Abstract:Biofilms are physical barriers composed of extracellular polymeric substances (EPS) that enable planktonic bacteria to resist host responses and antibacterial treatments, complicating efforts to clear bacteria such as Pseudomonas aeruginosa (P. aeruginosa) and thereby contributing to persistently chronic infections. As such, it is critical to develop a robust antimicrobial strategy capable of effectively eradicating P. aeruginosa biofilms and to further address aggressive clinical infection. In this study, ultrasound-activatable targeted nanoparticles were designed by using poly(lactic-co-glycolic acid) (PLGA) nanoparticles to encapsulate phase-transformable perfluoropentane (PFP) and the antibiotic meropenem via a double emulsion approach, followed by conjugation with anti-P. aeruginosa antibodies. In this strategy, ultrasound exposure can trigger PFP to produce microbubbles, inducing ultrasonic cavitation effects that can disrupt EPS components and allow nanoparticles to release meropenem to kill P. aeruginosa directly and accelerate the associated wound healing. These nanoparticles eradicated biofilms effectively and cleared bacteria in vitro as well as exhibited potent anti-infective activity in vivo. In summary, this study demonstrates the efficacy of a sonobactericidal strategy as a means of effectively and reliably eliminating biofilms.

Abstract:PurposeMethicillin-resistant Staphylococcus aureus (MRSA) infection at impaired wound is associated with high risks of developing to persistent bacterial infections since bacterial biofilm is easy to form in MRSA infected wounds. An advanced therapeutic approach to effectively penetrate and eliminate bacterial biofilm and to accelerate cell proliferation and migration at the wound is crucial.MethodsThe poly(ε-caprolactone)-monomethoxyl poly (ethylene glycol) (PCL-mPEG) micelles loaded with Quercetin and Rifampicin (QRMs) were prepared. Bacterial biofilm proliferation and elimination effect of QRMs were evaluated with confocal laser scanning microscopy. Antibacterial assay was further performed to detect antibacterial activity and mechanism. The cell scratch assay and cellular uptake were performed in HaCaT skin epithelial cells.ResultsOur results showed that the small sized QRMs could penetrate the interior of MRSA biofilm to disperse and eradicate biofilm. Then, antibiotics are released and accumulated in the acidic biofilm environment. QRMs could kill bacteria through increasing bacterial membrane permeability and altering membrane potential and membrane fluidity. Moreover, QRMs improved intracellular and cytoplasmic delivery efficiency of drugs to epithelial cells, and in the scratch test, presented a stronger ability to promote migration and proliferation of HaCaT cells compared with free drugs. Hemolysis test further proved good biocompatibility of QRMs.ConclusionsQRMs could potentially be used as a novel dual-functional nanotherapeutic for anti-bacterial infection by eradicating biofilm and accelerating cells proliferation at MRSA infected wound.

Abstract:The misuse and abuse of antibiotics have given rise to a severe problem of the drug resistance of bacteria. Solving this problem has been a vitally important task in the modern medical arena. In this work, an antimicrobial peptide (AMP), BF2b, and gold nanorods (AuNRs) were used to develop a specific drug delivery system for killing methicillin-resistant Staphylococcus aureus (MRSA). On the one hand, BF2b has unique anti-bacterial performance and has a lower tendency than traditional antibiotics to engender the drug resistance of bacteria. On the other hand, AuNRs have diverse distinct properties, such as photo-thermal conversion, which can be employed for photo-thermal sterilization. We aimed to integrate the anti-bacterial activity of BF2b and the photo-thermal sterilization of AuNRs to kill drug-resistant bacteria. Fourier-transform infrared spectroscopy, microBCA and zeta potential measurements were utilized to characterize the product, AuNR@PEG/BF2b. Transmittance electron microscopy, UV-vis spectroscopy and photothermal conversion measurement were conducted to verify the stability and photothermal conversion capacity of AuNR@PEG/BF2b. Cell viability and hemolysis assay were carried out to test the biocompatibility of AuNR@PEG/BF2b. Finally, the in vitro and in vivo experiments were performed to demonstrate the excellent bactericidal activity of AuNR@PEG/BF2b.

