Abstract:Ester-containing deltamethrin pesticides are widely used in farmland and have inevitable side effects on the biosphere and human health. Microbia have been used for efficient degradation of deltamethrin, but the related mechanism and enzyme characteristics have not been elucidated. In this study, a species Brevibacillus parabrevis BCP-09 could degrade up to 75 mg L −1 deltamethrin with a degradation efficiency of 95.41%. Proteomic and genomic methods were used to explore its degradation mechanism. Enzymes belonged to hydrolases, oxidases and aromatic compound degrading enzymes were expressed enhanced and might participate in the deltamethrin degradtion. RT-PCR experiment and enzyme activity analysis verified the degradation of deltamethrin by bacterial protein. Additionally, the formation of endospores can help strain BCP-09 resist the toxicity of deltamethrin and enhance its degradation. This study supplies a scientific evidence for the application of Brevibacillus parabrevis BCP-09 in the bioremediation of environmental pollution and enriches the resources of deltamethrin-biodegradable proteins.

Abstract:A fungal strain capable of utilizing chlorpyrifos as sole carbon and energy sources was isolated from soil by enrichment cultivation approach. The half-lives of degradation (DT50) for chlorpyrifos at concentrations of 1, 10, and 100 mg l(-1) by the fungal strain DSP in mineral salt medium were measured to be 2.03, 2.93, and 3.49 days, respectively. Two cell-free extracts [E (1:10) and E (1:20)] from the fungal strain DSP in bran-glucose medium were prepared and used to enhance chlorpyrifos degradation on vegetables. Compared with the controls, the DT50 of chlorpyrifos were reduced by 70.3%, 65.6%, 80.6%, 80.6%, and 86.1%, and by 53.8%, 43.2%, 66.0%, 54.3%, and 67.7% on E (1:20) and E (1:10) treated pakchoi, water spinach, Malabar spinach, haricot beans, and pepper, respectively. The 7-day residual values (R (7)) of chlorpyrifos on E (1:10) treated vegetables were all lower than the corresponding maximum residue levels of European Union (EU MRLs), except that the R (7) value on haricot beans was slightly higher than the corresponding EU MRLs. The results indicate that cell-free extracts could rapidly degrade chlorpyrifos residues on vegetables.
Characterization of a Novel Cyfluthrin-Degrading Bacterial Strain Brevibacterium Aureum and Its Biochemical Degradation Pathway.

Abstract:Brevibacterium aureum DG-12, a new bacterial strain isolated from active sludge, was able to degrade and utilize cyfluthrin as a growth substrate in the mineral medium. Response surface methodology using central composite rotatable design of cultural conditions was successfully employed for optimization resulting in 88.6% degradation of cyfluthrin (50 mg L-1) within 5 days. The bacterium degraded cyfluthrin by cleavage of both the carboxylester linkage and diaryl bond to form 2,2,3,3-tetramethyl-cyclopropane-methanol, 4-fluoro-3-phenexy-benzoic acid, 3,5-dimethoxy phenol, and phenol, and subsequently transformed these compounds with a maximum specific degradation rate, half-saturation constant and inhibition constant of 1.0384 day(-1), 20.4967 mg L-1, and 141.9013 mg L-1, respectively. A novel degradation pathway for cyfluthrin was proposed based on analysis of these metabolites. In addition, this strain was found capable of degrading a wide range of synthetic pyrethroid insecticides. Our results suggest that B. aureum DG-12 may be an ideal microorganism for bioremediation of the pyrethroid-contaminated environments. (C) 2013 Elsevier Ltd. All rights reserved.
Primary Study on the Characteristics of Crude Pyrethroid Pesticide-Degrading Enzyme Extracted from Pseudomonas diminuta Strain M5R14

