Abstract:Purpose: Overweight/obese individuals show impairments in executive functions such as inhibitory control. However, the neural mechanisms underlying these disturbances-and specifically, whether or not they involve altered activation of the specific prefrontal cortex regions-are not yet fully understood. Methods: The motivational dimensional model of affect suggests that high approach-motivated positive affect (e.g., desire) may impair executive function. In the present study, we investigated individual differences in neural responses to videos of food stimuli, and examined brain activity during a cognitive task in an approach-motivated positive state using functional near-infrared spectroscopy (fNIRS). In Experiment 1, in 16 healthy young adults, we tested whether prefrontal cortex activation differed during a food video clip versus a neutral video clip. Then, after viewing each video clip, we tested for differences in executive function performance and prefrontal cortex activation during a Stroop task. Experiment 2 was the same, except that we compared 20 overweight/obese with 20 healthy young adults, and it incorporated only the food video clip. Results and conclusions: The results of both experiments indicated that food stimuli decrease activation in regions of the prefrontal cortex related to executive function. This study also suggests that overweight/obese might consciously suppress their responses to a desired stimulus, yet here it seems that effect was less pronounced than in healthy controls. Food-related inhibitory control deficits in young male adults with obesity: Behavioral and ERP evidence from a food-related go/no-go task

Abstract:Purpose: Obesity poses a pervasive challenge to global public health, which is linked to adverse physical health outcomes and cognitive decline. Cognitive function, particularly food-related cognitive function, plays a critical role in sustaining a healthy weight and mitigating the progression of obesity. The aim of this study was to investigate the behavioral and neuroelectronic aspects of food-related inhibitory functions in young adult males with obesity. Methods: Forty-nine participants with obesity and healthy-weight were recruited (BMI = 35.83 ± 5.06 kg/m2 vs. 22.55 ± 1.73 kg/m2, age = 24.23 ± 4.55 years vs. 26.00 ± 3.97 years). A food-related Go/No-go task which included 6 distinct blocks in a randomized order was conducted to investigate the general and food-related inhibitory control. 180 stimulus images from the Food Picture Database encompassing high-calorie food, low-calorie food, and neutral images were selected. Behavioral (Go RT, Go ACC, No-go ACC) and event-related potential measures (N2 and P3 amplitude) during the food-related Go/No-go task were measured. Results: The main findings indicated that the group with obesity exhibited lower No-go accuracy, slower go reaction times, and smaller P3 amplitudes in high-calorie, low-calorie foods, and neutral picture, compared to the normal-weight group, but with no group difference in N2. Additionally, high-calorie food induced larger N2 and P3 amplitude than the neutral stimuli. Conclusions: Young male adults with obesity exhibit poorer inhibitory control in both food and non-food domains, specifically in slower reaction time and reduced accuracy, featuring difficulties in neural resource recruitment during the inhibitory control process. Additionally, the P3 component could serve as sensitive indicators to reveal the neural mechanisms of inhibitory control deficits in obesity, while the N2 and P3 components may differentiate the neural differences between high-calorie foods and non-foods in inhibitory control processing. Food, especially high-calorie food, induces more neural resources and may exacerbate the poor inhibitory ability towards food in obesity. Targeted interventions such as exercise interventions, cognitive training as well as neuromodulation interventions are warranted in the future to improve impaired general and food-related inhibitory functions in the obese populations, offering both theoretical and practical frameworks for obesity prevention and treatment.
“Free won't” of food in overweight and normal-weight adults: Comparison of neurocognitive correlates of intentional and reactive inhibitions

