plant world, in particular in vegetables and fruit. These pieces of scientific evidence have led to an increase in the demand for functional foods and drinks enriched in polyphenols, so that plant materials are more and more requested. The availability of food and agricultural wastes has adverse effects on the economy, environment, and human health. On the other hand, these materials are a precious source of bioactive compounds as polyphenols. Their recovery and reuse from wastes are according to the circular economy strategy, which has introduced the "zero waste concept." However, the process is convenient from an economic and environmental point of view only if the final products are standardized and obtained using sustainable and industrial technologies. In this panorama, this paper describes an industrial and sustainable platform for the production of micronized powders and extracts enriched in polyphenols from Olea europaea L. and Vitis vinifera L. wastes that are useful for food, cosmetics, and pharmaceuticals sectors. The platform is based on drying plant materials, extraction of polyphenols through membrane technologies with water, and, when necessary, the concentration of the final fractions under vacuum evaporation. All powders and extracts were characterized by high-performance liquid chromatography-diode array detector-mass spectrometry analysis to define the qualitative and quantitative content of bioactive compounds and insure their standardization and reproducibility. The chromatographic profiles evidenced the presence of secoiridoids, flavones, flavonols, anthocyanins, hydroxycinnamic acids, catechins, and condensed tannins. An overview of the biological activities of the main polyphenols present in Olea europaea L. and Vitis vinifera L. powders and extracts is reported because of biomedical applications.

Abstract:In the framework of introducing green strategies for food processing, the industrial orientation has shifted towards the replacement of conventional petroleum-based solvents with alternative eco-friendly ones. On this basis, the objective of this study was to synthesize a novel, tertiary, food-grade deep eutectic solvent, composed of glycerol, citric acid, and L-proline (GL-CA-Pro), and to test it as a solvent for the extraction of polyphenols from agri-food waste biomass. After an initial screening on various common residual materials (apple peels, lemon peels, orange peels, red grape pomace, olive leaves), evidence emerged that indicated GL-CA-Pro was more effective than other DESs commonly used for polyphenol extraction. Furthermore, extracts from red grape pomace (RGP) and olive leaves (OLL) were shown to contain higher level of total polyphenols and increased antioxidant activity. Process optimization for those two materials with the response surface methodology revealed that the major difference pertained to the extraction time. In addition, for both materials, GL-CA-Pro was shown to provide higher total polyphenol yields (53.25 and 42.48 mg gallic acid equivalents per g of dry mass, respectively) compared to water and 60% aqueous ethanol. However, the chromatographic analyses for OLL suggested aqueous ethanol was a more suitable solvent for some principal polyphenolic constituents. The RGP extract produced with GL-CA-Pro exhibited significantly stronger antioxidant effects compared to the aqueous and hydroethanolic extracts, but the outcome for the OLL extracts was diversified. It was concluded that GL-CA-Pro is a very efficient solvent for RGP polyphenols, but its efficiency regarding OLL was comparable to that of aqueous ethanol.

Abstract:Agri-food by-products, obtained as waste from the food industry, negatively impact the global economy and the environment. In order to valorize waste materials from fruit juices and tomato sauces as upcycled materials rich in health-promoting compounds, they were characterized in terms of polyphenolic and protein content. The results obtained were compared with those collected for their final products. The recovery of polyphenols was performed via ultrasound-assisted extraction (UAE). A high-performance liquid chromatography–diode array detector (HPLC-DAD) method was developed and validated to depict the quali-quantitative polyphenolic profile of both the by-products and the final products. The antioxidant capacity of the resulting extracts was characterized by UV-Vis spectrophotometric assays in terms of total phenolic content (TPC) and total antioxidant status (TAS). Moreover, the protein content was assessed with the Kjeldahl method too. The results highlighted a significant quantity of polyphenols remaining in peach, apricot, and apple by-products, which were able to exert an antioxidant activity (in the range of 4.95 ± 5.69 × 10−1 to 7.06 ± 7.96 × 10−1 mmol Trolox 100 g−1 of dry weight (DW) sample). Conversely, the tomato by-products were highly rich in proteins (11.0 ± 2.00 to 14.4 ± 2.60 g of proteins 100 g−1 DW). The results proved that all by-products may potentially be sustainable ingredients with nutritional and functional value in a circular bio-economy prospect.

