Abstract:Hair analysis plays an important role in forensic toxicology and biomonitoring tests. However, cosmetic treatments cause changes to the hair. Thus, a better understanding of the hair's structure and the factors that influence its composition is critical. It is known that oxidative treatments modify the hair chemical, structural and mechanical properties. These treatments also cause degradation of the melanin as well as of the structures present in the hair cuticle and cortex. Considering that the literature is unanimous regarding the increase in hydrophilicity and porosity promoted in human hair by bleaching, in this work we investigated how this oxidative damage is triggered. By combining several techniques, inelastic and quasi-elastic neutron scattering, differential scanning calorimetry, thermal gravimetric analysis and X-ray diffraction, we were able to connect the chemical and structural changes to a subtle dynamic modification of the proton mobility in the hair fibers. In addition, alterations in the thermal behavior evidenced a small denaturation of <i>α</i>-keratin intermediate filaments and a slight decrease in the amount of confined water in the hair fibers. Moreover, data obtained by neutron spectroscopy indicated that bleaching attacks the thioester groups of the proteins causing larger proton mobility of the hydrogenous components (water, protein and/or lipids).

Abstract:The use of a kinetic formalism, with a time‐dependent rate constant, allows for describing hair fibre swelling while absorbing moisture and provides hints about the entanglement of the keratin chains inside the fibre. Objective The rate process of moisture sorption by human hair has been analysed in order to hints for helping deepen knowledge on the hair structure and to explain the behaviour of hair in response to moisture. Methods The isotherms of moisture sorption by hair were recorded, via dynamic vapour sorption (DVS), for untreated (virgin) and treated (heat‐shaped and bleached) hair, as well as for separated cuticle and cortex. Results By considering that, during moisture uptake, the hair fibres also swell, it is possible to introduce a time‐dependent rate constant for describing the kinetics of the moisture sorption. This model allows for clearly separating the moisture sorption processes occurring in Cuticle and in Cortex and for proposing a role of chain entanglement in the two main compartments of the fibre. It may also provide some hints on the structural changes occurring in the fibre after different cosmetic treatments. The influence of the weight of the sample on the kinetics of the sorption process has also been noted and quantified. The analysis pointed to a transition occurring at around 30% relative humidity, assigned to the opening of the hair inner structure, and accommodation of more water molecules. This allowed for an estimate of the value of the activation energy of the water molecules reacting with the active sites, which was found to be in good agreement with results published in the literature. Conclusion The analysis of the kinetics of moisture sorption by hair was shown not only to provide information on the chain entanglement inside the fibre and the effect of cosmetic treatments but also to demonstrate and quantify the influence of fibre density on the sorption process. It is thus suggested that, along with examination of the hysteresis, the analysis of sorption kinetics helps reveal a more complete picture of hair moisture management. Résumé Objectif Le processus de taux de sorption de l'humidité par les cheveux humains a été analysé afin d'aider à approfondir les connaissances sur la structure des cheveux et expliquer le comportement des cheveux en réponse à l'humidité. Méthodes les isothermes de la sorption de l'humidité par les cheveux ont été enregistrés, via la sorption dynamique de la vapeur (DVS), pour les cheveux non traités (vierge) et traités (thermoformés et peroxydés), ainsi que pour les cuticules et le cortex séparés. Résultats en considérant que, lors de l'absorption de l'humidité, les fibres capillaires gonflent également, il est possible d'introduire une constante de taux linéaire pour décrire la cinétique de la sorption de l'humidité. Ce modèle permet de séparer clairement les processus de sorption d'humidité survenant dans les cuticules et dans le cortex et de proposer un rôle d'enchevêtrement des chaînes dans les deux principaux compartiments de la fibre. Il peut également fournir des indices sur les changements structurels survenant dans la fibre après différents traitements cosmétiques. L'influence du poids de l'échantillon sur la cinétique du processus de sorption a également été notée et quantifiée. L'analyse a indiqué une transition se produisant à environ 30 % d'humidité relative, attribuée à l'ouverture de la structure interne des cheveux, et à la présence d'un plus grand nombre de molécules d'eau. Cela a permis d'estimer la valeur de l'énergie d'activation des molécules d'eau réagissant avec les sites actifs, ce qui s'est avéré bien en accord avec les résultats publiés dans la littérature. Conclusion il a été démontré que l'analyse de la cinétique de la sorption de l'humidité par les cheveux fournit non seulement des informations sur l'enchevêtrement des chaînes à l'intérieur de la fibre et sur l'effet des traitements cosmétiques, mais démontre et quantifie également l'influence de la densité des fibres sur le processus de sorption. Il est donc indiqué que, parallèlement à l'examen de l'hystérésis, l'analyse de la cinétique de sorption permet d'obtenir un panorama de la gestion de l'humidité par les cheveux.

