Effects of Cortical Stimulation on Auditory‐responsive Thalamic Neurones in Anaesthetized Guinea Pigs

Abstract:In the present study, we investigated neuronal responses to acoustic stimuli and cortical stimulation in the medial geniculate body (MGB) through in vivo intracellular recordings in anaesthetized guinea pigs. Of the 54 neurones examined with acoustic stimuli, 36 showed excitatory postsynaptic potential (EPSP) responses and 19 showed inhibitory postsynaptic potential (IPSP) responses to acoustic stimuli. Of the 36 EPSP neurones examined with corticofugal modulation, 29 received corticofugal depolarization, 3 corticofugal inhibition, and 4 showed no effect. Of the 19 IPSP neurones, 17 received corticofugal inhibition and 2 were not affected. The mean amplitude of the EPSPs evoked by acoustic stimuli was similar to that evoked by the electrical cortical stimulation (9.19 ± 5.55 mV versus 9.22 ± 5.16 mV). There was a significant correlation between the parameters of the EPSPs evoked by an acoustic stimulus and those evoked by cortical stimulation. The mean amplitude of the IPSP evoked by electrical cortical stimulation was significantly greater than that evoked by acoustic stimuli (11.6 ± 3.8 mV versus 9.1 ± 3.7 ms, P < 0.05). Seven auditory EPSP and 7 IPSP neurones were examined with corticofugal modulation and labelled with Neurobiotin. Of the 7 EPSP neurones, 5 showed excitatory responses to cortical stimulation and 2 demonstrated no effects. Four of the 5 neurones that received corticofugal depolarization were located in the lemniscal MGB and 1 in the non‐lemniscal MGB; of the remaining 2, 1 was located in the lemniscal and the other in the non‐lemniscal MGB. Of the 7 IPSP neurones, 1 received an excitatory corticofugal input followed by an inhibitory input and 4 received only an inhibitory corticofugal input, while the remainder demonstrated no corticofugal effects. All 7 neurones were located in the non‐lemniscal MGB. The result that both ascending and descending inputs caused similarly shaped EPSPs reflects a neuronal endogenous characteristic irrespective of the physical locations of the synapses. The IPSP responses to both acoustic stimuli and electrical cortical stimulation are likely to be caused by feedback from the thalamic reticular nucleus.
Multiple Visuotopically Organized Subdivisions of the Lateral Pulvinar/Central Lateral Inferior Pulvinar Project into Thin and Thick Stripe Compartments of V2 in Macaques.

Abstract:The lateral and central lateral inferior pulvinar (PL/PIcl) of primates has been implicated in playing an important role in visual processing, but its physiological and anatomical characteristics remain to be elucidated. It has been suggested that there are two complete visuotopic maps in the PL/PIcl, each of which sends afferents into V2 and V4 in primates. Given that functionally distinct thin and thick stripes of V2 both receive inputs from the PL/PIcl, this raises the possibility of a presence of parallel segregated pathways within the PL/PIcl. To address this question, we selectively injected three types of retrograde tracers (CTB-488, CTB-555, and BDA) into thin or thick stripes in V2 and examined labeling in the PL/PIcl in macaques. As a result, we found that every cluster of retrograde labeling in the PL/PIcl included all three types of signals next to each other, suggesting that thin stripe- and thick stripe-projecting compartments are not segregated into domains. Unexpectedly, we found at least five topographically organized retrograde labeling clusters in the PL/PIcl, indicating the presence of more than two V2-projecting maps. Our results suggest that the PL/PIcl exhibits greater compartmentalization than previously thought. They may be functionally similar but participate in multiple cortico-pulvinar-cortical loops.
Modulatory Effect of Cortical Activation on the Lemniscal Auditory Thalamus of the Guinea Pig.

