Abstract:For conductive hydrogels applied in biosensors, wearable devices and so forth, multifunctionality is an inevitable trend of development to meet various practical requirements and enhance human experience. Herein, inspired by nanocomposite, double-network (DN) and mussel chemistry, a new Graphene oxide@Dopamine/Alginate/Poly (acrylic acid-co-acrylamide) [GO@DA/Alginate/P(AAc-co-AAm)] hydrogel was fabricated through one-pot in-situ radical copolymerization. GO@DA nanofillers, prepared via GO confined DA polymerization, imparted the hydrogel with remarkable adhesiveness. Alginate/P(AAc-co-AAm) DN matrix, physically and chemically crosslinked by Fe3+ and N,N'-Methylenebisacrylamide, made hydrogels ultrastretchable, self-healing and biocompatible. With contents of DA and alginate accurately regulated, the tensile strength, elongation, adhesion strength and conductivity of the optimal hydrogel could reach 320.2 kPa, 1198 %, 36.9 kPa and 3.24 +/- 0.12 S/m, respectively. What's more notable was that the synergistic integration of repeatable adhesiveness, strain sensitivity, use stability, self-healing ability and biocompatibility provided such hydrogels with tremendous possibility of practical application for strain sensors.
Mechanically tuneable physical nanocomposite hydrogels from polyelectrolyte complex templated silica nanoparticles for anionic therapeutic delivery

Abstract:Hydrogels have shown great promise for drug delivery and tissue engineering but can be limited in practical applications by poor mechanical performance. The incorporation of polymer grafted silica nanoparticles as chemical or physical crosslinkers in in situ polymerised nanocomposite hydrogels has been widely researched to enhance their mechanical properties. Despite the enhanced mechanical stiffness, tensile strength, and self-healing properties, there remains a need for the development of simpler and modular approaches to obtain nanocomposite hydrogels. Herein, we report a facile protocol for the polyelectrolyte complex (PEC) templated synthesis of organic-inorganic hybrid poly(ethylenimine) functionalised silica nanoparticles (PEI-SiNPs) and their use as multifunctional electrostatic crosslinkers with hyaluronic acid (HA) to form nanocomposite hydrogels. Upon mixing, electrostatic interactions between cationic PEI-SiNPs and anionic HA resulted in the formation of a coacervate nanocomposite hydrogel with enhanced mechanical stiffness that can be tuned by varying the ratios of PEI-SiNPs and HA present. The reversible electrostatic interactions within the hydrogel networks also enabled self-healing and thixotropic properties. The excess positive charge present within the PEI-SiNPs facilitated high loading and retarded the release of the anionic anti-cancer drug methotrexate from the nanocomposite hydrogel. Furthermore, the electrostatic complexation of PEI-SiNP and HA was found to mitigate haemotoxicity concerns associated with the use of high molecular weight PEI. The method presented herein offers a simpler and more versatile strategy for the fabrication of coacervate nanocomposite hydrogels with tuneable mechanical stiffness and self-healing properties for drug delivery applications.

Abstract:Microfluidics is emerging as a promising tool to control physicochemical properties of nanoparticles and to accelerate clinical translation. Indeed, microfluidic-based techniques offer more advantages in nanomedicine over batch processes, allowing fine-tuning of process parameters. In particular, the use of microfluidics to produce nanoparticles has paved the way for the development of nano-scaled structures for improved detection and treatment of several diseases. Here, ionotropic gelation is implemented in a custom-designed microfluidic chip to produce different nanoarchitectures based on chitosan-hyaluronic acid polymers. The selected biomaterials provide biocompatibility, biodegradability and non-toxic properties to the formulation, making it promising for nanomedicine applications. Furthermore, results show that morphological structures can be tuned through microfluidics by controlling the flow rates. Aside from the nanostructures, the ability to encapsulate gadolinium contrast agent for magnetic resonance imaging and a dye for optical imaging is demonstrated. In conclusion, the polymer nanoparticles here designed revealed the dual capability of enhancing the relaxometric properties of gadolinium by attaining Hydrodenticity and serving as a promising nanocarrier for multimodal imaging applications.
Mussel-inspired Tough Hydrogels with Self-Repairing and Tissue Adhesion

