Tumor and Stem Cell Biology Genetic Ablation of Metadherin Inhibits Autochthonous Prostate Cancer Progression and Metastasis

Abstract:Metadherin (MTDH) overexpression in diverse cancer types has been linked to poor clinical outcomes, but definitive genetic proof of its contributions to cancer remains incomplete. In particular, the degree to which MTDH may contribute to malignant progression in vivo is lacking. Here, we report that MTDH is amplified frequently in human prostate cancers where its expression levels are tightly correlated with prostate cancer progression and poor disease-free survival. Furthermore, we show that genetic ablation of MTDH in the transgenic adenomcarcinoma of mouse prostate (TRAMP) transgenic mouse model of prostate cancer blocks malignant progression without causing defects in the normal development of the prostate. Germline deletion of Mtdh in TRAMPmice prolonged tumor latency, reduced tumor burden, arrested progression of prostate cancer at well-differentiated stages, and inhibited systemicmetastasis to distant organs, thereby decreasing cancer-related mortality 10-fold. Consistent with these findings, direct silencing of Mtdh in prostate cancer cells decreased proliferation in vitro and tumor growth in vivo, supporting an epithelial cell–intrinsic role of MTDH in prostate cancer. Together, our findings establish a pivotal role for MTDH in prostate cancer progression and metastasis and define MTDH as a therapeutic target in this setting. Cancer Res; 74(18); 5336–47. 2014 AACR.
Multi-dimensional analysis of heat-induced soybean protein hydrolysate gels subjected to ultrasound-assisted pH pretreatment

Abstract:This study aimed to evaluate the gelation characteristics of soybean protein hydrolysate (SPH) extracted by enzyme-assisted aqueous extraction. Specifically, the changes in gelation behaviors for heat-induced (95°C, 20 min) SPH dispersions treated with pH (pH 3, 5, 9; pH 7 as control) and ultrasound (U; 240 W, 30 min) were investigated. The results showed that typical gel behavior with high elastic nature in the viscoelasticity and network structures were observed during the heating process, where the disulfide bond played a dominant role in the gel network formation of all the samples. Notably, the heat-induced aggregation in the SPH gels was mainly formed by the association of the basic B polypeptide in 11S and β subunit in 7S. The most superior SPH gel was formed at pH 7 when assisted by ultrasonication during the heating process. This as-synthesized gel showed a uniform filamentous structure and exhibited the more excellent textural, rheological and thermal properties than those of the samples formed under acidic and alkaline conditions. These results are of great value in revealing the gelation mechanism of SPH.

Abstract:To explore greener modifier of konjac glucomannan, a new type of thermal irreversible gel using konjac glucomannan (KGM) and basic amino acids (L-lysine, and L-arginine) was prepared in the present study. The effects of different concentrations (1%, 2%, 3%, 4%, and 5%) of these two basic amino acids on the structure and properties of KGM hydrogels were mainly studied. The results showed that there was a better texture characteristic of composite gel as the concentration of amino acids was above 4%. Besides, with the increase in amino acid concentrations, the pore size of composite gel was denser and the syneresis of composite gel was more obvious, respectively. The rheological results indicated that the storage modulus (G ') was influenced by the concentration of amino acids and temperature. It was found that the two basic amino acids removed the acetyl group of KGM molecule and changed the original crystalline domain of KGM as evidenced by FT-IR and X-ray diffraction (XRD) analysis. Furthermore, compared to L-lysine, L-arginine could exert a greater effect on the KGM gel. In conclusion, the two basic amino acids (L-lysine, and L-arginine) could remarkedly improve the structure and properties of natural KGM and could be used as modifiers of the KGM hydrosol.
Effects of ultrasound pretreatment on the enzymatic hydrolysis of soy protein isolates and on the emulsifying properties of hydrolysates

