Abstract:The japonica rice lines growing in field were used as experimental materials.The effects of genotype,low temperature pretreatment of spikes,2,4-D,2,4,5-T combined with sucrose or maltose in induction medium on in vitro culture of rice anther were investigated.The culture response of anthers was critical to the donor genotype and a large number of callus and green plantlets were obtained from high responding lines.The culture response of anther from pretreatment of spikes at 5℃ for 8 days was higher than that of spikes pretreated at 5℃ for 4 days.The culture response of anther were increased by adding 0.5mg/L 2,4,5-T and 50g/L sucrose in induction medium.Large quantity of embryo like structure and green plantlets were obtained from in vitro culture of microspores of 05P-27 and 05P-40 lines.
Development of In Vitro Anther Culture for Doubled Haploid Plant Production in Indica Rice (Oryza sativa L.) Genotypes

Abstract:Anther culture is an efficient biotechnological tool in modern plant breeding programs to produce new varieties and parental lines in hybrid seed productions. However, some bottlenecks-low induction rate, genotype dependency, albinism-restrict the widespread utilization of in vitro anther culture in rice breeding, especially in Oryza sativa ssp. indica (indica) genotypes, while an improved efficient protocol can shorten the process of breeding. Three different induction media (N6NDK, N6NDZ, Ali-1) and four plant regeneration media (mMSNBK1, MSNBK3, MSNBKZ1, MSNBKZ2) were tested with five indica rice genotypes to increase the efficiency of in vitro androgenesis (number of calli and regenerated green plantlets). The production of calli was more efficient on the N6NDK medium with an average 88.26 calli/100 anthers and N6NDZ medium with an average of 103.88 calli/100 anthers as compared to Ali-1 with an average of 6.96 calli/100 anthers. The production of green plantlets was greater when calli was produced on N6NDK medium (2.15 green plantlets/100 anthers) compared when produced on to N6NDZ medium (1.18 green plantlets/100 anthers). Highest green plantlets production (4.7 green plantlets/100 anthers) was achieved when mMSNBK1 plant regeneration medium was used on calli produced utilizing N6NDK induction medium. In the best overall treatment (N6NDK induction medium and mMSNBK1 plant regeneration medium), four tested genotypes produced green plantlets. However, the genotype influenced the efficiency, and the green plantlets production ranged from 0.4 green plantlets/100 anthers to 8.4 green plantlets/100 anthers. The ploidy level of 106 acclimatized indica rice plantlets were characterized with flow cytometric analyses to calculate the percentage of spontaneous chromosome doubling. Altogether, 48 haploid-, 55 diploid-, 2 tetraploid- and 1 mixoploid plantlets were identified among the regenerant plantlets, and the spontaneous chromosome doubling percentage was 51.89%. Utilization of DH plants have been integrated as a routine method in the Hungarian rice breeding program. The tetraploid lines can be explored for their potential to offer new scopes for rice research and breeding directions in the future.
Breeding of an Indica-compatible Japonica Photoperiod-sensitive Genic Male Sterile Line in Rice and Its Heterosis

Abstract:The incomplete fertility of japonica x indica rice hybrids has inhibited breeders' access to the substantial level of heterotic potential of these hybrids.As hybrid sterility is caused by an allelic interaction at a small number of loci,it is possible to overcome it by simple introgression at the major sterility loci.Here we report the use of marker-assisted backcrossing to transfer into the elite japonica variety.Zhendao 88 a photoperiod-and thermo-sensitive male sterility gene from Lunhui 422S(indica) and the yellow leaf gene from the line Yellow 249(indica).The microsatellite markers RM276,RM455,RM141 and RM185 were used to tag the fertility genes S5,S8,S7 and S9,respectively.Line 509S is a true-breeding photoperiod-and thermo-sensitive genic male sterile(PTGMS) line,which morphologically closely resembles the japonica-type.Genotypic analysis showed that the genome of 509S had about 92% japonica DNA.Nevertheless,hybrids between 509S and japonica varieties sufferred from a level of hybrid sterility,although the line is highly cross-compatible with indica types,with the resulting hybrids expressing a significant degree of heterosis.Besides,the results suggested that segment substitution on fertility loci based on known information and marker-assisted selection was an effective approach to utilizing the heterosis of rice inter-subspecies.

