Abstract:The migration of sperm cells in a female reproductive tract is responsible for the successful fertilization of the female egg. In this research work, the effect of the surrounding fluids on the motion of sperm cells has been studied using a microfluidic channel. To analyze the motility of sperm, primary motility parameters such as velocity, beat frequency, amplitude, and derived parameters such as linearity, straightness, and wobble have been measured. The results indicate that sperms possess higher progressive motility in non-Newtonian fluids compared to Newtonian fluids in the same viscosity range. The motion of the sperm shows an inverse relationship between the amplitude of the head trajectory and the beat frequency of the flagella. Numerical studies were performed to measure the drag force on these sperm. The trajectories of the flagella, forces acting on sperm, power generated, pulling power, and efficiency of the sperm motion through the fluid medium have been investigated and a relationship between the force and rotation of the flagella has been established. The results show that the flagella also change their shape based on the properties of the surrounding fluid. This study aims to improve our understanding of issues related to infertility diagnosis and help design in-vitro experiments required for sperm separation.

Abstract:A major puzzle in biology is how mammalian sperm determine and maintain the correct swimming direction during the various phases of the sexual reproduction process. Whilst chemotaxis is assumed to dominate in the immediate vicinity of the ovum, it is unclear which biochemical or physical cues guide spermatozoa on their long journey towards the egg cell. Currently debated mechanisms range from peristaltic pumping to temperature sensing (thermotaxis) and direct response to fluid flow variations (rheotaxis), but little is known quantitatively about their relative importance. Here, we report the first quantitative experimental study of mammalian sperm rheotaxis. Using microfluidic devices, we investigate systematically the swimming behavior of human and bull sperm over the whole range of physiologically relevant shear rates and viscosities. Our measurements show that the interplay of fluid shear, steric surface-interactions and chirality of the flagellar beat leads to a stable upstream spiraling motion of sperm cells, thus providing a generic and robust rectification mechanism to support mammalian fertilisation. To rationalise these findings, we identify a minimal mathematical model that is capable of describing quantitatively the experimental observations. The combined experimental and theoretical evidence supports the hypothesis that the shape and beat patterns of mammalian sperm cells have evolved to optimally exploit rheotaxis for long-distance navigation.

Abstract:The migratory abilities of motile human spermatozoa in vivo are essential for natural fertility, but it remains a mystery what properties distinguish the tens of cells which find an egg from the millions of cells ejaculated. To reach the site of fertilization, sperm must traverse narrow and convoluted channels, filled with viscous fluids. To elucidate individual and group behaviors that may occur in the complex three-dimensional female tract environment, we examine the behavior of migrating sperm in assorted micro-channel geometries. Cells rarely swim in the central part of the channel cross-section, instead traveling along the intersection of the channel walls (`channel corners'). When the channel turns sharply, cells leave the corner, continuing ahead until hitting the opposite wall of the channel, with a distribution of departure angles, the latter being modulated by fluid viscosity. If the channel bend is smooth, cells depart from the inner wall when the curvature radius is less than a threshold value close to 150 micron. Specific wall shapes are able to preferentially direct motile cells. As a consequence of swimming along the corners, the domain occupied by cells becomes essentially 1-dimensional. This leads to frequent collisions and needs to be accounted for when modeling the behavior of populations of migratory cells and considering how sperm populate and navigate the female tract. The combined effect of viscosity and three-dimensional architecture should be accounted for in future in vitro studies of sperm chemoattraction.

Abstract:To fertilize eggs, sperm must pass through narrow, complex channels filled with viscoelastic fluids in the female reproductive tract. While it is known that the topography of the surfaces plays a role in guiding sperm movement, sperm have been thought of as swimmers, i.e., their motility comes solely from sperm interaction with the surrounding fluid, and therefore, the surfaces have no direct role in the motility mechanism itself. Here, we examined the role of solid surfaces in the movement of sperm in a highly viscoelastic medium. By visualizing the flagellum interaction with surfaces in a microfluidic device, we found that the flagellum stays close to the surface while the kinetic friction between the flagellum and the surface is in the direction of sperm movement, providing thrust. Additionally, the flow field generated by sperm suggests slippage between the viscoelastic fluid and the solid surface, deviating from the no-slip boundary typically used in standard fluid dynamics models. These observations point to hybrid motility mechanisms in sperm involving direct flagellum-surface interaction in addition to flagellum pushing the fluid. This finding signifies an evolutionary strategy of mammalian sperm crucial for their efficient migration through narrow, mucus-filled passages of the female reproductive tract.

