Abstract:The limited solubility and stability of pea proteins hinder their utilization in liquid formulations. In this study, protein glutaminase (PG) was employed to modify pea protein isolates (PPIs) and obtain deamidated PPI with varying degrees of deamidation (DD, 10–25%). The solubility and thermal stability of these deamidated PPI samples were assessed, and a comprehensive analysis, including SDS-PAGE, zeta potential, FTIR, surface hydrophobicity, and intrinsic fluorescence, was conducted to elucidate the mechanism behind the improvement in their functional properties. The results reveal that PG modification greatly enhances the solubility and heat stability of PPI, with the most notable improvements observed at higher DD (>20%). PG modification increases the net charge of PPI, leading to the unfolding and extension of the protein structures, thus exposing more hydrophobic groups. These structural changes are particularly pronounced when DD exceeds 20%. This increased electrostatic repulsion between carboxyl groups would promote protein unfolding, enhancing interactions with water and hindering the aggregation of unfolded protein in the presence of salts at elevated temperatures (supported by high-performance size exclusion chromatography and transmission electron microscopy). Accordingly, PG-mediated deamidation shows promise in enhancing the functional properties of PPI.

Coarse Grained Molecular Dynamics Simulations of SubMicron Liquid Cylinders and Jets

Abstract:A coarse-grained molecular dynamics (MD) method, dissipative particle dynamics (DPD), is employed to simulate the breakup of liquid sub-micron liquid cylinders and jets. Consistent with prior findings employing MD, Rayleigh’s criterion for capillary breakup of inviscid liquid cylinders is shown to be applicable. Thermal fluctuations are the primary mechanism inducing instability and this leads to the formation of almost monodisperse drops. The parameters varied in the study include the cylinder radius, thermal length scale, viscosity, and surface tension. The breakup time does not show the same scaling dependence as in capillary breakup of liquid cylinders at the macroscale. The time variation of the radius at the point of breakup agrees with prior theoretical predictions from expressions derived with the assumption that thermal fluctuations lead to breakup. The thermal fluctuations accelerate the breakup of liquid jets at the submicron scale. The time evolution of minimum jet radius as given by prior theoretical analysis is recovered. Introduction The formation of drops from the breakup of sub-micron scale liquid cylinders and jets is important in many current and emerging applications, such as nanoscale machining, super-fine ink-jet printing, and drug-/gene-delivery to biological cells. The breakup of liquid cylinders and atomization of liquid jets at the macroscale has been a topic of study for over a century [14, 12], but sub-micron scale applications have gained in importance only recently. Computational studies of two-phase flows at the macroscale are complex because of the need to track highly deforming interfaces and develop numerical schemes which can be employed for compressible (gas) and incompressible (liquid) fluids [3]. At the sub-micron scale, the assumption that the fluid properties can be represented by statistical averaging may not hold. At this scale, molecular dynamics (MD) is an obvious choice as a numerical technique [10]. For example, it has been applied to study the behaviour of nanodrops [5] and mixing processes at the nanoscale [8]. Nevertheless, the method is computationally expensive. There are approaches which are computationally less expensive but which sacrifice precise information about dimensions and scales because of lack of adequate information about coarse graining. These approaches are, however, very useful to determine trends in physical behaviour at the sub-micron scale. In this work, the dissipative particle dynamics (DPD) approach will be applied to study the breakup of liquid nanocylinders and nanojets [4,6,7]. An important question here is whether the method can capture the physics associated with sub-micron liquid cylinder and jets. The Computational Model The DPD method is particle-based in which each particle represents millions of molecules. The governing equations are written down for the particles and the equations contain quantities which represent the collective behaviour of the millions of molecules they represent. Abstract:This study investigated the effects of transglutaminase (TGase) treatment on the taste profiles and peptide compositions of pea protein isolate hydrolysates (PPIH) produced by Flavourzyme and protease 11,039. TGase significantly increased umami intensity by 50% and dramatically decreased bitterness by 40% in PPIH. While protein recovery, degree of hydrolysis, and peptide nitrogen percentage remained unaffected, TGase altered the relative proportions of taste compounds. This was evident through increased levels of umami, hydrophilic, and sweet-bitter amino acids in non-free amino acids, indicating the formation of peptides with umami potential. UPLC-MS/MS analysis confirmed that TGase facilitated the generation of longer peptides with higher umami potential compared to the short peptides in untreated PPIH. Simultaneously, the reduction in bitter components and the generation of umami substances contributed to the decrease in bitterness. Receptor-peptide docking studies confirmed that the newly formed peptides had higher binding affinities. Overall, TGase modification led to a shift in the substance profiles of PPIH, resulting in the creation of peptides with enhanced umami characteristics. These findings demonstrate the potential of combining TGase treatment with enzymatic hydrolysis to modify taste properties and develop flavorful plant-based protein ingredients.

