Abstract:Impact statementRecent methodological advances in network neuroscience allow integrating connectivity information from different scales or aspects through multilayer network models. Utilizing such models, we found common and specific neurodevelopmental abnormalities in multiband functional brain networks between major depressive disorder (MDD) and schizophrenia (SCH), and the abnormalities cannot be uncovered by individually analyzing brain networks within each frequency interval. These findings have important implications for understanding shared and unique neural mechanisms underlying MDD and SCH and highlight the necessity of integrating connectivity information from different frequency intervals to search for new biomarkers of the diseases. Introduction: Both major depressive disorder (MDD) and schizophrenia (SCH) are characterized by neurodevelopmental abnormalities; however, transdiagnostic and diagnosis-specific patterns of such abnormalities have rarely been examined, particularly in large-scale functional brain networks via advanced multilayer network models.Methods: Here, we collected resting-state functional magnetic resonance imaging data from 45 MDD patients, 64 SCH patients, and 48 healthy controls (HCs; 13-45 years old), and we constructed functional networks in different frequency intervals. The frequency-dependent networks were then fused by multiplex network models, followed by graph-based topological analyses.Results: We found that functional networks of the patients showed common neurodevelopmental abnormalities in the right ventromedial parietooccipital sulcus (opposite correlations with age to HCs), whereas functional networks of the MDD patients exhibited specific alterations in the left superior parietal lobule and right precentral gyrus with respect to cross-frequency interactions. These findings were quite different from those from brain networks within each frequency interval, which revealed SCH-specific neurodevelopmental abnormalities in the right superior temporal gyrus (opposite correlations with age to the other two groups) in 0.027-0.073 Hz, and SCH-specific alterations in the left superior temporal gyrus and bilateral insula in 0.073-0.198 Hz. Finally, multivariate analysis of age prediction revealed that the subcortical network lost prediction ability in both patient groups, whereas the visual network exhibited additional prediction ability in the MDD patients.Discussion and Conclusion: Altogether, these findings demonstrate transdiagnostic and diagnosis-specific neurodevelopmental abnormalities and alterations in large-scale functional brain networks between MDD and SCH, which have important implications for understanding shared and unique neural mechanisms underlying the diseases.

Abstract:Altered functional brain networks have been a typical manifestation that distinguishes major depressive disorder (MDD) patients from healthy control (HC) subjects in functional magnetic resonance imaging (fMRI) studies. Recently, rich club and diverse club metrics have been proposed for network or network neuroscience analyses. The rich club defines a set of nodes that tend to be the hubs of specific communities, and the diverse club defines the nodes that span more communities and have edges diversely distributed across different communities. Considering the heterogeneity of rich clubs and diverse clubs, combining them and on the basis to derive a novel indicator may reveal new evidence of brain functional integration and separation, which might provide new insights into MDD. This study for the first time discussed the differences between MDD and HC using both rich club and diverse club metrics and found the complementarity of them in analyzing brain networks. Besides, a novel index, termed "active index", has been proposed in this study. The active index defines a group of nodes that tend to be diversely distributed across communities while avoiding being a hub of a community. Experimental results demonstrate the superiority of active index in analyzing MDD brain mechanisms.
Connectome Analysis of Functional and Structural Hemispheric Brain Networks in Major Depressive Disorder

Abstract:Neuroimaging studies have shown topological disruptions of both functional and structural whole-brain networks in major depressive disorder (MDD). This study examined common and specific alterations between these two types of networks and whether the alterations were differentially involved in the two hemispheres. Multimodal MRI data were collected from 35 MDD patients and 35 healthy controls, whose functional and structural hemispheric networks were constructed, characterized, and compared. We found that functional brain networks were profoundly altered at multiple levels, while structural brain networks were largely intact in patients with MDD. Specifically, the functional alterations included decreases in intra-hemispheric (left and right) and inter-hemispheric (heterotopic) functional connectivity; decreases in local, global and normalized global efficiency for both hemispheric networks; increases in normalized local efficiency for the left hemispheric networks; and decreases in intra-hemispheric integration and inter-hemispheric communication in the dorsolateral superior frontal gyrus, anterior cingulate gyrus and hippocampus. Regarding hemispheric asymmetry, largely similar patterns were observed between the functional and structural networks: the right hemisphere was over-connected and more efficient than the left hemisphere globally; the occipital and partial regions exhibited leftward asymmetry, and the frontal and temporal sites showed rightward lateralization with regard to regional connectivity profiles locally. Finally, the functional–structural coupling of intra-hemispheric connections was significantly decreased and correlated with the disease severity in the patients. Overall, this study demonstrates modality- and hemisphere-dependent and invariant network alterations in MDD, which are helpful for understanding elaborate and characteristic patterns of integrative dysfunction in this disease.

