Abstract:Poly(vinylidene fluoride) (PVDF) membrane was pre-irradiated by electron beam, and then poly(ethylene glycol) methyl ether methacrylate (PEGMA) was grafted onto the membrane surface in the aqueous solution. The degree of grafting was significantly influenced by the pH value of the reaction solution. The surface chemical changes were characterized by the Fourier transform infrared attenuated total reflection spectroscopy (FTIR-ATR) and X-ray photoelectron spectroscopy (XPS). Combining with the analysis of the nuclear magnetic resonance proton and carbon spectra (1H NMR and 13C NMR), PEGMA was mainly grafted onto the membrane surface. Morphological changes were characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM). The porosity and bulk mean pore size changes were determined by a mercury porosimeter. The surface and bulk hydrophilicity were evaluated on the basis of static water contact angle, dynamic water contact angle and the dynamic adsorption process. Furthermore, relative high permeation fluxes of pure water and protein solution were obtained. All these results demonstrate that both hydrophilicity and fouling resistance of the PVDF membrane can be improved by the immobilization of hydrophilic comb-like polymer brushes on the membrane surface.
PEG-functionalized aliphatic polycarbonate brushes with self-polishing dynamic antifouling properties

Abstract:Hydrophilic antifouling polymers provide excellent antifouling effects under usual short-term use conditions, but the long-term accumulation of contaminants causes them to lose their antifouling properties. To overcome this drawback, surface-initiated ring-opening graft polymerization (SI-ROP) was performed on the surface of the material by applying the cyclic carbide monomer 4'-(fluorosulfonyl)benzyl-5-methyl-2-oxo-1,3-dioxane-5-carboxylate (FMC), which contains a sulfonylfluoride group on the side chain, followed by a "sulfur(IV)-fluorine exchange" (SuFEx) post click modification reaction to link the hydrophilic polyethylene glycol (PEG) to the polyFMC (PFMC) brush, and a novel antifouling strategy for self-polishing dynamic antifouling surfaces was developed. The experimental results showed that the antifouling surface could effectively prevent the adsorption of proteins such as bovine serum albumin (BSA, ∼96.4%), fibrinogen (Fg, ∼87.8%) and lysozyme (Lyz ∼69.4%) as well as the adhesion of microorganisms such as the bacteria Staphylococcus aureus (S. aureus) (∼87.5%) and HeLa cells (∼67.2%). Moreover, the enzymatically self-polished surface still has excellent antifouling properties. Therefore, this modification method has potential applications in the field of biosensors and novel antifouling materials.

Abstract:Brush copolymers with poly(ethylene glycol) (PEG) as side chains have received wide attentions due to their unique properties and potential applications. In this article,the synthesis strategies,including“grafting through”and“grafting onto”,their specific advantages and limitations are reviewed,with an emphasis on brush PEG copolymers based on biodegradable materials,such as polyesters. In each strategy,various polymerization techniques,such as ionic polymerization,radical polymerization,ring opening polymerization and their compatible partner various coupling reactions, are employed. Meanwhile,the surface morphologies of the micelles built from linear PEG copolymers and brush PEG copolymers are illustrated in this article,and the effects of the architecture of PEG brush on the formed nanoparticles’ properties in drug delivery are briefly discussed. Well-defined brush PEG structure can prolong circulation times,resulting in improvement in drug efficacy. However,the effective synthetic methods as well as the micelles’ in vivo and in vitro properties still need ongoing and further researches.
Reducing Non-Specific Binding and Uptake of Nanoparticles and Improving Cell Targeting with an Antifouling PEO-b-PγMPS Copolymer Coating

