Abstract:Economy of Pakistan is heavily dependent upon agriculture and extensive use of pesticide is quiet common to enhance the crop yield. Imidacloprid is among the first choice pesticides in Pakistan and it has been reported that through run off along with water it ends up in water bodies affecting non target aquatic fauna. Through the present investigation, we are reporting the effects of Imidacloprid on the fatty acids composition of a non-target, commercially important carp: Labeo rohita. Fish were exposed to sub lethal concentration of Imidacloprid (120 mgL1) for 2, 4 and 8 days (short term) as well as for 16, 32 and 64 days (long term experimental conditions). Pesticide untreated controls were also maintained for each treatment. Following the specific Imidacloprid exposure, fatty acid composition (%) was determined in the muscle of all experimental groups by using gas chromatography. Fish exposed to Imidacloprid for 8 days had reduced Palmitic acid (p = 0.02) and elevated muscle Arachidic acid (p < 0.001) than control group. Labeo rohita exposed to the pesticide for 32 days had elevated muscle Oleic (p = 0.02) and Linoleic acid (p = 0.02) while fish exposed to Imidacloprid to 64 days had reduced muscle Palmitic (p = 0.04) and Oleic acid (p = 0.03). In conclusion, we are reporting that the exposure to sub lethal concentration of Imidacloprid disturb the muscle fatty acid composition of Labeo rohita that may affect its food quality. The effects were more pronounced under long term experimental conditions and were probably due to potentiating lipid peroxidation and disturbed fish metabolism upon Imidacloprid exposure.
Short- and Long-Term Exposure to Imidacloprid Disturbs the Elemental Composition and Free Amino Acid Profile in Muscles of Labeo Rohita

Abstract:Imidacloprid [1-(6-chloro-3-pyridylmethyl)-N-nitro-imidazolidin-2-ylideneamine] is among the most extensively used pesticide in Pakistan. The aim of this investigation was to report the changes in concentration of different elements and amino acid in the muscle of Labeo rohita upon imidacloprid exposure. Juvenile Labeo rohita (N = 480) were treated with 120mgL−1 imidacloprid under short and long-term experimental conditions. At the end of each experiment, fish muscle was used for elemental concentration (determination by an atomic absorption spectrophotometer) and amino acid estimation (by high-performance liquid chromatography (HPLC)). Our results indicated that sodium, potassium, calcium, magnesium, iron, aluminum, and lead concentrations were significantly different (P < 0.05) when compared between imidacloprid-treated and untreated L. rohita and changes were more pronounced in short-term experimental groups. HPLC-generated data analysis showed that proline, isoleucine, and glycine concentrations increased while the levels of phenylalanine, alanine, and leucine decreased significantly (P < 0.05) in muscles of fish exposed to imidacloprid under short-term conditions. On the other hand, serine, arginine, and glycine increased significantly and alanine decreased significantly in L. rohita exposed to pesticide under long-term experimental conditions. It is concluded that imidacloprid (120 mg L−1) exposure to L. rohita for a variable duration has significantly changed the elemental composition and studied amino acid concentrations in fish muscles making it unsuitable for human consumption.