Abstract:At present, most brain functional studies are based on traditional frequency bands to explore the abnormal functional connections and topological organization of patients with depression. However, they ignore the characteristic relationship of electroencephalogram (EEG) signals in the time domain. Therefore, this paper proposes a network decomposition model based on Improved Empirical Mode Decomposition (EMD), it is suitable for time-frequency analysis of brain functional network. On the one hand, it solves the problem of mode mixing on original EMD method, especially on high-density EEG data. On the other hand, by building brain function networks on different intrinsic mode function (IMF), we can perform time-frequency analysis of brain function connections. It provides a new insight for brain function connectivity analysis of major depressive disorder (MDD). Experimental results found that the IMFs waveform decomposed by Improved EMD was more stable and the difference between IMFs was obvious, it indicated that the mode mixing can be effectively solved. Besides, the analysis of the brain network, we found that the changes in MDD functional connectivity on different IMFs, it may be related to the pathological changes for MDD. More statistical results on three network metrics proved that there were significant differences between MDD and normal controls (NC) group. In addition, the aberrant brain network structure of MDDs was also confirmed in the hubs characteristic. These findings may provide potential biomarkers for the clinical diagnosis of MDD patients.

Abstract:Background: Existing studies have shown that the brain network of major depression disorder (MDD) has abnormal topologies. However, constructing reliable MDD brain networks is still an open problem. New method: This paper proposed a reliable MDD brain network construction method. First, seven connectivity methods are used to calculate the correlation between channels and obtain the functional connectivity matrix. Then, the matrix is binarized using four binarization methods to obtain the EEG brain network. Besides, we proposed an improved binarization method based on the criterion of maximizing differences between groups: the adaptive threshold (AT) method. The AT can automatically set the optimal binarization threshold and overcome the artificial influence of traditional methods. After that, several network metrics are extracted from the brain network to analyze inter-group differences. Finally, we used statistical analysis and Fscore values to compare the performance of different methods and establish the most reliable method for brain network construction. Results: In theta, alpha, and total frequency bands, the clustering coefficient, global efficiency, local efficiency, and degree of the MDD brain network decrease, and the path length of the MDD brain network increases. Comparison with existing methods: The results show that AT outperforms the existing binarization methods. Compared with other methods, the brain network construction method based on phase-locked value (PLV) and AT has better reliability. Conclusions: MDD has brain dysfunction, particularly in the frontal and temporal lobes.

Abstract:This study investigates the abnormal brain structural connectivity in patients with major depressive disorder (MDD) through a modularization approach.Diffusion tensor imaging (DTI) data were collected from 20 MDD patients and 20 age-matched controls.The structural modularity characteristics and their relationships with clinical variables were examined.Results indicated that MDD patients exhibited reorganized modules with widely damaged global inter-module connectivity in limbic system and altered intra-module connectivity in left hemisphere.Furthermore, the module degree in right rolandic operculum, left posterior cingulate gyrus, right cuneus and right fusiform gyrus were negatively correlated with the depression severity.Additionally, a positive relationship was also found between the duration of disease and participant coefficient in left median cingulate.The current findings highlight altered modular organization in MDD patients, which might contribute to the pathogenesis of the disease.

