Abstract:We aimed to investigate the effects of Clostridium butyricum-, Bacillus subtilis-, and Bacillus licheniformis-based potential probiotics on the growth performance, intestinal morphology, immune responses, and caecal short chain fatty acids (SCFAs) and microbial structure in broiler chickens. Three treatment groups containing a total of 1200 one-day-old AA broilers were included: birds fed with a basal diet only (Con), birds fed with added 10(10) probiotics cfu/kg (ProL), and birds fed with added 10(11) probiotics cfu/kg (ProH). The dietary probiotics significantly improved the final and average body weights and serum immunoglobulins A, M, and Y. The probiotics also enhanced the ileal morphology and improved the caecal acetate, butyrate, and propionate contents. Furthermore, 16S rRNA sequencing revealed that dietary compound probiotics modulated the caecal microflora composition as follows: (1) all birds shared 2794 observed taxonomic units; (2) treatment groups were well separated in the PCA and PCoA analysis; (3) the relative abundance of Parabacteroides, Ruminococcaceae_UCG-014, Barnesiella, Odoribacter, [Eubacterium_coprostanoligenes_group], [Ruminococcus]_torques_group, and Butyricimonas significantly varied between treatments. The compound probiotics improved the growth performance, serum immune responses, the ratio of ileal villus height to crypt depth, and major caecal SCFAs in broiler chickens. The dietary C. butyricum-, B. subtilis-, and B. licheniformis-based probiotics improved overall broiler health and would benefit the poultry industry.

Abstract:Background Different types of exogenous protease supplements have a positive impact on animal performance, but their effects on the nutritional value of meat and the gut microbial community of broilers have not been extensively studied. The objective of this investigation was to determine the impact of supplementation with a novel alkaline protease derived from Bacillus licheniformis (at doses of 0, 100, 200, 300, and 400 g/t) on the fatty acid and amino acid profiles, inosine monophosphate (IMP) levels, total volatile basic nitrogen (TVB-N) content found within the breast muscle, as well as the impact on the cecal microbiota and metabolites. Results Supplementation with 200–400 g/t of the novel protease resulted in a significant elevation in the concentration of essential amino acids ( P < 0.001), flavor amino acids ( P < 0.001), and total protein ( P = 0.013) within the breast muscle. Results derived from the 16S rRNA sequencing and untargeted metabolomics analysis of the cecal content revealed that the novel protease reshaped the cecal microbial and metabolite profiles. In particular, it led to increased relative abundances of Bacteroides , Lactobacillus , Alistipes , and Eubacterium , while simultaneously causing a reduction in the metabolites of D-lactic acid and malonic acid. Moreover, correlation analyses unveiled significant relationships between distinct microbes and metabolites with the contents of IMP, fatty acids, and amino acids in the broiler's breast muscle. Conclusion In summary, the novel protease regulated the intestinal microbial community and metabolism, thereby inducing changes in the compositions of fatty acids and amino acids profiles, as well as IMP levels in broiler meat. These alterations significantly contributed to the enhancement of the nutritional value and flavor of the meat. Graphical abstract

