Abstract:Aflatoxin M1 (AFM1) is the main secondary metabolites of aflatoxin B1 (AFB1) framed in the liver and discharged into milk when people and animals polish off AFB1-polluted food. The persist from feed to milk in dairy cows is affected by different dietary and physiological elements, including taking care of regimens, rate of ingestion, rate of absorption, animal health, hepatic biotransformation limit, and real milk production of the animals. AFM1 exposure might cause both intense and constant toxicity. In humans, AFM1 exposure is related with cancer-causing nature, genotoxicity, mutagenicity, and teratogenicity. In dairy animals, persistent openness to AFs can decrease execution, debilitate liver capability, compromise invulnerable capability, and increment sickness susceptibility. AFM1 openness represents a worry for the worldwide populace, especially for babies and youngsters who drink milk in bounteous amounts and thus, are more powerless to unfavorable impacts. Occurrence of Aflatoxin B1 in Total Mixed Rations and Aflatoxin M1 in Raw and Commercial Dairy Milk in Northern China During Winter Season

Abstract:This study aims to investigate the occurrence of aflatoxin B1 (AFB1) in total mixed rations (TMR) and aflatoxin M1 (AFM1) in raw and commercial milk in northern China during winter season. A total of 22 feed samples, as well as the corresponding 84 raw milk and 69 commercial milk samples were randomly collected from north-west, northeast and north areas of China from November 2019 to January 2020. High performance liquid chromatography and enzyme linked immunosorbent assay method were applied to detect AFB1 and AFM1. All samples presented AFB1 (30?370 ng/L) and AFM1 (10?430 ng/L) levels under the legal limit of China for raw milk (10 ?g/L) and commercial milk (500 ng/L). While 31.8% of TMRs and 34.5% of raw milk samples exceeded the European Union (EU) legal limit for AFM1 in raw milk (5 ?g/L) and commercial milk (50 ng/L). Compared with the samples from northeast and north areas of China with the similar AFB1 concentration, samples from northwest area of China exhibited higher AFM1 level (mean = 130 ng/L), thus presenting a higher transfer ratio from AFB1 to AFM1. A Linear relationship was observed between AFB1 in TMRs and AFM1 in raw milk. In conclusion, all samples from northern China presented AFB1 and AFM1 levels under Chinese legal limit and only about 30% of the tested samples exceeded the EU legal limit. However, the aflatoxin contamination in northwest area of China should be paid more attention to due to the high AFM1 levels and high variance in milk samples.

Abstract:The objective of this study was to investigate the occurrence of aflatoxin M-1 (AFM(1)) in Chinese liquid milk products. A total of 190 liquid milk samples, including 168 ultrahigh-temperature-treated milk samples and 22 pasteurized milk (PM) samples, were collected in August 2019. A screening assay with the Charm rapid test kit and a confirmation method with high-performance liquid chromatography were used for AFM(1) analysis. Nine (4.74%) samples were screened positive, of which 5 (2.11%) samples were confirmed with concentration levels of 0.022 to 0.049 mu g/kg. The AFM(1) levels confirmed were all below the maximum residue levels set by China, the European Union, the United States, and the Codex Alimentarius Commission. The detection rate of AFM(1) in domestic milk samples was 3.39%, while no AFM(1) was detected in samples of imported milk. The prevalences of AFM(1) detected in three groups of brands were as follows: group I, the major brands of China. 2.70%; group II, the local city domestic brands, 4.55%; and group III, the brands imported into China, 0. The detection rate of AFM(1) was significantly higher in PM samples (9.09%) than in ultrahigh-temperature-treated samples (1.19%) (P < 0.05). Although the residue level of AFM(1) did not exceed the maximum residue levels in any of the samples, the higher detection rate in local Chinese brands, especially in PM samples, deserves the attention of the Chinese government and consumers.
Prevalence and concentration of aflatoxin M1 in breast milk in Africa: a meta-analysis and implication for the interface of agriculture and health

