Abstract:Marked regional differences in prevalence of pneumoconiosis are apparent in the US despite comparable dust exposure. In the present study, we examined the ability of 28 coal samples to release bioavailable iron (BAI) and calcium, as well as other metals such as Cr, Ni, Cu, and Co, from three coal mine regions in Utah (UT), West Virginia (WV), and Pennsylvania (PA), respectively. BAI is defined as iron (both Fe2+ and Fe3+) released by the coals in 10 mM phosphate solution, pH 4.5, which mimics conditions of the phagolysosomes in cells. We found that coals from the UT, WV, and PA regions released average levels of BAI of 9.6, 4658.8, and 12149 parts per million (ppm, w/w), respectively, which correlated well with the prevalence of pneumoconiosis from that region (correlation coefficient r = 0.92). The low levels of BAI in the UT coals were due to the presence of calcite (CaCO3), which was shown to be preferentially acid solubilized before iron compounds. Release of iron by two coal samples from the PA and UT regions was further examined in vitro in human lung epithelial A549 cells. We found that the coal from PA, with a high prevalence of pneumoconiosis, released BAI in a dose-dependent manner, both in tissue culture media and in A549 cells. At 2 microg/cm2, levels of lipid peroxidation induced by the PA coal were increased 112% over control cells at 24 h treatment, and were sustained at this level for 3 days. The coal from UT, with a low prevalence of pneumoconiosis, induced a marginal increase in cellular iron at 5 and 10 microg/cm2 treatments and had no effect on lipid peroxidation. Calcium levels in the cells treated with the PA and UT coals were 8.6 and 11.5 micromoles/10(6) cells, respectively, and were significantly higher than that in the controls (5.3 micromoles/10(6) cells) [corrected]. Our results suggest that the differences in the BAI content in the coals may be responsible for the observed regional differences in the prevalence of pneumoconiosis. Therefore, BAI may be a useful characteristic of coal for predicting coal's toxicity.

Abstract:Haber et al. (1) have made a careful and detailed study of the effects of disease prior to an outbreak on estimates of vaccine efficacy following the outbreak. Their calculations are based on the assumption that the vaccine affords "all or nothing" protection, that is, that a proportion of vaccinees are wholly protected against infection while the remainder are totally unprotected. One issue of considerable practical importance in estimating vaccine efficacy in outbreak investigations is whether to include or exclude individuals infected prior to the outbreak from the attack rate denominators. The authors review various options and conclude that, under most circumstances, including prior cases in the denominators will not produce serious bias. However, previous work (2) has shown that, while this is indeed correct for "all or nothing" vaccines, it is not so for other types of vaccines. In particular, for so-called "leaky" vaccines in which the effect of vaccination is to reduce disease incidence in all vaccinees, without giving complete protection to any, considerable bias may occur resulting in low or even negative vaccine efficacies in outbreaks or long-term follow-up studies of age-specific efficacy. For example, consider a secondary school outbreak of pertussis in 15-year-old students. Suppose, for simplicity, that vaccination took place close to birth, that the average annual incidence of pertussis prior to the outbreak was 10 percent, and that the vaccine efficacy is 80 percent, reducing the annual incidence to 2 percent. By the age of 15 years, 22.3 percent (100 exp<-0.1 X 15)) of those unvaccinated are susceptible, compared with 74.1 percent (100 exp(-0.2 X 0.1 X 15)) of those vaccinated. The vaccine efficacy during the outbreak (which is the age-specific efficacy at the age of 15 years) calculated by including prior cases in the attack rate denominators is then 34 percent (100 X (1 (0.2 X 0.741)/0.223)), rather than 80 percent. If the average annual incidence in the 0to 15-year age group had been 15 percent, the observed efficacy during the outbreak would have been negative: —21 percent.