Abstract:The potential heterosis present in japonica x indica inter-subspecific hybrids is great,and represents a clear next step in the search for increasing the economic yield of rice.However,the problems of too tall plants,too long maturity and partial sterility in these hybrids have inhibited their use in hybrid rice breeding.Aiming at these limitative factors,some basic solutions for figuring out these issues were proposed on gthe basis of breeding practice.Besides,the results suggested that the marker-assisted selection was an effective approach to utilizing the heterosis of rice inter-subspecies.

Abstract:Recently Agrobacterium-mediated transformation of rice has been widely used in japonica transformation.However,the transformation of indica rice is still difficult,mainly because the induction of embryogenic callus is not routine and the rate of plant regeneration from transformed calli is low.There are big genetic differences between these 2 subspecies,and indica rice is particularly genotype-dependent.This paper briefly reviewed the history of agrobacterium-mediated transformation of indica rice and recent progress achieved.It also introduced various factors affecting indica rice transformation,analyzed problems existing in its research and provided strategy to improve the efficiency of transformation.The establishment of high efficient indica rice transformation system will accelerate the process of applying transformation technology in rice breeding programs and promote the studies on rice functional genomics.

Abstract:Inheritance of three anther and culture characters, callus induction, green plant regeneration and culture efficiency was studied using incomplete diallel crosses with a gamete model. It was suggested that callus induction was mainly controlled by gametic additive effects and with less effect of the maternal effects. Green plant regeneration was mainly determined by maternal effects with less influence of gametic additive effects. Culture efficiency was controlled by gametic additive, maternal and cytoplasmic effects. Cultivar Lunhui 422 showed positive genetic effects for all three traits and was a very good parent for rice anther culture breeding. Significant positive heterosis was observed for callus induction. Both gametic additive and maternal correlations contributed to the significant genotypic and phenotypic correlations between callus induction and green plant regeneration suggesting these two traits to be linked.

Abstract:Hybrid sterility restricts the utilization of superior heterosis of indica-japonica inter-subspecific hybrids. In this study, we report the identification of RHS12, a major locus controlling male gamete sterility in indica-japonica hybrid rice. We show that RHS12 consists of two genes (iORF3/DUYAO and iORF4/JIEYAO) that confer preferential transmission of the RHS12-i type male gamete into the progeny, thereby forming a natural gene drive. DUYAO encodes a mitochondrion-targeted protein that interacts with OsCOX11 to trigger cytotoxicity and cell death, whereas JIEYAO encodes a protein that reroutes DUYAO to the autophagosome for degradation via direct physical interaction, thereby detoxifying DUYAO. Evolutionary trajectory analysis reveals that this system likely formed de novo in the AA genome Oryza clade and contributed to reproductive isolation (RI) between different lineages of rice. Our combined results provide mechanistic insights into the genetic basis of RI as well as insights for strategic designs of hybrid rice breeding.

Abstract:Exploitation of heterosis has brought significant advance in plant breeding and agricultural production, although its genetic basis is still poorly understood. In this study, a total of 66 chromosome segment substitution (CSS) lines, derived from a cross between japonica rice inbred line Asominori (as the recurrent parent) and indica rice inbred line IR24 (as the donor parent), were used to investigate the genetic basis of heterosis in indica × japonica inter-subspecific rice hybrids. Each CSS line was crossed with the background parent Asominori, and the heterosis of F1 hybrids was estimated by comparing the F1 performance with its two parental lines. Field experiments were carried out across six different environments to evaluate yield and yield-related traits in the 66 CSS lines and their 66 corresponding F1 hybrids. Quantitative trait loci (QTL) analyses were conducted using a likelihood ratio test based on the stepwise regression. Thirty-six QTL were identified with significant effects in CSSL, 21 with significant effects in hybrids and 13 with significant effects in both. On the basis of average dominance degree, of all the 70 QTL affecting yield-related agronomic traits, 28.6% (20) showed an overdominance, 35.7% (25) a partial dominance and 30% (21) an additive effect, indicating that all effects contribute to trait variation in japonica–indica rice hybrids. Effects of these QTL were examined to identify Indica rice chromosome segments of interest for the improvement of japonica inbred lines and hybrids.