Abstract:Methods for examining cationic polymer deposition on hair are well known and polymers such as Polyquaternium-10 have enjoyed a significant commercial impact on shampoos and body washes as unique conditioning materials. It was recently reported that hair can be examined using a new microscopic called Digital Volumetric Imaging or DVI (10). By employing fluorescent dyes, deposition of cationic oligosaccharides onto damaged blond hair fibers was discussed. Because hair auto-fluorescences, the microscope allows for examination of hair fibers directly including viewing of the cuticle, cortex and melanin within the cortex and careful imaging even distinguishes the medulla of the hair fiber. In this paper, examination of six virgin hair types including: 1) Afro-American, 2) Asian, 3) European brown, 4) red, 5) blond and 6) gray was conducted looking for differences that each hair type brings to the visualizing technique. Digital manipulation of the fluorescent data allows for examination of interior hair fiber structures as well as the development of animated movies of three dimensional hair fiber structures.
Influence of internal structures of hair fiber on hair appearance. I. Light scattering from the porous structure of the medulla of human hair.

Abstract:In this study the influence of the medulla structure on hair appearance was examined. Hair with a porous medulla gave a whitish and lusterless appearance because of light scattering from the fiber center, whereas in the cases without pores, a clear and brilliant appearance was observed. The optical influences of the medulla pores were measured by a spectral goniophotometer, and obtained data were analyzed in terms of the CIE L*a*b* color system. Both contrasts in lightness and apparent color (chroma and hue) decreased in the hair with medulla pores, and the decreases in contrast caused a whitish and lusterless appearance. The distribution of the amount of medulla pores was investigated for Japanese females. The histogram was further analyzed by hair care behavior of individual panelists, and it was found that the pores in the medulla can be generated in a heat-drying process.

Abstract:For severe forms of psoriasis ciclosporin has now been proven to be a clinically effective therapeutic regimen. Beside inhibiting T-cell activation, ciclosporin exerts various effects on other cells of the immune system, e.g. Langerhans cells, mast cells and endothelial cells. The influence of ciclosporin on these cells may be relevant to an understanding of its beneficial effect in psoriasis treatment. New information about the molecular mechanism of action regarding calcineurin-regulated signal transduction pathways provide further insight into the pharmacological effects. On the basis of new findings concerning the mode of action of ciclosporin and the results of recent multicentre studies in psoriasis, new guidelines for therapy are given.

Abstract:BACKGROUND: The main objective of this study was to determine the lipid profile of brown and white Caucasian hair fibres and the effects of lipids on the properties of fibres.MATERIALS AND METHODS: To determine the structures of white and brown hair lipid bilayers, cross sections of fibres of both hair types were examined using synchrotron-based μ-FTIR mapping. Dynamic vapour sorption (DVS) analyses were also performed to determine the differences in the barrier function of both fibres.RESULTS: Spatial identification of lipids showed that a great amount of lipids was present in the medulla of fibres of both hair types, but important differences were also observed between cuticles of the different fibres. The cuticle of a white hair fibre showed a significant decrease in its lipid content, but did not show differences in the lateral packing order with respect to the cuticle of a brown hair fibre. The cortex and medulla of the white hair fibre also exhibited a significant decrease in its lipid content but with a higher lateral packing order than brown hair. Using DVS analysis, it was found that the water dynamics of white hair fibres differed from those of brown hair fibres, showing a decrease in their total capacity to absorb water and an increase in the velocity of the exchange of water with the environment.CONCLUSION: The results of both techniques demonstrated a high correlation between the characteristics of the lipids located in the cuticle and the water dynamics of the fibres.