Abstract:Background: Major depressive disorder (MDD) is known to be characterized by altered brain functional connectivity (FC) patterns. However, whether and how the features of dynamic FC would change in patients with MDD are unclear. In this study, we aimed to characterize dynamic FC in MDD using a large multi-site sample and a novel dynamic network-based approach. Methods: Resting-state functional magnetic resonance imaging (fMRI) data were acquired from a total of 460 MDD patients and 473 healthy controls, as a part of the REST-meta-MDD consortium. Resting-state dynamic functional brain networks were constructed for each subject by a sliding-window approach. Multiple spatio-temporal features of dynamic brain networks, including temporal variability, temporal clustering and temporal efficiency, were then compared between patients and healthy subjects at both global and local levels. Results: The group of MDD patients showed significantly higher temporal variability, lower temporal correlation coefficient (indicating decreased temporal clustering) and shorter characteristic temporal path length (indicating increased temporal efficiency) compared with healthy controls (corrected p < 3.14 x 10(-3)). Corresponding local changes in MDD were mainly found in the default-mode, sensorimotor and subcortical areas. Measures of temporal variability and characteristic temporal path length were significantly correlated with depression severity in patients (corrected p < 0.05). Moreover, the observed between-group differences were robustly present in both first-episode, drug-naive (FEDN) and non-FEDN patients. Conclusions: Our findings suggest that excessive temporal variations of brain FC, reflecting abnormal communications between large-scale bran networks over time, may underlie the neuropathology of MDD.
Dynamic Community Structure in Major Depressive Disorder: A Resting-State MEG Study.

Abstract:BACKGROUND:Major Depressive Disorder (MDD), characterized by depressed mood or anhedonia, is associated with altered functional connectivity (FC) within and between large scale networks such as the Default Mode Network (DMN), the Central Executive Network (CEN) and the Salience Network (SN). Since aberrant FC exhibits temporal variability and could give rise to distorted reconfiguration of functional brain networks, an in-depth analysis of the community structure could provide further insight into the synchrony of networks. We hypothesized that alterations in dynamic network community structure in MDD could be temporally accompanied by disrupted conscious states of these three networks.METHODS:26 MDD patients and 25 healthy controls were scanned using a whole-head resting-state Magnetoencephalography (MEG) machine. A novel multilayer modularity framework explored the functional modulation of these networks. Recruitment (R) and integration (I) provided the strength of interaction within networks or across networks, respectively.RESULTS:The brain regions in the DMN, CEN and SN were transiently integrated and segmented in both patients and controls. R of CEN and I of SN were significantly greater in MDD compared to controls.CONCLUSION:Intrinsic resting-state networks dynamically interact and reorganize into distinct functional modules in both patients and controls. However, the CEN "hyper-intertwines" with itself and SN "hyper-integrates" among the network of interest in depressed patients compared to controls. Network-level alterations in R and I revealed a more generalized system-level effect rather than a focal-wise effect from a neural dynamic perspective. This could potentially highlight an abnormal network-based mechanism in depression.

Abstract:Objective To examine the changes of functional network modularity in treatment-na(i)ve,first episode major depressive disorder (MDD) patients and the relationship between modularity related indicators and clinical severity.Methods 50 treatment-naive,first episode patients with MDD and 37 age,gender,and education level matched healthy control (HC) were included.They all received fMRI scanning during quiet,eye closed and awake state.We used the 90 automated anatomical labeling (ALL) template to construct functional connectivity matrix.Several modularity related indicators,including global modularization,intra-module connectivity,inter-module connectivity,were computed.The differences in these indicators were compared between MDD group and HC group.The Pearson correlation analysis were performed to show correlation between the regions showing modularity changes and severity of disease.Result There were 4 modules in MDD patients and 3 modules in HC group.MDD patients showed significant decline in modularity degree in the left middle orbitofrontal gyrus,the inferior orbitofrontal gyrns,the superior occipital gyrus,and the rolandic operculum.No significant correlation were found between modularity degree values of these four regions and the severity of disease (r=0.21,P＞0.05).No significant differences were found in global modularization,intra connectivity and inter connectivity between two groups (r=0.11,P＞0.05).Conclusion MDD patients may have changed connectivity in orbitofrontal cortex,superior occipital gyrus and rolandic operculum,which are relevant nodes of neural network basis of MDD.
Identifying Subgroups of Major Depressive Disorder Using Brain Structural Covariance Networks and Mapping of Associated Clinical and Cognitive Variables