Abstract:Breast milk is one of the many distinct forms of food that can be contaminated with aflatoxin M 1 (AFM 1 ). They may be consumed by eating contaminated foods, such as contaminated meat and crops, which would then be present in breast milk and cause health problems, including nervous system disorders and cancers of the lungs, liver, kidneys, and urinary tract. However, the prevalently inconsistent explanation of prevalence and concentration remains a big challenge. Thus, this meta-analysis was conducted to determine the prevalence and concentration of harmful chemicals in breast milk in an African context. The databases MEDLINE, PubMed, Embase, SCOPUS, Web of Science, and Google Scholar were searched for both published and unpublished research. To conduct the analysis, the collected data were exported to Stata version 18. The results were shown using a forest plot and a prevalence with a 95% confidence interval (CI) using the random-effects model. The Cochrane chi-square (I2) statistics were used to measure the studies' heterogeneity, and Egger's intercept was used to measure publication bias. This review included twenty-eight studies with 4016 breast milk samples and newborns. The analysis showed the overall prevalence and concentration of aflatoxin M 1 in breast milk were 53% (95% CI 40, 65; i 2 = 98.26%; P = 0.001). The pooled mean aflatoxin M 1 concentration in breast milk was 93.02 ng/l. According to this study, the eastern region of Africa was 62% (95% CI 39–82) profoundly affected as compared to other regions of the continent. In subgroup analysis by publication year, the highest level of exposure to aflatoxins (68%; 95% CI 47–85) was observed among studies published from 2010 to 2019. This finding confirmed that more than half of lactating women's breast milk was contaminated with aflatoxin M 1 in Africa. The pooled mean aflatoxin M 1 concentration in breast milk was 93.02 ng/l. According to this study, the eastern region of Africa was profoundly affected compared with other regions. Thus, the government and all stakeholders must instigate policies that mitigate the toxicity of aflatoxins in lactating women, fetuses, and newborns.

Abstract:Aflatoxin M1 (AFM1) contamination of milk affects the general population with particular attention to children who frequently consume milk as part of complementary food. This study determined AFM1 contamination of cow's milk and estimated the health risk of dietary AFM1 through consumption of cow's milk among children (6 to 36 months) in the Magadu ward of Morogoro region in Tanzania. A total of 165 mother-baby pairs were recruited and interviewed on child feeding practices with a focus on feeding of cow's milk in the past 24 h. Alongside the interview, 100 raw cows' milk samples were collected from subsampled respondent households and were analyzed for AFM1 using enzyme-linked immunosorbent assay (ELISA). The results showed that about 35% of the surveyed children consumed cow's milk in the form of plain milk, incorporated in porridge and/or tea. The amount consumed varied from 62.5 to 500 mls with a median of 125 (125, 250) mls at a frequency of 1 to 2 times a day. All raw cows' milk (100%) samples ( n = 100) were found contaminated with AFM1 at concentrations ranging from 0.052 to 9.310 μg/L and median of 2.076 μg/L (1.27, 2.48). All samples were contaminated by AFM1 at levels above the limits of 0.05 μg/L of raw milk set by the Tanzania Bureau of Standard and the European Union, while 97% exceeded 0.5 μg/L set by the US Food and Drug Administration. Exposure to AFM1 due to consumption of cow's milk ranged from 0.0024 to 0.077 μg/kg bw per day with a median of 0.019 (0.0016, 0.026) μg/kg bw per day, while the margin of exposure (MOE) ranged from 5.19 to 166.76 and median 20.68 (15.33, 25.40) implying high risk of public health concern. This study recommends that advocacy on consumption of cows' milk to combat undernutrition in children should consider a holistic approach that considers the milk's safety aspect.

Abstract:Aflatoxin M1 (AFM1), a group 1 carcinogen, is a risk factor to be monitored in milk. This study aimed to investigate the occurrence of AFM1 in milk in Xinjiang, China, and to assess the risk of exposure for milk consumers in different age-sex groups. A total of 259 milk samples including pasteurized milk (93 samples), extended-shelf-life (ESL) milk (96), and raw donkey milk (70) were collected in Xinjiang from January to March in 2022. The AFM1 content of the milk samples was detected using a validated ELISA method. Of the 259 total samples analyzed for AFM1, 84 (32.4%) samples were contaminated at levels greater than the detection limit of 5 ng/L, with the maximum level of 16.5 ng/L. The positive rates of AFM1 in pasteurized milk and ESL milk were 43.0% (n = 40) and 45.8% (n = 44), respectively, and AFM1 was undetectable in donkey milk. The estimated daily intakes of AFM1 in each age group were lower than the hazard limits and were similar between male and female milk consumers. Therefore, the AFM1 contamination of milk in Xinjiang is low but still needs to be continuously monitored considering that children are susceptible to AFM1.

