Abstract:Although phosphoenolpyruvate carboxylases (PEPC) are reported to be involved in fatty acid accumulation, nitrogen assimilation, and salt and drought stresses, knowledge on the function of PEPC genes is still limited, particularly on the bacterial-type PEPC gene. To investigate the physiological functionality of Atppc4 , an Arabidopsis bacterial-type PEPC gene, Atppc4 , was specifically suppressed by artificial microRNA (amiRNA) in Arabidopsis . Transgenic plants with constitutively expressed Atppc4 -amiRNA exhibited substantially decreased accumulation of Atppc4 transcripts, whereas other three plant-type PEPC genes, Atppc1 , Atppc2 and Atppc3 , were significantly upregulated in roots. The PEPC activity was improved about 5.1 times in roots of Atppc4 -amiRNA transgenic lines. This result indicates that transcription of bacterial-type and plant-type PEPC genes in plants is interacted with each other in plants. The bacterial-type PEPC genes, Atppc4 , may play an important role in modulating the transcription of plant-type PEPC genes. The effects of Atppc4 on seed lipid content and fatty acid composition were not detected in this research. This indicated that Atppc4 might be independent of plant lipid accumulation. However, the root development was found to be related with Atppc4 . Root elongation of transgenic plants was significantly inhibited. The inhibition can be partially relieved by salt treatment. The results showed that specific downregulation of the bacterial-type PEPC gene, Atppc4 , by artificial microRNA improved salt tolerance in Arabidopsis . The improved salt tolerance may be related with the improved PEPC activity.
Genome-Wide Identification and Analysis of the Phosphoenolpyruvate Carboxylase Gene Family in Suaeda aralocaspica, an Annual Halophyte With Single-Cellular C4 Anatomy

Abstract:Phosphoenolpyruvate carboxylase (PEPC) plays pivotal roles in the carbon fixation of photosynthesis and a variety of metabolic and stress pathways. Suaeda aralocaspica belongs to a single-cellular C 4 species and carries out a photosynthetic pathway in an unusually elongated chlorenchyma cell, which is expected to have PEPCs with different characteristics. To identify the different isoforms of PEPC genes in S. aralocaspica and comparatively analyze their expression and regulation patterns as well as the biochemical and enzymatic properties in this study, we characterized a bacterial-type PEPC (BTPC; SaPEPC-4) in addition to the two plant-type PEPCs (PTPCs; SaPEPC-1 and SaPEPC-2) using a genome-wide identification. SaPEPC-4 presented a lower expression level in all test combinations with an unknown function; two SaPTPCs showed distinct subcellular localizations and different spatiotemporal expression patterns but positively responded to abiotic stresses. Compared to SaPEPC-2 , the expression of SaPEPC-1 specifically in chlorenchyma cell tissues was much more active with the progression of development and under various stresses, particularly sensitive to light, implying the involvement of SaPEPC-1 in a C 4 photosynthetic pathway. In contrast, SaPEPC-2 was more like a non-photosynthetic PEPC. The expression trends of two SaPTPCs in response to light, development, and abiotic stresses were also matched with the changes in PEPC activity in vivo (native) or in vitro (recombinant), and the biochemical properties of the two recombinant SaPTPCs were similar in response to various effectors while the catalytic efficiency, substrate affinity, and enzyme activity of SaPEPC-2 were higher than that of SaPEPC-1 in vitro. All the different properties between these two SaPTPCs might be involved in transcriptional (e.g., specific cis -elements), posttranscriptional [e.g., 5′-untranslated region (5′-UTR) secondary structure], or translational (e.g., PEPC phosphorylation/dephosphorylation) regulatory events. The comparative studies on the different isoforms of the PEPC gene family in S. aralocaspica may help to decipher their exact role in C 4 photosynthesis, plant growth/development, and stress resistance.