Abstract:Soy protein isolate (SPI) was modified by ultrasound pretreatment (200 W, 400 W, 600 W) and controlled papain hydrolysis, and the emulsifying properties of SPIH (SPI hydrolysates) and USPIH (ultrasound pretreated SPIH) were investigated. Analysis of mean droplet sizes and creaming indices of emulsions formed by SPIH and USPIH showed that some USPIH had markedly improved emulsifying capability and emulsion stabilization against creaming during quiescent storage. Compared with control SPI and SPIH-0.58% degree of hydrolysis (DH), USPIH-400W-1.25% (USPIH pretreated under 400W sonication and hydrolyzed to 1.25% DH) was capable of forming a stable fine emulsion (d43=1.79 μm) at a lower concentration (3.0% w/v). A variety of physicochemical and interfacial properties of USPIH-400W products have been investigated in relation to DH and emulsifying properties. SDS-PAGE showed that ultrasound pretreatment could significantly improve the accessibility of some subunits (α-7S and A-11S) in soy proteins to papain hydrolysis, resulting in changes in DH, protein solubility (PS), surface hydrophobicity (H0), and secondary structure for USPIH-400W. Compared with control SPI and SPIH-0.58%, USPIH-400W-1.25% had a higher protein adsorption fraction (Fads) and a lower saturation surface load (Γsat), which is mainly due to its higher PS and random coil content, and may explain its markedly improved emulsifying capability. This study demonstrated that combined ultrasound pretreatment and controlled enzymatic hydrolysis could be an effective method for the functionality modification of globular proteins.

Abstract:Abstract In order to explore the impacts of limited hydrolysis combined with glycation modification on the structure, functional properties, allergenicity, and volatile compounds of silkworm pupae protein, the crude silkworm pupae protein was firstly extracted by alkaline-solution method, followed by Alcalase-treated limited hydrolysis for 10-30 min. Xylose was subsequently added and reacted for 1 h at 95 °C. Afterwards, the structure, functionality, allergenicity, and volatile compounds before and after modification were analysed. The molecular weight of modified silkworm pupae protein was remarkably decreased. The solubility was improved from 4.7 to 28.6%. The foaming ability was elevated by 21%. The in vitro digestibility was elevated by 34.0-44.4%. Furthermore, the abundance of six potential allergic proteins was remarkably reduced. In addition, the contents of off-flavour compounds in modified silkworm pupae proteins substantially decreased. Overall, limited hydrolysis combined with glycation modification can improve the functionality and flavour of silkworm pupae protein, while reduce the allergenicity.

Abstract:The purpose of this study was to investigate the influence of two-step sequential cross-linking induced by laccase and heat treatment on the conformation and gelling properties of low-salt myofibrillar proteins (MP) modified by konjac glucomannan (KGM) and ultrasound treatments. Adding sugar beet pectin (SBP), irrespective of laccase crosslinking, significantly decreased the fluorescence intensity of low-salt MPs (MP, UMP, UMP-KGM). Laccase crosslinking caused the exposure of hydrophobic and Tyr residues, induced moderate aggregation of MPs, and enhanced rheological properties and gel strength of composite MP gels, especially in the UMP-KGM sample. This result could also be corroborated by the highest WHC, improved textural characteristics, enhanced intermolecular interactions, and increased percentage in t-g-g conformation of S–S bridges in the SBP modified UMP-KGM gel after laccase crosslinking. Therefore, the dual crosslinking could be used to regulate the protein-protein and protein-polysaccharide interactions and improve the gel properties of low-salt meat products.

Abstract:This research focused on the unique aggregation behavior of l-Lysine (L-Lys) and l-Arginine (L-Arg) on quinoa protein (QP) gelation under microwave (MW) irradiation. The results of pH and conductivity manifested that the discrepancy of the influence of two alkaline amino acids on protein was due to the diversity of their electrochemical characteristics rather than to aggregation induced by pH-dominated acid-base transitions. Solubility, turbidity and particle size, ζ-potential results exhibited that L-Lys enhanced the ordered aggregation of protein induced by MW heating more than L-Arg. Circular dichroism (CD), UV absorption spectra, surface hydrophobicity (H0) and sulfhydryl groups demonstrated that L-Lys denaturized and unfolded proteins more vigorously than L-Arg, exposing more hydrophobic residues and boosting the hydrophobicity of the protein's surface. These findings emphasized that L-Lys and MW had a more pronounced synergistic effect on the strength and water holding capacity (WHC) of protein gels than L-Arg. Scanning electron microscopy (SEM) also revealed a denser porous network microstructure in the QP gels with L-Lys added.

