Abstract:The increasing demand for tea consumption calls for the development of more products with distinct characteristics. The sensory quality of tencha is significantly determined by innate differences among tea cultivars. However, the correlations between the chemical composition and sensory traits of tencha are still unclear. To enhance the understanding of the flavor formation mechanism in tencha and further to develop new cultivars resources, we investigated non-volatiles and volatile metabolites as well as sensory traits in tencha from different tea cultivars (Camellia sinensis cv. Yabukita, Longjing 43 and Baiye 1); the relationships between the flavor traits and non-volatiles/volatiles were further evaluated by partial least squares - discriminate analysis (PLS-DA), multiple factor analysis (MFA) and multidimensional alignment (MDA) analysis. A total of 64 non-volatiles and 116 volatiles were detected in all samples, among which 71 metabolites were identified as key flavor-chemical contributors involving amino acids, flavonol glycosides, flavones, catechins, ketones, alcohols, hydrocarbons, aldehydes, esters and acids. The levels of taste-related amino acids, flavonol glycosides and gallic acid varied significantly among the tencha samples made from different tea cultivars. All the samples exhibited typical quality characteristics of tencha. The tencha from Camellia sinensis cv. Longjing 43 and Camellia sinensis cv. Baiye 1 (cultivated in the open) exhibited higher levels of amino acids and gallic acid, which were associated with the umami taste and mellow taste of tea infusion. Abundant flavonol glycosides were related to the astringency, while partial tri-glycosides specifically quercetin-3-O-galactoside-rhamnoside-glucoside and total of flavonol galactoside-rhamnoside-glucoside were associated with mellow taste. The floral alcohols were identified as significant contributors to the refreshing aroma traits of tencha. The green, almond-like, acidic and fruity odorants were associated with a green and fresh aroma, while the green, cheesy and waxy odorants such as ketones, esters, acids and hydrocarbons were associated with seaweed-like aroma. This study provides insight into sensory-related chemical profiles of tencha from different tea cultivars, supplying valuable information on flavor and quality identification for tencha.
Enhancing the Flavor Profile of Strong-Flavored Green Tea from Sichuan: Evaluation and Optimization Study

Abstract:This study evaluated seven taste subfactors of 64 green teas from Sichuan province and categorized them into three types: fresh and mellow (FM), strong and thick (ST), and fresh and strong (FS). The FM-type showed excellent freshness and sweetness, while the ST-type exhibited bitterness, astringency, and strength. Component analysis showed higher water extract and tea polyphenols in the ST-type, with the soluble sugar content correlating positively with taste strength but not sweetness, highlighting the complexity of flavor perception. More exhaustively, we used targeted metabolomics to examine the relevant constituent fractions. The levels of epigallocatechin gallate, epicatechin gallate, epigallocatechin, epicatechin, dihydromyricetin and rutin were highest in ST, and positively correlated with bitterness and astringency. Umami amino acids were found mostly in FM, and were positively correlated with freshness. To optimize the taste quality of ST green tea, two methods were investigated: tea bag confinement leaching and aroma enhancement. Both methods effectively reduced bitterness and astringency. Tea bag confinement leaching can be widely applied to enhance the utilization of summer tea raw materials and elevate the sensory quality of middle- and low-grade green teas. While promising for seasoned tea flavor enhancement and bitter suppression, the aroma enhancement technique requires further research.

Abstract:Baking is widely accepted for aroma enhancement of black tea, and studies have mainly focused on the aroma or chemical substances under a specified baking condition. Understanding of the feedback between aroma substances and characteristics is urgently needed. Therefore, a mutual feedback platform (SES/MS) combined sensory evaluation system (SES) with gas chromatography–mass spectrometry (GC-MS) was established. Based on this platform, we found that baking at 90 °C for 4 h or 5 h could maintain the primary aroma attributes and increase characteristic aroma attributes—these were considered to be the best baking conditions for Yunnan congou black tea. Meanwhile, 47 volatiles were identified, among which, pyrrole and benzaldehyde appeared to have a caramel aroma, and 2-furanmethanol and α-terpineol presented a baked aroma. This study reveals the dynamic change of aroma profiles and compounds during the aroma enhancement, and provides an optional template for researchers, focused on the relationship between quality and aroma attributes of teas.
Unraveling the contributing factors of stale odor in Longjing tea through a sensomics approach

Abstract:Longjing tea is renowned for its fresh aroma and high value. However, during storage, the emergence of an off-flavor known as "stale odor" can significantly impact the flavor quality and economic benefits of Longjing tea. Yet, the specific volatiles responsible for this stale odor in Longjing tea remain unknown. In this study, Longjing tea samples with varying degrees of stale odor intensity were analyzed using simultaneous distillation extraction coupled with gas chromatography-mass spectrometry (SDE-GC-MS). Through odor activity value (OAV) and fractional omission testing, hexanoic acid and trans-2-nonenal were identified as the primary contributors to the stale odor. Moreover, the concentration of hexanoic acid was found to be valuable in predicting the intensity of the stale odor in Longjing tea. The oxidative degradation of linoleic acid was proved as the generation pathway of stale odor in Longjing tea. These findings provide essential theoretical principles for Longjing tea production and preservation.