Abstract:Food-related inhibition plays a critical role in the manifestation of overweight. Previous research has focused exclusively on stimulus-driven (reactive) inhibition, which is different from intentional inhibition that refers to an internally generated decision to “stop”. This study investigated the food-related neurophysiological correlates of intentional and reactive inhibitions in overweight and normal-weight adults. We compared 35 overweight participants (OWs) and 34 normal-weight participants (NWs) on performance and electroencephalography-based measures during a food-related go/no-go/choose task. In this task, participants made reactive responses to an instructed go/no-go target or made intentional choices whether to execute or inhibit a keypress when presented with a free-choice target. Our results mainly showed, 1) for group-difference, N2a amplitudes of OWs were less negative than that of NWs in the intentional trials; 2) for source difference, N2a amplitudes were less negative in reactive condition than in intentional condition uniquely in OWs. Moreover, comparison across intentional responses revealed that P2 amplitudes in no-go trials were lower than in go trials. Additionally, a greater body mass index correlated with lower intentional no-go-P2 and reactive go/no-go-P2 amplitudes. These findings suggest that overweight is associated with deficits in food-related intentional inhibition, which is segregated from reactive inhibition. The individual differences in premotor inhibition during free-choice situations might provide an explanation for overeating behaviors in overweight adults' daily life. Further, our results refine the ERP marker of intentional inhibition from N2 to N2a, which could be an essential neural mechanism underlying the “free won't” of food in OWs.

Abstract:Meeting the energy demands of ultraendurance cycling requires careful planning and monitoring of food and fluid intake. This case study presents the nutrient intake of a cyclist while training for and competing in the Race Across AMerica (RAAM). Carbohydrate accounted for 65% of the calories consumed during training (4,743 kcal), 75% during 24-hr races (10,343 kcal), and 78% during RAAM (8,429 kcal). Gastrointestinal complaints during RAAM included nausea, feeling of fullness, and abdominal distension. Although probably exacerbated by sleep deprivation, these problems were all diet related. Based on this experience, it appears that by controlling the carbohydrate concentration of beverages, limiting dietary fiber, and relying on carbohydrate as the primary energy source, one could both control gastrointestinal symptoms and promote optimal performance in training and in ultramarathon cycling.
Mild hunger elicits attentional desensitization to visual food cues in healthy, non-obese individuals

Abstract:Introduction Food is a vital human need, and the human visual system is finely tuned to detect and respond to food cues in the environment. The omnipresence of food cues across various settings has been linked to the prevalence of obesity in susceptible populations. However, the influence of the post-prandial state on visual attention to food stimuli remains poorly understood. This study aimed to elucidate how a 12 hour fast affects visual attention to food and non-food stimuli in healthy, non-obese individuals. Methods Visual attention was assessed by measuring the total duration of visual fixations on stimuli presented on a computer screen, using a screen-based eye tracker (Tobii X2-60). Participants were divided into two groups: those who had fasted for 12 hours and those tested within two hours after consuming breakfast (satiated state). Additionally, performance on the Food Stroop task and electrodermal activity (EDA) responses were measured to evaluate attentional interference and physiological arousal, respectively. Salivary samples were also collected to assess levels of alpha-amylase and cortisol. Results Fasted participants exhibited a progressive decline in visual attention toward food stimuli compared to satiated individuals, reflecting a satiated state. This effect was independent of the palatability of the depicted food items and was not observed with stimuli representing non-food items. The Food Stroop task revealed no differences between fasting and satiated participants, indicating that the presence of food-related stimuli does not differentially impact attentional interference under varying hunger states. Moreover, no significant variations were observed in EDA responses across participant groups and stimulus types, suggesting that the modulation of visual attention to food cues by hunger is independent of physiological arousal. Interestingly, satiated subjects exhibited higher levels of salivary alpha-amylase, which was inversely related to their subjective hunger ratings. No differences in salivary cortisol levels were found between groups. Discussion The findings indicate a novel influence of mild hunger on the processing of visual food cues, independent of physiological arousal. The decline in visual attention to food stimuli in fasted individuals suggests that satiety modulates visual processing. The lack of differences in attentional interference and physiological arousal between fasting and satiated states further supports the notion that visual attention to food cues is primarily driven by hunger-related mechanisms rather than stress. Additionally, the inverse relationship between salivary alpha-amylase levels and hunger ratings implies that alpha-amylase may serve as a marker of satiety rather than stress.

