Abstract:In this study, bimetallic lanthanide coordination polymer nanoparticles (Tb-GMP-Eu CPNs) were developed for sensing alkaline phosphatase (ALP) by utilizing guanine monophosphate (GMP) as the bridge ligand to self-assemble with Tb3+ and Eu3+ in Tris-HCl buffer solution. In this polymer framework, an efficient energy transfer from Tb3+ to Eu3+ occurred because the emission energy of Tb3+ matched well with the absorption energy of Eu3+. As a result, the synthesized Tb-GMP-Eu CPNs exhibited the characteristic red emission of Eu3+ when excited at 290 nm. When ALP was introduced into the Tb-GMP-Eu CPNs system, it would destruct the combination between Tb and GMP by a dephosphorylation process, resulting in interruption of energy transfer from Tb3+ to Eu3+ and therefore the fluorescence quenching of Eu3+. Based on these facts, a fluorescence assay based on bimetallic lanthanide coordination polymer nanoparticles for ALP detection was developed. Such a Tb-GMP-Eu CPNs sensor exhibited sensitive response to ALP with a detection limit down to 0.004 U L-1. Moreover, the successful application of ALP inhibitor evaluation and determination of ALP in serum samples have been demonstrated. Therefore, the proposed method has great potential application for ALP detection in biological and clinical diagnosis.

Abstract:The abnormal level of alkaline phosphatase (ALP) in human blood stream indicates health problem. A novel fluorescent sensing system based on competitive coordination with europium ions (Eu 3+ ) integrated in a non-luminescent lanthanide-metal organic framework nanosheets (NO 2 -Eu-MOF NS) was developed for ALP analysis in human serum. The fluorescence signal is turned-on by the antenna ligand (1,4-phenylenediphosphonic acid (PEPP)) that competitively coordinates with Eu 3+ ions from the nanosheet to produce emissive species (Eu x (PEPP) y (NO 2 BDC) z ) in-situ. 4-Nitrophenyl phosphate (PNPP) is a substrate of ALP and specifically hydrolyzed into 4-nitrophenol. PNPP exhibits a strong absorption in the UV-Vis region (250–400 nm), which covers the excitation wavelength (280 nm) of Eu x (PEPP) y (NO 2 BDC) z . Therefore, the existing of PNPP causes Eu x (PEPP) y (NO 2 BDC) z non-emissive and, after hydrolysis with ALP, Eu x (PEPP) y (NO 2 BDC) z can be excited and give a detectable fluorescence signal. Based on the fluorescence change, quantitative analysis of ALP activity is thus achievable. This biosensing system was demonstrated in the determination of ALP activity with good linear relationship in the range of 5–1000 U L −1 . The limit of detection was 1.1 U L −1 (S/N = 3). The result reveals that the biosensor exhibits excellent sensing performance and anti-interference ability. It could be potentially utilized for clinical routine analysis of ALP activity in human serum.

Abstract:The development of sensitive and accurate detection of inorganic pyrophosphate (PPi) and pyrophosphatase activity (PPase) is important as they play vital roles in biological systems. However, it is still not satisfactory for most of the analytical methods for PPi and PPase because of their Cu2+-dependence and poor accuracy. Although the metal ion triggered aggregation-induced emission (AIE) of metal nanoclusters (NCs) offers a new approach to design a Cu2+-free strategy for the accurate determination of PPi and PPase recently, current methods are all focused on utilizing pure metal NCs. Alloy NCs incorporating the advantages of diverse metal usually can achieve improved behaviors in the application, such as enhanced sensitivity and stability. In this work, glutathione stabilized alloy Au/Ag NCs were synthesized via a simple method and used for the fluorescence detection of PPi and PPase based on a Zn2+-regulated AIE strategy. The controlled release of Zn2+ by PPi and PPase could regulate the AIE of Au/Ag NCs and be employed to response PPi concentration and PPase activity. This method processes simple procedure, high sensitivity and stability, and low toxicity. In addition, we also studied the AIE behaviors of this Au/Ag NCs and offer some fundamental understanding of the AIE properties of water-soluble alloy NCs. This study not only provides a straightforward and new approach for PPi and PPase determination but a basis for further study on the AIE properties of alloy NCs and their application.
IgA immune responses against acetaldehyde adducts and biomarkers of alcohol consumption in patients with IgA glomerulonephritis.

