Abstract:Projection-based 3D bioprinting(PBP):a powerful method to fabricate 3D cellular structures Three-dimensional(3D)bioprinting has played an impor-tant role in tissue engineering and regenerative medicine areas over the past decade[1].Different from traditional cell cultures in Petri dishes,3D bioprinting can build bionic structures with a better potential to become artificial organ substitutes[2-4].With the development of photocurable bio-materials,the projection-based 3D printing method has been successfully applied in biological research[5,6].It pos-sesses significant advantages in precision and speed due to its unique manufacturing principle.On the one hand,light-dependent solidification allows it to build models with resolutions around 50 μm(theoretically a minimum of 1 μm)[7].On the other hand,its surface-to-entity printing process provides duplicates in batches without extra time require-ment[8].Therefore,it has attracted the attention of many researches devoted to the field of 3D cell culture.

Abstract:Projection-based 3D printing is a vat polymerization printing method that works by generating bitmaps as dynamic masks to project onto a photosensitive material surface for layer-by-layer curing. Projection-based 3D printing has the highest resolution/time for manufacturing ratio among all 3D printing technologies; however, projection-based 3D bioprinting, which uses bioinks as printing materials that contain cells and/or biomolecules, suffers from low printing resolution, with a substantial gap between the theoretical and the actual resolution. In this Review, we summarize the steps and challenges to achieve high-resolution projection-based 3D bioprinting and provide pragmatic optimization strategies for tissue engineering and regenerative medicine applications. Projection-based 3D bioprinting has the highest resolution/time for manufacturing ratio among all 3D printing technologies. This Review discusses the main components of projection-based 3D bioprinting and summarizes the necessary steps and challenges to improve bioprinting resolution.

Abstract:Three-dimensional (3D) printing technology is expected to solve the organ shortage problem. However, owing to the accuracy limitations, it is difficult for the current bioprinting technology to achieve an accurate control of the spatial position and distribution of a single cell or single component droplet. In this study, to accurately achieve the directional deposition of different cells and biological materials in the spatial position for the construction of large transplantable tissues and organs, a high-precision multichannel 3D bioprinter with submicron-level motion accuracy is designed, and concurrent and synergistic printing methods are proposed. Based on the high-precision motion characteristics of the gantry structure and the requirements of concurrent and synergistic printing, a 3D bioprinting system with a set of 6 channels is designed to achieve six-in-one printing. Based on the Visual C++ environment, a control system software that integrates the programmable multi-axis controller (PMAC) motion, pneumatic, and temperature control subsystems was developed and designed. Finally, based on measurements and experiments, the 3D bioprinter and its control system was verified to fulfil the requirements of multichannel, concurrent, and synergistic printing with submicron-level motion accuracy, significantly shortening the printing time and improving the printing efficiency. This study not only provides an equipment basis for printing complex heterogeneous tissue structures, but also improves the flexibility and functionality of bioprinting, and ultimately makes the construction of complex multicellular tissues or organs possible.

Abstract:Three-dimensional（3 D） printing is a novel promising technology based on 3 D imaging and layer-by-layer additive fabrication.It has a profound influence on all aspects of our lives and is playing an increasing important role in many areas including engineering, manufacturing, art, education and medicine. "3 D bioprinting" has been put forward with the technical progress in 3 D printing and might be a possible way to solve the serious problem of human organ shortage in tissue engineering and regenerative medicine. Many research groups flung them into this area and have already made some gratifying achievements.However, it is a long way to fabricate a live organ. Many elements lead to the limitation of 3 D bioprinting. This review introduces the background and development history of 3 D bioprinting, compares different approaches of 3 D bioprinting and illustrates the key factors of the printing process. Meanwhile, this review also points out existing challenges of 3 D bioprinting and has a great prospect. Some points proposed in this review might be served as reference for the research of this field.