Abstract:The mussel-inspired polymeric hydrogels have been attractively explored owing to their self-repairing or adhesive property when the catechol groups of dopamine could chelate metal ions. However, it was a challenge for self-repairing hydrogels owning high mechanical properties. Herein, a synergistic strategy was proposed by combining catechol-Fe3+ complexes and hydrophobic association. The resulting hydrogels exhibited seamless self-repairing behavior, tissue adhesion and high mechanical property. Moreover, the pH-dependent stoichiometry of catechol-Fe3+ and temperature-sensitive hydrophobic association endue hydrogels with pH/thermo responsive characteristics. Subsequently, the self-repairing rate and mechanical property of hydrogels were investigated at different pH and temperature. This bio-inspired strategy would build an avenue for designing and constructing a new generation of self-repairing, tissue-adhesive and tough hydrogel. (C) 2017 Elsevier B.V. All rights reserved.

Abstract:A method for grafting dopamine onto TEMPO-oxidized chitin nanofibers (TOChN) was developed, achieving a surface grafting rate of 54 % through the EDC/NHS reaction. This process resulted in the formation of dopamine-grafted TOChN (TOChN-DA). Subsequently, an adherent, highly sensitive, fatigue-resistant conductive PAM/TOChN-PDA/Fe 3+ (PTPF) hydrogel was successfully synthesized based on the composition of polyacrylamide (PAM) and TOChN-DA, which exhibited good cell compatibility, a tensile strength of 89.42 kPa, and a high adhesion strength of 62.56 kPa with 1.2 wt% TOChN-DA. Notably, the PTPF hydrogel showed stable adherence to various surfaces, such as rubber, copper, and human skin. Specifically, the addition of FeCl 3 contributed to a multifunctional design in the PTPF interpenetrating network (IPN) hydrogel, endowing it with conductivity, cohesion, and antioxidant properties, which facilitated sensitive motion and acoustics monitoring. Moreover, the PTPF hydrogel demonstrated exceptional fatigue resistance and sensing stability, maintaining performance at 50 % strain over 1000 cycles. These attributes render the PTPF hydrogel a promising candidate for advanced biosensors in medical and athletic applications.

Abstract:A novel mussel-inspired adhesive hydrogel with enhanced adhesion based on methacrylated catechol-chitosan (MCCS) and dopamine methacrylate (DMA) was prepared via photopolymerization. The structure and morphology of the MCCS/DMA adhesive hydrogel were investigated by using FTIR, NMR, XRD, TG, and SEM. The rheological and texture properties, swelling and degradation characteristics, as well as the adhesion mechanism of the hydrogels were also examined. These results revealed that the MCCS/DMA hydrogels have a dense double cross-linking network structure with porous internal microstructures, and exhibited controllable swelling and degradation properties, good thermostability, and stable rheological characteristics. Furthermore, the adhesive mechanism of MCCS/DMA hydrogel has been confirmed by the FTIR and 2D correlation FTIR spectroscopy. Additionally, the results of in vitro cytotoxicity assessment indicated that the resulting hydrogels have good cytocompatibility. Overall, the MCCS/DMA adhesive hydrogel may have potential applications in medical bioadhesives.