Abstract:In the present study, we investigated the point-to-point modulatory effects from the auditory cortex to the thalamus in the guinea pig. Corticofugal modulation on thalamic neurons was studied by electrical activation of the auditory cortex. The modulation effect was sampled along the frontal or sagittal planes of the auditory thalamus, focusing on the ventral division (MGv) of the medial geniculate body (MGB). Electrical activation was targeted at the anterior and dorsocaudal auditory fields, to which the MGv projects and from which it assumptively receives reciprocal projections. Of the 101 MGv neurons examined by activation of the auditory cortex through passing pulse trains of 100-200 microA current into one after another of the three implanted electrodes (101 neurons x 3 stimulation sites = 303 cases), 208 cases showed a facilitatory effect, 85 showed no effect, and only 10 cases (7 neurons) showed an inhibitory effect. Among the cases of facilitation, 63 cases showed a facilitatory effect >100%, and 145 cases showed a facilitatory effect from 20-100%. The corticofugal modulatory effect on the MGv of the guinea pig showed a widespread, strong facilitatory effect and very little inhibitory effect. The MGv neurons showed the greatest facilitations to stimulation by the cortical sites, with the closest correspondence in BF. Six of seven neurons showed an elevation of the rate-frequency functions when the auditory cortex was activated. The comparative results of the corticofugal modulatory effects on the MGv of the guinea pig and the cat, together with anatomical findings, hint that the strong facilitatory effect is generated through the strong corticothalamic direct connection and that the weak inhibitory effect might be mainly generated via the interneurons of the MGv. The temporal firing pattern of neuronal response to auditory stimulus was also modulated by cortical stimulation. The mean first-spike latency increased significantly from 15.7 +/- 5.3 ms with only noise-burst stimulus to 18.3 +/- 4.9 ms (n = 5, P < 0.01, paired t-test), while the auditory cortex was activated with a train of 10 pulses. Taking these results together with those of previous experiments conducted on the cat, we speculate that the relatively weaker inhibitory effect compared with that in the cat could be due to the smaller number of interneurons in the guinea pig MGB. The corticofugal modulation of the firing pattern of the thalamic neurons might enable single neurons to encode more auditory information using not only the firing rate but also the firing pattern.
Visually-evoked activity and variable modulation of auditory responses in the macaque inferior colliculus

Abstract:How multisensory cues affect processing in early sensory brain areas is not well understood. The inferior colliculus (IC) is an early auditory structure that is visually responsive (Porter et al., 2007; Bulkin & Groh, 2012b, 2012a), but little is known about how visual signals affect the IC's auditory representation. We explored how visual cues affect both spiking and local field potential (LFP) activity in the IC of two monkeys performing a task involving saccades to auditory, visual, or combined audiovisual stimuli. We confirm that LFPs are sensitive to the onset of fixation lights as well as the onset of visual targets presented during steady fixation. The LFP waveforms evoked by combined audiovisual stimuli differed from those evoked by sounds alone. In single-unit spiking activity, responses were weak when visual stimuli were presented alone, but visual stimuli could modulate the activity evoked by sounds in a stronger way. Such modulations could involve either increases or decreases in activity, and whether increases or decreases were observed was variable and not obviously correlated with the responses evoked by visual or auditory stimuli alone. These findings indicate that visual stimuli shape the IC's auditory representation in flexible ways that differ from those observed previously in multisensory areas.

Modulation of auditory responses by visual inputs in the mouse auditory cortex

Abstract:Abstract So far, our understanding on the role of the auditory cortex (ACX) in processing visual information has been limited to infragranular layers of the ACX, which have been shown to respond to visual stimulation. Here, we investigate the neurons in supragranular layers of the mouse ACX using 2-photon calcium imaging. Contrary to previous reports, here we show that more than 20% of responding neurons in layer2/3 of the ACX respond to full-field visual stimulation. These responses occur by both excitation and hyperpolarization. The primary ACX (A1) has a greater proportion of visual responses by hyperpolarization compared to excitation likely driven by inhibitory neurons of the infragranular layers of the ACX rather than local layer 2/3 inhibitory neurons. Further, we found that more than 60% of neurons in the layer 2/3 of A1 are multisensory in nature. We also show the presence of multisensory neurons in close proximity to exclusive auditory neurons and that there is a reduction in the noise correlations of the recorded neurons during multisensory presentation. This is evidence in favour of deep and intricate visual influence over auditory processing. The results have strong implications for decoding visual influences over the early auditory cortical regions. Significance statement To understand, what features of our visual world are processed in the auditory cortex (ACX), understanding response properties of auditory cortical neurons to visual stimuli is important. Here, we show the presence of visual and multisensory responses in the supragranular layers of the ACX. Hyperpolarization to visual stimulation is more commonly observed in the primary ACX. Multisensory stimulation results in suppression of responses compared to unisensory stimulation and an overall decrease in noise correlation in the primary ACX. The close-knit architecture of these neurons with auditory specific neurons suggests the influence of non-auditory stimuli on the auditory processing.

