Abstract:Biodegradable injectable hydrogels have been extensively studied and evaluated in various medical applications such as for bulking agents, drug delivery reservoirs, temporary barriers, adhesives, and cell delivery matrices. Where injectable hydrogels are intended to facilitate a healing response, it may be desirable to encourage rapid cellular infiltration into the hydrogel volume from the tissue surrounding the injection site. In this study, we developed a platform technique to rapidly form pores in a thermally responsive injectable hydrogel, poly(NIPAAm-co-VP-co-MAPLA) by using mannitol particles as porogens. In a rat hindlimb muscle injection model, hydrogels incorporating porosity had significantly accelerated cellular infiltration. To influence the inflammatory response to the injected hydrogel, enzymatically digested urinary bladder matrix (UBM) was mixed with the solubilized hydrogel. The presence of UBM was associated with greater polarization of the recruited macrophage population to the M2 phenotype, indicating a more constructive foreign body response. The hybrid hydrogel positively affected the wound healing outcomes of defects in rabbit adipose tissue with negligible inflammation and fibrosis, whereas scar formation and chronic inflammation were observed with autotransplantation and in saline injected groups. These results demonstrate the value of combining the effects of promoting cell infiltration and mediating the foreign body response for improved biomaterials options soft tissue defect filling applications. STATEMENT OF SIGNIFICANCE:Our objective was to develop a fabrication process to create porous injectable hydrogels incorporating decellularized tissue digest material. This new hydrogel material was expected to exhibit faster cellular infiltration and a greater extent of pro-M2 macrophage polarization compared to control groups not incorporating each of the functional components. Poly(NIPAAm-co-VP-co-MAPLA) was chosen as the representative thermoresponsive hydrogel, and mannitol particles and digested urinary bladder matrix (UBM) were selected as the porogen and the bioactive decellularized material components respectively. In rat hindlimb intramuscular injection models, this new hydrogel material induced more rapid cellular infiltration and a greater extent of M2 macrophage polarization compared to control groups not incorporating all of the functional components. Abstract:Synthetic nanofibrillar hydrogels resembling a natural extracellular matrix have emerged as a promising material in the biomedical field; however, achievement of multifunctional nanofibrillar hydrogels with good biocompatibility is still an important and challenging task. Herein, we report a fluorescent nanofibrillar hydrogel prepared with polyethyleneimine (PEI) and cellulose nanocrystals (CNCs) via one-step hydrothermal treatment. The nanofibrillar hydrogels are mainly formed from CNCs cross-linked by carbon dots (CDs) through hydrogen bonding and hydrophobic interactions. The nanostructures and the mechanical properties of the hydrogels are controlled by weight ratios of CNCs to PEI and the total concentration of CNCs and PEI. In comparison with other hydrogels from CNCs and CDs, the nanofibrillar hydrogels exhibit better mechanical properties with relatively high Young's moduli. The hydrogels show relatively low cytotoxicity and good biocompatibility, which are demonstrated by in vitro cell culture and in vivo subcutaneous implantation in rats. Abstract:Gelatine is a biocompatible and natural polymer with chemical properties similar to the extracellular matrix. However, it has poor mechanical properties and sensitive to enzymatic biodegradation that limits its application in 3D scaffold fabrication. Cellulose nanofibrous (CNF) offers biocompatibility, high surface area and excellent mechanical properties with slow in-vivo degradation. To fine tune their properties, CNF, and gelatine (CNF-GEL) were blended to form biocomposite aerogels. Epichlorohydrin (EPH) was incorporated into CNF-GEL as a chemical crosslinker to investigate its effect on the physiochemical, mechanical, and biological properties of the biocomposite aerogels both in-vitro and in-vivo. Regardless of the composition of the prepared aerogels, they possessed porosity of >90% with the pore size of 7-135 mu m, which was confirmed in the morphological analysis. The presence of EPH improved the chemical interaction between CNF and gelatine, hence enhanced the compressive strength compared to uncrosslinked samples. The formulation of crosslinked CNF-GEL 90:10 offered the highest compressive strength of 61.35 kPa. The in-vitro and in-vivo studies showed adequate cytocompatibility, cell viability and cell attachment in the optimal crosslinked formulation with tuned enzymatic degradation. Antimicrobial property was also achieved in the optimal scaffold by incorporating curcumin as an antimicrobial agent. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:A theoretical treatment of transient grating diffraction is derived for gratings that are spatially nonuniform in the direction perpendicular to the sample surface. This treatment is readily generalized to any four-wave mixing experiment. Both reflection and transmission geometries of diffraction are examined for the standard transient grating case, in which both grating excitation beams are incident upon the same side of the sample For samples in which the grating amplitude perpendicular to the sample surface varies slowly relative to the optical wavelength, the reflection geometry is shown to probe only the surface or the interface, while the transmission geometry probes the bulk of the sample. An experimental example using four transient grating geometries (two reflection, two transmission) is shown to yield significantly different temporal responses, illustrating the nature of the theoretical predictions. Abstract:Increasing demand for long-distance high-power transmission and advances in Voltage Source Converter technology create a case for embedded VSC-HVDC transmission solutions. Numerous aspects have to be considered in order to fully understand the influence of embedded VSC-HVDC links on inter-area transfer capability. This paper focuses on the influence of embedded VSC-HVDC on power system stability. Maximum transfer capability of a long corridor connecting two synchronized areas and subjected to a permanent fault is used as an indicator to tune the design and operation parameters of the HVDC link and provide a comparison with alternative AC reinforcement of the corridor. The investigation is carried out first on a simple radial test system and then up-scaled to a case study on the modified IEEE 39-bus test system. The study is performed using generic VSC controller design and positive-sequence RMS simulations with the software DIgSILENT PowerFactory.