Abstract:Phototherapy has emerged to eradicate recalcitrant bacteria without causing drug resistance, but it is often accompanied by considerable limitations owing to a high tolerance of recalcitrant bacteria to heat and oxidative damage, leading to low efficiency of monotherapy and unwanted side effects. Assuming that employing antimicrobial peptides (AMPs) to disrupt bacterial membranes could reduce bacterial tolerance, a multifunctional "on-demand" nanosystem based on zeolitic imidazolate framework-8 (ZIF-8) with metal ions for intrinsic antibacterial activity was constructed to potently kill methicillin-resistant Staphylococcus aureus (MRSA). Then, microneedles (MNs) were used to transdermally deliver the ZIF-8-based nanosystem for localized skin infection. After MNs insertion, the nanoplatform could specifically deliver the loaded therapeutic components to bacterial infection sites through employing hyaluronic acid (HA) as a capping agent, thus realizing the "on-demand" payload release triggered by excess hyaluronidase secreted by MRSA. The prepared nanosystem and MNs were confirmed to exert an amplified triple therapy originating from membranolytic effect, phototherapy, and ion therapy, thus displaying a powerful bactericidal and MRSA biofilm destruction ability. This intelligent antimicrobial strategy may bring a dawn of hope for eradicating multidrug-resistant bacteria and biofilms.
Biofilm microenvironment-activated multimodal therapy nanoplatform for effective anti-bacterial treatment and wound healing

Abstract:Antimicrobial drug development faces challenges from bacterial resistance, biofilms, and excessive inflammation. Here, we design an intelligent nanoplatform utilizing mesoporous silica nanoparticles doped with copper ions for loading copper sulfide (DM/Cu 2+ -CuS). The mesoporous silica doped with tetrasulfide bonds responds to the biofilm microenvironment (BME), releasing Cu 2+ ions, CuS along with hydrogen sulfide (H 2 S) gas. The release of hydrogen sulfide within 72 h reached 793.5 μM, significantly higher than that observed with conventional small molecule donors. H 2 S induces macrophages polarization towards the M2 phenotype, reducing inflammation and synergistically accelerating endothelial cell proliferation and migration with Cu 2+ ions. In addition, H 2 S disrupts extracellular DNA within biofilms, synergistically photothermal enhanced peroxidase-like activity of CuS to effectively eradicate biofilms. Remarkably, DM-mediated consumption of endogenous glutathione enhances the anti-biofilm activity of H 2 S and improves oxygen species (ROS) destruction efficiency. The combination of photothermal therapy (PTT), chemodynamic therapy (CDT), and gas treatment achieves sterilization rates of 99.3% and 99.6% against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ), respectively, in vitro under 808 nm laser irradiation. Additionally, in vivo experiments demonstrate a significant biosafety and antibacterial potential. In summary, the H 2 S donor developed in this study exhibits enhanced biocompatibility and controlled release properties. By integrating BME-responsive gas therapy with antibacterial ions, PTT and CDT, a synergistic multimodal strategy is proposed to offer new therapeutic approaches for wound healing. Statement of Significance The advanced DMOS/Cu 2+ -CuS (DMCC) multimodal therapeutic nanoplatform has been developed for the treatment of drug-resistant bacterial wound infections and has exhibited enhanced therapeutic efficacy through the synergistic effects of photothermal therapy, chemodynamic therapy, Cu 2+ ions, and H 2 S. The DMCC exhibited exceptional biocompatibility and could release CuS, Cu 2+ , and H 2 S in response to elevated concentrations of glutathione within the biofilm microenvironment. H 2 S effectively disrupted the biofilm structure. Meanwhile, peroxidase activity of CuS combined with GSH-mediated reduction of Cu 2+ to Cu + generated abundant hydroxyl radicals under acidic conditions, leading to efficient eradication of pathogenic bacteria. Furthermore, both H 2 S and Cu 2+ could modulate M2 macrophages polarization and regulate immune microenvironment dynamics. These strategies collectively provide a novel approach for developing antibacterial nanomedical platforms.