Abstract:Coal mining has also been associated with adverse environmental and health impacts including cancer and respiratory disorders, with the presence of thermal power plants exacerbating the problem of heavy metal pollution. Minimal studies have been conducted on the environmental impacts, health risks, and bioaccessibility of heavy metals in coal mine areas. Consequently, samples of street dust were collected from different locations in the Singrauli mine complex and analysed. Heavy metals (Cu, Ni, Zn, Cr, Co, As, and Mo) were found to be higher than the background concentration, with the maximum concentration was found in areas close to the Thermal Power Plants, like Near Vindyachal TPP, Near Shakti Nagar TPP, and Anpara. The highest geo-accumulation index value was found for Co, Mo, Zn, and As, indicating moderate to strong pollution levels. Health risk assessment (for both adults and children) revealed that Cr and Fe posed significantly higher Hazard Quotient and Hazard Index (HI) values, indicating significant non-carcinogenic threats. Moreover, Carcinogenic Risk (CR) values for Cd, Cr, and Ni indicated a risk of carcinogenicity to the public exposed to road dust. The study also examined the bioaccessibility of the metals, which showed that the gastric phase accumulated a higher percentage of Ni (42.52%), Pb (34.79%), Co (22.22%), As (20%) and Cu (15%) than the intestinal phase. Strong positive correlation was observed between metal concentration (Cu, Pb, Cr, Fe, Zn, and Mn), HI, and CR of adult and child, while bioaccessibility of intestinal phase was positively correlated with gastric phase of metals (Cu, Ni, Co, As, and Mn).

Abstract:The present study aimed to examine elemental contamination near the residential areas of four coal thermal power plants in India. Water and surface soil samples were collected from various geographic sites around Chennai, Korba, Rupnagar, and Udupi power plants. The study identified aluminum (Al), arsenic (As), chromium (Cr), cobalt (Co), copper (Cu), cadmium (Cd), iron (Fe), lithium (Li), magnesium (Mg), manganese (Mn), mercury (Hg), nickel (Ni), zinc (Zn), and lead (Pb) in water and soil samples. Further, soil morphology was also studied using the SEM and EDX. Additionally, using non-cancerous and cancerous risk indices, human health hazards through consuming contaminated water and soil were estimated. Results indicated that Fe and Mn concentrations in water exceeded the standard limits, i.e ., the Fe (0.36 mg/l) at site 2 (Chennai), 0.490 mg/l at site 1, and 1.907 mg/l at site 3 (Udupi), while Mn was found as 0.28 mg/l at site 4 (Chennai), 0.27 mg/l at site 3 (Udupi), and 1.52 mg/l at site 4 (Rupnagar). In the case of soil, the maximum metal concentration (mg/kg) was found in Rupnagar, followed by Chennai, Korba, and Udupi. The hazard index (HI) for non-cancerous and cancerous risks was found within the USEPA standard limits, indicating insignificant health risks to the adjoining population of coal thermal power plants. This study provides baseline information regarding the elemental composition in water and soil and the associated health risk status around Indian coal thermal power plants.

Abstract:This study evaluates the cytotoxic and genotoxic effects of PM2.5 collected from an open-cast coal mining area in northern Colombia. Cyclohexane (CH), dichloromethane (DCM), and acetone (ACE) extracts were obtained using Soxhlet extraction to isolate compounds of different polarities. Human lymphocytes were exposed to the extracted compounds, and cytotoxicity and genotoxicity were assessed using the cytokinesis block micronucleus (CBMN) and comet assays, incorporating FPG and ENDO III enzymes to detect oxidative DNA damage. Chemical analysis revealed that the organic fractions consisted mainly of modified hydrocarbons and volatile organic compounds. The CBMN assay showed a significant increase in micronuclei in binucleated (MNBN) and mononucleated (MNMONO) cells and nucleoplasmic bridges (NPB) in exposed lymphocytes. The comet assay revealed substantial oxidative DNA damage, particularly with the ACE extract, which significantly increased oxidized purines and pyrimidines. DCM induced similar effects, while CH showed moderate effects. CREST immunostaining revealed aneugenic activity, particularly in cells exposed to ACE and DCM extracts. These results suggest that polar fractions of PM2.5, likely containing metals and modified PAHs, contribute to DNA damage and chromosomal instability. The study highlights the need to monitor the composition of PM2.5 in mining regions to implement stricter environmental policies to reduce exposure and health risks.

Abstract:(1) Background: Despite the encouraging indications regarding the suitability (biocompatibility) of iron carbide nanoparticles (ICNPs) in various biomedical applications, the published evidence of their biosafety is dispersed and relatively sparse. The present review synthesizes the existing nanotoxicological data from in vitro studies relevant to the diagnosis and treatment of cancer. (2) Methods: A systematic review was performed in electronic databases (PubMed, Scopus, and Wiley Online Library) on December 2023, searching for toxicity assessments of ICNPs of different sizes, coatings, and surface modifications investigated in immortalized human and murine cell lines. The risk of bias in the studies was assessed using the ToxRTool for in vitro studies. (3) Results: Among the selected studies (n = 22), cell viability emerged as the most frequently assessed cellular-level toxicity endpoint. The results of the meta-analysis showed that cell models treated with ICNPs had a reduced cell viability (SMD = −2.531; 95% CI: −2.959 to −2.109) compared to untreated samples. A subgroup analysis was performed due to the high magnitude of heterogeneity (I2 = 77.1%), revealing that ICNP concentration and conjugated ligands are the factors that largely influence toxicity (p < 0.001). (4) Conclusions: A dose-dependent cytotoxicity of ICNP exposure was observed, regardless of the health status of the cell, tested organism, and NP size. Inconsistent reporting of ICNP physicochemical properties was noted, which hinders comparability among the studies. A comprehensive exploration of the available in vivo studies is required in future research to assess the safety of ICNPs’ use in bioimaging and cancer treatment.