Abstract:With excellent biocompatibility and unique physiochemical properties,nanocelluloses including cellu-lose nanocrystals(CNCs)and cellulose nanofibrils(CNFs)are promising candidates for preparing biomed-ical hydrogels.CNCs and CNFs are different in morphology and surface charges.Herein,CNCs and two CNFs(CNFs-C,Carboxylated CNFs;CNFs-P,Phosphorylated CNFs)were synthesized and applied to fabri-cate hydrogels through metal crosslinking.Aluminum crosslinking was found to be the best choice for enhancing the strength.This study systematically compared the morphologies,storage modulus,loss fac-tor,continuous shear ramp,self-healing,swelling,in vitro degradation and injectable properties of the fabricated hydrogels.Further,a radar chart is summarized as guidelines to direct the rational selection to meet the specific requirements of further biomedical applications.At the same nanocellulose concen-tration and after Al3+crosslinking,CNCs hydrogels had strong water holding capacity twice as much as that of CNFs hydrogels.While CNFs hydrogels showed higher hardness and stronger resistance to degra-dation than that of CNCs.These results provide detailed insights into nanocellulose hydrogels,making it possible to use these guidelines to select hydrogels for desired performance.

Abstract:Biopolymer hydrogels are an attractive class of materials for wound dressings and other biomedical applications because of their ease of use and availability from biomass. Here, we present a hydrogel formation approach based on alginate and chitosan. Alginate is conventionally cross-linked using multivalent ions such as Ca2+ but in principle any polycationic species can be used such as polyelectrolytes. Exchanging the cross-linking Ca2+ ions partially with chitosan, which at pH 7 has available positive charges as well as good interactions with Ca2+, leads to an improved Young's modulus. This gel is non-toxic to mammalian cells and hence allows conveniently for stem cell encapsulation since it is based on two-component mixing and gel formation. Additionally, the chitosan is known to have a bactericidal effect which is retained when using it in the alginate⁻chitosan gel formation and the formed hydrogels displayed bactericidal effects against P. aeruginosa and S. aureus. The combination of anti-bacterial properties, inclusion of stem cells, and the hydrogel nature would provide an ideal environment for complex wound healing.

