Abstract:A 120-day feeding trial was conducted to evaluate the effect of different stocking densities on growth, the non-specific immunities, antioxidant status and digestive enzyme activities of Oreochromis niloticus fingerlings under a zero-water exchange biofloc system. Tilapias (0.51 ± 0.05 g) were randomly distributed in twelve tanks, each with 300 L water. The experimental design was completely randomized using three replications with four treatments 166 orgs m−3 (LD, low density), 333 orgs m−3 (MD, middle density) and 600 orgs m−3 (HD, high density) with glucose added as biofloc groups, and a clear water group without glucose added as a control 333 orgs m−3. The fish cultured in LD and MD group showed higher final body weight. For the digestive enzymes, the lipase, trypsin, and amylase activities were all depressed in HD group and control group. Regarding the immune and antioxidant abilities, significantly lower values (P < 0.05) of the lysozyme, complement 3, and glutathione were observed for the fish that reared in the control group and HD group. The stress indicator, the cortisol, 5-hydroxytryptamine, and glucose concentrations were also depressed in HD group and control group, meanwhile the alanine aminotransferase, aspartate transaminase and alkaline phosphatase were all higher in HD group and control group. The significant higher survival was observed in the LD and MD group after Vibrio harveyi challenge test. The results of the experiment indicated that the biofloc in situ had the effects of anti-crowding stress.

Abstract:Optimizing fish performance depends on several factors, with dietary protein levels and rearing temperature playing important roles. In this study, Nile tilapia fingerlings (Oreochromis niloticus) weighing an average of 20.00 ± 1.26 g were divided into nine groups (in three replicates). Each group was subjected to different water temperatures (26 °C, 28 °C, and 30 °C) and received one of three dietary protein levels (20%, 25%, and 30%) for two months. Our findings indicate that higher temperatures, particularly at 30 °C, increased water electrical conductivity and total dissolved salts, especially noticeable in fish fed 25% or 30% crude protein (CP). Lower total ammonia nitrogen levels were observed at 28 °C with 25% CP, 30 °C with 30% CP, and 26 °C with 30% CP. Hepatic growth hormone receptor 1 and insulin-like growth factor 1 expression gradually rose with higher dietary CP percentages in fish at 26 °C but declined in those at 30 °C, albeit remaining higher than in the 28 °C groups with 25% CP. Fish at 28 °C showed the best final body weights and growth performance when fed 20% or 25% CP, with no significant difference between these groups. Hepatic leptin expression did not differ significantly among groups, but hepatic fatty acid binding protein expression notably increased in fish fed 30% CP at both 26 °C and 30 °C compared to those at 28 °C with 25% CP. Within the same temperature group, fish fed 30% CP exhibited higher globulin levels, particularly thriving at 28 °C or 30 °C. Hepatic mucin-like protein expression significantly increased across all groups, especially in fish at 30 °C with 30% CP compared to those at 28 °C with 25% CP. Hepatic lysozyme expression also increased notably in fish at 30 °C with 30% CP. Notable changes in superoxide dismutase, catalase, and glutathione peroxidase expression were observed, with the highest serum superoxide dismutase and catalase activities recorded in fish at 30 °C with 25% CP. Overall, dietary protein levels of 25% and 30%, combined with temperatures of 28 °C and 30 °C, yielded favorable outcomes, particularly favoring 28 °C with 25% protein.