Abstract:Modern society's keen regard for aesthetics made hair products an integral part of a multi‐billion‐dollar cosmetic industry. Hair care products (e.g., shampoos and conditioners) and chemical treatments (e.g., bleaching and permanent waving) result in various effects on the morphological attributes of hair. Generally, water adsorbed on the hair surface is known to significantly dictate the hair's mechanical characteristics (smoothness and friction), and hair's macroscopic wettability has been commonly used to indicate its surface properties. However, an approach to selectively characterize the hydration water in the hair surface is required to accurately understand the intermolecular events between the hair and its vicinal water. In this paper, we successfully obtained the infrared (IR) absorption spectra of the hydration water of human hair. We employed the multivariate curve resolution‐alternating least square (MCR‐ALS) method to separate the hydration and bulk water spectra from the whole spectra. Comparing the IR spectra of the hydration water of chemically untreated and bleached hair samples, we conclude that water molecules form strong hydrogen bonds with the bleached hair surface due to the destruction of the hair's hydrophobic outer layer and the consequent formation of hydrophilic residues.

Abstract:We use differential scanning calorimetry ( DSC ) to examine ten pure standard fiber samples and five polyester-cotton composite fiber samples to look for fiber evidence. The results show that the characteristics of DSC curves such as typical molten peak shape and position can serve as the useful criteria for differentiating fiber samples. Quantitative analyses of polyester-cotton composite fiber samples yield precise results with absolute errors generally being less than 3. 0%, which confirms the feasibility of using the DSC method for forensic identification.

Abstract:Human hair has become a promising non-invasive matrix in assessing exposure to environmental organic pollutants (OPs). However, exogenous contaminants, which were absorbed into the hair via sweat, sebum, and air particles/dust, could contribute to OP levels in hair and interfere with the precise exposure assessment. So far, the microscopic mechanisms underlying the absorption of exogenous OPs into hair remain inadequately understood. This study focused on the in-situ investigation of the diffusion processes of exogenous OPs into the hair structure using secondary ion mass spectrometry (SIMS) and isotopic tracer techniques. Results showed that the relative signal intensities of deuterium-labeled tris(1,3-dichloro-2-propyl) phosphate (TDCPP), 1-hydroxypyrene (1-OH-Pry), and bisphenol A (BPA) in the hair cortex were notably elevated after a 6-hour exposure. Diffusion coefficients of contaminants were related to their molecular weight, and absorption volumes to their water solubility and molecular structures. Exposure duration and solvent influenced the rate of diffusion and absorption volumes. The distribution of deuterium-labeled molecules in exposed hair samples after washing with two different solvents (acetone or water) was similar to that before washing. Our findings revealed the diffusion of OPs in hair cross-sections, indicating exogenous contributions to contaminants that are biologically incorporated into the hair.

Abstract:Human hair is a complex nanocomposite fiber whose physical appearance and mechanical strength are governed by a variety of factors like ethnicity, cleaning, grooming, chemical treatments and environment. Characterization of mechanical properties of hair is essential to develop better cosmetic products and advance biological and cosmetic science. Hence the behavior of hair under tension is of interest to beauty care science. Human hair fibers experience tensile forces as they are groomed and styled. Previous researches about tensile testing of human hair were seemingly focused on the longitudinal direction, such as elastic modulus, yield strength, breaking strength and strain at break after different treatment. In this research, experiment of evaluating the mechanical properties of human hair, such as Young's modulus and Poisson's ratio, was designed and conducted. The principle of the experimental instrument was presented. The system of testing instrument to evaluate the Young's modulus and Poisson's ratio was introduced. The range of Poisson's ratio of the hair from the identical person was evaluated. Experiments were conducted for testing the mechanical properties after acid, aqueous alkali and neutral solution treatment of human hair. Explanation of Young's modulus and Poisson's ratio was conducted base on these results of experiments. These results can be useful to hair treatment and cosmetic product.