Abstract:BACKGROUND: The present study was conducted to investigate the effects of dietary novel alkaline protease from Bacillus licheniformis on the growth performance, meat quality, antioxidant status and intestinal morphology of broilers. In total, 4000 broilers were randomly assigned into five groups and treated with normal control, normal control + 100 mg kg(-1) protease, normal control + 200 mg kg(-1) protease, normal control + 300 mg kg(-1) protease and normal control + 400 mg kg(-1) protease. RESULTS: Supplementing protease impacted final body weight (linear, P = 0.003; quadratic, P = 0.006) and decreased feed conversion rate (linear, P = 0.036) in broilers. Moreover, dietary protease significantly increased breast muscle rate (linear, P = 0.005; quadratic, P = 0.021) and decreased drip loss (linear, P < 0.001; quadratic, P < 0.001). In addition, dietary protease notably increased protein digestibility (linear, P = 0.001; quadratic, P = 0.006) and trypsin activity (linear, P = 0.002; quadratic, P = 0.009) in jejunum. Light microscopy revealed that the jejunum villi in the 300 mg kg(-1) and 400 mg kg(-1) groups exhibited greater height and a denser arrangement compared to those in the control group. The addition of protease decreased malondialdehyde content (linear, P < 0.001; quadratic, P < 0.001) and increased total antioxidant capacity (linear, P = 0.001; quadratic, P < 0.001) in pectoral muscles. CONCLUSION: The results of the present study suggest that dietary novel alkaline protease from B. licheniformis improved growth performance by affecting trypsin activity, protein digestibility, antioxidant capacity and intestinal health. Abstract:This study aimed to investigate the supplemental effects of probiotic Bacillus subtilis fmbJ (BS fmbJ) on growth performance, antioxidant capacity, and meat quality of broiler chickens. A total of 240 day-old male Arbor Acres (AA) broiler chickens were randomly allotted to 4 treatments and raised for 6 wk. Each treatment had 6 replicate pens with ten birds per replicate. Birds in the control group (CON) were fed diets without BS fmbJ and antibiotics. The BS groups were fed the basal diets with BS fmbJ at 2 × 1010 cfu/kg (BS-1 group), BS fmbJ at 3 × 1010 cfu/kg (BS-2 group), BS fmbJ at 4 × 1010 cfu/kg (BS-3 group) without antibiotics for 42 d. In the study, dietary supplementation with BS fmbJ significantly improved (P < 0.05) the average daily gain (ADG), average daily feed intake (ADFI), and feed conversion ratio (FCR) of broilers from 21 to 42 d and 1 to 42 d. At 42 d, the final body weight was increased (P < 0.05) in BS-2 group compared with that in CON. Dietary BS fmbJ significantly increased (P < 0.05) serum IgA and IgG concentrations of broilers after 42 days raising. The glutathione (GSH), glutathione reductase (GR), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) activity of serum and liver were increased (P < 0.05), and methane dicarboxylic aldehyde (MDA) contents in serum and liver were decreased (P < 0.05) by BS fmbJ added into the broiler diets. Dietary supplementation with BS fmbJ significantly decreased (P < 0.05) reactive oxygen species (ROS) contents in liver mitochondria of broilers. Additionally, the expression of antioxidant enzyme gene including nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase 1 (HO-1), superoxide dismutase (SOD), and glutathione peroxidase (GPx) were improved (P < 0.05) by BS fmbJ added into the broiler diets. Among measuring items of chicken breast meat quality, the drip loss, cooking loss, shear force, L*24 h, a*24 h, b*45 min, and b*24 h values were influenced (P < 0.05) by BS fmbJ provided in the diet. Based on these results, Bacillus subtilis fmbJ could be acted as a beneficial feed additive with antioxidant capacity in broiler diets.

Abstract:This paper aimed to study the dietary effects of Bacillus subtilis fmbj (BS fmbj) on growth performance, small intestinal histomorphology, and its antioxidant capacity of broilers at 21 d of raising. A total of 300 1-d old male Arbor Acres broilers were randomly assigned to 5 groups: broilers fed the basal diets with 0 g/kg BS fmbj (CON), 0.2 g/kg BS fmbj (BS-1), 0.3 g/kg BS fmbj (BS-2), 0.4 g/kg BS fmbj (BS-3), and 0.5 g/kg BS fmbj (BS-4). The results showed that there were no differences in the growth performance among treatments during the trail. Dietary BS fmbj in broiler diets increased (P < 0.05) the serum immunoglobulin A (IgA) and immunoglobulin G (IgG) concentration, and enhanced the secretory immunoglobulin A (sIgA) level of small intestine (jejunum and ileum) compared with those in the CON group. The BS groups could improve (P < 0.05) the values of villus length, villus width, crypt depth, and villus area of small intestine compared with that in the CON group. Compared with the CON group, the BS group increased (P < 0.05) small intestinal antioxidant capacity and its mitochondrial antioxidant capacity, and also improved the antioxidant related-gene expression. The BS group exerted a lower (P < 0.05) level of oxidative damages in small intestine than that of the CON group. In conclusion, dietary BS fmbj in broiler diets was potential to improve the small intestinal histomorphology, small intestinal antioxidant capacity, and its mitochondrial antioxidant capacity. Thus this BS fmbj might be considered to be an important additive for the poultry industry.