Abstract:Pyriproxyfen (PPF) mimics a natural hormone in insects and disrupts their growth. It is a well-known synthetic insecticide and aromatic juvenile hormone analog frequently used in agriculture and vegetable crops to control various insect species. At present, scanty information is available about the possible potential threats of PPF in aquatic organisms. Therefore, in this study, different toxico-pathologic endpoints of PPF like DNA damage, biomarkers of oxidative stress, and status of antioxidant enzymes were determined in Labeo rohita (freshwater fish). In our study, 60 active, free from any external obvious ailments, same size, age, and body mass were randomly allocated to four glass aquaria (T0-T3) separately containing 100 L water. The fish present in groups T1, T2, and T3 were administered PPF dissolved in water 300, 600, and 900 μg/L for 30 days. Different tissues including the blood and visceral organs were obtained from each fish on days 10, 20, and 30 of the experiment. Results on various morphological and nuclear changes in red blood cells of PPF-exposed Labeo rohita fish including pear-shaped erythrocytes, spherocytes, red blood cells with a blebbed nucleus, micronucleus, and nuclear remnants were significantly increased. Our results on genotoxicity (comet assay) recorded significantly ( P ≤ 0.05 ) increased DNA damage in various tissues of insecticide-exposed fish. The results on oxidative stress profile (reactive oxygen species and thiobarbituric acid reactive substances) and antioxidant enzymes (reduced glutathione superoxide dismutase, peroxidase, and catalase) in multiple tissues of Labeo rohita fish concluded significantly ( P ≤ 0.05 ) higher quantity of biomarkers of oxidative stress and lower concentrations of different antioxidant enzymes in treated fish. Hence, the findings of our experimental research determine that PPF could induce adverse toxic impacts on multiple tissues of Labeo rohita fish.

Abstract:Fishes have been widely used as a representative to estimate the health of an aquatic ecosystem. In the present study, Labeo rohita was selected for biomarker study against decabromodiphenyl ether (BDE-209), a persistent organic pollutant (POP), as it is a widely used Indian carp. The results suggested significant effects on the optimum metabolism of Labeo rohita . After 48 to 72 h of exposure, most of the biomarkers such as lactate dehydrogenase (LDH), creatine kinase (CK), serum glutamic pyruvic transaminase (SGPT), serum glutamic oxaloacetic transaminase (SGOT), and hepatosomatic index (HSI) increased drastically indicating the higher index of tissue and liver damage. On the contrary, succinic dehydrogenase (SDH), acetylcholinesterase (AChE), and alkaline phosphatase (ALP) showed a reverse trend suggesting the shifting of fish metabolism towards anaerobic respiration mode because of induced stress. Increased catalase (CAT) activity was also observed, which indicated increased abundance of reactive hydroxyl species and therefore a possible oxidative stress in fishes. It is further suggested to understand and examine the biotransformation characteristics and degradation pathways of polybrominated diphenyl ether (PBDE)s, which would be useful to comprehend their environmental fate.

Abstract:λ-cyhalothrin, a synthetic type II pyrethroid, has become increasingly popular for control of aphids, butterfly larvae, and beetles, replacing other agricultural chemicals. As a result of which, residues of this synthetic pesticide are being reported across the globe in natural water, which poses a serious threat to aquatic life. Therefore, the present study was designed to understand the toxicity effects of λ-cyhalothrin on behaviour, oxidative stress and neurotoxicity in a vertebrate aquatic model, zebrafish ( Danio rerio ). The fish were exposed to 0.129, 0.194 and 0.388 μg/L corresponding to 5%, 10% and 20% of 96hLC 50 (1.94 μg/L) for 28 days. Upon exposure to the highest concentration (0.388 μg/L), the test animal exhibited significant alterations in behavioural patterns like number of entries to the top zone (n), decrease in average speed (m/s) and decrease in time spent in top zone (s). Moreover, the shoaling test demonstrated a significant decrease ( p < 0.05) in the relative time spent by the tested fish (%) near the stimulus fish. The change in behavioural alterations might be linked to a significant decrease ( p < 0.05) in the brain acetylcholine esterase activity. Furthermore, the present study also illustrates oxidative stress exerted by λ-cyhalothrin through an increase in the production of reactive oxygen species, which is again clearly depicted by a significant increase ( p < 0.05) in Superoxide dismutase, Catalase and Glutathione peroxidase activities. Overall, the present study systematically demonstrates the chronic effects of λ-cyhalothrin on adult fish behaviour and physiology, which will contribute to assessing the risks of λ-cyhalothrin to organismal health.

