Abstract:Simultaneous delivery of therapeutic agents and energy by magnetic nanoparticles (MNPs) at targeted sites can boost cancer therapy and alleviate side effects. To achieve this goal, however, the magnetic fluid hyperthermia (MFH) usually exhibits the unsufficient thermal efficiency due to their narrow magnetization curves. Besides, an inappropriately large administration concentration also causes health deterioration as shown in an animal model. In this study, the core–shell cube that enhances the coercivity and magnetization related to single‐compositional MNPs by elaborately tuning their interface relaxation via the magnetocrystalline and surface anisotropy is developed. Néel and Brownian relaxation can be adjusted by the particles’ structures to maximize the hyperthermia efficacy upon an alternating‐magnetic‐field (AMF). Furthermore, temozolomide and lactoferrin‐coated CoFe2O4@Fe3O4 core–shell cubes are rapidly internalized by targeting cancer cells and penetrate into tumor spheroids while subjecting to AMF. The targeted cubes with the capabilities of enhanced coercivity, AMF‐induced drug penetration into tumors, and magnetothermal ablation for cancer therapy display potentials for clinical uses. The shape effects of core/shell on hard/soft magnetic materials reflected to their Brownian relaxation time (τB) and Néel relaxation time (τN), facilitating the controls of surface anisotropy (Ks), hyperthermia effects. Via conjugation of temozolomide (TMZ) and lactoferrin (Lf), the magnetic core–shell heterostructures facilitate the penetrative delivery to tumor spheroids, chemotherapy treatment efficacy, and intracellular mechanical destruction on a microfluidic chip.

Abstract:The adoption of magnetic hyperthermia as either a stand-alone or adjunct therapy for cancer is still far from being optimised due to the variable performance found in many iron oxide nanoparticle systems, including commercially available formulations. Herein, we present a reproducible and potentially scalable microwave-based method to make stable citric acid coated multi-core iron oxide nanoparticles, with exceptional magnetic heating parameters, viz. intrinsic loss parameters (ILPs) of up to 4.1 nH m(2) kg(-1), 35% better than the best commercial equivalents. We also probe the core-to-core magnetic interactions in the particles via remanence-derived Henkel and ΔM plots. These reveal a monotonic dependence of the ILP on the magnetic interaction field Hint, and show that the interactions are demagnetising in nature, and act to hinder the magnetic heating mechanism.
Multi-modal Mn-Zn Ferrite Nanocrystals for Magnetically-Induced Cancer Targeted Hyperthermia: a Comparison of Passive and Active Targeting Effects.

Abstract:The high performance and increased tumor-targeting accumulation of magnetic nanocrystals (MNCs) are the most important considerations in cancer targeted magnetic hyperthermia (TMH). To achieve these goals, our study was firstly done using well-established fluorescence/magnetic Mn-Zn ferrite MNCs (core size: 14 nm) as multi-modal imaging contrast agents and highly-efficient "heat generators", which were coated with a biocompatible PEG-phospholipid (DSPE-PEG2000) and further modified by a cyclic tripeptide of arginine-glycine-aspartic acid (RGD). By using a mouse model bearing breast carcinoma (4T1), we then systematically compared PEGylated MNCs (MNCs@PEG)- and RGD-PEGylated MNCs (MNCs@RGD)-mediated tumor targeting abilities by intravenous administration. The MNCs@PEG-based passive targeting could successfully accumulate at the tumor due to the enhanced permeability and retention (EPR) effects, but the non-targeted localization might make the MNCs@PEG "leaking" from larger pores of tumor fenestrated vascular networks. Our designed MNCs@RGD, simultaneously functionalized with PEG and RGD ligands, might promote a synergistic effect including efficient tumor vasculature active targeting and EPR-mediated passive targeting, improving total MNC concentration and retention time in tumor tissues. By combining fluorescence/magnetic resonance (MR)/thermal multi-modal imaging-guided diagnostics and continuous TMH treatment under an alternating current magnetic field (ACMF, 2.58 kA m(-1), 390 kHz), the tumor surface could be heated to approximately 43-44 °C based on the MNC-mediated repeated injections. Sufficient temperature elevation induced the apoptosis of tumor cells, and inhibited the tumor angiogenesis. Compared with MNCs@PEG, the active MNCs@RGD-based tumor targeting MR image was significantly more efficient due to both the higher and long-lasting tumor accumulation, but its antitumor efficacy was not obviously improved in the TMH treatments. To achieve a singularly promising tumor TMH therapy, a greatly increased MNC content in tumor was needed. This insight indicated that not only the tumor vasculature targeting, but also the active tumor cells targeting of MNCs should receive considerable attention in future clinical TMH therapy application.

