Abstract:Food safety is a global concern, and conventional methods for detecting foodborne bacteria, such as polymerase chain reaction and enzyme-linked immunosorbent assay, often involve time-consuming processes, specialized equipment, or specific recognition of particular bacterial strains. There is an urgent need for more efficient and convenient detection methods for foodborne pathogens. This study addresses this need by introducing an easily constructed fluorescent sensor array for the identification of various foodborne bacteria. The sensor array comprises carbon quantum dots (CQDs) functionalized with ampicillin, polymyxin, and gentamicin, each exhibiting different affinities for binding with specific bacteria. Leveraging machine learning algorithms, the proposed sensor array enables rapid, accurate, and highly sensitive identification of foodborne pathogens. This approach offers a convenient solution for the development of rapid, accurate, and hypersensitive detection methods for multiple bioactive samples. In summary, the fluorescent sensor array in this study holds promise for advancing the field of foodborne bacteria detection.

Optimization of Sensors Array as an Electronic Nose in Identifying Adulterate Milk

Abstract:The electronic nose was widely used in the food area. A single couldn’t assess the quality of odor because its complexity. The sensors array could resolve this problem. The sensors array was different when different food was identified. The adulterate milk were identified by electronic nose, and the signals were analyzed by variance analysis, PCA (principal component analysis) and clustering analysis. The best sensors array made up of 8 sensors was obtained. The new sensors array was applied to identify the adulterate milk. The data was analyzed by LDA analysis (Linear Discriminant Analysis). The plot of 8-sensor array could distinguish the milk adulterated with powdered milk of different proportion. So the electronic nose with 8-sensor array had the ability of distinguishing the adulterate milk well as electronic nose with the 10-sensor array.

Abstract:Culture plating is worldwide accepted as the gold standard for quantifying viable foodborne pathogens. However, it is time-consuming (1-2 days) and requires specialized laboratory and personnel. This study reported a deep learning enhanced digital microfluidic platform for multiplex detection of viable foodborne pathogens. The new method used a Time-Lapse images driven EfficientNet-Transformer Network (TLENTNet) to type and quantify the bacteria through spatiotemporal features of bacterial growth and digital enumeration of bacterial culture. First, the bacterial sample was prepared with LB medium and injected into a pre-vacuumed microfluidic chip with an array of 800 microwells to encapsulate at most one bacterium in each well. Then, a programmed sliding microscopic platform was used to scan all microwells every 15 min, capturing time-lapse images of bacterial growth within each microwell. Finally, the TLENTNet was used to facilitate bacterial typing and quantification. Under optimal conditions, this platform was able to detect four bacterial species (S.typhimurium, E. coli O157:H7, S. aureus and B. cereus) with an average accuracy of 97.72% and a detection limit of 63 CFU/mL in 7 h.

Abstract:The contamination of mycotoxins is a serious problem around the world. It has detrimental effects on human beings and leads to tremendous economic loss. It is essential to develop a rapid and non-destructive method for contamination recognition particularly for early alarm. In this study, the whole-cell biosensor array was constructed and employed for rapid recognition of wheat contamination by combining with machine learning algorithms. Seven key VOCs were explored through univariate coupling to multivariate analysis of orthogonal partial least squares-discrimination analysis (OPLS-DA) models. The promoters of dnaK , katG , oxyR , soxS obtained from the stress-responsive of key VOCs were fused to the bacterial operon and fabricated on the whole-cell biosensor. The constructed whole-cell biosensor array was consisted with four kinds of sensors and 18 sensor unit. The bioluminescent intensity combined with linear machine learning algorithm of partial least squares discriminant analysis (PLS-DA) and non-linear algorithms of back propagating artificial neural network (BP-ANN) and least square support vector machine (LS-SVM) were employed to establish discrimination models for mold contamination especially for early warning. The Monte-Carlo strategy was performed to generate thirty subsets for modeling to give more reliable results. As a result, the whole-cell biosensor combined with non-linear algorithm of LS-SVM was practicable for detecting mold identification for wheat early-warning with the accuracy of 97.24%. Additionally, this study provides practical and effective methods not only for wheat quality guarantee and supervision but also for other foodstuffs.