Abstract:The present study used scanning electron microscopy (SEM) to assess the toxicity of sub-lethal concentrations of deltamethrin (0.035, 0.007 and 0.0007 mg L −1 ) and permethrin (0.93, 0.093 and 0.0093 mg L −1 ) on the ultrastructure of the scales of Anabas testudineus (Bloch, 1792) during a 21 day exposure. The oxygen uptake of the fish during deltamethrin (0.007 and 0.0007 mg L −1 ) and permethrin (0.093 and 0.0093 mg L −1 ) exposure was also investigated. The SEM studies revealed abnormal morphological alterations and modifications of fish scales, which were concentration-dependent. Deltamethrin-exposed fish showed severe deformation and fusion of two circuli in different rows, thereby disrupting the normal radii pattern. This fusion was probably caused by the cyanide moiety of the pesticide. On the other hand, permethrin characteristically produced a thick mucus layer over the scale surface. The different concentrations of both the pesticides affected circuli pattern with severe breakage of circuli and loss of lepidonts present over their ridges. Erythrocyte extrusions were also seen at several places over the scale surface. There was significant reduction of oxygen uptake in fish exposed to permethrin at both the concentrations, but in deltamethrin treatments significant reduction occurred only at the higher concentration of 0.007 mg L −1 , though this was less than the lower permethrin concentration of 0.0093 mg L −1 , and moreover effects of both the pyrethroids on oxygen consumption increased at higher concentrations.

Multiple biomarkers based appraisal of deltamethrin induced toxicity in silver carp (Hypophthalmichthys molitrix)

Abstract:Deltamethrin (DLM) is α-cyano (type II) synthetic pyrethroid. DLM exposure leads to strong neurotoxic effects and a number of complex toxicological syndromes. The current study assessed DLM mediated oxidative stress, behavioral, hematological, histopathological, and biochemical toxic effects on silver carp (Hypophthalmichthys molitrix). Exposure to an acute concentration (2 μg/L) of DLM resulted in different behavioral inconsistencies and a time-dependent significant (P < 0.05) change in the hematology and serum biochemistry of silver carp. A significant (P < 0.05) increase in the activities of reactive oxygen species, lipid peroxidation, and antioxidant enzymes whereas a significant decrease in total protein contents in the liver, gills, brain, and muscle tissues were observed. DLM exposure increased the activities of metabolic enzymes in the gills, muscles, and liver of silver carp. A significant (P < 0.05) increase in DNA damage in peripheral blood erythrocytes was evident. DLM exposure led to a time-dependent significant (P < 0.05) increase in the whole-body cortisol and blood glucose level, while a significant decrease in acetylcholine esterase activity in the brain, liver, and muscle tissues. Different histopathological changes in the liver, gills, brain, and intestine were observed, however, no significant change in the gross anatomy and morphometric parameters of the fish was observed. The current study provides valuable information for devising better strategies regarding environmental management, chemicals' risk assessment, biodiversity conservation, and monitoring of the aquatic organisms. DLM was concluded to be highly toxic to fish. The extensive use of DLM should be prohibited or allowed under strict environmental laws; otherwise, it might lead to the extermination of the susceptible wildlife, such as commercially very valuable but nearly threatened silver carp.

