Abstract:BACKGROUND: Long term successful root canal treatment requires effective debridement and disinfection of root canal system. Persistent periradicular lesions are usually associated with Enterococccus faecalis. Prompt research for natural alternatives for irrigation is mainly due to the constant increase in antibiotic resistant strains and side effects caused by synthetic drugs. Sodium hypochlorite; the gold standard for irrigation has many disadvantages. Therefore, the present study was aimed to explore newer irrigants probably be as more effective and at the same time would be less irritating to the tissues than NaOCl.MATERIAL AND METHODS: Ninety extracted human mandibular premolars were biomechanically prepared, vertically sectioned, placed in tissue culture wells exposing the root canal surface to E. faecalis to form a biofilm. At the end of 3rd week, all groups were irrigated with 3 ml of test solutions and control for 10 minutes. The samples were then scraped with a scalpel, inoculated on tryptone soy agar plates and incubated for 24 hours at 37ºC. The plates were then subjected to digital colony counter and evaluated for E. faecalisgrowth. The growth was statistically analysed by ANOVA & Post Hoc Tukey tests.RESULTS: Chitosan + Chlorhexidine, NaOCl and Chlorhexidine showed no statistically significant difference, whereas all the other inter‑group differences were statistically significant (P<0.05).CONCLUSIONS: Chitosan + Chlorhexidine, Chlorhexidine and Propolis were found to be as efficacious as sodium hypochlorite. The use of natural alternatives as root canal irrigation solutions might prove to be advantageous considering several unfavorable properties of NaOCl. Surgical satellite pharmacies in institutions with anesthesiology training programs for physicians.

Abstract:The results of a survey on the use of surgical satellite pharmacies in hospitals with anesthesiology training programs are reported. In June 1990 a questionnaire was mailed to 158 directors of anesthesiology training programs for physicians. The questionnaire solicited information on the presence of surgical satellite pharmacies in the training hospitals and the nature of the services provided, including accounting for controlled substances. Responses were received from 102 program directors and their designees, for a 65% response rate. Some respondents returned questionnaires completed by affiliated hospitals; a total of 137 responses were accumulated. Surgical satellite pharmacies were present in 46 (34%) of the 137 hospitals. Of those 46 satellite pharmacies, only 14 dispensed all controlled substances to anesthetic-administration areas. Most of the satellite pharmacies provided services at least eight hours per day. All 46 pharmacies dispensed controlled substances, 31 provided i.v. admixtures, and 12 dispensed all i.v. solutions. Accountability for controlled drugs was provided through a daily inventory count (45 satellite pharmacies), daily comparison of agents received and returned (36), review of the anesthesia record (31), random audits of individual providers (18), or quantitative or qualitative analysis of residual drugs (16). Accountability was considerably better in hospitals with surgical satellite pharmacies than in hospitals without them. Surgical satellite pharmacies provided increased accountability for controlled substances in institutions with anesthesiology training programs but did not use all the available methods for preventing drug diversion.
Comparing the Biofilm Removal Capacity of NaOCl, Povidone-Iodine, Chlorhexidine, Curcumin, and Triphala as Endodontic Irrigants