Abstract:Tissue defects were treated by invasive processes such as implanting the scaffold to the defected area. Hydrogel scaffolds with the well-defined internal structure are significant for tissue engineering. In this work, preparation and characterization of hydrogels based on collagen and modified nanocrystalline cellulose (CNCs) have been reported. The surface morphology of hydrogels with different ratios of CNCs has been determined. The effects of polymer ratio on the resulting hydrogels, such as pore morphology, swelling ratio, degradation rate and mechanical properties were examined for a possible non-aggressive tissue engineering application. Subsequently, studies on cytotoxicity of hydrogels to fibroblasts have been defined.

Abstract:Nanocellulose has been highlighted as one of the most promising biomaterials for biomedical applications with the potential to outperform conventional polymeric materials. However, the design of nanocellulose-based biomaterials for wound healing still requires precise control over biophysical and biochemical cues to direct a series of cellular activities in healing processes. The bioactive cellulose nanofibrils scaffolds with controlled release of basic fibroblast growth factors (bFGFs) were constructed for potential wound healing applications. The Quartz Crystal Microbalance with Dissipation monitoring (QCM-D) analysis revealed the polyion complex interaction between the positively charged bFGFs with the negatively charged cellulose nanofibrils, which mimics the interaction between bFGFs and heparin sulfate in extracellular matrix in the body. Such association enables not only the storage of bFGFs in a readily available form and from where it is slowly released, but also potential protection of it from denaturation. The release profile of bFGFs from the CNF scaffolds was tailored by tuning the CNF surface chemistry and in situ deconstruction of the scaffolds. The in situ enzymatic deconstruction of the scaffolds provides a possibility to tune the bioavailability of bFGFs for cell growth and proliferation, and more importantly, to balance the scaffolds degradation and new tissue formation in wound healing. The MTT assay of cells proliferation and fluorescence imaging of cells cultured in the 3D scaffolds showed that the CNF scaffolds loaded with bFGF can significantly facilitate the proliferation of the cells, even if only a small amount of bFGF was loaded. Enzymatic deconstruction of the CNFs network further increases the bFGFs bioavailability, and promotes cell proliferation. This work may serve as an important step toward the development of nanocellulose-based biomaterials with tailored biophysical and biochemical cues for wound healing and other biomedical applications.

Abstract:Gelatin (GA), hyaluronic acid (HA) and cellulose nanocrystals (CNC) are promising materials for skin wound care. In this study the GA-HA-CNC hydrogels were prepared by cross-linking and freeze-drying. The composition and mechanism of GA-HA-CNC hydrogels were confirmed by FTIR. The morphology and pore size were obtained by SEM. We accessed the physical property from rheological results and swelling ratio. NIH-3T3 cells were inoculated into the hydrogels and cultured for different days, then we analyzed the cytotoxicity of the prepared hydrogels by CCK-8 methods and live/dead pictorial diagram using staining kits. FTIR revealed the combination between GA, HA and CNC was attributed to the amide bond and hydrogen bonding. SEM results showed that the drying GA-HA-CNC hydrogels were spongy, with the pore diameter about 80–120 µm. CNC significantly enhanced the property of the hydrogels and play a vital role according to the rheology and swelling results. The cells culture results showed that NIH-3T3 cells can attached to, grow, and proliferate well on the GA-HA-CNC hydrogels. In conclusion, the natural GA-HA-CNC hydrogel has great potential for the skin wound repair.

Abstract:Hydrogels based on natural, biodegradable materials have gained considerable interest in the medical field due to their improved drug delivery profiles and tissue-mimicking architecture. In this regard, this study was devoted to the preparation and characterization of new physically crosslinked hydrogels based on carboxymethyl cellulose and an unconventional crosslinking agent, phytic acid. Phytic acid, in addition to its antioxidant and antibacterial effects, can improve the biological properties and stability of gels, without adding toxicity. Fourier transform infrared (FTIR) spectroscopy, rheological studies and thermal analysis confirmed the hydrogel formation. The influence of the ratio between the cellulose derivative and the crosslinker upon the morphological structure and water uptake was evidenced by scanning electron microscopy (SEM) and swelling measurements in simulated body fluids. Furthermore, procaine was entrapped within the hydrogels and used as a model drug for in vitro studies, which highlighted the dependence of the drug release on the phytic acid content of the matrix. The materials demonstrated antibacterial effects against Escherichia coli and Staphylococcus aureus bacteria. The biocompatibility was assessed on fibroblast cells, and according to our results, hydrogels can improve cell viability highlighting the potential of these systems as therapeutic scaffolds for skin tissue engineering.