Abstract:One of the major limitations impeding the sensitivity and specificity of biomarker targeted nanoparticles is non-specific binding by biomolecules and uptake by the reticuloendothelial system (RES). We report the development of an antibiofouling polysiloxane containing amphiphilic diblock copolymer, poly(ethylene oxide)-block-poly(γ-methacryloxypropyl trimethoxysilane) (PEO-b-PγMPS), for coating and functionalizing high quality hydrophobic nanocrystals such as iron oxide nanoparticles and quantum dots. These PEO-b-PγMPS-coated nanocrystals were colloidally stable in biological medium and showed low non-specific binding by macromolecules after incubation with 100% fetal bovine serum. Both in vitro experiments with macrophages and in vivo biodistribution studies in mice revealed that PEO-b-PγMPS copolymer-coated nanocrystals have an antibiofouling effect that reduces non-specific cell and RES uptake. Surface functionalization with amine groups was accomplished through co-crosslinking the polysiloxane coating layer and (3-Aminopropyl)trimethoxysilane in aqueous solution. Tumor integrin αvβ3 targeting peptide cyclo-RGD ligands were conjugated on the nanoparticles through a heterobifunctional linker. The resulting integrin αvβ3 targeting nanoparticle conjugates showed improved cancer cell targeting with a stronger affinity to U87MG glioma cells, which have a high expression of αvβ3 integrins, but minimal binding to MCF-7 breast cancer cells with low expression of αvβ3 integrins.
Conducting Polymer-Infused Electrospun Fibre Mat Modified by POEGMA Brushes as Antifouling Biointerface

Abstract:Biofouling on surfaces, caused by the assimilation of proteins, peptides, lipids and microorganisms, leads to contamination, deterioration and failure of biomedical devices and causes implants rejection. To address these issues, various antifouling strategies have been extensively studied, including polyethylene glycol-based polymer brushes. Conducting polymers-based biointerfaces have emerged as advanced surfaces for interfacing biological tissues and organs with electronics. Antifouling of such biointerfaces is a challenge. In this study, we fabricated electrospun fibre mats from sulphonated polystyrene-block-poly(ethylene-ran-butylene)-block-polystyrene (sSEBS), infused with conducting polymer poly(3,4-ethylenedioxythiophene) (PEDOT) (sSEBS-PEDOT), to produce a conductive (2.06 ± 0.1 S/cm), highly porous, fibre mat that can be used as a biointerface in bioelectronic applications. To afford antifouling, here the poly(oligo (ethylene glycol) methyl ether methacrylate) (POEGMA) brushes were grafted onto the sSEBS-PEDOT conducting fibre mats via surface-initiated atom transfer radical polymerization technique (SI-ATRP). For that, a copolymer of EDOT and an EDOT derivative with SI-ATRP initiating sites, 3,4-ethylenedioxythiophene) methyl 2-bromopropanoate (EDOTBr), was firstly electropolymerized on the sSEBS-PEDOT fibre mat to provide sSEBS-PEDOT/P(EDOT-co-EDOTBr). The POEGMA brushes were grafted from the sSEBS-PEDOT/P(EDOT-co-EDOTBr) and the polymerization kinetics confirmed the successful growth of the brushes. Fibre mats with 10-mers and 30-mers POEGMA brushes were studied for antifouling using a BCA protein assay. The mats with 30-mers grafted brushes exhibited excellent antifouling efficiency, ~82% of proteins repelled, compared to the pristine sSEBS-PEDOT fibre mat. The grafted fibre mats exhibited cell viability >80%, comparable to the standard cell culture plate controls. Such conducting, porous biointerfaces with POEGMA grafted brushes are suitable for applications in various biomedical devices, including biosensors, liquid biopsy, wound healing substrates and drug delivery systems.