Abstract:The mesoporosity of carbons can be determined using traditional methods such as gas physisorption or mercury intrusion, as well as by an alternative technique, thermoporometry (TPM). TPM focuses on determining porosity in the wet state. If water is used as probe liquid, the mesopores larger than 10 nm are hard to evaluate due to the overlapping of pore and bulk melting peaks. To overcome this problem, octamethylcyclotetrasiloxane (OMCTS) is used. Due to the fact that OMCTS undergoes solid-solid phase change in the same temperature range as melting in small mesopores, its use for mesopores below 10 nm is problematic. Thus, the applicability of OMCTS as a probe liquid for TPM characterization of small mesopores was tested in this work. The separation of melting in small pores from solid-solid transition was elaborated by several approaches like changes of heating rate, implementation of prefreezing step, deconvolution of experimental DSC records with the use of basic kinetic equation as underlying functions). It was found that the prefreezing step and a measurement of pure OMCTS under the same conditions as expected for TPM measurement is useful since the solid-solid transition depends on the measurement conditions. The usefulness of the suggested measurement conditions and deconvolution method was successfully demonstrated on carbonaceous samples with exclusively small mesopores (5 nm), as well as carbons containing both small and large mesopores. The results indicate that this approach enables the investigation of the entire mesoporous range and makes OMCTS a more versatile probe liquid for TPM.

Abstract:The hair is a filamentous biomaterial consisting of the cuticle , the cortex and the medulla , all held together by the cell membrane complex. The cortex mostly consists of helical keratin proteins that spiral together to form coiled-coil dimers, intermediate filaments, micro-fibrils and macro-fibrils. We used X-ray diffraction to study hair structure on the molecular level, at length scales between ∼3–90 Å, in hopes of developing a diagnostic method for diseases affecting hair structure allowing for fast and noninvasive screening. However, such an approach can only be successful if common hair treatments do not affect molecular hair structure. We found that a single use of shampoo and conditioner has no effect on packing of keratin molecules, structure of the intermediate filaments or internal lipid composition of the membrane complex. Permanent waving treatments are known to break and reform disulfide linkages in the hair. Single application of a perming product was found to deeply penetrate the hair and reduce the number of keratin coiled-coils and change the structure of the intermediate filaments. Signals related to the coiled-coil structure of the α -keratin molecules at 5 and 9.5 Å were found to be decreased while a signal associated with the organization of the intermediate filaments at 47 Å was significantly elevated in permed hair. Both these observations are related to breaking of the bonds between two coiled-coil keratin dimers.

