Abstract:Drought is among the most severe environmental stressors, imposing detrimental effects on plant growth and development. In this study, we have identified a class III peroxidase gene, OsPrx114, which was induced under PEG (Polyethylene glycol) and drought conditions and found to be localized in both the plasma membrane and endoplasmic reticulum. The promoter region of OsPrx114 encompasses cis-elements, including ABRE (ABA response elements), MBS (MYB binding elements), and W-box, which are likely contributors to drought tolerance. In comparison to Nipponbare (Nip) plants, the overexpression of OsPrx114 enhanced drought tolerance by reducing the accumulation of reactive oxygen species (ROS) through the upregulation of enzyme activities, such as peroxide dismutase (POD) and catalase (CAT). This was accompanied by an increase in proline (Pro) content and a decrease in malondialdehyde (MDA) content. Furthermore, a series of assays including yeast one-hybrid, electrophoretic mobility shift, and dual luciferase assays, demonstrated that the transcription factor OsWRKY50 binds to the W-box (TTGACC) within the promoter of OsPrx114, thereby activating its transcription. OsWRKY50 plays a positive role in regulating OsPrx114-mediated drought resistance by mitigating ROS accumulation in rice. These findings offer a molecular foundation for comprehending the function of the OsWRKY50-OsPrx114 module in response to drought stress in rice.
CRISPR/Cas knockout of the NADPH oxidase gene OsRbohB reduces ROS overaccumulation and enhances heat stress tolerance in rice

Abstract:Summary Heat stress (HS) has become a major factor limiting crop yields worldwide. HS inhibits plant growth by ROS accumulation, and NADPH oxidases (Rbohs) are major ROS producers in plants. Here, we show that CRISPR/Cas knockout of the OsRbohB (OsRbohB‐KO) significantly increased rice tolerance to HS imposed at various different growth stages. We produced OsRbohB‐KO and OsRbohB‐overexpression (OsRbohB‐OE) lines in a japonica cultivar, Nipponbare. Compared with nontransgenic wild‐type (WT) plants, the OsRbohB‐KO lines showed a significant increase in chlorophyll contents (5.2%–58.0%), plant growth (48.2%–65.6%) and grain yield (8.9%–20.5%), while reducing HS‐induced ROS accumulation in seeds (21.3%–33.0%), seedlings (13.0%–30.4%), anthers (13.1%–20.3%) and grains (9.7%–22.1%), under HS conditions. Analysis of yield components revealed that the increased yield of OsRbohB‐KO plants was due to increased starch synthetase activity, spikelets per panicle (2.0%–9.3%), filled spikelets (4.8%–15.5%), percentage of filled spikelets (2.4%–6.8%) and 1000‐grain weight (2.9%–7.4%) under HS conditions during the reproductive stage. Grain milling and appearance quality, and starch content were also significantly increased in OsRbohB‐KO plants under HS conditions during the mature stage. Furthermore, OsRbohB‐KO significantly upregulated the expression levels of heat shock‐related genes, OsHSP23.7, OsHSP17.7, OsHSF7 and OsHsfA2a, in rice seedlings and grains under long‐term HS conditions. Conversely, OsRbohB‐OE resulted in phenotypes that were opposite to OsRbohB‐KO in most cases. Our results suggest that suppression of OsRbohB provides an effective approach for alleviating heat damage and improving grain yield and quality of rice under long‐term HS conditions.

