Abstract:The rapid development of the biopharmaceutical industry has gradually shifted the focus of R&D to integrated continuous manufacturing. In this study, the effects of residence time and protein concentration on the performance of twin-column periodic counter-current chromatography for continuous antibody capture with new affinity resin Praesto Jetted A50 were evaluated. The results showed that the continuous process exhibited better performance with higher productivity, higher capacity utilization and lower buffer consumption when compared with the batch process. Meanwhile, the increase of protein concentration could increase productivity, while it showed minor impact on capacity utilization and buffer consumption. Furthermore, monoclonal antibody (mAb) continuous capture from clarified cell culture supernatant was investigated. The results showed that the qualities of the products obtained from the continuous and batch processes were similar. The performance of the continuous capture process at 1 min residence time was significantly better than the batch process with the improvements in productivity (37.5 g/L/h) and capacity utilization (77.8%) as well as reducing buffer consumption by 60%. Finally, effects of Protein A resins on the continuous process were discussed, and the results indicated that the Protein A resins with higher dynamic binding capacity at low residence time would be more suitable for continuous processes. In general, continuous capture under the suitable operation conditions shows good performance and has great potential for mAb production.

Abstract:Antibody, especially monoclonal antibody has become an important therapeutic drug which has a broad market demand and prospect.Nowadays the bottleneck of antibody production has shifted from antibody expression to downstream processing.The typical antibody purification process starts with protein A affinity chromatographic to capture the antibody and includes two subsequent polishing steps for impurity removal, such as ionic exchange, hydrophobic interaction or hydroxyapatite chromatography.There are some limitations restricting the development in antibody industry.Therefore, the researchers are committed to develop novel purification methods, including mixed-mode chromatography (MMC) and short peptide biomimetic chromatography which remedy the weakness of traditional methods.According to the recent advances in the purification of monoclonal antibody drugs, protein A affinity chromatography and the novel chromatographic methods and their applications in monoclonal antibody purification are reviewed in the present article.
Process design and application of twin-column continuous chromatography for antibody affinity separation

Abstract:In order to improve process throughput and reduce operation cost of Protein A affinity chromatography, a twin-column continuous chromatography method was studied for monoclonal antibody (mAb) separation. Effects of residence time in the column and loading protein concentration were investigated, and the separation conditions were optimized for mAb capture from cell culture broth. Two typical Protein A affinity resin (MabSelect SuRe and UniMab) were used and the breakthrough curves were measured under different residence times and IgG concentrations, and their effects on the productivity, resin capacity utilization and buffer consumption were evaluated. The results indicate that the twin-column continuous chromatography method has higher productivity and capacity utilization, and lower buffer consumption when comparing to traditional batch chromatography at low residence times. The separation performance of continuous chromatography for mAb capture from cell culture broth was similar to batch chromatography with purity > 97%, yield > 87%, aggregates < 2% and host cell proteins < 300 ppm. However, the capacity utilization increased from 20% ~30% to 70% ~80%, and the buffer consumption reduced to 1/5~1/4 of traditional batch processes. The results demonstrate that the twin-column continuous chromatography method shows great potential for the mAb production.

Abstract:Frontal chromatography has seen increased interest for protein purification, in particular as a polishing step in downstream processes for therapeutic proteins production, as for example the purification of monoclonal antibodies (mAbs) from high molecular weight impurities, e.g., aggregates, using cation exchange resins. In this work we introduce a new two-column continuous process implementing frontal chromatography. The design procedure and its performance, compared to classical batch technology, are discussed. This represents an additional option in the realisation of optimised continuous downstream processing of therapeutic proteins.