Abstract:BACKGROUND: Identifying data-driven subtypes of major depressive disorder (MDD) holds promise for parsing the heterogeneity of MDD in a neurobiologically informed way. However, limited studies have used brain structural covariance networks (SCNs) for subtyping MDD. METHODS: This study included 145 unmedicated patients with MDD and 206 demographically matched healthy control subjects, who underwent a structural magnetic resonance imaging scan and a comprehensive neurocognitive battery. Patterns of structural covariance were identified using source-based morphometry across both patients with MDD and healthy control subjects. K-means clustering algorithms were applied on dysregulated structural networks in MDD to identify potential MDD subtypes. Finally, clinical and neurocognitive measures were compared between identified subgroups to elucidate the profile of these MDD subtypes. RESULTS: Source-based morphometry across all individuals identified 28 whole-brain SCNs that encompassed the prefrontal, anterior cingulate, and orbitofrontal cortices; basal ganglia; and cerebellar, visual, and motor regions. Compared with healthy control subjects, individuals with MDD showed lower structural network integrity in three networks including default mode, ventromedial prefrontal cortical, and salience networks. Clustering analysis revealed two MDD subtypes based on the patterns of structural network abnormalities in these three networks. Further profiling revealed that patients in subtype 1 had younger age of onset and more symptom severity as well as greater deficits in cognitive performance than patients in subtype 2. CONCLUSIONS: Overall, we identified two MDD subtypes based on SCNs that differed in their clinical and cognitive profile. Our results represent a proof-of-concept framework for leveraging these large-scale SCNs to parse heterogeneity in MDD.

Abstract:Background: Major Depressive Disorder (MDD) is common and disabling, but its neural pathophysiology remains unclear. Functional brain network studies in MDD have largely had limited statistical power and data analysis approaches have varied widely. The REST-meta-MDD Project of resting-state fMRI (R-fMRI) addresses these issues. Methods: The 25 research groups in China composing the REST-meta-MDD Project contributed R-fMRI data of 1,300 patients with MDD and 1,128 normal controls (NCs). The data were preprocessed locally with a standardized protocol prior to aggregated group analyses. We focused on functional connectivity (FC) within the default mode network (DMN), frequently reported to show increased FC in MDD. Outcomes: We found decreased instead of increased DMN FC in MDD compared to NCs. We found FC reduction only in recurrent MDD, not in first-episode drug-naive MDD. Decreased DMN FC was associated with medication usage but not with MDD duration or severity. Exploratory analyses also revealed alterations of local intrinsic activity in MDD. Interpretation: We confirmed the key role of DMN in MDD but found reduced rather than increased FC within the DMN. Future studies should test whether decreased DMN FC mediates treatment response. All the R-fMRI indices have been made publicly available via the R-fMRI Maps Project. The REST-meta-MDD model can be generalized to longitudinal MDD studies and to other psychiatric disorders. A Study of Major Depressive Disorder Based on Resting-State Multilayer EEG Function Network

Abstract:Depression is a complex mental disease with its pathological mechanism unclear. To depict the complete picture of the abnormal information interaction in a depressed brain, this study is the first to apply fully connected multilayer brain functional (FCMBF) network framework and proposed composite FCMBF (CFCMBF) network framework, combined with graph theory to analyze the within-frequency coupling (WFC) and cross-frequency coupling (CFC) of sensor-layer and source-layer electroencephalography (EEG) signals in relevant subjects. Results showed that in the sensor-layer FCMBF network, depressive patients showed significantly reduced functional connectivity, as well as abnormal global and local information processing abilities of the network, and these network properties were significantly correlated with depressive symptoms. In addition, from the perspective of depression recognition, we found that the sensor-layer CFCMBF network could achieve better classification accuracy, especially when using the overlapping degree of node under the right center region, its accuracy could reach $86.88\% pm 9.25 \%$ . More importantly, the construction of the CFCMBF network has higher time efficiency and less information loss, since it not only measures the WFC and CFC between brain region representative signals (BRRSs) extracted from different brain regions, but also measures these two couplings between all nodes within each brain region. Although the FCMBF network contains more complete information by calculating WFC and CFC between all nodes distributed in each region, it will result in an enormous computational cost. In summary, this study proved the utility of multilayer brain network in revealing the abnormal brain interaction patterns of depression, and our proposed method might provide methodological support for efficient depression recognition research based on multilayer brain networks.

