Abstract:Cancer is one of the most fatal diseases for decades. Aggregation‐induced emission luminogens (AIEgens) have been recently used as molecular imaging or therapeutic agents in cancers, due to the advantages of large Stokes shift, high quantum yield, great biocompatibility, and strong photostability. AIEgens can specifically target different types of cancer via diverse targeting strategies. AIEgen‐based fluorescence imaging, especially near‐infrared imaging, demonstrated deep penetration and suitable signal‐to‐noise ratio, which allows reliable in vivo cancer imaging. Combined with other imaging modalities, AIEgen‐based multimodal imaging could provide multidimensional cancer hallmarks from different perspectives. In addition, AIEgen‐based phototherapy can be used for photodynamic therapy and photothermal therapy, which facilitate ablation of cancer cells with good biosafety and high therapeutic effects in vivo. AIEgens nanoparticles fabricated with some specific chemicals, drugs, or siRNA, could display synergistic therapeutic effects for cancers. This paper comprehensively describes the current status and future perspectives of AIEgens, which have showed a great potential for the future preclinical and clinical translation on in vivo molecular imaging and theranostics in cancer.

Abstract:Optical molecular imaging and image-guided theranostics benefit from special and specific imaging agents, for which aggregation-induced emission luminogens (AIEgens) have been regarded as good candidates in many biomedical applications. They display a large Stokes shift, high quantum yield, good biocompatibility, and resistance to photobleaching. Neurological diseases are becoming a substantial burden on individuals and society that affect over 50 million people worldwide. It is urgently needed to explore in more detail the brain structure and function, learn more about pathological processes of neurological diseases, and develop more efficient approaches for theranostics. Many AIEgens have been successfully designed, synthesized, and further applied for molecular imaging and image-guided theranostics in neurological diseases such as cerebrovascular disease, neurodegenerative disease, and brain tumor, which help us understand more about the pathophysiological state of brain through noninvasive optical imaging approaches. Herein, we focus on representative AIEgens investigated on brain vasculature imaging and theranostics in neurological diseases including cerebrovascular disease, neurodegenerative disease, and brain tumor. Considering different imaging modalities and various therapeutic functions, AIEgens have great potential to broaden neurological research and meet urgent needs in clinical practice. It will be inspiring to develop more practical and versatile AIEgens as molecular imaging agents for preclinical and clinical use on neurological diseases.

Abstract:Biophotonics has seen significant advancements with the development of optical imaging techniques facilitating the noninvasive detection of biologically relevant species. Aggregation-induced emission (AIE) materials have emerged as a novel class of luminogens exhibiting enhanced luminescence or photodynamic efficiency in the aggregated state, making them ideal for biomedical applications. The intracellularly controlled aggregation of aggregate-induced emission luminogens (AIEgens) enables high-resolution imaging of intracellular targets and diagnosis of related diseases, and disease therapy is allowed by exploiting the novel properties of aggregates. This review provides an in-depth analysis of the strategies employed to modulate the aggregation of AIEgens, focusing on the importance of molecular modifications to improve hydrophilicity and achieve precise control over the intercellular aggregation of AIEgens. Furthermore, the representative applications of AIEgens in bioimaging, such as enzyme activity monitoring, protein tracking, organelle function monitoring, and in vivo tumor-specific therapeutics, are reviewed. Additionally, we outline the challenges and future opportunities for AIE research, emphasizing the importance of the strategies for realizing the precisely controllable aggregation of AIEgens inside cells and the need for extending AIEgens' absorption and emission wavelengths. This review aims to elucidate the rational development of multi-modality AIEgens for advanced biomedical applications.

Abstract:High-quality fluorescence bioimaging requires organic exogenous contrast agents with biocompatibility, brightness, and photostability. The recently discovered and rapidly developed aggregation-induced emission luminogens (AIEgens) are excellent candidates because they exhibit enhanced emission upon the restriction of intramolecular motions (RIM) effect. AIEgens are individually lit up once bound with biomolecules or influenced by the surrounding bio-environment, and abundant biological information can be acquired on the basis of high-sensitivity fluorescence imaging. In addition, AIEgen-incorporated nanoparticles possess bright fluorescence and improved photostability, which is beneficial to long-term bioimaging with high contrast and spatial resolution. In this review, we summarize the latest advances in AIEgen-based fluorescence bioimaging, as well as the relevant applications in theranostics.