Abstract:BACKGROUND The pathogenesis of IgA glomerulonephritis (IgAGN) involves intense deposition of IgAs within the glomerulus. Although previous studies have shown that heavy drinking frequently leads to the generation of IgA antibodies against neo-antigens induced by ethanol metabolites and tissue deposition of IgAs, the associations between alcohol consumption, IgA immune responses, and kidney disease have not been examined. METHODS A total of 158 IgAGN patients (96 men, 62 women) were classified as abstainers (n = 38), moderate drinkers (n = 114), and heavy drinkers (n = 6) based on self-reported alcohol consumption. The reference population included 143 individuals (99 men, 44 women) who were either apparently healthy abstainers (n = 31), moderate drinkers (n = 43), or heavy drinkers devoid of liver disease (n = 69). The assessments included various biomarkers of alcohol consumption: carbohydrate-deficient transferrin (CDT), glutamyl transferase, gamma-CDT (combination of GGR and CDT), mean corpuscular volume (MCV), tests for liver and kidney function, serum immunoglobulin A (IgA), and specific IgA antibodies against acetaldehyde-protein adducts. RESULTS In male IgAGN patients, drinking status was significantly associated with MCV, p < 0.001; CDT, p < 0.01; and gamma -CDT, p < 0.05. In the reference population, all biomarkers and anti-adduct IgA levels were found to vary according to drinking status. In IgAGN patients, anti-adduct IgA levels were elevated in 63% of the cases but the titers did not associate with self-reported ethanol intake. CONCLUSIONS These data indicate high levels of IgA antibodies against acetaldehyde-derived antigens in IgAGN patients, which may hamper the use of the immune responses as markers of alcohol consumption among such patients. Future studies on the pathogenic and prognostic significance of anti-adduct immune responses in IgAGN patients are warranted.
Multifunctional lanthanide metal-organic framework based ratiometric fluorescence visual detection platform for alkaline phosphatase activity

Abstract:A turn-on/off ratiometric fluorescence detection platform based on multifunctional lanthanide metal-organic framework (Ln-MOF) and an enzymatic cascade reaction is proposed for alkaline phosphatase (ALP) activity assay. L-phosphotyrosine is hydrolyzed to levodopa (L-dopa) by two steps of enzymatic reaction. L-dopa further reacts with naphthoresorcinol to produce carboxyazamonardine with strong emission at 490 nm. In this process, multifunctional Ln-MOF (Cu@Eu-BTC, BTC is the 1,3,5-benzenetricarboxylic acid) acts not only as a nanozyme to catalyze the fluorogenic reaction between L-dopa and naphthoresorcinol but also as a fluorescence internal standard. The emission of Cu@Eu-BTC at 620 nm is quenched by phosphate anions, and the dual-response ratiometric fluorescence (F490/F620) can be achieved. A good linear relationship was obtained between őĒ(F490/F620) and ALP activity in the range 0.3‚Äď24 U L‚ąí1 with the detection limit of 0.02 U L‚ąí1. In addition,¬†a portable assay tube was designed for visual and point-of-care testing of ALP activity by color variation (ratiometric chromaticity). Both the ratiometric fluorescence detection and the visual detection methods were successfully applied to monitor ALP activity in human serum samples with recovery between 95.5%‚Äď109.0% and 94.0%‚Äď110.1%, and relative standard deviation less than 8.1% and 9.5%, respectively. As far as we know, this is the first report of ALP activity assay assisted by multifunctional Ln-MOF.