Abstract:Dietary phospholipids supplementation is essential for normal development of fish larvae due to their limited phospholipid biosynthetic ability. The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, antioxidant capacity and lipid metabolism of larval hybrid grouper. The larvae (40 day after hatching, 0.37 +/- 0.01 g) were fed the diet with graded level of phospholipids (1.87, 3.61, 5.53, 7.25 and 9.69%) four times daily for 23 days. Results showed that the specific growth rate of grouper larvae was significantly improved with the increase of dietary phospholipids, and optimal dietary phospholipids was estimated to be 9.17%. Fatty acids composition of whole fish body and liver was highly reflective of dietary fatty acid profile. In addition, the increase of dietary phospholipids resulted in a significantly elevated hepatic crude lipid and triglyceride content in a dose-dependent manner. The analysis of gene expression revealed that increased dietary phospholipids did not produce any statistical effect on the expression of lipogenesis genes including LXRalpha, FAS and ACC, but significantly facilitated the expression of DGAT2, a rate-limiting enzyme involved in triglyceride synthesis. The expression of genes, CPT1, involved in I3-oxidation was not significantly influenced as dietary phospholipids increased. However, phospholipids inclusion significantly reduced the expression of ATGL, an rate-limiting lipolytic enzyme. Additionally, the supplementation of dietary phospholipids significantly elevated the activity of antioxidant relate enzymes, CAT, SOD and T-AOC, but significantly decreased the MDA content, an end product of fatty acid peroxidation. In summary, dietary phospholipids supplementation was beneficial in improving growth performance and antioxidant capacity of grouper larvae, and meanwhile, promoted hepatic lipid deposition through regulating the synthesis and hydrolysis of triglyceride.

Abstract:The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, and lipid metabolism in juvenile hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lancolatus ♂). Five isonitrogenous and isolipidic diets with graded levels of phospholipids (14.10, 32.10, 52.50, 70.20, and 91.30 g/kg) were fed to triplicate groups of juvenile grouper (initial body weight 13.00 ± 0.02 g) for 61 days. Results showed that dietary phospholipids produced no statistical influence on growth performance of grouper. The fatty acids profiles of experimental fish are correlated well with dietary fatty acids composition. The increase of dietary phospholipids significantly elevated the crude lipid content in whole fish body, liver and muscle. Meanwhile, the increased dietary phospholipids significantly promoted hepatic triglyceride (TG) accumulation. The results of gene expression analysis revealed that dietary phospholipids did not significantly affect the expression of lipogenesis related genes, FAS, ACC and DGAT2. In addition, high dietary phospholipids inclusion significantly decreased the expression of fatty acids oxidation related genes, PPARɑ and CPT1. In summary, juvenile grouper don’t have a specific requirement for dietary phospholipids which inclusion aggravated lipid deposition through the regulation of fatty acids oxidation at the transcriptional level.

Abstract:The objectives of this study were to investigate the effect of different dietary lipid levels on the serum biochemistry, antioxidant responses, initial immunity, and mTOR pathway of juvenile genetic improvement-farmed tilapia (GIFT, Oreochromis niloticus). Six groups of the juveniles (initial body weight 20.66 ± 1.33 g) in triplicate were fed for 90 days using six iso-nitrogen diets with different lipid levels (0.35%, 3.35%, 6.35%, 9.35%, 12.35%, and 15.35%). The main results were as follows: Compared with the control group (0.35%), the diets with different lipid supplementation significantly improved (p < 0.05) the contents of total protein (TP), albumin (ALB), globulin (GLB), glucose (GLU), triglyceride (TG), total cholesterol (T-CHO), high-density lipoprotein (HDL), low-density lipoprotein (LDL), total antioxidant capacity (T-AOC), malondialdehyde (MDA), complement 3 (C3), and immunoglobulin M (IgM), the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), lysozyme (LYZ), and alkaline phosphatase (ALP), the expression level of phosphatidylinositol-3-kinase (PI3K), Akt protein kinase B (Akt), and mammalian target of rapamycin (mTOR) genes in juvenile tilapia. However, diets with different lipid supplementation significantly reduced (p < 0.05) the expression level of tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) genes of juvenile tilapia. In conclusion, the with different lipid supplementation could significantly affect the serum biochemistry, antioxidant responses, initial immunity, and mTOR pathway of juvenile tilapia.