Abstract:<p>The effect of dual-frequency ultrasound treatment with different working modes on the lysinoalanine (LAL) formation and structural characterization of rice dreg protein isolates (RDPI) was studied during alkaline exaction processing. Ultrasonic notably decreased the LAL amount of RDPI and enhanced the protein dissolution rate. The LAL content of RDPI, especially sequential dual frequency 20/40 kHz, decreased by 12.02 % (P < 0.05), compared to non-sonicated samples. Herein, the protein dissolution rate was higher. The changes in sulfhydryl groups was positively correlated with the LAL formation. The amino acids (AA) such as threonine (Thr), lysine (Lys), and arginine (Arg) were reduced, resulting in a decrease in LAL content following sonication. Besides, ultrasonication altered protein secondary structure by reducing random coil and β-sheet contents, while α-helix and β-turn contents increased. Alterations in the surface hydrophobicity, particle size, particle size distribution, and microstructure indicated more irregular fragment with microparticles of RDPI by sonochemical treatment. Thus, ultrasound treatment may be a new and efficacious process for controlling the LAL generation in prepared-protein food(s) during alkali extraction.</p>

Abstract:Glycated conjugates were prepared by heating aqueous dispersions of soy protein isolate (SPI) and maltodextrin (MD) after ultrasonic treatment (138 W/cm2, 20 kHz) for 5, 15, and 25 min. The effect of the ultrasonic pretreatment on the structure, physicochemical characteristics, and acid-induced gelation properties of the resulting SPI–MD conjugates was investigated. The analyses of free amino groups, degree of graft, intermediates, and browning intensity confirmed the formation of SPI–MD conjugates. The ultrasonic treatment promoted the glycation reaction, which led to the increase in the β-type ordered secondary structure and less compact tertiary conformation of conjugates. The ultrasonic treatment for 5 min increased the isoelectric point, surface hydrophobicity (H0), free sulfhydryl (-SH) groups, and particle size of the conjugates. These changes in the structural and physicochemical properties of the conjugates affected their acid-induced gelation. Glucono-δ-lactone was used to form acid gels at 25 °C. Although the glycation with MD deteriorated the acid-induced gelation properties of SPI, the 5 min ultrasonic conjugates formed gels faster than the untreated conjugate, and their gels exhibited higher storage modulus (G′), gel strength, and water holding capacity (WHC), as well as denser microstructure, than the untreated conjugate gels. However, prolonged ultrasonic treatment (15 or 25 min) weakened the gelation properties of the conjugates by promoting the glycation reaction and reducing the H0. Thus, an optimum ultrasonic pretreatment can be used to modify the structure of soy protein glycation conjugates to control their gelation properties.

Abstract:As a versatile ingredient, soy protein isolate (SPI) has been widely used in the food industry and the gelling property of SPI can be further modified using exogenous polysaccharides to achieve desired functionalities and nutritional properties. Therefore, in this study, the effect and mechanism of Lycium barbarum polysaccharides (LBP) on the gelling behavior of soy protein isolate (SPI) were investigated. It was found that the gel strength, water-holding capacity, and viscoelastic properties of SPI gel were improved with the addition of LBP (0.5–2.5%, w/v) in a concentration-dependent manner. Thermogravimetric analysis indicated that the thermal stability of SPI was also improved with the addition of LBP. The FTIR spectroscopy revealed that LBP addition had no significant effect on the α-helix and random coil content of SPI but significantly increased the β-sheet content and reduced the β-turn content. Furthermore, through intrinsic fluorescence, surface hydrophobicity, free sulfhydryl content, and protein solubility analysis, it was concluded that the electrostatic and hydrophobic interactions played a critical role in the formation of the SPI-LBP gel composite. Interestingly, the disulfide bond formation was the least contributor to the binary gel composite network. Overall, LBP is not only an important nutritional supplement but also a promising functional ingredient for improving the gel properties of SPI.