Abstract:Fluorescent silver coordination polymer nanoparticles (Ag-TPA CPNs) were synthesized using a combination of terephthalic acid (TPA) and silver nitrate via an ultrarapid microwave-assisted strategy within 15 min. The Ag-TPA CPNs displayed a high fluorescent quantum yield (QY = 20.19%) and large Stokes shift (~200 nm), with two emission peaks at 490 nm and 520 nm under an excitation wavelength of 320 nm. A fluorescent "turn-off" method using fluorescent Ag-TPA CPNs was applied to detect the alkaline phosphatase (ALP) activity on the basis of the ALP-catalyzed hydrolysis of ascorbic acid 2-phosphate (AA2P) to ascorbic acid (AA), and the AA product triggered the reduction of Ag+ ions into silver nanoparticles. The fluorescent lifetime of Ag-TPA CPNs decreased from 3.93 ms to 3.80 ms after the addition of ALP, which suggests that this fluorescent "turn-off" detection of ALP activity is a dynamic quenching process. The fluorescent intensity had a linear relationship with the concentration of ALP in the range of 0.2-12 mU/mL (r = 0.991) and with a limit of detection (LOD) of 0.07 mU/mL. It showed high selectivity in ALP detection towards metal ions and amino acids, as well as other enzymes such as horseradish peroxidase, glucose oxidase, tyrosinase, trypsin, lysozyme, and superoxides. When it was applied for the fluorescent "turn-off" detection of ALP activity in serum samples, mean recovery levels ranging from 99.5% to 101.2% were obtained, with relative standard deviations (RSDs) lower than 4% accuracy. Therefore, it is an efficient and accurate tool for analyzing ALP levels in biosamples.

Abstract:A fluorescence sensing method for the quantification of alkaline phosphatase (ALP) was developed by integrating the strand displacement amplification with DNAzyme-catalytic recycling cleavage of molecular beacons. ALP can hydrolyze a 3′-phosphoralated primer into a 3′-hydroxy primer which can initiate the strand displacement amplification to produce the Mg 2+ -dependent DNAzyme. The DNAzyme can then catalyze the cleavage of the DNA molecular beacon labeled with FAM fluorophore at its 5′-end and BHQ1 quencher at its 3′-end, turning on the fluorescence of FAM fluorophore. The content of ALP in a sample can be deduced from the measured fluorescence intensity. Due to the cascading nature of its amplification strategy, the proposed method achieved sensitive and specific ALP detection in human serum samples. Its results were in good consistent with the corresponding values obtained by a commercial ALP detection kit. The limit of detection of the proposed method for ALP is about 0.015 U/L, lower than some methods recently reported in literature, demonstrating its potential for ALP detection in biomedical research and clinical diagnosis.

Abstract:A fluorimetric method is described for the determination of alkaline phosphatase (ALP) activity. It is based on the use of polyethyleneimine-coated silver nanoclusters (AgNCs), which display an intense blue fluorescence peaking at 450 nm (under 375 nm excitation). ALP catalyzes the dephosphorylation of the thiophosphate amifostine to generate a thiol that binds to the AgNCs and causes its fluorescence to be quenched. Under the optimal experimental conditions, fluorescence linearly drops in the 0.08-2.0 U L-1 ALP activity range, and the limit of detection is 0.02 U L-1. The method was successfully applied to the determination of ALP activity in spiked human serum samples. Graphical abstract Alkaline phosphatase (ALP) catalyzes the degradation of amifostine with a generation a thiol product. Abstract:Alkaline phosphatase (ALP) is a commonly used marker in clinical practice, and this enzyme is a key indicator for diagnosing various diseases. In this study, we describe the development of a reliable and novel fluorescent assay for ALP detection based on chitosan carbon dots (C-CDs, peak emission, 412 nm) and calcein (peak emission, 512 nm). In the presence of Eu3+ (which binds calcein), the fluorescence intensity of calcein is quenched. Utilizing the ALP-triggered generation of phosphate ions (PO43−) from the substrate p-nitrophenyl phosphate (pNPP), the Eu3+ ions bind PO43− (which shows a higher affinity toward Eu3+ than calcein), and the fluorescence of calcein is recovered. As a consequence, C-CDs fluorescence is decreased by inner filter effect (IFE). Exploiting these changes in the fluorescence intensity ratio of C-CDs and calcein, we developed a high sensitivity, accurate, and easily synthesized ratiometric fluorescence probe. Our novel fluorescent bioassay demonstrates good linear relationship in the 0.09–0.8 mU mL−1 range, with a low detection limit of 0.013 mU mL−1. The excellent applicability of this novel assay in HepG2 cells and human serum samples demonstrates that our novel method has excellent biomedical research and disease diagnosis prospects.