Abstract:Background: Major depressive disorder (MDD) is the leading cause of disability among all mental illnesses with increasing prevalence. The diagnosis of MDD is susceptible to interference by several factors, which has led to a trend of exploring objective biomarkers. Electroencephalography (EEG) is a non-invasive procedure that is being gradually applied to detect and diagnose MDD through some features such as functional connectivity (FC). Methods: In this research, we analyzed the resting-state EEG of patients with MDD and healthy controls (HCs) in both eyes-open (EO) and eyes-closed (EC) conditions. The phase locking value (PLV) method was utilized to explore the connection and synchronization of neuronal activities spatiotemporally between different brain regions. We compared the PLV between participants with MDD and HCs in five frequency bands (theta, 4-8 Hz; alpha, 8-12 Hz; beta1, 12-16 Hz; beta2, 16-24 Hz; and beta3, 24-40 Hz) and further analyzed the correlation between the PLV of connections with significant differences and the severity of depression (via the scores of 17-item Hamilton Depression Rating Scale, HDRS-17). Results: During the EO period, lower PLVs were found in the right temporal-left midline occipital cortex (RT-LMOC; theta, alpha, beta1, and beta2) and posterior parietal-right temporal cortex (PP-RT; beta1 and beta2) in the MDD group compared with the HC group, while PLVs were higher in the MDD group in LT-LMOC (beta2). During the EC period, for the MDD group, lower theta and beta (beta1, beta2, and beta3) PLVs were found in PP-RT, as well as lower theta, alpha, and beta (beta1, beta2, and beta3) PLVs in RT-LMOC. Additionally, in the left midline frontal cortex-right temporal cortex (LMFC-RT) and posterior parietal cortex-right temporal cortex (PP-RMOC), higher PLVs were observed in beta2. There were no significant correlations between PLVs and HDRS-17 scores when connections with significantly different PLVs (all p > 0.05) were checked. Conclusion: Our study confirmed the presence of differences in FC between patients with MDD and healthy individuals. Lower PLVs in the connection of the right temporal-left occipital cortex were mostly observed, whereas an increase in PLVs was observed in patients with MDD in the connections of the left temporal with occipital lobe (EO), the circuits of the frontal-temporal lobe, and the parietal-occipital lobe. The trends in FC involved in this study were not correlated with the level of depression. Limitations: The study was limited due to the lack of further analysis of confounding factors and follow-up data. Future studies with large-sampled and long-term designs are needed to further explore the distinguishable features of EEG FC in individuals with MDD.

Abstract:Major depressive disorder (MDD) is a common psychiatric condition associated with aberrant large-scale distributed brain networks. However, it is unclear how the network dysfunction in MDD patients is characterized by imbalance or derangement of network modular segregation. Fifty-one MDD patients and forty-three matched healthy controls (HC) were recruited in the present study. We analyzed intrinsic brain activity derived from resting-state functional magnetic resonance imaging (R-fMRI) and then examined brain network segregation by computing the participation coefficient (PC). Further intra- and inter-modular connections analysis were preformed to explain atypical PC. Besides, we explored the potential relationship between the above graph theory measures and symptom severity in MDD. Lower modular segregation of the frontal–parietal network (FPN) was found in MDD compared with the HC group. The MDD group exhibited increased inter-module connections between the FPN and cingulo-opercular network (CON), between the FPN and cerebellum (Cere), between the CON and Cere. At the nodal level, the PC of the anterior prefrontal cortex, anterior cingulate cortex, inferior parietal lobule (IPL), and intraparietal sulcus showed larger in MDD. Additionally, the inter-module connections between the FPN and CON and the PC values of the IPL were negatively correlated with depression symptom in the MDD group. These findings might give evidence about abnormal FPN in MDD from the perspective of modular segregation in brain networks.