Abstract:Development of novel materials with dual antibacterial and antifouling functions is important for many bio-applications. In this work, we design and synthesize a series of mixed polymer brushes grafted from silica surface as a new type of antibiofouling and antimicrobial coating, which combine poly(N-hydroxyethyl acrylamide) (polyHEAA) synthesized by atom transfer radical polymerization (SI-ATRP) and cationic poly(trimethylamino) ethyl methacrylate chloride, polyMETAC) synthesized by photoiniferter-mediated polymerization (SI-PIMP). The surface composition, charge and morphology of the resultant mixed polymer brushes are carefully characterized and optimized by tuning ATRP and PIMP initiator ratios. The mixed polymer brushes possess strong bacterial killing ability from polyMETAC chains and high antifouling property from polyHEAA chains. At the optimal conditions, the mixed polyHEAA/polyMETAC brushes exhibit excellent anti-bacterial adhesion property by retaining ultralow cell adhesion of (similar to)3.0 x 105 cells/cm2 and high contact killing efficiency by killing more than 90% attached bacteria of Escherichia coil and Staphylococcus. Long-term antibacterial performance indicated the cell adhesion was retained at ultralow level of 5 x 105 cells/cm2 even after 72 h exposure to bacteria solution. The results indicate that surface grafting with polyHEAA and polyMETAC can be potentially useful for long-term biomedical and biomaterial applications.

Abstract:Polyphosphoester (PPE) brush coatings are developed as anti‐biofouling and degradable surfaces, addressing the critical issue of non‐biodegradability in synthetic brush coatings. Utilizing surface‐initiated organocatalytic ring‐opening polymerization, PPE brushes demonstrate controlled degradability and an enhanced resistance to protein adsorption and bacterial adhesion. This performance makes them promising materials for anti‐biofouling applications in medical devices and artificial implants. The implementation of polymer brush coatings has proven crucial in biomedical and biotechnological applications for controlling biological interactions. Despite the extensive research and use of synthetic polymer coatings, concerns regarding their long‐term non‐biodegradability have arisen. This issue is significant as non‐degradable polymers can lead to complications such as inflammatory responses and bioaccumulation, highlighting the need for polymers that degrade in a controlled manner. To address this, polyphosphonate brushes are synthesized as anti‐biofouling coatings on silicon surfaces via controlled surface‐initiated organocatalytic ring‐opening polymerization (SI‐ROP) of cyclic phospholanes. 2‐Ethyl‐2‐oxo‐1,3,2‐dioxaphospholane and 2‐hexyl‐2‐oxo‐1,3,2‐dioxaphospholane are chosen as the monomers to prepare hydrophilic and hydrophobic brushes with thicknesses up to 55 nm, with a proven degradability in aqueous media. In addition, protein adsorption, bacterial adhesion, and biofilm formation are investigated as a function of the polyphosphonate side chain. Compared to non‐coated surfaces and polyester brushes, hydrophilic poly(2‐ethyl‐2‐oxo‐1,3,2‐dioxaphospholane) brushes have an enhanced resistance toward fouling of albumin, fibrinogen, and diluted human serum proteins, as well as toward Escherichia coli and Staphylococcus aureus bacteria. The stability and anti‐biofouling performance of hydrophilic polyphosphonate brushes position them as an attractive option as degradable materials for anti‐biofouling matrices on medical devices and/or lubricious coatings for artificial implant technologies.

Fabrication of Transparent PEGylated Antifouling Coatings via One-Step Pyrogallol Deposition

Abstract:Antifouling coatings are critical for many biomedical devices. A simple and universal technique used to anchor antifouling polymers is important in order to expand its applications. In this study, we introduced the pyrogallol (PG)-assisted immobilization of poly(ethylene glycol) (PEG) to deposit a thin antifouling layer on biomaterials. Briefly, biomaterials were soaked in a PG/PEG solution and PEG was immobilized onto the biomaterial surfaces via PG polymerization and deposition. The kinetics of PG/PEG deposition started with the deposition of PG on the substrates, followed by the addition of a PEG-rich adlayer. However, prolonged coating added a top-most PG-rich layer, which deteriorated the antifouling efficacy. By controlling the amounts of PG and PEG and the coating time, the PG/PEG coating was able to reduce more than 99% of the adhesion of L929 cells and the adsorption of fibrinogen. The ultrathin (tens of nanometers) and smooth PG/PEG coating was easily deposited onto a wide variety of biomaterials, and the deposition was robust enough to survive harsh sterilization conditions. Furthermore, the coating was highly transparent and allowed most of the UV and Vis light to pass through. The technique has great potential to be applied to biomedical devices that need a transparent antifouling coating, such as intraocular lenses and biosensors.
Understanding the Oxidative Stability of Antifouling Polymer Brushes.