Abstract:Plant-based protein foods as suitable alternative protein sources have recently received increased global interest. The scientific community is exploring effective modification approaches to enhance the functionality of plant-based proteins for expanded utilization. Deamidation has shown great potential for structural modifications and improving the processing efficiency of proteins. In this review, we firstly revisit the enzyme reaction mechanism of protein-glutaminase and its fundamental differences from other enzymatic methods for the deamidation of proteins. Then, the latest advances regarding the suitability of protein-glutaminase modifications for improving the functional properties (e.g., solubility, emulsifying and foaming properties, flavor, and reduction in allergenicity) of plant-based proteins are overviewed. Finally, we address the potential prospect associated with the use of protein-glutaminase in plant-based protein foods, such as meat, dairy, and egg alternatives. This review provides a novel perspective for the design of plant-based protein foods by using protein-glutaminase in order to match animal counterparts in taste and texture, and to fuel widespread adoption.

Abstract:Pea protein is an emerging functional ingredient in structuring plant-based meat analogs. Due to its hypoallergenic, non-GMO, and high nutritional value , pea protein has succeeded in capturing the interests of scientists around the globe. Nevertheless, the beany flavor is a significant impediment to consumer acceptance of pea protein products, creating a gap for further research on the formation mechanism and removal methods of beany flavor is essential for the robust development of pea protein food and nutritional products. Herein, this review summarizes the key beany flavor compounds, their formation mechanisms, and interactions between pea protein and off-flavor compounds, along with the extrapolation of the extraction methods, detection methods, sensory evaluation, and the application of omics techniques for flavor analyses. The possible metabolic pathways of related microorganisms to reduce beany flavor in pea protein are also discussed. The formation of beany flavor is a complex process; the lipoxygenase-hydroperoxide lyase mechanism is to be held responsible for the main formation pathway, interaction between flavor compounds and pea protein, as well as the interaction between flavor compounds, collectively impact the overall flavor profiles. Modern food flavor analysis techniques combined with sensory evaluation provide more accurate support for the identification and analyzing beany flavor. Microbial fermentation is to be considered a thought-provoking strategy for decreasing off-flavor. At the same time, its metabolic mechanism requires further exploration, still proving to be an allusion for the development and delivery of plant-based foods with a lessened beany flavor.

Identification of volatile and odor-active compounds in pea protein fractions obtained by a modified extraction method using fermentation

Abstract:This study investigates the aromatic composition of pea albumin and globulin fractions obtained through either fermentation or conventional acidification using hydrochloric acid (control) toward the isoelectric point of pea globulins. Different lactic acid bacteria were used including S. thermophilus (ST), L. plantarum (LP), and their coculture (STLP). The volatile compounds were extracted by solvent-assisted flavor evaporation technique and quantified by gas chromatography-mass spectrometry (GC-MS). Odor-active compounds (OAC) were further characterized by gas chromatography-olfactometry (GC-O). In total, 96 volatile and 36 OACs were identified by GC-MS and GC-O, respectively. The results indicated that the protein fractions obtained by conventional acidification were mainly described by green notes for the presence of different volatile compounds such as hexanal. However, the samples obtained by fermentation had a lower content of these volatile compounds. Moreover, protein fractions obtained by coculture fermentation were described by volatile compounds associated with fruity, floral, and lactic notes. PRACTICAL APPLICATION: The insights from this study on pea protein aroma could find practical use in the food industry to enhance the sensory qualities of plant-based products. By utilizing fermentation methods and specific lactic acid bacteria combinations, manufacturers may produce pea protein with reduced undesirable green notes, offering consumers food options with improved flavors. This research may contribute to the development of plant-based foods that not only provide nutritional benefits but also meet consumer preferences for a more appealing taste profile.