Abstract:Bacterial resilience within biofilms, rendering them up to 1000 times more resistant to antibiotic drugs, poses a formidable challenge. This study introduces a targeted biofilm eradication strategy, termed "target-penetration-killing-eradication", implemented through magnetic micro-robotic technology. Specifically, we present the development of a magnetic-guided nano-antibacterial platform designed for alternating magnetic field (AMF) controlled vancomycin release in the eradication of Staphylococcus aureus biofilms. To address the issue of premature vancomycin release in physiological conditions, we employed a temperature-sensitive linking agent, 4,4'-azobis(4-cyano valeric acid), facilitating the conjugation of vancomycin onto Fe3O4/CS nanocomposites, resulting in the novel construct Fe3O4@CS-ACVA-VH. The release mechanism adheres to first-order kinetics and Fickian diffusion, with each 10-min AMF treatment releasing approximately 8.4 ± 1.1% of vancomycin. The potency of vancomycin in the release solution was similar to that of the original drug (MIC: 7.4 ± 3.5 vs. 5.6 μg/mL). Fe3O4@CS-ACVA-VH exhibited sustained antibacterial efficacy, inhibiting bacterial growth for four consecutive days and preventing the formation of bacterial biofilms on its surface. Contact-inhibition bacterial activity of Fe3O4@CS-ACVA-VH against S. aureus was 0.046875 mg/mL. Conceptually validating our approach, we emphasize Fe3O4@CS-ACVA-VH's exceptional ability to penetrate S. aureus biofilms under static magnetic field attraction. Furthermore, the nano-platform offers the unique advantage of on-demand vancomycin release through alternating magnetic field stimulation, effectively clearing a larger biofilm area. This multifunctional nano-platform demonstrates magnetic-guided biofilm penetration followed by controlled vancomycin release, presenting a promising strategy for enhanced biofilm eradication.

Abstract:Chronic wounds frequently become infected with bacterial biofilms which respond poorly to antibiotic therapy. Aminoglycoside antibiotics are ineffective at treating deep-seated wound infections due to poor drug penetration, poor drug uptake into persister cells, and widespread antibiotic resistance. In this study, we combat the two major barriers to successful aminoglycoside treatment against a biofilm-infected wound: limited antibiotic uptake and limited biofilm penetration. To combat the limited antibiotic uptake, we employ palmitoleic acid, a host-produced monounsaturated fatty acid that perturbs the membrane of gram-positive pathogens and induces gentamicin uptake. This novel drug combination overcomes gentamicin tolerance and resistance in multiple gram-positive wound pathogens. To combat biofilm penetration, we examined the ability of sonobactericide, a non-invasive ultrasound-mediated-drug delivery technology to improve antibiotic efficacy using an in vivo biofilm model. This dual approach dramatically improved antibiotic efficacy against a methicillin-resistant Staphylococcus aureus (MRSA) wound infection in diabetic mice.

Sono‐Triggered Biomimetically Nanoantibiotics Mediate Precise Sequential Therapy of MRSA‐Induced Lung Infection