Abstract:This study investigated the effects of different ultrasonic power and ultrasonic time on the structure and emulsifying properties of pea protein isolate (PPI)–arabinose conjugates. An examination of the absorbance and color development of PPI-d-arabinose (Ara) conjugates found that compared with traditional heating, the degree of glycosylation of protein reached the maximum when the ultrasonic treatment power was 150 and the treatment time was 30 min. Structural analysis revealed that the content of disordered structures (β-turn and random coil) of the protein conjugates increased, the maximum emission wavelength of the fluorescence spectrum was red-shifted, and the UV second-order derivative values decreased. The protein structure unfolded, exposing more hydrophobic groups on the molecular surface. Ultrasonic treatment improved the emulsification of protein conjugates. The emulsifying activity index (EAI) increased to 19.7 and 19.3 m2/g, and the emulsifying stability index (ESI) also increased. The contact angle and zeta potential also demonstrate that ultrasonic power has a positive effect on emulsion stability. Based on examining the thermal stability of the emulsion, the ultrasonic treatment increased the thermal denaturation resistance of the protein. This result confirms that mild sonication can increase the degree of glycosylation reaction and improve the emulsification properties of protein–Ara conjugates, providing a theoretical basis for developing foods with excellent emulsification properties.

Abstract:In this study, whey protein isolate (WPI) was used as raw material to study the effects of l -lysine (Lys) on the gel properties and structure of WPI, and the application of modified WPI in fish sausage was studied. The results showed that with the increase of Lys concentration, the gel properties and water holding capacity of WPI were greatly improved, while the Lys concentration exceeding 4 mg/mL would produce adverse reactions, such as the decrease of chromaticity and gel hardness. At Lys concentration of 1.6 mg/mL, WPI showed the highest texture properties and good water holding capacity, which depended on the dense microstructure and reduced pore size. This was because WPI crosslinks with Lys through electrostatic interaction, reducing electrostatic repulsion and promoting its crosslinking and polymerization, which was manifested by the change of WPI structure and the increase of particle size. In addition, the hardness, chewiness, and cooking loss capacity of the surimi added with modified WPI were better than those of the pure fish sausage, corn starch and WPI single treatment group. This study can provide a theoretical basis for the study of WPI modified gel and provide guidance for the modified WPI used as gelling agent in surimi products.

Abstract:In this study, the effects of ultrasound treatment on the texture, physicochemical properties and protein structure of composite gels prepared by salted egg white (SEW) and cooked soybean protein isolate (CSPI) at different ratios were investigated. With the increased SEW addition, the ζ -potential absolute values, soluble protein content, surface hydrophobicity and swelling ratio of composite gels showed overall declining trends ( P <0.05), while the free sulfhydryl (SH) contents and hardness of exhibited overall increasing trends ( P <0.05). Microstructural results revealed that composite gels exhibited denser struture with the increased SEW addition. After ultrasound treatment, the particle size of composite protein solutions significantly decreased ( P <0.05), and the free SH contents of ultrasound-treated composite gels were lower than that of untreated composite gels. Moreover, ultrasound treatment enhanced the hardness of composite gels, and promoted the conversion of free water into non-flowable water, and when ultrasonic power exceeded 150 W, the hardness of composite gels could not be further enhanced. FTIR results indicated that ultrasound treatment facilitated the composite protein aggregates to form a more stable gel structure. The improvement of ultrasound treatment on the properties of composite gels was mainly by promoting the dissociation of protein aggregates, and the dissociated protein particles further interacted to form denser aggregates through disulfide bond, thus facilitating the crosslinking and reaggregation of protein aggregates to form denser gel structure. Overall, ultrasound treatment is an effective approach to improve the properties of SEW-CSPI composite gels, which can improve the potential ultilization of SEW and SPI in food processing.