Overexpressing heat-shock protein OsHSP50.2 improves drought tolerance in rice

Abstract:OsHSP50.2, an HSP90 family gene up-regulated by heat and osmotic stress treatments, positively regulates drought stress tolerance probably by modulating ROS homeostasis and osmotic adjustment in rice. Heat-shock proteins (HSPs) serve as molecular chaperones for a variety of client proteins in abiotic stress response and play pivotal roles in protecting plants against stress, but the molecular mechanism remains largely unknown. Here, we report an HSP90 family gene, OsHSP50.2, which acts as a positive regulator in drought stress tolerance in rice (Oryza sativa). OsHSP50.2 was ubiquitously expressed and its transcript level was up-regulated by heat and osmotic stress treatments. Overexpression of OsHSP50.2 in rice reduced water loss and enhanced the transgenic plant tolerance to drought and osmotic stresses. The OsHSP50.2-overexpressing plants exhibited significantly lower levels of electrolyte leakage and malondialdehyde (MDA) and less decrease of chlorophyll than wild-type plants under drought stress. Moreover, the OsHSP50.2-overexpressing plants had significantly higher SOD activity under drought stress compared with the wild type. These results imply that OsHSP50.2 positively regulates drought stress tolerance in rice, probably through the modulation of reactive oxygen species (ROS) homeostasis. Additionally, the OsHSP50.2-overexpressing plants accumulated significantly higher content of proline than the wild type under drought stress, which contributes to the improved protection ability from drought stress damage via osmotic adjustment. Our findings reveal that OsHSP50.2 plays a crucial role in drought stress response, and it may possess high potential usefulness in drought tolerance improvement of rice.
OsASR5 Enhances Drought Tolerance Through a Stomatal Closure Pathway Associated with ABA and H2 O2 Signalling in Rice.

Abstract:Drought is one of the major abiotic stresses that directly implicate plant growth and crop productivity. Although many genes in response to drought stress have been identified, genetic improvement to drought resistance especially in food crops is showing relatively slow progress worldwide. Here, we reported the isolation of abscisic acid, stress and ripening (ASR) genes from upland rice variety, IRAT109 (Oryza sativa L. ssp. japonica), and demonstrated that overexpression of OsASR5 enhanced osmotic tolerance in Escherichia coli and drought tolerance in Arabidopsis and rice by regulating leaf water status under drought stress conditions. Moreover, overexpression of OsASR5 in rice increased endogenous ABA level and showed hypersensitive to exogenous ABA treatment at both germination and postgermination stages. The production of H2 O2 , a second messenger for the induction of stomatal closure in response to ABA, was activated in overexpression plants under drought stress conditions, consequently, increased stomatal closure and decreased stomatal conductance. In contrast, the loss-of-function mutant, osasr5, showed sensitivity to drought stress with lower relative water content under drought stress conditions. Further studies demonstrated that OsASR5 functioned as chaperone-like protein and interacted with stress-related HSP40 and 2OG-Fe (II) oxygenase domain containing proteins in yeast and plants. Taken together, we suggest that OsASR5 plays multiple roles in response to drought stress by regulating ABA biosynthesis, promoting stomatal closure, as well as acting as chaperone-like protein that possibly prevents drought stress-related proteins from inactivation.

Abstract:CODESSA Pro derivative descriptors were calculated for a data set of 426 azeotropic mixtures by the centroid approximation and the weighted-contribution-factor approximation. The two approximations produced almost identical four-descriptor QSPR models relating the structural characteristic of the individual components of azeotropes to the azeotropic boiling points. These models were supported by internal and external validations. The descriptors contributing to the QSPR models are directly related to the three components of the enthalpy (heat) of vaporization.
Overexpression of OsMYB48-1, a Novel MYB-Related Transcription Factor, Enhances Drought and Salinity Tolerance in Rice

Abstract:MYB-type transcription factors (TFs) play essential roles in plant growth, development and respond to environmental stresses. Role of MYB-related TFs of rice in drought stress tolerance is not well documented. Here, we report the isolation and characterization of a novel MYB-related TF, OsMYB48-1, of rice. Expression of OsMYB48-1 was strongly induced by polyethylene glycol (PEG), abscisic acid (ABA), H2O2, and dehydration, while being slightly induced by high salinity and cold treatment. The OsMYB48-1 protein was localized in the nucleus with transactivation activity at the C terminus. Overexpression of OsMYB48-1 in rice significantly improved tolerance to simulated drought and salinity stresses caused by mannitol, PEG, and NaCl, respectively, and drought stress was caused by drying the soil. In contrast to wild type plants, the overexpression lines exhibited reduced rate of water loss, lower malondialdehyde (MDA) content and higher proline content under stress conditions. Moreover, overexpression plants were hypersensitive to ABA at both germination and post-germination stages and accumulated more endogenous ABA under drought stress conditions. Further studies demonstrated that overexpression of OsMYB48-1 could regulate the expression of some ABA biosynthesis genes (OsNCED4, OsNCED5), early signaling genes (OsPP2C68, OSRK1) and late responsive genes (RAB21, OsLEA3, RAB16C and RAB16D) under drought stress conditions. Collectively, these results suggested that OsMYB48-1 functions as a novel MYB-related TF which plays a positive role in drought and salinity tolerance by regulating stress-induced ABA synthesis.