Abstract:Parameters of liquor color difference, taste related constituents and aroma related volatiles in 23 Chinese green tea (Camellia sinensis) samples were analyzed by color difference meter, high performance liquid and gas chromatographs respectively. Seven variables were extracted by principal component analysis from the data sets corresponding to the liquor color, taste and aroma and linear regression of total score of sensory preference (TSSP) upon the 7 extracted variables produced a significant relationship: TSSP = 105.82–0.56 (ΔE)−0.15 (total catechins) + 0.32 (geraniol) + 0.53 (GCG) + 0.44 (linalool oxide I) −0.54 (ascorbic acid) + 0.24 (n-valeraldehyde) (p = 0.003, standard error of the estimation = 2.97). The regressive relationship simplifies the process of chemical and instrumental assessment of sensory preference of green tea and could be used for green tea quality assessment.

Abstract:Longjing tea is one of China's most renowned green teas and is enjoyed by tea enthusiasts all over the world. Although its unique aroma is the most important quality attribute, the key aroma compounds were not yet systematically analysed. A sensory-guided screening of the aroma distillate isolated from a high-grade Longjing green tea infusion by application of an aroma extract dilution analysis revealed 60 aroma-active domains in the gas chromatogram with flavour dilution (FD) factors in the range from 32 to the highest FD factor of 4098, as found for 3-methylnonane-2,4-dione (hay-like, aniseed-like) and vanillin (vanilla-like). A quantitation of the 42 aroma compounds eliciting the highest FD factors by means of stable isotope dilution assays (SIDA) followed by a calculation of odour activity values (OAV; ratio of concentration to odour threshold) showed that 30 aroma compounds exceeded their odour threshold in the tea beverage. By mixing 30 purified reference compounds of these odorants in water in exactly the same concentrations as determined in the tea beverage, the genuine aroma of the Longjing tea could successfully be simulated. In ensuing studies, quantitative changes during the traditional manufacturing process of the tea leaves as well as differences induced by the hot water treatment were systematically evaluated based on SIDAs. The results indicated the great importance of the initial pan-frying for the formation of key aroma compounds in the tea leaves. A comparison of the concentrations of selected key aroma compounds in the finished tea leaves and the tea infusion prepared thereof indicated further that large quantities of, for example, 2-methylbutanal, dimethyl sulphide, and 3-methylnonane-2,4-dione were obviously formed from yet unknown precursors by the hot water treatment.

Abstract:Withering is a key process that affects the aroma of Keemun black tea (KBT). In this study, the aroma composition of KBT through natural withering, sun withering, and warm-air withering was analysed using gas chromatography-mass spectrometry. The results revealed significant differences in the three samples. Gas chromatography-olfactometry and aroma extract dilution analysis were performed with screening through a relative odour activity value (rOAV) > 1. In total, 11 aroma-active compounds (geraniol, (Z)-4-heptenal, 1-octen-3-ol, (E)-β-ionone, 3-methylbutanal, linalool, β-damascenone, (E, E)-2,4-decadienal, methional, (E, E)-2,4-nonadienal, and (E)-2-nonenal) were found to be responsible for the differences in aroma caused by different withering methods. Linalool (rOAV, 161) and geraniol (rOAV, 785) were responsible for the higher flowery and fruity aromas when sun withering was applied, whereas methional (rOAV, 124) contributed to the intense roasty aroma when warm-air withering was employed. Moreover, our results were verified by quantitative descriptive analysis and addition experiments.