Abstract:Background: Major Depressive Disorder (MDD) is a pervasive mental health issue with significant diagnostic challenges. Electroencephalography (EEG) offers a non-invasive window into the neural dynamics associated with MDD, yet the diagnostic efficacy is contingent upon the appropriate selection of EEG features and brain regions. Methods: In this study, resting-state EEG signals from both eyes-closed and eyes-open conditions were analyzed. We examined band power across various brain regions, assessed the asymmetry of band power between the hemispheres, and integrated these features with clinical characteristics of MDD into a diagnostic regression model. Results: Regression analysis found significant predictors of MDD to be beta2 (16–24 Hz) power in the Prefrontal Cortex (PFC) with eyes open ( B = 20.092, p = 0.011), beta3 (24–40 Hz) power in the Medial Occipital Cortex (MOC) ( B = −12.050, p < 0.001), and beta2 power in the Right Medial Frontal Cortex (RMFC) with eyes closed ( B = 24.227, p < 0.001). Asymmetries in beta1 (12–16 Hz) power with eyes open ( B = 28.047, p = 0.018), and in alpha (8–12 Hz, B = 9.004, p = 0.013) and theta (4–8 Hz, B = −13.582, p = 0.008) with eyes closed were also significant predictors. Conclusion: The study confirms the potential of multi-region EEG analysis in improving the diagnostic precision for MDD. By including both neurophysiological and clinical data, we present a more robust approach to understanding and identifying this complex disorder. Limitations: The research is limited by the sample size and the inherent variability in EEG signal interpretation. Future studies with larger cohorts and advanced analytical techniques are warranted to validate and refine these findings.

Abstract:Major depressive disorder (MDD) and bipolar disorder (BD) share clinical features, which complicates their differentiation in clinical settings. This study proposes an innovative approach that integrates structural connectome analysis with machine learning models to discern individuals with MDD from individuals with BD. High-resolution MRI images were obtained from individuals diagnosed with MDD or BD and from HCs. Structural connectomes were constructed to represent the complex interplay of brain regions using advanced graph theory techniques. Machine learning models were employed to discern unique connectivity patterns associated with MDD and BD. At the global level, both BD and MDD patients exhibited increased small-worldness compared to the HC group. At the nodal level, patients with BD and MDD showed common differences in nodal parameters primarily in the right amygdala and the right parahippocampal gyrus when compared with HCs. Distinctive differences were found mainly in prefrontal regions for BD, whereas MDD was characterized by abnormalities in the left thalamus and default mode network. Additionally, the BD group demonstrated altered nodal parameters predominantly in the fronto-limbic network when compared with the MDD group. Moreover, the application of machine learning models utilizing structural brain parameters demonstrated an impressive 90.3% accuracy in distinguishing individuals with BD from individuals with MDD. These findings demonstrate that combined structural connectome and machine learning enhance diagnostic accuracy and may contribute valuable insights to the understanding of the distinctive neurobiological signatures of these psychiatric disorders.

Abstract:Major depressive disorder (MDD) is recognized as a severe mental illness with imbalanced interactions among brain networks. However, the detailed mechanisms of large-scale network dysfunction and their clinical implications are not fully understood. To explore the neurological basis of altered connectivity within the brain, the current case-control study aimed to examine large-scale connectivity coherence in MDD using resting-state functional magnetic resonance imaging data from 1,148 individuals with MDD and 1,079 healthy volunteers across nine research centers. Global brain connectivity (GBC) was estimated and compared between groups. Compared with healthy volunteers, individuals with MDD had decreased GBC in sensorimotor/visual networks and increased GBC mainly in default mode networks (DMNs) (voxel-level P < 0.001, cluster-level P < 0.05). These main findings were consistent across different clinical states of MDD, indicating their independence from clinical factors (P < 0.05, FDR-corrected). Further seed connectivity revealed that individuals with MDD had heightened connectivity between DMNs and primary sensory cortices, but reduced connectivity within primary sensory cortices (voxel-level P < 0.001, cluster-level P < 0.05). The findings suggest a network imbalance toward the DMNs at the expense of the sensorimotor/visual networks in individuals with MDD experiencing a depressive episode. These alterations, involving both higher-order cognitive systems and low-level sensory systems, could provide insights into understanding the multifaceted clinical and cognitive deficits observed in MDD. Using a large multisite dataset of people with major depressive disorder and healthy controls, the authors show global brain connectivity alterations in the sensorimotor, visual, and default mode networks.