Abstract:Purpose: To determine the clinical characteristics and outcome of bacterial meningitis in adult patients using CT, MRI and CSF culture. Methods: Patients admitted to the university hospital with clinical diagnosis of meningitis were included in the present study. All patients were subjected to clinical examination as well as CT, MRI scanning and CSF-bacteriological culture. Results: The age distribution of the patients shows that there is high frequency of symptoms found in the age group between ages 40 - 45. CSF in the present study included WBS, RBC, glucose and protein, which is associated with the adverse outcome. Meningitis can be confirmed by many methods, in the present study three different methods were used, which included CT scan, MRI and CSF analysis and bacterial culture. In CT scan, among the predictors of meningitis, persons with TB (P = 0.037), patients with HIV (P = 0.036) and stroke (P = 0.036) showed significant association. Abstract:Background: Cellulose Nanofibrils (CNFs) are natural nanomaterials with nanometer dimensions. Compared with ordinary cellulose, CNFs own good mechanical properties, large specific surface areas, high Young's modulus, strong hydrophilicity and other distinguishing characteristics, which make them widely used in many fields. This review aims to introduce the preparation of CNFs-based hydrogels and their recent biomedical application advances. Methods: By searching the recent literatures, we have summarized the preparation methods of CNFs, including mechanical methods and chemical mechanical methods, and also introduced the fabrication methods of CNFs-based hydrogels, including CNFs cross-linked with metal ion and with polymers. In addition, we have summarized the biomedical applications of CNFs-based hydrogels, including scaffold materials and wound dressings. Results: CNFs-based hydrogels are new types of materials that are non-toxic and display a certain mechanical strength. In the tissue scaffold application, they can provide a micro-environment for the damaged tissue to repair and regenerate it. In wound dressing applications, it can fit the wound surface and protect the wound from the external environment, thereby effectively promoting the healing of skin tissue. Conclusion: By summarizing the preparation and application of CNFs-based hydrogels, we have analyzed and forecasted their development trends. At present, the research of CNFs-based hydrogels is still in the laboratory stage. It needs further exploration to be applied in practice. The development of medical hydrogels with high mechanical properties and biocompatibility still poses significant challenges.

Abstract:Advanced wound dressings improve wound healing by releasing antibacterial agents, accelerating wound closure, and reporting (sensing) changes in the wound's state. The challenge with the release of antibacterial agents such as drugs, peptides, or nanoparticles is their unregulated administration. In addition, bacteria resistance to antibiotics stimulates the search for new types of antibacterial wound dressings. Here, we report a new approach to antibacterial wound dressings by utilizing a nanocolloidal hydrogel with strong Fe3+ ion sequestration capability, thus depriving bacteria of much-needed ionic iron and suppressing bacteria growth. The hydrogel was derived from cellulose nanocrystals decorated with carbon dots (C-dot/CNCs). Upon Fe3+ ion uptake by the nanofibrillar hydrogel, the photoluminescence of the hydrogel was quenched, due to adsorption of ions to the C-dot surface, thus reporting on the removal of ionic iron from the medium. The hydrogel suppressed the growth of antibiotic-resistant Gram-negative Escherichia coli, antibiotic-resistant Pseudomonas aeruginosa, and Gram-positive Staphylococcus aureus and was noncytotoxic for human fibroblasts. Wound dressings were readily fabricated using three-dimensional (3D) printing. Abstract:Acute wounds subject to frequent deformations are difficult to be treated because the healing process was easily interfered by external mechanical forces. Traditional wound dressings have limited efficacy because of their poor mechanical properties and skin adhesiveness and difficulty in the delivery of therapeutic drugs effectively. As such, tough and skin-adhesive wound dressings with sustainable and stimuli-responsive drug release properties for treatment of those wounds are highly desirable. For this purpose, we have developed a mechano-responsive poly(sulfobetaine methacrylate) hydrogel which aims to control the delivery of antibiotic drug upon application of mechanical forces. Diacrylated Pluronic F127 micelles were used as a macro-cross-linker of the hydrogel and loaded with hydrophobic antimicrobial drugs. The micelle-cross-linked hydrogel has excellent mechanical properties, with the ultimate tensile strength and tensile strain of up to 112 kPa and 1420%, respectively, and compressive stress of up to 1.41 MPa. Adhesiveness of the hydrogel to the skin tissue was ∼6 kPa, and it did not decrease significantly after repetitive adhesion cycles. Protein adsorption on the hydrogel was significantly inhibited compared to that on commercial wound dressings. Because of the mechano-responsive deformation of micelles, the release of drug from the hydrogel could be precisely controlled by the extent and cycles of mechanical loading and unloading, endowing the hydrogel with superior antibacterial property against both Gram-positive and Gram-negative bacteria. In addition, drug penetration into the skin tissue was enhanced by mechanical stress applied to the hydrogel. The micelle-cross-linked zwitterionic hydrogel also showed good cell biocompatibility, negligible skin irritation, and healing capacity to acute skin wounds in mice. A 3D bioprinted antibacterial hydrogel dressing of gelatin/sodium alginate loaded with ciprofloxacin hydrochloride