Abstract:Efficient detection of food is important for an organism's survival. The results of previous experimental studies are consistent with this statement: food is detected in photographs faster than non-food items. Moreover, fat content modulates the speed of food detection. However, it is not known whether such sensitivity to the fat content of food is modulated by participants' internal states. To investigate these issues, we measured reaction times during a visual search task in which participants in fasting and postprandial states detected high-fat food (fast food), low-fat food (Japanese diet), and non-food (kitchen utensils) targets within crowds of non-food distractors (cars). Our results indicate that both hungry and satiated groups detected food targets more rapidly than non-food targets. The high-fat foods were detected more rapidly than low-fat foods in the hungry group, whereas no difference was observed between reaction times when satiated participants detected high-fat and low-fat food targets. These results suggest that food captures our visual attention even when we are satiated, and that fat detection efficiency is heightened when we are hungry.

Abstract:Background The loss of self-control or inability to resist tempting/rewarding foods, and the development of less healthful eating habits may be explained by three key neural systems: (1) a hyper-functioning striatum system driven by external rewarding cues; (2) a hypo-functioning decision-making and impulse control system; and (3) an altered insula system involved in the translation of homeostatic and interoceptive signals into self-awareness and what may be subjectively experienced as a feeling. Methods The present study examined the activity within two of these neural systems when subjects were exposed to images of high-calorie versus low-calorie foods using functional magnetic resonance imaging (fMRI), and related this activity to dietary intake, assessed by 24-hour recall. Thirty youth (mean BMI = 23.1 kg/m 2 , range = 19.1 - 33.7; age =19.7 years, range = 14 - 22) were scanned using fMRI while performing food-specific go/nogo tasks. Results Behaviorally, participants more readily pressed a response button when go trials consisted of high-calorie food cues (HGo task) and less readily pressed the response button when go trials consisted of low-calorie food cues (LGo task). This habitual response to high-calorie food cues was greater for individuals with higher BMI and individuals who reportedly consume more high-calorie foods. Response inhibition to the high-calorie food cues was most difficult for individuals with a higher BMI and individuals who reportedly consume more high-calorie foods. fMRI results confirmed our hypotheses that (1) the "habitual" system (right striatum) was more activated in response to high-calorie food cues during the go trials than low-calorie food go trials, and its activity correlated with participants’ BMI, as well as their consumption of high-calorie foods; (2) the prefrontal system was more active in nogo trials than go trials, and this activity was inversely correlated with BMI and high-calorie food consumption. Conclusions Using a cross-sectional design, our findings help increase understanding of the neural basis of one’s loss of ability to self-control when faced with tempting food cues. Though the design does not permit inferences regarding whether the inhibitory control deficits and hyper-responsivity of reward regions are individual vulnerability factors for overeating, or the results of habitual overeating.
The effect of hunger state on hypothalamic functional connectivity in response to food cues

Abstract:The neural underpinnings of the integration of internal and external cues that reflect nutritional status are poorly understood in humans. The hypothalamus is a key integrative area involved in short- and long-term energy intake regulation. Hence, we examined the effect of hunger state on the hypothalamus network using functional magnetic resonance imaging. In a multicenter study, participants performed a food cue viewing task either fasted or sated on two separate days. We evaluated hypothalamic functional connectivity (FC) using psychophysiological interactions during high versus low caloric food cue viewing in 107 adults (divided into four groups based on age and body mass index [BMI]; age range 24-76 years; BMI range 19.5-41.5 kg/m2 ). In the sated compared to the fasted condition, the hypothalamus showed significantly higher FC with the bilateral caudate, the left insula and parts of the left inferior frontal cortex. Interestingly, we observed a significant interaction between hunger state and BMI group in the dorsolateral prefrontal cortex (DLPFC). Participants with normal weight compared to overweight and obesity showed higher FC between the hypothalamus and DLPFC in the fasted condition. The current study showed that task-based FC of the hypothalamus can be modulated by internal (hunger state) and external cues (i.e., food cues with varying caloric content) with a general enhanced communication in the sated state and obesity-associated differences in hypothalamus to DLPFC communication. This could potentially promote overeating in persons with obesity.
Food go/no-go training alters neural circuits for food evaluation for appetite reduction