Abstract:Hypothesis Multicore flower-like iron oxide nanoparticles (IONPs) are among the best candidates for magnetic hyperthermia applications against cancers. However, they are rarely investigated in physiological environments and their efficacy against cancer cells has been even less studied. The combination of magnetic hyperthermia, using multicore IONPs, with selected bioactive molecules should lead to an enhanced activity against cancer cells. Experiments Multicore IONPs were synthesized by a seeded-growth thermal decomposition approach. Then, the cytotoxicity, cell uptake, and efficacy of the magnetic hyperthermia approach were studied with six cancer cell lines: PANC1 (pancreatic carcinoma), Mel202 (uveal melanoma), MCF7 (breast adenocarcinoma), MB231 (triple-negative breast cancer line), A549 (lung cancer), and HCT116 (colon cancer). Finally, IONPs were modified with a chemotherapeutic drug (SN38) and tumor suppressor microRNAs (miR-34a, miR-182, let-7b, and miR-137), to study their activity against cancer cells with and without combination with magnetic hyperthermia. Findings Two types of multicore IONPs with very good heating abilities under magnetic stimulation have been prepared. Their concentration-dependent cytotoxicity and internalization have been established, showing a strong dependence on the cell line and the nanoparticle type. Magnetic hyperthermia causes significant cell death that is dramatically enhanced in combination with the bioactive molecules.

Abstract:Few-layer, carbon-coated, iron (C/Fe) magnetic nanoparticles (MNPs) were synthesized with controlled sizes ranging from 7 to 9 nm. The additional loading of two anti-cancer drugs, doxorubicin and erlotinib, was achieved through - stacking onto the carbon shells. Controlled release of the drugs was successfully triggered by radio frequency (RF) heating or pH variation. Based on the experimental results, C/Fe MNPs act as heat-inducing agents and are able to thermally destroy cancer cells when RF is applied. It was found that the combination of anti-cancer drugs (in particular a low dose of doxorubicin) and RF treatment demonstrates a synergistic effect in inducing cell death in pancreatic cancer cells. Our findings demonstrate that MNPs can be used as highly efficient multimodal nanocarrier agents for an integrated approach to cancer treatment involving triggered delivery of antineoplastic drugs and RF-induced thermal therapy.

Nanomagnetic Actuation of Hybrid Stents for Hyperthermia Treatment of Hollow Organ Tumors

Abstract:This paper describes a magnetic nanotechnology that locally enables hyperthermia treatment of hollow organ tumors by using polymer hybrid stents with incorporated magnetic nanoparticles (MNP). The hybrid stents are implanted and activated in an alternating magnetic field to generate therapeutically effective heat, thereby destroying the tumor. Here, we demonstrate the feasibility of nanomagnetic actuation of three prototype hybrid stents for hyperthermia treatment of hollow organ tumors. The results show that the heating efficiency of stent filaments increases with frequency from approximately 60 W/gFe (95 kHz) to approximately 250 W/gFe (270 kHz). The same trend is observed for the variation of magnetic field amplitude; however, heating efficiency saturates at approximately 30 kA/m. MNP immobilization strongly influences heating efficiency showing a relative difference in heating output of up to 60% compared to that of freely dispersed MNP. The stents showed uniformly distributed heat on their surface reaching therapeutically effective temperatures of 43 °C and were tested in an explanted pig bile duct for their biological safety. Nanomagnetic actuation of hybrid stents opens new possibilities in cancer treatment of hollow organ tumors.
Low Dimensional Assemblies of Magnetic MnFe$_2$O$_4$ Nanoparticles and Direct In Vitro Measurements of Enhanced Heating Driven by Dipolar Interactions: Implications for Magnetic Hyperthermia