Abstract:AMV@NanoCip, a novel biomimetic nanomedicine, integrates PBP2a antibody‐presenting nanovesicles with self‐assembled ciprofloxacin nanoparticles exhibiting sonodynamic properties. This formulation demonstrates exceptional MRSA‐targeting and antibacterial efficacy, achieving 99.99% sterilization in vitro and significant bacterial reduction in a pneumonia model. The sequential treatment strategy, combining adhesion, membrane disruption, and synergistic therapy, presents a promising approach for combating bacterial infections. Bacterial‐induced lower respiratory tract infections are a growing global health concern, exacerbated by the inefficacy of conventional antibiotics and delivery methods to effectively target the lower respiratory tract, leading to suboptimal therapeutic outcomes. To address this challenge, this work engineers PBP2a antibody‐presenting membrane nanovesicles (AMVs) specifically designed to target the penicillin‐binding protein variant on the surface of methicillin‐resistant Staphylococcus aureus (MRSA). Concurrently, this work develops pure ciprofloxacin nanoparticles (NanoCip) that, for the first time, exhibits exceptional self‐generated sonodynamic properties, attributed to hydrogen‐bond‐driven self‐assembly, while maintaining their inherent pharmacological efficacy. These NanoCip particles are integrated with AMVs to create a novel biomimetic nanomedicine, AMV@NanoCip. This formulation demonstrated remarkable MRSA‐targeting affinity in both in vitro and in vivo models, significantly enhancing antibacterial activity. Upon ultrasound stimulation, AMV@NanoCip achieves over 99.99% sterilization of MRSA in vitro, with a reduction exceeding 5.14 Log CFU. Prokaryotic transcriptomic analysis further elucidates the synergistic mechanisms by which AMV@NanoCip, coupled with ultrasound, disrupts the MRSA exoskeleton. In a MRSA‐induced pneumonia animal model, AMV@NanoCip+US results in a substantial bacterial load reduction in the lungs (99.99%, 4.02 Log CFU). This sequential treatment strategy (adhesion‐membrane disruption‐synergistic therapy) offers significant promise as an innovative therapeutic approach for combating bacterial infections.

A multifunctional nanoplatform for precision-guided therapeutic intervention in bacterial infection

Abstract:Skin wound infection has become a global clinical problem in recent years. Curcumin (Cur) and polylysine (PLL) are natural products with strong antibacterial properties. However, the poor water solubility and low stability of Cur and the cationic toxicity of PLL limit their application. In this study, we synthesized a macromolecular hyaluronic acid (HA)-curcumin drug (HC) via esterification. HC was attracted by electrostatic interactions with positively charged PLL to form a spherical nanocomplex (HCP) with hyaluronidase (HAase) and pH dual response under ultrasonication. HCP was found to target the bacterial infection microenvironment and release Cur and PLL for synergistic antibacterial action. In addition, HCP was proven to exhibit good biocompatibility and broad spectrum antibacterial activity to bacterial strains S. aureus and E. coli and antibacterial biofilm activities in vitro. In vivo experiments showed that HCP could inhibit pathogens and promote wound healing. These results prove that HCP can be used as a new strategy for the treatment and management of infected wounds.
Carrier-Free Binary Self-Assembled Nanomedicines Originated from Traditional Herb Medicine with Multifunction to Accelerate MRSA-Infected Wound Healing by Antibacterial, Anti-Inflammation and Promoting Angiogenesis