Abstract:Tilapia tolerate hypoxia; thus, they are an excellent model for the study of hypoxic adaptation. In this study, we determined the effect of acute hypoxia stress on the antioxidant capacity, metabolism, and gill/liver ultrastructure of male genetically improved farmed tilapia (GIFT, Oreochromis niloticus). Fish were kept under control (dissolved oxygen (DO): 6.5 mg/L) or hypoxic (DO: 1.0 mg/L) conditions for 72 h. After 2 h of hypoxia stress, antioxidant enzyme activities in the heart and gills decreased, while the malondialdehyde (MDA) content increased. In contrast, in the liver, antioxidant enzyme activities increased, and the MDA content decreased. From 4 to 24 h of hypoxia stress, the antioxidant enzyme activity increased in the heart but not in the liver and gills. Cytochrome oxidase activity was increased in the heart after 4 to 8 h of hypoxia stress, while that in the gills decreased during the later stages of hypoxia stress. Hypoxia stress resulted in increased Na+-K+-ATP activity in the heart, as well as hepatic vacuolization and gill lamella elongation. Under hypoxic conditions, male GIFT exhibit dynamic and complementary regulation of antioxidant systems and metabolism in the liver, gills, and heart, with coordinated responses to mitigate hypoxia-induced damage.

Abstract:Objective We tested the hypothesis that fat metaplasia on MRI of the sacroiliac joints (SIJ) increases the propensity for new bone formation in the spine of patients with spondyloarthritis. Methods We assessed baseline T1-weighted and short τ inversion recovery SIJ MRIs from patients in the Follow Up Research Cohort in Ankylosing Spondylitis (FORCAST). Radiographic progression was assessed using the modified Stoke Ankylosing Spondylitis Spine Score (mSASSS). Structural and inflammatory lesions were scored using the Spondyloarthritis Research Consortium of Canada (SPARCC) SIJ structural and SPARCC SIJ inflammation scores, respectively. Radiographic progression was compared in cases with and without definite MRI lesions (score ≥2 or <2) and the extent of MRI lesions at baseline was compared in patients with and without radiographic progression. The predictive capacity of MRI SIJ lesions for radiographic progression in the spine was assessed in univariate and multivariate regression analyses. Results The extent of MRI structural lesions in the SIJ at baseline was significantly greater in those patients who had spinal radiographic progression on follow-up (p=0.003, 0.02, 0.003 for fat metaplasia, backfill and ankylosis, respectively). Also, radiographic progression was significantly greater in cases with definite baseline SIJ ankylosis (p=0.008). In multivariate regression that included all types of MRI lesions and was adjusted for age, sex, symptom duration, duration of follow-up, CRP, baseline mSASSS and treatment, the extent of SIJ fat metaplasia and ankylosis at baseline were independently associated with radiographic progression. Conclusions SIJ ankylosis and fat metaplasia but not inflammatory lesions increase the propensity for radiographic progression in the spine.

Abstract:THIS work was undertaken with a view to ascertaining the part played by the spleen in certain acute and chronic infective and noninfective diseases. It must be pointed out that this work consists solely of the study of the histological and cytological changes, together with the bacteriological findings in these cases. Special attention was directed to the question of the fate of the erythrocytes in the spleen, in so far as microscopical examination can determine. The cases examined, 87 in number, comprised 46 examples of acute infect'ive and non-infective diseases, 28 cases of diseases presenting a chronic course, and 13 of chronic diseases with an acute terminal infection.

Abstract:An 8-week feeding trial was carried out to assess the effects of dietary vitamin E on growth, fillet texture, and antioxidant capacity of genetically improved farmed tilapia (GIFT), Oreochromis niloticus. In total, 240 fish (initial body weight 79.37 g) were assigned to four triplicate groups (20 fish per replicate) and fed diets supplemented with one of the four vitamin E (dl-α-tocopheryl acetate) concentrations: 0 (control diet), 40, 200, and 1000 mg kg−1. Fish fed the control diet had significantly poorer growth (weight gain; 185.29 vs 214.25–219.28%), feed efficiency (63.99 vs 70.44–72.30%), and survival rate (86.67 vs 95–100%) than those fed vitamin E-supplemented diets (P < 0.05). Protein percentage in muscle of the fish fed the 40 mg kg−1 diet was significantly higher than that of the control group (19.95 vs 18.65%) (P < 0.05). Based on texture profile analysis, fish in the control group had significantly lower cohesiveness (0.43 vs 0.47–0.50), resilience (0.25 vs 0.28–0.31), and chewiness (340.8 vs 406.4–504.5 g) than fish in the other groups (P < 0.05). Vitamin E supplementation significantly enhanced superoxide dismutase activity in serum and muscle and catalase activity in serum (P < 0.05). Malondialdehyde contents in serum and muscle significantly decreased (P < 0.05). These results suggest that no less than 40 mg kg−1 vitamin E improve growth performance, fillet flesh quality, and antioxidant capacity of GIFT. On the basis of second-order polynomial regression, the optimum vitamin E supplemented level for GIFT is about 573.8–582.3 and 655–752.5 mg kg−1 based on fillet texture and serum antioxidant capacity, respectively.