Abstract:Skin plays a crucial role in human physiological functions, however, it was vulnerable to bacterial infection which delayed wound healing. Nowadays, designing an individual wound dressing with good biocompatibility and sustaining anti-infection capability for healing of chronic wounds are still challenging. In this study, various concentrations of the ciprofloxacin (CIP) were mixed with gelatine (Gel)/sodium alginate (SA) solution to prepare Gel/SA/CIP (GAC) bioinks, following the fabrication of GAC scaffold by an extrusion 3D bioprinting technology. The results showed that the GAC bioinks had good printability and the printed GAC scaffolds double-crosslinked by EDC/NHS and CaCl2 had rich porous structure with appropriate pore size, which were conducive to drug release and cell growth. It demonstrated that the CIP could be rapidly released by 70% in 5 min, which endowed the GAC composite scaffolds with an excellent antibacterial ability. Especially, the antibacterial activities of GAC7.5 against Escherichia coli and Staphylococcus aureus within 24 h were even close to 100%, and the inhibition zones were still maintained 14.78 ± 0.40 mm and 14.78 ± 0.40 mm, respectively, after 24 h. Meanwhile, GAC7.5 also demonstrated impressive biocompatibility which can promote the growth and migration of L929 and accelerate wound healing. Overall, the GAC7.5 3D bioprinting scaffold could be used as a potential skin dressing for susceptible wounds with excellent antibacterial activity and good biocompatibility to meet urgent clinical needs.
In vitro toxicity assessment of hydrogel patches obtained by cation-induced cross-linking of rod-like cellulose nanocrystals

Abstract:With the purpose of designing active patches for photodynamic therapy of melanoma, transparent and soft hydrogel membranes (HMs) have been fabricated by cation-induced gelation of rod-like cellulose nanocrystals (CNCs) bearing negatively charged carboxylic groups. Na+ , Ca2+ , Mg2+ have been used as cross-linkers of cellulose nanocrystal (CNC). The biosafety of this material and of its precursors has been evaluated in vitro in cell cultures. Morphological changes, cell organelles integrity, and cell survival with the tetrazolium salt reduction (MTT) assay were utilized as tests of cytotoxicity. Preliminary investigation was performed by addition of the hydrogel components to the cell culture medium and by incubations of the CNC-HM in direct and indirect contact with a confluent monolayer of A375 melanoma cells. Direct contact assays suffered from interference of physical stress. Careful evaluation of cytotoxicity was obtained considering the overall picture provided by microscopy and biochemical tests performed with the CNC-HM in indirect contact with two melanoma cell lines (A375, M14) and human fibroblasts. CNCs have been demonstrated to be a safe precursor material and CNC-HMs have a good biocompatibility provided that the excess of cations, in particular of Ca2+ is removed. These results indicate that CNC and can be safely used to fabricate biomedical devices such as transparent hydrogel patches, although attention must be paid to the fabrication procedure.
Grid computing of spatial statistics: using the TeraGrid for G i* (d) analysis