Abstract:Fluorescent biomedical materials can visualize subcellular structures and therapy processes in vivo. The aggregation-induced emission (AIE) phenomenon helps suppress the quenching effect in the aggregated state suffered by conventional fluorescent materials, thereby contributing to design strategies for fluorescent biomedical materials. Photoresponsive biomedical materials have attracted attention because of the inherent advantages of light; i.e., remote control, high spatial and temporal resolution, and environmentally friendly characteristics, and their combination with AIE facilitates development of fluorescent molecules with efficient photochemical reactions upon light irradiation. In this review, organic compounds with AIE features for biomedical applications and design strategies for photoresponsive AIE luminogens (AIEgens) are first summarized briefly. Applications are then reviewed, with the employment of photoresponsive and AIE-active molecules for photoactivation imaging, super-resolution imaging, light-induced drug delivery, photodynamic therapy with photochromic behavior, and bacterial targeting and killing being discussed at length. Finally, the future outlook for AIEgens is considered with the aim of stimulating innovative work for further development of this field.

Abstract:Luminogens with aggregation-induced emission (AIEgens) have been widely applied in the field of photodynamic therapy. Among them, aggregation-induced emission photosensitizers (AIE-PSs) are demonstrated with high capability in fluorescence and photoacoustic bimodal imaging, as well as in fluorescence imaging-guided photodynamic therapy. They not only improve diagnosis accuracy but also provide an efficient theranostic platform to accelerate preclinical translation as well. In this short review, we divide AIE-PSs into three categories. Through the analysis of such classification and construction methods, it will be helpful for scientists to further develop various types of AIE-PSs with superior performance.

Abstract:Considerable efforts have been made into developing aggregation-induced emission fluorogens (AIEgens)-containing nano-therapeutic systems due to the excellent properties of AIEgens. Compared to other fluorescent molecules, AIEgens have advantages including low background, high signal-to-noise ratio, good sensitivity, and resistance to photobleaching, in addition to being exempt from concentration quenching or aggregation-caused quenching effects. The present review outlines the major developments in the biomedical applications of AIEgens-containing systems. From a literature survey, the recent AIE works are reviewed and the reasons why AIEgens are chosen in various biomedical applications are highlighted. The research activities on AIEgens-containing systems are increasing rapidly, therefore, the present review is timely.

Abstract:Aggregation-induced emission (AIE) enables "Turn-On" imaging generally through single aggregation of the AIE luminogen (AIEgen). Dual aggregrations of the AIEgen might further enhance the imaging intensity and the consequent sensitivity. Herein, we rationally designed a near-infrared (NIR) AIEgen Ac-Trp-Glu-His-Asp-Cys(StBu)-Pra(QMT)-CBT (QMT-CBT) which, upon caspase1 (Cas1) activation, underwent a CBT-Cys click reaction to form cyclic dimers QMT-Dimer (the first aggregation) and assembled into nanoparticles (the second aggregation), turning the AIE signal "on" for enhanced imaging of Alzheimer's disease (AD). Molecular dynamics simulations validated that the fluorogen QMT in QMT-NPs stacked much tighter with each other than in the single aggregates of the control compound Ac-Trp-Glu-His-Asp-Cys(tBu)-Pra(QMT)-CBT (QMT-CBT-Ctrl). Dual aggregations of QMT rendered 1.9-, 1.7-, and 1.4-fold enhanced fluorescence intensities of its single aggregation in vitro, in cells, and in a living AD mouse model, respectively. We anticipate this smart fluorogen to be used for sensitive diagnosis of AD in the clinic in the near future.