Abstract:On the increase of major depressive disorders (MDD), many researchers paid attention to their recognition and treatment. Existing MDD recognition algorithms always use a single time-frequency domain method method, but the single time-frequency domain method is too simple and is not conducive to simulating the complex link relationship between brain functions. To solve this problem, this paper proposes a recognition method based on multi-layer brain functional connectivity networks (MBFCN) for major depressive disorder and conducts cognitive analysis. Cognitive analysis based on the proposed MBFCN finds that the Alpha-Beta1 frequency band is the key sub-band for recognizing MDD. The connections between the right prefrontal lobe and the temporal lobe of the extremely depressed disorders (EDD) are deficient in the brain functional connectivity networks (BFCN) based on phase lag index (PLI). Furthermore, potential biomarkers by the significance analysis of depression features and PHQ-9 can be found.

Abstract:Background: Accumulated evidence has illuminated the topological infrastructure of major depressive disorder (MDD). However, the changes of topological properties of anatomical brain networks in remitted major depressive disorder patients (rMDD) remain an open question. The present study provides an exploratory examination of pattern changes among current major depressive disorder patients (cMDD), EMDD patients and healthy controls (HC) by means of a pattern recognition analysis.Methods: Twenty-eight cMDD patients (age range: 22-54, mean age: 39.57), 15 EMDD patients (age range: 23-53, mean age: 3840) and 30 HC (23-54, mean age: 35.57) were enrolled. For each subject, we computed five kinds of weighted white matter (WM) networks via employing five physiological parameters (i.e. fractional anisotropy, mean diffusivity, lambda(1), lambda(2) and lambda(3)) and then calculated three network measures of these weighted networks. We treated these measures as features and fed into a feature selection mechanism to choose the most discriminative features for linear support vector machine (SVM) classifiers.Results: Linear SVM could excellently distinguish the three groups with the 100% classification accuracy of recognizing cMDD/rMDD from HC, and 97.67% classification accuracy of recognizing cMDD from rMDD. The further pattern analysis found two types of discriminative patterns among cMDD, rMDD and HC. (i) Compared with HC, both cMDD and EMDD exhibited the similar deficit patterns of node strength primarily involving the salience network (SN), default mode network (DMN) and frontoparietal network (FPN). (ii) Compared with cMDD and rMDD showed the altered pattern of intra-communicability within DMN and inter-communicability between DMN and the other sub-networks including the visual recognition network (VRN) and SN.Limitations: The present study had a limited sample size and a lack of larger independent data set to validate the methods and confirm the findings.Conclusions: These findings implied that the impairment of MOD was closely associated with the alterations of connections within SN, DMN and FPN, whereas the remission of MDD was benefitted from the network compensatory of intra-communication within DMN and inter-communication between DMN and the other sub-networks (i.e., VRN and SN). Dynamic cortical connectivity alterations associated with Major Depressive Disorder: an EEG study

Abstract:There are various depressive subtypes identified in patients with major depressive disorder (MDD). Depression with psychotic symptoms is usually known to be a severe type of depression that includes symptoms such as delusions and/or hallucinations, and remains a common condition that is often underrecognized and inadequately treated in clinical practice. Electroencephalography (EEG) biomarkers have been implicated to classify healthy and psychopathological neural signals using machine learning algorithms. In this study, we sought to identify cortical functional connectivity metrics that differentiate network manifestation of different depressive subtypes and healthy controls. We first performed replication analyses to obtain the principal functional connectivity microstates across each independent group (healthy controls, psychotic depressions and nonpsychotic depressions). Next, we examined temporal functional connectivity dynamics in each group. The results show that fundamental dynamic functional connectivity microstates are highly reproducible, both within and across participants. Based on the temporal and sequential parameters (mean duration, fractional windows and transition number) derived from dynamic functional connectivity analysis, we found inter-group differences across healthy and MDD subgroups statistically significant. These results show that the principal FC microstates dynamics are essential neural biomarkers distinctly associated with depression clinical phenotypes.Clinical relevance-Our findings suggest that a network-level feature, that may reflect the neurobiological difference between different depression subtypes, and healthy controls, and in turn may contribute towards a scalable EEG-based assisted diagnostic tool.