Abstract:One Pseudomonas diminuta strain M5R14 was selected as object in the present study, which was isolated from the tame sludge in water course of Pesticide Factory of Hangzhou and could degrade a variety of strains of synthetic pyrethroid insecticides. The inner and extra-cell crude enzyme of Pseudomonas diminuta strain M5R14 prepared by ultrasonic break up and centrifugal separation methods, and the characteristics of crude pyrethroid pesticide-degrading enzyme were primary studied. The results showed that pyrethroid pesticides degrading enzyme excreted from the strain of was intracellular enzyme, the best pH for bifenthrin, fenpropathrin and cypermethrin degradation was 6.5, as well as the most appropriate temperature was 35°C. Besides, the Michaelis constant and maximum degradation rate varied due to pyrethroid species. The Michaelis constant to such 3 pyrethroid pesticides were 4162.73, 1092.57, 1171.97 μmol/L respectively, and the maximum degradation rate to such 3 pyrethroid pesticides were 0.152, 0.038, 0.043 μmol/(min·mg) protein. The degradation process of crude degrading enzymes to such 3 pyrethroid pesticides was fitted for first-order kinetic equation and the half life (t 1/2) were 119.51, 113.63, 108.30 min, respectively. Crude degrading enzymes for the three pyrethroid insecticides could be maintained high degradation activity when the pH ranging from 5.0 to 8.0, and the temperature was 20°C to 50°C, indicating the degrading enzymes were with large scope of pH stability and thermalstability. Therefore, there's higher potential in controlling the residuals of synthetic pyrethroid insecticides.

Abstract:Commonly used flame retardants, such as polybrominated diphenyl ethers, are extremely persistent in the environment, causing serious environmental risks. Certain strains of bacteria are able to degrade several low brominated congeners of PBDEs aerobically. However, the aerobic degradation pathway is not yet well understood, particularly at the genetic level. In this study, we isolated Cupriavidus sp. WS from the environment that could degrade diphenyl ether (DE), 4-bromodiphenyl ether, and 4,4'-bromodiphenyl ether. DE was completely degraded in 6 days without any detectable end-product. Using transposon mutagenesis, several DE degradation-deficient mutants were obtained. Knocking out bphA1, bphA2, and bphA3 eliminated the ability of the Cupriavidus sp. WS bacterium to degrade DE, indicating that the bph genes play a crucial role in DE degradation by this strain. The specific roles of bphA, bphB, and bphC were identified by systematically expressing these genes in Escherichia coli. The dihydrodiol product of BphA was dehydrogenated into 2,3-dihydroxydiphenyl ether by BphB. 2,3-Dihydroxydiphenyl ether was then decomposed into phenol and 2-pyrone-6-carboxylic acid by BphC. Thus, BphA, BphB, and BphC act sequentially in the aerobic degradation of DE, 4-bromodiphenyl ether, and 4,4'-dibromodiphenyl ether by the Cupriavidus sp. WS bacterium.
Cloning,Analysis and Fusion Expression of Methyl Parathion Hydrolase

Abstract:Methyl parathion(MP) is a kind of organophosphorous pesticide which was widely used by farmers all over the world in 1990's.It is effective in controlling pesticide but harmful to human beings.Bioremediation is an effective and economic method to treat environment that has been polluted by hazardous organic compounds,so researchers paid much attention in this area.Most of which focused on isolating functional bacteria,studying its degrading mechanism,and cloning degradation related genes.DLL 1 was isolated from soil polluted by MP for a long time and identified as Pseudomonas putida .It could degrade MP to CO 2 and H2 O [9] .DLL E4 with higher degrading ability was obtained by UV and LiCl mutagenesis of DLL 1 [10] .A big plasmid was found in DLL E4 but very difficult to be cured,and mutant M -P + losing activity of MP hydrolase by nitriguanidine mutagenesis also had this plasmid.So shotgun cloning method was chosen to clone the MP degradation gene( mpd ) from DLL E4.Total DNA was digested by Sau3AI,the fragments of 2～4 kb were recovered and inserted in pUC19 at BamHI site.The recombinant plasmids were transferred to DH5α.Six positive clones (pDT1～6) with MP hydrolase activity were selected on LB+MP plate,and pDT3 was chosen to be sequenced for its smallest molecular weight (inserted fragment 2 5 kb).After sequencing and analyzing of the inserted fragment,769～1794 region was considered as the most possible ORF and this region was cloned by PCR and ligated with pET 32a to construct an expressing vector pET MP.pET MP was transferred to E.coli BL21(DE3).The inducing time,concentration of IPTG and lactose were also studied.The result showed that the molecular weight of MP hydrolase was about 36 kDa,the iptimal inducing time and concentration for lactose were 2h and 2%(V/V) respectively. mpd was primary located in chromosome DNA in DLL E4 by PCR.
Isolation and Identification of Degradation Bacteria Enterobacter Aerogenes for Pyrethriods Pesticide Residues and Its Degradation Characteristics