Abstract:Phosphoenolpyruvate carboxylase (PEPC) plays a crucial role in the initial carbon fixation process in C 4 plants. However, its nonphotosynthetic functions in Haloxylon ammodendron , a C 4 perennial xerohalophytic shrub, are still poorly understood. Previous studies have reported the involvement of PEPC in plant responses to abiotic stresses such as drought and salt stress. However, the underlying mechanism of PEPC tolerance to drought stress has not been determined. In this study, we cloned the C 4 -type PEPC gene HaPEPC1 from H. ammodendron and investigated its biological function by generating transgenic Arabidopsis plants with ectopic expression of HaPEPC1 . Our results showed that, compared with WT (wild-type) plants, ectopic expression of HaPEPC1 plants exhibited significantly greater germination rates and chlorophyll contents. Furthermore, under drought stress, the transgenic plants presented increased root length, fresh weight, photosynthetic capacity, and antioxidant enzyme activities, particularly ascorbate peroxidase and peroxidase. Additionally, the transgenic plants exhibited reduced levels of malondialdehyde, H 2 O 2 (hydrogen peroxide), and O 2 − (superoxide radical). Transcriptome analysis indicated that ectopic expression of HaPEPC1 primarily regulated the expression of genes associated with the stress defence response, glutathione metabolism, and abscisic acid (ABA) synthesis and signalling pathways in response to drought stress. Taken together, these findings suggest that the ectopic expression of HaPEPC1 enhances the reduction of H 2 O 2 and O 2 − in transgenic plants, thereby improving reactive oxygen species (ROS) scavenging capacity and enhancing drought tolerance. Therefore, the HaPEPC1 gene holds promise as a candidate gene for crop selection aimed at enhancing drought tolerance.

Transformation of Beta-Lycopene Cyclase Genes from Salicornia Europaea and Arabidopsis Conferred Salt Tolerance in Arabidopsis and Tobacco

Abstract:Inhibition of lycopene cyclization decreased the salt tolerance of the euhalophyte Salicornia europaea L. We isolated a β-lycopene cyclase gene SeLCY from S. europaea and transformed it into Arabidopsis with stable expression. Transgenic Arabidopsis on post-germination exhibited enhanced tolerance to oxidative and salt stress. After 8 and 21 d recovery from 200 mM NaCl treatment, transgenic lines had a higher survival ratio than wild-type (WT) plants. Three-week-old transgenic plants treated with 200 mM NaCl showed better growth than the WT with higher photosystem activity and less H(2)O(2) accumulation. Determination of endogenous pigments of Arabidopsis treated with 200 mM NaCl for 0, 2 or 4 d demonstrated that the transgenic plants retained higher contents of carotenoids than the WT. Furthermore, to compare the difference between SeLCY and AtLCY from Arabidopsis, we used viral vector mediating ectopic expression of SeLCY and AtLCY in Nicotiana benthamiana. Although LCY genes transformation increased the salt tolerance in tobacco, there is no significant difference between SeLCY- and AtLCY-transformed plants. These findings indicate that SeLCY transgenic Arabidopsis improved salt tolerance by increasing synthesis of carotenoids, which impairs reactive oxygen species and protects the photosynthesis system under salt stress, and as a single gene, SeLCY functionally showed no advantage for salt tolerance improvement compared with AtLCY.

Abstract:High salinity and nitrogen (N) deficiency in soil are two key factors limiting crop productivity, and they usually occur simultaneously. Here we firstly found that H+-PPase is involved in salt-stimulated NO3- uptake in the euhalophyte Salicornia europaea. Then, two genes (named SeVP1 and SeVP2) encoding H+-PPase from S.europaea were characterized. The expression of SeVP1 and SeVP2 was induced by salt stress and N starvation. Both SeVP1 or SeVP2 transgenic Arabidopsis and wheat plants outperformed the wild types (WTs) when high salt and low N occur simultaneously. The transgenic Arabidopsis plants maintained higher K+/Na+ ratio in leaves and exhibited increased NO3- uptake, inorganic pyrophosphate-dependent vacuolar nitrate efflux and assimilation capacity under this double stresses. Furthermore, they had more soluble sugars in shoots and roots and less starch accumulation in shoots than WT. These performances can be explained by the up-regulated expression of ion, nitrate and sugar transporter genes in transgenic plants. Taken together, our results suggest that up-regulation of H+-PPase favours the transport of photosynthates to root, which could promote root growth and integrate N and carbon metabolism in plant. This work provides potential strategies for improving crop yields challenged by increasing soil salinization and shrinking farmland. Improved oxidative tolerance in suspension-cultured cells of C4-pepctransgenic rice by H2O2 and Ca(2+) under PEG-6000