Abstract:Antibiotic-resistant bacteria pose considerable risks to global health, particularly through transmission in the food chain. Herein, we developed the artificial intelligence-driven quantification of antibiotic-resistant bacteria in food using a color-encoded multiplex hydrogel digital loop-mediated isothermal amplification (LAMP) system. The quenching of unincorporated amplification signal reporters (QUASR) was first introduced in multiplex digital LAMP. During amplification, primers labeled with different fluorophores were integrated into amplicons, generating color-specific fluorescent spots. While excess primers were quenched by complementary quenching probes. After amplification, fluorescent spots in red, green, and blue emerged in hydrogels, which were automatically identified and quantified using a deep learning model. Methicillin-resistant Staphylococcus aureus and carbapenem-resistant Escherichia coli in real fruit and vegetable samples were also successfully detected. This artificial intelligence-driven color-encoded multiplex hydrogel LAMP offers promising potential for the digital quantification of antibiotic-resistant bacteria in the food industry.

Abstract:Biohazards, which may occur at all supply chain stages, pose significant threats to food safety and public health. Addressing these concerns and enhancing food safety necessitates a nondestructive pathogen surveillance approach capable of continuously and simultaneously detecting multiple pathogens. Detecting and differentiating low concentrations of pathogenic bacteria amid high background microflora levels in foods is challenging, requiring technology with high sensitivity and robust discriminatory capability. This study introduces an artificial neural network-driven paper chromogenic array sensor (ANN-PCA) technique developed for the nondestructive, continuous, and simultaneous detection of Salmonella Enteritidis ( SE ) and Escherichia coli O157:H7 ( Ec ) from a high background microflora in shredded cheddar cheese. This method enables accurate detection of SE and Ec in monoculture and cocktail culture while distinguishing them from a high level of background microflora (~7.5 log CFU/g), with accuracies ranging from 72 ± 11% to 92 ± 3%. In addition, SE and Ec were successfully identified at concentrations as low as 1 log CFU/g within one day, with an accuracy of 72 ± 11%. This approach exhibits promising potential for integration into a digitalized, smart, and resilient nondestructive surveillance system for real-time pathogen detection in foods throughout the supply chain without enrichment, incubation, or other sample preparation steps.

Abstract:Abstract Background The identification of foodborne pathogenic bacteria types plays a crucial role in food safety and public health. In consideration of long culturing times, tedious operations and the desired specific recognition elements in conventional methods, the alternative fluorescent sensor arrays can offer a high-effective approach in bacterial identification by using multiple cross-reactive receptors. Herein, we achieve this goal by constructing an upconversion fluorescent sensor array based on anti-stokes luminogens featuring a series of functional lanthanide-doped upconversion nanoparticles (UCNPs) with phenylboronic acid, phosphate groups, or imidazole ionic liquid. The prevalent spotlight effect of microorganism and the electrostatic interaction between UCNPs and bacteria endow such sensor array an excellent discrimination property. Results Seven common foodborne pathogenic bacteria including two Gram-positive bacteria ( Staphylococcus aureus and Listeria monocytogenes ) and five Gram-negative bacteria ( Escherichia coli , Salmonella , Cronobacter sakazakii , Shigella flexneri and Vibrio parahaemolyticus ) are precisely identified with 100% accuracy via linear discriminant analysis (LDA). Furthermore, blends of bacteria have been identified accurately. Bacteria in real samples (tap water, milk and beef) have been effectively discriminated with 92.1% accuracy. Conclusions Current fluorescence sensor array is a powerful tool for high-throughput bacteria identification, which overcomes the time-consuming bacteria culture and heavy dependence of specific recognition elements. The high efficiency of whole bacterial cell detection and the discrimination capability of life and death bacteria can brighten the application of fluorescence sensor array.