Abstract:By incubation experiment, the bacterial strain labeled as M6R9 was isolated from the tame sludge in water course of Pesticide Factory of Hangzhou, and was identified as Enterobacter aerogenes, which had highly efficient degradation for Bifenthrin, Fenpropathrin and Cypermethrin. By investigating the physiological characteristics of the strain, the results show that the bacterium is a gram-negative aerobe bacilli, size is (0.8-1.9) microm x (0.5-1.0) microm, and is capable of utilizing Bifenthrin, Fenpropathrin and Cypermethrin as sole carbon source. Under the condition of ventilation, (25-30) degrees C, inoculated amount at D(415 nm) 0.2, pH 7.0, pesticide concentration 100 mg x L(-1) and vibrational speed 180 r x min(-1), the degradation efficiencies to Bifenthrin, Fenpropathrin and Cypermethrin are the highest by strain M6R9. Under such condition, in the mixture culture medium with 100 mg x L(-1) Bifenthrin, Fenpropathrin and Cypermethrin, the degradation ratios are 55.74%, 55.11% and 55.96% after culturing 3 d, respectively, the degradation processes are fitted for first-order kinetic equation and the half lives (t(1/2)) are 65.4,70.7 and 68.6 h respectively. The degradation ability of Enterobacter aerogenes M6R9 on Bifenthrin, Fenpropathrin and Cypermethrin is positively correlated to inoculated amount,vibrational speed and ventilation.
Study on Degrading Performance of Two Efficient Mixed Strains for Synthetic Pyrethroid Insecticides Degradation

Abstract:Two efficient strains of synthetic pyrethroid insecticides-degrading bacteria,M6R9 and M5R14,isolated from the tame sludge of sewer in pesticide factory of Hangzhou,which can degrade bifenthrin,fenpropathrin and cypermethrin simultaneously,were identified as Enterobacter aerogenes and Pseudomonas diminuta,respectively.By comparing degradation rates of the two strains independently and in mixed types, it′s found that the degradation rate of bifenthrin,fenpropathrin and cypermethrin was related to inoculated amount at OD_(415nm),and the degradation process was fitted to first-order kinetic equation.Under the optimum inoculated amount at OD_(415nm)=0.2 (the mixed strains M6R9 and M5R14 respectively inoculated OD_(415nm)=0.1 at equal proportion,while the single strain M6R9 or M5R14 inoculated OD_(415nm)=0.2),the degradation rate of mixed strains for bifenthrin,fenpropathrin and cypermethrin (100 mg/L) increased by 2.5% ,3.4% ,2.3% or 14.5% ,14.6% ,15.5% ,and the half-life (T1/2) was shorten by 8.1,14.8,13.1 h or 40.3,50.7,46.4 h,compared with the single strain M6R9 or M5R14 after culturing 3 days in the condition of pH 7.0,30 ℃,180 r/min.It′s proposed that there′s a synergistic effect between M6R9 and M5R14 on bifenthrin,fenpropathrin and cypermethrin.
Microbial Fermentation as an Efficient Method for Eliminating Pyrethroid Pesticide Residues in Food: A Case Study on Cyfluthrin and Aneurinibacillus aneurinilyticus D-21