Abstract:Three-dimensional (3D) bioprinting is a rapidly growing technology that has been widely used in tissue engineering, disease studies, and drug screening. It provides the unprecedented capacity of depositing various types of biomaterials, cells, and biomolecules in a layer-by-layer fashion, with precisely controlled spatial distribution. This technology is expected to address the organ-shortage issue in the future. In this review, we first introduce three categories of 3D bioprinting strategies: inkjet-based printing (IBP), extrusion-based printing (EBP), and light-based printing (LBP). Biomaterials and cells, which are normally referred to as “bioinks,” are then discussed. We also systematically describe the recent advancements of 3D bioprinting in fabricating cell-laden artificial tissues and organs with solid or hollow structures, including cartilage, bone, skin, muscle, vascular network, and so on. The development of organs-on-chips utilizing 3D bioprinting technology for drug discovery and toxicity testing is reviewed as well. Finally, the main challenges in current studies and an outlook of the future research of 3D bioprinting are discussed.

Abstract:3D bioprinting has the capability to create 3D cellular constructs with the desired shape using a layer-by-layer approach. Inkjet 3D bioprinting, as a key component of 3D bioprinting, relies on the deposition of cell-laden droplets to create native-like tissues/organs which are envisioned to be transplantable into human body for replacing damaged ones. Benefiting from its superiorities such as high printing resolution and deposition accuracy, inkjet 3D bioprinting has been widely applied to various areas, including, but not limited to, tissue engineering and drug screening in pharmaceutics. Even though inkjet 3D bioprinting has proved its feasibility and versatility in various fields, the current applications of inkjet 3D bioprinting are still limited by the printing technique and material selection. This review, which specifically focuses on inkjet 3D bioprinting, firstly summarizes the techniques, materials, and applications of inkjet 3D bioprinting in tissue engineering and drug screening, subsequently discusses the major challenges that inkjet 3D bioprinting is facing, and lastly summarizes potential solutions to those challenges.
Modeling the Printability of Photocuring and Strength Adjustable Hydrogel Bioink During Projection-Based 3D Bioprinting.

Abstract:As a projection based three-dimensional printing method, digital light processing bioprinting (DLPBP) has higher printing resolution and is suitable for constructing finer structures to mimic tissues when compared to extrusion based bioprinting. However, there is a lack of understanding about printing behavior during DLPBP. Herein, a photo crosslinking theory for ink was established and a specified amount of light absorber was added to control crosslinking depth. Then, a standardized methodology was established to quantitatively evaluate printing resolution using different parameters. Complex biostructures, such as the ear, hand, and heart, were precisely printed after understanding the mechanism. Additionally, the mechanical properties of printed samples were accurately adjusted by changing the hydrogel concentration, as well as the degree of substitution and photocrosslinking time. The tissue types printed were from ultra-soft tissues, such as liver (6-8 kPa) to soft tissue, such as the skin (0.3-0.4 MPa). A branching vessel with cells in a real tensile modulus was printed as a demonstration. After 1 week of culture, proliferation and function of human umbilical vein endothelial cells were characterized. Overall, we made it possible to print a mimic complex tissue with high precision, required physical properties and functionalized living cells.

Abstract:3D bioprinting is an emerging technology that uses computer printing technology and unique bio-ink components to create artificial organs and biomedical objects. It is an interdisciplinary development technology integrating biology, chemistry, materials science, life science and other disciplines, and the technology behind biological 3D printing has improved in its development over the past few decades. Precisely printed biomaterials could benefit both tissue engineering and regenerative medicine. 3D bioprinting technology makes it possible to print tissues and organs, providing favorable conditions for organ transplantation and medical experiments. This paper mainly reviews the development of bioprinting technology and related concepts. According to the morphological change and chemical composition, the commonly used bioprinting materials (bio-inks) and their characteristics are described. The main methods of bioprinting are summarized, including traditional inkjet 3D bioprinting, extrusion 3D bioprinting, laser-assisted 3D bioprinting, photocuring 3D bioprinting and emerging 4D bioprinting and in situ bioprinting technologies. Finally, the paper introduces the development trend of bioprinting in the transplantation of skin, blood vessels and complex organs, as well as the precision research of tumors. In addition, current research challenges and prospects for future bioprinted 3D printing materials are discussed.