Abstract:<h3 class="Heading">Background</h3>Drought is a major abiotic stress factor that influences the yield of crops. Basic leucine zipper motif (bZIP) transcription factors play an important regulatory role in plant drought stress responses. However, the functions of a number of bZIP transcription factors in rice are still unknown.<h3 class="Heading">Results</h3>In this study, a novel drought stress-related bZIP transcription factor, OsbZIP62, was identified in rice. This gene was selected from a transcriptome analysis of several typical rice varieties with different drought tolerances. OsbZIP62 expression was induced by drought, hydrogen peroxide, and abscisic acid (ABA) treatment. Overexpression of OsbZIP62-VP64 (OsbZIP62V) enhanced the drought tolerance and oxidative stress tolerance of transgenic rice, while osbzip62 mutants exhibited the opposite phenotype. OsbZIP62-GFP was localized to the nucleus, and the N-terminal sequence (amino acids 1–68) was necessary for the transcriptional activation activity of OsbZIP62. RNA-seq analysis showed that the expression of many stress-related genes (e.g., OsGL1, OsNAC10, and DSM2) was upregulated in OsbZIP62V plants. Moreover, OsbZIP62 could bind to the promoters of several putative target genes and could interact with stress/ABA-activated protein kinases (SAPKs).<h3 class="Heading">Conclusions</h3>OsbZIP62 is involved in ABA signalling pathways and positively regulates rice drought tolerance by regulating the expression of genes associated with stress, and this gene could be used for the genetic modification of crops with improved drought tolerance.

Abstract:Drought stress adversely affects crop growth and productivity worldwide. In response, plants have evolved several strategies in which numerous genes are induced to counter stress. High mobility group (HMG) proteins are the second most abundant family of chromosomal proteins. They play a crucial role in gene transcriptional regulation by modulating the chromatin/DNA structure. In this study, we isolated a novel HMG gene, OsHMGB707, one of the candidate genes localized in the quantitative trait loci (QTL) interval of rice drought tolerance, and examined its function on rice stress tolerance. The expression of OsHMGB707 was up-regulated by dehydration and high salt treatment. Its overexpression significantly enhanced drought tolerance in transgenic rice plants, whereas its knockdown through RNA interference (RNAi) did not affect the drought tolerance of the transgenic rice plants. Notably, OsHMGB707-GFP is localized in the cell nucleus, and OsHMGB707 is protein-bound to the synthetic four-way junction DNA. Several genes were up-regulated in OsHMGB707-overexpression (OE) rice lines compared to the wild-type rice varieties. Some of the genes encode stress-related proteins (e.g., DREB transcription factors, heat shock protein 20, and heat shock protein DnaJ). In summary, OsHMGB707 encodes a stress-responsive high mobility group protein and regulates rice drought tolerance by promoting the expression of stress-related genes.