Abstract:Background and aim: A sessile multicellular organism that is immersed in a self-produced matrix of extracellular polymeric substances and has its cells firmly attached to a surface is referred to as a microbial biofilm. When it comes to pulp and periradicular pathosis, biofilms are crucial. To reduce the number of microorganisms in the root canal and assist in treating periapical pathosis, endodontic therapy must include decontamination of the system of tooth root canals through biomechanical preparation and irrigation of the root canal. This study compares sodium hypochlorite (NaOCl), povidone-iodine, chlorhexidine, curcumin, and triphala as endodontic irrigating solutions regarding their capacity to eliminate biofilm from root canals. Materials and methods: A total of 60 patients were included if they had pulpitis. Two specific samples (samples A and B) were chosen for analysis from a collection of samples so that their bacterial composition is most similar to that of acute pulpitis. The suspensions of bacterial cells from this polymicrobial culture have been collected from frozen stock and then regrown by inoculation on Columbia agar base (Basingstoke, UK) with the addition of vitamin K1, hemin, and 5% (v/v) calf blood. The pureness of the suspensions was assessed using colony morphology and Gram staining. Analytical profile index (API) 20A tests or automated test for bacteria (ATB) ID 32A tests were initially used to identify the isolates. These polymicrobial cultures' in vitro biofilms were developed using membrane filters made of cellulose nitrate. The tested irrigating solutions were as follows: 5.25% sodium hypochlorite, 10% triphala, 0.2% chlorhexidine gluconate, 10% povidone-iodine, and 5% curcumin (CUR). On the other hand, phosphate-buffered saline was taken as a control agent. Results: As the standard of excellence in endodontic irrigation, NaOCl has eliminated all germs in sample A following 15 minutes of culture and in both of the specimens after 40 minutes. Iodine also eliminated all germs after 40 minutes of administration, indicating that it would be worth exploring using iodine as a potential endodontic irrigant. Iodine achieved total bacterial elimination after 40 minutes in both samples; however, it was less effective after 15 minutes. Our findings indicate that iodine solution is the most suitable alternative after the supremely effective NaOCl, although it requires longer contact times to generate the necessary and recognized broad-spectrum antibacterial properties, including in the case of biofilms. Furthermore, curcumin also showed significant results after NaOCl and iodine. Conclusion: The antibacterial potency of each studied irrigant was significant, supporting their usage in endodontics. It was observed that NaOCl has the maximum antibacterial activity.

Abstract:Aim. This study aimed to evaluate the antibacterial effect of sodium hypochlorite gel and four types of intracanal medicaments. Materials and Methods. The agar diffusion method was used to evaluate the antibacterial effect of five medicaments (sodium hypochlorite gel (NaOCl), chlorhexidine gel (CHX), calcium hydroxide paste (CH), Ledermix, and Diapex plus) against Enterococcus faecalis (E. faecalis), Staphylococcus aureus (S. aureus), and Escherichia coli (E. coli). The zone of inhibition around each medicament was measured in millimeters, after 48 hours of incubation at 37°C. The antibacterial effects of medicaments against each microbial strain and the sensitivity of microorganisms towards each medicament were compared using the one-way ANOVA and Games–Howell post hoc tests. The level of significance was set to p < 0.05 . Results. All medicaments showed variable inhibition zones for all bacterial strains except Diapex Plus which showed no antibacterial activity. NaOCl gel exhibited the most significant inhibition zones for all bacterial strains followed by CHX gel, Ledermix, and CH. However, the effect of CHX and CH paste against S. aureus was statistically similar, while the effect of CH against E. faecalis was significantly higher than the Ledermix. Conclusion. Sodium hypochlorite gel displayed the highest antibacterial activity among tested medicaments and can be recommended as a potent intracanal medicament. Chlorhexidine gel showed a significantly higher antibacterial effect when compared with Ledermix and calcium hydroxide. Calcium hydroxide demonstrated stronger antibacterial activity against E. faecalis than Ledermix. Diapex Plus exhibited no antibacterial effect.

Abstract:Background. This in vitro study aimed to compare the antibacterial effect of different concentrations of sodium hypochlorite on elimination of Enterococcus faecalis from root canal systems of primary teeth with or without a passive sonic irrigation system (EndoActivator). Methods. The root canals of 120 extracted single-rooted primary incisors were prepared using the crown-down technique. The teeth were autoclaved and inoculated with E. faecalis. The infected samples were then randomly divided into 6 experimental groups of 15 and positive and negative control groups as follows: group 1: 0.5% sodium hypochlorite solution; group 2: 2.5% sodium hypochlorite solution; group 3: 5% sodium hypochlorite solution; group 4: 0.5% sodium hypochlorite solution + sonic activation; group 5: 2.5% sodium hypochlorite solution + sonic activation; and group 6: 5% sodium hypochlorite solution + sonic activation. Microbiological samples were collected before and after disinfection procedures and the colony-forming units were counted. Statistical analyses were performed using the two-way ANOVA and post hoc Duncan's tests in cases of significant difference. Results. There were no significant differences between the groups in any of the variables (concentration of antiseptic or use of sonic irrigation system). Conclusion. Use of passive sonic irrigation systems in endodontic treatment of single-rooted primary teeth is of no benefit compared to regular needle irrigation. The results of this study also recommends use of lower concentrations of sodium hypochlorite solution (0.5%) for irrigation of the root canal system rather than higher concentrations given approximately equal efficacy.
Antibacterial Ability of Sodium Hypochlorite Activated with PUI vs. XPF File against Bacteria Growth on Enterococcus faecalis Mature Biofilm