Abstract:Conventional electrophysiological technique Was used to determine the distribution of auditory-visual multisensory neurons and to study the auditory-visual information integration in rat cortex. 130 auditory-visual bimodal neurons including 65 AN neurons, 28 v-A neurons and 37 a-A neurons were isolated. These bimodal neurons located in the boundary area between auditory and visual cortex were mainly in the dorsal bank of auditory cortex. There are divided strip trends in the distribution of different types of bimodal neurons that most of v-A neurons were recorded in the cortex close to auditory cortex, a-V neurons were mostly appeared in the part adjacent to visual cortex and a majority of AN neurons were between of them. One point must be pointed out that bimodal neurons were presented not uniformly but in the way of patchy distribution. According to the different integration effects, all bimodal neurons were divided into three types: enhancement, modulation and depression. The temporal disparity among combinations of two stimuli was showed to be a critical factor influencing the integration of auditory and visual information. At the same time the best interstimulus interval time under which the strongest interaction can be evoked has been thoroughly studied and found that it has somewhat regular arrangement that 98 of the 130 (75%) auditory-visual bimodal neurons in this experiment have the best integration among the 30 similar to 50 ms interval time. The results suggest that the auditory-visual bimodal neurons exist in the rat cortex having the similar multisensory and auditory-visual integration characteristics of the multimodal neurons in the other animals' analogy cortex.
Correlation of Neural Response Properties with Auditory Thalamus Subdivisions in the Awake Marmoset.

Abstract:As the information bottleneck of nearly all auditory input that reaches the cortex, the auditory thalamus serves as the basis for establishing auditory cortical processing streams. The functional organization of the primary and nonprimary subdivisions of the auditory thalamus is not well characterized, particularly in awake primates. We have recorded from neurons in the auditory thalamus of awake marmoset monkeys and tested their responses to tones, band-pass noise, and temporally modulated stimuli. We analyzed the spectral and temporal response properties of recorded neurons and correlated those properties with their locations in the auditory thalamus, thereby forming the basis for parallel output channels. Three medial geniculate body (MGB) subdivisions were identified and studied physiologically and anatomically, although other medial subdivisions were also identified anatomically. Neurons in the ventral subdivision (MGV) were sharply tuned for frequency, preferred narrowband stimuli, and were able to synchronize to rapid temporal modulations. Anterodorsal subdivision (MGAD) neurons appeared well suited for temporal processing, responding similarly to tone or noise stimuli but able to synchronize to the highest modulation frequencies and with the highest temporal precision among MGB subdivisions. Posterodorsal subdivision (MGPD) neurons differed substantially from the other two subdivisions, with many neurons preferring broadband stimuli and signaling changes in modulation frequency with nonsynchronized changes in firing rate. Most neurons in all subdivisions responded to increases in tone sound level with nonmonotonic changes in firing rate. MGV and MGAD neurons exhibited responses consistent with provision of thalamocortical input to core regions, whereas MGPD neurons were consistent with provision of input to belt regions.
Comparison of Auditory-Vocal Interactions Across Multiple Types of Vocalizations in Marmoset Auditory Cortex