This trend is further aggravated as the overuse antibiotics and the bacterial resistance to all known antibacterial agents. Therefore, new therapeutic alternatives are urgently needed. Methods: Enlightenment the combination usage of traditional herb medicine, one carrier-free binary nanoparticles (GA-BBR NPs) was discovered, which was self-assembled from gallic acid and berberine through electrostatic interaction, π-π stacking and hydrophobic interaction; and it could be successfully prepared by a green, cost-effective and "one-pot" preparation process. Results: The nanoparticles exhibited strong antibacterial activity and biofilm removal ability against multidrug-resistant S. aureus (MRSA) by downregulating mRNA expression of rpsF, rplC, rplN, rplX, rpsC, rpmC and rpsH to block bacterial translation mechanisms in vitro and in vivo, and it had well anti-inflammatory activity and a promising role in promoting angiogenesis to accelerate the wound healing on MRSA-infected wounds model in vivo. Additionally, the nanoparticles displayed well biocompatibility without cytotoxicity, hemolytic activity, and tissue or organ toxicity. It occurs when tissue is destroyed or cellular integrity is compromised due to mechanical, physiological, or metabolic related issues; and if the trauma untreated, chronic wounds can easily develop. 1–4 In chronic wounds, bacteria can multiply and cause serious infections. Infections lead to delay healing, increase wound size, tissue decay, and even severe systemic damages. In the initial stages of chronic wound formation, gram-positive bacteria, especially Staphylococcus aureus , are predominate. 5 Once the bacteria have settled in the wound, they will multiply rapidly to form a mature biofilm. Abstract:Highlights • A nanoplatform equipped with "disruption and killing" functions is designed to enhance the effectiveness of conventional antibiotics for biofilm eradication. • The nanoplatform can degrade the polysaccharides of extracellular polymeric substances to disrupt the structural integrity of biofilms. • Under near-infrared irradiation, the nanoplatform facilitates the release and the bacterial uptake of rifampicin to effectively eliminate the bacteria within biofilms. Due to the absence of timely and effective therapies, infections induced by bacterial biofilms have been widely acknowledged as a significant global public health concern. In modern times, aside from surgical intervention (when appropriate), antibiotics are the sole clinical option for treating biofilm-associated infections. However, the rise of drug resistance, as well as the poor therapeutic effects of current treatment regimens in eliminating biofilms highlight the requirement for novel strategies to enhance the accessibility of antibiotics in the "post-antibiotic era". The current study presents a multifunctional nanoplatform equipped with a "disruption and killing" function to enhance the effectiveness of conventional antibiotics for the eradication of biofilms. Herein, mesoporous silica nanoparticles were employed as carriers to encapsulate the model antibiotic rifampicin (Rif). Subsequently, the nanoparticles were coated with layers of the tannic acid/iron ion (TA/Fe) complex and immobilized with α-amylase. The α-amylase present in the outer layer can degrade the polysaccharides of extracellular polymeric substances (EPS), which in turn disrupts the structural integrity of the biofilms, thus facilitating the entry of the nanoplatform. When exposed to near-infrared (NIR) light, the TA/Fe complex layers can generate heat, which facilitates the release of Rif and increases the bacterial uptake of Rif by damaging the bacterial cell membrane, ultimately resulting in the elimination of bacteria within biofilms. The in vitro experiments demonstrated that this nanoplatform effectively eliminated over 99% of biofilms formed by Staphylococcus aureus and Pseudomonas aeruginosa when exposed to NIR radiation for 10 min. Additionally, in vivo experimental findings further validated the extensive therapeutic efficacy of this nanoplatform against biofilm-infected wounds, accelerating the rate of healing and reducing inflammatory reactions. To summarize, this nanoplatform provides a novel avenue to improve the effectiveness of conventional antibiotics in eradicating bacterial biofilms. Facile Assembly of Multifunctional Antibacterial Nanoplatform Leveraging Synergistic Sensitization between Silver Nanostructure and Vancomycin

Abstract:The emergence of antibiotic-resistant bacterial strains renders the conventional antibiotic therapy less efficient. The integration of two distinct bactericides into one compact platform provides a promising strategy to realize a combinational antimicrobial therapy. In this work, an efficient chemo-Ag nanohybrid antibacterial platform was facilely developed based on the integration of vancomycin-carrying polydopamine with silver nanoparticles (). The as-synthesized antibacterial nanohybrid inherited the intrinsic properties of both bactericides to achieve a synergistic antibacterial performance against both Staphylococcus aureus and Escherichia coli strains by attacking bacteria from two distinct fronts. Through this combinational therapy, the efficiency of antibiotic against S. aureus was significantly improved by reducing drug dosage with less opportunity for imparting drug resistance. In addition, this antibacterial nanohybrid, with innate photothermal properties, could achieve auxiliary hyperthermia-assisted bacterial inactivation in the meantime. Furthermore, the outstanding in vivo bacteria-killing activity and wound-healing acceleration were demonstrated in a S. aureus-infected mouse skin defect model. Taken together, this bactericidal nanohybrid could achieve sustained antibiotic release and wipe out bacteria more effectively in a synergistic way, thus reducing the emergence of antibiotic resistance. This work holds great potential to advance the development of novel antibacterial agents and combinational strategies as a promising supplement of antibiotics in the near future.