Abstract:A clinical trial was undertaken to examine the effects of a potent cyclooxygenase inhibitor, flurbiprofen, on both developing and established gingivitis in humans. 21 subjects with healthy gingiva abstained from all oral hygiene procedures for 21 days. 7 subjects were prescribed flurbiprofen, 50 mg b.d. beginning from baseline and a control group (Cl, n = 14) were given placebo. Gingival redness and bleeding on probing were assessed at baseline, 7, 14 and 21 days. Crevicular fluid (GCF) samples were also taken to determine concentrations of PGE2, TxB2 and LTB4 at baseline and at 21 days. Results show that flurbiprofen significantly inhibited the development of redness and bleeding (p < 0.001) effects which were associated with a significant inhibition of TxB2 (p < 0.05). There were no apparent flurbiprofen effects on GCF-PGE2 or GCF-LTB4 during this 21-day gingivitis, model To assess the effects of flurbiprofen on established experimental gingivitis, the model was extended to 28 days. On day 21, the Cl group was subdivided into 2 groups of 7 subjects. One group was prescribed flurbiprofen (50 mg b.d.) for 7 days and controls (C2) continued to take placebo. All subjects continued to abstain from tooth cleaning. Pretreatment (day 21) and post-treatment (day 28) comparisons showed that flurbiprofen again significantly inhibited bleeding (p < 0.001), but did not affect redness. Control subjects demonstrated a significant elevation in gingival bleeding on day 28, and this was associated with significant rises in GCF-PGE2 (p < 0.001), GCF-TxB2 (p < 0.01) and GCF-LTB4 (p < 0.05).(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:This study was conducted to evaluate the effect of dietary protein level and amino acid supplementation on tilapia nursing performance under biofloc system conditions at cold suboptimum temperature. Four experiment diets were examined; 30% CP diet as a positive control (30P), 22 % CP diet as negative control (22P), 22 % CP diet supplemented with 1% lysine (22PL) and 22 % CP diet supplemented with 0.5% threonine (22PT). Formulated diets were tested under biofloc system conditions in three replicates for each treatment. 12 experimental tanks 1000 L were used, each tank was filled with 400 L. each tank was stocked with 50 fries (6.20±0.01 gm. /fish). The experiment was extended for 97 days. Fish fed on a 22PT diet had the highest significantly the best values of weight gain (WG), feed intake (FI), feed convergent ratio (FCR), and specific growth rate (SGR) (11.10, 16.95, 1.55 and 1.40, respectively). Also, 22PT diet groups record the highest protein content of dried microbial floc and wet whole-body. The lowest total ammonia nitrite (TAN) values were recorded for the fish groups fed on a 22PT diet. It could be concluded that under biofloc system conditions, a low protein diet supplemented with threonine can be used as a tool to improve biofloc system performance during tilapia nursing at cold suboptimal water temperature.
Sodium chloride alleviates oxidative stress and physiological responses induced by extreme winter cold in genetically improved farmed tilapia (GIFT; Oreochromis niloticus )