Abstract:Hybrids between rice subspecies indica and japonica display strong heterosis. However, semi-sterility of inter-subspecific hybrids between indica and japonica varieties is a major obstacle for application of hybrid vigor in rice production. Semi-sterility was previously ascribed to allelic interaction at a number of different loci, whereas, wide-compatibility varieties can overcome hybrid sterility. Variety Nekken 2, which is a source of wide compatibility genes, showed sterility when crossed to the Korean variety Yeong Pung. Genetic and cytological analyses revealed that the semi-sterility was caused by partial abortion of the embryo sac. Genome-wide analysis of the backcross population, Nekken 2/Yeong Pung//Nekken 2 identified two independent loci for hybrid sterility on chromosomes 1 and 12, explaining 18.99 and 18.03% of the phenotypic variance, respectively. To confirm this result, another population of the same backcross containing 216 individuals was tested at a different site in a different year. The locus on chromosome 12 was detected again. Based on the study, the stable QTL on chromosome 12 appeared to be different from previously reported genes for this trait, and was designated as S35(t) .

Abstract:Heading date (HD) is a key trait for the adaptation of rice cultivar to a specific growing region. Here, we report conventional and marker-assisted breeding strategies using genetic information related to the determination of HD, where the breeding objectives were to avoid the delayed heading common in indica × japonica hybrids, to increase the efficiency in selecting hybrid rice combinations having a suitable growth duration, and to develop cultivars with target growth duration by quantitative trait locus (QTL) pyramiding. The allelic constitution at the major HD loci was determined for a set of 109 leading Chinese rice cultivars by crossing them with HD tester lines. It was shown that the late heading in indica × japonica hybrids can be overcome by replacing the strong photoperiod-sensitivity allele Se-1 n with Se-1 e . A breeding strategy to enable the selection of hybrid combinations with suitable growth duration was proposed, based on HD genotypic information in rice. Meanwhile, a QTL analysis for HD was conducted over five years based on a recombinant inbred line population, derived from two parents Asominori (japonica) and IR24 (indica). Four QTLs, located on chromosomes 2, 3, 6, and 8, respectively, could be detected in all five years, indicating they were stably expressed QTL. According to this QTL information, and taking Asominori as an example, the HD genotypes for improving the growth duration were designed, and the best breeding selection schemes were determined by use of a genetic breeding simulation tool. Results obtained in this study demonstrate that genetic information related to HD can make a significant contribution to rice breeding.

Abstract:Asian cultivated rice (Oryza sativa L.) and African cultivated rice (Oryza glaberrima Steud.) are the two main cultivated rice species in the world, with strong heterosis in their F1 hybrids. However, hybrid sterility is a major barrier, although significant heterosis has been observed. In this study, an F1 pollen semi-sterility locus, S19, was identified on rice chromosome 3 by using near-isogenic lines derived from repeated backcross and marker-assisted selection. The typical pollen semi-sterility was observed in F1 hybrids between S19-NIL and Dianjingyou 1. Cytological study of pollen developmental stages indicated that pollen abortion occurred at the late binucleate stage because of a starch accumulation obstacle in some pollen grains. Molecular analysis revealed that the semi-sterility was caused by the abortion of most male gametophytes carrying the S19 allele from the japonica variety Dianjingyou 1. In a population of 12,780 F2 plants derived from S19-NIL/Dianjingyou 1, the S19 locus was fine-mapped to a chromosomal region of 54 kb based on BAC clones of cv. Nipponbare. Interestingly, an addition of a DNA fragment of about 89 kb to the 54-kb region was found in S19-NIL based on BAC clones of O. glaberrima. Gene prediction analysis identified 12 open reading frames (ORF) based on the region of Dianjingyou 1, while 32 ORFs were predicted in S19-NIL. Map-based cloning of this gene will help us to understand the underlying mechanism of hybrid sterility between the two cultivated rice species.
Study on heterosis of inter-subspecies between indica and japonica rice ( Oryza sativa L.) using chromosome segment substitution lines