Abstract:This study was designed to evaluate the usage of Bacillus subtilis fmbj (BS fmbj) in broiler diets, and its effects on the antioxidant capacity of broilers at an early age. A total of 240 day-old male Arbor Acres (AA) broilers were randomly assigned to 4 groups, namely the control (CON) group (fed basal diets with 0 cfu/kg BS fmbj), the BS-1 group (fed basal diet with 0.2 × 1011 cfu/kg BS fmbj), BS-2 group (fed basal diet with 0.3 × 1011 cfu/kg BS fmbj), and BS-3 group (fed basal diet with 0.4 × 1011 cfu/kg BS fmbj). No differences were found in the average daily gain (ADG), average daily feed intake (ADFI), and feed conversion ratio (FCR) among the different treatments at 21 d. The BS groups showed lower (P < 0.05) levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) than that in the CON group at 21 d of growth. The dietary BS fmbj increased (P < 0.05) the antioxidant activity in the serum, liver, and hepatic mitochondria, but decreased (P < 0.05) the serum and hepatic malondialdehyde (MDA) levels compared to those in the CON group at 21 d of feeding. The BS groups showed lower (P < 0.05) level of hepatic mitochondrial reactive oxygen species (ROS), protein carbonyls (PC), and 8-hydroxy-2-deoxyguanosine (8-OHdG); however, in this group, higher (P < 0.05) levels of mitochondrial membrane potential (MMP), adenosine triphosphate (ATP), and mitochondria DNA (mtDNA) were determined compared to those in the birds of the CON group at 21 d. The BS group showed increased (P < 0.05) levels of antioxidant related-gene expression in the liver and hepatic mitochondria compared to that in the CON group. In conclusion, BS fmbj (0.3 g/kg in broiler diets) has the potential to improve the antioxidant status of the body as well as the hepatic mitochondrial function and thus, appears to be an important additive for both the consumer and the industry.
Effects of Dietary Bacillus subtilis HC6 on Growth Performance, Antioxidant Capacity, Immunity, and Intestinal Health in Broilers

Abstract:This study aimed to investigate the impact of Bacillus subtilis HC6 on the growth performance, immunity, antioxidant capacity, and intestinal health of broilers. A total of 180 one-day-old white feather broilers were randomly divided into two experimental groups, each comprising six replicates of fifteen chicks from 1 to 50 d of age. The groups were either fed a basal diet (CON) or the same diet supplemented with 5 × 108 cfu/kg of Bacillus subtilis HC6 (BS). Our results indicated that compared with the CON, dietary supplementation with BS increased feed efficiency during d 21–50 and d 1–50 (p < 0.05). Moreover, BS supplementation enhanced antioxidant capacity in the serum and liver, and also decreased the activity of diamine oxidase and the level of endotoxins (p < 0.05). Additionally, BS treatment increased the villi height in the jejunum and ileum, increased the ratio of villus height/crypt depth in the ileum, upregulated the expression of tight junction proteins in the jejunal mucosa, and downregulated the levels of IL-22 and IFN-γ on day 50 (p < 0.05). Principal coordinates analysis yielded clear clustering of two groups; dietary BS increased the relative abundance of Bacteroidales_unclassified (genus) and Olsenella (genus), and decreased the abundance of genera Alistipes on day 50, which identified a strong correlation with FCR, serum differential metabolites, or differential gene expression in the jejunal mucosa by spearman correlation analysis. The PICRUSt2 analysis revealed that supplementation with BS enriched the pathways related to xenobiotics biodegradation and metabolism, carbohydrate metabolism, energy metabolism, signaling molecules and interaction, the digestive system, and transport and catabolism. These results demonstrated that dietary BS increased feed efficiency, antioxidant capacity, and the mRNA expression of pro-inflammatory cytokines in the jejunal mucosa; and decreased the activity of diamine oxidase in serum, which might be attributed to the modulation of community composition and the functions of cecal microbiota in white-feathered broilers.