Abstract:The objectives of the present study were to assess the antibacterial effectiveness of two sodium hypochlorite (NaOCl) concentrations (2.5% and 5.25%) activated by means of two techniques, passive ultrasonic irrigation (PUI) and XP-endo® Finisher (FKG Dentaire SA, La Chaux-de-Fonds, Switzerland) (XPF) against bacteria growth in intracanal mature biofilm. Our aim was to determine if the effect of heating up NaOCl at body temperature (BT) contributed to an improvement of the efficacy of XPF. Sixty-two single-canal human roots previously instrumented were infected with E. faecalis inoculum at 0.5 McFarland and incubated at 37 °C for two weeks. Twelve specimens were randomly selected as positive control, and the remaining fifty were divided into five experimental groups (n = 10). The first two were irrigated with 2.5 vs. 5.25% NaOCl at room temperature (RT), activated with PUI, and the other three were irrigated with XPF. Of these three, two were irrigated using 2.5 vs. 5.25% NaOCl at RT and one was irrigated with 5.25% NaOCl at BT. Our results showed that NaOCl was effective in biofilm removal for all experimental groups (p > 0.05), especially in the groups irrigated with 5.25% NaOCl at room temperature (RT) activated with PUI and the group treated with 5.25% NaOCl at BT with XPF. These groups were the most successful ones (p < 0.001). NaOCl, activated with XPF, was as effective as PUI in biofilm removal from the apical third of the canal when it was used at higher concentration and heated up. This study indicates that XPF only reached the efficacy of PUI when NaOCl was heated up.
Effectiveness of calcium and sodium hypochlorite in association with reciprocating instrumentation on decontamination of root canals infected with Enterococcus faecalis

Abstract:The aim of this study was to evaluate the antimicrobial activity of sodium and calcium hypochlorite utilising reciprocating instrumentation. Sixty root canals were inoculated with E. faecalis for 14 days. Samples were divided into six groups according to decontamination protocol: G1: no treatment, G2: distilled water, G3: 2.5% sodium hypochlorite, G4: 2.5% calcium hypochlorite, G5: 5.25% sodium hypochlorite and G6: 5.25% calcium hypochlorite. Instrumentation was performed with Wave One reciprocating system (Dentsply Sirona Endodontics, York, PA, USA) in groups G2 to G6. Colony-forming units (CFUs) counting was performed and the data were subjected to Anova and Tukey (α = 0.05). Group 1 and 2 showed the highest mean contamination, with a significant difference between them (P < 0.05). Groups 3, 4, 5 and 6 showed the lowest contamination means with no significant difference between them (P < 0.05). Sodium and calcium hypochlorite, in association with reciprocating instrumentation, can be an effective decontamination protocol in root canals infected with E. faecalis.