Abstract:The oolong tea aroma is shown to consist of cultivar aroma and technical aroma in this study based on the aroma differences between oolong tea products of cultivars of different suitability, as determined by correlation analysis of olfactory, sensory, electronic nose, and GC-MS data. Human senses were significantly affected by the aroma components, which included eight terpene metabolites (β-Ocimene, (Z)-Furan linalool oxide, linalool, (3E)-4,8-Dimethyl-1,3,7-nonatriene, (E)-Pyranoid linalool oxide, γ-Elemene, Humulene, (Z,E)-α-Farnesene), three carotenoid metabolites (β-Ionone, (Z)-Geranylacetone and 6-methyl-5-Hepten -2-one), three lipid metabolites ((Z)-3-Hexenyl (Z)-3-hexenoate, Butanoic acid hexyl ester, and (Z)-Jasmone), four amino acid metabolites (Methyl salicylate, Geranyl isovalerate, indole, and Phenylethyl alcohol) ), and six thermal reaction products (2-Pentylfuran, Octanal, Decanal, (E,E)-2,4-Nonadienal, (Z)-2-Decenal, and (E)-2-Undecenal). Meanwhile, several aroma compounds (such as (E)-Nerolidol and α-Farnesene), mainly comprising the "technical aroma" formed in the processing mode, were noted to be less closely related to cultivar suitability. Abstract:Despite extensive studies, the relationship between the quality/quantity of tea odorants and oolong tea quality remains unclear. To investigate the key components affecting Bao-chung tea quality, we collected samples of different grades from a tea-tasting competition and determined the content and composition of volatile components and individual catechins using gas chromatography–mass spectrometry and high-performance liquid chromatography. We used an electronic nose (E-nose) to collect odor component signals and established a quality recognition model. The different tea grades did not significantly differ in catechin content, but their specific odor intensity and proportion of odor components varied significantly. Linear discriminant analysis showed that the intensity and proportion of volatile organic compounds could be used for distinguishing the different grades of Bao-chung tea. By combining different quantities of indole, linalool, and butanoic acid and proportions of p-cymene, cis-β-ocimene, nonanal, allo-ocimene, cis-jasmone, and α-farnesene, the ability to distinguish among Bao-chung tea grades was significantly improved. Our results revealed that the quality of Bao-chung tea should be evaluated based on the combined perception of odor component intensity and proportion rather than solely relying on the concentration or composition of specific compounds. Therefore, individuals can judge a Bao-chung tea grade based on the combined perception of odor component intensity and proportion. The E-nose can be used to identify Bao-chung tea grades based on its ability to determine the odorant composition.

Abstract:Flavour determinants in tea infusions and their changes during manufacturing processes were studied using Camellia sinensis cultivars 'Bai-Sang Cha' ('BAS') possessing significant floral scents and 'Fuding-Dabai Cha' ('FUD') with common green tea odour. Metabolite profiling based on odour activity threshold revealed that 'BAS' contained higher levels of the active odorants β-ionone, linalool and its two oxides, geraniol, epoxylinalool, decanal and taste determinant catechins than 'FUD' (p<0.05). Enhanced transcription of some terpenoid and catechin biosynthetic genes in 'BAS' suggested genetically enhanced production of those flavour compounds. Due to manufacturing processes, the levels of linalool and geraniol decreased whereas those of β-ionone, linalool oxides, indole and cis-jasmone increased. Compared with pan-fire treatment, steam treatment reduced the levels of catechins and proportion of geraniol, linalool and its derivatives, consequently, reducing catechin-related astringency and monoterpenol-related floral scent. Our study suggests that flavour determinant targeted modulation could be made through genotype and manufacturing improvements.

Abstract:Citrus blend black teas are popular worldwide, due to its unique flavor and remarkable health benefits. However, the aroma characteristics, aroma profiles and key odorants of it remain to be distinguished and cognized. In this study, the aroma profiles of 12 representative samples with three different cultivars including citrus (Citrus reticulata), bergamot (Citrus bergamia), and lemon (Citrus limon) were determined by a novel approach combined head space-solid phase microextraction (HS-SPME) with comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (GC×GC-TOFMS). A total of 348 volatile compounds, among which comprised esters (60), alkenes (55), aldehydes (45), ketones (45), alcohols (37), aromatic hydrocarbons (20), and some others were ultimately identified. The further partial least squares discrimination analysis (PLS-DA) certified obvious differences existed among the three groups with a screening result of 30 significant differential key volatile compounds. A total of 61 aroma-active compounds that mostly presented green, fresh, fruity, and sweet odors were determined in three groups with gas chromatography-olfactometry/mass spectrometry (GC-O/MS) assisted analysis. Heptanal, limonene, linalool, and trans-β-ionone were considered the fundamental odorants associated with the flavors of these teas. Comprehensive analysis showed that limonene, ethyl octanoate, copaene, ethyl butyrate (citrus), benzyl acetate, nerol (bergamot) and furfural (lemon) were determined as the characterized odorants for each type.