Abstract:Food go/no-go training has been traditionally categorized as a type of inhibitory training that decreases the desire for high-calorie food consumption. This training requires participants to either respond or withhold their responses to presented items with go cues or food items with no-go cues, respectively. Recent findings have suggested that this training may devalue food items associated with no-go cues instead of facilitating inhibitory control, leading to reduced food consumption. We thus hypothesized that food go/no-go training would alter the brain response to food items with no-go cues in food evaluation regions. To examine this hypothesis, we conducted a repeated measures functional magnetic resonance imaging using food images in healthy participants, who underwent 3 weeks of food go/no-go training (n = 26) using high- and low-calorie food items paired with no-go cues (no-go food) and go cues (go food), respectively, and control training (n = 24). The food go/no-go training reduced the ratings for the desire to eat no-go foods and increased the ratings for go foods. The reduction in no-go food rating was positively associated with a decrease in daily snack intake. The neural responses in the food evaluation regions increased for go foods. Moreover, the functional connectivity of those regions was altered. The food go/no-go training did not decrease impulsivity traits or increase restrained eating, which are associated with inhibitory control. Overall, food go/no-go training influenced the brain regions associated with food evaluation, thus devaluating no-go foods and reducing the daily snack intake. Accordingly, food go/no-go training could promote healthier food choices.

Abstract:Introduction: In the modern obesogenic environment, heightened reactivity to food-associated cues plays a major role in overconsumption by evoking appetitive responses. Accordingly, functional magnetic resonance imaging (fMRI) studies have implicated regions of the salience and rewards processing in this dysfunctional food cue-reactivity, but the temporal dynamics of brain activation (sensitization or habituation over time) remain poorly understood. Methods: Forty-nine obese or overweight adults were scanned in a single fMRI session to examine brain activation during the performance of a food cue-reactivity task. A general linear model (GLM) was used to validate the activation pattern of food cue reactivity in food > neutral contrast. The linear mixed effect models were used to examine the effect of time on the neuronal response during the paradigm of food cue reactivity. Neuro-behavioral relationships were investigated with Pearson's correlation tests and group factor analysis (GFA). Results: A linear mixed-effect model revealed a trend for the time-by-condition interactions in the left medial amygdala [t(289) = 2.21, β = 0.1, P = 0.028], right lateral amygdala [t(289) = 2.01, β = 0.26, P = 0.045], right nucleus accumbens (NAc) [t(289) = 2.81, β = 0.13, P = 0.005] and left dorsolateral prefrontal cortex (DLPFC) [t(289) = 2.58, β = 0.14, P = 0.01], as well as in the left superior temporal cortex [42 Area: t(289) = 2.53, β = 0.15, P = 0.012; TE1.0_TE1.2 Area: t(289) = 3.13, β = 0.27, P = 0.002]. Habituation of blood-oxygenation-level-dependent (BOLD) signal during exposure to food vs. neutral stimuli was evident in these regions. We have not found any area in the brain with significant increased response to food-related cues over time (sensitization). Our results elucidate the temporal dynamics of cue-reactivity in overweight and obese individuals with food-induced craving. Both subcortical areas involved in reward processing and cortical areas involved in inhibitory processing are getting habituated over time in response to food vs. neutral cues. There were significant bivariate correlations between self-report behavioral/psychological measures with individual habituation slopes for the regions with dynamic activity, but no robust cross-unit latent factors were identified between the behavioral, demographic, and self-report psychological groups. Discussion: This work provides novel insights into dynamic neural circuit mechanisms supporting food cue reactivity, thereby suggesting pathways in biomarker development and cue-desensitization interventions.