Abstract:Pea protein is widely used as an alternative protein source in plant-based products. In the current study, we fermented pea protein to reduce off-flavor compounds, such as hexanal, and to produce a suitable fermentate for further processing. Laboratory fermentations using 5% (w/v) pea protein suspension were carried out using four selected lactic acid bacteria (LAB) strains, investigating their growth and acidification capabilities in pea protein. Rapid acidification of pea protein was achieved with Lactococcus lactis subsp. lactis strain LTH 7123. Next, this strain was co-inoculated together with either the yeasts Kluyveromyces lactis LTH 7165, Yarrowia lipolytica LTH 6056, or Kluyveromyces marxianus LTH 6039. Fermentation products of the mixed starter cultures and of the single strains were further analyzed by gas chromatography coupled with mass spectrometry to quantify selected volatile flavor compounds. Fermentation with L. lactis LTH 7123 led to an increase in compounds associated with the "beany" off-flavors of peas, including hexanal. However, significant reduction in those compounds was achieved after fermentation with Y. lipolytica LTH 6056 with or without L. lactis LTH 7123. Thus, fermentation using co-cultures of LAB and yeasts strains could prove to be a valuable method for enhancing quality attributes of pea protein-based products.
Synergistic Improvement of Quinoa Protein Heat-Induced Gel Properties Treated by High-Intensity Ultrasound Combined with Transglutaminase

Abstract:BACKGROUNDQuinoa protein is enriched with a wide range of amino acids, including all nine essential amino acids necessary for the human body, and in appropriate proportions. However, as the main ingredient of gluten-free food, it is difficult for quinoa to form a certain network structure for lack of gluten protein. The aim of this work was to enhance the gel properties of quinoa protein. Therefore, the texture characteristics of quinoa protein treated with different ultrasound intensities coupled with transglutaminase (TGase) were investigated. RESULTSThe gel strength of quinoa protein gel increased markedly by 94.12% with 600 W ultrasonic treatment, and the water holding capacity increased from 56.6% to 68.33%. The gel solubility was reduced and free amino content increased the apparent viscosity and the consistency index. Changes in the free sulfhydryl group and hydrophobicity indicated that ultrasound stretched protein molecules and exposed active sites. The enhanced intrinsic fluorescence intensity at 600 W demonstrated that ultrasonic treatment affected the conformation of quinoa protein. New bands emerged in sodium dodecylsulfate-polyacrylamide gel electrophoresis indicating that high-molecular-weight polymers were generated through TGase-mediated isopeptide bonds. Furthermore, scanning electron microscopy showed that the gel network structure of TGase-catalyzed quinoa protein was more uniform and denser, thereby improving the gel quality of quinoa protein. CONCLUSIONThe results suggested that high-intensity ultrasound combined with TGase would be an effective way to develop higher-quality quinoa protein gel. & COPY; 2023 Society of Chemical Industry.