Abstract:A 6-week trial was designed to investigate the effects of dietary sodium chloride supplementation on physiological, metabolic, and molecular stress response parameters. The findings showed that (1) there were no significant differences between sodium chloride supplementation groups (0.05S, 0.1S, and 0.15S) and the control group ( P > 0.05), except for the 0.2S diet, which showed better final body weight, weight gain rate, specific growth rate, and feed conversion ratio than the control group ( P < 0.05). (2) The hypothermic stress experiment results showed that the survival rates in the 0.1S and 0.15S diets were significantly higher than the control group ( P < 0.05). (3) Transcription results showed that these enriched pathways in the gill were mainly energy metabolism and apoptosis pathways, while the major enrichment pathways in the liver were mainly amino acid metabolism and carbohydrate metabolism. (4) The plasma parameter results showed, compared to the control group, the 0.15S diet significantly increased the plasma GLU, TG contents, and Na + and K + concentrations and decreased the plasma ALT activity ( P < 0.05). In addition, the 0.1S diet increased the plasma ALB content and Cl − concentration ( P < 0.05). The gill Na + /K + -ATPase activity decreased markedly when the fish were fed the 0.1S and 0.15S diets ( P < 0.05). The antioxidant enzyme activity results showed that the 0.1S and 0.15S diets significantly increased the T-SOD activities ( P < 0.05). Gene expression results showed that compared to the control group, the 0.1S and 0.15S diets up-regulated the expression of gys , hsp70 , mlcp , mlc , myosin , tnt mRNA, and down-regulated the akt , gk , and erk mRNA expression. Based on the regression analysis, the optimum dietary sodium chloride levels range from 0.10 % to 0.13 % of the diet, which could facilitate energy regulation, improve the immune response, and ultimately strengthen the cold resistance of GIFT.

Abstract:This study investigated the effect of lipid levels on growth, body composition, lipid metabolism enzymes activity and related genes expression in hybrid grouper (♀ Epinephelus fuscointestinestatus × ♂ E. lanceolatu ). Three diets with different lipid levels of 6% (low-lipid feed, LF), 11% (medium-lipid feed, MF) and 16% (high-lipid feed, HF) were designed and each diet was fed to triplicate groups of 30 fish each (6.73 ± 0.00g) for eight weeks. The results revealed the following. 1) The maximum weight gain rate (WGR), specific growth rate (SGR), protein efficiency ratio (PER) and the lowest feed conversion ratio (FCR) values were found in the HF group. There was no significant difference in the survival rate (SR) among the groups ( P > 0.05). 2) Condition factor (CF), hepatosomatic index (HSI) and visceralsomatic index (VSI) increased with increasing dietary lipid. 3) Crude lipid content in the whole-body and muscle increased significantly as the dietary lipid level increased ( P < 0.05). 4) Principal component analysis of fatty acids showed that sum n-3 polyunsaturated fatty acids (∑n-3PUFA)/sum n-6 polyunsaturated fatty acids (∑n-6PUFA) was significantly higher in the HF group than in the other two groups ( P < 0.05). 5) Biochemical serum indexes showed higher triglycerides (TG), total cholesterol (TC), alkaline phosphatase (AKP) and low-density lipoprotein-cholesterol (LDL-C) levels but lower high-density lipoprotein-cholesterol (HDL-C) level in the HF group. 6) Liver lipid metabolism enzymes activity and biochemical indicators showed that fish fed the MF diet had higher lipoprotein lipase (LPL), hepatic lipase (HL) and triglyceride lipase (ATGL) activity but lower fatty acid synthase (FAS) activity and very-low density lipoprotein-cholesterol (VLDL-C) level. 7) The relative expression levels of lipoprotein lipase ( lpl ), adipocyte-type fatty acid-binding protein ( a-fabp ), liver-type fatty acid-binding protein ( l-fabp ), heart-type fatty acid-binding protein ( h-fabp ) and hormone-sensitive lipase ( hsl ) genes in the fish liver were higher in the MF group with increasing dietary lipid ( P < 0.05), while the fatty acid synthase ( fas ) gene was expressed at lower relative level ( P < 0.05). 8) Histological oil-red sections of the liver showed that fish in the HF group had larger and more hepatic lipid droplets than those fed the other diets. It was concluded that fish fed the HF diet had better growth performance but this diet was not conducive to the progress of lipid metabolism, resulting in lipid accumulation and affecting the quality of the fish. This study provides useful information for the formulation of environmentally friendly and cost-effective diets for hybrid grouper.