Abstract:To understand the molecular responses of PC (Overexpressing the maize C4-pepc gene, which encodes phosphoenolpyruvate carboxylase (PEPC)), to drought stress at cell level, we analyzed changes in the levels of signaling molecules (hydrogen peroxide (H2O2), calcium ion (Ca(2+)), and nitric oxide (NO)) in suspension-cultured PC and wild-type (WT) rice (Oryza sativa L.) cell under drought stress induced by 20% polyethylene glycol 6000 (PEG-6000). Results demonstrated that PC improved drought tolerance by enhancing antioxidant defense, retaining higher relative water content, survival percentages, and dry weight of cells. In addition, PEPC activity in PC under PEG treatment was strengthened by addition of H2O2 inhibitor, dimethylthiourea (DMTU) and NO synthesis inhibitor, 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO), respectively, while that in PC was weakened by addition of free calcium chelator, ethylene glycol-bis(b-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA) + calcium channel outflow inhibitor, ruthenium red (RR) + plasma membrane channel blocker La(NO3)3, but EGTA + RR did not. Results also showed that NO and Ca(2+) was lying downstream of H2O2 in drought-induced signaling. Calcium ion was also involved in the expression of C4-pepc in PC. These results suggested that PC could improve oxidative tolerance in suspension-cultured cells and the acquisition of this tolerance required downregulation of H2O2 and the entry of extracellular Ca(2+) into cells across the plasma membrane for regulation of PEPC activity and C4-pepc expression.
Overexpression of the Polygonum Cuspidatum PcDREB2A Gene Encoding a DRE-Binding Transcription Factor Enhances the Drought Tolerance of Transgenic Arabidopsis Thaliana

Abstract:Plants have evolved complex signaling networks that enable them to adapt to adverse environmental conditions. The dehydration-responsive element-binding (DREB) transcription factors are important for plant responses to abiotic stresses. In this study, a new member of the AP2/ERF transcription factor gene family, PcDREB2A, was cloned and characterized from Polygonum cuspidatum, a traditional Chinese medicinal herb. PcDREB2A, which includes a typical AP2 domain, was clustered in the A-2 subgroup of the DREB subfamily. At the seedling stage, PcDREB2A expression was induced by cold, salt, and drought stresses. A yeast one-hybrid assay and an analysis of transiently transformed tobacco revealed that PcDREB2A can specifically bind to the DRE motif and transactivate reporter gene expression. Following 200 and 250 mM mannitol treatments, the PcDREB2A-overexpressing Arabidopsis thaliana lines had longer roots and a significantly higher fresh weight than the wild-type plants. Furthermore, under drought stress conditions, the PcDREB2A-overexpressing A. thaliana plants accumulated less malondialdehyde than the control plants. These results indicate that PcDREB2A encodes a novel DREB transcription factor in P. cuspidatum. Furthermore, the data generated in this study may be useful for researchers and breeders interested in genetically engineering plants to increase drought tolerance without inhibiting growth.

Abstract:SUMMARY Salinity poses a significant challenge to plant growth and crop productivity by adversely affecting crucial processes, including photosynthesis. Efforts to enhance abiotic stress tolerance in crops have been hindered by the trade‐off effect, where increased stress resistance is accompanied by growth reduction. In this study, we identified and characterized a plastocyanin gene (PaPC) from the Antarctic moss Polytrichastrum alpinum, which enhanced photosynthesis and salt stress tolerance in Arabidopsis thaliana without compromising growth. While there were no differences in growth and salt tolerance between the wild type and Arabidopsis plastocyanin genes (AtPC1 and AtPC2)‐overexpressing plants, PaPC‐overexpressing plants demonstrated superior photosynthetic efficiency, increased biomass, and enhanced salt tolerance. Similarly, PaPC‐overexpressing rice plants exhibited improved yield potential and photosynthetic efficiency under both normal and salt stress conditions. Key amino acid residues in PaPC responsible for this enhanced functionality were identified, and their substitution into AtPC2 conferred improved photosynthetic performance and stress tolerance in Arabidopsis, tobacco, and tomato. These findings not only highlight the potential of extremophiles as valuable genetic resources but also suggest a photosynthesis‐based strategy for developing stress‐resilient crops without a growth penalty.