Abstract:In this paper, a new formulation of biodegradable and bioresorbable chitosan-based hydrogel for controlled drug release was investigated. A chitosan–dendrimer–hydroxyapatite hydrogel, obtained by covalently grafting chitosan powder with an hyperbranched PAMAM dendrimer followed by in-situ precipitation of hydroxyapatite and gelification, was synthesized and characterized by FTIR, NMR, TGA, XRD and rheological studies. The hydrogels have been also doped with an anti-inflammatory drug (ketoprofen) in order to investigate their drug release properties. Chemical and chemical-physical characterizations confirmed the successful covalent functionalization of chitosan with PAMAM and the synthesis of nanostructured hydroxyapatite. The developed hydrogel made it possible to obtain an innovative system with tunable rheological and drug-releasing properties relative to the well-known formulation containing chitosan and hydroxyapatite powder. The developed hydrogel showed different rheological and drug-releasing properties of chitosan matrix mixed with hydroxyapatite as a function of dendrimer molecular weight; therefore, the chitosan–dendrimer–hydroxyapatite hydrogel can couple the well-known osteoconductive properties of hydroxyapatite with the drug-release behavior and good processability of chitosan–dendrimer hydrogels, opening new approaches in the field of tissue engineering based on biopolymeric scaffolds.

Abstract:Conductive hydrogels with tunable multifunctionality show great potential in wearable smart devices. In this work, inspired by the nano-structured architecture strategy and natural mussel adhesive mechanism, a conductive hydrogel (AgPMP hydrogel) was fabricated through facile direct in-situ polymerization. Silver nanowires (Ag NWs) and polydopamine (PDA) chains were integrated into the poly(N-isopropylacrylamide-co-acrylamide) (P(NIPAM-co-AM)) gel matrix, with activated PNIPAM microgels (MGs) serving as nano-crosslinkers. The overall internal structure of the hydrogel presented a heterogeneous cross-linked mesh-like structure with numerous irregular wrinkles. The AgPMP hydrogel demonstrated significantly enhanced mechanical strength, achieving a remarkable stretchability of 1140% attributed to the nano-microstructured architecture built by AM-based network, nano-crosslinkers and Ag NWs. Moreover, due to the synergistic effect of PDA and Ag NWs, the hydrogel exhibited impressive photothermal antibacterial activity. Meanwhile, in the presence of abundant active catechol groups, this hydrogel possessed adequate tissue-adhesive and self-healing abilities as well as good antioxidant property. Notably, the AgPMP hydrogel exhibited outstanding strain-sensitive conductivity. The resultant strain sensor owned high sensitivity with a guage factor (GF) up to 8.35 at strain of 250%. The stable response signals during continuous and gradual cycling proved its electrical durability and stability. Combined with the great merits of above performance, this sensor holds great promise for detecting huaman movements. The relative resistance change ranged from 16% to 64% as the finger bending angle changed from 30° to 90°. Thanks to its simple fabrication process, this multifunctional conductive hydrogel is expected to have widespread potential applications in tissue engineering, soft electronic skin and human-health monitoring.

Abstract:Hydrogels, with 3D hydrophilic polymer networks and excellent biocompatibilities, have emerged as promising biomaterial candidates to mimic the structure and properties of biological tissues. The incorporation of nanomaterials into a hydrogel matrix can tailor the functions of the nanocomposite hydrogels to meet the requirements for different biomedical applications. However, most nanomaterials show poor dispersion in water, which limits their integration into the hydrophilic hydrogel network. Mussel-inspired chemistry provides a mild and biocompatible approach in material surface engineering due to the high reactivity and universal adhesive property of catechol groups. In order to attract more attention to mussel-inspired nanocomposite hydrogels, and to promote the research work on mussel-inspired nanocomposite hydrogels, we have reviewed the recent advances in the preparation of mussel-inspired nanocomposite hydrogels using a variety of nanomaterials with different forms (nanoparticles, nanorods, nanofibers, nanosheets). We give an overview of each nanomaterial modified or hybridized by catechol or polyphenol groups based on mussel-inspired chemistry, and the performances of the nanocomposite hydrogel after the nanomaterial’s incorporation. We also highlight the use of each nanocomposite hydrogel for various biomedical applications, including drug delivery, bioelectronics, wearable/implantable biosensors, tumor therapy, and tissue repair. Finally, the challenges and future research direction in designing mussel-inspired nanocomposite hydrogels are discussed.