Abstract:Many motile biological cells navigate along concentration gradients of signaling molecules: This chemotaxis guides for instance sperm cells from marine invertebrates, which have to find egg cells in the ocean. While chemotaxis has been intensively studied for idealized conditions of rotationally symmetric gradients in still water, natural gradients are usually distorted, e.g., by turbulent flows in the ocean. Recent experiments and direct numerical simulations with sperm cells and bacteria surprisingly suggest the existence of an optimal flow strength at which chemotaxis is more effective than for still water. We use sperm chemotaxis in simple shear flow as a prototypical example to understand the origin of such an optimal flow strength theoretically: We quantify how flow accelerates spreading of signaling molecules released by the egg, but distorts the resulting concentration field into long and thin filaments. The competition between these two effects sets an optimal flow strength that maximizes sperm-egg encounter. We characterize how sperm cells `surf' along concentration filaments, typical for scalar turbulence, revealing a general navigation paradigm in the presence of flow. We compare both simulation and theory with previous experimental results and find good agreement.

Abstract:International Journal of Modern Physics B, Ahead of Print. The sperm propelling mechanism has been proposed as a possible resource for soft micro-robots in confined spaces, with potential applications in biomedical engineering. Human sperm cells essentially swim through the non-Newtonian liquid (cervical mucus) to reach their target. Thus, sperm cells swimming through non-Newtonian fluids is not vital only for physiology, but also for the fabrication of swimming micro-robots. Inspired by these remarkable applications, we examine the basic mechanics of spermatozoa motility using an undulating sheet model. This undulating sheet is bounded between two rigid walls which is self-propeling in the negative axial direction. The liquid around the spermatozoa is taken as Carreau fluid with electro-osmotic properties. The application of the lubrication approximation results in the reduction of momentum equations. The resulting ODE is solved numerically via the finite difference method and MATLAB's built-in routine bvp5c. The unknowns that are present in the boundary conditions are refined by the root-finding algorithm. Power losses, cell speed, flow rate, velocity of the fluid, and streamline pattern are visualized by graphs. The findings of this study have important implications for the designing and optimization of electrically controlled microswimmers.

Abstract:Investigating sperm locomotion in the presence of an external fluid flow and geometries simulating the female reproductive tract can lead to a better understanding of sperm motion during the fertilization process. In this study, using a microfluidic device featuring a stricture that simulates the biophysical properties of narrow junctions inside the female reproductive tract, we observed the gate-like role the stricture plays to prevent sperm featuring motility below a certain threshold from advancing towards the fertilization site. At the same time, all sperm slower than the threshold motility accumulate before the stricture and swim in a butterfly-shaped path between the channel walls which maintains the chance of penetrating the stricture and thus advancing towards the egg. Interestingly, the accumulation of sperm before the stricture occurs in a hierarchical manner so that sperm with higher velocities remain closer to each other and as the sperm velocity drops, they spread further apart.

Abstract:We briefly describe the similarities of the experiments of sperm motion in microfluidic "strictures" by Zafeeani et al. in 2019 (Sci. Adv. 5, eaav21111, 2019) and those by Altshuler et al. in 2013 (Soft Matter 9, 1864, 2013). We shortly discuss the hydrodynamic elements justifying the strong resemblance between the two types of experiments, and suggest that other previous results in E. coli motion (Soft Matter 11, 6248, 2015) may shed further light on the understanding of sperm migration.

Abstract:Sperm motility analysis is an indispensable test in male infertility diagnosis. The current microscopic method cannot select the sperm of specific velocity for further in vitro Fertilization (IVF) usage or research purposes. Here, we report a novel and simple microfluidic device capable of balancing sperm's swimming speed with local flow velocity. The results suggested that sperm motility was quantitatively graded and sperm with different average straight line velocity (VSL) 40.5 ± 3.0, 27.2 ± 0.6 and 18.7 ± 0.6 μm/s respectively were automatically trapped in corresponding regions. Sperm motility classification (SMC) and sperm trapping may facilitate both the diagnosis of male infertility and further studies on sperm physiological behavior.