Abstract:Auditory-vocal interaction, the modulation of auditory sensory responses during vocal production, is an important but poorly understood neurophysiological phenomenon in nonhuman primates. This sensory-motor processing has important behavioral implications for self-monitoring during vocal production as well as feedback-mediated vocal control for both animals and humans. Previous studies in marmosets have shown that a large portion of neurons in the auditory cortex are suppressed during self-produced vocalization but have primarily focused on a single type of isolation vocalization. The present study expands previous analyses to compare auditory-vocal interaction of cortical responses between different types of vocalizations. We recorded neurons from the auditory cortex of unrestrained marmoset monkeys with implanted electrode arrays and showed that auditory-vocal interactions generalize across vocalization types. We found the following: 1) Vocal suppression and excitation are a general phenomenon, occurring for all four major vocalization types. 2) Within individual neurons, suppression was the more general response, occurring for multiple vocalization types, while excitation tended to be more specific to a single vocalization type. 3) A subset of neurons changed their responses between different types of vocalization, most often from strong suppression or excitation for one vocalization to unresponsive for another, and only rarely from suppression to excitation. 4) Differences in neural responses between vocalization types were weakly correlated with passive response properties, measured by playbacks of acoustic stimuli including recorded vocalizations. These results indicate that vocalization-induced modulation of the auditory cortex is a general phenomenon applicable to all vocalization types, but variations within individual neurons suggest possible vocalization-specific coding.

Abstract:Single neurons in the primate auditory cortex exhibit vocalization-related modulations (excitatory or inhibitory) during self-initiated vocal production. Previous studies have shown that these modulations of cortical activity are variable in individual neurons' responses to multiple instances of vocalization and diverse between different cortical neurons. The present study investigated dynamic patterns of vocalization-related modulations and demonstrated that much of the variability in cortical modulations was related to the acoustic structures of self-produced vocalization. We found that suppression of single unit activity during multi-phrased vocalizations was temporally specific in that it was maintained during each phrase, but was released between phrases. Furthermore, the degree of suppression or excitation was correlated to the mean energy and frequency of the produced vocalizations, accounting for much of the response variability between multiple instances of vocalization. Simultaneous recordings of pairs of neurons from a single electrode revealed that the modulations by self-produced vocalizations in nearby neurons were largely uncorrelated. Additionally, vocalization-induced suppression was found to be preferentially distributed to upper cortical layers. Finally, we showed that the summation of all auditory cortical activity during vocalization, including both single and multi-unit responses, was weakly excitatory, consistent with observations from studies of the human brain during speech.
Laminar Subnetworks of Response Suppression in Macaque Primary Visual Cortex

Abstract:Cortical inhibition plays an important role in information processing in the brain. However, the mechanisms by which inhibition and excitation are coordinated to generate functions in the six layers of the cortex remain unclear. Here, we measured laminar-specific responses to stimulus orientations in primary visual cortex (V1) of awake monkeys (male, Macaca mulatta ). We distinguished inhibitory effects (suppression) from excitation, by taking advantage of the separability of excitation and inhibition in the orientation and time domains. We found two distinct types of suppression governing different layers. Fast suppression (FS) was strongest in input layers (4C and 6), and slow suppression (SS) was 3 times stronger in output layers (2/3 and 5). Interestingly, the two types of suppression were correlated with different functional properties measured with drifting gratings. FS was primarily correlated with orientation selectivity in input layers ( r = –0.65, p < 10 –9 ), whereas SS was primarily correlated with surround suppression in output layers ( r = 0.61, p < 10 –4 ). The earliest SS in layer 1 indicates the origin of cortical feedback for SS, in contrast to the feedforward/recurrent origin of FS. Our results reveal two V1 laminar subnetworks with different response suppression that may provide a general framework for laminar processing in other sensory cortices. SIGNIFICANCE STATEMENT This study sought to understand inhibitory effects (suppression) and their relationships with functional properties in the six different layers of the cortex. We found that the diversity of neural responses across layers in primary visual cortex (V1) could be fully explained by one excitatory and two suppressive components (fast and slow suppression). The distinct laminar distributions, origins, and functional roles of the two types of suppression provided a simplified representation of the differences between two V1 subnetworks (input network and output network). These results not only help to elucidate computational principles in macaque V1, but also provide a framework for general computation of cortical laminae in other sensory cortices.