Abstract:This case report describes the third recurrence of a poorly differentiated follicular thyroid carcinoma that led to destruction of the thyroid cartilage. Curative surgery required hemilaryngectomy. Because of three previous operations (including neck dissection) and repeated external radiation, the typical laryngoplastic procedures and muscle flaps were not applicable. We therefore employed a stretched jejunal patch to cover the defect so that the neoglottic slit rendered respiration and phonation possible. Thus this approach resulted in functional reconstruction and obviated the need for a tracheostoma. We conclude that the use of a jejunal transplant represents a viable alternative to local reconstruction, particularly in cases complicated by previous surgery and external radiation.
Synergistic effect-mediated fluorescence switching of nitrogen-doped carbon dots for visual detection of alkaline phosphatase

Abstract:The natural enzyme alkaline phosphatase (ALP), which is associated with a large number of diseases, plays an essential role in clinicopathological examinations. Therefore, it is of great significance to quantify the ALP activity levels. In this work, a switchable fluorescence assay of detecting alkaline phosphatase (ALP) activity was proposed using Fe3+ to adjust the fluorescence of nitrogen-doped carbon dots (CDs). Wherein, CDs chelated with Fe3+, leading to a remarkable and dynamic fluorescence quenching. However, ascorbic acid (AA) and phosphate group (Pi), resulting from the enzymatic reaction between ascorbic acid-2-phosphate (AAP) and ALP, enabled the great fluorescence recovery, which was possibly related with the synergistic effect including the chemical redox of AA towards Fe3+ and the formation of a stable FePO4 complex. Based on this synergistic effect-mediated switching mechanism, the indirect detection of ALP activity was developed in the range of 15–445 U/L with the limit of detection at 2.576 U/L (3σ/k) and the limit of quantitation at 8.587 U/L (10σ/k), which could be observed by the naked-eyes. Then, the proposed method was further applied to the detection of ALP activity in human serum and urine samples with satisfactory results, which was in good agreement with the result obtained by ALP commercial kit. This proposed synergistic effect from AA and Pi provides a new insight for ALP activity detection.