Abstract:This experiment was conducted to investigate the effects of Clostridium butyricum on growth per-formance, antioxidant capacity, immune function and serum biochemical parameters of broilers.A total of 540 one-day-old Ross 308 broilers were randomly assigned to 3 groups with 6 replicates per group and 30 birds per replicate for a 42-day trial.Dietary treatments were:an antibiotic-free basal diet ( control group, CON) , basal diet supplemented with 10 mg/kg colistin sulfate and 50 mg/kg bacitracin zinc ( antibiotic group, ANT) , and basal diet supplemented with 3×108 CFU/kg Clostridium butyricum (Clostridium butyricum group, CB).The results showed as follows:1) the body weight ( BW) , average daily gain ( ADG) and average daily feed in-take ( ADFI) of broilers in CB and ANT were significantly increased ( P<0.05) compared with those in CON. 2 ) The activities of serum glutathione peroxidase ( GSH-Px) and total superoxide dismutase ( T-SOD) in broil-ers at 42 days of age were significantly enhanced ( P<0.05) compared with those in CON, and GSH-Px activi-ty was 60.00%higher ( P<0.05) than that in ANT.At 7 and 21 days of age, serum T-SOD activity of broilers in ANT was significantly increased ( P<0.05) compared with that in CON.3) Compared with CON, serum immunoglobulin A ( IgA) content in broilers at 21 days of age in CB and ANT was increased by 28.70%( P<0.05) and 26.46%( P<0.05) , and serum immunoglobulin G ( IgG) content in broilers at 7, 21 and 42 days of age was increased by 36.60%( P<0.05) , 37.77%( P<0.05) and 27.03%( P<0.05) , respectively.Serum IgG content of broilers at 7 and 42 days of age in ANT was significantly higher ( P<0.05) than that in CON. Serum immunoglobulin M ( IgM) content of broilers at 21 and 42 days of age in CB was significantly increased compared with CON, while 7.92%( P>0.05) and 47.62%( P<0.05) higher than that in ANT.4) Serum to-tal protein ( TP) content of broilers in CB was significantly higher ( P<0.05) than that in CON, especially in-creased by 31.33%( P<0.05) and 52.27%( P<0.05) compared with that of broilers at 21 and 42 days of age in ANT.Meanwhile, serum ammonia content of broilers in CB was significantly decreased ( P<0.05) com-pared with that of broilers at 21 and 42 days of age in CON.These results indicated that supplementation with Clostridium butyricum in diet can enhance serum antioxidant capacity and immune function, promote protein metabolism, and then improve the growth performance of broilers;moreover, it can aslo reduce ammonia emis-sions from broilers.

Abstract:This study was conducted to determine the effects of B.subtilis on meat quality,nutrient digestibility and serum biochemical parameters of broilers.216 1-day-old Ross 308 broilers were randomly divided into the control group(basal diet)and the treatment group(basal diet supplemented with 105 CFU/g B.subtilis).Each group had three replicates 36 broilers each(half male and half female),the experiment lasted for 42 days.The results showed that dehydration rate of pectorales was decreased significantly,while the lightness(L*),redness(a*),and yellowness(b*)values of pectorales and crureus were improved(P >0.05)compared with the control group.The addition of B.subtilis increased the digestibility of dry matter(P<0.01),crude protein(P<0.01),ether extract(P<0.01)and crude ash(P<0.05)evidently.The level of serum BA,UA,T-CHL and TG were decreased significantly(P<0.05),while the activity of AKP was improved(P<0.01)compared to the control group.In conclusion,the addition of B.subtilis in the diets could improve meat quality,the digestibility of feed nutrient through impacting serum biochemical parameters of broiler.
Effects of Broccoli Stem and Leaf Meal on Broiler Performance, Skin Pigmentation, Antioxidant Function, and Meat Quality

Abstract:Three hundred sixty 1-d-old Ross 308 male broilers were used to study the effects of broccoli stem and leaf meal (BSLM) on growth performance, skin pigmentation, antioxidant function, and meat quality. The chicks were fed 4 diets containing different levels (0, 4.0, 8.0, and 12.0%) of BSLM as partial replacement for corn and soybean meal for a period of 42 d. The results showed that dietary supplementation of BSLM had no effect (P > 0.05) on growth performance. As compared with control, dietary 4%, 8%, and 12% BSLM increased (P < 0.05) b value (yellowness) both in shank and breast skin, increased (P < 0.05) the concentrations of xanthophylls in abdominal fat and breast skin, improved (P < 0.05) total antioxidant capability, lowered malondialdehyde concentration, and decreased drip loss percentage of breast muscle. Dietary 8% and 12% BSLM decreased (P < 0.05) shank L values (lightness), increased (P < 0.05) shank a value (redness), and increased (P < 0.05) the activities of superoxide dismutase and catalase of breast muscle as compared with control. The results indicated that dietary supplementation of BSLM in broiler chickens improved the poultry products quality with the more skin pigmentation and the less drip loss percentage of breast meat. The more skin pigmentation mainly related to the high amount of xanthophylls in BSLM. The decreased meat drip loss fed BSLM may be caused by the antioxidative function of BSLM.
A combination of selenium and Bacillus subtilis improves the quality and flavor of meat and slaughter performance of broilers