Abstract:Anaerobically fermented pickled tea (PT) can be produced by spontaneous fermentation (SF) or yeast-enhanced fermentation (YF). Aroma and taste characteristics of PT during YF and SF were investigated using sensory evaluation, odour activity, aroma character impact values, HS-SPME-GC–MS, UPLC-QQQ-MS/MS, and spectrophotometry, annotating 198 volatile and 115 non-volatile components. The main contributing volatile components were β-ionone, and 1-octanol, promoted by YF and SF, and yielding floral and fruity aromas respectively. Additionally, compared with SF, YF promoted the formation of citronellol yielding a floral aroma, inhibited the stale aroma of methoxybenzenes, and reduced bitter, astringent, and sour tastes. Furthermore, partial least-squares regression analysis identified the main components related to the 'acides aroma' of PT as linalool oxide, n -decanoic acid, hexanoic acid, 3,7-dimethyl-2,6-octadienoic acid, 3-methyl-1-dodecyn-3-ol, and nerolidol. This application could be used as methodology for the comprehensive analysis of tea aroma and taste and these results can act as guidelines for PT production and quality control.

Pseudomonas cepacia lipase--mediated transesterification reactions of hydrocinnamates.

Abstract:Use of lipase from Pseudomonas cepacia in transesterifcation reactions of ethyl hydrocinnamate with different alcohols has been examined. Among the alcohols tested, viz., n-butanol, iso-amyl alcohol, benzyl alcohol, n-octanol and 1-phenylethanol, only n-butanol yielded the transesterified product. Among the solvents tested, viz., n-heptane, n-hexane, cyclohexane, toluene, diisopropylether and n-butanol, the initial rate of transesterification proceeded in the order cyclohexane > n-heptane > n-hexane > diisopropylether > n-butanol > toluene. Using hexane as the solvent and a substrate to enzyme ratio of 1:5, the substrate to alcohol ratio was varied to maximize the yield. n-Butyl hydrocinnamate was obtained in 92% yield in 48 hr by employing a 1:1 (wt/wt) ratio of ethyl hydrocinnamate to lipase and a 1:5 (vol/vol) ratio of ethyl hydrocinnamate to n-butanol in hexane.
Volatolomics-assisted characterization of the key odorants in green off-flavor black tea and their dynamic changes during processing

Abstract:Aroma plays a pivotal role in the quality of black tea. However, the acceptability of black tea is greatly limited by the green off-flavor (GOF) resulting from the inappropriate processing control. In this study, the key odorants causing GOF were investigated by volatolomics, and their dynamic changes and formation pathways were in-depth understood. Significant alterations in volatile metabolites were observed in the withering stage. A total of 14 key odorants were identified as contributors to GOF, including 2-methylpropanal, 3-methylbutanal, 1-hexanol, nonanal, (E, E)-2,4-heptadienal, benzaldehyde, linalool, (E, E)-3,5-octadiene-2-one, β-cyclocitral, phenylacetaldehyde, (E, E)-2,4-nonadienal, methyl salicylate, geraniol, and β-ionone. Among them, (E, E)-2,4-heptadienal (OAV = 3913), characterized by fatty, green, and oily aromas, was considered to be the most important contributor causing GOF. Moreover, it was found that lipid degradation served as the primary metabolic pathway for GOF. This study provides a theoretical foundation for off-flavor control and quality improvement of black tea.
Characterization of stale odor in green tea formed during storage: Unraveling improvements arising from reprocessing by baking

Abstract:Changes in key odorants and aroma profiles of three types of green tea during storage and after baking treatment were determined using headspace solid-phase microextraction–gas chromatography–mass spectrometry/olfactometry, odor activity value (OAV) and orthogonal partial least-squares discriminant analysis. As the stale odor developed during storage, the content of aldehydes decreased, whereas the contents of ketones and heterocycles (furans, pyrroles, etc.) increased. Key odorants, including ( E , E )-2,4-heptadienal, α -terpineol, ( E , E )-2,4-nonadienal and ( E , E )-2,4-decadienal, appeared to be mainly responsible for the stale odor. After baking treatment, the stale odor was significantly improved, the contents of aldehydes and heterocycles were significantly increased and the content of alcohols was significantly decreased. Of these, 2-pentylfuran, hexanal, nonanal and limonene, which increased after baking, contribute the green, fruity and roasted aromas of green tea, because of their high OAVs, aroma intensities and variable importance in projection values. This study has improved understanding of the chemical changes resulting in stale odor development in green tea during storage and improvements arising from baking treatment, as well as providing a theoretical basis for the improvement of storage conditions and green tea flavor.