Abstract:Auxin/indoleacetic acid (Aux/IAA) proteins play an important regulatory role in the developmental process of plants and their responses to stresses. A previous study has shown that constitutive expression of OsIAA18 , an Aux/IAA transcription factor gene of rice improved salt and osmotic tolerance in transgenic Arabidopsis plants. However, little work is known about the regulatory functions of the OsIAA18 gene in regulating the abiotic stress tolerance of rice. In this study, the OsIAA18 gene was introduced into the rice cultivar, Zhonghua 11 and the OsIAA18 overexpression in rice plants exhibited significantly enhanced salt and drought tolerance compared to the wild type (WT). Moreover, overexpression of OsIAA18 in rice increased endogenous levels of abscisic acid (ABA) and the overexpression of OsIAA18 in rice plants showed hypersensitivity to exogenous ABA treatment at both the germination and postgermination stages compared to WT. Overexpression of OsIAA18 upregulated the genes involved in ABA biosynthesis and signaling pathways, proline biosynthesis pathway, and reactive oxygen species (ROS)-scavenging system in the overexpression of OsIAA18 in rice plants under salt and drought stresses. Proline content, superoxide dismutase (SOD), and peroxidase (POD) activities were significantly increased, whereas malonaldehyde (MDA), hydrogen peroxide (H 2 O 2 ), and superoxide anion radical (O 2 – ) content were significantly decreased in the transgenic plants under salt and drought stresses. Taken together, we suggest that OsIAA18 plays a positive role in drought and salt tolerance by regulating stress-induced ABA signaling. The OsIAA18 gene has a potential application in genetically modified crops with enhanced tolerance to abiotic stresses.

Abstract:In plants, auxin and ABA play significant roles in conferring tolerance to environmental abiotic stresses. Earlier studies have been shown that some Aux/IAA genes, with important signaling factors in the auxin pathway, were induced in response to drought and other abiotic stresses. However, the mechanistic links between Aux/IAA expression and general drought response remain largely unknown. In this study, OsIAA20, a rice Aux/IAA protein, shown with important roles in abiotic stress. Phenotypic analyses revealed that OsIAA20 RNAi transgenic rice reduced drought and salt tolerance; whereas, OsIAA20 overexpression plants displayed the opposite phenotype. Physiological analyses of OsIAA20 RNAi rice grown under drought or salt stress showed that proline and chlorophyll content significantly decreased, while malondialdehyde content and the ratio of Na+/ K+ significantly increased. In addition, OsIAA20down-regulation reduced stomatal closure and increased the rate of water loss, while transgenic plants overexpressing OsIAA20 exhibited the opposite physiological responses. Furthermore, an ABA-responsive gene, OsRab21, was down-regulated in OsIAA20 RNAi rice lines and upregulated in OsIAA20 overexpression plants. Those results means OsIAA20 played an important role in plant drought and salt stress responses, by an ABA dependent mechanism, and it will be a candidate target gene used to breed abiotic stress tolerance.

Abstract:Calmodulin-like (CML) genes regulate plant growth, development, and responses to abiotic stresses such as salinity and drought. Many genes encoding CML proteins have been identified from rice (Oryza sativa), but their functions remain largely unknown. Our characterization of one putative CML gene, OsDSR-1 (O. sativa Drought Stress Response-1), showed that its protein binds Ca2+ and displays Ca2+-dependent conformational changes. In contrast to wild-type (WT) and OsDSR-1-RNA interference (OsDSR-1-Ri) plants, transgenic rice plants that overexpress OsDSR-1 were significantly more drought tolerant and had increased sensitivity to abscisic acid. Furthermore, their concentrations of free proline and soluble sugars and the activities of reactive oxygen species-scavenging enzymes as well as the transcript levels of many ROS-scavenging and stress-related genes were significantly enhanced under drought stress. Much less hydrogen peroxide and malondialdehyde accumulated in OsDSR-1-overexpressing (OsDSR-1-OE) plants than in either the Ri or WT plants. All of these results suggest that OsDSR-1 plays important roles in conferring tolerance to drought in rice by decreasing the occurrence of oxidative damage.

Abstract:Haloacid dehalogenase-like hydrolase (HAD) superfamily have been shown to get involved in plant growth and abiotic stress response. Although the various functions and regulatory mechanism of HAD superfamily have been well demonstrated, we know little about the function of this family in conferring abiotic stress tolerance to rice. Here, we report OsHAD3 , a HAD superfamily member, could affect drought tolerance of rice. Under drought stress, overexpression of OsHAD3 increases the accumulation of reactive oxygen species and malondialdehyde than wild type. OsHAD3 -overexpression lines decreased but antisense-expression lines increased the roots length under drought stress and the transcription levels of many well-known stress-related genes were also changed in plants with different genotypes. Furthermore, overexpression of OsHAD3 also decreases the oxidative tolerance. Our results suggest that overexpression of OsHAD3 could decrease the drought tolerance of rice and provide a new strategy for improving drought tolerance in rice.