Abstract:With the recent advances of additive manufacturing, three-dimensional (3D) bioprinting has enabled the printing of biocompatible materials, cells and supporting components into complex 3D functional living tissues to address the need for tissues and organs suitable for transplantation. As vascularization is generally identified as one of the main technological barriers for building 3D organs, the freeform printing of 3D biological vascular constructs is of great importance to prove the overall feasibility of 3D bioprinting techniques. In this study, vascular-like constructs are fabricated by both laser- and inkjet-based printing techniques using a liquid support-based approach, which utilizes a calcium chloride-based solution as a cross-linking agent and buoyancy-based support material. 3D constructs such as Y-shaped tubular constructs have been successfully fabricated from alginate-based and cell-laden bioinks with a satisfactory post-printing cell viability.

Abstract:Three-dimensional (3D) printing, also called additive manufacturing (AM) or rapid prototyping (RP), is a layer by layer manufacturing method and now has been widely used in many areas such as organ printing, aerospace and industrial design. Now 3D printed microfluidics attract more and more interests for its rapid printing in the lab. In this review, we focused on the advances of 3D printed microfluidic chips especially the use in the chemistry and biology (vascularization and organs on chips). Based on the brief review of different 3D printing methods, we discussed how to choose the suitable 3D printing methods to print the desired microfluidics. We predict that microfluidics will be evolved from 2D chips to 3D cubes, printed hydrogel-based microfluidics will be reported and widely used, sensors & actuators could be integrated in the microfluidics during printing, and rapid assembling chips with printed microfluidic modules will be popular in the near future.

Abstract:Biomanufacturing of tissues/organs in vitro is our big dream, driven by two needs: organ transplantation and accurate tissue models. Over the last decades, 3D bioprinting has been widely applied in the construction of many tissues/organs such as skins, vessels, hearts, etc., which can not only lay a foundation for the grand goal of organ replacement, but also be served as in vitro models committed to pharmacokinetics, drug screening and so on. As organs are so complicated, many bioprinting methods are exploited to figure out the challenges of different applications. So the question is how to choose the suitable bioprinting method? Herein, we systematically review the evolution, process and classification of 3D bioprinting with an emphasis on the fundamental printing principles and commercialized bioprinters. We summarize and classify extrusion-based, droplet-based, and photocuring-based bioprinting methods and give some advices for applications. Among them, coaxial and multi-material bioprinting are highlighted and basic principles of designing bioinks are also discussed.

Abstract:Three-dimensional (3D) bioprinting of soft large-scale tissues in vitro is still a big challenge due to two limitations, (i) the lack of an effective way to print fine nutrient delivery channels (NDCs) inside the cell-laden structures above the millimetre level; (ii) the need for a feasible strategy to vascularize NDCs. Here, a novel 3D bioprinting method is reported to directly print cell-laden structures with effectively vascularized NDCs. Bioinks with desired tissue cells and endothelial cells (ECs) are separately and simultaneously printed from the outside (mixed with GelMA) and inside (mixed with gelatin) of a coaxial nozzle. As a result, the printed large-scale tissue consists of sheath-core fibers. At the same time, when the core fibers are dissolved to generate channels, the ECs deposit and adhere to the channels automatically. With this method, 3D cell-laden, vascularized tissue constructs (⩾1 cm) with a long-term culture (⩾20 d) are firstly reported. Specifically, vascularized cancer tissue constructs and osteogenic tissue constructs were generated. Considering the above advantages, this advanced bioprinting strategy has significant potential for building large-scale vascularized tissue constructs for applications in tissue engineering, and possibly even in regenerative medicine and organ repair.