Abstract:Recent studies have shown that spontaneous pre-stimulus fluctuations in brain activity affect higher-order cognitive processes, including risky decision-making, cognitive flexibility, and aesthetic judgments. However, there is currently no direct evidence to suggest that pre-choice activity influences value-based decisions that require self-control. We examined the impact of fluctuations in pre-choice activity in key regions of the reward system on self-control in food choice. In the functional magnetic resonance imaging (fMRI) scanner, 49 participants made 120 food choices that required self-control in high and low working memory load conditions. The task was designed to ensure that participants were cognitively engaged and not thinking about upcoming choices. We defined self-control success as choosing a food item that was healthier over one that was tastier. The brain regions of interest (ROIs) were the ventral tegmental area (VTA), putamen, nucleus accumbens (NAc), and caudate nucleus. For each participant and condition, we calculated the mean activity in the 3-s interval preceding the presentation of food stimuli in successful and failed self-control trials. These activities were then used as predictors of self-control success in a fixed-effects logistic regression model. The results indicate that increased pre-choice VTA activity was linked to a higher probability of self-control success in a subsequent food-choice task within the low-load condition, but not in the high-load condition. We posit that pre-choice fluctuations in VTA activity change the reference point for immediate (taste) reward evaluation, which may explain our finding. This suggests that the neural context of decisions may be a key factor influencing human behavior.

Unraveling Interoceptive Processing and Action Dynamics: Exploring Neural and Psychological Responses to Food Cues Using fMRI

Abstract:Interoception, the perception of body signals, which is crucial for maintaining metabolic homeostasis, is asserted to play a vital role in obesity. Despite conceptual assumptions that impaired insular interoceptive processing contributes to overeating behaviors predominantly through modulating motor cortices, this link has not been extensively explored. Therefore, to further investigate neural mechanisms underlying insula-based interoceptive processing, this pre-registered (PMC9003175) study assessed blood oxygenation level-dependent (BOLD) responses via functional magnetic resonance tomography (fMRI) in 45 healthy participants (31 females/14 males, age 35.78 plus-or-minus sign 10 years, BMI 29.52 plus-or-minus sign 3.5 kg/m2) during a block-designed food cue reactivity task. Region of interest (insula) and whole brain voxel-wise correlation analyses explored neural correlates of visceral interoception. Furthermore, group factor analysis (GFA) unveiled coherence patterns between neural (fMRI) and psychological/behavioral measures. At the psychological level, self-reported hunger (P < 0.01, d = 0.82) and food craving (P < 0.01, d = 0.68) significantly increased, while craving control (P = 0.04, d = 0.37) decreased after cue exposure. Voxel-wise correlation analysis identified positive correlations (P < 0.01) between visceral interoception and activation of the precentral gyrus (PrG or motor cortex), insula, inferior frontal gyrus (IFG), posterior cingulate cortex (PCC), and superior parietal lobule (SPL). Moreover, altered functional connectivity dynamics were noted within the insula-PrG-IFG network during food cue exposure, with a significant reduction of IFG-PrG connections (P = 0.05). Interestingly, GFA identified a cross-unit latent factor across neural and psychological/behavioral measures. Overall, our findings indicate that altered interoceptive processing (insula activity), motor planning (motor cortex activity), and diminished inhibitory control (negative IFG-PrG connectivity) collectively contribute to food craving generation and potentially subsequent actions toward food consumption.

Hunger and BMI modulate neural responses to sweet stimuli: fMRI meta-analysis

Abstract:Objective: Consuming sweet foods, even when sated, can lead to unwanted weight gain. Contextual factors, such as longer time fasting, subjective hunger, and body mass index (BMI), may increase the likelihood of overeating. Nevertheless, the neural mechanisms underlying these moderating influences on energy intake are poorly understood. Methods: We conducted both categorical meta-analysis and meta-regression of factors modulating neural responses to sweet stimuli, using data from 30 functional magnetic resonance imaging (fMRI) articles incorporating 39 experiments (N = 995) carried out between 2006 and 2019. Results: Responses to sweet stimuli were associated with increased activity in regions associated with taste, sensory integration, and reward processing. These taste-evoked responses were modulated by context. Longer fasts were associated with higher posterior cerebellar, thalamic, and striatal activity. Greater self-reported hunger was associated with higher medial orbitofrontal cortex (OFC), dorsal striatum, and amygdala activity and lower posterior cerebellar activity. Higher BMI was associated with higher posterior cerebellar and insular activity. Conclusions: Variations in fasting time, self-reported hunger, and BMI are contexts associated with differential sweet stimulus responses in regions associated with reward processing and homeostatic regulation. These results are broadly consistent with a hierarchical model of taste processing. Hunger, but not fasting or BMI, was associated with sweet stimulus-related OFC activity. Our findings extend existing models of taste processing to include posterior cerebellar regions that are associated with moderating effects of both state (fast length and self-reported hunger) and trait (BMI) variables.