Abstract:This study aimed to investigate the effects of the combination of selenium and Bacillus subtilis (Se-BS) on the quality and flavor of meat and slaughter performance of broilers. A total of 240 one-day-old Arbor Acres broilers were randomly allotted to four treatments of a basal diet supplemented with no selenium (control), sodium selenite (SS), BS, or Se-BS and raised for 42 days. Compared with the control group, Se-BS significantly increased the carcass weight, the half-eviscerated weight, the completely eviscerated weight, the carcass rate, and redness in broiler muscles; improved the antioxidant state by increasing glutathione peroxidase (GPx) and glutathione S-transferase activities, the total antioxidant capacity, and GPx-1 and thioredoxin reductase 1 messenger RNA (mRNA) levels; promoted biological activity by increasing the contents of glutamate, phenylalanine, lysine, and tyrosine; and increased Se and five types of nitrogenous volatile substances in muscles. On the other hand, Se-BS treatment decreased the shear force, drip loss, and the malondialdehyde, glutathione, and lead contents in muscles. Se-BS exerted a better effect on slaughter performance, the physicochemical quality of meat, the redox status, the amino acid contents, the trace element contents, and volatile substances compared with SS and BS. In conclusion, Se-BS had a positive effect on the quality and flavor of meat and slaughter performance of broilers, suggesting that Se-BS may be a beneficial feed additive.

Abstract:Heat stress is the most common environmental stressor in poultry production, negatively affecting growth performance, meat quality, and welfare. Therefore, the aim of this study was to compare the nutritional effects of dietary supplementation with selenomethionine, Bacillus subtilis (BS), and a combination of selenomethionine and BS on broilers challenged with heat stress. A total of 300 21-day-old male broilers (Ross 308) were randomly assigned to 5 groups with 6 replicates of 10 broilers per each: control group (CON, broilers raised at 22 ± 2 °C), heat stress exposure group (HS, broilers raised at 32 ± 2 °C for 8 h/d), HSS group (HS group supplemented with 0.3 mg/kg selenomethionine), HSB group (HS group supplemented with 1 × 109 cfu/kg BS), and HSBS group (HS group supplemented with 0.3 mg/kg selenomethionine and × 109 cfu/kg BS). The experiment lasted for 21 d. The results indicated that, compared to the CON group, heat stress reduces (P < 0.05) broiler growth performance and damages the meat quality in breast and thigh muscles. Dietary supplementation with selenomethionine and BS did not improve the growth performance of broilers under heat stress. However, compared to the HS group, the HSS, HSB, and HSBS groups showed significantly increased (P < 0.05) pH45 min, redness (a*) and yellowness (b*), muscle fiber density, intramuscular fat, triglyceride content, and expression levels of Myf5, CAPN 2, FM, SLC27A1, A-FABP, H-FABP, APOB-100, and ACC in breast and thigh muscles. Meanwhile, these groups showed reduced (P < 0.05) lightness (L*), drip loss, shear force, muscle fiber cross-sectional area, and FM gene expression level. The HSBS group showed greater improvement in the physicochemical quality of muscle and volatile substances compared to the HSS and HSB groups. In conclusion, selenomethionine and BS improved meat quality and flavor in broilers under heat stress by modulating muscle fiber composition and characteristics, as well as increasing intramuscular fat deposition.