Abstract:As a role for oxygen free radicals has been suggested in gentamicin (G) nephrotoxicity, we tested the hypothesis that exogenously administered glutathione (GSH), able to restore intracellular antioxidant potential, could be useful in reducing damage. Adult Sprague-Dawley rats were injected with saline (n = 30), subcutaneous (s.c.) G 100 (n = 23) and 150 mg/kg/day (n = 14), or s.c. G at the same dosages plus intraperitoneal (i.p.) GSH 1200 mg/kg/day (n = 24 and 14, respectively) for 7 days. In the G-100-day protocol, GSH-treated rats showed significantly lower renal G content (2.79 +/- 0.8 vs. 3.61 +/- 1.4 micrograms/mg prot) coupled with lower plasma urea (153 +/- 79 vs. 188 +/- 61 mg/dL) and creatinine levels (1.63 +/- 1 vs. 2.45 +/- 1 mg/dL). As to renal oxidant/antioxidant balance, local GSH was increased (0.32 +/- 0.01 vs. 0.19 +/- 0.01 microgram/mg prot) while lipid peroxidation, determined by production of thiobarbituric acid reactive substances (TBARS), was decreased (0.35 +/- 0.02 vs. 0.52 +/- 0.02 nmol/mg prot). In the G-150-mg protocol, GSH-treated rats showed no differences in renal gentamicin content or in blood urea and creatinine levels, in spite of a significantly lower renal TBARS production and a significantly higher GSH content. Urine enzyme excretion did not significantly change in GSH-treated vs. not-GSH-treated rats in both protocols. We conclude that: (a) GSH interferes with G nephrotoxicity mainly via a reduction in G uptake; (b) the oxidative renal stress is not crucial in inducing renal damage. In fact, when increased G dosages blunt the ability of GSH in reducing G uptake, no substantial protection is demonstrated.

This review mainly overviews the current state-of-art advances of AIE fluorophores achieving the superb performance in brain imaging and therapy, which facilitate deep tissue penetration, high contrast to autofluorescence and efficient blood-brain barrier (BBB) crossing by rational molecular design and functionalized strategies. Abstract:Photoacoustic imaging (PAI) is a rapidly emerging modality in biomedical research with the advantages of noncontact operation, high optical resolution, and deep penetration. Great efforts and progress in the development of PAI agents with improved imaging resolution and sensitivity have been made over the past 2 decades. Among them, organic agents are the most promising candidates for preclinical/clinical applications due to their outstanding in vivo properties and facile biofunctionalities. Motivated by the unique properties of aggregation-induced emission (AIE) luminogens (AIEgens), various optical probes have been developed for bioanalyte detection, multimodal bioimaging, photodynamic/photothermal therapy, and imaging-guided therapeutics. In particular, AIE-active contrast agents have been demonstrated in PAI applications with excellent performance in imaging resolution and tissue permeability in vivo. This paper presents a brief overview of recent progress in AIE-based agents in the field of photoacoustic imaging. In particular, we focus on the basic concepts, data sorting and comparison, developing trends, and perspectives of photoacoustic imaging. Through numerous typical examples, the way each system realizes the desired photoacoustic performance in various biomedical applications is clearly illustrated. We believe that AIE-based PAI agents would be promising multifunctional theranostic platforms in clinical fields and will facilitate significant advancements in this research topic.

Abstract:Surgery is still one of the most important means of treating tumors. Fluorescence imaging-mediated surgical navigation and intraoperative phototherapy are considered to be effective ways to improve surgical outcomes. However, the lack of ideal imaging/phototherapy agents continues to be a barrier to their clinical application. Traditional imaging/phototherapy agents present some challenges in living organisms, such as aggregation caused quenching (ACQ), which cause a burst of fluorescent signal or loss of photosensitivity. The introduction of aggregation-induced emission (AIE) luminogens (AIEgens) promises to compensate for the challenges posed by ACQ fluorophores, in addition to their advantages such as high brightness/photostability/photosensitivity in aggregated states, large Stokes shift and excellent biocompatibility. This review focuses on some of the recently emerged AIE probes that are promising to assist the surgery, and systemically summarizes the strategies to solve the issues encountered during the applications of surgery navigation and intraoperative phototherapy. We emphasize the molecular design concept of the AIE probes, reveal their applications in intraoperative imaging and phototherapy, and comment on the existent insufficiency. This review aims to provide concepts for the design of AIE probes for addressing surgical navigation and intraoperative phototherapy, and to present the challenges faced in their clinical translation in order to enhance surgical outcomes in the future.

Abstract:Luminogens with aggregation-induced emission (AIEgens) characteristics have been well developed and applied in various areas such as bio-imaging, theranostics, organic photoelectronics and chemo/bio sensors. However, most of the reported AIEgens suffer from the disadvantages of complex organic synthesis and high cost, as well as being environmentally unfriendly and hard to degrade, which have largely limited their real applications. In this work, we discovered berberine chloride, a natural isoquinoline alkaloid isolated from Chinese herbal plants, as an unconventional rotor-free AIEgen with bright solid-state emission and water-soluble characteristics. Single crystal structure analysis and optical property, viscosity, and host-guest interaction studies suggested that intramolecular vibration and twisted intramolecular charge transfer were responsible for the AIE phenomenon of berberine chloride. Moreover, berberine chloride was biocompatible and could specifically target lipid droplets in a fluorescence turn-on and wash-free manner, demonstrating the great potential of natural products as promising AIE probes.