Abstract:Ureteric stenting allows palliative treatment of strictures resulting from trauma, surgery, neoplasm or from scarring following treatment of malignancy. Retrograde and antegrade approaches can be used (Mitty et al, 1986; Shore et al, 1987) but, in a small proportion of patients, neither approach is successful, and patients with ureters which are completely occluded present a problem. We describe a combined antegrade and retrograde technique analogous to that used in biliary stenting (Robertson et al, 1987; Freeman et al, 1991) as a useful method in such cases. Three cases are described which illustrate the salient features. A 56-year-old man was admitted for insertion of a stent across an obstructed right distal ureteric orifice. His well-differentiated bladder tumour had been treated by several endoscopic resections over the previous 5 years and one course of intravesical chemotherapy had been given 16 months earlier.
Effects of Fermentation with Tetragenococcus halophilus and Zygosaccharomyces rouxii on the Volatile Profiles of Soybean Protein Hydrolysates

Abstract:The effects of fermentation with lactic acid bacteria (LAB) and yeast on the aroma of samples were analyzed in this work. The volatile features of different soybean hydrolysates were investigated using both GC-MS and GC-IMS. Only 47 volatile flavor compounds (VFCs) were detected when using GC-IMS, while a combination of GC-MS and GC-IMS resulted in the identification of 150 compounds. LAB-yeast fermentation could significantly increase the diversity and concentrations of VFCs (p < 0.05), including alcohols, acids, esters, and sulfurs, while reduce the contents of aldehydes and ketones. Hierarchical clustering and orthogonal partial least squares analyses confirmed the impact of fermentation on the VFCs of the hydrolysates. Seven compounds were identified as significant compounds distinguishing the aromas of different groups. The partial least squares regression analysis of the 25 key VFCs (ROAV > 1) and sensory results revealed that the treatment groups positively correlated with aromatic, caramel, sour, overall aroma, and most of the key VFCs. In summary, fermentation effectively reduced the fatty and bean-like flavors of soybean hydrolysates, enhancing the overall flavor quality, with sequential inoculation proving to be more effective than simultaneous inoculation. These findings provided a theoretical basis for improving and assessing the flavor of soybean protein hydrolysates.

Abstract:ABSTRACT: The idea that plasmids replicate within hosts at the expense of cell metabolic energy and preformed cellular blocks depicts plasmids as a kind of molecular parasites that, even when they may eventually provide plasmid-carrying strains with growth advantages over plasmid-free strains, doom hosts to bear an unavoidable metabolic burden. Due to the consistency with experimental data, this idea was rapidly adopted and used as a basis of different hypotheses to explain plasmid-host interactions. In this article we critically discuss current ideas about plasmid effects on host metabolism, and present evidence suggesting that the complex interaction between plasmids and hosts is related to the alteration of the cellular regulatory status.

Abstract:The development of plant-based meat analogs is currently hindered by the beany flavor generated by raw soybean protein and extrusion processing. Wide concern has led to extensive research on the generation and control of this unwanted flavor, as an understanding of its formation in raw protein and extrusion processing and methods through which to control its retention and release are of great significance for obtaining ideal flavor and maximizing food quality. This study examines the formation of beany flavor during extrusion processing as well as the influence of interaction between soybean protein and beany flavor compounds on the retention and release of the undesirable flavor. This paper discusses ways to maximize control over the formation of beany flavor during the drying and storage of raw materials and methods to reduce beany flavor in products by adjusting extrusion parameters. The degree of interaction between soybean protein and beany compounds was found to be dependent on conditions such as heat treatment and ultrasonic treatment. Finally, future research directions are proposed and prospected. This paper thus provides a reference for the control of beany flavor during the processing, storage, and extrusion of soybean raw materials used in the fast-growing plant-based meat analog industry.