Abstract:Poly(oligoethylene glycol methacrylate) (POEGMA) and zwitterionic polymer brushes have been widely used for constructing biocompatible or antifouling surfaces, and their oxidative stability is very important to the practical application. Herein, POEGMA, poly(sulfobetaine methacrylate) (PSBMA), poly(2-(methacryloyloxy)ethyl phosphorylcholine) (PMPC), and poly(carboxybetaine methacrylate) (PCBMA) were grafted on quartz crystal microbalance (QCM) chips via surface-initiated atom transfer radical polymerization (SI-ATRP). XPS and MS analyses demonstrate that the mass loss of these polymer brushes in oxidative environment is due to the scission of the polymer-anchoring segments. Molecular simulation further illustrates this mass loss mechanism should be always true for those polymer brushes anchored on different substrates. In situ QCM monitoring indicates that, compared with zwitterionic polymethacrylates, POEGMA brushes show the lowest mass loss rate mainly due to their cross-linked structures. This study sheds light on the contradictory reports about the oxidative stability of POEGMA and zwitterionic polymethacrylate brushes up to now, and highlights the important role of the polymer-anchoring segments playing in the oxidative stability of polymer brushes.
A green, versatile, and facile strategy for anti-biofouling surface with ultra-high graft density polyethylene glycol

Abstract:Implantable catheters are susceptible to severe complications due to non-specific protein adhesion on their surfaces. Polyethylene glycol (PEG) coatings, the gold standard for resistance to non-specific protein adhesion, present a challenge in achieving high-density grafting, which significantly restricts their use as anti-biofouling coatings. Herein, we exploited the strong interaction between polyphenols (PCs) and polycations (K6-PEG) to graft PEG onto the surface of PC-Cu (A network of metal polyphenols composed of proanthocyanidins and metal copper ions, with expectation for the coating with excellent resistance to non-specific protein adhesion (PC-Cu@K6-PEG). The introduction of K6-PEG resulted in enhanced stability and modulus of PC-Cu, as well as a reduction in the surface adhesion energy and contact angle of PC-Cu. In contrast to previously reported PEG coatings, PC-Cu@K6-PEG exhibited a markedly elevated grafting density of PEG (4.06 chains/nm2), which was more than double the highest value previously reported (1.9 chains/nm2), due to the diffusing ability of K6-PEG throughout the PC-Cu networks. PC-Cu@K6-PEG displays robust resistance to a variety of proteins, microbials, and platelet attachment, thereby preventing thrombosis. The coating ability of PC-Cu onto diverse substrates, combined with the simple, straightforward and environmentally benign process of fabricating PC-Cu@K6-PEG, suggests that this strategy has significant potential for use in anti-biofouling surfaces.

Abstract:Poly(ethylene glycol) (PEG), a linear polymer known for its "stealth" properties, is commonly used to passivate the surface of biomedical implants and devices, and it is conjugated to biologic drugs to improve their pharmacokinetics. However, its antigenicity is a growing concern. Here, the antigenicity of PEG is investigated when assembled in a poly(oligoethylene glycol) methacrylate (POEGMA) "bottlebrush" configuration on a planar surface. Using ethylene glycol (EG) repeat lengths of the POEGMA sidechains as a tunable parameter for optimization, POEGMA brushes with sidechain lengths of two and three EG repeats are identified as the optimal polymer architecture to minimize binding of anti‐PEG antibodies (APAs), while retaining resistance to nonspecific binding by bovine serum albumin and cultured cells. Binding of backbone‐ versus endgroup‐selective APAs to POEGMA brushes is further investigated, and finally the antigenicity of POEGMA coatings is assessed against APA‐positive clinical plasma samples. These results are applied toward fabricating immunoassays on POEGMA surfaces with minimal reactivity toward APAs while retaining a low limit‐of‐detection for the analyte. Taken together, these results offer useful design concepts to reduce the antigenicity of polymer brush‐based surface coatings used in applications involving human or animal matrices.