Abstract:We investigate the dynamics and hydrodynamics of a human spermatozoa swimming freely in 3D. We simultaneously track the sperm flagellum and the sperm head orientation in the laboratory frame of reference via high-speed high-resolution 4D (3D+t) microscopy, and extract the flagellar waveform relative to the body frame of reference, as seen from a frame of reference that translates and rotates with the sperm in 3D. Numerical fluid flow reconstructions of sperm motility are performed utilizing the experimental 3D waveforms, with excellent accordance between predicted and observed 3D sperm kinematics. The reconstruction accuracy is validated by directly comparing the three linear and three angular sperm velocities with experimental measurements. Our microhydrodynamic analysis reveals a novel fluid flow pattern, characterized by a pair of vortices that circulate in opposition to each other along the sperm cell. Finally, we show that the observed sperm counter-vortices are not unique to the experimental beat, and can be reproduced by idealised waveform models, thus suggesting a fundamental flow structure for free-swimming sperm propelled by a 3D beating flagellum.

Abstract:In recent decades, humans have faced greater challenges in reproduction. Assisted reproductive technology is the most prominent approach for addressing this problem. Current clinical screening methods simply consider the motility or morphology of the sperm. However, as the spermatozoa need to navigate over a long distance in the female reproductive tract and survive the natural screening processes therein, these methods are imperfect. Many approaches have been undertaken to study the chemotaxis and thermotaxis navigation behavior of spermatozoa, but few of these have involved integrated screening that considers motility, chemotaxis, and thermotaxis based on the biological environment of the human body. Current routine sperm evaluation techniques are inadequate and fail to simultaneously provide conclusive evidence for the thermotactic and chemotactic characteristics of sperm. Thus, such screening of functional spermatozoa will be an advancement in assisted reproduction. In this study, we developed a fully integrated biomimetic microfluidic system for screening sperm for their characteristics when exposed to temperature and chemical gradients. Based on our results, we showed that spermatozoa were attracted by temperature and chemical gradients in the physiological range. Moreover, we ascertained a suitable temperature gradient range for thermotaxis and statistically proved that the thermotactic and chemotactic responses are not linked. Here, we report the first quantitative study of functional sperm during thermotaxis and chemotaxis, and our analysis of the difference in motility caused by different conditions. More broadly, we foresee the clinical application of these biologically motivated parameters and characteristics in assisted reproduction in humans.

Abstract:The hydrodynamic behavior of swimming cells in a bounded domain filled with Oldroyd 8-constant fluid is expounded. Stokes equations normally hold for bodies swimming with slow speed, having small scales, and interacting with highly viscous medium. Besides fluid properties, gravitational force and angle of inclination are apt to enhance/reduce motility. In this work, we explore the motion of swimming sperms with gravitational effects. The two-dimensional infinite sheet (spermatozoa) is assumed in the channel center. The passage is declined with surface roughness effects. An appropriate numerical approach, the implicit finite difference method (IFDM) is adopted to solve the resulting differential equation. The speed of spermatozoa is simulated by Newton's root finder. Energy losses are also calculated numerically. The assistive factors are highlighted in the discussion section. Sperms that attach or interact with the cervix face challenges if the surface is irregular, potentially impacting their ability to move further.

Abstract:This paper presents a theoretical study of a non-linear rheological fluid transport in an axisymmetric tube by cilia. However, an attempt has been made to explain the role of cilia motion on the transport of fluid through the ductus efferentes of the male reproductive tract. Ostwald-de Waele power-law viscous fluid has been considered to represent the rheological fluid. To analyze pumping by means of a sequence of beat of cilia from row-to-row of cilia in a given row of cells and from one row of cells to the next (metachronal wave movement), we consider the conditions that the corresponding Reynolds number is small enough for inertial effects to be negligible and the wavelength to diameter ratio is large enough for the pressure to be considered uniform over the cross-section. Analyses and computations of the detailed fluid motion reveal that the time-averaged flow rate is dependent on $\epsilon$, a non-dimensional measure involving the mean radius $a$ of the tube and the cilia length. Thus, flow rate significantly varies with the cilia length. Moreover, the flow rate has been reported near to the estimated value $6\times 10^{-3}$$ml/h$ for human efferent ducts if $\epsilon$ is near by 0.4. The estimated value was suggested by Lardner and Shack [1] in human, based on the experimental observations for flow rates in efferent ducts of other animals, e.g., rat, ram and bull. In addition, the nature of the rheological fluid, i.e., the value of the fluid index $n$ strongly influences various flow-governed characteristics. The interesting feature of the paper is that the pumping improves with the thickening behavior for small values of $\epsilon$ or in free pumping ($\Delta P=0$) and pumping ($\Delta P>0$) regions.
Two-dimensional slither swimming of sperm within a micrometre of a surface