Abstract:Numerous findings from functional neuroimaging research suggest that overweight may be associated with alterations in reactive inhibition. However, there is a dearth of research investigating the functional connectivity that mediates intentional inhibition in overweight individuals. To explore this issue, 55 overweight and 45 normal-weight adults completed an assessment consisting of a resting-state functional magnetic resonance imaging scan, a behavioural task measuring food-specific intentional inhibition, and a 1-year longitudinal measurement of BMI change. A seed-based approach was employed to examine the group-difference of the resting-state functional connectivity (rsFC) of the medial frontal cortex (MFC) (dorsal fronto-medial cortex [dFMC], pre-supplementary motor area, and premotor cortex) regions involved in intentional inhibition. Compared with normal-weight adults, the overweight individuals exhibited higher rsFC between the MFC seeds and (i) cerebellum, (ii) postcentral gyrus, (iii) middle temporal gyrus, and (iv) posterior cingulate cortex, while lower rsFC strength were observed between MFC seeds and (i) putamen and (ii) insula. The overweight individuals with higher dFMC–cerebellum rsFC strength showed poorer performance in food-specific intentional inhibition and gained more weight a year later than those of normal-weight participants. Results suggested that altered functional connections between MFC and regions associated with reward and maladaptive eating may be key neural mechanisms of food-specific intentional inhibition in overweight status. Therefore, individuals are encouraged to make informed decisions about their health and reduce their consumption of obesogenic foods from the perspective of intentional inhibition.

Abstract:Background: Response inhibition, an important domain of executive function (EF), involves the ability to suppress irrelevant or interfering information and impulses. Previous studies have shown impairment of response inhibition in high functioning autism (HFA) and attention deficit hyperactivity disorder (ADHD), but more recent findings have been inconsistent. To date, almost no studies have been conducted using functional imaging techniques to directly compare inhibitory control between children with HFA and those with ADHD.Method: Nineteen children with HFA, 16 age-and intelligence quotient (IQ)-matched children with ADHD, and 16 typically developing (TD) children were imaged using functional near-infrared spectroscopy (NIRS) while performing Go/No-go and Stroop tasks.Results: Compared with the TD group, children in both the HFA and ADHD groups took more time to respond during the No-go blocks, with reaction time longest for HFA and shortest for TD. Children in the HFA and ADHD groups also made a greater number of reaction errors in the No-go blocks than those in the TD group. During the Stroop task, there were no significant differences between these three groups in reaction time and omission errors. Both the HFA and ADHD groups showed a higher level of inactivation in the right prefrontal cortex (PFC) during the No-go blocks, relative to the TD group. However, no significant differences were found between groups in the levels of oxyhemoglobin concentration in the PFC during the Stroop task.Conclusion: Functional brain imaging using NIRS showed reduced activation in the right PFC in children with HFA or ADHD during an inhibition task, indicating that inhibitory dysfunction is a shared feature of both HFA and ADHD.

Abstract:In our food-rich environment we must constantly resist appealing food in order to maintain a healthy lifestyle. Previous studies have found that food-specific inhibition training can produce changes in eating behaviour, such as a reduction in snack consumption. However, the mechanisms that drive the effect of inhibition training on eating behaviour remain unknown. Identifying the mechanism underlying food-specific inhibition training could lead to more targeted training interventions increasing the potential efficacy of such interventions. In the current study, we investigated directly whether training-induced effects on inhibitory control might underlie the predicted change in eating behaviour. Healthy individuals who scored high on uncontrolled eating were randomly assigned to receive six online training sessions over six consecutive days of either food-specific response inhibition training (active group; n = 21) or response inhibition training without food stimuli (control group; n = 20). We measured pre- and post-training inhibitory control in the context of food and food cue sensitivity, as well as food consumption in a bogus taste test. As expected, food-specific inhibition training decreased snack consumption in the bogus taste test relative to control training. However, the active training did not improve inhibitory control towards food, nor did it reduce food cue sensitivity above and beyond the control training. Future studies are needed to investigate the potential underlying mechanism of food-specific inhibition training, as it remains unclear what drives the reliable effect on eating behaviour.