Abstract:Aim: This study aims to evaluate and compare the antimicrobial effect of Photosensitized Nanoparticles, Diode Laser (DL), and Conventional and Sonic Irrigation Systems against Enterococcus faecalis in root canals – An in vitro Study. Materials and Methods: The root canals of 104 extracted human single-rooted teeth were prepared, and E. faecalis was incubated in the root canals for 1 week. The teeth were randomly divided into four experimental groups and one control group ( n = 20): No treatment was done, sodium hypochloride (NaOCl) group ( n = 20): Canals irrigated with5 ml of 2.5% NaOCl for 60 s, NaOCl and Endoactivator group ( n = 20): Canals irrigated with 5 ml of 2.5% NaOCl for 60 s followed by activation of NaOCl with Endoactivator for 30 s, NaOCl and 910 nm DL group ( n = 20): (1 W 5 times for 5 s) canals irrigation with5 ml of 2.5% NaOCl for 60 s followed by Photodynamic Therapy (PDT) with 910 nm DL, silver nanoparticles (AgNPs)/Indocyanine green (ICG)/DL group ( n = 20): DL modified PDT with 5 min of irrigation with 5 mL AgNPs (100 ppm)/ICG/910 nm DL (200 mW, 30 s). A reduction in colony count was assessed by counting colony-forming units by Digital Colony Counter. Results: Significant reductions were noted in E. faecalis colony counts in all groups ( P 0.05). Conclusions: NaOCl and 910 nm DL were more successful in reducing the number of E. faecalis colony counts followed by NaOCl and Endoactivator group but it can also be concluded that PDT with ICG, an 910 nm DL, and AgNPs has the potential to be used as an adjunct for disinfection of the root canal system.

Abstract:Purpose: To evaluate the antimicrobial effect of chitosan as root canal irrigant on Enterococcus faecalis in comparison to 3%NaOCL in experimentally infected primary anterior root canals. Materials and methods: Access cavity and mechanical preparation of root canals of 30 extracted primary anterior teeth which were then sterilized at autoclave at 1210 for 15 minutes. These teeth inoculated with Enterococcus faecalis with reference strain ATCC 19433.Then grouped into three groups which were irrigated with 3%NaOCl, 0.5% chitosan and saline respectively. Samples were cultured on bile auscline agar incubated at 370 for 24 hours. Counting of colony forming units formed before and after irrigation was performed. Results: Chitosan had higher antimicrobial effect than saline and lower than NaOCl. Statistical analysis between the groups revealed that there was a statistically significant difference between the three irrigants (P-value = 0.001). Conclusion: Chitosan solution can be used as a natural alternative to sodium hypochlorite in root canal irrigation, but may need to increase its concentration or the time of application inside the root canals and also should be supported by in vivo studies.

Abstract:ABSTRACT Aim: The purpose of this systematic review was to evaluate the superiority of sodium hypochlorite (NaOCl) liquid or NaOCl gel in disinfecting root canals. Methods: Online electronic databases such as PubMed-MEDLINE, Scopus, and Embase were explored for suitable studies. In this evaluation, papers were deemed eligible for the data extraction if they included in vitro experiments using NaOCl liquid or NaOCl gel for canal irrigation and its influence on the smear layer, as well as their elimination of the bacteria from the root canal. The review was registered in International Prospective Register of Systematic Reviews with registration number CRD420223441126. Results: One hundred and sixty-one articles were found after initial screening and four articles were included for qualitative synthesis. Overall risk of bias was “high.” Two studies did not show significant variations among 2.5% NaOCl solution and gel. Conclusion: As per rigorous investigation done, NaOCl solution proved more efficient against Enterococcus faecalis than its gel form. More research is needed to determine the best effective NaOCl type, concentration, duration, and treatment regimens.
Effect of Active Oxygen Fluid (Blue®m) as a Root Canal Irrigant Against Enterococcus Faecalis

Abstract:Purpose: To evaluate the antimicrobial effect of a new active oxygen fluid (Blue®m) as a root canal irrigant against Enterococcus faecalis compared to sodium hypochlorite (NaOCl). Material and methods: Forty-five extracted single-canaled human teeth were selected, received root canal preparation, autoclaved, and contaminated with Enterococcus faecalis. The specimens were randomly allocated into three groups: Group (A) served as the negative control, receiving irrigation with saline (n = 15); Group (B) was irrigated with 5.25% NaOCl (n = 15); and Group (C) was irrigated with 10 mL of Blue®m (n = 15). Microbial sampling from the root canals was performed before and after irrigation. The difference between the pre-irrigation and post-irrigation colony-forming units (CFU/mL) was calculated. The data was analysed using a one-way ANOVA followed by post-hoc Tukey tests. The significance level was set at 5%. Results: Blue®m statistically significantly reduced the bacterial load compared to saline (p = 0.009), but NaOCl was most effective, outperforming both (p 0.0001). Conclusion: Irrigation with Blue®m demonstrated antibacterial efficacy against Enterococcus faecalis, but it was not as effective as NaOCl.