Abstract:Sperm motion near surfaces plays a crucial role in fertilization, but the nature of this motion has not been resolved. Using total internal reflection fluorescence microscopy, we selectively imaged motile human and bull sperm located within one micron of a surface, revealing a distinct two-dimensional (2D) ‘slither’ swimming mode whereby the full cell length (50–80 μm) is confined within 1 μm of a surface. This behaviour is distinct from bulk and near-wall swimming modes where the flagellar wave is helical and the head continuously rotates. The slither mode is intermittent (∼1 s, ∼70 μm), and in human sperm, is observed only for viscosities over 20 mPa·s. Bull sperm are slower in this surface-confined swimming mode, owing to a decrease in their flagellar wave amplitude. In contrast, human sperm are ∼50% faster—suggesting a strategy that is well suited to the highly viscous and confined lumen within the human fallopian tube.

Rheotaxis of Active Droplets

Abstract:Rheotaxis is a well-known phenomenon among microbial organisms and artificial active colloids, wherein the swimmers respond to an imposed flow. We report the first experimental evidence of upstream rheotaxis by spherical active droplets. It is shown that the presence of a nearby wall and the resulting strong flow-gradient at the droplet level is at the root of this phenomenon. Experiments with optical cells of different heights reveal that rheotaxis is observed only for a finite range of shear rates, independent of the bulk flow-rate. We conjecture that the flow induced distortion of an otherwise isotropic distribution of filled/empty micelles around the droplet propels it against the flow. We also show that nematic droplets exhibit elastic stress-induced oscillations during their rheotactic flight. A promising potential of manipulating the rheotactic behavior to trap as well as shuttle droplets between target locations is demonstrated, paving way to potentially significant advancement in bio-medical applications.

Abstract:Head rotation in human spermatozoa is essential for different swimming modes and fertilisation, as it links the molecular workings of the flagellar beat with sperm motion in three-dimensional (3D) space over time. Determining the direction of head rotation has been hindered by the symmetry and translucent nature of the sperm head, and by the fast 3D motion driven by the helical flagellar beat. Analysis has been mostly restricted to two-dimensional (2D) single focal plane image analysis, which enables tracking of head centre position but not tracking of head rotation. Despite the conserved helical beating of the human sperm flagellum, human sperm head rotation has been reported to be uni- or bi-directional, and even to intermittently change direction in a given cell. Here, we directly measure the head rotation of freely swimming human sperm using multi-plane 4D (3D+t) microscopy and show that: (1) 2D microscopy is unable to distinguish head rotation direction in human spermatozoa; (2) head rotation direction in non-capacitating and capacitating solutions, for both aqueous and viscous media, is counterclockwise (CCW), as seen from head to tail, in all rotating spermatozoa, regardless of the experimental conditions; and (3) head rotation is suppressed in 36% of spermatozoa swimming in non-capacitating viscous medium, although CCW rotation is recovered after incubation in capacitating conditions within the same viscous medium, possibly unveiling an unexplored aspect of the essential need of capacitation for fertilisation. Our observations show that the CCW head rotation in human sperm is conserved. It constitutes a robust and persistent helical driving mechanism that influences sperm navigation in 3D space over time, and thus is of critical importance in cell motility, propulsion of flagellated microorganisms, sperm motility assessments, human reproduction research, and self-organisation of flagellar beating patterns and swimming in 3D space.

Abstract:In this fluid mechanics video, we explore the kinematics of chemotaxing sperm cells (sea urchin, \textit{Arbacia punctulata}) swimming in a chemoattractant gradient. We demonstrate that the complex swimming trajectories resulting in chemotactic behavior (`turn-and-run' motility) are comprised of several distinct flagellar maneuvers. These motility patterns likely play an important role optimizing chemotaxic motility and navigation, when the sperm cells are subjected external fluid flows.

Lab on a Chip - Miniaturisation for Chemistry and Biology

Abstract:Sperm motility is a prerequisite for male fertility. Enhancing the concentration of motile sperms in assisted reproductive technologies – for human and animal reproduction – is typically achieved through aggressive methods such as centrifugation. Here, we propose a passive technique for the amplification of motile sperm concentration, with no externally imposed forces or flows. The technique is based on the disparity between probability rates, for motile cells, of entering and escaping from complex structures. The effectiveness of the technique is demonstrated in microfluidic experiments with microstructured devices, comparing the trapping power in different geometries. In these micro-traps, we observe an enhancement of cells' concentration close to 10, with a contrast between motile and non-motile cells increased by a similar factor. Simulations of suitable interacting model sperms in realistic geometries reproduce quantitatively the experimental results, extend the range of observations and highlight the components that are key to the optimal trap design.