Abstract:Magnetic fluid hyperthermia (MFH), the procedure of raising the temperature of tumor cells using magnetic nanoparticles (MNPs) as heating agents, has proven successful in treating some types of cancer. However, the low heating power generated under physiological conditions makes necessary a high local concentration of MNPs at tumor sites. Here, we report how the in vitro heating power of magnetically soft MnFe$_2$O$_4$ nanoparticles can be enhanced by intracellular low-dimensional clusters through a strategy that includes: a) the design of the MNPs to retain N\'eel magnetic relaxation in high viscosity media, and b) culturing MNP-loaded cells under magnetic fields to produce elongated intracellular agglomerates. Our direct in vitro measurements demonstrated that the specific loss power (SLP) of elongated agglomerates ($SLP=576\pm33$ W/g) induced by culturing BV2 cells in situ under a dc magnetic field was increased by a factor of 2 compared to the $SLP=305\pm25$ W/g measured in aggregates freely formed within cells. A numerical mean-field model that included dipolar interactions quantitatively reproduced the SLPs of these clusters both in phantoms and in vitro, suggesting that it captures the relevant mechanisms behind power losses under high-viscosity conditions. These results indicate that in situ assembling of MNPs into low-dimensional structures is a sound possible way to improve the heating performance in MFH.

Abstract:Alternative cancer treatments such as photothermal therapy (PTT) and magnetic hyperthermia (MHT) techniques have been studied to show promising potential as supplementary modalities. However, such techniques have their own limitations; for instance, highly concentrated intratumoral injections of magnetic nanoparticles are required to compensate their low specific loss power under safe and low magnetic field intensity for the MHT, while the PTT has limitations in the treatment of deep-seated tumors due to low light penetration. Here, the decoration of the multi-walled carbon nanotube (MWCNT) surface by magnetic nanoparticles (≈8.5 nm) was achieved by a hydrothermal method and the development of MWCNT/Mn0.5Zn0.5Fe2O4 (MZFC) hybrids for magneto-photothermal dual-mode cancer therapy. The obtained specific loss power of the MZFC hybrids is found to be at least 1 order of magnitude higher, with improvement from ∼19 W/g to 225 W/g, under the excitation of both an AMF with a magnetic field intensity of 6.4 kA/m and a frequency of 300 kHz and a simultaneous NIR laser of 0.5 W/cm2 irradiation. The synergistic utilization of the photothermal and magnetic properties of MZFC effectively diminishes the required magnetic field amplitude and NIR laser power density. Our in vitro cell experiments confirmed that the thermal effects mediated by the MZFC after endocytosis delivered enhanced cytotoxicity in the presence of dual excitation of NIR laser and AMF. These findings indicate that MZFC nanohybrids possess significant potential as targeted nanoheating agents for hyperthermia applications.

Abstract:Here, magnetic nanostructures made of chain‐like iron oxide nanocubes and photothermal dyes enwrapped in amphiphilic polymers, are developed. The obtained nanostructures are eradicating, in vitro, thermo‐resistant glioblastoma cells at temperature as low as 41 °C only when this therapeutic temperature is reached by a combination of MH and PTT of clinical conditions, thus offering new alternative approaches to treat thermo‐resistant cancer. A self‐assemble amphiphilic diblock copolymer that can incorporate iron oxide nanocubes (IONCs) in chain‐like assemblies as heat mediators for magnetic hyperthermia (MHT) and tuneable amounts of IR780 dye as agent for photothermal therapy (PTT) is developed. MHT‐heating performance of photobeads in viscous media have the same heat performances in water at magnetic field conditions of clinical use. Thanks to IR780, the photobeads are activated by infrared laser light within the first biological window (808 nm) with a significant enhancement of photo‐stability of IR780 enabling the raise of the temperature at therapeutic values during multiple PTT cycles and showing unchanged optical features up to 8 days. Moreover, the photobeads fluorescent signal is preserved once internalized by glioblastoma multiforme (GBM) cells. Peculiarly, the photobeads are used as toxic agents to eradicate thermo‐resistant GBM cells at mild heat, as low as 41 °C, with MHT and PTT both of clinical use. Indeed, a high U87 GBM cell mortality percentage is obtained only with dual MHT/PTT while each single treatment dose not provide the same cytotoxic effects. Only for the combined treatment, the cell death mechanism is assigned to clear sign of apoptosis as observed by structural/morphological cell studies and enhanced lysosome permeability.