Abstract:In this work, a novel approach for simple and sensitive determination of alkaline phosphatase (ALP) is developed on the basis of an inner filter effect of p-nitrophenylphosphate (PNPP) on the fluorescence of gold nanoclusters (AuNCs). AuNCs with a high quantum yield of 12% were synthesized by one-pot strategy and were directly applied as fluorescent substance. When AuNCs were mixed with PNPP, the fluorescence of the AuNCs was remarkably quenched or was decreased via the inner filter effect since the absorption spectrum of PNPP overlaps well with the excitation spectrum of the AuNCs. While in the presence of ALP, PNPP was catalytically hydrolyzed into p-nitrophenol, which has different absorption characteristics from those of PNPP, resulting in the recovery of the AuNCs fluorescence. Thus, a novel “turn-on” fluorescent sensor for detecting ALP was established with a detection limit as low as 0.002 U/L (signal-to-noise ratio of 3). Abstract:The conditioni broadlv defined by the title of this paper was brought to our notice bv the clinical observation of six children at the Hospital for Sick Children, Great Ormond Street, all of whomn presented the symptoms of wasting, constipation, and regurgitation of undigested food from the cesophaguis. Although the term ' cardiospasm 'was applied to these children for the purposes of the hospital records, the underlving morbid condition which existed at the lower end of the cesophagus remained in most cases obscure. In the hope of reaching a clearer understanding of the pathological basis of these caves. the hospital records were searched, and six additional cases of similar natture were found. Of the total of twelve children with cesoph.ageal obstructio), five have died. With one exception these have come to post-mortem examination, and the pathologist's reports have been available. In addition, radiological evidence has been forthcoming in all buit two cases. To be complete. this article should incluide that form of obstruction met (luring the first two weeks of life due to complete atresia of the Wesophagus. Instances of this condition have been frequenitly recorded, and in 1917 Cautlev )ublished a full review of the subject. The syrmptoms are well defined, and the condition is incompatible with life, so that fturther allusion to this group will not be made. Acquired stenosis such as follow s upon the ingestion of caustics is also excluded from consideration. The study of the cases at our disposal was simplified by grouping them according to the level in the cesophagus at which the obstruction occurred, and we shall consider them in this manner. The first and largest group consists of 10 children who had obstruction at the level of the seventh thoracic vertebra, halfway between the level of the bifurcation of the trachea and the diaphragm. It is to these cases that we wish to draw particular attention, because they fcrnm a clearly defined clinical entity, which is, hoNever, apt to be confused with the condition called ' cardiospasm ' or 'achalasia of the cardia.' Abstract:Abnormal expression of alkaline phosphatase (ALP) in serum has received considerable attention in health monitoring and disease diagnosis. However, conventional optical analysis based on a single signal must compromise background interference and limited sensitivity in trace analysis. As an alternative candidate, the ratiometric approach depends on the self-calibration of two independent signals in a single test to minimize interferences from the background for accurate identification. Here, a carbon dot/cobalt-metal organic framework nanocoral (CD/Co-MOF NC) mediated fluorescence-scattering ratiometric sensor has been developed for simple, stable, and highly sensitive detection of ALP. ALP-responsive phosphate production was used to coordinate cobalt ion and collapse the CD/Co-MOF NC, resulting in the recovery of fluorescence signal from dissociative CDs and the decrease of second-order scattering (SOS) signal from the cracked CD/Co-MOF NC. The ligand-substituted reaction and the optical ratiometric signal transduction provide a rapid and reliable chemical sensing mechanism. The ratiometric sensor effectively converted ALP into a ratio signal of fluorescence-scattering dual emission throughout a wide linear concentration range of six orders of magnitude with a detection limit of 0.6 mU/L. In addition, self-calibration of fluorescence-scattering ratiometric method can reduce background interference and improve sensitivity in serum, approaching recoveries of ALP from 98.4% to 101.8%. Due to the above advantages, the CD/Co-MOF NC mediated fluorescence-scattering ratiometric sensor readily provides rapid and stable quantitative detection of ALP as a promising in vitro analytical method for clinical diagnostics.

Abstract:Alkaline phosphatase (ALP) is of extraordinary importance in living organisms and widely serves as a labeling enzyme in biochemical assays. Conventional methods based on the colorimetric readout of chemicals have issues in the photosensitivity and the difficulty in visibly distinguishing the color intensity. Herein, we developed a plasmonic assay for detecting ALP activity and its inhibitors based on gold nanoparticles (AuNPs) with good stability and naked-eye readout. Ag(I) specifically oxidizes TMB to produce positively charged oxTMB, which electrostatically aggregates negatively charged AuNPs. ALP as an enzyme specifically hydrolyzes a substrate to generate ascorbic acid, which reduces Ag(I) and modulates the aggregation of AuNPs. This plasmonic ALP assay has a linear detection range of 5–50 mU/mL with a limit of detection of 0.39 mU/mL. In combination with a smartphone, it enables monitoring ALP inhibitors including dimethoate and arsenic that are environmental contaminants. This plasmonic sensing strategy with good stability, visible readout, high specificity, low cost, and ease of operation provides a promising platform for enzyme detection and environmental monitoring.

Abstract:In this work, a novel and sensitive fluorescence sensing system for alkaline phosphatase (ALP) was constructed using a bifunctional copper metal-organic framework (Cu@MOF) nanozyme, which had excellent oxidase-mimetic activity and fluorescence properties. Owing to the presence of 2-amino-1,4-benzenedicarboxylic acid (1,4-BDC-NH2) ligand, Cu@MOF displays excellent fluorescence performance at 444 nm. Additionally, Cu2+ endows the oxidase-like activity of Cu@MOF, which could trigger p-phenylenediamine (PPD) to be oxidized to a brown product (PPDox) and quench the photoluminescence of Cu@MOF through the inner filtration effect (IFE). As the preferential affinity of ATP for Cu2+, the catalytic activity of Cu@MOF was significantly reduced once ATP was added, thus PPD could not be oxidized and fluorescence was recovered. In the presence of ALP, ATP was hydrolyzed to adenosine and Pi, which allowed Cu@MOF to regain its catalytic activity and continued to catalyze the generation of PPDox. The fluorescence of Cu@MOF was therefore weakened once again. The ALP activity was directly proportional to the degree of decrease in fluorescence intensity. Thus, this novel fluorescence sensing strategy had a linear range of 0.5-60 U/L and the limit of detection was 0.14 U/L. The established sensing method could also be used to for ALP inhibitors screening, and achieved satisfactory results in determining the level of ALP activity in human serum.