Abstract:The increasing prevalence of coal mine dust-related lung diseases in coal miners calls for urgent and meticulous scrutiny of airborne respirable coal mine dust (RCMD), specifically focusing on particles at the nano-level. This necessity is driven by expanding research, including the insights revealed in this paper, that establish the presence and significantly increased toxicity of nano-sized coal dust particles in contrast to their larger counterparts. This study presents an incontrovertible visual proof of these tiny particulates in samples collected from underground mines, utilizing advanced techniques such as scanning electron microscopy (SEM) and energy-dispersive spectroscopy (EDS). The intricate elemental composition of nano-sized coal dust identified through EDS analysis reveals the presence of elements such as silica and iron, which are known to contribute to lung pathologies when inhaled over prolonged periods. The outcomes of the statistical analyses reveal significant relationships between particle size and elemental composition, highlighting that smaller particles tend to have higher carbon content, while larger particles exhibit increased concentrations of elements like silica and aluminum. These analyses underscore the complex interactions within nano-sized coal dust, providing critical insights into their behavior, transport, and health impacts. The nano-sized coal dust could invade the alveoli, carrying these toxic elements from where they are impossible to exhale. The revelation of nano-sized coal dust's existence and the associated health hazards necessitate their incorporation into the regulatory framework governing the coal mining industry. This study lays the groundwork for heightened protective measures for miners, urging the invention of state-of-the-art sampling instruments, comprehensive physicochemical profiling of RCMD nanoparticles, and the pursuit of groundbreaking remedies to neutralize their toxic impact. These findings advocate for a paradigm shift in how the coal mining industry views and handles particulate matter, proposing a re-evaluation of occupational health standards and a call to action for protecting coal miners worldwide.

Abstract:Coal mine workers are continuously exposed to respirable coal mine dust (RCMD) in workplaces, causing severe lung diseases. RCMD characteristics and their relations with dust toxicity need further research to understand the adverse exposure effects to RCMD. The geographic clustering of coal workers' pneumoconiosis (CWP) suggests that RCMD in the Appalachian region may exhibit more toxicity than other geographic regions such as the Rocky Mountains. This study investigates the RCMD characteristics and toxicity based on geographic location. Dissolution experiments in simulated lung fluids (SLFs) and in vitro responses were conducted to determine the toxicity level of samples collected from five mines in the Rocky Mountains and Appalachian regions. Dust characteristics were investigated using Fourier-transform infrared spectroscopy, scanning electron microscopy, the BET method, total microwave digestion, X-ray diffraction, and X-ray photoelectron spectroscopy. Inductively coupled plasma mass spectrometry was conducted to determine the concentration of metals dissolved in the SLFs. Finer particle sizes and higher mineral and elemental contents were found in samples from the Appalachian regions. Si, Al, Fe, Cu, Sr, and Pb were found in dissolution experiments, but no trends were found indicating higher dissolutions in the Appalachian region. In vitro studies indicated a proinflammatory response in epithelial and macrophage cells, suggesting their possible participation in pneumoconiosis and lung diseases development.

Abstract:The coal sample taken from Yinchuan,coal-fired PM2.5 was sampled by fixed source dilution channel and PM2.5 suspension was extracted by ultrasonic water-bath method.Human umbilical vein endothelial cells were treated with PM2.5 suspension at various concentrations and exposed for different times(6h,12h,and 24h).The MTS assay was used to measure the effect of PM2.5 on cell proliferation of EA.hy926.Coal-fired PM2.5 suspension decreased the viability of vascular endothelial cells at different times and exposure time groups of 12h and 24h showed significance statistically in comparison to the solvent control.Compared with the same concentration group of 12h and 6h,exposure time groups of 24h had statistically significant differences.Coal-fired PM2.5 could significantly decrease the viability of hman umbilical vein endothelial cells in dose-dependent and time-dependent manner.Vascular endothelial injury might be one of the possible mechanisms of cardiovascular toxicity caused by coal-fired PM2.5.