Abstract:The massive quantities of geographic information that are collected by modern sensing technologies are difficult to use and understand without data reduction methods that summarize distributions and report salient trends. Statistical analyses, therefore, are increasingly being used to analyze large geographic data sets over a broad spectrum of spatial and temporal scales. Computational Grids coordinate the use of distributed computational resources to form a large virtual supercomputer that can be applied to solve computationally intensive problems in science, engineering, and commerce. This paper presents a solution to computing a spatial statistic, G i* (d) using Grids. Our approach is based on a quadtree‐based domain decomposition that uses task‐scheduling algorithms based on GridShell and Condor. Computational experiments carried out on the TeraGrid were designed to evaluate the performance of solution processes. The Grid‐based approach to computing values for G i* (d) shows improved performance over the sequential algorithm while also solving larger problem sizes. The solution demonstrated not only advances knowledge about the application of the Grid in spatial statistics applications but also provides insights into the design of Grid middleware for other computationally intensive applications. On Low-Concentration Inks Formulated by Nanocellulose Assisted with Gelatin Methacrylate (GelMA) for 3D Printing toward Wound Healing Application

Abstract:Cellulose nanofibrils (CNFs) in the form of hydrogels stand out as a platform biomaterial in bioink formulation for 3D printing because of their low cytotoxicity and structural similarity to extracellular matrices. In the present study, 3D scaffolds were successfully printed with low-concentration inks formulated by 1 w/v % 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO)-oxidized CNF with less than 1 w/v % gelatin methacrylate (GelMA). Quartz crystal microbalance with dissipation monitoring (QCM-D) measurements showed strong interaction between the two biopolymers. The UV cross-linking ability of GelMA (≤1 w/v %) was enhanced in the presence of TEMPO-oxidized CNFs. Multiple factors including strong physical interaction between CNF and GelMA, in situ cross-linking of CNF by Ca2+, and UV cross-linking of GelMA enabled successful 3D printing of low-concentration inks of CNF/GelMA into scaffolds possessing good structural stability. The mechanical strength of the scaffolds was tuned in the range of 2.5 to 5 kPa. The cell culture with 3T3 fibroblasts revealed noncytotoxic and biocompatible features for the formulated inks and printed scaffolds. More importantly, the incorporated GelMA in the CNF hydrogel promoted the proliferation of fibroblasts. The developed low-concentration CNF/GelMA formulations with a facile yet effective approach to fabricate scaffolds showed great potential in 3D printing for wound healing application.

Abstract:Electrospun nanofibrous membranes of natural polymers, such as gelatin, are fundamental in the design of regenerative devices. Crosslinking of electrospun fibres from gelatin is required to prevent dissolution in water, to retain the original nanofibre morphology after immersion in water, and to improve the thermal and mechanical properties, although this is still challenging to accomplish in a controlled fashion. In this study, we have investigated the scalable manufacture and structural stability in aqueous environment of a UV-cured nanofibrous membrane fabricated by free surface electrospinning (FSES) of aqueous solutions containing vinylbenzylated gelatin and poly(epsilon-caprolactone) dimethacrylate (PCL-DMA). Vinylbenzylated gelatin was obtained via chemical functionalisation with photopolymerisable 4-vinylbenzyl chloride (4VBC) groups, so that the gelatin and PCL phase in electrospun fibres were integrated in a covalent UV-cured co-network at the molecular scale, rather than being simply physically mixed. UV-cured nanofibrous membranes did not dissolve in water and showed enhanced thermal and mechanical properties, with respect to as-spun samples, indicating the effectiveness of the photo-crosslinking reaction. In addition, UV-cured gelatin/PCL membranes displayed increased structural stability in water with respect to PCL-free samples and were highly tolerated by G292 osteosarcoma cells. These results therefore support the use of PCL DMA as hydrophobic, biodegradable crosslinker and provide new insight on the scalable design of water insoluble, mechanical competent gelatin membranes for healthcare applications.