Abstract:Heterosis for yield and its component traits between chromosome segments from IR24, an indica variety, and the counterparts from 02428, a japonica rice, was investigated by using a hybrid F 1 population composed of 63 combinations between 02428 and IR24 chromosome segment substitution lines (CSSLs) with the genetic background of Asominori, a japonica variety. Significant differences in heterosis for yield and yield-component traits were observed among the crosses. Analysis of graphical genotyping showed that 14 substituted segments were responsible for yield heterosis. All of them were from all the 12 chromosomes of IR24 except chromosomes 8 and 10. Six segments at the intervals of RFLP markers, such as X132-G1340-R459, X48-C393A, R288-R1854, R2918-X52, X257-C1350 and R367-X189-2-X24-2 on chromosomes 2, 3, 4, 11 and 12 respectively, had very significant heterosis for yield at the level of P ⩽ 0.005 based on t -test, individually increasing the hybrid yield by more than 35% compared with the control cross “Asominori×02428”. Most of IR24 chromosome segments were found to have no significant hybrid effect for yield and yield-component traits, and one segment located at R2171 on chromosome 6 possessed significant negative effect with 27% of yield decrease. Advantages of using CSSLs in the heterosis studies were discussed and approaches of the partial and genome-wide exploitation of rice heterosis between indica and japonica by molecular marker-assisted selection were then proposed.

Abstract:Abstract Exploitation of biotechnological tools in conventional breeding strategies is the need of the hour for overcoming limitations in rice production and productivity. In addition, improvement in quantity and quality along with resistance to climatic and disease stress in rice require immediate attention. Anther culture has proven its efficiency by instantaneously fixing homozygosity through diploidization of regenerated haploid plants. Therefore, androgenesis provides an efficient platform for developing inbred lines in a short period of time. Although anther culture shows its efficiency in speeding up breeding in several crop species, including rice, associated limitations still prevent the exploitation of its optimum potential. Although anther culture is well exploited in japonica rice breeding, its application in indica rice is limited because of inherent recalcitrant genetic backgrounds. The success of anther culture is determined by several factors that limit the efficiency of androgenesis. Identified constraints are early anther necrosis, poor-callus response, and proliferation, and low green-plant regeneration, along with the most frustrating albinism associated with indica rice, which has been considerably clarified. This chapter details the method of androgenesis and scope for improving the applicability of anther culture producing doubled haploids of rice in order to use it as a complementary tool for precision breeding.

Abstract:Isolated microspore culture is a useful biotechnological technique applied in modern plant breeding programs as it can produce doubled haploid (DH) plants and accelerate the development of new varieties. Furthermore, as a single-cell culture technique, the isolated microspore culture provides an excellent platform for studying microspore embryogenesis. However, the reports on isolated microspore culture are rather limited in rice due to the low callus induction rate, poor regeneration capability, and high genotypic dependency. The present study developed an effective isolated microspore culture protocol for high-frequency androgenesis in four japonica rice genotypes. Several factors affecting the isolated microspore culture were studied to evaluate their effects on callus induction and plantlet regeneration.

Abstract:Oxidative stress is one of the major causes of aging considered at both the cellular and organismal levels. There is evidence that this agent may also constitute a common element connecting the aging of cells of higher organisms with a similar process occurring in eukaryotic and prokaryotic unicellulars. The aim of this paper was to present the current state of knowledge on the role of oxidative stress in aging of human somatic cells, human as a whole as well as the yeast Saccharomyces cerevisiae and bacteria Caulobacter crescentus, and Escherichia coli.

Abstract:Rice ( Oryza sativa L.) is an important staple crop. The exploitation of the great heterosis that exists in the inter-subspecific crosses between the indica and japonica rice has long been considered as a promising way to increase the yield potential. However, the male and female sterility frequently occurred in the inter-subspecific hybrids hampered the utilization of the heterosis. Here we report that the inter-subspecific hybrid sterility in rice is mainly affected by the genes at Sb , Sc , Sd and Se loci for F 1 male sterility and the gene at S5 locus for F 1 female sterility. The indica -compatible japonica lines (ICJLs) developed by pyramiding the indica allele ( S -i) at Sb , Sc , Sd and Se loci and the neutral allele ( S -n) at S5 locus in japonica genetic background through marker-assisted selection are compatible with indica rice in pollen fertility and in spikelet fertility. These results showed a great promise of overcoming the inter-subspecific hybrid sterility and exploiting the heterosis by developing ICJLs.