Abstract:The field of hydrogels has rapidly expanded in recent years. A growing theme is the development of hydrogels that are bioinspired, with tailorable physicochemical properties. Marine mussels utilize their byssus to adhere themselves to rocks and thereby decrease the turbulent buffeting of the intertidal zone. An increase in pH is a noticeable phenomenon during the mussel byssus secretion and formation processes. In this study, by mimicking the acidic-to-basic pH switch that occurs during byssus formation, a series of bioinspired, pH-initiated hyaluronic acid (HA) hydrogels that possess tunable cohesive and adhesive properties were developed. Specifically, four different kinds of hyaluronic acid (HA) hydrogel crosslinking networks were fabricated by utilizing the multifaceted nature of catechol chemistry, including metal-catechol coordination, quinone-catechol dismutation, quinone-amine Michael addition and Schiff base reactions, or quinone-thiol Michael addition reactions. Our research demonstrated that pH, as one of the most important environmental variables for mussels, plays a vital role in regulating the cohesion and adhesion behaviors. And regarding the development of new materials, this biomimetic strategy also demonstrates alternative and tunable crosslinking approaches that can overcome the limitations of current HA hydrogels.
Supramolecular polymer/peptide hybrid hydrogels with tunable stiffness mediated by interchain acid-amide hydrogen bonds

Abstract:Hydrogels are a class of biomaterials used in the field of tissue engineering and drug delivery. Many tissue engineering applications depend on the material properties of hydrogel scaffolds, such as mechanical stiffness, pore size, and interconnectivity. In this work, we describe the synthesis of peptide/polymer hybrid double-network (DN) hydrogels composed of supramolecular and covalent polymers. The DN hydrogels were prepared by combining the self-assembled pentafluorobenzyl diphenylalanyl aspartic acid (PFB-FFD) tripeptide for the first network and the polymeric PNIPAM-PEGDA copolymer for the second network. During this process, self-assembled peptide nanostructures are cross-linked to the polyacrylamide group in the polymer network through non-covalent interactions. The PNIPAM-PEGDA:PFB-FFD hydrogel exhibited higher mechanical stiffness ( G ′ ∼2 kPa) than the PNIPAM-PEGDA copolymer. Moreover, PNIPAM-PEGDA:PFB-FFD hydrogel shows a decrease in pore size (∼1.2 μm) compared to the original copolymer (∼5.2 μm), with the structural framework of highly interconnected fibrous peptide network. The mechanical stiffness of hydrogels was systematically investigated by rheological analysis in response to various variables, including UV exposure time, concentration of peptides, and amino acid functionalization. Modulating the time of UV irradiation resulted in PNIPAM-PEGDA:PFB-FFD hydrogels with a four-fold increase in stiffness. The influence of amino acid side chains and terminal charge of peptides on the strength of DN hydrogels was also investigated using pentafluorobenzyl diphenylalanyl lysine (PFB-FFK). Interestingly, PFB-FFK, which has an amine group on the side chain, does not exhibit the DN structures. The mechanical properties and pore sizes of PNIPAM-PEGDA:PFB-FFK hydrogel were very similar to those of the PNIPAM-PEGDA copolymer due to poor cross-linking. The biocompatibility of the hydrogel materials was tested with the hMSC cell line using the MTT method, and the results indicate that the materials are non-toxic and potentially useful for biological applications.