Abstract:Objectives Osmanthus black tea (OBT) is produced by reprocessing black tea while it absorbs the fragrance of Osmanthus fragrans and this scenting process is crucial for forming the unique aroma of OBT. This work intended to reveal the effects of scenting parameters, the types of base tea, and scenting technologies on the aroma quality of OBT.Materials and Methods In this study, volatile compounds of OBTs with different scenting parameters, types of base tea, and scenting technologies were analyzed by solid-phase microextraction and gas chromatography-mass spectrometry. Weighted gene co-expression network analysis and orthogonal partial least squares discriminant analysis were used to identify the key volatile compounds of OBT.Results More than 80 volatile compounds of OBT were found, and six compounds, alpha-ionone, beta-ionone, gamma-decalactone, dihydro-beta-ionone, dihydro-beta-ionol, and 6-pentyl-2-pyrone, were identified as the key volatiles of OBT. As scenting time (ST) increased, the key volatile compounds in OBT showed an upward trend or increased initially and then decreased. Meanwhile, the contents of key volatile compounds increased as the ratio of flowers to tea (RFT) increased. In addition, base teas with a high proportion of pekoes produced higher-quality OBT by absorbing more key volatile compounds from O. fragrans. Comparing OBTs made from the fast-scenting process and the traditional scenting process (TS), showed that higher aroma quality was obtained through repeated scenting rounds of TS.Conclusions Appropriate long ST and high RFT, as well as base tea with a high proportion of pekoes and TS with repeated scenting rounds are beneficial for the accumulation of odorants and the improvement of aroma quality in OBT.
Characterization of the Key Aroma Compounds of Three Kinds of Chinese Representative Black Tea and Elucidation of the Perceptual Interactions of Methyl Salicylate and Floral Odorants

Abstract:Jinjunmei (JJM), Keemun (KM), and Dianhong (DH) are the representative black teas in China, and they have always been favored by consumers. In this study, we aim to obtain the aroma characteristic information of volatile components in black tea samples through headspace solid-phase microextraction (HS-SPME), solvent-assisted flavor evaporation (SAFE), and gas chromatography-mass spectrometry combined with gas chromatography-olfactometry technology. The results showed that 70 compounds including α-methylbenzyl alcohol (isomer of β-phenylethanol) were identified as odorants. Among them, 39 compounds such as linalool and geraniol showed a high degree of aroma contribution. Furthermore, the Feller’s additive model was used to explore the perceptual interactions among the methyl salicylate and the floral compounds (10 groups): five groups of binary compounds showed masking effect after mixing, one group showed additive effect, and four groups showed synergistic effect. The ratio (R) was compared with the aroma index (n) of Steven’s law, which found a high-fitness exponential relationship. The results of this study help to provide additional and new theoretical guidance for improving the aroma quality of black tea.

Abstract:Aroma is an indispensable factor that substantially impacts the quality assessment of black tea. This study aims to uncover the dynamic alterations in the sweet and floral aroma black tea (SFABT) throughout various manufacturing stages using a comprehensive analytical approach integrating gas chromatography electronic nose, gas chromatography–ion mobility spectrometry (GC-IMS), and gas chromatography–mass spectrometry (GC-MS). Notable alterations in volatile components were discerned during processing, predominantly during the rolling stage. A total of 59 typical volatile compounds were identified through GC-IMS, whereas 106 volatile components were recognized via GC-MS throughout the entire manufacturing process. Among them, 14 volatile compounds, such as linalool, β-ionone, dimethyl sulfide, and 1-octen-3-ol, stood out as characteristic components responsible for SFABT with relative odor activity values exceeding one. This study serves as an invaluable theoretical platform for strategic controllable processing of superior-quality black tea.

Abstract:Steamed green tea has a long history and unique aroma, but little is known about its key aroma components. In this study, 173 volatiles in steamed green tea were identified using solvent-assisted flavor evaporation and headspace-solid phase microextraction plus two chromatographic columns of different polarities. Aroma extract dilution analysis revealed 48 highly aroma-active compounds with flavor dilution factors 64-1024. Internal standards were used to calculate odorant active value (OAV), and 11 OAV > 1 key aroma compounds were determined. Omission test identified eight substances, including dimethyl sulfide, (E)-β-ionone, cis-jasmone, linalool, nonanal, heptanal, isovaleraldehyde and (Z)-3-hexenol, as the key aroma active compounds of steamed green tea. With the increase of withering degree, the content of these substances increased first and then decreased except for heptanal and cis-jasmone. Moreover, the water content of 62 % was suggested to be an appropriate withering degree during the processing of steamed green tea.