Synergistic and On-Demand Release of Ag-AMPs Loaded on Porous Silicon Nanocarriers for Antibacteria and Wound Healing

Abstract:Due to the abuse of antibiotics, antimicrobial resistance is rapidly emerging and becoming a major global risk for public health. Thus, there is an urgent need for reducing the use of antibiotics, finding novel treatment approaches, and developing controllable release systems. In this work, a dual synergistic antibacterial platform with on-demand release ability based on silver nanoparticles (AgNPs) and antimicrobial peptide (AMP) coloaded porous silicon (PSi) was developed. The combination of AgNPs and AMPs (Tet-213, KRWWKWWRRC) exhibited an excellent synergistic antibacterial effect. As a carrier, porous silicon can efficiently load AgNPs and AMP under mild conditions and give the platform an on-demand release ability and a synergistic release effect. The AgNPs and AMP coloaded porous silicon microparticles (AgNPs-AMP@PSiMPs) exhibited an acid pH and reactive oxygen species (ROS)-stimulated release of silver ions (Ag+) and AMPs under bacterial infection conditions because of oxidation and desorption effects. Moreover, the release of the bactericide could be promoted by each other due to the interplay between AgNPs and Tet-213. In vitro antibacterial tests demonstrated that AgNPs-AMP@PSiMPs inherited the intrinsic properties and synergistic antibacterial efficiency of both bactericides. In addition, wound dressing loaded with AgNPs-AMP@PSiMPs showed outstanding in vivo bacteria-killing activity, accelerating wound-healing, and low biotoxicity in aStaphylococcus aureus-infected rat wound model. The present work demonstrated that PSiMPS might be an efficient platform for loading the antibiotic-free bactericide, which could synergistically and on-demand release to fight wound infection and promote wound healing.

Abstract:Wound biofilms represent an elusive conundrum in contemporary treatment and diagnostic options, accredited to their escalating antibiotic resistance and interference in chronic wound healing processes. Here, we developed mesoporous polydopamine (mPDA) nanoparticles, and grafted with rhodamine B (Rb) as biofilm lipase responsive detection probe, followed by π − π stacking mediated ciprofloxacin (CIP) loading to create mP-Rb@CIP nanoparticles. mPDA NPs with a melanin structure could quench fluorescence emissions of Rb. Once encountering biofilm in vivo , the ester bond in Rb and mPDA is hydrolyzed by elevated lipase concentrations, triggering the liberation of Rb and restore fluorescence emissions to achieve real-time imaging of biofilm-infected wounds. Afterwards, the 808 nm near-infrared (NIR) illumination initiates a spatiotemporal controlled antibacterial photothermal therapy (PTT), boosting its effectiveness through photothermal-triggered CIP release for synergistic biofilm eradication. The mP-Rb@CIP platform exhibits dual diagnostic and therapeutic functions, efficaciously treating biofilm-infected wounds in vivo and in vitro . Particularly, the mP-Rb@CIP/NIR procedure expedites wound-healing by alleviating oxidative stress, modulating inflammatory mediators, boosting collagen synthesis, and promoting angiogenesis. Taken together, the theranostic nanosystem strategy holds significant potential for addressing wound biofilm-associated infections.