Abstract:An effective magnetic nanocrystals (MNCs)-mediated theranostics strategy as a combination of simultaneous diagnostics and heating treatment of tumors by using magnetic resonance imaging (MRI) and alternating current magnetic field (ACMF) is successfully developed. In this strategy, we had firstly synthesized a well-established Mn–Zn ferrite MNCs coated with PEG-phospholipids (1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol copolymers, DSPE-PEG2000). The monodisperse PEGylated MNCs with core–shell structure (15 nm) exhibited excellent performance, such as high magnetism of 98 emu g−1 Fe, relaxivity coefficient (r2) of 338 mm−1 s−1, and specific absorption rate (SAR) value of 324 W g−1 Fe. It was proved that the obtained MNCs with an average diameter of 48.6 nm can drastically minimize the recognition and phagocytosis of macrophages, simultaneously improve their biocompatibility in vitro. These advantages endowed them with efficient passive targeting ability in vivo for prominent tumor MRI and magnetically induced heating when exposed to ACMF, based on enhanced permeability and retention (EPR) effects. To ensure sufficient accumulation of MNCs within tumors for targeted hyperthermia, we described the use of MNCs with a well-tolerated intravenous single dose of 18 mg Fe/kg mouse body weight, achieving repeatedly injection and hyperthermia within a subcutaneous breast cell carcinoma mouse model. With an ACMF of 12 A at 390 kHz, the tumor surface sites could be heated to approximately 43 °C in 30 min based on MNCs-mediated intravenous injections. The long-lasting hyperthermia could effectively induce the apoptosis of tumor cells, inhibit the angiogenesis of tumor vessels, and finally suppress the tumor growth within a certain period of time.
Optimizing the magnetic field strength and concentration of silica coated cobalt ferrite nanoparticles for magnetic hyperthermia

Abstract:A therapeutic breakthrough in cancer treatment has recently been made by using magnetic nanoparticles (MNPs) for heating in hyperthermia therapy. Unapproachable tumors are being effectively destroyed by focused heat produced by MNPs. The major challenges regarding practical application of this therapy include the control and improvement of induction heating ability of MNPs and hyperthermia temperature range between 42–47 °C, for secure treatment at targeted area. Here, in this study, we have reported the synthesis and characterization of core–shell structured silica-coated cobalt ferrite nanoparticles (SiO2 coated CoFe2O4 MNPs), which are potential candidates for use as heat source in magnetic hyperthermia therapy. The SiO2 coated CoFe2O4 MNPs were synthesized using the reverse micelle method, with the SiO2 coating performed simultaneously during MNP synthesis. Various analytical tools were utilized for the characterization. The structural measurements were probed by X-ray diffraction (XRD). Monodisperse MNPs with nearly spherical core–shell structure was revealed by Transmission electron microscopy (TEM) results. The average diameter of MNPs obtained from TEM analysis was 15 nm. The surface coating of cobalt ferrite MNPs with silica was verified by Fourier transform infrared spectrometry (FTIR). The saturation magnetization values obtained using vibrating sample magnetometer (VSM) measurements were 45.74 emu/g, revealing the superparamagnetic nature of the nanoparticles. The heating efficiency of different concentrations of synthesized nanoparticles was evaluated in an aqueous solution under an alternating magnetic field of strength 5.5 kA/m at a frequency of 260 kHz. A saturation temperature of 42 °C was attained at an optimum concentration of 1.7 mg/mL, while a magnetic field strength of 3.9 kA/m achieved 42 °C at a fixed concentration of 3.5 mg/ml. The obtained specific absorption rate (SAR) values for all the samples were between 56.23 to 100.1 W/g. The correlation between SAR values, MNP concentration, and applied magnetic field strength was also examined. The high saturation magnetization, efficient heating rates, and high SAR values make our synthesized silica-coated cobalt ferrite nanoparticles promising candidates for magnetic hyperthermia treatments, potentially improving therapeutic outcomes for cancer patients.