Abstract:The microbial fermentation of food has emerged as an efficient means to eliminate pesticide residues in agricultural products; however, the specific degradation characteristics and mechanisms remain unclear. In this study, a Gram-positive bacterium, Aneurinibacillus aneurinilyticus D-21, isolated from fermented Pixian Douban samples exhibited the capability to degrade 45 mg/L of cyfluthrin with an efficiency of 90.37%. Product analysis unveiled a novel cyfluthrin degradation pathway, involving the removal of the cyanide group and ammoniation of the ester bond into an amide. Whole genome analysis discovered the enzymes linked to cyfluthrin degradation, including nitrilase, esterase, carbon-nitrogen ligases, and enzymes associated with aromatic degradation. Additionally, metabolome analysis identified 140 benzenoids distributed across various aromatic metabolic pathways, further substantiating D-21's catabolic capability toward aromatics. This study underscores the exceptional pyrethroid degradation prowess of A. aneurinilyticus D-21, positioning it as a promising candidate for the biotreatment of pesticide residues in food systems.

Abstract:Organophosphorus pesticides are highly toxic phosphate compounds with the general structure of O=P(OR)3 and threaten human health seriously. Methyl parathion hydrolase from microbial is an important enzyme to degrade organophosphorus pesticides (OPs) into less toxic or nontoxic compounds likep-nitrophenol and diethyl phosphate. Here, a gene encoding methyl parathion hydrolase from Azohydromonas australica was firstly cloned and expressed in Escherichia coli. The recombinant hydrolase showed its optimal pH and temperature at pH 9.5 and 50 °C. Leveraging 1 mM Mn2+, the enzyme activity was significantly enhanced by 29.3-fold, and the thermostability at 40 and 50 °C was also improved. The recombinant MPH showed the specific activity of 4.94 and 16.0 U/mg towards methyl parathion and paraoxon, respectively. Moreover, A. australica MPH could effectively degrade various of OPs pesticides including methyl parathion, paraoxon, dichlorvos and chlorpyrifos in a few minutes, suggesting a great potential in the bioremediation of OPs pesticides.

Abstract:A bacterial strain labeled M5R14 was isolated from the sludge in the watercourse of the Pesticide Factory of Hangzhou,and was identified as Pseudomonas diminuta.The strain showed highly efficient degradation of bifenthrin,fenpropathrin and cypermethrin.The physiological characteristics of the strain were investigated,and the bacterium is a gram-negative aerobic coccibacillus,of size 0.3～0.7 μm,which is capable of utilizing bifenthrin,fenpropathrin and cypermethrin as sole carbon sources.The degradation of bifenthrin,fenpropathrin and cypermethrin by strain M5R14 was highest under the conditions of aeration,30℃,inoculation to OD415nm=0.2,pH 6.0～7.0,pesticide concentration 100 mg·L-1 and shaking speed 180 r·min-1.Under such conditions,in culture medium with 100 mg·L-1 bifenthrin,fenpropathrin and cypermethrin together,the degradation ratios are 43.78%,43.91% and 43.75% after incubating for 3 days.Furthermore,the degradation processes fit a first-order kinetic equation and the half lives(t1/2) were 97.6,106.6 and 101.9 h,respectively.The ability of Pseudomonas diminuta M5R14 to degrade bifenthrin,fenpropathrin and cypermethrin is positively correlated to the amount of inoculum,shaking speed and aeration.
Biodegradation of λ-cyhalothrin through cell surface display of bacterial carboxylesterase