Abstract:Biomimetic nanovesicles co‐assembled by macrophage membranes and artificial lipids are developed for delivering antibiotics to treat bacterial infections. The macrophage membrane endows the nanovesicles with the biodetoxification capacity, while the artificial lipid membrane can be engineered to increase the fluidity and bacterial anchoring. Injection of this system can effectively eliminate S. aureus in the infected tissue and reduce inflammation. Tissue bacterial infections are a major pathological factor in many diseases. Effects on this aspect are in focus for the development of coordinated therapeutic strategies for bacterial killing and anti‐inflammation. Here, inspired by the biodetoxification capacity of immune cells, multifunctional biomimetic nanovesicles (MφM‐LPs) that are co‐assembled by macrophage membranes and artificial lipids to deliver antibiotics for treating bacterial infections, are presented. The macrophage membrane endows the MφM‐LPs with the capacity of lipopolysaccharide and inflammatory cytokine neutralization, while the artificial lipid membrane can be further engineered to increase the fluidity and anchor to bacteria. In addition, the MφM‐LPs can deliver sufficient ciprofloxacin with controllable release to accomplish an excellent antibacterial activity and biodetoxification capacity in vitro. Based on these advantages, it is demonstrated in a mouse model of Staphylococcus aureus (S. aureus) focal infection, that a single injection of the biomimetic nanovesicles can effectively anchor to and eliminate S. aureus in the infected tissue and reduce inflammatory cytokine levels. Thus, the tissue regeneration and collagen deposition can be accelerated. These results indicate the potential values of integrating natural and artificial membrane materials as a multifunctional biomimetic drug delivery system to treat bacterial infections.

Abstract:The use of opioid medications for analgesia is associated with concerns about adverse side effects and the potential for development of physical dependence, tolerance, or addiction. Pain often is undertreated, which may provoke drug-seeking behavior by patients. Physicians must assess requests for more pain medication as stemming from either undertreatment of pain, development of physical tolerance, or addiction. Important tools for addiction screening include the use of questionnaires, patient interviews, and lab tests. In this study, the physiological and behavioral consequences of chronic pain and its treatment with opioids, along with guidelines for prescribing opioid pain medication, are presented.

Abstract:A "natural" olfactory learning paradigm was used to assess the effects of an altered perinatal olfactory environment on the development of odor-elicited behavior in young rats. Infant rats (from 3 to 12 days of age) reared by dams fed a eucalyptol-adulterated diet responded to the odor of eucalyptol with high levels of activity, mouthing, and probing, and also demonstrated a marked preference for that odor in a two-choice situation (Experiment 1). The effects were observed only in pups whose dam was fed eucalyptol-adulterated diet and were not observed in pups exposed to nonmaternal sources of odor. The intensity of behavioral activity elicited by eucalyptol odor varied, to some extent, with the concentration of the eucalyptol in the dam's diet during prenatal exposure (Experiment 2). Cross-fostering revealed that these effects were due almost entirely to the pups' postnatal exposure to the eucalyptol odor (Experiment 3). These findings indicate that novel odors specifically associated with the mother can gain control over orientation and ingestion-related behaviors.
Multi-functional Nanofilms Capable of Angiogenesis, Near-Infrared-triggered Anti-Bacterial Activity and Inflammatory Regulation for Infected Wound Healing