Controlled Synthesis and Surface Modification of Magnetic Nanoparticles with High Performance for Cancer Theranostics Combining Targeted MR Imaging and Hyperthermia

Abstract:Magnetic nanoparticles (MNPs) are of great scientific interest for the researchers from a wide range of biomedicine applications, including clinical magnetic resonance imaging (MRI), cancer magnetic hyperthermia, triggered drug release, and catalysis. Especially, the high performance of these MNPs, such as controllable size and morphology, superior magnetism, high magnetically induced heating effects, favorable biocompatibility, accurate targeting ability, and long circulation, is crucial for their effective application in clinical diagnosis and therapy of disease. In a brief overview, the controlled chemical design and synthesis, the formation mechanism, as well as the surface functional strategy of MNPs are firstly presented. High-quality MNPs are typically prepared through a thermal decomposition of organometallic compounds in high-boiling organic solvent containing surfactants, in which the shape, size, composition, surface states, magnetic characteristics, biocompatibility, and some other important features are synthetically controlled. Understanding the interplay of these parameters is critical for the effective imaging and therapeutics, which include MRI relaxivity, heat emission, and attractive forces. We then discuss approaches to constructing versatile MNP probes as MRI contrast agents with high sensitivity, with which the sensitive and target-specific observation of biological imaging at the molecular and cellular levels is possible. Finally, we present an overview of recent breakthroughs in the development of MNP-based magnetic hyperthermia. The purpose of this section is to address different factors that affect the targeting, heating, and biodistribution to safely control the therapeutic efficacy of targeted magnetic hyperthermia (TMH).

Abstract:Iron Oxide Nanoparticles (IONPs) hold the potential to exert significant influence on fighting cancer through their theranostics capabilities as contrast agents (CAs) for magnetic resonance imaging (MRI) and as mediators for magnetic hyperthermia (MH). In addition, these capabilities can be improved by doping IONPs with other elements. In this work, we report the synthesis and characterization of single‐core and alloy ZnFe novel magnetic nanoparticles (MNPs), with improved magnetic properties and more efficient magnetic‐to‐heat conversion. Remarkably, our results challenge classical nucleation and growth theories, which cannot fully predict the final size/shape of these nanoparticles and, consequently, their magnetic properties, implying the need for further studies to better understand the nanomagnetism phenomenon. On the other hand, leveraging the enhanced properties of these new NPs, successful tumor therapy by MH was achieved following their intravenous administration and tumor accumulation via the EPR effect. Notably, these results were obtained using a single low dose of MNPs and a single exposure to clinically suitable alternating magnetic fields (AMF). Therefore, as far as we are aware, we demonstrate, for the first time, the successful application of intravenously administered MNPs for MRI‐tracked MH tumor therapy in passively targeted tumor xenografts using clinically suitable conditions. Multi‐Chambered Core/Shell Supraparticles for Real‐Time, Full‐Time Diagnosis and Treatment Integration of Tumors

Abstract:Multi‐chambered core/shell magnetic nanoparticles are developed as personalized and precise nanocarriers to provide real‐time, full‐time diagnosis and treatment integration of tumors. The supraparticles possess unique properties of hierarchical structure and full‐time release ability, showing in vitro precise uptake against cancer cells and in vivo multimodal imaging capability. To a certain extent, theranostic nanoplatforms promote tumor treatment efficiency. However, timely monitoring of the critical stages and signal sustainability of the entire process is challenging. In this study, multi‐chambered core/shell magnetic nanoparticles (MC‐MNPs) as drug and imaging agent multi‐loaded nanocarriers with a synergistic release function are reported. Supraparticles with stable chambers are formed by the supercooling self‐assembly of several core/shell magnetic nanoparticles composed of amphiphilic copolymers as the core and hydrophilic magnetic iron oxide nanoparticles as the shell. Desalinized doxorubicin and coumarin 6 are stored in different cavities of nanocarriers, and chitosan is used as an outer encapsulation layer. Based on their construction properties, MC‐MNPs can exhibit gradient‐degraded and steady‐released controllability in the tumor environment. Furthermore, real‐time accumulation situations and full‐time diagnostic signals of nanocarriers are thoroughly demonstrated using fluorescence imaging and T2‐weighted magnetic resonance imaging before and after magnetic hyperthermia in targeted tumors under an alternating magnetic field. Thus, MC‐MNPs as theranostic nanocarriers exhibit great potential for the timely monitoring and full‐time guidance of tumor treatment.