Abstract:This study was aimed to measure the dietary effects of probiotic Bacillus subtilis strain fmbj (BS fmbj) on antioxidant capacity and oxidative stability of chicken breast meat during storage. Treatment groups were fed the basal diet with BS fmbj at 0 g/kg (CON), 0.2 g/kg (BS-1), 0.3 g/kg (BS-2), or 0.4 g/kg (BS-3) doses without antibiotics. During 8 days of storage at 4°C, BS-2 group showed a significant improvement (P < 0.05) on meat quality (pH, Drip loss, Cooking loss, Shear force, color L*, a*, b*), free radical scavenging activity (DPPH, ABTS+, H2O2), tissues antioxidant enzyme capacity (SOD, CAT, GSH-Px, GSH, T-SH), mitochondria antioxidant enzyme capacity (MnSOD, GPx, GSH), mRNA expression of antioxidant genes (Nrf2, HO-1, SOD, CAT, GSH-Px) and mitochondrial function genes (avUCP, NRF1, NRF2, TFAM, PGC-1α), oxidative damage index (MDA, ROS, PC, 8-OHdG), and MMP level in chicken breast meat as compared to the CON group. These results indicate that dietary BS fmbj in broiler diets can protect breast meat against the storage-induced oxidative stress by improving their free radical scavenging capacity and antioxidant activity during 8 days of storage at 4°C.

Abstract:Three hundred one-day-old broilers were randomly divided into two groups.Each group had 3 replicates with 50 broilers per replication.The control group was fed with the basic diets and the treatment group was fed with the basic diets supplemented with 0.75 g /kg bacillus subtilis preparation.The whole experiment lasted 77 days.The earlier stage was from 1st to 35th day,and the later was from the 36th to 77th day.Results were as follows:compared with the control,the average body weight and daily gain in the treatment group increased by 6.75% (P0.05) and 8.22% (P0.05).In the later stage,the increase was more obvious (6.72% and 13.69%,P0.05).Besides,feed intakes of the treatment group was slightly higher than that of thecontrol (P0.05).For the gain ratios of the treatment group,there was an increase in the earlier stage (P0.05),but a decrease in the later.Levels of total antioxidant capacity(T-AOC),activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) of the test group were higher than those of the control group by 25.75%(P0.05)、20.10%(P0.05)and 18.12%(P0.05),and maleic dialdehyde (MDA) decreased correspondingly compared with the control (P0.05).
Effects of Dietary Supplementation of Glutamine on Meat Quality and Antioxidant Indexes of Broilers.

Abstract:The experiment was conducted to study the effects of glutamine on meat quality, flavor compounds and antioxidant indexes of broilers. Three hundred and sixty 1-day-old Avian broilers were randomly divided into four groups with 6 replicates in each group and 15 broilers in each replicate. Birds in control group were fed basal diet and those in test groups were fed basal diet added with 0. 5% , 1.0% and 1.5% glutamine, respectively. Eighteen birds from each group were slaughtered on the 42th day to determine related indexes. The results showed that compared with control group, in the three test groups water holding capacity and pH of meat were increased (P0. 05); the thickness of subcutaneous fat and intermuscular fat depth were decreased (P0. 05);the contents of collagen, essential amino acids, flavor amino acids, glutamate and phenylalanine in breast muscle were significantly increased (P0. 05), respectively. The contents of inosinic acid were increased in 1. 0% and 1. 5% Gln groups(P0. 05); and the content of malondialdehyde (MDA) was significantly decreased in 1. 0% Gln group (P0. 05); the activity of SOD tended to increase (P0. 05). In 0. 5% and 1. 0% Gln groups, the activities of SOD and GSH-Px in serum were significantly increased (P0. 05). It was concluded that adding glutamine into the diets could improve the meat quality, fresh flavor and antioxidant capacity of broilers, and the 0. 5% Gln group had the best effect.
Benefit of Dietary Supplementation with Bacillus subtilis BYS2 on Growth Performance, Immune Response, and Disease Resistance of Broilers