Abstract:Pyrethroids are the third widespread used insecticides globally which have been extensively applied in agricultural or household environments. Due to continuous applications, pyrethroids have been detected both in living cells and environments. The permanent exposure to pyrethroids have caused substantial health risks and ecosystem concerns. In this work, a λ-cyhalothrin (one kind of pyrethroid insecticides) degrading bacterium Bacillus velezensis sd was isolated and a carboxylesterase gene, CarCB2 was characterized. A whole cell biocatalyst was developed for λ-cyhalothrin biodegradation by displaying CarCB2 on the surface of Escherichia coli cells. CarCB2 was successfully displayed and functionally expressed on E. coli cells with optimal pH and temperature of 7.5 and 30 °C, using p-NPC4 as substrate, respectively. The whole cell biocatalyst exhibited better stability than the purified CarCB2, and approximately 120%, 60% or 50% of its original activity at 4 °C, 30 °C or 37 °C over a period of 35 d was retained, respectively. No enzymatic activity was detected when incubated the purified CarCB2 at 30 °C for 120 h, or 37 °C for 72 h, respectively. Additionally, 30 mg/L of λ-cyhalothrin was degraded in citrate-phosphate buffer by 10 U of the whole cell biocatalyst in 150 min. This work reveals that the whole cell biocatalyst affords a promising approach for efficient biodegradation of λ-cyhalothrin, and might have the potential to be applied in further environmental bioremediation of other different kinds of pyrethroid insecticides.

Abstract:Organophosphate compounds are widely used in pesticides to control weeds, crop diseases, and insect pests. Unfortunately, these synthetic compounds are hazardous and toxic to all types of living organisms. In the present work, Escherichia coli was bioengineered to achieve methyl parathion (MP) degradation via the introduction of six synthetic genes, namely, opdS , pnpAS , pnpBS , pnpCS , pnpDS , and pnpES , to obtain a new transformant, BL-MP. MP and its subsequent decomposition intermediates were completely degraded by this transformant to enter the metabolites of multiple anabolic pathways. The MP-degraded strain created in this study may be a promising candidate for the bioremediation of MP and potential toxic intermediates.

Analysis of the performance of the efficient di-(2-ethylhexyl) phthalate-degrading bacterium Rhodococcus pyridinovorans DNHP-S2 and associated catabolic pathways

Abstract:The widespread use of plastic has led to the global occurrence of phthalate esters (PAEs) pollution. PAEs can be effectively removed from polluted environments by microbe-mediated degradation. Di-(2-ethylhexyl) phthalate (DEHP) has the highest residual concentration in agricultural soil-contaminated areas compared to other PAEs in most of China. The Rhodococcus pyridinovorans DNHP-S2 microbial isolate identified was found to efficiently degrade DEHP. Within a 72 h period, the bacteria were able to degrade 52.47% and 99.75% of 500 mg L −1 DEHP at 10 °C and 35 °C, respectively. Dimethyl phthalate (DMP) was first identified as an intermediate metabolite of DEHP, which is different from the previously reported DEHP catabolic pathway. Genomic sequencing of DNHP-S2 identified benzoate 1,2-dioxygenase and catechol 2,3/1,2-dioxygenase as potential mediators of DEHP degradation, consistent with the existence of two downstream metabolic pathways governing DEHP degradation. Three targets DEHP metabolism-related enzymes were found to be DEHP-inducible at the mRNA level, and DNHP-S2 was able to mediate the complete degradation of DEHP at lower temperatures, as confirmed via RT-qPCR. DNHP-S2 was also found to readily break down other PAEs including DMP, di-n-butyl phthalate (DBP), di-n-octyl phthalate (DnOP), and n-butyl benzyl phthalate (BBP). Together, these results thus highlight DNHP-S2 as a bacterial strain with great promise as a tool for the remediation of PAE pollution. In addition to providing new germplasm and genetic resources for use in the context of PAE degradation, these results also offer new insight into the potential mechanisms whereby PAEs undergo catabolic degradation, making them well-suited for use in PAE-contaminated environments.