Abstract:Three-dimensional (3D) bioprinting technologies play significant roles in various facets of the medical field, such as bioengineering, tissue repair, scaffolds, biomedical devices, and drug. As a versatile manufacturing technology, 3D bioprinting is able to overcome the constraints of other conventional methods and shows potential for future advancements in the field of biology. Nevertheless, the existing 3D bioprinting technologies still grapple with significant challenges in materials, equipment, and applications. Therefore, it is essential to select appropriate bioprinting method in alignment with the required application. In this review, we aim to cover the development, classification, and application of 3D bioprinting, with a particular emphasis on the fundamental printing principles. Additionally, we discuss the potential of 3D bioprinting in terms of materialization, structuralization, and functionalization, highlighting its prospective applications. We firmly believe that 3D printing technology will witness widespread adoption in the future, as it has the potential to address the limitations associated with multi-size, multi-material, multi-cell, and high-precision bioprinting.

Abstract:With the advances of stem cell research, development of intelligent biomaterials and three-dimensional biofabrication strategies, highly mimicked tissue or organs can be engineered. Among all the biofabrication approaches, bioprinting based on inkjet printing technology has the promises to deliver and create biomimicked tissue with high throughput, digital control, and the capacity of single cell manipulation. Therefore, this enabling technology has great potential in regenerative medicine and translational applications. The most current advances in organ and tissue bioprinting based on the thermal inkjet printing technology are described in this review, including vasculature, muscle, cartilage, and bone. In addition, the benign side effect of bioprinting to the printed mammalian cells can be utilized for gene or drug delivery, which can be achieved conveniently during precise cell placement for tissue construction. With layer-by-layer assembly, three-dimensional tissues with complex structures can be printed using converted medical images. Therefore, bioprinting based on thermal inkjet is so far the most optimal solution to engineer vascular system to the thick and complex tissues. Collectively, bioprinting has great potential and broad applications in tissue engineering and regenerative medicine. The future advances of bioprinting include the integration of different printing mechanisms to engineer biphasic or triphasic tissues with optimized scaffolds and further understanding of stem cell biology.

Abstract:A major challenge in 3D extrusion bioprinting is the limited number of bioink that fulfills the opposing requirements for printability with requisite rheological properties and for functionality with desirable microenvironment. Here, this limitation is addressed by developing a generalizable strategy for formulating a cell-laden microgel-based biphasic (MB) bioink. The MB bioink comprises two components, that is, microgels in close-packed condition providing excellent rheological properties for extrusion bioprinting, and a hydrogel precursor that forms a second polymer network to integrate the microgels together, providing post-printing structural stability. This strategy enables the effective printing of a range of hydrogels into complex structures with high shape fidelity. The MB bioink offers great mechanical tunability without compromising printability, and hyperelasticity with superb cyclic compression and stretch endurance. Moreover, the microgels and hydrogel precursor of the MB bioink can encapsulate different types of cells, together creating a heterogeneous cellular microenvironment at microscale. It is successfully demonstrated that hepatocytes and endothelial cells with spatial cell patterning by using MB bioink induce the cellular reorganization and vascularization, leading to enhanced hepatic functions. The proposed MB bioink expands the palette of available bioinks and opens numerous opportunities for the biomedical applications such as tissue engineering and soft robotics.

Abstract:Three-dimensional (3D) bioprinting technology has emerged as a powerful biofabrication platform for tissue engineering because of its ability to engineer living cells and biomaterial-based 3D objects. Over the last few decades, droplet-based, extrusion-based, and laser-assisted bioprinters have been developed to fulfill certain requirements in terms of resolution, cell viability, cell density, etc. Simultaneously, various bio-inks based on natural–synthetic biomaterials have been developed and applied for successful tissue regeneration. To engineer more realistic artificial tissues/organs, mixtures of bio-inks with various recipes have also been developed. Taken together, this review describes the fundamental characteristics of the existing bioprinters and bio-inks that have been currently developed, followed by their advantages and disadvantages. Finally, various tissue engineering applications using 3D bioprinting are briefly introduced.