Abstract:Multiple IR microspectroscopy techniques were used to create a spectral histology of the hair and hair follicle, revealing the presence of unsaturated lipids in the hair medulla, the concentration of glycogen in the outer root sheath of the follicle, the distribution of calcium soaps at the surface of the intact hair, and the different maturation stages of the hair follicle progenitor cells. Objective Today, there is only limited knowledge of the spatial organization of hair chemistry. Infrared microspectroscopy is a well‐established tool to provide such information and has significantly contributed to this field. In this study, we present new results combining multiple infrared microspectroscopy methods at different length scales to create a better chemical histology of human hair, including the hair follicle, hair shaft, hair medulla and hair cuticle. Methods We used hyperspectral IR imaging & spectroscopy (HIRIS) and synchrotron‐radiation FTIR microspectroscopy (SR‐μFTIR) to measure transversal hair sections and SR‐μFTIR to obtain high‐resolution maps of longitudinal sections from the hair shaft and from the hair follicle. We used optical photothermal IR microspectroscopy (OPTIR) to analyse the cuticle surface of intact hairs. Results By mapping longitudinal sections of the human hair follicle with confocal SR‐μFTIR, we report the first demonstration of glycogen presence in the outer root sheath of the hair follicle by spectroscopy, and its quantification at the micron scale. Spectral maps, combined with machine learning‐based analysis, enabled us to differentiate the various layers of the hair follicle and provided insights into the chemical changes that occur during hair formation in the follicle. Using HIRIS and SR‐μFTIR to analyse the hair medulla in transversal sections of human hairs, we report here, for the first time by vibrational spectroscopy methods, the detection of unsaturated lipids at very low concentrations in the medulla. By analysing longitudinal sections of the hair shaft with SR‐μFTIR, we found that calcium carboxylates are present in large regions of the hair cuticle, and not just in small focal areas as previously thought. We then use OPTIR to analyse the hair cuticle of intact hairs at submicron resolution without sectioning and report the distribution of calcium carboxylates at the surface of intact hair for the first time. Conclusion These new findings illustrate the potential of infrared microspectroscopy for imaging the chemical composition of human hair and may have implications for biomedical research or cosmetology. Résumé Objectif Aujourd'hui, les connaissances sur l'organisation spatiale de la chimie capillaire sont limitées. La microspectroscopie infrarouge est un outil bien établi pour fournir de telles informations et a largement contribué à ce domaine. Dans cette étude, nous présentons de nouveaux résultats combinant plusieurs méthodes de microspectroscopie infrarouge à différentes échelles de longueur pour créer une meilleure histologie chimique des cheveux humains, y compris le follicule pileux, la tige pilaire, la moelle pilaire et la cuticule pilaire. Méthodes Nous avons utilisé l'imagerie et la spectroscopie hyperspectrales infrarouges (Hyperspectral IR Imaging & Spectroscopy, HIRIS) et la microspectroscopie IRTF par rayonnement synchrotron (synchrotron‐radiation FTIR microspectroscopy, SR‐μFTIR) pour mesurer des coupes transversales de cheveux, et la SR‐μFTIR pour obtenir des cartes à haute résolution des coupes longitudinales de la tige pilaire et du follicule pileux. Nous avons utilisé la microspectroscopie photothermique infrarouge optique (Optical Photothermal IR microspectroscopy, OPTIR) pour analyser la surface des cuticules de cheveux intacts. Résultats En cartographiant les coupes longitudinales du follicule pileux humain avec la SR‐μFTIR confocale, nous rapportons la première démonstration par spectroscopie de la présence de glycogène dans la gaine de la racine externe du follicule pileux, et sa quantification à l'échelle du micron. Les cartes spectrales, combinées à une analyse basée sur l'apprentissage automatisé, nous ont permis de différencier les différentes couches du follicule pileux et de mieux comprendre les changements chimiques qui surviennent pendant la formation des cheveux dans le follicule. En utilisant la méthode HIRIS et la SR‐μFTIR pour analyser la moelle pilaire dans les coupes transversales des cheveux humains, nous rapportons ici, pour la première fois par des méthodes de spectroscopie vibrationnelle, la détection de lipides insaturés à de très faibles concentrations dans la moelle. En analysant les coupes longitudinales de la tige pilaire par SR‐μFTIR, nous avons constaté que les carboxylates de calcium sont présents dans de vastes régions de la cuticule pilaire, et pas seulement dans de petites zones focales comme on le pensait auparavant. Abstract:Mammalian hair, as a key feature related to endothermy, physical protection, and camouflage, has a long geohistorical record that dates back even to the Mesozoic. However, the limited information on the taphonomy of hair hinders the understanding of its evolutionary significance in deep time. To address this, we conducted taphonomic experiments to simulate the transformation process of black hair. The changes in the morphological structure, microscopic characteristics, and chemical composition of hair during the simulated fossilization process were documented by scanning electron microscopy, Fourier transform infrared (FTIR) spectroscopy, and Raman spectroscopy. Our results revealed that damage to the morphology of hair began at a mild temperature (150–175°C), and hair transformed into a fluid-like mass at a relatively high temperature (200°C). In addition, the morphological structure of melanosomes had relatively greater preservation potential than keratins did, and the microscopic features of melanosomes were persistently stable. The Raman and FTIR spectra indicated that the characteristic melanin and keratin peaks changed as the temperature increased. Although the chemical signals were not lost completely, the variations in the characteristic amide bands in the FTIR spectra indicate that the keratins severely degraded at high temperatures (>200°C), whereas the characteristic melanin bands in the Raman spectra suggested that melanin signals were still recognizable, even at high temperatures (>200°C). These results were confirmed by principal component analysis (PCA) and principal component analysis linear discriminant analysis (PCA–LDA). In addition, the Raman signals of all the samples in the simulated experiments were distinguished from the signals of common geological carbonaceous materials (humic acid), which suggested that Raman spectroscopy can be used to detect melanin in fossils.
Chemical and physical treatments damage Caucasian and Afro-ethnic hair fibre: analytical and image assays