Abstract:Esterase, as a type of powerful catabolic enzyme for the degradation of pyrethroid pesticides (PYRs), appears promising in improving the quality of crops and the environment contaminated by pesticide residues. The purpose of this research is to provide a detailed introduction to the enzymatic properties, optimal production and immobilization conditions, and the degradation ability of Est804 for PYRs. The study on enzymatic properties indicated that Est804 was an alkaline esterase with an optimal pH of 8.0 and a broad optimal temperature in the range of 35−50°C. The optimal activity of free Est804 was calculated to be 112.812 U, and the specific enzyme activity was 48.97 U/mg. The kinetic parameters of Est804 were K m = 0.613 mM, k cat = 12,371 s –1 , and V m = 0.095 mM/min. The results of the fermentative optimization demonstrated that the optimal conditions included 1.5% of inoculation amount, 30 mL of liquid volume, 28°C of the fermentation temperature, and 18 h of the fermentation time. The optimal medium consists of 15.87 g of yeast powder, 8.00 g of glycerol, and 9.57 g of tryptone in 1 L of liquid. The optimized enzyme activity was 1.68-fold higher than that before optimization. Immobilized Est804 exhibited the highest activity under the optimum preparation conditions, including 0.35 g of chitosan dosage, 0.4 mL of an enzyme, and 4 h at 40°C for adsorption. The degradation rates of Cypermethrin (CYP), fenpropathrin (FE), and lambda-cyhalothrin (LCT) by Est804 within 30 min were 77.35%, 84.73%, and 74.16%, respectively. The present study indicated that Est804 possesses great potential for the treatment of pesticide residues on crops and environmental remediation, conducive to the development of SGNH family esterase against pyrethroid accumulation.

Abstract:Cypermethrin is popularly used as an insecticide in households and agricultural fields, resulting in serious environmental contamination. Rapid and effective techniques that minimize or remove insecticidal residues from the environment are urgently required. However, the currently available cypermethrin-degrading bacterial strains are suboptimal. We aimed to characterize the kinetics and metabolic pathway of highly efficient cypermethrin-degrading Bacillus thuringiensis strain SG4. Strain SG4 effectively degraded cypermethrin under different conditions. The maximum degradation was observed at 32 °C, pH 7.0, and a shaking speed of 110 rpm, and about 80% of the initial dose of cypermethrin (50 mg·L−1) was degraded in minimal salt medium within 15 days. SG4 cells immobilized with sodium alginate provided a higher degradation rate (85.0%) and lower half-life (t1/2) of 5.3 days compared to the 52.9 days of the control. Bioaugmentation of cypermethrin-contaminated soil slurry with strain SG4 significantly enhanced its biodegradation (83.3%). Analysis of the degradation products led to identification of nine metabolites of cypermethrin, which revealed that cypermethrin could be degraded first by cleavage of its ester bond, followed by degradation of the benzene ring, and subsequent metabolism. A new degradation pathway for cypermethrin was proposed based on analysis of the metabolites. We investigated the active role of B. thuringiensis strain SG4 in cypermethrin degradation under various conditions that could be applied in large-scale pollutant treatment.

Abstract:Pesticide residues in soil and food are of concern for human health. Biodegradation is an attractive option for reducing these residues, but more insight into the mechanism of degradation is necessary to ensure that toxic deadend products are not produced in the process. In this study, a β-cypermethrin (β-CY) degrading bacterium which we named GW-01 was isolated from a sheep's rumen chyme. HPLC analyses of culture extracts revealed that train GW-01 degraded ∼ 60 % of 100 mg/L β‑CY within 7 d. Strain GW-01 transiently accumulated 3-phenoxybenzoic acid (3-PBA), phenol, and benzoic acid during β-CY degradation. Genomic, transcriptomic, and proteomic analyses of GW-01 during degradation of β-CY and its metabolites provided evidence that an α/β- hydrolase and a cytochrome aa3 quinol oxidase were responsible for the degradation of β-CY and 3-PBA, respectively. Chlorohydrolase was implicated in the degradation of phenol to catechol. Transcriptional and proteomic evidence was also found for upregulation of the benzoic acid degradation pathway and meta-pathway (catechol-2,3-dioxygenase), respectively. Based on these results a pathway for β-CY degradation is propose that provides greater insight into the mechanism of pyrethroid degradation by microorganisms.