Abstract:L-carnitine (LC) and α-Ketoglutarate (AKG) are important growth promoters used in aquafeed. The study aimed to evaluate the incorporation of LC and AKG at different ratios in the diet of tilapia (initial weight 1.38 ± 0.03 g) in order to facilitate lipid utilization and protein synthesis. Fish were fed six isonitrogenous (~ 30 g/100 g CP) and isolipidic (~ 6 g/100 g CL) diets containing graded LC/AKG ratios of 0 (Control), 0.11, 0.42, 1.00, 2.33 and 9.00 in six treatments for 60 days. Fish fed with LC/AKG ratios 2.33 and 9.00 showed significantly (P < 0.05) higher percentage weight gain, specific growth rate and protein efficiency ratio. Feed conversion ratio in fish-fed diets with LC/AKG ratio 9.00 improved significantly (p < 0.05) than other treatments. The whole-body protein content of tilapia and digestive enzyme activity were significantly higher with higher weight gain. The body lipid content was significantly lower in the LC/AKG ratio 9.00. The liver antioxidant parameters and activity of the immune components were significantly higher in the LC/AKG ratio 9 group. The lower serum triglyceride and cholesterol level was also recorded in LC/AKG ratio 9 group. The histology of the intestine and liver showed increased villi area and decreased lipid droplets, respectively, in tilapia fed with higher LC/AKG ratios. It was concluded from the above results that the higher LC and lower AKG (LC/AKG ratio 9.00) combination attributed maximum lipid utilization and higher protein efficiency and thus better growth performance in tilapia. This was also reflected in activity of digestive enzymes, antioxidant enzymes and immune status in tilapia.

Abstract:Marine red yeast is a good feed additive for the aquaculture industry that can promote the growth of aquatic animals, and significantly improve their antioxidant capacity, survival rate, and nonspecific immune ability of the body. Our hypothesis is that dietary supplementation with marine red yeast could affect the serum biochemistry, muscle composition, muscle fatty acid composition, lipid metabolism enzyme activity, and expression of antioxidant and inflammatory genes in juvenile genetically improved farmed tilapia (GIFT, Oreochromis niloticus). Five diets with different levels of marine red yeast (0 %, 0.25 %, 0.50 %, 0.75 %, and 1.00 %) were used to feed juvenile GIFT (initial weight: 21.12 ± 0.86 g) for 60 days. The main results are as follows: Compared with the control group (0 % marine red yeast), the contents of total protein (TP), albumin (ALB), high-density lipoprotein (HDL), and free fatty acid (FFA) in the serum of juvenile GIFT tilapia significantly increased (P < 0.05), while the contents of glucose (GLU), triglyceride (TG), total cholesterol (T-CHO), low-density lipoprotein (LDL), alanine aminotransferase (ALT) significantly decreased (P < 0.05). The contents of crude protein, crude fat, and docosahexaenoic acid + eicosapentaenoic acid (DHA+EPA) in the muscle significantly increased (P < 0.05), while the contents of moisture and monounsaturated fatty acids (∑MUFAs) significantly decreased (P < 0.05). The activities of acetyl CoA carboxylase α (ACCα), glucose-6-phosphate dehydrogenase (G6PD), and stearoyl-CoA desaturase (SCD) in the liver significantly increased (P < 0.05). The expression levels of catalase (cat), alkaline phosphatase ( alp ), nuclear factor erythroid 2-related factor 2 ( nrf2 ), lysozyme ( lyz ), glutathione S-transferase (gst), glutathione peroxidase (gsh-px), and superoxide dismutase (sod) genes in the liver significantly increased (P < 0.05). The expression levels of tumor necrosis factor α ( tnf-α ), interferon-γ ( inf-γ ), interleukin 6 ( il -6), interleukin 8 ( il -8), interleukin 1β ( il-1β ), and transforming growth factor-β ( tgf-β1 ) genes in the liver, spleens, and head kidney significantly increased (P < 0.05), while the expression level of interleukin 10 ( il -10) gene significantly decreased (P < 0.05). In conclusion, the addition of different levels of marine red yeast could significantly affect the serum biochemistry, muscle composition, muscle fatty acid composition, lipid metabolism enzyme activity, and expression of antioxidant and inflammatory genes in juvenile GIFT tilapia. Based on the results, the optimal dietary marine red yeast level was 0.50 %.