Abstract:The present study aims to investigate the hydrogeochemical evolution of heavy metals and assesses impacts of mining activities on the groundwater resources and potential human health risks in the coal mining areas of Ib valley coalfield. In this perspective, a total of one hundred and two mine water and groundwater samples were collected from different locations. The water samples were analysed for some selected heavy metals i.e. Mn, Cu, Pb, Zn, Ni, Co, As, Se, Al, Sr, Ba, Cd, Cr, V and Fe using ICP-MS. In addition, pH and SO 4 2− concentration were also measured following APHA procedure. The water pH in the Ib valley coalfields ranged from 3.26 to 8.18 for mine water and 5.23 to 8.52 for groundwater, indicating acidic to alkaline nature of water. Mn in mine water and Zn in groundwater environment were observed as the most dominant metals. The water hazard index (WHI) reflects that around 80% of mine water are non-toxic (WHI<5), 5% slightly toxic (5 10) and 15% extremely toxic (WHI>15). Relatively high pH and low concentration of dissolved metals and SO 4 2− in groundwater as compared to mine water indicate lesser impact of mining activities. The calculated drinking water quality index (DWQI) suggests that Mn, Al, Ni and Fe in mine water and Mn, Fe, Ni and Pb in groundwater were the major objectionable metals which caused the water quality deterioration for drinking uses. Further, the non-carcinogenic health risk assessment for adult male, female and child populations identifies Co, Mn, Ni as the key elements making the water hazardous for human health. Comparatively higher ratio of ingestion rate and body weight in child population might be causing higher health risks in child population as compared to adult male and adult female population.

Abstract:Indoor air pollution from the combustion of biomass fuel and associated health risks is a critical issue in developing countries. Concentrations of PM2.5 and PM10 are measured in Birbhum, West Bengal, during 2017–2018. PM2.5-bound elemental concentrations of twelve metals are determined in rural kitchens. The results showed higher toxicological risks in BMF (1.15) than the LPG users (0.14). The risk of non-carcinogenic exposure related with dermal contact and ingestion was observed in the acceptable limits (HQ < 1) for all age groups, and the risk associated with inhalation exposure from Cr, Ni, As, and Mn exceeded the acceptable limit. Results also suggest that carcinogenic risks from ingestion and dermal contact are within the acceptable limit (1 × 10−4–1 × 10−6) except Cr and As which were found to exceed the range. The deposition flux (Dφ) for multiple metals in the head airway region, tracheobronchial region, and alveolar regions was found to be higher in teenagers as compared to other groups, whereas the value was lower in infants. Further, it was notified from the Dφ that the metals could pass through the head airways and harm the tracheobronchial tree and alveolar region, increasing the risk of human health.

Abstract:Airborne particulates in iron ore mining are a risk factor for adverse human lung effects. In this study, fine particulates deposited on surfaces of about 1.5 m above the ground and 6 meters from a milling unit of the Gol-e-Gohar iron ore mine were collected through wipe sampling. Dust particles less than 5 μm in diameter were separated with an electronic sieve. Aliquots were prepared from the sieved iron ore dust estimated to be equivalent to respiratory exposure in the iron ore mill in the concentrations of 1, 5, 10, 50, 100, and 250 μg/mL, which were intended to represent equivalent inhaled doses from working one month to a working life (25 years) in the mine. The airborne concentration of respirable particles was about five times the threshold limit value given (TLV ® ) for iron oxide published by the American Conference of Governmental Industrial Hygienists. The in vitro toxicity range was estimated to be equivalent to an accumulated dose associated with working from one month to a working life in the mine. Treatment of the A549 cells resulted in decreased dehydrogenase activity and cell glutathione content and increased reactive oxygen species (ROS) generation, mitochondrial membrane permeability, and cell apoptosis-necrosis rates. The results of this study revealed the possibility of lung damage at cell doses for respirable airborne iron oxide particles estimated to be equivalent to accumulated lifetime exposures among Gol-e-Gohar miners. Further studies are recommended to investigate the effect of actual contaminants in the workplace on the occurrence of health effects on workers.