Constitutive and dark-induced expression of Solanum tuberosum phosphoenolpyruvate carboxylase enhances stomatal opening and photosynthetic performance of Arabidopsis thaliana

Abstract:The effect of constitutive and dark-induced expression of Solanum tuberosum phosphoenolpyruvate carboxylase (PEPC) on the opening state of stomata and photosynthetic performance in Arabidopsis thaliana plants was studied. Transcript accumulation analyses of the A. thaliana dark-induced (Din10 and Din6) and the Pisum sativum asparagine synthetase 2 promoters (Asn2) in transiently transformed tobacco leaves showed that Din10 promoter induced more DsRed accumulation in the dark compared to the other din genes. Overexpression of PEPC under the control of the constitutive enhanced CaMV 35S (p35SS) and dark-induced Din10 promoter in stably transformed A. thaliana plants increased the number of opened stomata in dark adapted leaves. Gas exchange measurements using A. thaliana plants transgenic for p35SS-PEPC and Din10-PEPC revealed a marked increase in stomatal conductance, transpiration, and dark respiration rates measured in the dark compared to wild-type plants. Moreover, measurement of CO(2) assimilation rates at different external CO(2) concentrations (C(a) ) and different light intensities shows an increase in the CO(2) assimilation rates in transgenic Arabidopsis lines compared to wild-type plants. This is considered as first step towards transferring the aspects of Crassulacean acid metabolism-like photosynthetic mechanism into C3 plants.
A CBL-interacting Protein Kinase TaCIPK27 Confers Drought Tolerance and Exogenous ABA Sensitivity in Transgenic Arabidopsis.

Abstract:Drought is one of the major environmental stresses to plants. The calcium sensor, calcineurin B-like (CBL) proteins, and their interacting protein kinases (CIPK) play important roles in responding to abiotic stresses. In this study, we functionally characterized a CIPK gene from Triticum aestivum designated TaCIPK27. The transcriptional levels of TaCIPK27 were increased both in roots and leaves after treatment with polyethylene glycol 8000, abscisic acid and H2O2. Besides, TaCIPK27 interacted with AtCBL1, AtCBL3, AtCBL4, AtCBL5 and AtCBL9 in yeast two-hybrid assays. Ectopic overexpression of TaCIPK27 positively regulates drought tolerance in transgenic Arabidopsis compared with controls, which was demonstrated by seed germination and survival rates experiments, as well as the detection of physiological indices including ion leakage, malonic dialdehyde and H2O2 contents and antioxidant enzyme activities under normal and drought conditions. Moreover, higher concentration of endogenous abscisic acid was detected under drought in TaCIPK27 transgenic plants. In addition, TaCIPK27 transgenic plants were more sensitive to exogenous abscisic acid treatment at seed germination and seedling stage. The expression levels of somedrought stress and abscisic acid related genes were up-regulated in TaCIPK27 transgenic plants. The results suggest that TaCIPK27 functions as a positive regulator under drought partly in an ABA-dependent pathway.
Joint expression of Zmpepc, Zmppdk, and Zmnadp-me is more efficient than expression of one or two of those genes in improving the photosynthesis of Arabidopsis

Abstract:This study assessed the effects of seven combinations of maize (Zea mays) genes phosphoenolpyruvate carboxylase (pepc), pyruvate phosphate dikinase (ppdk), and NADP-malic enzyme (nadp-me), on the photosynthesis of Arabidopsis. The photosynthetic rate, carboxylation efficiency, and shoot-dry-weight of Zmpepc (PC), Zmpepc + Zmppdk (PCK), Zmpepc + Zmnadp-me (PCM), and Zmpepc + Zmppdk + Zmnadp-me (PCKM) were significantly higher than those of the control wild-type (WT), with a trends to be PCKM > PCK > PC and PCM > WT. This indicated that Zmpepc was a prerequisite for improved photosynthetic performance; Zmppdk had a positive effect on Zmpepc, and the triple gene combination had the most significant synergistic effects. PCKM significantly enhanced activity of photosystem (PS)II (K, J phase) and PSI, light energy absorption (ABS/CSm) and conversion (TRo/ABS), and electron transfer (ETo/TRo). PCKM up-regulated 18 photosynthesis-related proteins, among which, 11 were involved in light reaction resulting in improved light-energy absorption and conversion efficiency, electron transfer, activity and stability of PSII and PSI, and the ATP and NADPH production. The remaining seven proteins were involved in dark reaction. The up-regulation of these proteins in PCKM improved the coordinated operation of light and dark reaction, increasing the photosynthesis and dry weight ultimately. These results also provide a promising strategy for the genetic improvement of the photosynthetic performance of C<sub>3</sub> crops by inserting major C<sub>4</sub> photosynthetic genes.