Abstract:Sutili, Fernando Jonas ; Battisti, Eduardo Kelm ; Gressler, Luciane Tourem ; Kreutz, Luiz Carlos ; A 60-day feeding trial was conducted to evaluate the impact of dietary β-glucan in combination with citral and eugenol on the growth, whole body composition, and innate immunity of Nile tilapia (Oreochromis niloticus) weighing 9.16 g, under cold suboptimal temperatures (20oC). The treatments included a control (Co); β-glucan (G) at 1.0 g/kg/diet; citral and eugenol (CE) at 0.5 g/kg each; and a combination of β-glucan, citral, and eugenol (GCE) at the same concentrations (1.0, 0.5 and 0.5 g/kg, respectively). Although no significant differences were observed in growth parameters post-trial, proximate analysis indicated an increase in body protein and a decrease in body fat content in the GCE group compared to the Co and G groups, respectively. Enhanced respiratory burst activity of blood leukocytes and myeloperoxidase activity were recorded in the GCE group, indicating improved innate immune responses. Serum complement hemolytic activity was significantly higher in the G group, with no additional benefits from the CE or GCE treatments. Furthermore, the G and GCE groups displayed increased serum lysozyme activity when compared to the control. These findings suggest that the combination (GCE) of these feed additives may offer a beneficial strategy for promoting health in tilapia reared under stressful conditions.

Abstract:The hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus) is a new species of grouper crossed from giant grouper (E. lanceolatus) as the male parent and brown-marbled grouper (E. fuscoguttatus) as the female parent. We hypothesized that optimal levels of dietary protein may benefit liver function. High-lipid diets are energetic feeds that conserve protein and reduce costs, and are a hot topic in aquaculture today. Therefore, the objective of the research is to investigated the effects of dietary protein level in high-lipid diets on serum and liver biochemistry, liver histology, and liver immune and antioxidant indexes and gene mRNA expression of the juvenile hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus). Six iso-lipidic (161 g/kg) diets were formulated containing graded levels of protein (510 as control, 480,450, 420, 390 and 360 g/kg). Each treatment consisted of three replicates and 30 fish (6.70 ± 0.02 g) in one replicate. After an 8-week feeding experiment, the results indicated the following: (1) With the decreasing of dietary protein level, the specific growth rate (SGR) of groupers increased gradually and then decreased; SGRs of the 390 and 360 g/kg groups were significantly lower than other groups (p < 0.05). (2) In terms of serum and liver, the activity of antioxidant enzymes such as catalase (CAT) and superoxide dismutase (SOD), and the total antioxidant capacity (T-AOC) content, and the activity of immune enzymes such as lysozyme (LYS) and immunoglobulin (IgM) was significantly increased under the appropriate protein level. (3) Based on liver histology, we know that high or low dietary protein levels cause liver damage. (4) Dietary protein levels can significantly affect the mRNA expression levels of an anti-inflammatory factor gene (tgfβ), pro-inflammatory factor genes (il6, il8), heat shock proteins, and antioxidant and immune genes (hsp70 and hsp90, gpx, nrf2, keap1). It is concluded that the appropriate protein level can promote the growth performance of groupers, improve antioxidant activity and immune enzyme activity in serum and liver, and enhance the expression of immune genes.