Abstract:The effects of chlorpyrifos, a frequently detected organophosphate in aquatic ecosystems, on biochemical (protein and glycogen) contents and oxidative enzyme activities (catalase and lipid peroxidation) in liver, muscle and gill tissues of three freshwater fish Trichogaster fasciata , Mystus vittatus and Heteropneustes fossilis were evaluated after 21-day exposure to 1 and 10% of 96 h LC 50 of this pesticide, which were 1.63 and 16.3 μg L −1 ; 5.87 and 58.7 μg L −1 and 2.12 and 21.2 μg L −1 , respectively. On comparing with control, significant reductions in protein concentration were found in liver, muscle and gill of the three fishes treated with both higher as well as lower concentrations of the pesticide except in gill of M. vittatus and liver of H. fossilis treated with the lower concentrations. Glycogen content reductions were significant in the liver and muscle of the fishes, as well as gill tissue of T. fasciata treated with the two pesticide concentrations. Significant elevations of catalase activity were found in liver of the three fishes treated with the higher concentrations, in muscle tissues of both T. fasciata and M. vittatus treated with both the concentrations and in gills of the three fishes except H. fossilis treated with the lower concentration of the pesticide Significant elevations of lipid peroxidation level were also found in liver of all the three fish species treated with the higher concentrations, in the muscle tissue of M. vittatus as well as in the gill of T. fasciata and H. fossilis treated with both the concentrations of the pesticide. Chlorpyrifos exposed gill ultrastructure of T. fasciata, M. vittatus and H. fossilis revealed concentration-dependent effects of the pesticide on gill surface ultrastructure which include distortion of primary and secondary lamellae, deterioration of pavement cell and microridge structures, extrusion of red blood cells (RBCs), secretion of mucous layer on filament, sloughing of primary lamellae and clumping of secondary lamellae. The present study parameters could serve as useful biomarkers for evaluating the risk of pesticide toxicity to fish. These findings also point out the possible health risks to the consumers of these fish captured from contaminated water bodies.

Impact of continuous Triazophos exposure on Labeo rohita: Physiological, biochemical, and histological alterations and IBRv2 index assessment

Abstract:Pesticides are commonly used in agriculture and aquaculture. Triazophos, an organophosphate-based pesticide, is widely used in agriculture to control many insect pests. Due to its high photochemical stability and mode of action, Triazophos could persist in the aquatic ecosystem and cause toxic effects on non-target organisms. We have studied the potential toxic effects of Triazophos on L. rohita. Primarily, we determined the median lethal concentration (LC50) of Triazophos for 24 and 96 h. Next, we studied acute (96 h, LC50-96 h) toxicity. Then, we studied chronic (35 days, 1/10th LC50-24 h Treatment I: 0.609 mg/L, 1/5th LC50-96 h Treatment II: 1.044 mg/L) toxicity. We analyzed blood biomarkers such as hematology (Hb, Hct, RBC, WBC, MCV, MCH and MCHC), prolactin, cortisol, glucose and protein levels. Concurrently, we analyzed tissue biomarkers such as glycogen, GOT, GPT, LDH and histopathology. IBRv2 index assessment method was also to evaluate the Triazophos toxicity. Studied hematological, hormonal, biochemical and enzymological biomarkers were affected in Triazophos treated groups when compare to the control group. The changes in these biomarkers were statistically significant at the 0.05 alpha level. Triazophos exposed fish shown a severe degenerated primary and secondary lamellae, lamellar fusion, hypertrophy and telangiectasia in the gills. In the hepatic tissue, it caused moderate necrosis, blood congestion, distended sinusoids with minor vacuolation, prominent pyknotic nuclei, hypertrophy, cloudy swelling of cells, lipid accumulation and fibrotic lesions. In the renal tissue, Triazophos caused thickening of Bowman's capsule, hyaline droplets degeneration, irregular renal corpuscle, congestion, cellular swelling, degeneration of tubular epithelium, necrosis, shrunken glomerulus, vacuolated glomerulus, hypertrophy, exudate and edema. IBRv2 analysis suggested that tissue biomarkers are highly sensitive to Triazophos toxicity and prolonged exposure could cause serious health effects like acute toxicity in fish. Triazophos could cause multiorgan toxicity at studied concentrations.