Abstract:To understand the adaptive mechanisms, and the strategy and direction of evolution of exploiting plants for living in oasis-desert transitional areas where there was a water gradient, mechanisms of C-4 photosynthesis and antioxidative protection were studied in leaves and assimilating shoots of C-3 desert shrub Hedysarum scoparium under water gradient accompanied with high irradiance and high temperature in simulated water gradient trial site that lies in the desert. In this study, C-4 pathway was detected in the leaves and assimilating shoots of H. scoparium and the antioxidative enzymatic system was found to be very important in the protection against oxidants, both increasing considerably antioxidative enzymes and C-4 photosynthetic enzymes under increasing water stress in C-3 plant. The results indicated that the C-4 photosynthetic response of assimilating shoots was more positive than that of leaves with water condition exacerbate. It was proposed that the direction of evolution of organs of C-4 photosynthesis and positive interaction between antioxidative enzymes and C-4 photosynthetic key enzymes in C-3 desert species were important factors for plant growth, survival and reproduction in water gradient environment.
Halophyte Nitraria billardieri CIPK25 promotes photosynthesis in Arabidopsis under salt stress

Abstract:The calcineurin B-like (CBL)-interacting protein kinases (CIPKs), a type of plant-specific genes in the calcium signaling pathway, function in response to adverse environments. However, few halophyte derived CIPKs have been studied for their role in plant physiological and developmental adaptation during abiotic stresses, which inhibits the potential application of these genes to improve environmental adaptability of glycophytes. In this study, we constructed Nitraria billardieri CIPK25 overexpressing Arabidopsis and analyzed the seedling development under salt treatment. Our results show that Arabidopsis with NbCIPK25 expression exhibits more vigorous growth than wild type plants under salt condition. To gain insight into the molecular mechanisms underlying salt tolerance, we profiled the transcriptome of WT and transgenic plants via RNA-seq. GO and KEGG analyses revealed that upregulated genes in NbCIPK25 overexpressing seedlings under salt stress are enriched in photosynthesis related terms; Calvin-cycle genes including glyceraldehyde-3-phosphate dehydrogenases (GAPDHs) are significantly upregulated in transgenic plants, which is consistent with a decreased level of NADPH (GAPDH substrate) and increased level of NADP + . Accordingly, NbCIPK25 overexpressing plants exhibited more efficient photosynthesis; soluble sugar and proteins, as photosynthesis products, showed a higher accumulation in transgenic plants. These results provide molecular insight into how NbCIPK25 promotes the expression of genes involved in photosynthesis, thereby maintaining plant growth under salt stress. Our finding supports the potential application of halophyte-derived NbCIPK25 in genetic modification for better salt adaptation.

Convergent molecular evolution of phosphoenolpyruvate carboxylase gene family in C 4 and crassulacean acid metabolism plants

Abstract:Phosphoenolpyruvate carboxylase (PEPC), as the key enzyme in initial carbon fixation of C 4 and crassulacean acid mechanism (CAM) pathways, was thought to undergo convergent adaptive changes resulting in the convergent evolution of C 4 and CAM photosynthesis in vascular plants. However, the integral evolutionary history and convergence of PEPC in plants remain poorly understood. In the present study, we identified the members of PEPC gene family across green plants with seventeen genomic datasets, found ten conserved motifs and modeled three-dimensional protein structures of 90 plant-type PEPC genes. After reconstructing PEPC gene family tree and reconciled with species tree, we found PEPC genes underwent 71 gene duplication events and 16 gene loss events, which might result from whole-genome duplication events in plants. Based on the phylogenetic tree of the PEPC gene family, we detected four convergent evolution sites of PEPC in C 4 species but none in CAM species. The PEPC gene family was ubiquitous and highly conservative in green plants. After originating from gene duplication of ancestral C3-PEPC, C4-PEPC isoforms underwent convergent molecular substitution that might facilitate the convergent evolution of C 4 photosynthesis in Angiosperms. However, there was no evidence for convergent molecular evolution of PEPC genes between CAM plants. Our findings help to understand the origin and convergent evolution of C 4 and CAM plants and shed light on the adaptation of plants in dry, hot environments.