Abstract:A strain of Bacillus subtilis (B. subtilis) BYS2 was previously isolated from Mount Tai, which is located in Tai'an City in the Shandong Province of China. The strain was then stored in the Environmental Microbiology Laboratory at Shandong Agricultural University. To evaluate the effect of the bacterium preparation in broiler production, we fed the bacterium (106 CFU/g) to 1-day-old broilers and continued this feeding for 6 weeks to analyze its effect on growth and immune performance. We found that the average weight of the bacterium-fed group increased by 17.19% at weeks 5 compared to the control group (P < 0.05). The height of the villi in the duodenum and jejunum and the ratio of villi to crypt were significantly increased in the bacterium-fed group at weeks 5 (P < 0.05). Also, the IgG in the serum of broilers in the experimental group increased by 31.60% (P < 0.05) and IgM 30.52% (P < 0.05) compared with those in the control group. The expressions of the major pattern recognition receptors (PRRs), antiviral proteins, pro-inflammatory cytokines, and β-defensins were significantly higher than those in the control group (P < 0.05). Meanwhile, the bursa immune organ indices of broilers in the experimental group were significantly higher than those in the control group (P < 0.05). Also, after 5 weeks of continuous feeding, when infected with Escherichia coli (E. coli) O1K1 and Newcastle disease virus (NDV) F48E8, the content of bacteria and virus in tissues and organs of the experimental group decreased significantly, and the survival rate of infected chickens increased by 31.1% and 17.7%, respectively (P < 0.05). These results show that the anti-infective B. subtilis BYS2 could, to some extent, replace antibiotics to promote growth, improve innate immunity, and enhance disease resistance in broilers.

Abstract:This trial was conducted to investigate the adhesion of Bacillus amyloliquefaciens to Caco-2 cells, the effects of Bacillus amyloliquefaciens on the mucosal immunity, cecal volatile fatty acids and microbial diversity in broiler chickens. Three hundred and sixty 1-d-old Ross 308 chicks were randomly divided into 3 dietary treatments groups, which birds fed with basal diet, basal diet supplemented with colistine sulfate, and basal diet supplemented with Bacillus amyloliquefaciens. Polymerase chain reaction denaturing gradient gel electrophoresis was used to analyze the change of cecal microflora, and Gas Chromatography was used to analyze the cecal volatile fatty acids. Data showed that: 1) Bacillus amyloliquefaciens had a good adhesion ability to epithelial cells; 2) the supplementation of Bacillus amyloliquefaciens significantly increased the concentration of ileal mucosal secretory IgA and interleukin 6, decreased (P Lass Than 0.05) the concentration of tumor necrosis factor-a; 3) resulted in the change of cecal microbial community, higher levels of acetic acid, methylacetic and isovaleric acid in the birds. Thus, we considered that Bacillus amyloliquefaciens enhanced the mucosal immunity, increased the cecal concentration of major volatile fatty acids and the diversity microflora community in broilers.
Intestinal Morphologic and Microbiota Responses to Dietary Bacillus spp. in a Broiler Chicken Model

Abstract:Dietary inclusion of probiotic Bacillus spp. beneficially affect the broiler chickens by balancing the properties of the indigenous microbiota causing better growth performance. The effects of three Bacillus spp. on the growth performance, intestinal morphology and the compositions of jejunal microflora were investigated in broiler chickens. A total of 480 1-day-old male Arbor Acres broilers were randomly divided into four groups. All groups had six replicates and 20 birds were included in each replicate. The control birds were fed with a corn-soybean basal diet, while three treatment diets were supplemented with Bacillus coagulans TBC169, B. subtilis PB6, and B. subtilis DSM32315 with a dosage of 1 × 109 cfu/kg, respectively. The experiment lasted for 42 days. The compositions and diversity of jejunal microflora were analyzed by MiSeq high-throughput sequencing. The B. coagulans TBC169 group showed marked improvements of growth performance, nutrient digestibility and intestinal morphology compared with the other B. subtilis treatments. B. coagulans TBC169 supplementation improved the average body weight (BW), average daily weight gain (ADG), total tract apparent digestibility of crude protein and gross energy (GE), and reduced feed conversion rate (FCR) compared with the control group (P < 0.05). The villus height to crypt depth ratio (VH/CD) of jejunum and duodenum was increased in the birds fed with B. coagulans TBC169 compared with the control group (P < 0.05). However, two B. subtilis treatments presented more positive variation of the jejunum microflora of chickens than that in the B. coagulans TBC169 group. B. subtilis PB6 and B. subtilis DSM32315 treatments improved the diversity of jejunal microbiota on day 21 compared with the control (P < 0.05), while which were decreased on day 42 (P < 0.05). The supplementation with B. coagulans TBC169 significantly improved the proportion of Firmicutes, otherwise two B. subtilis significantly improved the proportion of Proteobacteria, Bacteroidetes, Actinobacteria, and Acidobacteria at the phylum level during starter phase and decreased the proportion of Bacteroidetes during growing phase compared with the control. The supplementation with B.subtilis DSM32315 significantly improved the proportion of Clostridiales during starter phase, whereas two B. subtilis significantly improved the proportion of Pseudomonas, Burkholderia, Prevotella, DA101 during growing phase at the genus level compared with the control. In conclusion, the dietary supplementation with probiotic Bacillus spp. strains improved body weight and intestinal morphology in broiler chickens, which might be associated with the gut microbiota.