Abstract:Organophosphorus pesticides form a diverse group of chemicals, having a wide range of physicochemical properties with crucial toxicological actions and endpoints. These are extensively used to control pests of different food (fruits, vegetables, tea, etc.) and non-food (tobacco, cotton, etc.) crops. Malathion is an important widely used organophosphorus pesticide but its hepatotoxic effects on fish are not well studied. Therefore, the current study was designed to investigate the hepatotoxic effects of Malathion on rohu (Labeo rohita) fish in a semi-static system using different parameters. The LC50 of Malathion was found to be 5 µg/L for rohu for 96 h through Probit analysis and was used for further toxicity testing. To find the hepatotoxic effects of Malathion, changes in different biochemical indices including protein contents, Lipid Peroxidation (LPO), activities of four protein metabolic enzymes [Aspartate Aminotransferase (AAT), Lactate Dehydrogenase (LDH), Alanine Aminotransferase (AlAT), and Glutamate Dehydrogenase (GDH)], seven antioxidant enzymes [Catalase (CAT), Superoxide Dismutase (SOD), Peroxidase (POD), Glutathione (GSH), Glutathione Reductase (GR), Glutathione-s-transferase (GST), and Glutathione Peroxidase (GSH-Px)], DNA damage [in term of comet tail length, tail moment, DNA percentage in tail, and olive tail moment], reactive oxygen species (ROS), and Histopathological alterations were assayed. Malathion exposure led to a time-reliant significant (P < 0.05) decrease in protein contents and a significant (P < 0.05) increase in ROS, LPO, enzymatic activities, and DNA damage. The histopathological examination of the liver showed different changes including hepatic necrosis, fatty infiltration, hemorrhage vacuolation, glycogen vacuolation, congestion, and cellular swelling. The current study clearly revealed Malathion as a potent hepatotoxic pesticide; therefore the injudicious, indiscriminate and extensive use of Malathion should be prohibited or at least reduced and strictly monitored.

Abstract:Aquatic organisms are simultaneously exposed to multiple hazardous chemicals that can be released into water bodies. The current study aimed to evaluate the effect of sublethal concentration (1/50 96 h-LC 50 ) of two formulated pesticides: ethoprophos, bispyribac-sodium, and their combination for 1, 2, 3, and 4 weeks on oxidative stress, genotoxic response, and gill morphology in Nile tilapia. This study is the first to demonstrate the toxic effects of ethoprophos and bispyribac-sodium mixture on the commercial important species, Oreochromis niloticus . The results showed that the 96 h-LC 50 values of ethoprophos and bispyribac-sodium were 4.8 and 0.064 mg/L, respectively. Additionally, exposure to individual or combined pesticides induced a significant increase in the level of malondialdehyde (MDA), glutathione S-transferase (GST), and 8-hydroxy-2-deoxyguanosine (8-OHdG), as well as a notable decline in reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) levels at all time of exposure. Furthermore, there were alterations in ultrastructure of the gill samples, including erosive lesions on the primary and secondary lamellae, fusion of microridges, and excessive mucus secretions on the epithelium. The data clearly demonstrate that the negative effects of the tested compounds are time-dependent and are more severe in combination than in a single compound. Collectively, our results indicated that the interaction of ethoprophos and bispyribac-sodium might be largely synergistic and provide new insights into the molecular mechanisms of fish confronting these substances.

Abstract:The effect of combined exposure to decabromodiphenyl ether (BDE-209) and lead (Pb) on neurodevelopment of zebrafish (Danio rerio) larvae was investigated. Zebrafish embryos were exposed to Pb (0, 5, 10, 20 µg/L) and BDE-209 (0, 50, 100, 200 µg/L), either alone or in combination (Mix1: 5 + 50 µg/L, Mix2: 10 + 100 µg/L, Mix3: 20 + 200 µg/L) for up to 144 h post-fertilization. Growth of secondary motoneuron axons and expression of genes related to central nervous system development was significantly inhibited in Mix3 co-exposure group. A significant increase in reactive oxygen species (ROS), lipid peroxidation, DNA damage, and perturbation of the antioxidant system was detected in the Mix3 group compared to single-toxicant treatments or control. Depressed locomotor activity was recorded in the Mix2 and Mix3 groups. Addition of N-acetyl cysteine to Mix3 eliminated excessive ROS, and protected against lipid peroxidation, DNA damage, and locomotor dysfunction. Pb uptake was increased in the presence of BDE-209, but BDE-209 bioconcentration and the ability to metabolize BDE-209 were decreased in the presence of Pb. These results suggest that BDE-209 and Pb have a synergistic disruptive effect on neurodevelopment in zebrafish larvae by enhanced generation of ROS, which is a major factor that contributes to developmental neurotoxicity.
Effect of nanoformulated cypermethrin, a parasiticide in Labeo rohita fingerlings: impact on biochemical, haematological, and stress response