Abstract:Abstract Sensory-motor interactions in the auditory system play an important role in vocal self-monitoring and control. These result from top-down corollary discharges, relaying predictions about vocal timing and acoustics. Recent evidence suggests such signals may be two distinct processes, one suppressing neural activity during vocalization and another enhancing sensitivity to sensory feedback, rather than a single mechanism. Single-neuron recordings have been unable to disambiguate due to overlap of motor signals with sensory inputs. Here, we sought to disentangle these processes in marmoset auditory cortex during production of multi-phrased ‘twitter’ vocalizations. Temporal responses revealed two timescales of vocal suppression: temporally-precise phasic suppression during phrases and sustained tonic suppression. Both components were present within individual neurons, however, phasic suppression presented broadly regardless of frequency tuning (gating), while tonic was selective for vocal frequencies and feedback (prediction). This suggests that auditory cortex is modulated by concurrent corollary discharges during vocalization, with different computational mechanisms.

Neural Integration of Audiovisual Sensory Inputs in Macaque Amygdala and Adjacent Regions

Abstract:Integrating multisensory inputs to generate accurate perception and guide behavior is among the most critical functions of the brain. Subcortical regions such as the amygdala are involved in sensory processing including vision and audition, yet their roles in multisensory integration remain unclear. In this study, we systematically investigated the function of neurons in the amygdala and adjacent regions in integrating audiovisual sensory inputs using a semi-chronic multi-electrode array and multiple combinations of audiovisual stimuli. From a sample of 332 neurons, we showed the diverse response patterns to audiovisual stimuli and the neural characteristics of bimodal over unimodal modulation, which could be classified into four types with differentiated regional origins. Using the hierarchical clustering method, neurons were further clustered into five groups and associated with different integrating functions and sub-regions. Finally, regions distinguishing congruent and incongruent bimodal sensory inputs were identified. Overall, visual processing dominates audiovisual integration in the amygdala and adjacent regions. Our findings shed new light on the neural mechanisms of multisensory integration in the primate brain.

Parallel Streams of Direct Corticogeniculate Feedback from Mid-level Extrastriate Cortex in the Macaque Monkey

Abstract:First-order thalamic nuclei receive feedforward signals from peripheral receptors and relay these signals to primary sensory cortex. Primary sensory cortex, in turn, provides reciprocal feedback to first-order thalamus. Because the vast majority of sensory thalamocortical inputs target primary sensory cortex, their complementary corticothalamic neurons are assumed to be similarly restricted to primary sensory cortex. We upend this assumption by characterizing morphologically diverse neurons in multiple mid-level visual cortical areas of the primate ( Macaca mulatta ) brain that provide direct feedback to the primary visual thalamus, the dorsal lateral geniculate nucleus (LGN). Although the majority of geniculocortical neurons project to primary visual cortex (V1), a minority, located mainly in the koniocellular LGN layers, provide direct input to extrastriate visual cortex. These "V1-bypassing" projections may be implicated in blindsight. We hypothesized that geniculocortical inputs directly targeting extrastriate cortex should be complemented by reciprocal corticogeniculate circuits. Using virus-mediated circuit tracing, we discovered corticogeniculate neurons throughout three mid-level extrastriate areas: MT, MST, and V4. Quantitative morphological analyses revealed nonuniform distributions of unique cell types across areas. Many extrastriate corticogeniculate neurons had spiny stellate morphology, suggesting possible targeting of koniocellular LGN layers. Importantly though, multiple morphological types were observed across areas. Such morphological diversity could suggest parallel streams of V1-bypassing corticogeniculate feedback at multiple stages of the visual processing hierarchy. Furthermore, the presence of corticogeniculate neurons across visual cortex necessitates a reevaluation of the LGN as a hub for visual information rather than a simple relay.

Sensory-motor Interaction in the Primate Auditory Cortex During Self-Initiated Vocalizations.