Abstract:The food environment can interact with cognitive processing and influence eating behaviour. Our objective was to characterize the impact of implicit olfactory priming on inhibitory control towards food, in groups with different weight status. Ninety-one adults completed a modified Affective Shifting Task: they had to detect target stimuli and ignore distractor stimuli while being primed with non-attentively perceived odours. We measured reactivity and inhibitory control towards food pictures. Priming effects were observed on reactivity: participants with overweight and obesity were slower when primed with pear and pound cake odour respectively. Common inhibitory control patterns toward foods were observed between groups. We suggest that non-attentively perceived food cues influence bottom-up processing by activating distinguished mental representations according to weight status. Also, our data show that cognitive load influences inhibitory control toward foods. Those results contribute to understanding how the environment can influence eating behaviour in individuals with obesity.

Abstract:Postprandial increases of gastrointestinal hormones are associated with reduced energy intake, partially through direct effects on the brain. However, it remains unknown whether the fasting levels of gastrointestinal hormones are associated with altered brain activity in response to visual food stimuli. Thus, we performed a whole‐brain regression analysis to assess the association between fasting brain activations according to fMRI during viewing of highly desirable vs. less desirable food images with fasting levels of five gastrointestinal hormones (GLP‐1, GLP‐2, oxyntomodulin, glicentin and GIP) in thirty‐six subjects with obesity cross‐sectionally. We observed that fasting blood levels of GIP were inversely associated with the activation of attention‐related areas (visual cortices of the occipital lobe, parietal lobe) and of oxyntomodulin and glicentin with reward‐related areas (insula, putamen, caudate for both and additionally with orbitofrontal cortex for glicentin) and hypothalamus when viewing highly desirable as compared to less desirable food images. Future studies are needed to confirm whether fasting levels of oxyntomodulin, glicentin and GIP are associated with the activation of brain areas involved in appetite regulation and with energy intake in people with obesity.This article is protected by copyright. Abstract:The lack of inhibitory control toward foods may cause unhealthy eating behavior and lead to obesity. However, previous research failed to reach consensus on the alterations in event-related potential (ERP) markers of inhibitory control, i.e., N2 and P3. We hypothesized that the ERP effects of inhibitory control reported in previous food-based Go/NoGo studies might be obscured by non-inhibitory processes associated with stimulus probability. We designed two food-based Go/NoGo tasks composed of stimuli with the same type and frequency of occurrence (60% non-foods, 20% high-calorie foods, 20% low-calorie foods), one with response inhibition toward high- and the other toward low-calorie foods. Such an experimental design allowed us to isolate neural activity associated with inhibitory control from that associated with non-inhibitory processes by constructing ERP difference waves between NoGo and Go trials with the same frequency of occurrence. Electroencephalography data were collected from 32 obese participants and 29 normal-weight controls. Obese participants showed significantly lower accuracy in NoGo trials than normal-weight controls in both tasks. ERP data suggested inhibition-related effects for P3 (P3d) but not N2 in the difference waves, and obese participants showed significantly decreased P3d amplitudes than normal-weight controls in both tasks. In addition, we found that across both groups, individuals with larger waist-to-hip ratios showed smaller P3d amplitudes in both tasks, while such correlations between body mass index and P3d amplitude were only observed in the high-calorie task. Our findings suggest that the decreased effect of P3, not N2, might reflect the neural substrate of inhibitory control deficits in obese people. Thus, P3 could serve as an important neural marker in the future development of new therapeutic strategy that aims to improve inhibitory control in obesity.