Abstract:Abstract Many plant protein ingredients have unpleasant flavor (e.g., green, beany, bitterness) characteristics. Isolation of the protein fraction will reduce the intensity of the undesirable flavors but in most cases the flavor issue is not eliminated. The objective of this review is to provide information about undesirable flavors associated with plant proteins and approaches to mitigate these flavors. The focus of the review will be on flavor associated with plant proteins from pulses, soybean, and oilseeds (e.g., hempseed, flaxseed). While other plant proteins exist, they will not be covered extensively in this limited review. The undesirable flavors are defined as volatile organic compounds (VOCs) and non‐volatile organic compounds (non‐VOCs). Example VOCs include aldehydes, ketones, and isobutyl pyrazines whereas saponins, bitter peptides, and phenolics are among the most common non‐VOCs. The raw material used as the source for the protein dictates the undesirable flavors remaining in the protein ingredient. The intensity of the binding of the VOCs and non‐VOCs to protein serves as the basis for the detection and difficulty in removing these compounds from the protein. A discussion about flavor binding and processing methods to mitigate unwanted flavors in protein ingredients will be provided. Methods presented will focus on extraction and thermal treatment. Although flavor masking is an approach to minimize undesirable, this topic will not be covered. Understanding the compounds responsible for the flavor issues and methods to minimize their impacts on sensory characteristic of protein ingredients can provide manufacturers with solutions to addressing flavor issues inherent to plant proteins.
Direct Classification of Volatile Organic Compounds in Heat-Treated Glutathione-Enriched Yeast Extract by Headspace-Gas Chromatography-Ion Mobility Spectrometry (HS-GC-IMS)

Abstract:Yeast extract (YE) is tremendously used as a flavor enhancer in foods because of its composition. However, different types of YE have different aromatic profiles. This study focuses on the thermal treatment of glutathione-enriched yeast extract (GSH-YE) at different temperatures with the addition of Maillard precursors to investigate any change in its volatile organic profile. Headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) was utilized for the instrumental analysis. A total number of 104 volatile compounds were analyzed through this approach. Qualitative and quantitative analysis of the samples were made in order to calculate the concentration of volatile compounds in samples. Sensory evaluation was performed to compare the overall aroma descriptions with the volatile profile of the heat-treated samples. Sensory scores revealed that an increase in the thermal temperature significantly affected the meaty and roasted aroma of GSH-YE. Many sulfur-containing volatile compounds including 2-methyl-3-furanthiol, 2-furfurylthiol, 2-methyl-3-(methylthio)furan, dimethyl trisulfide, 2-acetylthiazol, and some furans and pyrazines related to meaty and roasted odor notes were reported in this study. Significant increase in the amounts of furfural and dimethyl sulfide was observed in samples having higher thermal temperatures as compared with the control treatment. The temperature of 100 °C with adequate amounts of Maillard precursors in GSH-YE was considered an optimal condition for generating ample amounts of 2-methyl-3-methylthiofuran and 2-furfurylthiol. The 3D graphical data confirms the potential application of HS-GC-IMS as a more potent instrumental approach in the determination of volatile compounds in yeast extract.

Abstract:Defatted peanut meal protein hydrolysates (DPMHs) usually have a bitter taste. γ-Glutamylation by Bacillus amyloliquefaciens l-glutaminase was introduced to DPMH to reduce its bitterness and generated a γ-glutamylated product (DPMH-G). Extra l-glutamine (l-Gln) (5% w/w) was added to DPMH, and the mixture was then γ-glutamylated (DPMH-G-Q). Results showed that γ-glutamylation decreased the bitterness of the products and also enhanced their kokumi, umami, and salty taste, especially for DPMH-G-Q. Bitter amino acids and bitter peptides were found to be substrates (acceptors) of the synthesized γ-[Glu](1,2)-AAs and γ-Glu-AA-AAs, respectively. The production yield of γ-[Glu](1,2)-AAs was only 0.69/100 g for DPMH-G and 2.30/100 g for DPMH-G-Q, which was much lower than that of γ-Glu-AA-AAs (5.73/100 g for DPMH-G and 18.72/100 g for DPMH-G-Q). The improvement in taste attributes of DPMH might mainly be due to the consumption of bitter dipeptides and the production of γ-Glu-AA-AAs. In DPMH-G-Q, eight γ-Glu-AA-AAs were identified, including γ-Glu-Ile-Lys, γ-Glu-Ala-Ile, γ-Glu-Leu-Leu, γ-Glu-Phe-Leu, γ-Glu-Thr-Leu, γ-Glu-Ile-Met, γ-Glu-Val-Leu, and γ-Glu-Ser-Tyr, which were first time reported. They all can enhance umami, salty, and kokumi taste with a threshold value between 1.61 ± 0.21-2.16 ± 0.19, 1.65 ± 0.19-2.23 ± 0.20, and 0.67 ± 0.21-1.00 ± 0.22 mM, respectively. Insufficient l-Gln restricted the formation of γ-glutamyl peptides, and this was why DPMH-G had a lower yield and variety than DPMH-G-Q. This also suggested that l-glutaminase is selective to different substrates. Overall, this study provides a new method to reduce the bitterness of protein hydrolysates and also improve the taste by synthesizing γ-glutamyl tripeptides.