Abstract:Cypermethrin is a pyrethroid insecticide used in aquaculture farms against ectoparasites. The conventional pesticide formulations are now being found to be greatly ineffective at low dosages, and their indiscriminate usage may cause undesired effects in ecosystems. So, the current study was designed to develop a nanoformulation of cypermethrin that would have many advantages over conventional pesticide formulations, and the toxic effects of this formulation were then tested on the carp Labeo rohita fingerlings. The 96-h LC 50 was estimated at 0.018 mg L −1 after the acute toxicity study. Chronic toxicity studies were carried out, exposing the fish to two sublethal doses via 1/10th (C1) and 1/50th (C2) of 96-h LC 50 for a period of 45 days. The nanoformulation exposure caused a significant reduction in the brain AChE enzyme activity. Catalase and glutathione-s-transferase enzyme activity in the gills and liver increased significantly, as did superoxide dismutase enzyme activity in the kidney. Serum total protein, triglycerides, cholesterol, and some serum enzyme activities decreased. In contrast, white blood cells, red blood cells, haemoglobin, and hematocrit levels decreased only in fish exposed to a higher dose. As baseline information, these findings may aid in understanding the toxic effect of nanoformulated cypermethrin on finfish.

Abstract:The purpose was to clarify the mechanism of normal incremental amplitude responses in compound muscle action potentials (CMAPs) and single fiber action potentials (SFAPs) produced by repeated high frequency electrical stimulation. Whereas the amplitude response of CMAPs and SFAPs increased during stimulation at 20 c.p.s. of the right flexor carpi ulnaris muscles of 50 rats, their durations gradually decreased. Incremental responses of the CMAPs and SFAPs evoked by repeated stimulation were suppressed in 25 rats by 30 mg/kg of magnesium sulfate and in 25 rats by 120 mg/kg of dantrolene sodium. We speculate that the incremental responses of CMAPs and SFAPs evoked by repeated stimulation were suppressed by the unchanged shape of the muscle after the injection of those chemicals.
Impact of triflumezopyrim (insecticide) on blood and serum biochemistry of freshwater fish, subadult Labeo rohita (Hamilton, 1822)

Abstract:The impact of insecticides on water bodies can be evaluated by studying the physiology and health of fish. These chemicals pose a significant and direct risk to aquatic ecosystems and hence aquatic animals like fish. Therefore, this study was designed to investigate the toxic effects of sub-lethal concentrations of triflumezopyrim, a novel synthetic insecticide commonly used in crop protection, on the biochemical and hematological parameters of freshwater sub-adult Labeo rohita L. Here, fish were subjected to sub-lethal doses of triflumezopyrim at various concentrations (1.41 ppm, 3.27 ppm, and 4.97 ppm) for 21 days. In addition, fish were grown in an insecticide-free control group to determine the toxic effects of the insecticides. The obtained data on various biochemical and hematological parameters of the fish were analyzed using state-of-the-art statistics. Our findings suggest that triflumezopyrim exposure has a significant effect on blood variables. However, variable response, such as mean corpuscular volume, hemoglobin concentration, neutrophils, and eosinophils were unaffected by insecticide treatment, regardless of the dose. Our interhematological parameter analysis revealed varying interrelationships at various insecticide doses. Similarly, triflumezopyrim exposure significantly affected the response of serum biomarker profiles i.e., the difference in mean treatment pairs (control and three doses of triflumezopyrim) had a significant effect on the response of most serum profile parameters. For example, cholesterol response was inversely related to serological markers, including total protein, alanine aminotransferase, and aspartate aminotransferase, under triflumezopyrim exposure. The relationship pattern and grouping dynamics of the blood and serum parameters changed drastically under various insecticide doses according to correlation and cluster analyses. Our results showed that repeated exposure of fish to various sub-lethal concentrations of triflumezopyrim negatively affected their physiology and serology, which is highly harmful to the organism.