Abstract:Background: The objective of this study is to study the adverse effects of coal mining environment on workers to discover early effective biomarkers. Methods: The molecular epidemiological study was conducted with 502 in-service workers, who were divided into miner and auxiliary. We measured the individual levels of dust exposure for participants. Clinical examinations were conducted by qualified doctors. Peripheral blood was collected to measure biochemistry, hemogram, and karyocyte apoptosis. Results: All workers were healthy who have not found with any diseases that can be diagnosed medically in the physical examination and showed no difference in dust exposure level, age, height, weight, and body mass index between groups. The working years of miners were lower than that of auxiliaries ( p < 0.001). Compared with auxiliaries, the concentration and percentage of lymphocytes ( p = 0.040, p = 0.012), basophils ( p = 0.027, p = 0.034), and red blood cells ( p < 0.001) and the concentration of hemoglobin of miners were lower ( p < 0.001). The percentage of neutrophils ( p = 0.003), the concentration of mean corpuscular hemoglobin concentration ( p = 0.002), and the proportion of karyocyte apoptosis in miners were higher ( p < 0.001). Miners presented higher blood urea nitrogen ( p < 0.001), ratio of blood urea nitrogen to creatinine ( p < 0.001), the high density lipoprotein cholesterol ( p < 0.001), lower creatinine ( p < 0.05), and cholesterol ( p < 0.001). Conclusion: The coal mining environment impacted mining workers' immune function, renal function, and the hematopoietic system, including BUN/CRE, HGB, RBC, and LYMPH, which could be used as early biomarkers to screen the health of coal miners.

Geochemical and mineralogical assessment of environmentally sensitive elements in Neyveli lignite deposits, Cauvery Basin, India

Abstract:This research work presents an examination of the concentrations and modes of occurrence of environmentally sensitive elements within lignite deposits, located in Neyveli, within the Cauvery Basin of India. Coal is one of the most complex geologically formed materials, consisting of organic and inorganic matter. The inorganic mineral matter including the crystalline minerals, non-crystalline mineraloids, and elements with non-mineral associations. These lignite samples underwent complete analysis encompassing macroscopic, microscopic and geochemical assessments. The analysis reveals that the total mineral matter (MM) content, comprising significant proportions of sulphides, carbonate and argillaceous components. Geochemical characterization further elucidates the lignite's properties, with proximate analysis yielding values such as ash, volatile matter and fixed carbon and the Ultimate components analysis reveals the carbon, hydrogen, nitrogen, sulphur and oxygen. Inorganic mineral matters play a significant role in coal utilization, and also such modes of occurrence of elements provide useful geochemical information on coal formation and coal-bearing basin evolution. In this paper, we assess the associations of elements and minerals, as well as the associations of selected elements including environmentally-sensitive (e.g., S, As, U, and Hg), and some major elements (e.g., Ca, Mg, Fe, Al, and Ti) that have largely occurred in non-mineral forms in these low-rank coals. And also, comparative analysis is conducted between the concentrations of elements within the lignite samples and the values reported for World Clarke Brown Coals (WCBC). Particularly, some of these elements exhibit significantly high environmental sensitivity, demanding careful consideration in lignite extraction and utilization practices.
Chemical Speciation, Bioavailability and Human Health Risk Assessment of Metals in Surface Dust from an Industrial Cluster in India

Abstract:In this study, distribution of metals in different geochemical forms, their mobility and bioavailability in bulk surface dust samples of Bhiwadi industrial cluster (BIC) in Rajasthan, India, was assessed by modified Community Bureau of Reference (m-BCR) sequential extraction procedure. Potential risk of metals in surface dust to environment and human health was evaluated using Contamination factor (Cf), Mobility Factor (MF) and Risk Assessment Code (RAC), and carcinogenic and non-carcinogenic health risk. Residual fraction contained significant amount of metals as Cd(55.86%), Cr(86.05%), Fe(90.06%), Mn(69.94%), Ni(66.08%), and V(71.80%). Pb(52.43%) was present in reducible fraction, while Cu was equally distributed in reducible (27.66%) and oxidizable (28.20%) fractions. Zn was equally distributed in acid exchangeable (33.15%) and reducible (35.01%) fractions. High Cf values were observed for Zn (1.32-16.98), followed by Pb (0.38-11.23) and Cu (0.26-8.22). RAC indicated high risk of Cd, Mn, Ni and Zn to environment due to their high mobility and toxic nature. Zn, Pb, Cu and Cd showed highest mobility (potential bioavailability) in samples collected around metal casting, electroplating, and automobile part industries. Data indicated that metals can bio-available with the changes in redox conditions in environment. Ingestion was major pathway for carcinogenic and non-carcinogenic health risks followed by dermal and inhalation. Hazardous Index value (6.32) indicated higher susceptibility of children for non-carcinogenic risk as compared to adults. Carcinogenic risk of Cr, Cd, Ni and Pb was higher than acceptable levels in surface dust, suggesting a high risk of cancer to exposed population.