Abstract:Background: As a very resistant microorganism in infected root canals, Enterococcus faecalis (E. faecalis) can stubbornly survive lethal challenges and invade dentinal tubules, making it the most persistent pathogen in root canal treatment. Enterococcus faecalis (E. faecalis) is the most commonly encountered microorganism detected in persistent root canal infections. These bacteria possess certain virulence factors, invade dentinal tubules and resist nutritional deprivation. Proper irrigation is an essential step for success in root canal therapies which is achieved by using excellent endodontic irrigants. Aim: To evaluate the antimicrobial efficacy of Sodium Hypochlorite (NaOCl), Silver Diamine Fluoride (SDF), Chitosan Nanoparticles (CNPs) root canal irrigants against the bacterial strain of Enterococcus Faecalis (E. faecalis) using agar well diffusion method. Materials and Methods: In this in-vitro study, the test materials were manipulated in accordance with the manufacturer’s instructions. The antimicrobial properties of root canal irrigants were evaluated by using agar diffusion method using bacterial strain of Enterococcus faecalis (ATCC 29212). 0.25 mL of each irrigant was placed on to 6.5 mm diameter blotting papers which were placed in 7 mm diameter wells on the Mueller Hinton agar plates. Later, E. faecalis strains were inoculated with sterile cotton swab on to the agar plates. Results: The p-value Sodium Hypochlorite showed the greatest zone of inhibition followed by SDF and Chitosan Nanoparticles respectively (p Sodium Hypochlorite showed the greatest zone of inhibition followed by SDF, Bioactive Glass Nanoparticles and Chitosan Nanoparticles respectively. Conclusion: Sodium Hypochlorite was the most effective root canal irrigant followed by SDF ,whereas Chitosan Nanoparticles was the least efficacious compared to the rest against Enterococcus Faecalis.

Abstract:Objectives: Enterococcus faecalis (E. faecalis), an infecting microorganism of the root canals, is difficult to eliminate during endodontic therapy. In this study, the effect of root canal disinfection with sodium hypochlorite (NaOCl) and chlorhexidine (CHX) was evaluated on planktonic and biofilm forms of E. faecalis in comparison with antimicrobial photodynamic therapy (aPDT) as an alternative strategy for root canal disinfection. Materials and Methods: In this study, E. faecalis (ATCC 29212) was used. The experimental procedures included aPDT with curcumin (CUR) and indocyanine green (ICG) as photosensitizers, irrigation with 5.25% NaOCl, 0.2% and 2.0% CHX solutions as traditional endodontic irrigating solutions, and the control group. The antibacterial and anti-biofilm potentials were assessed by counting the colony forming units and also using the crystal violet assay, respectively. Results: According to the results, E. faecalis biofilm was disrupted by 65.3%, 81.0% and 92.6% using 0.2% CHX, 2.0% CHX, and 5.25% NaOCl, respectively (P<0.05). In addition, CUR- and ICG-mediated aPDT displayed a significant reduction in E. faecalis count (90.2% and 82.5%, respectively) and its biofilm (83.6% and 75.2%, respectively) in comparison to the control group (P<0.05). Conclusions: APDT has a high potential for elimination of E. faecalis and is almost equivalent to NaOCl and CHX. It can be used as an adjucnt to conventional endodontic irrigating solutions.