Abstract:Introduction: Butachlor (BTL) and triadimefon (TDF), the widely used herbicide and fungicide, are unavoidable enter into the aquatic environment. However, there were limited study regarding to the joint toxicity of these two pesticides on fish at present.Aim: To evaluate the potential thyroid-disrupting toxicity and exposed to different concentrations of BTL mixed with TDF.Materials and methods: Zebrafish embryo (n=3) were exposed to 0.01 and 0.05 fold of LC50 from the acute joint toxicity test, of which 0.32 mg/L (BTL) and 9.41 mg/L (TDF) for single or mixture agents (BTL: 0.0064 mg/L, 0.032 mg/L; TDF: 0.1882 mg/L, 0.9410 mg/L; co-exposure: 0.0032 mg/L BTL + 0.0941 mg/L TDF, 0.016 mg/l BTL + 0.4705 mg/L TDF) after 10-day post-fertilization. Hatching, malformation, survival rates and thyroid hormones (THs), genes expression involved in HPT-axis of embryos were measured and detected in control and separately/co-exposure treatments. THs contents were evaluated by ELISA kit and the expression levels of genes were determined by RT-PCR.Results: Hatching, malformation and survival rates of embryos exposed to single BTL exhibited no statistically significant difference from the control besides decreased of high concentration in survival rates. Exposure to TDF reduced hatching, survival rate and increased malformation. The combined exposure to BTL and TDF resulted in greater adverse effects on embryonic development. BTL exposure significantly increased free T-3 and T-4 contents. Elevated free T-3 content was also observed in the larvae exposed with single BTL. Co-exposure of the two pesticides caused greater enhanced of T-3 and T-4 levels. Furthermore, gene data showed BTL up-regulated the mRNA expression of tpo, tsh beta, tg, ttr, dio2, TDF up regulated the mRNA expression of tpo, tr alpha, ttr, dio2 and down-regulated tr beta gene. The mixture of the two pesticides caused up-regulation mRNA expression of tra, tr beta, tg, ttr, dio2.Conclusion: BTL and TDF resulted in adverse effects on zebrafish embryonic development and caused thyroid endocrine disruption, BTL and TDF have a synergistic effect on development and thyroid endocrine by enhanced level of thyroid hormone. Abstract:PBDEs are toxic, lipophilic, hydrophobic, and persistent artificial chemicals, characterized by high physical and chemical stability. Although PBDEs are known to disturb hormone signaling, many effects of 2,2',4,4',5 - pentain polybrominated diphenyl ethers (BDE-99) in fish remain unclear. The current study investigates the effects of BDE-99 in Oreochromis niloticus where sixty-four juvenile fish were orally exposed to 0.294, 2.94, 29.4 ng g-1 of BDE-99, every 10 days, during 80 days. The results showed histopathological findings in liver and kidney, increasing acetylcholinesterase activity in muscle, disturbs in the antioxidant system in liver and brain and decreasing the plasmatic levels of vitellogenin in females. According to multivariate analysis (IBR), the higher doses are related to the interaction of oxidative and non-oxidative enzymes. The present study provided evidence of deleterious effects after sub-chronic exposure of BDE 99 to O. niloticus, increasing the knowledge about its risk of exposure in fish.