Abstract:A novel fluorescence assay is proposed through activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP) strategy for alkaline phosphatase (ALP) activity detection. First of all, 2-bromo-2-methylpropionic acid (BMP) was employed as the initiator to modify on the surface of the magnetic nanoparticle (Fe3O4-MNP) by amide bonding. Then, ascorbic acid (AA) produced by ALP catalyzed the phosphate group removal from L-ascorbic acid 2-phosphate sesquimagnesium salt hydrate (AAPS), which underwent a redox reaction with Cu(II) and the product Cu(I) triggered the ARGET ATRP reaction. Finally, a strong fluorescent signal could be detected at 514 nm due to numerous fluorescent monomers being grafted to the Fe3O4-MNPs surface (Ex = 490 nm, Em = 514 nm). Under optimal experimental conditions, the linear range of this fluorometric assay for ALP activity was 1-80 mU mL-1, and the detection limit was 0.68 mU mL-1. The method exhibited excellent selectivity and satisfactory results were obtained in the inhibition rate and human serum experiments. Therefore, this ALP activity detection strategy has great potential for clinically relevant disease detection and drug screening. Abstract:Alkaline phosphatase (ALP), as an immunological label, is widely used in biochemical assays. Here, a simple yet effective strategy for ALP activity detection was proposed on the basis of in situ formation of Prussian blue nanoparticles and polychromatic superposition effect. Firstly, ascorbic acid, a product from ALP-catalyzed hydrolysis of 2-phospho-L-ascorbic acid (AAP), converted yellow ferricyanide into ferrocyanide. Then, the specific reaction between ferrocyanide and ferric ions (Fe3+) initiated the generation of Prussian blue nanoparticles in situ. Meanwhile, the residual AAP chelated with Fe3+, and a stable Fe3+-AAP complex was quickly formed. When Prussian blue nanoparticles mixed with brown Fe3+-AAP complex and yellow ferricyanide at different ratios, a distinct color variation was presented. Therefore, a sensitive multicolor assay of ALP activity with a detection limit of 1.0 U/L was realized by simply blending commercially available reagents. Furthermore, magnetic sandwich and competitive sensing platforms for multiple biomarkers detection were constructed by combining the ALP-regulated multicolor system with the well-developed aptasensor. The feasibility of the sensors was convincingly demonstrated by using thrombin and prostate specific antigen as model targets. In addition, the proposed multicolor strategy was employed for evaluating inhibition efficiency, and shows potential in visual screening of enzyme inhibitors. Such a facile, sensitive and low-cost sensing strategy provides a new perspective to develop universal platforms of point-of-care testing.

Abstract:lkaline phosphatase (ALP) plays significant roles in regulating intracellular processes and is an important biomarker connected to several diseases. In this work, one facile and sensitive sensing platform based on CQD-silver nanocomposites (CQD-silver NPs) for colorimetric detection of alkaline phosphatase (ALP) was introduced. ALP triggers the removal of the phosphate group of ascorbic acid 2-phosphate (AA2P), which is then transformed into ascorbic acid (AA). The as-obtained AA can easily cause significant aggregation of monodispersed NPs and cause the system color to turn from bright yellow to gray. Based on the color change of the ratio of 490 nm/630 nm, ALP was sensitively and selectively detected. Under the optimum, the established method showed linearity for ALP in the range of 0.1-50 U L-1 and the detection limit was low at 0.035 U L-1, and it was subjected to ALP inhibitor screening from goji berry extract. These results indicated that the colorimetric system can be used as a simple tool for visual and fast evaluation of ALP activity as well as providing an alternative to screen ALP inhibitors.