Abstract:Rapid and accurate identification of foodborne pathogens improves public health. Currently employed methods are time-consuming, sensitive to environmental factors, and complex. This study develops a colorimetric sensor for detecting multiple bacteria with one probe using double-enzyme-induced colorimetry. Based on alkaline phosphatase (ALP) in bacteria decomposes L-ascorbic acid 2-magnesium phosphate salt hydrate into ascorbic acid (AA). Manganese dioxide flowers (MnO 2 NFs) can oxidize TMB to etch gold nanorods (Au NRs), which can be inhibited by AA reduction to produce rich colors. Bacteria with varying ALP levels can be identified based on color changes and plasmon resonance wavelength signals produced from Au NRs. Furthermore, the conversion of RGB signals to digital signals and the use of linear discriminant analysis (LDA) allowed 99.57% accuracy in identifying multiple bacteria. It can simultaneously identify five foodborne pathogens across diverse environments (shrimp, meat, milk, etc.). This method may be useful for the rapid and simple identification of foodborne illnesses.

Abstract:Foodborne illnesses caused by Salmonella and Staphylococcus aureus are a significant public health concern, leading to societal and economic repercussions. It is important to develop a simple and straightforward bacteria detection and identification method. A triple-probe multiplex rolling circle amplification technique has been developed to simultaneously detect Salmonella Typhimurium and S. aureus. This method utilizes fluorophore-labeled long padlock probes targeting S. Typhimurium invA and S. aureus glnA specific genes, along with a pH-based detection approach for direct visual identification. The multiplex hyperbranched saltatory rolling circle amplification assay at 30 °C has showed promising results with synthetic targets within 30 min and real bacteria within 2 h after establishing the detection settings. The assay is specific for S. aureus and S. Typhimurium, with a limit of detection of 39 μM for fluorescence and 78 μM for colorimetric. In the simulative test of this method for the detection of S. Typhimurium and S. aureus in milk, the limit of detection for the fluorescence signal after 2 h of amplification was 10 CFU/mL and 5 CFU/mL, respectively. The detection method was evaluated to be stable enough to detect pathogen for 3.29 months. Consequently, this triple-probe-multiplex rolling circle amplification method displays notable specificity, sensitivity, as well as ease of interpretation when testing food samples for harmful pathogens.
Mechanisms of solvent evaporation encapsulation processes: prediction of solvent evaporation rate.

Abstract:The mechanism of organic solvent evaporation during microencapsulation and its role during microsphere hardening has been investigated. Evaporation and encapsulation studies were carried out in a jacketed beaker, filled with aqueous hardening solution, which was maintained at constant temperature and constant stirring rate in the turbulent regime. Evaporation of dissolved methylene chloride (MC), ethyl acetate (EA), and acetonitrile (ACN) was examined by the decline in organic solvent concentration in the hardening bath, which was monitored by gas chromatography. The evaporation from the bath followed first-order kinetics under dilute conditions (e.g., MC < 3 mg/mL), yielding an overall permeability coefficient, P. The value of P was theoretically related to the Kolmogorov length-scale of turbulence under conditions that favor liquid-side transport control. According to theory, factors that favored liquid-phase control (as opposed to gas-phase control) were those that favored a high Henry's law constant [i.e., elevated temperature near the normal boiling point (bp) of the organic solvent] and properties of the dissolved organic solvent (i.e., low normal bp and low aqueous solubility). These theoretical hypotheses were confirmed by (1) correlating the experimentally determined P with process variables raised to the appropriate power according to theory, r(2) = 0.95 (i.e., P approximately rotational speed, omega(3/4), impeller diameter, d (5/4), volume of hardening bath, V(-1/4), and the product of kinematic viscosity and diffusion coefficient, nu(-5/12)D (2/3)), and (2) illustrating that at constant temperature, the tendency of the evaporation system to obey liquid-side transport control follows the same order of increasing Henry's law constant (i.e., MC > EA > ACN). To establish the relationship of evaporation with microsphere hardening, the decline in MC concentration was determined in both the continuous and dispersed polymer phases during microencapsulation. By applying a mass balance with respect to MC in the hardening bath, the cumulative hardening profile of the microspheres was accurately predicted from the interpolating functions of the kinetics of MC loss from the bath with and without polymer added. These results have potential use for microsphere formulation, design of encapsulation apparatus, and scale up of microsphere production.
In-field Detection of Multiple Pathogenic Bacteria in Food Products Using a Portable Fluorescent Biosensing System