Abstract:Cells of the obligate halophile Halobacterium cutirubrum were grown to the stationary phase in the presence of 32P-orthophosphate, 35S-sulfate, or 14C-labelled precursors (acetate, malonate, mevalonate, glycerol). 32P was incorporated into the phosphatide components in proportion to their relative concentrations, thus: phosphatidyl glycerophosphate (diether analogue) > unidentified minor phosphatide > phosphatidyl glycerol (diether analogue). 35S-Sulfate was incorporated mostly into the glycolipid sulfate ester component, but small amounts of 35S also appeared in a more polar sulfolipid and in a minor unidentified sulfolipid.14C-Mevalonate was the most efficient precursor for the phytanyl chains, followed in decreasing order by acetate and glycerol. Only traces of 14C from the malonate precursor were incorporated, and these appeared entirely in the phytanyl groups. Critical Parameters to Improve Pancreatic Cancer Treatment Using Magnetic Hyperthermia: Field Conditions, Immune Response, and Particle Biodistribution

Abstract:Magnetic hyperthermia (MH) was used to treat a murine model of pancreatic cancer. This type of cancer is generally characterized by the presence of dense stroma that acts as a barrier for chemotherapeutic treatments. Several alternating magnetic field (AMF) conditions were evaluated using three-dimensional (3D) cell culture models loaded with magnetic nanoparticles (MNPs) to determine which conditions were producing a strong effect on the cell viability. Once the optimal AMF conditions were selected, <i>in vivo</i> experiments were carried out using similar frequency and field amplitude parameters. A marker of the immune response activation, calreticulin (CALR), was evaluated in cells from a xenograft tumor model after the MH treatment. Moreover, the distribution of nanoparticles within the tumor tissue was assessed by histological analysis of tumor sections, observing that the exposure to the alternating magnetic field resulted in the migration of particles toward the inner parts of the tumor. Finally, a relationship between an inadequate body biodistribution of the particles after their intratumoral injection and a significant decrease in the effectiveness of the MH treatment was found. Animals in which most of the particles remained in the tumor area after injection showed higher reductions in the tumor volume growth in comparison with those animals in which part of the particles were found also in the liver and spleen. Abstract:For in vivo magnetic hyperthermia tests, which are typically conducted on small animal models, one of the objectives is the design of alternating current (AC) magnetic field applicators able to guarantee an effective activation of magnetic nanoparticles (MNPs). During therapy application, it is critical to optimize heat deposition due to MNPs and minimize side effects in healthy tissues. For an accurate treatment planning, it is required to carefully select the geometry of the applicator coils and their location with respect to the body, as a function of the position and size of the tumour target region. Additionally, one should preliminary estimate the impact of experimental conditions on the MNP heating efficiency and thus on their capability to induce a temperature increase in tissues. Biophysical constraints have also to be taken into account in the choice of AC magnetic field parameters (frequency and amplitude), to avoid eddy current effects as much as possible. In this study, we present realistic simulations of preclinical tests on a mouse model, evaluating thermal response under various experimental conditions. We investigate different field applicator configurations, including helical, Helmholtz and pancake coils, while also analysing the role of the amplitude and frequency of the supply current, as well as of the type and administered dose of MNPs. The temperature increase in tissues, resulting from the heating effects due to AC magnetic field exposure and MNP activation, is calculated by means of in-house finite element models that solve the low-frequency electromagnetic field problem and the bioheat transfer equation. This in silico approach, which is applicable to any type of field applicators and MNPs, has been demonstrated to provide useful insights for the optimization of in vivo experiments, enabling the design of safer and more effective treatments.