Abstract:Abstract Background Phosphoenolpyruvate carboxylase (PEPC) plays an important role in the primary metabolism of higher plants. Several studies have revealed the critical importance of PEPC in the interaction of carbon and nitrogen metabolism. However, the function mechanism of PEPC in nitrogen metabolism is unclear and needs further investigation. Results This study indicates that transgenic rice expressing the sugarcane C4-PEPC gene displayed shorter primary roots and fewer crown roots at the seedling stage. However, total nitrogen content was significantly higher in transgenic rice than in wild type (WT) plants. Proteomic analysis revealed that there were more differentially expressed proteins (DEPs) responding to nitrogen changes in transgenic rice. In particular, the most enriched pathway “glutathione (GSH) metabolism”, which mainly contains GSH S-transferase (GST), was identified in transgenic rice. The expression of endogenous PEPC , GST and several genes involved in the TCA cycle, glycolysis and nitrogen assimilation changed in transgenic rice. Correspondingly, the activity of enzymes including GST, citrate synthase, 6-phosphofructokinase, pyruvate kinase and ferredoxin-dependent glutamate synthase significantly changed. In addition, the levels of organic acids in the TCA cycle and carbohydrates including sucrose, starch and soluble sugar altered in transgenic rice under different nitrogen source concentrations. GSH that the substrate of GST and its components including glutamic acid, cysteine and glycine accumulated in transgenic rice. Moreover, the levels of phytohormones including indoleacetic acid (IAA), zeatin (ZT) and isopentenyladenosine (2ip) were lower in the roots of transgenic rice under total nutrients. Taken together, the phenotype, physiological and biochemical characteristics of transgenic rice expressing C 4 -PEPC were different from WT under different nitrogen levels. Conclusions Our results revealed the possibility that PEPC affects nitrogen metabolism through regulating GST, which provide a new direction and concepts for the further study of the PEPC functional mechanism in nitrogen metabolism.

Abstract:Alteration in atmospheric carbon dioxide concentration and other environmental factors are the significant cues of global climate change. Environmental factors affect the most fundamental biological process including photosynthesis and different metabolic pathways. The feeding of the rapidly growing world population is another challenge which imposes pressure to improve productivity and quality of the existing crops. C4 plants are considered the most productive, containing lower photorespiration, and higher water-use & N-assimilation efficiencies, compared to C3 plants. Besides, the C4-photosynthetic genes not only play an important role in carbon assimilation but also modulate abiotic stresses. In this review, fundamental three metabolic processes (C4, C3, and CAM) of carbon dioxide assimilation, the evolution of C4-photosynthetic genes, effect of elevated CO2 on photosynthesis, and overexpression of C4-photosynthetic genes for higher photosynthesis were discussed. Kranz-anatomy is considered an essential prerequisite for the terrestrial C4 carbon assimilation, but single-celled C4 plant species changed this well-established paradigm. C4 plants are insensitive to an elevated CO2 stress condition but performed better under stress conditions. Overexpression of essential C4-photosynthetic genes such as PEPC, PPDK, and NADP-ME in C3 plants like Arabidopsis, tobacco, rice, wheat, and potato not only improved photosynthesis but also provided tolerance to various environmental stresses, especially drought. The review provides useful information for sustainable productivity and yield under elevated CO2 environment, which to be explored further for CO2 assimilation and also abiotic stress tolerance. Additionally, it provides a better understanding to explore C4-photosynthetic gene(s) to cope with global warming and prospective adverse climatic changes.

Populus euphratica Phospholipase Dδ Increases Salt Tolerance by Regulating K+/Na+ and ROS Homeostasis in Arabidopsis