Abstract:Foodborne diseases caused by pathogenic bacteria have increasingly become a major public health issue worldwide. Rapid, simple, and accurate detection methods are urgently needed for in-field screening of bacterial pathogens. In our previous work, rapid detection methodologies have been established based on fluorescent nanobiosensors for simultaneous separation and detection of multiple foodborne pathogenic bacteria. In this research, a portable fluorescent biosensing system was designed and built and further assessed for in-field detection of three main types of bacterial pathogens that have been associated with the outbreaks of foodborne illness. Using the developed fluorescent nanobiosensor coupled with nanobead-based immunomagnetic separation, we conducted blind tests with the portable device to simultaneously detect E. coli O157:H7, L. monocytogenes, and S. Typhimurium in different food products in three cities selected from three big agricultural provinces in China. Specificity tests showed low interference of this multiplex biosensor from non-targets in food samples. The detection could be done from sampling to results within 60 min. Limits of detections of this method for E. coli O157:H7, L. monocytogenes, and S. Typhimuriuin were determined to be 10(2), 10(3), and 10(3) CFU/mL in lettuce, shrimp, and ground beef, respectively. Recovery tests were also investigated and this method was evaluated to be accurate comparing with the gold standard culturing method. Therefore, it is feasible for this portable fluorescence biosensing system to be used in rapid and in-field screening of multiple foodborne pathogenic bacteria in foods, such as vegetables, livestock meat, and sea food. And together with fluorescent nanobiosensors, it provides a promising alternative tool to traditional culturing method, or even conventional ELISA and PCR based methods. (C) 2016 Elsevier Ltd. Abstract:Recurrent incidents of economically motivated adulteration have long-lasting and devastating effects on public health, economy, and society. With the current food authentication methods being target-oriented, the lack of an effective methodology to detect unencountered adulterants can lead to the next melamine-like outbreak. In this study, an ensemble machine-learning model that can help detect unprecedented adulteration without looking for specific substances, that is, in a non-targeted approach, is proposed. Using raw milk as an example, the proposed model achieved an accuracy and F1 score of 0.9924 and 0. 0.9913, respectively, when the same type of adulterants was presented in the training data. Cross-validation with spiked contaminants not routinely tested in the food industry and blinded from the training data provided an F1 score of 0.8657. This is the first study that demonstrates the feasibility of non-targeted detection with no a priori knowledge of the presence of certain adulterants using data from standard industrial testing as input. By uncovering discriminative profiling patterns, the ensemble machine-learning model can monitor and flag suspicious samples; this technique can potentially be extended to other food commodities and thus become an important contributor to public food safety.

Abstract:A direct nanoprinting technique is used to fabricate a pit arrangement on the surface of an organic amorphous emissive layer. A pit arrangement with a closely packed hexagonal pattern with a 400 nm pitch is successfully fabricated within 60 s. Pit depth is more than 70 nm and depends on press pressure. The fabricated structure is considered to be a photonic crystal band edge laser, and laser oscillation is confirmed by optical excitation. It is also confirmed that the oscillation threshold can be controlled simply by changing the press pressure used in the direct nanoprinting procedure. The results show that the use of the direct nanoprinting technique enables the fabrication of organic nanostructure devices such as photonic crystal band edge lasers with an organic emissive material.