Abstract:Aflatoxin M1 (AFM(1)) is a main cause of hepatocarcenogenoma in Chinese population. Measurement of aflatoxin exposure in human may help in providing clear evidence for the exposure of specific environmental pollutants in certain population. "One child policy" in China offered parents more careful to choose safe food during pregnancy, but no reports published on the efficacy of their endeavor. In present study, we aimed to assess the exposure of AFM(1) in Chinese pregnant women. The urine samples were collected from 600 volunteers from Zhejiang province, China and the urinary concentration of AFM(1) was measured using ELISA kit. AFM(1) was detected in 84% of the pregnant women. The geometric mean and 95th percentile concentration of AFM(1) in pregnant women were 50.3 ng/L and 633.5 ng/L, respectively. Our results point out that pregnant women especially are at the high risk of exposure to AFM(1). Our results also indicate that although "one child policy" offered parents to pay more attention for the selection of safe food, but detection of AFM(1) in urine of pregnant women indicate that more foods containing AFM(1) still need to be detected. Highest exposure of AFM(1) in pregnant women indicates that awareness campaigns must be started especially in the rural areas of China regarding the possible hazardous effects of AFM(1) exposure in pregnant women.

Abstract:Abstract Aflatoxin M 1 (AFM 1 ) is a major metabolite of aflatoxin B 1 occurring in many feed materials commonly used in the ration of dairy cows. The level of contamination of feed materials is currently increasing in many parts of the world due to climate change. These changes emphasise the necessity to monitor AFM 1 levels in milk and dairy products as a precautionary measure to protect consumer health risk. In the current study, the AFM 1 levels were measured in raw, pasteurised, and ultra-high temperature processed (UHT) milk commercially available in Thailand. In total, 900 milk samples were analysed, making this investigation one of Thailand’s first large-scale monitoring of milk contamination. Over a period of three consecutive years (2019 to 2021), each year 100 individual samples of either AFM 1 , pasteurised and UHT milk were collected, extracted using an immunoaffinity column for AFM 1 and then quantified by a validated liquid chromatography analysis with fluorescence detection. The measured AFM 1 levels ranged between 65-1,810 ng/l, 12-87 ng/l, and 7-38 ng/l in the raw, pasteurised, and UHT milk, respectively, in 2019. In 2020, the corresponding AFM 1 concentration range was 58-1,232 ng/l, 15-65 ng/l, and 7-29 ng/l and 52-1,432 ng/l, 20-59 ng/l, and 7-33 ng/l in 2021, respectively. According to international guidance documents, a direct comparison and formal risk analysis revealed that the measured AFM 1 levels of the processed milk samples showed good compliance with the US regulatory limits and the Codex Alimentarius Commission recommendations. However, the latter applies particularly to milk and dairy products intended for the consumption of infants and young children, considering their relatively high consumption of milk and dairy products and the specific sensitivity of infants and toddlers under the age of 6 years to mutagenic and genotoxic contaminants in foods.

Abstract:The study gathered publications from 2003 to 2018 reporting the occurrence of aflatoxin M1 (AFM1) in samples of milk and milk products from Latin America. Results showed that in 3547 milk and 969 dairy samples, the percentage of incidence and concentration levels for AFM1 were of 67% and 0.001–23.10 μg/kg for milk and 63% and 0.001–18.12 μg/kg for dairy products, respectively. In addition, the highest estimated daily intakes (EDIs) of AFM1 were obtained for Brazil, Costa Rica, Colombia, and Mexico (2.4, 1.0, 1.2 and 20.9 ng/kg bw/day, respectively). However, all average DI values calculated for the analyzed studies reported in the period of 15 years for the countries of Latin America were above that calculated by Joint FAO/WHO Expert Committee for Latin American countries (0.058 ng/kg bw/day).