Abstract:The increasing demand for efficient and robust processes in the purification of monoclonal antibodies (mAbs) has recently brought frontal chromatography to the forefront. Applied during the polishing step, it enables the removal of high molecular weight aggregates from the target product, achieving high purities. Typically, this process is operated in batch using a single column, which makes it intrinsically subjected to a purity-yield tradeoff. This means that high purities can only be achieved at the cost of lowering the product yield and vice versa. Recently, a two-column continuous implementation of frontal chromatography, referred to as Flow2, was developed. Despite being able of alleviating the purity-yield tradeoff typical of batch operations, the increase in the number of process parameters complicates its optimal design, with the risk of not exploiting its full potential. In this study, we developed an ad hoc design procedure (DP) suitable for the optimization of both batch frontal chromatography and Flow2 in terms of purity, yield, and productivity. This procedure provided similar results as a multiobjective optimization based on genetic algorithm but with lower computational effort. Then, batch and Flow2 operated at their optimal conditions were compared. Besides showing a more favorable Pareto front of yield and productivity at a specified purity, the Flow2 process demonstrated improved robustness compared to the batch process with respect to modifications in the loading linear velocity, washing buffer ionic strength and loading time, thus providing an appealing operation for integrated processes.
A Novel Dextran-Grafted Tetrapeptide Resin for Antibody Purification

Abstract:Short peptide biomimetic affinity chromatography as a novel antibody separation chromatography is a potential alternative to protein A chromatography. However, if directly attaching ligand to matrix, the adsorption capacity and mass transfer rate would be affected by pore blockage and steric effect. Grafting resin is an effective method to solve this problem by using polymer as a bridge between matrix and ligand. In this work, a novel resin was prepared by grafting a tetrapeptide to the dextran-grafted matrix. Then, the adsorption properties for human immunoglobin G and BSA were determined. The results showed the saturation adsorption capacity could reach to 133 mg/g resin at pH 8.9 with a significantly low dissociation constant (0.03 mg/mL). The influence of flow rates to dynamic binding capacity of this resin was less than that of the non-grafted resin. The separation performance of the resin showed monoclonal antibody could be well isolated from the Chinese hamster ovary culture supernatant at pH 9.0 with the purity of 93.0% and yield of 84.7% by one step. Overall, this resin could achieve higher binding capacity by the possible of gaining higher ligand density, indicating its potential significance for separation in larger scale systems.

Abstract:Peptide ligands have been developed as powerful alternatives to Protein A for antibody purification. In this study, a tetrapeptide library with two aromatic residues, one arginine and one aromatic/aliphatic residue (Aro-Aro-R-Aro/Ali) was constructed considering molecular interactions and applications. The library had 128 peptide ligands and was screened by flexible docking and molecular dynamics simulation. The results indicated that Ac-YFRH was the best ligand, and then was coupled onto an aminated argarose matrix. The results of isotherm adsorption showed that the binding capacity of human immunoglobulin G (IgG) was high at pH 7-8 and low at pH 4-5. The IgG adsorption also showed obvious salt-independence, and the addition of salt could reduce unspecific adsorption caused by electrostatic interaction. After the optimization of pH and salt addition, high IgG purity of 98.4% could be obtained with the recovery of 89.4% from BSA-containing feedstock. The purification of monoclonal antibody from CHO cell culture supernatant was tested, and the purity and recovery reached 98.0% and 79.5%, respectively. The results demonstrated that the new resin with the tetrapeptide YFRH ligand had high affinity and flexible operation window for IgG purification. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Affinity biomimetic chromatography is a novel bioseparation technology for the biological active substance, particularly showing good performance for antibody purification with the advantages of low cost, stable ligand structure, high specificity and so on. The critical point of affinity biomimetic chromatography is the specially-designed biomimetic ligand. The biomimetic ligands used currently include chemical synthetic ligand and short peptide ligand, which are normally obtained by rational design and screening. For a particular target protein, instead of selecting the ligand randomly, it is necessary to build a reasonable ligand library by rational designing method and screen the suitable mimetic ligand by high-throughput screening technology. According to the recent advances in affinity biomimetic chromatography, the designing and screening of ligands used for affinity mimetic chromatography and the applications in antibody purification are reviewed in the present article.
A high-throughput approach to developing and optimizing mixed-mode membrane chromatography for protein purification