Abstract:The migration of organochlorine pesticides (OCPs) and cypermethrin residues from internal organs to edible tissues of ice-held Labeo rohita (rohu) was investigated in this study. The liver (246 μg/kg) had the highest level of ∑OCP residues, followed by the gills (226 μg/kg), intestine (167 μg/kg), and muscle tissue (54 μg/kg). The predominant OCPs in the liver and gut were endosulfan (53–66 μg/kg), endrin (45–53 μg/kg), and dichloro-diphenyl-trichloroethane (DDT; 26–35 μg/kg). The ∑OCP residues in muscle increased to 152 μg/kg when the entire rohu was stored in ice, but they decreased to 129 μg/kg in gill tissues. On days 5 and 9, the total OCPs in the liver increased to 317 μg/kg and 933 μg/kg, respectively. Beyond day 5 of storage, total internal organ disintegration had led to an abnormal increase in OCP residues of liver-like mass. Despite a threefold increase in overall OCP residues by day 9, accumulation of benzene hexachloride (BHC) and heptachlor was sixfold, endrin and DDT were fourfold, aldrin was threefold, and endosulfan and cypermethrin were both twofold. Endosulfan, DDT, endrin, and heptachlor were similarly lost in the gills at a rate of 40%, while aldrin and BHC were also lost at 60 and 30%, respectively. The accumulation of OCP residues in tissues has been attributed to particular types of fatty acid derivatives. The study concluded that while pesticide diffusion to edible tissues can occur during ice storage, the levels observed were well below the allowable limit for endosulfan, endrin, and DDT.

Abstract:Use of pesticides or insecticides can be highly toxic to aquatic life forms due to leaching and agricultural runoff, rains or flood. Fipronil (FP) is a GABA receptor inhibitor, while buprofezin (BPFN) is an insect growth regulator. Presently, we exposed groups of aquaria acclimated carp fish (Cyprinus carpio) for 96h to sub-lethal concentrations of fipronil (400μgL(-1); 9.15×10(-7)molL(-1)) and buprofezin (BPFN, 100mgL(-1); 1.072×10(-6)molL(-1)) singly or in combination. The extent of damage was assessed at biochemical, hematological, molecular biological and histopathological level. Results obtained in treated fish were compared statistically with those of control non-treated fish and also among treatment groups. Significance level was p<0.05. Compared to control, serum total protein and globulin concentrations decreased significantly (p<0.0001) in fish treated with FP; while albumin concentration remained unaltered with all treatments. Glucose concentration decreased significantly (p<0.002) in fish treated with FP. In contrast, combined FP+BPFN treatment and BPFN treatment caused insignificant elevation of glucose concentration. Hematological assessment demonstrated significant decrease in red blood cell and thrombocyte counts, hemoglobin concentration and hematocrit percent; while white blood cell count showed an increase in all treatment groups (p<0.0001). Blood smears from pesticide treated fish revealed aberrant erythrocyte morphologies which included necrosis, micronuclear formation and hyperchromatosis. DNA laddering assay carried out on whole blood demonstrated excessive smear formation in combined FP+BPFN and BPFN treatment groups but no smear formation was noticeable in FP treated fish. Compared to control, whole blood DNA content increased significantly in the combined FP+BPFN and BPFN treatment groups (p<0.001 and p<0.009). With all treatments histopathological changes observed in the gills were: epithelial uplifting and necrosis of lamellae, lamellar atrophy, disruption of cartilaginous core, fusion and disorganization of lamellae and telangiectasia. In liver these were: karyorrhexis, hepatocellular hypertrophy, nuclear hypertrophy, melanomacrophage aggregates and central vein contraction, while in the kidney: deterioration of glomerulus and dilatation of Bowman's space, dilatation of renal tubules, thyroidisation, altered tubular lumen, nuclear hypertrophy, cellular atrophy, and cellular necrosis were the outcome. Our study revealed that FP and BPFN produce highly toxic effects on fish when given in combination or singly. To our knowledge, this is the first report on toxicity caused by FP and BPFN in single and combined state.