Abstract:Glycated conjugation of plant protein such as soy protein isolate (SPI) with saccharides is one popular strategy to modify the physicochemical characteristics of these plant protein resources, which may be affected by the glycation methods including dry-heating and wet-heating. In this study, the impact of these two glycation methods on the rheological and emulsifying properties of a binary system made by SPI-gum Arabic (GA) was studied. The results indicated that dry-heating conjugates had higher viscosity and more elastic characteristics than those wet-heating conjugates. The emulsifying properties of SPI-GA conjugates by different preparation routes were evaluated by various oil phases including eugenol, cinnamaldehyde and soybean oil. Overall, emulsions stabilized by dry-heating conjugates showed lower zeta-potential value than those with wet heating conjugates. The interfacial properties of these conjugates were compared using soybean oil emulsion as a model. Higher emulsifying ability and stability were obtained by emulsions with dry-heating conjugates, which was attributed to their more compact structures, higher protein adsorption capacity and thicker viscoelastic films formed at the interface, and therefore enhanced electrostatic repulsion between droplets. The findings in this study are useful for fabrication and utilization of protein-polysaccharide glycation conjugates as emulsifiers in functional foods.

Abstract:In this study, we fed the larval of Bombyx mori silkworms with nanodroplets of liquid metal (LM) coated with microgels of marine polysaccharides to obtain stretchable silk. Alginate-coated liquid metal nanodroplets (LM@NaAlg) were prepared with significant chemical stability and biocompatibility. This study demonstrates how the fed LM@NaAlg acts on the as-spun silk fiber. We also conducted a series of characterizations and steered molecular dynamics simulations, which showed that the LM@NaAlg additions impede the conformation transition of silk fibroins from the random coil and α-helix to the β-sheet by the formation of hydrogen bonds between LM@NaAlg and the silk fibroins, thus enhancing the elongation at the breakpoints in addition to the tensile properties. The intrinsically highly stretchable silk showed outstanding mechanical properties compared with regular silk due to its 814 MPa breaking strength and a breaking elongation of up to 70%—the highest reported performance so far. We expect that the proposed method can expand the fabrication of multi-functional silks.

Abstract:As artificial extracellular matrix-like materials, silk-elastin-like protein (SELP) hydrogels, with excellent mechanical properties, high tunability, favorable biocompatibility, and controlled degradability, have become an important candidate in biomedical materials. In this study, SELP is composed of silk-like (GAGAGS) and elastin-like (GXGVP) tandem repeats, in which X residues are set as tyrosine and lysine. Furthermore, SELP polymers are prepared via SpyTag/SpyCatcher. To explore a gentler and more efficient enzymatic crosslinking method, an innovative method was invented to apply laccase to catalyze the formation of SELP hydrogels. Gelation could be successfully achieved in 2–5 min . SELP hydrogels mediated by laccase had the characteristic of low swelling rate, which could maintain a relatively stable shape even when immersed in water, and hence had the potential to be further developed into injectable biomaterials. Additionally, SELP hydrogels cross-linked by laccase showed excellent biocompatibility verified by L929 and HEK 293 T cells with cell viability >93.8 %. SELP hydrogels also exhibit good properties in sustained drug release and cell encapsulation in vitro. This study demonstrates a novel method to construct SELP hydrogels with excellent biocompatibility and expands the possibility of SELP-based material applications in biomedical fields.

Comparison of Gelation Time and Polyalcohol Effect on Hydrogels from Domestic and Wild Silk Fibroins

Abstract:Silk fibroin (SF) hydrogels were obtained from both domestic (Bombyx mori) and wild (Antheraea pernyi) silkworms from aqueous silk fibroin solutions at room temperature. The gelation time of the Antheraea pernyi (A. pernyi) SF solution was significantly shorter than that of the Bombyx mori (B. mori) SF solution. The secondary structures of the two kinds of hydrogels were also compared. In order to further reduce the gelation time, various amounts of polyethylene glycol (PEG) were blended with the silk fibroins of A. pernyi and B. mori. The gelation time of both A. pernyi SF and B. mori SF decreased with the increased amount of PEG. After freeze-drying, the hydrogels were characterized through X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), and Raman spectroscopy. Results showed that the addition of polyalcohol did not change the main secondary structure of the hydrogels. However, the addition of polyalcohol did reduce the gelation time and triggered additional formation of β-sheets.