Abstract:The influence of chain length of a fluorinated side chain of an asymmetric polymer brush with poly(ethylene glycol) (PEG) side chains on the antifouling property was systematically investigated so as to obtain more knowledge for deepening our understanding of the structure-(antifouling)-property relationship of asymmetric polymer brush. A series of asymmetric polymer brushes, consisting of hydrophobic poly(pentafluoropropyl methacrylate) (PPTFMA) side chains with the number of repeat units of pentafluoropropyl methacrylate (PTFMA) ranging from 8 to 42 and hydrophilic PEG side chains, was first synthesized by sequential reversible addition-fragmentation chain transfer (RAFT) polymerization and atom transfer radical polymerization (ATRP). Subsequently, thin films were prepared by spin-casting the solution of these brushes onto indium tin oxide (ITO) and SiO2 substrates. Water contact angle analysis showed that the hydrophobicity of the film surface increased with the length of PPTFMA side chain. The. atomic force microscopy (AFM) measurement demonstrated that these films had a roughness of <3 and <10 nm onto ITO glass and SiO2 substrates, respectively. The antifouling behaviors of these films in bovine serum albumin (BSA) solution were evaluated by quartz crystal microbalance (QCM), which showed that the asymmetric brush surfaces had considerable antifouling performance with less protein adsorption in comparison with the bare surface. In addition, the films made from polymer brushes with shorter PPTFMA side chains exhibited better antifouling and fouling-release behaviors.

Abstract:PURPOSE Upon irradiation the active diacetylene layer of radiochromic films undergoes dose-dependent polymerization in the blue polydiacetylene form. Dose assessment is currently based on linear absorbance measurements. The scope of the present study was designed for the further understanding of the linear and, in particular, the nonlinear optical characteristics of such films and the utilization of these characteristics for devising alternative "reading" procedures. METHODS The nonlinear optical interactions of laser light with previously irradiated GafChromic(®) HD-810 and MD55-2 films were studied by z-scan techniques. The focused laser beam strikes on the film, which is located at various distances from the focal spot. The beam induces linear and nonlinear absorption and refraction. The measurement of the transmitted power (open-aperture z-scan) allows the study of the nonlinear absorption. On the other hand, the measurement of the axial region of the transmitted beam (closed-aperture z-scan) is related to both nonlinear absorption and refraction effects. A 10 mW, 633 nm, linearly polarized He/Ne laser was employed as the light source. The transmittance measurements were coupled with absorbance measurements carried out over the whole visible spectrum and at various reading temperatures, in an attempt to clarify the underlying mechanisms dictating the observed effects. These effects were incorporated to an open-aperture z-scan model, developed for the purpose of comparison to the corresponding experimental curves. RESULTS The transmittance data obtained by both open- and closed-aperture z-scan were found to be dose-dependent, thus allowing such transmittance techniques to be employed for dose assessment. Low power open-aperture z-scan measurements (sensitive to absorptive nonlinearities) revealed a stepwise two-photon excitation of the active layer, through an intermediate state. At higher laser intensities (and/or absorbed dose), the shapes of the z-scan curves were found to be more complicated. These shapes were attributed to a temperature-dependent (thermochromic) transition from the blue form to the conformer red form of the active material, a transition which appears to be irreversible at high intensities. This transition was incorporated into the open-aperture z-scan model which reproduced satisfactorily the corresponding experimental curves. Finally, the use of a chopped laser beam in the closed-aperture z-scan technique allowed artifact-free, high spatial resolution dose measurements using a laser beam of relatively high power. CONCLUSIONS The model developed in the present study elucidates some of the effects related to film reading and the causes of some of the artifacts reported in the literature. Two-dimensional dose distributions can be assessed by z-scan; however, the merits of such a technique relative to those currently in use have to be further investigated.
Molecular simulations and understanding of antifouling zwitterionic polymer brushes.