Abstract:Traditionally, wine production has been assumed as an environmentally friendly process. However, it requires a considerable amount of resources (e.g. water, fertilisers and organic amendments), and, at the same time, producing a large amount of waste and by‐products. Environmental analysis of the wine making process reports that organic solid wastes and wastewater represent the main effluents of the sector. This review aims to make an overview about the recent insights and results related to valorisation of grape pomace and how this approach, considering the number of dedicated studies, is now reaching its maturity. By‐products valorisation, a concept increasingly consolidated in the field of industrial residues management, holds immense potential for prospective production. In this regard, the valorisation of grape pomace is proving its effectiveness, obtaining remarkable results. Traditionally, wine production has been considered as an environmentally friendly process. However, it requires a considerable amount of resources and, on the other hand, produces a large amount of wastewater and organic wastes. The number of published works exploring alternative ways to valorize grape pomace has been increasing since 2013. By‐products valorisation holds immense potential for prospective production.

Abstract:The wine industry generates a large amount of waste, such as grape pomace, which contains compounds of interest. In this study, the ecological technologies of ultrasonic-assisted extraction (UAE) and microwave-assisted extraction (MAE) were investigated to recover compounds of interest from the grape by-product. A fractional factorial design was used to explore the best extraction conditions and Box-Behnken response surface (MSR) methodology was performed for optimization using water as solvent. The best extraction conditions (time: 15 min, temperature: 65 °C, and amplitude: 70 %) resulted in the highest concentrations of phenolic compounds, anthocyanins, and antioxidant activity by the ABTS and DPPH assay. These two green technologies, ultrasound and microwaves and water as the solvent, applying a statistical optimization model allowed to improve extraction yield sustainably to find the best extraction conditions. The compounds extracted have the potential to be used in the food industry.

Abstract:The winemaking sector is one of the most productive worldwide, and thus it also generates large amounts of by-products with high environmental impacts. Furthermore, global market trends and government regulations promote industrial alternatives based on sustainable production processes. As a result, several studies have focused their attention on the reuse of grape by-products in the agro-food chain. Vine shoots, grape stalks, and wine lees, although produced to a lesser extent than grape pomace, have increasingly been receiving attention for their applications in the food sector, since they are a good source of functional and bioactive compounds. In this framework, our review highlights the promising results obtained by exploiting the antioxidant and/or antimicrobial activity of vine shoots, grape stalks, and wine lees or their extracts to replace the most common oenological additives and to assay the activity against food pathogens. Further, innovative functional foods and sustainable food packaging have been formulated by taking advantage of polyphenols and fiber, as well as plant bio-stimulants, in order to obtain grapes and wines with high quality characteristics. Overall, these by-products showed the potential to be recycled into the food chain as functional additives for different products and applications, supporting the sustainability of the winemaking sector.

Grape Pomace Valorization: A Systematic Review and Meta-Analysis

Abstract:This systematic review aimed to collect data and analyze the possible use of grape pomace, a winemaking industry byproduct, in the production of fortified foods. The English articles found in Web of Science, Scopus, and Google Scholar, from January 2006 until May 2020, were used for the conduction of overview tables and meta-analysis. The systematic review emphasized the two main issues concerning grape pomace application to other food products: (i) grape pomace contains high amounts of health promoting compounds; and (ii) the use of grape pomace is influencing the waste management. The grape pomace has been used in the fortification of plant origin food, meat, fish, and dairy products, mainly due to higher polyphenols and dietary fiber contents. The fortification was declared as successful in all studied food types. The change of color, caused by polyphenolic compounds, was mainly observed as an adverse effect of the fortification. Higher levels of fortification also caused notable undesirable changes in texture. The most valuable influence of the grape pomace addition according to included papers and meta-analysis is certainly a higher nutritional quality and oxidative stability of fortified products, reflected as higher polyphenol and total dietary fiber content.