Abstract:Chronic infected wound healing is a critical challenge in clinical practice owing to the involvement of multiple physiological processes, including bacteria-related, inflammatory regulation and angiogenesis. Therefore, a multi-functional strategy with synergistic anti-bacterial, anti-inflammatory and pro-angiogenic effects should be developed. Owing to their biomimetic structural features and controlled delivery of active agents, electrospun nanofilms are promising biomaterials for the treatment of skin defects. In this study, we fabricated multi-functional nanofilms with pro-angiogenic, anti-bacterial and anti-inflammatory capacities. First, strontium (Sr) ions were incorporated into poly(L-lactic-co-caprolactone) (PLCL) nanofilms. Subsequently, polydopamine (PDA) and zinc oxide (ZnO) were decorated onto the surface of Sr-loaded PLCL nanofilms to prepare ZnO/PDA@PLCL@Sr nanofilms. In vitro results showed that ZnO/PDA@PLCL@Sr nanofilms were biocompatible, exhibited angiogenic activity and significantly inhibited the growth of Staphylococcus aureus and Escherichia coli upon near-infrared -light irradiation. Furthermore, ZnO/PDA@PLCL@Sr nanofilms were found to drive the transformation of macrophages into the M2 phenotype. In vivo results further validated that ZnO/PDA@PLCL@Sr nanofilms exhibited pro-angiogenic and anti-bacterial activities and regulated inflammation to accelerate wound -healing in a rat model of bacteria-infected skin defects. In conclusion, we successfully developed a multi-functional biomaterial with pro-angiogenic, anti-bacterial and anti-inflammatory capacities to treat chronic infected wounds.

Abstract:A synergistic antibacterial nanoparticles (MPH NPs) are prepared by loading antibacterial peptide polymyxin B (PMB) on Fe‐MOF (MIL‐100) with hyaluronic acid (HA) modification. MPH NPs have synergistic broad‐spectrum antibacterial properties on E. coli, S. aureus, and MRSA by chemodynamic therapy (CDT) and release of PMB. MPH NPs can be used for wound healing and lung infection therapy. The problem of antimicrobial resistance (AMR) caused by the abuse of antibiotics is becoming serious. The development of antibacterial materials with synergistic efficiency and treatment of deep tissue/organ infections is imminent. Herein, synergistic antibacterial nanoparticles (MPH NPs) are prepared by loading antibacterial peptide polymyxin B (PMB) on Fe‐based MOF (MIL‐100) with hyaluronic acid (HA) modification. MPH NPs exerts antibacterial effects by chemodynamic therapy (CDT) and the release of PMB. MPH NPs have broad‐spectrum antibacterial properties on Gram‐negative bacteria (E. coli, 100%), Gram‐positive bacteria (S. aureus, 98.5 %), and MRSA (98.4%). Importantly, MPH NPs not only promote the healing of infected wounds but also target lungs to accomplish organ infection therapy. Therefore, this study provides a new strategy for designing a synergetic anti‐AMR bacteria system and the function for deep tissue/organ infection therapy in the future.

Abstract:Background: Methicillin-resistant Staphylococcus aureus (MRSA) biofilm-associated bacterial infection is the primary cause of nosocomial infection and has long been an ongoing threat to public health. MRSA biofilms are often resistant to multiple antimicrobial strategies, mainly due to the existence of a compact protective barrier; thus, protecting themselves from the innate immune system and antibiotic treatment via limited drug penetration. Results: A hierarchically structured hydrogen sulfide (H2S)-releasing nano-disinfectant was presented, which was composed of a zinc sulfide (ZnS) core as a H2S generator and indocyanine green (ICG) as a photosensitizer. This nano-disinfectant (ICG-ZnS NPs) sensitively responded to the biofilm microenvironment and demonstrated efficient eradication of MRSA biofilms via a synergistic effect of Zn2+, gas molecule-mediated therapy, and hyperthermia. Physically boosted by released H2S and a near-infrared spectroscopy-induced hyperthermia effect, ICG-ZnS NPs destroyed the compactness of MRSA biofilms showing remarkable deep-penetration capability. Moreover, on-site generation of H2S gas adequately ameliorated excessive inflammation, suppressed secretion of inflammatory cytokines, and expedited angiogenesis, therefore markedly accelerating the in vivo healing process of cutaneous wounds infected with MRSA biofilms. Conclusion: ICG-ZnS NPs combined with NIR laser irradiation exhibited significant anti-biofilm activity in MRSA biofilms, can accelerate the healing process through deep-penetration and anti-inflammatory effectuation. The proposed strategy has great potential as an alternative to antibiotic treatment when combating multidrug-resistant bacterial biofilms.