Abstract:Aflatoxins (AFL) are fungal metabolites that can contaminate animal feed, causing toxic effects after consumption. Among the AFL, there are reports of the occurrence of aflatoxin M1 (AFM1) in milk, so the objective of this work was to evaluate the AFM1 content in bovine milk samples produced in the State of Amazonas-Brazil. Of the analyzed samples (n=30), 20% were found to be contaminated with AFM1 in the range of 0.093-0.320 μg/kg (Limit of detection= 0.0625 μg/mL). No sample exceeded current Brazilian legislation, but all exceeded the maximum European regulatory limit of 0.05 μg/kg. Risk analysis estimated a daily intake of 5.357 x 10-7 mg/kg body weight/day for men and 6.257 x 10-7 mg/kg body weight/day for women. The MOE result did not indicate concern in public health aspects. On the other hand, the occurrence of AFM1 in the state of Amazonas was observed in an unprecedented way and need further investigation including other dairy products.

Abstract:Introduction In Sidama, Ethiopia, animal-source foods can be difficult to access. Milk has important nutrients for child growth, but carries the risk of aflatoxin M 1 (AFM 1 ) contamination. AFM 1 is a metabolite of the mycotoxin aflatoxin B 1 (AFB 1 ) in dairy feed; cows secrete AFM 1 in milk when their feed contains AFB 1 produced by Aspergillus fungi in maize, nuts and oilseeds. It is unknown whether AFM 1 compromises child growth and health. Methods and analysis This protocol paper describes our study in Sidama to determine the impact of milk consumption and AFM 1 on child growth in the first 18 months of life. We will collect baseline and end-line data on dairy production, socioeconomic and nutritional factors of 1000 dairy-owning households with children ages 6–18 months at baseline; and gather samples of milk and dairy feed and child anthropometrics. We will conduct phone interviews every 6 months to ascertain changes in practices or child health. Dairy feed will be tested for AFB 1 ; milk for AFM 1 , pathogens and nutrients. Controlling for herd size, socioeconomic, nutritional and behavioural factors, we will determine the association between child anthropometrics and milk consumption, as well as AFM 1 exposure. We will examine whether AFM 1 exposure affects child growth in the first 18 months of life, and weigh the benefits and risks of milk consumption. Ethics and dissemination The protocol is approved by the Institutional Review Boards of the Ethiopian Public Health Institute (EPHI-IRB-481–2022), Michigan State University (STUDY00007996) and International Food Policy Research Institute (DSGD-23–0102). Written informed consent will be obtained from all participants, who may withdraw from the study at any time. Confidentiality of collected data will be given high priority during each stage of data handling. The study's findings will be disseminated through stakeholder workshops, local and international conferences, journal articles and technical reports.

Abstract:Aflatoxin M1 (AFM1) is a major carcinogenic compound found in milk and dairy products, posing a constant risk to consumers in the Mediterranean region. This study systematically reviewed AFM1 presence in these products in Mediterranean countries over a period of 20 years and estimated the relevant nutritional exposure for consumers. Using data from three databases, 596 articles were retrieved with 123 meeting the inclusion criteria. The frequency of AFM1 occurrence was 40% regardless of milk type, with non-cow milk showing an increasing trend. Moreover, the Estimated Daily Intake of AFM1 from milk and dairy product consumption and the associated Hazard Index were determined. Notably, Southeastern Mediterranean countries presented higher contamination levels, leading to elevated Estimated Daily Intake and Hhazard Index values. Thermally processed milk showed high contamination levels, and among dairy products, cheeses were the most contaminated. This review highlights AFM1 as a persistent hazard in the dairy sector, underscoring the need for effective interventions throughout the milk production stages.
Evaluation of Aflatoxins Occurrence and Exposure in Cereal-Based Baby Foods: An Update Review

Abstract:Purpose of review: The first stages of human life, which include the fetal period, infancy, and early childhood, are the most critical for human growth and development. This is the most vulnerable phase to health challenges due to the immature immune system and rapid development. Mycotoxins such as aflatoxins, ochratoxin A, patulin, fumonisins, zearalenone, and deoxynivalenol are secondary metabolites secreted by various fungal species, primarily Aspergillus, Fusarium, Penicillium, and Alternaria. Aflatoxins are one of the major mycotoxins produced in cereals and cereal-based foods by several species of Aspergillus, mainly Aspergillus flavus. In this context, this review provides a brief overview of the occurrence, exposure, legal regulations, and health effects of aflatoxins (B1, B2, G1, G2, and M1) in cereal-based baby foods and breast milk. Recent findings: Human aflatoxin exposure in utero and through breast milk, infant formulas, cereals, and cereal-based foods has been linked to various health consequences, including adverse birth outcomes, impaired growth and development, immune system suppression, and hepatic dysfunction. Recent evidence suggests that especially infants and children are more susceptible to aflatoxins due to their lower body weight, lowered capacity to detoxify harmful substances, more restrictive diet, immature metabolism and elimination, and faster rates of growth and development. It is essential for both food safety and infant and child health that aflatoxins in cereal and cereal-based products are precisely detected, detoxified, and managed.
A worldwide systematic literature review for aflatoxin M1 in infant formula milk: Human health risk assessment by Monte Carlo simulation