Abstract:Rice (Oryza sativa) responds to various abiotic stresses during growth. Plant-specific NAM, ATAF1/2, and CUC2 (NAC) transcription factors (TFs) play an important role in controlling numerous vital growth and developmental processes. To date, 170 NAC TFs have been reported in rice, but their roles remain largely unknown. Herein, we discovered that the TF OsNAC006 is constitutively expressed in rice, and regulated by H2O2, cold, heat, abscisic acid (ABA), indole-3-acetic acid (IAA), gibberellin (GA), NaCl, and polyethylene glycol (PEG) 6000 treatments. Furthermore, knockout of OsNAC006 using the CRISPR-Cas9 system resulted in drought and heat sensitivity. RNA sequencing (RNA-seq) transcriptome analysis revealed that OsNAC006 regulates the expression of genes mainly involved in response to stimuli, oxidoreductase activity, cofactor binding, and membrane-related pathways. Our findings elucidate the important role of OsNAC006 in drought responses, and provide valuable information for genetic manipulation to enhance stress tolerance in future plant breeding programs.

Abstract:Abiotic stress has been shown to limit the growth, development, and productivity of crops. Here, we characterized the function of a rice bZIP transcription factor OsbZIP16 in drought stress. Expression of OsbZIP16 was dramatically induced under drought conditions. Transient expression and transactivation assays demonstrated that OsbZIP16 was localized in the nucleus and had transactivation activity. At both the seedling and tillering stages, transgenic rice plants overexpressing OsbZIP16 exhibited significantly improved drought resistance, which was positively correlated with the observed expression levels of OsbZIP16. Representative downstream drought-inducible genes were observed to have significantly higher expression levels in transgenic rice plants than in the wild type plants under drought conditions. OsbZIP16 was shown to be induced by exogenous ABA treatment, while overexpression of OsbZIP16 was observed to make transgenic plants more sensitive to ABA than wild type plants were. Transcriptome analysis identified a number of differentially expressed genes between wild type plants and plants overexpressing OsbZIP16, many of which are involved in stress response according to their gene ontologies. Overall, our findings suggest that OsbZIP16 positively regulates drought resistance in rice.
Overexpression of OsVP1 and OsNHX1 Increases Tolerance to Drought and Salinity in Rice

Abstract:Drought and salinity are major abiotic stresses affecting rice production. To improve plant tolerance to salinity and drought, we overexpressed rice Na+/H+ exchangers (OsNHX1) and H+-pyrophosphatase in tonoplasts (OsVP1) in a japonica elite rice cultivar, Zhonghua 11. Compared with our wild-type control, transgenic plants overexpressing both genes incurred less damage when exposed to long-term treatment with 100 mM NaCl or water deprivation. Under high-saline conditions, the transformants accumulated less Na+ and malondialdehyde in the leaves, thereby allowing the plants to maintain a low level of leaf water potential and reduce stress-induced damage. Those transgenics also had higher photosynthetic activity during the stress period. Under those conditions, they also showed an increase in root biomass, which enabled more water uptake. These results suggest that OsVP1 and OsNHX1 improve the tolerance of rice crops against drought and salt by employing multiple strategies in addition to osmotic regulation.

Abstract:Gibberellin 2-oxidase (GA2ox) plays an important role in the GA catabolic pathway and the molecular function of the OsGA2ox genes in plant abiotic stress tolerance remains largely unknown. In this study, we functionally characterized the rice gibberellin 2-oxidase 8 (OsGA2ox8) gene. The OsGA2ox8 protein was localized in the nucleus, cell membrane, and cytoplasm, and was induced in response to various abiotic stresses and phytohormones. The overexpression of OsGA2ox8 significantly enhanced the osmotic stress tolerance of transgenic rice plants by increasing the number of osmotic regulators and antioxidants. OsGA2ox8 was differentially expressed in the shoots and roots to cope with osmotic stress. The plants overexpressing OsGA2ox8 showed reduced lengths of shoots and roots at the seedling stage, but no difference in plant height at the heading stage was observed, which may be due to the interaction of OsGA2ox8 and OsGA20ox1, implying a complex feedback regulation between GA biosynthesis and metabolism in rice. Importantly, OsGA2ox8 was able to indirectly regulate several genes associated with the anthocyanin and flavonoid biosynthetic pathway and the jasmonic acid (JA) and abscisic acid (ABA) biosynthetic pathway, and overexpression of OsGA2ox8 activated JA signal transduction by inhibiting the expression of jasmonate ZIM domain-containing proteins. These results provide a basis for a future understanding of the networks and respective phenotypic effects associated with OsGA2ox8.