Abstract:A Feeding trial was conducted to study the effects of different concentrations of CompositeBacillus sp.(Clostridium butyricum and Bacillus licheniformis,CBG) on growth performance,cecal microflora,and ammonia emission from excreta in broilers,to explore its action mechanism.Six hundred one-day-old Lingnan yellow-feathered broilers were randomly assigned to 4 groups with three replicate pens of 50 broilers each.There were 4 treatments: Control group,fed basal diet supplemented with 20 mg/kg Virginiamycin and 50 mg/kg Rroxarsone;and 3 experimental groups receiving basal diet supplemented with different concentrations of Composite Bacillus sp.(100,150,200 mg/kg,respectively).The experiment lasted for 50 days.The results showed that compared to the control,no significant differences were observed for growth performance after the supplementation of 100 mg/kg and 150 mg/kg Composit e Bacillus sp.(P >0.05).Composite Bacillus sp.had a significantly increasing effect on the populations of Lactobacillus of caecum at d 50(P <0.05).At 50 d of age,the pH value of caecum was significantly decreased after supplementation of 150 mg/kg and 200 mg/kg Composite Bacillus sp.preparation(P<0.05),and the group with 150 mg/ kg CBG got the optimal effect.With the supplementation of 150 mg/kg Composite Bacillus sp.,and uric acid content of its feces and caecum in day 50 was significant decreased(P<0.05).Composite Bacillus sp.significantly decreased the pH value of manure at day 43(P <0.05),and decreased the ammonia emission from excreta effectively,especially a better result happening on 150 mg/kg Composite Bacillus sp.group.Those indicated that Composite Bacillus sp.could regulate the intestinal microflora,decrease pH and the nitrogen content of feces and caecum,thus reduce the ammonia emission from manure.Results demonstrated that 150 mg/kg Composite Bacillus sp.is the best dosage in diets of broilers.

Abstract:AbstractThis study aimed to investigate the effects of selenium (Se)-enriched Bacillus subtilis (Se-BS) on growth performance, antioxidant capacity, immune status, and gut health in broilers. A total of 240 one-day-old Arbor Acres broilers were randomly allotted to four groups and fed with basal diet (control group), 0.30 mg/kg Se (SS group), 3 × 109 CFU/g B. subtilis (BS group), and 0.30 mg/kg Se + 3 × 109 CFU/g B. subtilis (Se-BS group) for 42 days. The results showed that Se-BS supplementation increased body weight (BW), average daily gain, the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), and peroxidase (POD), total antioxidant capacity (T-AOC), and the contents of interleukin (IL)-2, IL-4, and immunoglobulin (Ig) G in plasma, the index and wall thickness of the duodenum, the villus height and crypt depth of the jejunum, and GPx-1 and thioredoxin reductase 1 mRNA levels in liver and intestine and decreased feed conversion ratio (FCR) and plasma malondialdehyde (MDA) content compared with the control group on day 42 (P < 0.05). Compared with groups SS and BS, Se-BS supplementation increased BW, the activities of GPx, CAT, and POD, and the contents of IL-2, IL-4, and IgG in plasma, the index and wall thickness of the duodenum, the crypt depth and secretory IgA content of the jejunum, and GPx-1 mRNA levels in liver and intestine and decreased FCR and plasma MDA content on day 42 (P < 0.05). In conclusion, Se-BS supplementation effectively improved the growth performance antioxidant capacity, immune status, and gut health of broilers.