Abstract:This research was undertaken to find an efficient tissue culture system and Agrobacterium-mediated genetic transformation method for recalcitrant indica rice cultivars. For this, mature seeds of commercially important indica rice varieties, ASD16, ADT43, IR 64, and Pusa Basmati were cultured on MS and N6 medium supplemented with 2 mg l-1 2, 4-D + 30 g l-1 sucrose. The calli grown in N6 medium showed better friability and embryogenic response. Out of the four varieties tested, ASD16 and IR64 showed better callusing and embryogenic capacity as compared to ADT43 and Pusa Basmati. For genetic transformation studies, embryogenic calli of all the cultivars were co-cultivated with the Agrobacterium tumefaciens strain LBA 4404 harboring the binary vector pCambia 1305.1 with GUS gene. GUS assay was performed for the putative transformed calli and its activity was found to be qualitatively higher in ASD16 and IR64 than the other two varieties. The best responsive ASD16 transformed calli was regenerated and the putative transgenic lines were regenerated. ASD16 transformed calli were confirmed by GUS assay. PCR analysis confirmed the presence of both GUS and HPT genes in ASD16 transgenic lines.

Abstract:Anther culture technology,combined with cross breeding,induced mutation breeding and transgenic technology,has become an important technology in rice breeding. The anther culture ability of rice is a quantitative trait and the genotype is a key factor in the rice anther culture. Characteristics used for evaluating tissue culture ability was introduced firstly and then genotypic effects and genetic control of anther culture ability in rice were discussed,to provide some useful information for the improvement of rice anther culturability by genetic manipulations.

Abstract:Rice, one of the most important food crops and a model monocot plant, its genetic transformation has attracted widespread attention. Since the world’s first case of transgenic rice got success in 1988, the rice genetic transformation technology has been developed rapidly. In 1994, in a milestone work of Agrobacterium-mediated transformation of a number of japonica rice cultivars, rice genetic transformation system has been more advanced after nearly 20 years of development. Presently, Agrobacterium-mediated transformation and microparticle bombardment are the two most widely used methods for rice genetic transformation, while pollen tube pathway, electroporation and polyethylene glycol (PEG) medium are also employed in some laboratories. Agrobacterium transformation is the method of choice because it is of easy and low cost, the high transformation efficiency, the low copy number of transgenes inserted into the host genome and the stability of expression over generations. Nowadays more than 80% of the transgenic rice is transformed by the Agrobacterium method. Although there are a lot of reports in improving the methods and technology of rice transformation, difficulties and limitations still exist in rice genetic transformation. The transformation efficiency of several elite japonica and many indica varieties is still unsatisfactory as it is much lower. Some transformation protocols are laborious, time consuming, and highly genotype-dependent. Besides, there is a problem of somaclonal variations or somatic mutation frequently occurs in plant cells during in vitro culture. Therefore, it is very important to establish the high efficient, safety, large-scale and standard transformation systems for diverse varieties of rice. This paper briefly reviewed the history and development of rice genetic transformation system. Especially, the recent progress of rice transgenic technology for large scale performance in China was described. Many factors are known to affect the efficiency of genetic transformation, such as the genotype of the recipient plants, the type and age of the tissue being inoculated, the strain of Agrobacterium, the expression vector, the composition of the culture medium, as well as various conditions of tissue culture. Extensive research has been conducted to optimize the high-efficiency transformation system for different genotypes of rice. Various factors affecting the transformation efficiencies were explored. Modification in culture conditions for embryogenic calli induction and regeneration from mature seed and also the compatibility of the Agrobacterium strain were successful in examined transformation protocols. The highly efficient Agrobacterium-mediated transformation system for japonica and indica rice using immature embryos or mature embryo-derived calli as the explant has been established for the large-scale production of transgenic rice by integrating new technology method. Also the Agrobacterium-mediated co-transformation system has been preliminary developed and some experimental parameters for marker-free rice transformation have been optimized. However, a big gap between the update status in China and the advanced level in some renowned international biotechnology companies is still existed in either transformation efficiency or transformation scale of rice. It was concluded that bio-safety, high efficiency and large scale were the bottlenecks for production GM rice products and its industrialization. Future studies in this field should be focused on the use of commercial rice varieties, improvement of transformation efficiency, development of marker-free rice plants, introduction of multiple target genes, integration of foreign genes at specific site, etc. The review also analyzed the problems existing in large-scale rice transformation system and provided strategies to improve it. It may provide some useful information for efficient production of transgenic rice for practical use as well as for studying gene function.