Abstract:A robust biodegradable hydrogel is constructed from two components containing G4.0 PAMAM and DOPA, which displays excellent interconnected porous 3D networks, higher mechanical strength, low and stable swelling and good biocompatibility. Abstract:Hyaluronic acid-based hydrogels have been broadly used in medical applications due to their remarkable properties such as biocompatibility, biodegradability, super hydroscopicity, non-immunogenic effect, etc. However, the inherent weak and hydrophilic polysaccharide structure of pure hyaluronic acid (HA) hydrogels has limited their potential use in muco-adhesiveness, wound dressing, and 3D printing. In this research, we developed in-situ forming of catechol-modified HA hydrogels with improved mechanical properties involving blue-light curing crosslinking reaction. The effect of catechol structure on the physicochemical properties of HA hydrogels was evaluated by varying the content (0 - 40 %). The as-synthesized hydrogel demonstrated rapid prototyping, excellent wetting adhesiveness, and good biocompatibility. Furthermore, an optimized hydrogel precursor solution was used as a blue light-cured bio-ink with high efficiency and good precision and successfully prototyped a microstructure that mimicked the human hepatic lobule by using DLP 3D printing method. This catechol-modified HA hydrogel with tunable physicochemical and rapid prototyping properties has excellent potential in biomedical engineering.

Abstract:Alginate is a representative biocompatible natural polymer with low cost for a variety of biomedical applications, such as wound dressing, drug delivery systems, tissue scaffolds, and 3D bioprinting. Particularly, the rapid and facile gelation of alginate via ionic interactions with divalent cations has been used for in situ 3D hydrogel fiber formation, which is potentially applicable to engineering cell alignment. However, challenges in enhancing the mechanical properties of alginate hydrogel fibers under physiological conditions are unresolved because of their fast dissociation by ion exchange. Herein, we report a stabilization strategy for alginate hydrogel fibers through mussel-inspired catechol chemistry, which involves inter-catechol crosslinking within a few minutes under basic conditions. The fabrication of catechol-tethered alginate hydrogel fibers through wet-spinning enabled the design of mechanically strong 3D constructs consisting of fibers. Catechol-to-quinone oxidation followed by covalent crosslinking enhanced the tensile strength of a single fiber. Additionally, the ‘gluing’ capability of the catechol stabilized the interface among the fibers, thus retaining the shape fidelity of the 3D constructs and encapsulating the cell density during culture. Our findings will be useful for designing bioink materials specialized in fibrous-type tissue scaffolds with mechanical stability.

Abstract:Commercial or clinical tissue adhesives are currently limited due to their weak bonding strength on wet biological tissue surface, low biological compatibility, and slow adhesion formation. Although catechol-modified hyaluronic acid (HA) adhesives are developed, they suffer from limitations: insufficient adhesiveness and overfast degradation, attributed to low substitution of catechol groups. In this study, we demonstrate a simple and efficient strategy to prepare mussel-inspired HA hydrogel adhesives with improved degree of substitution of catechol groups. Because of the significantly increased grafting ratio of catechol groups, dopamine-conjugated dialdehyde-HA (DAHA) hydrogels exhibit excellent tissue adhesion performance (i.e., adhesive strength of 90.0 +/- 6.7 kPa), which are significantly higher than those found in dopamine-conjugated HA hydrogels (similar to 10 kPa), photo-cross-linkable HA hydrogels (similar to 13 kPa), or commercially available fibrin glues (2-40 kPa). At the same time, their maximum adhesion energy is 384.6 +/- 26.0 J m(-2), which also is 40-400-fold, 2-40-fold, and similar to 8-fold higher than those of the mussel-based adhesive, cyanoacrylate, and fibrin glues, respectively. Moreover, the hydrogels can gel rapidly within 60 s and have a tunable degradation suitable for tissue regeneration. Together with their cytocompatibility and good cell adhesion, they are promising materials as new biological adhesives.