Abstract:Mycotoxins, predominantly aflatoxin M1 (AFM1), require better quality control in infant formula milk (IFM). This study aims to use a systematic review study to estimate the amount and health risk assessment of AFM1 of IFM for infants less than one year old in the last two decades worldwide. For this purpose, a systematic search was conducted from June 27, 2000, to April 5, 2021. Two hundred and twelve articles and reports were obtained from various databases, and twenty of them were eventually chosen for the current study based on the study's selection criteria. Due to a lack of access to the raw data of the studies obtained, EXCEL re-simulated the data of each of these studies, keeping in mind the mean, standard deviation, and sample size of each study. The statistical analysis, health risk assessment, uncertainty analysis, and sensitivity analysis for AFM1 of IFM were then done on the simulated data by country, continent, and globally. Based on the compliance of AFM1 of IFM with the European Community (EC) standard (25 ng/kg), and also according to the results of health risk assessment, the quality of IFM provided to the market in most countries in the last two decades in terms of the AFM were at a high level and has been unauthorized. Therefore, it is necessary to pay special attention to continuous monitoring and control measures for AFM1 in consumer IFM over all countries worldwide. Due to the limited current information about AFM1 of IFM in the world, further studies are necessary.

Abstract:Aflatoxins (AFs), secondary metabolites produced by fungi, pose significant health risks, especially to children and elderly individuals. In developing countries such as Nepal, the tropical climate promotes fungal growth, leading to elevated levels of AF in animal feed and milk. In this study, we aimed to investigate the occurrence of aflatoxin M1 (AFM1) in dairy milk from the Kathmandu District and to assess husbandry practices contributing to contamination. We collected 84 milk samples, including raw milk from farms, retailers' milk, and packet milk, and analyzed them using the competitive enzyme-linked immunosorbent assay (c-ELISA) technique. We also interviewed farmers to gather information on feeding and storage practices. All the collected milk samples were contaminated with AFM1, with 97.6% of the samples exceeding the European Union (EU) maximum permissible limit of 50 ppt (0.05 μg/kg). The majority (98.5%) of the farms included paddy straw, and all farms (100%) included concentrate in their feed regimens. Only half (52%) of the farms had proper storage facilities. Straw was mostly stored in sacks outdoors or left open in a shed, while concentrates were stored in a closed room or shed. This study reveals very high levels of AFM1 contamination in the milk samples, presenting a serious public health issue, and recommends comprehensive surveillance and further investigations across the country, especially given the limited research and literature available.

Abstract:Evidence of the impact of exposure to multiple mycotoxins and environment enteric dysfunction (EED) on child growth is limited. Using data from a birth cohort study, the objectives of this study were to (a) quantify exposure to multiple mycotoxins (serum aflatoxin [AFB1 ] and ochratoxin A [OTA], urinary fumonisin [UFB1 ] and deoxynivalenol [DON]), as well EED (lactulose:mannitol [L:M] ratio); (b) examine the potential combined effects of multiple mycotoxin exposure and EED on growth. Multivariate regressions were used to identify associations between growth measurements (length, weight, anthropometric z-scores, stunting and underweight) at 24-26 months of age and exposure to mycotoxins and EED at 18-22 months (n = 699). Prevalence of AFB1 , DON, OTA and UFB1 exposure ranged from 85% to 100%; average L:M ratio was 0.29 ± 0.53. In individual mycotoxin models, AFB1 exposure was negatively associated with weight, WAZ, increased odds of stunting (odds ratio [OR]: 1.28, 95% confidence interval [CI]: 1.08, 1.52; p = 0.004) and underweight (OR: 1.18, 95% CI: 1.00, 1.38; p = 0.046). Irrespective of other mycotoxin exposure and presence of EED, AFB1 was negatively associated with length, weight, head circumference, LAZ and WAZ, and with increased odds of stunting and underweight, UFB1 was associated with increased odds of underweight, and DON was negatively associated with head circumference. EED was associated with the impaired length and weight. These findings suggest that certain mycotoxins and EED may have independent impacts on different facets of growth and that aflatoxin dominates such impacts. Thus, programs reducing exposure to mycotoxin and EED through multi-sectoral nutrition-sensitive interventions have the potential to improve child growth.