Abstract:Corynebacterium glutamicum was used to ferment wheat gluten hydrolysates (WGHs) to prepare flavour base. This study investigated the effect of hydrolysis degrees (DHs) and fermentation time on flavour of WGHs. During fermentation, the contents of amino nitrogen, total acid and small peptides increased, while the protein and pH value decreased. Succinic acid, GMP, and Glu were the prominent umami substances in fermented WGHs. The aromas of WGHs with different DHs could be distinguished by electronic nose and GC-IMS. Based on OAV of GC-MS, hexanal was the main compound in WGHs, while phenylethyl alcohol and acetoin were dominant after fermentation. WGHs with high DHs accumulated more flavour metabolites. Correlation analysis showed that small peptides (<1 kDa) could promote the formation of flavour substances, and Asp was potentially relevant flavour precursor. This study indicated that fermented WGHs with different DHs can potentially be used in different food applications based on flavour profiles.

Abstract:Fermentation with Weissella confusa A16 could improve the flavor of various plant-based sources. However, less is known about the influence of fermentation conditions on the profile of volatile compounds, dextran synthesis and acidity. The present work investigates the synthesis of potential flavor-active volatile compounds, dextran, acetic acid, and lactic acid, as well as the changes in viscosity, pH, and total titratable acidity, during fermentation of faba bean protein concentrate with W. confusa A16. A Response Surface Methodology was applied to study the effect of time, temperature, dough yield, and inoculum ratio on the aforementioned responses. Twenty-nine fermentations were carried out using a Central Composite Face design. A total of 39 volatile organic compounds were identified: 2 organic acids, 7 alcohols, 8 aldehydes, 2 alkanes, 12 esters, 3 ketones, 2 aromatic compounds, and 3 terpenes. Long fermentation time and high temperature caused the formation of ethanol and ethyl acetate and the reduction of hexanal, among other compounds linked to the beany flavor. Levels of dextran, acetic acid, and lactic acid increased with increasing temperature, time, and dough yield. Optimal points set for increased dextran and reduced acidity were found at low temperatures and high dough yield. Such conditions would result in hexanal, ethyl acetate and ethanol having a relative peak area of 35.9%, 7.4%, and 4.9%, respectively.

Abstract:Current food production methods and consumption behaviours are unsustainable and contribute to environmental harm. One example is food waste—around 38% of food produced is wasted each year. Here, we show that two common food waste products, wheat bran and brewer's spent grain, can successfully be upcycled via miso fermentation. During the fermentation process, kokumi γ-glutamyl peptides, known to increase mouthfulness, are produced; these include γ-ECG (oxidized), γ-EVG, γ-EV, γ-EE, γ-EF, and γ-EL. The profiles of kokumi peptides and volatile aroma compounds are correlated with koji substrate, pH, and enzymatic activity, offering straightforward parameters that can be manipulated to increase the abundance of kokumi peptides during the fermentation process. Correlation analysis demonstrates that some volatile aroma compounds, such as fatty acid ethyl esters, are correlated with kokumi peptide abundance and may be responsible for fatty, greasy, and buttery aromas. Consumer sensory analysis conveys that the bitter taste of vegetables, such as that in endives, can be dampened when miso extract containing kokumi peptides is added. This suggests that kokumi peptides, along with aroma volatile compounds, can enhance the overall flavour of plant-based products. This study opens new opportunities for cereal processing by-product upcycling via fermentation, ultimately having the potential to promote a plant-based diet.