In this work, we computationally studied the packing structure, surface hydration, and antifouling property of three zwitterionic polymer brushes of poly(carboxybetaine methacrylate) (pCBMA), poly(sulfobetaine methacrylate) (pSBMA), and poly((2-(methacryloyloxy)ethyl)phosporylcoline) (pMPC) brushes and a hydrophilic PEG brush using a combination of molecular mechanics (MM), Monte Carlo (MC), molecular dynamics (MD), and steered MD (SMD) simulations. We for the first time determined the optimal packing structures of all polymer brushes from a wide variety of unit cells and chain orientations in a complex energy landscape. Under the optimal packing structures, MD simulations were further conducted to study the structure, dynamics, and orientation of water molecules and protein adsorption on the four polymer brushes, while SMD simulations to study the surface resistance of the polymer brushes to a protein. The collective results consistently revealed that the three zwitterionic brushes exhibited stronger interactions with water molecules and higher surface resistance to a protein than the PEG brush. It was concluded that both the carbon space length between zwitterionic groups and the nature of the anionic groups have a distinct effect on the antifouling performance, leading to the following antifouling ranking of pCBMA > pMPC > pSBMA. This work hopefully provides some structural insights into the design of new antifouling materials beyond traditional PEG-based antifouling materials.
Bioinspired Dopamine and N-Oxide-Based Zwitterionic Polymer Brushes for Fouling Resistance Surfaces

Abstract:Biofouling is a great challenge for engineering material in medical-, marine-, and pharmaceutical-related applications. In this study, a novel trimethylamine N-oxide (TMAO)–analog monomer, 3-(2-methylacrylamido)-N,N-dimethylpropylamine N-oxide (MADMPAO), was synthesized and applied for the grafting of poly(MADMPAO) (pMPAO) brushes on quartz crystal microbalance (QCM) chips by the combination of bio-inspired poly-dopamine (pDA) and surface-initiated atom transfer radical polymerization technology. The result of ion adsorption exhibited that a sequential pDA and pMPAO arrangement from the chip surface had different characteristics from a simple pDA layer. Ion adsorption on pMPAO-grafted chips was greatly inhibited at low salt concentrations of 1 and 10 mmol/L due to strong surface hydration in the presence of charged N+ and O− of zwitterionic pMPAO brushes on the outer layer on the chip surface, well known as the "anti-polyelectrolyte" effect. During BSA adsorption, pMPAO grafting also led to a marked decrease in frequency shift, indicating great inhibition of protein adsorption. It was attributed to weaker BSA-pMPAO interaction. In this study, the Au@pDA-4-pMPAO chip with the highest coating concentration of DA kept stable dissipation in BSA adsorption, signifying that the chip had a good antifouling property. The research provided a novel monomer for zwitterionic polymer and demonstrated the potential of pMPAO brushes in the development and modification of antifouling materials.

Abstract:Polyurethane (PU) is a biopolymer that has been commonly used for biomedical applications. However, the biofouling phenomenon on the hydrophobic PU surface is one of the crucial issues that embarrassing its applications. Here, we report a facile & efficient approach to improve the anti-biofouling ability of the PU substrates. Active residues were firstly generated on the PU surface by using the low temperature air-plasma treatment, promoting the immobilization of the atom transfer radical polymerization (ATRP) initiators on the surface. Then, three types of zwitterionic polymer brushes, as well as PEG brushes, have been fabricated on the PU substrates through surface-initiated ATRP (SI-ATRP). Robust surface characterizations that capable of revealing the surface chemistry (including X-ray photoelectron spectroscopy (XPS) and wettability tests), and antifouling evaluations of the PU substrates (protein adsorption, platelet adhesion, and cell adhesion measurements) were performed. Results showed that three types of zwitterionic brushes have been successful grafted on the PU surface, respectively. And the three types of zwitterionic brushes, in general, significantly inhibited the protein adsorption, the platelet adhesion, and the cell adhesion on the PU surface, endowing a significantly improved anti-fouling ability to the PU substrates. Furthermore, we found that this facial zwitterionic surface modification did not compromise the mechanical property of the PU substrates. This strategy could be easily exploited to PU-based biomaterials to improve their performance in many applications.
Synergistically Improved Antifouling Efficiency of a Bioinspired Self-renewing Interface via a Borneol/ Boron Acrylate Polymer