Abstract:Cypermethrin is an important synthetic pyrethroid pesticide that widely used to control pests in agriculture. However, extensive use has caused its residue and the metabolite 3-phenoxybenzoic acid (3-PBA) to seriously pollute the environments and agricultural products. In this study, a highly efficient cypermethrin-degrading bacterial consortium was acclimated from long-term pyrethroid-contaminated soil. Two strains, designated JQ-L and A-3, were screened from the consortium, and identified as Rhodococcus sp. and Comamonas sp., respectively. Strain JQ-L transformed 100 mg/L of cypermethrin to 3-PBA within 60 h of incubation; however, 3-PBA could not be further degraded by the strain. Strain A-3 utilized 3-PBA as sole carbon for growth, and completely degraded 100 mg/L of 3-PBA within 15 h of incubation. Co-culture of JQ-L and A-3 completely degraded 100 mg/L of cypermethrin within 24 h of incubation. Furthermore, a complete catabolic pathway of cypermethrin and the metabolite 3-PBA by the co-culture was proposed. This study provided a promising strategy for efficient elimination of cypermethrin residue-contaminated environments and agricultural products.

Abstract:Methomyl is a widely used carbamate pesticide, which has adverse biological effects and poses a serious threat to ecological environments and human health. Several bacterial isolates have been investigated for removing methomyl from environment. However, low degradation efficiency and poor environmental adaptability of pure cultures severely limits their potential for bioremediation of methomyl-contaminated environment. Here, a novel microbial consortium, MF0904, can degrade 100% of 25 mg/L methomyl within 96 h, an efficiency higher than that of any other consortia or pure microbes reported so far. The sequencing analysis revealed that Pandoraea, Stenotrophomonas and Paracoccus were the predominant members of MF0904 in the degradation process, suggesting that these genera might play pivotal roles in methomyl biodegradation. Moreover, five new metabolites including ethanamine, 1,2-dimethyldisulfane, 2-hydroxyacetonitrile, N-hydroxyacetamide, and acetaldehyde were identified using gas chromatography-mass spectrometry, indicating that methomyl could be degraded firstly by hydrolysis of its ester bond, followed by cleavage of the C-S ring and subsequent metabolism. Furthermore, MF0904 can successfully colonize and substantially enhance methomyl degradation in different soils, with complete degradation of 25 mg/L methomyl within 96 and 72 h in sterile and nonsterile soil, respectively. Together, the discovery of microbial consortium MF0904 fills a gap in the synergistic metabolism of methomyl at the community level and provides a potential candidate for bioremediation applications.

Abstract:The heightened concern about the environmental impacts of pollutants drives interest in reducing their threats to humans and the environment. Bioremediating polluted sites under environmental stresses like biotic and abiotic poses significant challenges. This study aimed to isolate a bacterium that effectively degrades pyrethroids even under abiotic stresses involving heavy metals and biotic stresses with autochthonous factors. Here, a bacterial strain, Bacillus cereus BCS1 was isolated. The response surface methodology was established to quantify the environmental impacts on pyrethroid degradation. BCS1 effectively degraded pyrethroids across conditions at 21–36 °C, pH 6.5–8.0 and inoculum sizes 1.9–4.1 mg·L −1 , exceeding 90% degradation. Notably, over 84% of β -cypermethrin ( β -CP) was degraded even when exposed to various concentrations of lead (10–1000 mg·L −1 ), chromium (10–1000 mg·L −1 ), or cadmium (0.5–50 mg·L −1 ). Moreover, BCS1 significantly accelerated β -CP degradation in soil–plant systems, displaying biotic stress tolerance, with lower half-life values (10.1 and 9.5 d) in soil and higher removal (92.1% and 60.9%) in plants compared to controls (27.7 and 25.7 d), and (18.2% and 24.3%). This study presents a novel strain capable of efficiently degrading pyrethroids and displaying remarkable environmental stress resistance. Findings shed light on bioremediating organic pollutants in complex soil ecosystems.