Abstract:With the growing global concern over food safety, the rapid detection and disinfection of foodborne pathogens have become critical in public health. This study presents a novel machine learning-driven fluorescent sensor array utilizing aqueous CsPbBr 3 perovskite quantum dots (PQDs) for the rapid identification and eradication of foodborne pathogens. The relative signal intensity changes (ΔRGB) generated by the sensor array were analyzed using the machine learning algorithm—Support Vector Machine (SVM). The study achieved the identification and recognition of five pathogens and their mixtures within a concentration range of 1.0 × 10 3 to 1.0 × 10 7 CFU/mL with an accuracy rate of 100 %, and the limits of detection (LOD) for the pathogens were found to be low. Additionally, the array also showed excellent performance in the identification of pathogens in tap water, achieving an accuracy rate of 100 %. Furthermore, the fluorescent sensor array was capable of inactivating the pathogens with an efficiency of over 99 % within 30 min post-detection. This development provides an efficient and reliable tool for the field of food safety detection.

Abstract:Chemical-nose strategy has achieved certain success in the discrimination and identification of pathogens. However, this strategy usually relies on non-specific interactions, which are prone to be significantly disturbed by the change of environment thus limiting its practical usefulness. Herein, we present a novel chemical-nose sensing approach leveraging the difference in the dynamic metabolic variation during peptidoglycan metabolism among different species for rapid pathogen discrimination. Pathogens were first tethered with clickable handles through metabolic labeling at two different acidities (pH = 5 and 7) for 20 and 60 min, respectively, followed by click reaction with fluorescence up-conversion nanoparticles to generate a four-dimensional signal output. This discriminative multi-dimensional signal allowed eight types of model bacteria to be successfully classified within the training set into strains, genera, and Gram phenotypes. As the difference in signals of the four sensing channels reflects the difference in the amount/activity of enzymes involved in metabolic labeling, this strategy has good anti-interference capability, which enables precise pathogen identification within 2 h with 100% accuracy in spiked urinary samples and allows classification of unknown species out of the training set into the right phenotype. The robustness of this approach holds significant promise for its widespread application in pathogen identification and surveillance.

Abstract:Global food systems can benefit significantly from continuous monitoring of microbial food safety, a task for which tedious operations, destructive sampling, and the inability to monitor multiple pathogens remain challenging. This study reports significant improvements to a paper chromogenic array sensor - machine learning (PCA-ML) methodology sensing concentrations of volatile organic compounds (VOCs) emitted on a species-specific basis by pathogens by streamlining dye selection, sensor fabrication, database construction, and machine learning and validation. This approach enables noncontact, time-dependent, simultaneous monitoring of multiple pathogens ( Listeria monocytogenes , Salmonella , and E. coli O157:H7) at levels as low as 1 log CFU/g with over 90% accuracy. The report provides theoretical and practical frameworks demonstrating that chromogenic response, including limits of detection, depends on time integrals of VOC concentrations. The paper also discusses the potential for implementing PCA-ML in the food supply chain for different food matrices and pathogens, with species- and strain-specific identification.

Abstract:Fast and reliable identification of pathogenic bacteria is of upmost importance to human health and safety. Methods that are currently used in clinical practice are often time consuming, require expensive equipment, trained personnel, and therefore have limited applications in low resource environments. Molecular identification methods address some of these shortcomings. At the same time, they often use antibodies, their fragments, or other biomolecules as recognition units, which makes such tests specific to a particular target. In contrast, array-based methods use a combination of reporters that are not specific to a single pathogen. These methods provide a more data-rich and universal response that can be used for identification of a variety of bacteria of interest. In this report, we demonstrate the application of the excitation-emission spectroscopy of an environmentally sensitive fluorescent dye for identification of pathogenic bacterial species. 2-(4'-Dimethylamino)-3-hydroxyflavone (DMAF) interacts with the bacterial cell envelope resulting in a distinct spectral response that is unique to each bacterial species. The dynamics of dye-bacteria interaction were thoroughly investigated, and the limits of detection and identification were determined. Neural network classification algorithm was used for pattern recognition analysis and classification of spectral data. The sensor successfully discriminated between eight representative pathogenic bacteria, achieving a classification accuracy of 85.8% at the species level and 98.3% at the Gram status level. The proposed method based on excitation-emission spectroscopy of an environmentally sensitive fluorescent dye is a powerful and versatile diagnostic tool with high accuracy in identification of bacterial pathogens.