Abstract:This study aimed to compare the antimicrobial effect of Nano-Silver Irrigant, Sodium Hypochlorite and Chlorohexidine against Enterococcus Faecalis bifilm. 60 Sixty, recently extracted, sound maxillary anterior teeth with completely formed apices and straight roots were selected in this study. Canals were instrumented using the Protaper Next system (Dentsply Sirona) 1 mm shorter from the apical foramen up to size X5 ( size# 50/06). Sterility of the teeth after complete root canal cleaning and shaping were achieved by autoclaving for 15 min at 121ºC with the teeth immersed in distilled water. Enterococcus Faecalis Biofilm was prepared by mixing 5 mL of the bacterial inoculum with sterilized BHI, then inoculated in teeth samples for 60 days. Teeth were then divided after the incubation period into three groups; 20 teeth each, and were treated by delivering the each of the tested irrigating solution using sterile plastic syringes. The first group was treated with Nano-Silver 0.02mol/L (2000ppm) while the second was treated using with Sodium Hypochlorite (NaOCl) 5.25% and the third was treated usinwith Chlorohexidine (CHX) 2.0%. Serial rinses ensured 5 minutes contact period between the aseptic solution and the bacteria. Sterile paper points size 50 were introduced into the canals and maintained for 3 min for sample collection, after which the paper points were placed in test tubes containing 2ml of sterile saline and several dilutions were prepared. 100 µL of each dilution was applied to the blood agar culture plates and incubated at 37ºC for 24 hours. A classical bacterial counting technique was used for each group after the treatment application for the recovery of viable E. faecalis on agar plates The results showed that the NaOCl had the highest antibacterial activity with high statistically significant difference with the other groups followed by the chlorhexidine group which also show statistically significant difference than the other two groups, then came the nano silver irrigant group which showed the lowest antibacterial activity among all the groups with statistically significant difference with them.

Abstract:Bacterial resistance to antibiotics is as old as the living planet, i.e. over 3 billion years ago when bacterial life emerged together with its biological warfare weapons, the antibiotics. Bacteria can readily develop a diverse array of antibiotic resistance mechanisms, including selection of mutants which produce modified low-affinity drug targets, exhibit reduced drug permeability or increased drug efflux. Another pathway is through acquisition of mobile genetic determinants of drug inactivating enzymes, target protection or bypass mechanisms. Antibiotic resistance has accelerated and spread across the community of human and animal bacteria following massive use of antibiotics for over half-a century in human and veterinary medicine and agriculture. Today, the increasing prevalence of resistance among many bacterial pathogens is no longer counter-balanced by the launching of innovative and active antibiotics. This imbalanced "arms race" undermines the escalating use of broad-spectrum antibiotics and increases the risk of therapeutic failure, drug side effets and excess costs of care. The accumulation of antibiotic resistance genes and their epidemic dissemination is fuelled by the selective pressure associated with excess and inappropriate use of antibiotics combined with insufficient infection prevention and control measures. Confronting this threat effectively requires new strategies to be implemented globally, nationally, and locally across all healthcare and agricultural sectors.
Comparative evaluation of the efficacy of 1% and 3% Sodium hypochlorite in reducing the microbial counts in primary teeth root canals using Bioluminometer - A randomized clinical trial

Abstract:Purpose: Sodium hypochlorite (NaOCl) is commonly used to irrigate primary teeth during pulpectomy. Although high concentrations of NaOCl are effective, they pose a risk of toxic damage to periapical tissues if NaOCl penetrates through the apical foramen. Therefore, low concentrations of NaOCl are preferred to mitigate this risk. However, concerns persist regarding the antibacterial efficacy of low concentrations of NaOCl compared to high concentrations. The objective of this study was to assess and compare the efficacy of 1% and 3% NaOCl irrigation in reducing bacterial load within primary teeth root canals. Materials and methods: This clinical study involved forty participants divided into two groups. Group 1 (n = 20) received canal irrigation with 1 % NaOCl solution, while Group 2 (n = 20) received canal irrigation with 3 % NaOCl solution. Microbial samples were collected from the root canal using a paper point before and after irrigation. The samples were aseptically transferred to ultra-snap tubes and then analyzed using a Bioluminometer. The results were recorded. Results: Both groups exhibited a decrease in bacterial count after irrigation. The mean colony count post irrigation for 3 % NaOCl was 258.05 ± 28.61, and for 1 % NaOCl it was 267.60 ± 30.56. However, no statistically significant difference was observed upon intergroup comparison. Conclusion: This study shows that 1% NaOCl is equally effective as 3% NaOCl in reducing bacterial count in root canals. Thus, using 1% NaOCl as an irrigant is appropriate in clinical practice.