Abstract:Background: Sensory and structural characteristics of hair can be modified by chemical and physical treatments, as dyeing, brushing, but also by external factors, as sunlight radiation. However, quantitative data relating damage to the degree of hair curliness and treatments are missing. Objective: To evaluate the effect of chemical and physical treatments on different types of hair. Methods: In this study, we compared the effects of bleaching/dyeing, thioglycolate-based straightening, brushing/flat iron and UVA/Vis radiation on different types of hair: Caucasian (straight dark brown, straight blond, wavy dark brown and curly dark brown) and Afro-ethnic hair. Results: Bleaching/dyeing and UVA/Vis radiation increased combing work of Afro-ethnic hair tresses, indicating damage to the cuticle scales, which was confirmed by SEM images. Further, bleaching/dyeing caused wear on cuticles with high protein loss and reduction on the tryptophan content, independently of the hair curliness, which characterizes those procedures as very aggressive to hair structure. Straightening using ammonium thioglycolate, due to the oxidative treatment evolved, caused significant colour fading in brown/black tresses, while UVA/Vis radiation affected the colour of only blond hair tresses since they are poor in eumelanin, the photoprotective and photostable melanin molecule. Conclusion: The combination of analytical and image techniques contributed to understanding how the hair shape is related to the type of damage caused by several chemical and physical treatments. These findings contribute to the development of safety cosmetics that insure the beauty of curly hair, both Caucasian and Afro-ethnic hair.
Effect of Physical and Chemical Hair Removal Methods on Skin Barrier Function in vitro: Consequences for a Hydrophilic Model Permeant

Abstract:Background: Although very common in our society, the effect of hair removal on physiological skin parameters and on the ingress of applied chemicals has not been systematically investigated. Thus, as a first step, the aim of the present study was to elucidate the effect of hair removal through epilation (electric epilation, waxing) and depilation (dry and wet shaving, depilatory cream) on skin properties in vitro using the porcine ear model. Methods: Attenuated total reflection Fourier transform infrared spectroscopy, measurement of the transepidermal water loss (TEWL), visualization by capacitance-based contact imaging, confocal Raman spectroscopy (CRS), diffusion cell studies and tape stripping experiments were employed. Results: Increased TEWL and altered skin permittivity maps were observed. Decreased stratum corneum thickness was observed after waxing. Diffusion cell studies showed increased skin permeation especially in case of dry shaving, electric epilation and waxing. Conclusion: Considering CRS and diffusion cell data, a moderate if significant decrease in skin barrier function was found after hair removal by dry shaving (physical skin/material interaction) and epilation methods (plucking out the entire hair, for example, by electrical epilation and waxing). Subsequent experiments will include testing of different permeants covering a broad range of physicochemical properties in vitro and confirming our findings in vivo.

Abstract:Hair fiber is a key by-product of humane slaughter of pigs with considerable economic value. In the present study, we investigated the thermal properties of pig hair fiber using Differential Scanning Calorimetry (DSC) and thermogravimetric analysis (TGA). The DSC curve showed a broad endotherm (around 50–80°C) initially, followed by denaturation doublet peaks (229°C and 239°C) and finally a pyrolysis endotherm. The melting enthalpy of pig hair fiber was 9.93 J/g on dry basis. During TGA, distinct phases of initial weight loss due to loss of moisture and later through thermal degradation of protein around 238–240°C were observed. Mean thermal insulation and conductivity values of pig hair fiber were 0.068 ± 0.004 m2K/W and 0.029 ± 0.003 W/m/K, respectively. The thermal characteristics of pig hair fiber were similar to other keratin fibers of animal origin.