Abstract:Aflatoxin exposure has been implicated in adverse human health outcomes in areas where the toxin is prevalent in foods [1]. Consequently, many people may be chronically exposed to large amounts of the toxin worldwide. The effect of aflatoxin exposure is felt more in developing countries and this may be attributed to high levels of malnutrition coupled with chronic exposure to high levels of the toxin in their diets [2]. Consumption of high levels of aflatoxins has been associated with aflatoxicosis resulting in death, while chronic dietary exposure to aflatoxins is associated with liver and other cancers and poor growth outcomes in young children [3]. Exposure of infants to aflatoxins may be detrimental to their normal growth and other aspects of health later in life. Early linear growth retardation is associated with poor cognitive development, reduced physical productivity, a greater risk of poor pregnancy outcomes later in life, including low birth weight babies and a greater risk of obesity, coronary heart disease, diabetes and hypertension later in life [3]. Therefore, protection of fetuses, infants and young children from the effects of aflatoxins through maternal exposure would not only reduce the risk of them not realizing their cognitive potential, but also contribute to reducing the risk of non-communicable diseases whose prevalence is on the rise, not just in developed countries, but also in developing countries.

Abstract:Aflatoxins (AF) are a group of toxic secondary metabolites mostly produced by strains of Aspergillus spp. fungi. There are several types of aflatoxins (AFB1, AFG1, AFB2, AFG2, AFM1, AFM2, etc.), but the most toxic and the most common is AFB1. Aflatoxins have been shown to possess powerful carcinogenic, mutagenic and teratogenic properties. AFB1 can contaminate feed and food during the growth and/or storage under inappropriate conditions. When animals consume AFB1-contaminated feed, it undergoes various metabolic pathways and finally, it is metabolized into hydroxylated metabolite AFM1, which is possibly excreted in milk if the animal is lactating. This issue presents a huge concern regarding the representation of milk and dairy products in the human diet. Furthermore, climate changes have a significant impact on aflatoxin production. Therefore, it is necessary to develop and improve strategies for controlling and mitigating the occurrence of AFM1 in milk and dairy products. The aim of this paper is to provide an overview of the latest scientific literature regarding the occurrence of AFM1 in milk and dairy products.

Abstract:This paper aimed to present a review of the presence of aflatoxin M1 in milk and dairy products in Brazil, along with its characteristics and potential risks to human health. Overall, most studies analyzed during the defined period of this review (2013 to May 2023) reported average concentrations of AFM1 within the limits established by Brazilian legislation for milk, powdered milk, and cheese. However, considering the consumption of milk and dairy products by more vulnerable groups, such as children, two studies reported levels of AFM1 above the limits established for aflatoxins in different regions of Brazil. Thus, a more rigorous inspection by the responsible Brazilian authorities is necessary. Furthermore, since AFM1 is the only mycotoxin present in milk relevant to public health, it is important to continue conducting studies on the detection and determination of aflatoxins in milk and dairy products. This is necessary to monitor these contaminants and estimate the levels to which the population is exposed.

Abstract:As warranties on solar modules continue to expand, mechanical reliability of modules has become extremely important. Several industry standards exist to simulate conditions that modules will face in their lifetime. Among these standards are the IEC 61215/61646 and UL 1703. These tests specify the application of even pressure across the face of the module, at a given magnitude, for a given duration at room temperature (22° C). Effectively measuring and modeling these test standards will allow design of much more robust systems while maintaining cost efficiency. This paper will describe the development and validation of a standards-based mathematical model that enables rapid initial screening of module designs and components for improved module durability. The paper will be presented in four sections: 1.) the test setup and procedure; 2.) model development; 3.) model validation; and 4.) rapid parametric studies.