Abstract:Aim: To compare shock wave-enhanced emission photoacoustic streaming (SWEEPS) with sonic- and ultrasonically activated irrigation systems in removing Enterococcus faecalis biofilm from the root canal system. Methodology: Fifty human single-canalled mandibular premolars were included in the study. After access cavity preparation, the root canals were prepared to a standardized size and taper. Then, the entire root surface was covered with two layers of resin, and the root apices were sealed before sterilization. All root canals were inoculated with E. faecalis biofilm, and the samples were incubated aerobically for 2 weeks at 37 °C. Biofilm formation was confirmed by scanning electron microscopy. All samples were randomly divided into five groups (n = 10 each) based on their irrigation activation method as A (no treatment or negative control), B (no irrigation or positive control), C (sonically activated irrigation (SAI)), D (ultrasonically activated irrigation (UAI)), and E (needle irrigation activated by an Er: YAG laser device using a SWEEPS quartz tip (SWEEPS)). Then, dentine chips were retrieved, vortexed, and diluted for colony-forming unit counts. Data were analysed using analysis of variance and post-hoc Tukey tests (α = 5%). Results: All methods could significantly reduce E. faecalis biofilm compared with control so that the UAI, SWEEPS, and SAI groups indicated a 23.54%, 14.89%, and 7.81% biofilm reduction, respectively. UAI demonstrated a significantly more effective reduction of E. faecalis biofilm than SAI (p = 0.004). Conclusions: All irrigation activation methods significantly reduced E. faecalis biofilm, with ultrasonic use being the most effective.

Abstract:Objective: This study was done to evaluate the antibacterial effect of chitosan nano-particles (CNPs) root canal irrigant as a new alternative to Sodium hypochlorite (NaOCl) for disinfection of root canals inoculated with Enterococcus faecalis, with and without laser activation.Methodology: Sixty single rooted human premolars were decoronated, prepared and had their apical foramina sealed. E. faecalis were incubated in the root canals for 15 days. The teeth were then randomly divided into two experimental groups (n = 30) according to the disinfection protocol used. In Group I: disinfection was performed using the irrigant solutions only (Saline, 2.5% NaOCl, CNPs). Whereas in Group II, disinfection was done using the same irrigants followed by Diode laser at (980-nm) at 2 W output for 5 × 5 s. Intra-canal bacterial samples were taken before and after canal disinfection to determine the CFU count.Results: In group I, 2.5% NaOCl was as effective as CNP in eradication and significantly more effective than Saline (p = 0.008) in eradication of E. faecalis. In Group II, either 2.5% NaOCl or CNP in combination with diode laser irradiation showed a similarly high effect in bacterial eradication.Conclusions: Within the parameters used in this study, a combination therapy consisting of irrigation followed by diode laser irradiation should be utilized as an effective treatment modality for eliminating E. faecalis from root canal systems.

Abstract:Among black-tailed deer (Odocoileus hemionus sitkensis), roe deer (Capreolus capreolus), and reindeer (Rangifer tarandus), nitrogen levels in the contents of the rumen show consistent patterns of variation. Among deer of each species on the same range during the same season, the animal-derived nitrogen component of the rumen contents decreases as the vegetative nitrogen component increases. This inverse relationship may result from one, or more likely both, of the following causes. (1) The ability of the ruminant to maintain a fairly constant rumen environment to allow for its most efficient function. (2) An adaptability in these deer species to compensate for low nitrogen levels in the forage by recycling of nitrogen through the saliva and further conservation of ruminal nitrogen by recycling it through successive generations of microbial populations.