Abstract:A study to monitor and estimate respirable particulate matter (RPM), toxic trace metal concentrations in the work environment was carried out in different sections of an integrated steel manufacturing industry. The average RPM concentration observed varied according to the section blast furnace was 2.41 mg/m ; energy optimization furnace, 1.87 mg/m ; sintering plant, 0.98 mg/m ; continuous casting machine, 1.93 mg/m . The average trace metal concentration estimated from the RPM samples like iron, manganese, lead and chromium did not exceed ACGIH prescribed levels.

In Vitro Toxicological Assessment of Cobalt Ferrite Nanoparticles in Several Mammalian Cell Types

Abstract:Nanoparticles have been widely used in various fields due to the superior physicochemical properties and functions. As a result, human exposure to nanoparticles increases dramatically. Previous researches have shown that nanoparticles could travel through the respiratory, digestive system, or skin into the blood and then to the secondary organs such as the brain, heart, and liver. Besides, the nanoparticle toxicity is controversial and dependent on the sensitivity of the cell type, route of exposure, and condition, as well as their characteristics. Similarly, cobalt ferrite nanoparticles (CoFe2O4-NPs) have been used in different industrial fields, and have also various application possibilities in medical and biomedical fields. CoFe2O4-NPs induce toxic responses in various organisms such as human, mice, and algae. However, there is a serious deficit of information concerning their effects on human health and the environment. We aimed to investigate the toxic effects of CoFe2O4-NPs on liver (HepG2), colon (Caco-2), lung (A549), and neuron (SH-SY5Y) cells, which reflect different exposure routes in vitro, by using various toxicological endpoints. The cytotoxicity, genotoxicity, oxidative damage, and apoptosis induction of CoFe2O4-NPs (39 ± 17 nm) were evaluated. After 24 h, the nanoparticles decreased cell viability at ≤100 μg/mL, while increasing viability at >100 μg/mL. CoFe2O4-NPs induced DNA and oxidative damage with increased malondialdehyde (MDA) and 8-hydroxy deoxyguanosine (8-OHdG) levels and decreased glutathione (GSH) levels with no change in protein carbonyl (PC) levels. CoFe2O4-NPs had apoptotic effect in HepG2 and Caco-2 cells in a concentration-dependent manner and necrotic effects on SH-SY5Y and A549 cells. Consequently, the adverse effects of CoFe2O4-NPs should raise concern about their safety in consumer products.
The Cellular Toxicity of PM2.5 Emitted from Coal Combustion in Human Umbilical Vein Endothelial Cells.

Abstract:OBJECTIVE:To explore the relationship between different components of fine particulate matter (PM2.5) emitted from coal combustion and their cytotoxic effect in the vascular endothelial cells.METHODS:Coal-fired PM(2.5) was sampled using a fixed-source dilution channel and flow sampler. The sample components were analyzed by ion chromatography and inductively coupled plasma atomic emission spectroscopy (ICP-AES). The PM(2.5) suspension was extracted using an ultrasonic water-bath method and then human umbilical vein endothelial cells (EA.hy926) were treated with various concentrations of the PM(2.5) suspension. Cell proliferation, oxidative DNA damage, and global DNA methylation levels were used to measure the cellular toxicity of PM(2.5) emitted from coal combustion.RESULTS:Compared to other types of coal-fired PM(2.5) preparations, the PM2.5 suspension from Yinchuan coal had the highest cytotoxicity. PM(2.5) suspension from Datong coal had the highest toxic effect while that from Yinchuan coal had the lowest. Exposure to coal-fired PM(2.5) from Jingxi coal resulted in lower 8-hydroxy-2'-deoxyguanosine (8-OHdG) levels. At the same dose, PM(2.5) emitted from coal combustion could produce more severe DNA impairment compared to that produced by carbon black. Cell survival rate was negatively correlated with chloride and potassium ions content. The 5-methylcytosine (5-mC) level was positively correlated with Mn and negatively correlated with Zn levels. The 8 OHdG% level was positively correlated with both Mn and Fe.CONCLUSION:PM(2.5) emitted from coal combustion can decrease cell viability, increase global DNA methylation, and cause oxidative DNA damage in EA.hy926 cells. Metal components may be important factors that influence cellular toxicity.