Abstract:Membrane chromatography has been established as a viable alternative to packed-bed column chromatography for the purification of therapeutic proteins. Purification via membrane chromatography offers key advantages, including higher productivity and reduced buffer usage. Unlike column chromatography purification, the utilization of high-throughput screening in order to reduce development times and material requirements has been a challenge for membrane chromatography. This research focused on the development of a new, high-throughput screening technique for use in screening membrane chromatography conditions for monoclonal antibody purification. The developed screen utilizes a 96-well plate format, thereby allowing for the screening of multiple different membrane conditions at once. For this study, four mixed-mode cation exchange membranes and one cation exchange membrane were evaluated on the plate. The screen is performed in a similar manner to that of a resin slurry plate screen, however, instead of a single loading step, the antibody feed was loaded in 50 mg/ml increments up to a maximum loading of 450 mg/ml. Performing a similar, incremental loading on a resin plate would be impractical, as mixing times are substantially longer due to pore diffusion limitations. However, due to the significantly faster rate of mass transfer for membranes relative to resin, mixing times could be reduced by up to a factor of sixty on the membrane plate. Additional optimization showed that higher hydrophobicity can potentially lead to slower kinetics and mixing times that may need to be adjusted accordingly. The end result is a screen that has been proven to provide results comparable to those obtained on larger-scale membrane purification runs while also enabling exploration of a much greater operating space and significantly reducing the feed materials required.

Abstract:Monoclonal antibodies are conquering the biopharmaceutical market because they can be used to treat a variety of diseases. Therefore, it is very important to establish robust and optimized processes for their production. In this article, the first step of chromatography (Protein A chromatography) in monoclonal antibody purification was optimized with a focus on the critical elution step. Therefore, different buffers (citrate, glycine, acetate) were tested for chromatographic performance and product quality. Membrane chromatography was evaluated because it promises high throughputs and short cycle times. The membrane adsorber Sartobind® Protein A 2 mL was used to accelerate the purification procedure and was further used to perform a continuous chromatographic run with a four-membrane adsorber-periodic counter-current chromatography (4MA-PCCC) system. It was found that citrate buffer at pH 3.5 and 0.15 M NaCl enabled the highest recovery of >95% and lowest total aggregate content of 0.26%. In the continuous process, the capacity utilization of the membrane adsorber was increased by 20%.

Abstract:A 10-year retrospective review was conducted of incidental adnexal neoplasms found at the time of cesarean section. During the 10-year period between July 1,1976 and June 30, 1986, 17,902 cesarean sections were performed at Women's Hospital of the Los Angeles County/University of Southern California Medical Center. Adnexal neoplasms were identified with an incidence of one neoplasm per 197 cesarean sections. When a neoplasm is discovered during surgery, conservative surgery is recommended.

Abstract:In this study we compare a nanofiber device and a resin column, both with the same Protein A ligand, for the capture of four different monoclonal antibodies and a trivalent crystallizable fragments (Fc) of antibody. We show that the dynamic binding capacity of the nanofiber device varies slightly with residence time, with the transfer kinetics slower for the IgG1, than for the IgG4 antibody. Surface plasmon resonance shows that the trivalent Fc binds to multiple Protein A ligand sites due to its three repeating Fc units, whereas multiple IgG antibodies bind to a single Protein A ligand. The nanofiber device gives comparable eluate purity to the resin column but at lower antibody concentrations due to its larger housing volume and lower binding capacity. It can be used for up to 200 cycles without performance loss and gives consistent eluate quality when scaled from 0.4 mL to 3.75 mL. We show that the use of a single nanofiber device to process one 2000 L clarified harvest through multiple cycles can reduce the Cost of Goods fivefold compared with a resin column. Costs are potentially lower for up to thirteen 2000 L harvests if device reuse can be validated. Nanofiber devices are thus an attractive option for manufacture of pre-clinical or clinical trial material where the number of batches is limited.