Abstract:Phospholipase Dα (PLDα), which produces signaling molecules phosphatidic acid (PA), has been shown to play a critical role in plants adapting to salt environments. However, it is unclear whether phospholipase Dδ (PLDδ) can mediate the salt response in higher plants. PePLDδ was isolated from salt-resistant Populus euphratica and transferred to Arabidopsis thaliana to testify the salt tolerance of transgenic plants. The NaCl treatment (130 mM) reduced the root growth and whole-plant fresh weight of wild-type (WT) A. thaliana, vector controls (VC) and PePLDδ-overexpressed lines, although a less pronounced effect was observed in transgenic plants. Under salt treatment, PePLDδ-transgenic Arabidopsis exhibited lower electrolyte leakage, malondialdehyde content and H2O2 levels than WT and VC, resulting from the activated antioxidant enzymes and upregulated transcripts of genes encoding superoxide dismutase, ascorbic acid peroxidase and peroxidase. In addition, PePLDδ-overexpressed plants increased the transcription of genes encoding the plasma membrane Na+/H+ antiporter (AtSOS1) and H+-ATPase (AtAHA2), which enabled transgenic plants to proceed with Na+ extrusion and reduce K+ loss under salinity. The capacity to regulate reactive oxygen species (ROS) and K+/Na+ homeostasis was associated with the abundance of specific PA species in plants overexpressing PePLDδ. PePLDδ-transgenic plants retained a typically higher abundance of PA species, 34:2 (16:0–18:2), 34:3 (16:0–18:3), 36:4 (18:2–18:2), 36:5 (18:2–18:3) and 36:6 (18:3–18:3), under control and saline conditions. It is noteworthy that PA species 34:2 (16:0–18:2), 34:3 (16:0–18:3), 36:4 (18:2–18:2) and 36:5 (18:2–18:3) markedly increased in response to NaCl in transgenic plants. In conclusion, we suppose that PePLDδ-derived PA enhanced the salinity tolerance by regulating ROS and K+/Na+ homeostasis in Arabidopsis.

Abstract:It has previously been indicated that polyamines (PAs) are involved in the response to adverse environmental stresses in various plant species. Increased cellular PA levels or exogenous PA application can reduce the accumulation of reactive oxygen species (ROS) imposed by multiple abiotic stresses via increasing antioxidant enzyme activities. In this study, a novel spermidine synthase gene, designated as EsSPDS1, was cloned and characterized from the halophytic plant Eutrema salsugineum. Analyses of tissue-specific expression profiles revealed that EsSPDS1 was expressed in almost all tested organs including leaves, stems, flowers, roots and siliques. The expression of EsSPDS1 was obviously induced by polyethylene glycol (PEG) and high salinity treatments. Furthermore, the EsSPDS1 transcripts rapidly accumulated upon exposure to abscisic acid (ABA) treatments, implying that this gene was involved in ABA-mediated abiotic stress response. Transgenic tobacco plants harboring EsSPDS1 exhibited enhanced drought tolerance relative to wild-type (WT) plants. These transgenic plants had lower levels of malondialdehyde (MDA), less ion leakage (IL) and reactive oxygen species (ROS). However, relative water content (RWC) and activities of antioxidant enzymes including SOD and CAT of the transgenic plants was remarkably higher than those of WT plants under drought stress conditions. Collectively, our data suggested that the activation of EsSPDS1 expression in tobacco plants conferred drought tolerance, which might be attributed to enhance the ability to scavenge ROS levels.

Abstract:Phosphoenolpyruvate carboxylase (PEPC) plays a pivotal role in the photosynthetic CO2 fixation of C-4 plants. However, the functions of PEPCs in C-3 plants are less well characterized, particularly in relation to low atmospheric CO2 levels. Of the four genes encoding PEPC in Arabidopsis, PPC2 is considered as the major leaf PEPC gene. Here we show that the ppc2 mutants suffered a growth arrest when transferred to low atmospheric CO2 conditions, together with decreases in the maximum efficiency of PSII (F-v/F-m) and lower levels of leaf abscisic acid (ABA) and carbohydrates. The application of sucrose, malate, or ABA greatly rescued the growth of ppc2 lines under low CO2 conditions. Metabolite profiling analysis revealed that the levels of glycine and serine were increased in ppc2 leaves, while the abundance of photosynthetic metabolites was decreased under these conditions. The transcript levels of encoding enzymes involved in glycine or serine metabolism was decreased in ppc2 in an ABI5-dependent manner. Like the ppc2 mutants, abi5-1 mutants had lower photosynthetic rates and F-v/F-m compared with the wild type under photorespiratory conditions (i.e. low CO2 availability). However, the growth of these mutants was similar to that of the wild type under non-photorespiratory (low O-2) conditions. The constitutive expression of ABl5 prevented the growth arrest of ppc2 lines under low CO2 conditions. These findings demonstrate that PPC2 plays an important role in the acclimation of Arabidopsis plants to low CO2 availability by linking photorespiratory metabolism to primary metabolism, and that this is mediated, at least in part, through ABA- and ABI5-dependent processes.