Abstract:Plant respiratory burst oxidase homologs (Rbohs) are key enzymes that produce reactive oxygen species (ROS), which serve as signaling molecules regulating plant growth and stress responses. In this study, 39 TaRboh genes (TaRboh01–TaRboh39) were identified. These genes were distributed unevenly among the wheat genome's fourteen chromosomes, with the exception of homoeologous group 2 and 7 and chromosomes 4A, as well as one unidentified linkage group (Un). TaRbohs were classified into ten distinct clades, each sharing similar motif compositions and gene structures. The promoter regions of TaRbohs contained cis-elements related to hormones, growth and development, and stresses. Furthermore, five TaRboh genes (TaRboh26, TaRboh27, TaRboh31, TaRboh32, and TaRboh34) exhibited strong evolutionary conservation. Additionally, a Ka/Ks analysis confirmed that purifying selection was the predominant force driving the evolution of these genes. Expression profiling and qPCR results further indicated differential expression patterns of TaRboh genes between heat and drought stresses. TaRboh11, TaRboh20, TaRboh22, TaRboh24, TaRboh29, and TaRboh34 were significantly upregulated under multiple stress conditions, whereas TaRboh30 was only elevated in response to drought stress. Collectively, our findings provide a systematic analysis of the wheat Rboh gene family and establish a theoretical framework for our future research on the role of Rboh genes in response to heat and drought stress.

Abstract:Heat stress often results in the generation of reactive oxygen species, such as hydrogen peroxide, which plays a vital role as a secondary messenger in the process of abscisic acid (ABA)-mediated stomatal closure. Here, we characterized the rice (Oryza sativa) HEAT TOLERANCE AT SEEDLING STAGE (OsHTAS) gene, which plays a positive role in heat tolerance at the seedling stage. OsHTAS encodes a ubiquitin ligase localized to the nucleus and cytoplasm. OsHTAS expression was detected in all tissues surveyed and peaked in leaf blade, in which the expression was concentrated in mesophyll cells. OsHTAS was responsive to multiple stresses and was strongly induced by exogenous ABA. In yeast two-hybrid assays, OsHTAS interacted with components of the ubiquitin/26S proteasome system and an isoform of rice ascorbate peroxidase. OsHTAS modulated hydrogen peroxide accumulation in shoots, altered the stomatal aperture status of rice leaves, and promoted ABA biosynthesis. The results suggested that the RING finger ubiquitin E3 ligase OsHTAS functions in leaf blade to enhance heat tolerance through modulation of hydrogen peroxide-induced stomatal closure and is involved in both ABA-dependent and DROUGHT AND SALT TOLERANCE-mediated pathways.

Abstract:Various environmental stresses limit the plant growth and productivity. Earlier we reported the stress inducible dehydrin gene, OsDhn1, in rice. In this study we generated OsDhn1-overexpression transgenic rice plants (OsDhn1-OXs) to understand the dehydrin function. OsDhn1-OXs showed an enhanced drought and salt stress tolerance as judged by chlorophyll fluorescence (Fv/Fm), fresh and dry weight, water and chlorophyll content, and survival ratio. Furthermore, OsDhn1-OXs showed significantly increased tolerance to methyl viologen (MV)-induced oxidative stress. Under salt and drought stress condition, OsDhn1-OXs maintained relatively low level of H2O2 as compared to wild type plants. Taken together, it is suggested that OsDhn1 plays an important role in the stress tolerance via scavenging reactive oxygen species (ROS).