Abstract:3D bioprinting is a new 3D manufacturing technology, that can be used to accurately distribute and load microorganisms to form microbial active materials with multiple complex functions. Based on the 3D printing of human cells in tissue engineering, 3D bioprinting technology has been developed. Although 3D bioprinting technology is still immature, it shows great potential in the environmental field. Due to the precise programming control and multi-printing pathway, 3D bioprinting technology provides a high-throughput method based on micron-level patterning for a wide range of environmental microbiological engineering applications, which makes it an on-demand, multi-functional manufacturing technology. To date, 3D bioprinting technology has been employed in microbial fuel cells, biofilm material preparation, microbial catalysts and 4D bioprinting with time dimension functions. Nevertheless, current 3D bioprinting technology faces technical challenges in improving the mechanical properties of materials, developing specific bioinks to adapt to different strains, and exploring 4D bioprinting for intelligent applications. Hence, this review systematically analyzes the basic technical principles of 3D bioprinting, bioinks materials and their applications in the environmental field, and proposes the challenges and future prospects of 3D bioprinting in the environmental field. Combined with the current development of microbial enhancement technology in the environmental field, 3D bioprinting will be developed into an enabling platform for multifunctional microorganisms and facilitate greater control of in situ directional reactions.

Abstract:Over the last decades, the fabrication of 3D tissues has become commonplace in tissue engineering and regenerative medicine. However, conventional 3D biofabrication techniques such as scaffolding, microengineering, and fiber and cell sheet engineering are limited in their capacity to fabricate complex tissue constructs with the required precision and controllability that is needed to replicate biologically relevant tissues. To this end, 3D bioprinting offers great versatility to fabricate biomimetic, volumetric tissues that are structurally and functionally relevant. It enables precise control of the composition, spatial distribution, and architecture of resulting constructs facilitating the recapitulation of the delicate shapes and structures of targeted organs and tissues. This Review systematically covers the history of bioprinting and the most recent advances in instrumentation and methods. It then focuses on the requirements for bioinks and cells to achieve optimal fabrication of biomimetic constructs. Next, emerging evolutions and future directions of bioprinting are discussed, such as freeform, high-resolution, multimaterial, and 4D bioprinting. Finally, the translational potential of bioprinting and bioprinted tissues of various categories are presented and the Review is concluded by exemplifying commercially available bioprinting platforms.

Abstract:Three-dimensional bioprinting has emerged as one of the manufacturing approaches that could potentially fabricate vascularized channels, which is helpful to culture tissues in vitro. In this paper, we report a novel approach to fabricate 3D perfusable channels by using the combination of extrusion and inkjet techniques in an integrated manufacture process. To achieve this, firstly we investigate the theoretical model to analyze influencing factors of structural dimensions of the printed parts like the printing speed, pressure, dispensing time, and voltage. In the experiment, photocurable hydrogel was printed to form a self-supporting structure with internal channel grooves. When the desired height of hydrogel was reached, the dual print-head was switched to the piezoelectric nozzle immediately, and the sacrificial material was printed by the changed nozzle on the printed hydrogel layer. Then, the extrusion nozzle was switched to print the next hydrogel layer. Once the printing of the internal construct was finished, hydrogel was extruded to wrap the entire structure, and the construct was immersed in a CaCl2 solution to crosslink. After that, the channel was formed by removing the sacrificial material. This approach can potentially provide a strategy for fabricating 3D vascularized channels and advance the development of culturing thick tissues in vitro.

Abstract:Three-dimensional (3D) bioprinting has emerged as a class of promising techniques in biomedical research for a wide range of related applications. Specifically, stereolithography apparatus (SLA) and digital light processing (DLP)-based vat-polymerization techniques are highly effective methods of bioprinting, which can be used to produce high-resolution and architecturally sophisticated structures. Our review aims to provide an overview of SLA- and DLP-based 3D bioprinting strategies, starting from factors that affect these bioprinting processes. In addition, we summarize the advances in bioinks used in SLA and DLP, including naturally derived and synthetic bioinks. Finally, the biomedical applications of both SLA- and DLP-based bioprinting are discussed, primarily centered on regenerative medicine and tissue modeling engineering.