Abstract:Cancer comprehensive treatment has been fully recognized as it can provide an effective multi-modality approach for fighting cancers. This work evaluates the effects of a kind of novel solar-planet structured magnetic nanocomposites (MNCs) for magnetic thermochemotherapy. Amino silane coated magnetic nanoparticles (MNPs) as agent of magnetic mediated hyperthermia (MMH) for cancer treatment were prepared by the chemical precipitation method. Docetaxel (an anticancer drug) loaded polymeric nanoparticles (DNPs) composed of carboxylic-terminated poly (D,L-lactic-co-glycolic acid) (PLGA) with Vitamin E TPGS as emulsifier for sustained drug release were prepared by a modified solvent extraction/evaporation technique. Furthermore, the MNPs modified with amino groups could be covalently attached to the surface of carboxylic terminated DNPs to form the so-called solar-planet structured MNCs by 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide (NHS) crosslinking. The prepared solar-planet structure has been confirmed by fluorescent observation. Inductive heating property of the nanocomposite was evaluation by monitoring the temperature increase of the MNCs suspension under alternative magnetic field (AMF). Drug encapsulation efficacy and drug release of the magnetic nanocomposite were conducted by high performance liquid chromatography (HPLC). In vitro evaluation of the novel nanocomposite as mediator for thermochemotherapy was conducted on the U251 human glioma cells and the synergistic effect between MMH and docetaxel chemotherapy was confirmed. All the observation supports that solar-planet structured MNC is a novel and effective mediator for magnetic thermochemotherapy. The MNCs can realize cancer comprehensive treatment thus has great potential in clinical application.

Abstract:Nanoparticle-mediated thermotherapeutic research strives innovative, multifunctional, efficient, and safe treatments. Our study introduces a novel nanoplatform: the hollow magnetic vortex nanorings within a polydopamine layer (HMVNp), which exhibit dual functionality as magnetic and photothermal agents. Utilizing a "Dual-mode" approach, combining an alternating magnetic field (AMF) with near-infrared (NIR) laser irradiation, HMVNp demonstrated a significant enhancement in heating efficacy (58 ± 8 %, SAR = 1441 vs 1032 W/g) over traditional solid magnetite nanoparticles coated with polydopamine (SMNp). The unique geometry larger surface area to volume ratio facilitates efficient magnetic vortex dynamics and enhanced heat transfer. Addressing the challenge of heat resistant heat shock protein (Hsp) expression, encapsulated quercetin (Q) within HMVNp leverages tumor acidity and dual-mode thermal therapy to enhance release, showing a 28.8 ± 6.81 % increase in Q loading capacity compared to traditional SMNp. Moreover, HMVNp significantly improves contrast for both magnetic resonance imaging (MRI) and photoacoustic imaging (PAI), with an approximately 62 % transverse relaxation (R2 = 81.5 vs 31.6 mM -1 s −1 [Fe]). In vivo studies showed that while single treatments slowed tumor growth, dual-mode therapy with quercetin significantly reduced tumors and effectively prevented metastases. Our study highlights the potential of HMVNp/Q as a versatile agent in thermotherapeutic interventions, offering improved diagnostic imaging capabilities.

Abstract:Currently, one of the main causes of death in the world is cancer; therefore, it is urgent to obtain a precocious diagnosis, as well as boost research and development of new potential treatments, which should be more efficient and much less invasive for the patient. Magnetic hyperthermia (MH) is an emerging cancer therapy using nanoparticles, which has proved to be effective when combined with chemotherapy, radiotherapy and/or surgery, or even by itself, depending on the type and location of the tumor's cells. This article presents the results obtained by using a previously developed economic homemade hyperthermia device with different types of magnetite nanoparticles, with sizes ranging between 12 ± 5 and 36 ± 11 nm and presenting different shapes (spherical and cubic particles). These magnetic nanoparticles (MNPs) were synthesized by three different methods (co-precipitation, solvothermal and hydrothermal processes), with their final form being naked, or possessing different kinds of covering layers (polyethylene glycol (PEG) or citric acid (CA)). The parameters used to characterize the heating by magnetic hyperthermia, namely the Specific Absorption Rate (SAR) and the intrinsic loss power (ILP), have been obtained by two different methods. Among other results, these experiments allowed for the determination of which synthesized MNPs showed the best performance concerning hyperthermia. From the results, it may be concluded that, as expected, the shape of MNPs is an important factor, as well as the time that the MNPs can remain suspended in solution (which is directly related to the concentration and covering layer of the MNPs). The MNPs that gave the best results in terms of the SAR were the cubic particles covered with PEG, while in terms of total heating the spherical particles covered with citric acid proved to be better.