Abstract:Using supramolecular self-assembled nanocomposite materials made from protein and polysaccharide components is becoming more popular because of their unique properties, such as biodegradability, hierarchical structures, and tunable multifunctionality. However, the fabrication of these materials in a reproducible way remains a challenge. This study presents a new evaporation-induced self-assembly method producing layered hydrogel membranes (LHMs) using tropocollagen grafted by partially deacetylated chitin nanocrystals (CO-g-ChNCs). ChNCs help stabilize tropocollagen's helical conformation and fibrillar structure by forming a hierarchical microstructure through chemical and physical interactions. The LHMs show improved mechanical properties, cytocompatibility, and the ability to control drug release using octenidine dihydrochloride (OCT) as a drug model. Because of the high synergetic performance between CO and ChNCs, the modulus, strength, and toughness increased significantly compared to native CO. The biocompatibility of LHM was tested using the normal human dermal fibroblast (NHDF) and the human osteosarcoma cell line (Saos-2). Cytocompatibility and cell adhesion improved with the introduction of ChNCs. The extracted ChNCs are used as a reinforcing nanofiller to enhance the performance properties of tropocollagen hydrogel membranes and provide new insights into the design of novel LHMs that could be used for various medical applications, such as control of drug release in the skin and bone tissue regeneration.

Abstract:Dynamic chitosan-based hydrogels with enhanced antioxidant activity were synthesized through the formation of reversible imine linkages with 5-methoxy-salicylaldehyde. These hydrogels exhibited a porous structure and swelling capacity, influenced by the crosslinking degree, as confirmed by SEM and POM analysis. The dynamic nature of the imine bonds was characterized through NMR, swelling studies in various media, and aldehyde release measurements. The hydrogels demonstrated significantly improved antioxidant activity compared to unmodified chitosan, as evaluated by the DPPH method. This research highlights the potential of developing pH–responsive chitosan-based hydrogels for a wide range of biomedical applications.

Abstract:Glucose monitoring with high sensitivity and accuracy in the cerebrospinal fluid is a challenge for evaluating the role of glucose in the physiological and pathological processes. In this work, a ratiometric fluorescent probe for sensing glucose was developed. In the probe, the gold nanoclusters protected by ovalbumin played the role as the reference of fluorophore and the Alizarin Red S-3-aminophenyl boronic acid immobilized on the poly(N-acryloxysuccinimide) acted as both the response signal and specific recognition unit for sensing glucose. Once the ratiometric fluorescent probe reacted with glucose in the biological system, its fluorescence intensity at 567 nm was quenched, while the fluorescence intensity at 610 nm was essentially unchanged. In addition, the prepared ratiometric fluorescent probe showed higher stability against environmental effects. As a result, the present ratiometric fluorescent probe was successfully used for monitoring of glucose in the rat brain following the cerebral calm/ischemia.

Abstract:Advanced bioactive systems with defined macroscopic properties and spatio-temporal sequestration of extracellular biomacromolecules are highly desirable for next generation therapeutics. Here, chitosan hydrogels were prepared with neutral or negatively-charged crosslinkers in order to promote selective electrostatic complexation with charged drugs. Chitosan (CT) was functionalised with varied dicarboxylic acids, such as tartaric acid (TA), poly(ethylene glycol) bis(carboxymethyl) ether (PEG), 1.4-Phenylenediacetic acid (4Ph) and 5-Sulfoisophthalic acid monosodium salt (PhS), whereby PhS was hypothesised to act as a simple mimetic of heparin. ATR FT-IR showed the presence of C=O amide I, N-H amide II and C=O ester bands, providing evidence of covalent network formation. The crosslinker content was reversely quantified by 1H-NMR on partially-degraded network oligomers, so that 18 mol% PhS was exemplarily determined. Swellability, compressability, material morphology, and drug-loading capability were successfully adjusted based on the selected network architecture. Here, hydrogel incubation with model drugs of varied electrostatic charge, i.e. allura red (AR, --), methyl orange (MO, -) or methylene blue (MB, +), resulted in direct hydrogel-dye electrostatic complexation. Importantly, the cationic compound, MB, showed different incorporation behaviours, depending on the electrostatic character of the selected crosslinker. In light of this tuneable drug-loading capability, these CT hydrogels would be highly attractive as drug reservoirs towards e.g. the fabrication of tissue models in vitro.