Abstract:Underwater facilities are often perplexed by severe and ubiquitous biofouling. The widely applied commercial antifouling materials still have several challenges in static applications. Herein, a polymer containing isoborneol and borane (PBABs), the borneol derivative structure and grafted pyridine-triphenylborane (PTPB) as antifouling groups were prepared by radical polymerization. PBABs showed high antibacterial rates for Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) of up to 95.1% and 81.1%, respectively, confirming superior antibacterial adhesion propertys. More importantly, PBABs effectively reduced the expression of mussel adhesion protein, indicating superior antifouling propertys, resulting from the synergistic effect of multiple antifouling functional groups on the material's surface. Therefore, the PBABs have been evaluated as noncytotoxic, low-cost, easily synthesized, and mass-produced, which demonstrates their great potential for actual marine applications.

Abstract:In this work, we combined 3D printing technology and subsurface-initiated ring opening metathesis polymerization (ROMP) to construct a multi-scale brush-based antifouling surface. A biomimetic sharklet structured substrate embedded with ROMP initiator was prepared by 3D printing with commercial acrylic resin. Then, the embedded poly(ionic liquid) brushes were grafted onto the as-prepared biomimetic sharklet via subsurface-initiated ROMP of a novel ionic liquid monomer containing benzotriazole and imidazole groups ([BNIm][Br]). A series of characterization including infrared spectroscopy, X-ray photoelectron spectroscopy and atomic force microscope proved the successful grafting of poly([BNIm][Br]) brushes on the as-prepared surface. X-ray photoelectron spectroscopy etching and friction tests demonstrated that poly([BNIm][Br]) can be grafted not only on the surface but on the subsurface of the as-prepared surface, and the as-prepared poly(ionic liquid) brush-based surfaces showed satisfactory wear resistance compared to traditional surface initiated ROMP. Subsequently, we evaluated the anti-biofouling properties of poly([BNIm][Br]) brushes. The results indicated that poly(ionic liquid) brushes can obviously resist adhesion of bovine serum albumin and have good anti-bacterial activity against both E. coli and S. aureus. The as-prepared poly(ionic liquid) brush-based biomimetic surface also exhibited considerable antifouling performance for microalgae (Porphyridium and Dunaliella) due to the synergistic effect of the surface composition and microstructure.

Abstract:Zwitterionic polymers are suitable for replacing poly(ethylene glycol) (PEG) polymers because of their better antifouling properties, but zwitterionic polymers have poor mechanical properties, strong water absorption, and their homopolymers should not be used directly. To solve these problems, a reversible-addition fragmentation chain transfer (RAFT) polymerization process was used to prepare copolymers comprised of zwitterionic side chains that were attached to an ITO glass substrate using spin-casting. The presence of 4-vinylpyridine (4VP) and zwitterion chains on these polymer-coated ITO surfaces was confirmed using 1H NMR, FTIR, and GPC analyses, with successful surface functionalization confirmed using water contact angle, X-ray photoelectron spectroscopy (XPS), and atomic force microscopy (AFM) studies. Changes in water contact angles and C/O ratios (XPS) analysis demonstrated that the functionalization of these polymers with beta-propiolactone resulted in hydrophilic mixed 4VP/zwitterionic polymers. Protein adsorption and cell attachment assays were used to optimize the ratio of the zwitterionic component to maximize the antifouling properties of the polymer brush surface. This work demonstrated that the antifouling surface coatings could be readily prepared using a P4VP-modified method, that is, the functionality of P4VP to modify the prepared zwitterionic polymer. We believe these materials are likely to be useful for the preparation of biomaterials for biosensing and diagnostic applications.