Abstract:The ability of Nile tilapia to tolerate hypoxia, as well as low and high temperatures, presents a significant economic concern, as it adversely affects their growth and leads to increased mortality rates. A 42-day feeding trial was conducted to evaluate the impact of adding Bacillus amyloliquefaciens AV5 to a fish meal on the physiological response of Nile tilapia. Three meals were administered to fish (23.4 ± 0.3g) in triplicates. The diets included GC (without B. amyloliquefaciens AV5), G1 (10<sup>6</sup> cfu/g), and G2 (10<sup>8</sup> cfu/g). After the treatment trial, we assessed the antioxidant parameters, hemato-immunological indices, and stress-related genes in O. niloticus. Subsequently, we subjected the fish to hypoxia for 20h and low and high temperatures for 3h each. The findings demonstrated a significant rise in white blood cells, red blood cells, haemoglobin, and hematocrit levels in the blood of fish that were fed a meal supplemented with B. amyloliquefaciens AV5, compared to the control group (GC) (P<0.05), the serum of all fish groups that were supplemented with B. amyloliquefaciens AV5 exhibited an increase in catalase, total antioxidant capacity and superoxide dismutase activity and a decrease in lactate dehydrogenase, alanine aminotransferase, aspartate aminotransferase, pyruvate kinase, myeloperoxidase, glucose, cortisol (P < 0.05). In addition, all fish diet groups that received B. amyloliquefaciens AV5 as a supplement exhibited elevated levels of HIF-1α and HSP70 expression in their livers (P<0.05). Nile tilapia in the G2 diet, exhibited improved values in most evaluated indices under various stress settings (P<0.05). These data indicate that the G2 supplement may be used as a preventive measure to weaken the impacts of environmental stress on O. niloticus.

Abstract:High temperatures and low oxygen in aquatic environments, such as intensive aquaculture or in natural watersheds, inevitably cause stress in fish. Fish are exposed to high temperatures during the summer, which exacerbates hypoxia. Hypoxia (1.2 +/- 0.2 mg/L) under 20 degrees C (20 HG) and 26 degrees C (26 HG) was simulated to induce stress in largemouth bass (Micropterus salmoides). Related enzymes and genes involved in antioxidant, immune, and apoptotic responses were selected to explore the interactive effects of temperature and hypoxia on largemouth bass. The results showed that malondialdehyde (MDA) levels in plasma, gill, and liver increased in the 26 HG (p < 0.05). Liver superoxide dismutase (SOD) activity increased in the 26 HG. Peak SOD (SOD1, SOD2, SOD3a, and SOD3b), CAT, and GSH-Px mRNA levels in the gill and liver were observed at 12-24 h of stress. The levels of gill and liver total antioxidant capacity, catalase (CAT), glutathione peroxidase (GSH-Px) activities and other enzyme activities and genes in the 26 HG were higher than those in the 20 HG (p < 0.05). The gill and liver add phosphatase and alkaline phosphatase activities increased with time in the 26 HG (p < 0.05), while gill and liver lysozyme activities in the 26 HG were lower than those in the 20 HG (p < 0.05). Tumor necrosis factor-alpha mRNA level was upregulated in the gill and downregulated in the liver at 24 h in the 26 HG. Interleukin (IL)-1 beta and IL-8 mRNA levels were upregulated in the gill and liver in the 26 HG at 24 h, whereas IL-15 mRNA level was downregulated in the 26 HG at 12 h. Transforming growth factor-beta 1 mRNA level was upregulated in the gill in the 20 HG at 24 h, but downregulated in gill and liver in the 26 HG at 24 h. Similarly, IL-10, Hepcidin-1, and Hepcidin-2 showed lower expression levels in the 26 HG. Gill and liver caspase-3 activities were higher in the 26 HG (p < 0.05), and gill caspase-3 activity was higher than that in the liver. The mRNA levels of proapoptotic genes (caspase-3, caspase-8, and caspase-9) were higher in the 26 HG. The present study demonstrates the interactive effects of temperature and hypoxia on stress in largemouth bass gill and liver. These results will be helpful to understand the mechanisms of stress induced by temperature and hypoxia in fish and provide a theoretical basis for aquaculture management.