Abstract:Little is known about sensory-motor interaction in the auditory cortex of primates at the level of single neurons and its role in supporting vocal communication. The present study investigated single-unit activities in the auditory cortex of a vocal primate, the common marmoset (Callithrix jacchus), during self-initiated vocalizations. We found that 1) self-initiated vocalizations resulted in suppression of neural discharges in a majority of auditory cortical neurons. The vocalization-induced inhibition suppressed both spontaneous and stimulus-driven discharges. Suppressed units responded poorly to external acoustic stimuli during vocalization. 2) Vocalization-induced suppression began several hundred milliseconds prior to the onset of vocalization. 3) The suppression of cortical discharges reduced neural firings to below the rates expected from a unit's rate-level function, adjusted for known subcortical attenuation, and therefore was likely not entirely caused by subcortical attenuation mechanisms. 4) A smaller population of auditory cortical neurons showed increased discharges during self-initiated vocalizations. This vocalization-related excitation began after the onset of vocalization and is likely the result of acoustic feedback. Units showing this excitation responded nearly normally to external stimuli during vocalization. Based on these findings, we propose that the suppression of auditory cortical neurons, possibly originating from cortical vocal production centers, acts to increase the dynamic range of cortical responses to vocalization feedback for self monitoring. The excitatory responses, on the other hand, likely play a role in maintaining hearing sensitivity to the external acoustic environment during vocalization.

Abstract:Cross-modal plasticity within the visual and auditory cortices of early binocularly blind macaques is not well studied. In this study, four healthy neonatal macaques were assigned to group A (control group) or group B (binocularly blind group). Sixteen months later, blood oxygenation level-dependent functional imaging (BOLD-fMRI) was conducted to examine the activation in the visual and auditory cortices of each macaque while being tested using pure tones as auditory stimuli. The changes in the BOLD response in the visual and auditory cortices of all macaques were compared with immunofluorescence staining findings. Compared with group A, greater BOLD activity was observed in the bilateral visual cortices of group B, and this effect was particularly obvious in the right visual cortex. In addition, more activated volumes were found in the bilateral auditory cortices of group B than of group A, especially in the right auditory cortex. These findings were consistent with the fact that there were more c-Fos-positive cells in the bilateral visual and auditory cortices of group B compared with group A (p < 0.05). In conclusion, the bilateral visual cortices of binocularly blind macaques can be reorganized to process auditory stimuli after visual deprivation, and this effect is more obvious in the right than the left visual cortex. These results indicate the establishment of cross-modal plasticity within the visual and auditory cortices.
Distinct Superficial and deep laminar domains of activity in the visual cortex during rest and stimulation

Abstract:Spatial patterns of spontaneous neural activity at rest have previously been associated with specific networks in the brain, including those pertaining to the functional architecture of the primary visual cortex (V1). However, despite the prominent anatomical differences between cortical layers, little is known about the laminar pattern of spontaneous activity in V1. We address this topic by investigating the amplitude and coherence of ongoing local field potential (LFP) signals measured from different layers in V1 of macaque monkeys during rest and upon presentation of a visual stimulus. We used a linear microelectrode array to measure LFP signals at multiple, evenly spaced positions throughout the cortical thickness. Analyzing both the mean LFP amplitudes and between-contact LFP coherences, we identified two distinct zones of activity, roughly corresponding to superficial and deep layers, divided by a sharp transition near the bottom of layer 4. The LFP signals within each laminar zone were highly coherent, whereas those between zones were not. This functional compartmentalization was found not only during rest, but also when the receptive field was stimulated during a visual task. These results demonstrate the existence of distinct superficial and deep functional domains of coherent LFP activity in V1 that may reflect the intrinsic interplay of V1 microcircuitry with cortical and subcortical targets, respectively.

Abstract:Natural images comprise contours and boundaries defined by 1st-order luminance-modulated (LM) cues that are readily encoded by V1 neurons, and 2nd-order contrast-modulated (CM) cues that carry local, but not over-the-space, luminance changes. The neurophysiological foundations for CM processing remain unsolved. Here we used two-photon calcium imaging to demonstrate that V1 superficial-layer neurons respond to both LM and CM gratings in awake, fixating, macaques, with overall LM responses stronger than CM responses. Furthermore, adaptation experiments revealed that LM responses were similarly suppressed by LM and CM adaptation, with moderately larger effects by iso-orientation adaptation than by orthogonal adaptation, suggesting that LM and CM orientation responses likely share a strong orientation-non-selective subcortical origin. In contrast, CM responses were substantially more suppressed by iso-orientation than by orthogonal LM and CM adaptation, likely suggesting stronger orientation-specific intracortical influences for CM responses than for LM responses, besides shared orientation-non-selective subcortical influences. These results thus may indicate a subcortical-to-V1 filter-rectify-filter mechanism for CM processing: Local luminance changes in CM stimuli are initially encoded by orientation-non-selective subcortical neurons, and the outputs are half-wave rectified, and then summed by V1 neurons to signal CM orientation, which may be further substantially refined by intracortical influences.