Abstract:Central nervous system (CNS) diseases include Alzheimer's disease (AD), Parkinson's disease (PD), brain tumors, strokes, and other important diseases that are harmful and fatal to human beings. CNS diseases have the characteristics of high fatality rates, difficult diagnosis, and costly treatment. The diagnosis and treatment of CNS diseases by molecular imaging are usually limited by the depth of tissue penetration and the blood-brain barrier (BBB). Therefore, it is still a huge challenge to distinguish between the lesion and the surrounding parenchymal boundary with high sensitivity and specificity. Compared with traditional fluorophores with aggregation-caused quenching effect, luminogens with aggregation-induced emission (AIE) characteristics have strong near-infrared deep penetration, large Stokes shift, excellent biocompatibility, light stability, and desirable BBB permeability. In view of this, developing novel AIE-based materials for diagnostics and theranostics of CNS diseases is promising and of great significance. Herein, we highlight the recent research progress in this field with a special focus on near-infrared imaging and AIE nanorobots for CNS diseases. The design principle of AIE probes is discussed in detail, and the outlook is presented as well.

Abstract:A new concept of aggregation‐enhanced theranostics (AET) is put forward on the basis of tremendous aggregation‐enhanced superiorities of AIEgens in terms of diagnostic imaging, therapeutic intervention, and feasibility in construction of multi‐modality one‐for‐all theranostics. This review highlights the tremendous aggregation‐enhanced superiorities of AIE luminogens (AIEgens) in disease theranostics mainly involving diagnostic imaging (fluorescence and room temperature phosphorescence), therapeutic intervention (photodynamic therapy), and feasibility in construction of multi‐modality theranostics based on the experimental measurements and theoretical simulations. Additionally, the latest and advanced developments of AIEgens in disease theranostics in the aspect of corresponding strategies to design highly effective AIE‐active theranostics through triggering aggregation formation are comprehensively summarized. Moreover, a brief conclusion with the discussion of current challenges and future perspectives in this area is further presented.

Abstract:The concept of aggregation-induced emission (AIE) in purely organic luminescent molecules has drawn wide attention in the last two decades. Despite the many challenges, AIE-probes have opened versatile opportunities in many research fields. In particular, the emerging functional properties of room temperature phosphorescence (RTP) and thermally activated delayed fluorescence (TADF) have boosted the unique features of AIE luminogens (AIEgens). Thus, these luminescent materials extended the utility in sensing, imaging, optoelectronics and theranostic applications in biological field over the conventional fluorescent probe. Unlike the sensitivity of triplet state by oxygen and moisture, the long-lived phosphorescence and delayed fluorescence resulting from the enhanced intersystem crossing (ISC) and reverse intersystem crossing (RISC) from excited triplet state (T1) to excited singlet state (S1) in these luminophores gives rise to long lifetimes ranging from nanoseconds to milliseconds even up to seconds. As compared to traditional fluorescence molecules advanced AIE probes show high contrast imaging and deeper penetration depth, which have been demonstrated through near infrared I and II (NIR-I & NIR-II) fluorescence imaging, room temperature after-glow imaging and photoacoustic imaging. This chapter highlights the recent developments and principle of the efficient design of AIE probe with multi-functional properties evolved with new strategies for translational applications via fluorescence imaging, photoacoustic imaging and image-guided photodynamic/photothermal therapy (PDT/PTT) including future opportunities for AIEgens to advance the overall biomedical field.

Abstract:The in situ visualization of drugs can improve the understanding of their pharmacokinetics and mechanisms. Aggregation-induced emission (AIE) materials, which can aid in the visualization of drugs, are gradually being employed in pharmaceutical research due to their excellent fluorescence properties, good biocompatibility, and extremely high sensitivity. Herein, the progress of AIE materials in pharmaceutical research, including AIE carriers for drug delivery, AIE multifunctional prodrugs, and AIE compounds as bioactive reagents for theranostics is briefly described. Moreover, the opportunities and challenges of AIE materials in pharmaceutical research are discussed in depth. It is believed that versatile AIE materials hold great promise for the promotion of pharmacological research and can facilitate significant advancements in clinical fields.