Abstract:The structural and functional characteristics of the Protein A MabSelect resin are determined for a virgin sample and for samples removed from a column that had been operated in an antibody capture process which had shown losses in product recovery over fewer than 20 cycles. Compared to the virgin resin, the cycled samples show reduced porosity and apparent pore size based on inverse size exclusion chromatography while transmission electron microscopy (TEM) shows accumulation of foulants on the cycled resin. Adsorption isotherms, batch adsorption kinetics, and batch desorption kinetics, obtained using the antibody in purified form, show that the cycled samples have about 10% lower binding capacity and slower mass transfer. Confocal scanning laser microscopy shows, however, that different degrees of fouling exist for different beads in the cycled samples, which may correspond to the existence of areas exposed to minimal or no flow in the process column. Replacing the standard cleaning procedure with an improved multi-step cleaning protocol prevented the accumulation of foulants in the resin beads, as evident from TEM, and resulted in a stable operation with high recovery.

Abstract:The rapid increase in antibody demand is urgently calling for high throughput technologies to enhance the efficiency in downstream purification. Membrane chromatography using membrane adsorbents (MAs) has emerged as an attractive alternative. Nevertheless, despite the increasing research efforts made in the past decades, there is a lack of a comprehensive understanding of the structure – chemistry – performance relationship of MAs used in antibody purification, leaving the development of MAs in trial-and-error without a rational guideline. To fill the knowledge gaps, this work focuses on analyzing the key factors influencing the efficiency of bind-and-elute operation of MAs in antibody capture, aiming to bring logics to the choices of membrane materials and structure properties and surface/ligand chemistries in different chromatographic modes. Despite its essential role in antibody binding, the impact of the membrane pore structure was only considered by less than one quarter of the literature. Given a comparatively low antibody binding capacity for affinity MC (roughly in the order of 10–30 mg/ml bed volume), non-affinity types exhibit significant potential but only if the selectivity could be further improved. The common pore size range of the substrates reported in the literature was around 0.2–1 μm, however, when employing surface functionalization, the choices of the functionalization degree and layer thickness were not carefully made, leading to compromised binding and throughput. Scalable fabrication techniques such as electrospinning are investigated in other fields, but are still at the infancy stage for antibody capture applications. Although conventional affinity ligands are well-explored, there is a growing interest to explore alternative and cheaper affinity (e.g., peptides) or non-affinity ligands that can serve in the bind-and-elute of antibodies. With new ligands, it is essential to understand the binding and elution kinetics assisted by mathematical simulations, which however remain under-investigated. Overall, this review provided an in-depth analysis of current developments in research and new thoughts toward designing next-generation membrane chromatography for filling the supply gaps of antibody products.

Abstract:The germicidal influence of various antiseptics on the bacterial and fungal flora of leg ulcers was studied in vitro. Best effects were achieved by the triphenylmethane dyes brilliant green and methylrosaniline chloride. Chloramine and povidone iodine yielded average results, whereas the germicidal effects of potassium permanganate and eosin were not satisfactory. On the other hand, treatment with ozone resulted in sufficient inactivation of germs--with the only exceptions of Serratia and Klebsiella species.
Optimizing high throughput antibody purification by using continuous chromatography media

Abstract:The ability to engineer monoclonal antibodies (mAbs) with high specificity made mAbs the fastest growing segment in the drug market. mAbs represent 8 of the top 20 selling drugs with combined sales of more than 57 billion US$ per year. The ability to purify large numbers of mAbs with sufficient yields for initial screening campaigns has direct impact on the timelines of a project. Automated liquid handling (ALH)-based mAb purification platforms have been used to facilitate the production of large numbers of mAbs. However, the ongoing pressure to de-risk potential lead molecules at an early development stage by including bio-physical characterization of mAbs has further increased the demand to produce sufficient quantities from limited sample volumes. A bottleneck so far has been the limited dynamic binding capacity of these systems, which is partly due to the binding properties of commonly used Protein A affinity matrices. The present publication suggests that by using a Protein A matrix optimized for continuous chromatography applications the yields of ALH-based but also standard lab-scale mAb purifications can be significantly increased without the need to change established protocols.