Abstract:While it is well established that sensory cortical regions traditionally thought to be unimodal can be activated by stimuli from modalities other than the dominant one, functions of such foreign-modal activations are still not clear. Here we show that visual activations in early auditory cortex can be related to whether or not the monkeys engaged in audio-visual tasks, to the time when the monkeys reacted to the visual component of such tasks, and to the correctness of the monkeys' response to the auditory component of such tasks. These relationships between visual activations and behavior suggest that auditory cortex can be recruited for visually-guided behavior and that visual activations can prime auditory cortex such that it is prepared for processing future sounds. Our study thus provides evidence that foreign-modal activations in sensory cortex can contribute to a subject's ability to perform tasks on stimuli from foreign and dominant modalities.

Frontal-Auditory Cortical Interactions and Sensory Prediction During Vocal Production in Marmoset Monkeys

Abstract:The control of speech and vocal production involves the calculation of error between the intended vocal output and the resulting auditory feedback. Consistent with this model, recent evidence has demonstrated that the auditory cortex is suppressed immediately before and during vocal production, yet is still sensitive to differences between vocal output and altered auditory feedback. This suppression has been suggested to be the result of top-down signals containing information about the intended vocal output, potentially originating from motor or other frontal cortical areas. However, whether such frontal areas are the source of suppressive and predictive signaling to the auditory cortex during vocalization is unknown. Here, we simultaneously recorded neural activity from both the auditory and frontal cortices of marmoset monkeys while they produced self-initiated vocalizations. We found increases in neural activity in both brain areas preceding the onset of vocal production, notably changes in both multi-unit activity and local field potential theta-band power. Connectivity analysis using Granger causality demonstrated that frontal cortex sends directed signaling to the auditory cortex during this pre-vocal period. Importantly, this pre-vocal activity predicted both vocalization-induced suppression of the auditory cortex as well as the acoustics of subsequent vocalizations. These results suggest that frontal cortical areas communicate with the auditory cortex preceding vocal production, with frontal-auditory signals that may reflect the transmission of sensory prediction information. This interaction between frontal and auditory cortices may contribute to mechanisms that calculate errors between intended and actual vocal outputs during vocal communication.

Abstract:Previous studies have demonstrated that auditory cortex activity can be influenced by crosssensory visual inputs. Intracortical recordings in non-human primates (NHP) have suggested a bottom-up feedforward (FF) type laminar profile for auditory evoked but top-down feedback (FB) type for cross-sensory visual evoked activity in the auditory cortex. To test whether this principle applies also to humans, we analyzed magnetoencephalography (MEG) responses from eight human subjects (six females) evoked by simple auditory or visual stimuli. In the estimated MEG source waveforms for auditory cortex region of interest, auditory evoked responses showed peaks at 37 and 90 ms and cross-sensory visual responses at 125 ms. The inputs to the auditory cortex were then modeled through FF and FB type connections targeting different cortical layers using the Human Neocortical Neurosolver (HNN), which consists of a neocortical circuit model linking the cellular– and circuit-level mechanisms to MEG. The HNN models suggested that the measured auditory response could be explained by an FF input followed by an FB input, and the crosssensory visual response by an FB input. Thus, the combined MEG and HNN results support the hypothesis that cross-sensory visual input in the auditory cortex is of FB type. The results also illustrate how the dynamic patterns of the estimated MEG/EEG source activity can provide information about the characteristics of the input into a cortical area in terms of the hierarchical organization among areas.