Abstract:Monoclonal antibodies (mAbs) are commonly used biologic drugs for the treatment of diseases such as rheumatoid arthritis, multiple sclerosis, COVID-19 and various cancers. They are produced in Chinese hamster ovary cell lines and are purified via a number of complex and expensive chromatography-based steps, operated in batch mode, that rely heavily on protein A resin. The major drawback of conventional procedures is the high cost of the adsorption media and the extensive use of chemicals for the regeneration of the chromatographic columns, with an environmental cost. We have shown that conventional protein A chromatography can be replaced with a single crystallization step and gram-scale production can be achieved in continuous flow using the template-assisted membrane crystallization process. The templates are embedded in a membrane (e.g., porous polyvinylidene fluoride with a layer of polymerized polyvinyl alcohol) and serve as nucleants for crystallization. mAbs are flexible proteins that are difficult to crystallize, so it can be challenging to determine the optimal conditions for crystallization. The objective of this protocol is to establish a systematic and flexible approach for the design of a robust, economic and sustainable mAb purification platform to replace at least the protein A affinity stage in traditional chromatography-based purification platforms. The procedure provides details on how to establish the optimal parameters for separation (crystallization conditions, choice of templates, choice of membrane) and advice on analytical and characterization methods.
Development of a New Affinity Gold Polymer Membrane with Immobilized Protein A

Abstract:New and highly selective stationary phases for affinity membrane chromatography have the potential to significantly enhance the efficiency and specificity of therapeutic protein purification by reduced mass transfer limitations. This work developed and compared different immobilization strategies for recombinant Protein A ligands to a gold-sputtered polymer membrane for antibody separation in terms of functionalization and immobilization success, protein load, and stability. Successful, functionalization was validated via X-ray photoelectron spectroscopy (XPS). Here, a recombinant Protein A ligand was coupled by N-hydroxysuccinimide (NHS)/N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide (EDC) chemistry to carboxy-functionalized, gold-sputtered membranes. We achieved a binding capacity of up to 104 ± 17 mg of the protein ligand per gram of the gold-sputtered membrane. The developed membranes were able to successfully capture and release the monoclonal antibody (mAb) Trastuzumab, as well as antibodies from fresh frozen human blood plasma in both static and dynamic setups. Therefore, they demonstrated successful functionalization and immobilization strategies. The antibody load was tested using bicinchoninic acid (BCA), ultraviolet-visible spectroscopy (UV-vis) measurements, and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The outcome is a fully functional affinity membrane that can be implemented in a variety of different antibody purification processes, eliminating the need for creating individualized strategies for modifying the surface to suit different substrates or conditions.

Abstract:The aim of this study was to determine whether the +896 A→G substitution of the Toll-like receptor 4 (TLR4) gene, causing the Asp299→Gly change in the extracellular domain of TLR4, influences treatment response in recent-onset rheumatoid arthritis. 169 patients with rheumatoid arthritis were genotyped from the Finnish Rheumatoid Arthritis Combination Therapy trial, in which they were treated either with only one disease-modifying antirheumatic drug (DMARD) with or without prednisolone (single group), or with three DMARDs and prednisolone (combination group). Patients homozygotic for the wild-type +896A allele were compared with those having the polymorphic G allele in terms of early clinical response (at 6 months) by the 28-joint Disease Activity Score (DAS28). 1 of 20 (5%; (95% (confidence interval (CI) 1 to 5)) patients of the single group with TLR4 +896AG or GG and 29 of 67 (43%; (95% CI 31 to 56)) patients with AA were in remission (p = 0.001). DAS28 of the single group with TLR4 +896AG or GG was higher than with AA (p = 0.019). In the combination group, remission rates and DAS28 values were comparable between the genotypes. The polymorphic TLR4 +896G allele may impair treatment response to single DMARD treatment in recent-onset rheumatoid arthritis.
