Abstract:Variation in fermentation time may be an essential alternative to provide coffee beverages with different and unique sensory profiles. This work investigated the microbiological, chemical, and sensory changes in coffees submitted to different fermentation durations (0, 24, 48, 72, and 96 h). Self-induced anaerobiosis fermentation (SIAF) was used, and two treatments were performed: spontaneous fermentation and inoculation with S. cerevisiae CCMA0543. Microbiological analyses were performed, and the permanence of the inoculum was monitored. Chromatography (sugars, organic acids, and volatile compounds) was analyzed, and sensory analysis (temporal dominance of sensations - TDS) was performed. A total of 228 isolates were identified during spontaneous fermentation. The dominant bacteria and yeasts were Leuconostoc mesenteroides, Lactiplantibacillus plantarum, Staphylococcus warneri , Bacillus sp., Torulaspora delbrueckii , Hanseniaspora uvarum , and Meyerozyma caribbica . High concentrations of citric (18.67 mg.g − 1 ) and succinic (5.04 mg.g − 1 ) acids were detected at 96 h in SIAF fermentation. One hundred twenty-one volatile compounds were detected, but 22 were detected only in inoculated coffees. In spontaneous fermentation, 48 h of fermentation showed woody notes, while 72 h showed chestnuts. However, in the inoculated coffee, 72 h of fermentation showed high fruity dominance, and 96 h of fermentation was the only one with herbaceous notes. In addition, yeast inoculation increased the intensity of caramel notes in the first 48 h and increased the fruity flavor after 72 h of fermentation. Therefore, the type of fermentation (with or without inoculation) and the chosen fermentation time will depend on the sensorial profile the producer intends to obtain.

Characterization of bioactive, chemical, and sensory compounds from fermented coffees with different yeasts species

Abstract:Selected yeasts for coffee fermentation are correlated with changes in chemical compounds and beverage sensory characteristics. This work aimed to evaluate the chemical and sensory modifications of coffee fermented with one yeast (Saccharomyces cerevisiae CCMA 0543, Candida parapsilosis CCMA 0544, or Torulaspora delbrueckii CCMA 0684) and in co-inoculation (from two to two and the three together) by dry processing. Real-time PCR analyzes, total phenolic content and antioxidant activity (DPPH, ABTS, and FRAP), liquid and gas chromatography, and sensory analysis were performed. Caparaó coffees showed a higher C. parapsilosis (6.14 Log cell.g-1) population followed by S. cerevisiae (5.85 Log cell.g-1) and T. delbrueckii (4.64 Log cell.g-1). The total phenolic content has a strong and positive correlation with the fermentation time and the roasted beans and a moderate and positive correlation with DPPH, FRAP, and ABTS. Coffee inoculated with T. delbrueckii reduced caffeine concentration during the fermentation process. In co-cultivation, the trigonelline concentration showed the most significant decrease (around 4 mg.g-1) when inoculated with S. cerevisiae and T. delbrueckii. Detection of some organic acids and volatile compounds during fermentation may indicate that the starter cultures used different metabolic routes. All co-inoculation treatments presented the best sensory scores (>86 points). In the inoculated fermentation, fruity, citric, molasses, freshness, and wine notes appeared. The co-inoculated treatment with S. cerevisiae CCMA 0543, C. parapsilosis CCMA 0544, and T. delbrueckii CCMA 0684 was the best, considering the diversity of sensory notes descriptors and the final concentration of organic acids.

Abstract:Post-harvest processing and microbial fermentation of coffee fruits play an essential role in the metabolites formation that influence the nutritional and sensory quality of the beverage. Thus, the objective of this study was to analyze the effect of coffee cherries processing and fermentation conditions on the microbial communities and the chemical and sensory quality of the beverage. Induced fermentation changed in the bacteria and fungi communities (Treatments: T1, T3, and T7). Klebsiella sp. inoculation (T1) allowed an increase in richness of bacteria and 81 points in the sensory score over the fermentation time. However, there was a reduction in richness of microbial community in treatments with Saccharomyces cerevisiae (T3 and T7). An increase in the indexes of microbial diversity was observed in 72 h in treatment with coffee pulp (T2). In treatment with coffee cherries and spontaneous fermentation (T4) had a higher sensory score than other treatments, indicating a sensory gain from 36 to 72 h. Coffee cherries with superficial disinfection (T5) had a reduction in microbial profile, but did not change the final score of the beverage over the 72 h. In T6 (floaters fruits) was observed an alteration in the fungal community (36–72 h) and the lowest sensory score. The impact of adding water on coffee fermentation was dependent on time (T3 and T7). Furthermore, 5-hydroxymethylfurfural has a positive correlation with the final score of the beverage. Thus, microbial profile and sensory score of beverages are dependent of conditions of processing of coffee fruits and fermentation.

Abstract:This study evaluated the inoculation of Meyerozyma guilliermondii and Bacillus licheniformis, separately or in co-culture, in wet-processed conilon coffee. Wet fermentation was conducted for 48 h. Mesophilic bacteria, lactic acid bacteria, yeasts, and filamentous fungi were counted during fermentation. The inoculation of B. licheniformis and M. guilliermondii stimulated the multiplication of lactic acid bacteria. Acetic, citric, lactic, oxalic, malic, succinic, tartaric acids, glucose, and fructose were identified in all treatments at different concentrations. Methyl salicylate, 2-heptanol, 2-nonanol, and heptanone were found during fermentation. Methylpyrazine, 2,6-dimethylpyrazine, 2,5-dimethylpyrazine, and 3-ethyl-2,5-dimethylpyrazine identified after roasting assigned notes of "almond" and "chocolate" to the beverages. All treatments were classified as "premium," with the B. licheniformis treatment receiving the highest score. Bacillus licheniformis obtained better performance in fermentation, increasing coffee score and producing volatile compounds that provided positive sensory notes to the beverage.

Abstract:Climatic conditions, genotypes, and post-harvest processing methods influence coffee quality. Microbial fermentation during post-harvest processing has sparked researchers' interest due to the modulation of the sensory characteristics of coffee. However, the influence of microbial fermentation on different coffee genotypes has been little investigated. The objective of this study was to evaluate the chemical and sensory changes of Coffea arabica cultivars caused by different post-harvest processing and fermentations. Catucaí 785 and Catucaí Açú cultivars had better sensory evaluation than the Arara cultivar in the two years of sampling. However, dry fermentation proved to be a promising alternative to improve the sensory analysis of the Arara cultivar. Inoculation of Saccharomyces pastorianus yielded positive results in the chemical and sensory of the Catucaí Açú cultivar. Each coffee genotype is capable of expressing unique sensory and chemical attributes depending on the different post-harvest processing and fermentations applied.

Abstract:Background Coffee fermentation process influences the final coffee composition and the sensory aspects which define the quality of the beverage. In this study, coffee fruits underwent spontaneous self-induced anaerobic fermentation using samples with two percentages of immature fruits in submerged and solid-state processing. The effects on physicochemical composition and on the sensorial quality of coffee were evaluated. Results The two percentages of immature fruits corresponded to 11.0 and 0.3% of unripe fruits. The percentage of immature fruits significantly altered the initial content of sugars (sucrose, glucose, and fructose), ash, and titratable acidity. The water addition during the fermentative process did not significantly influence final moisture, proteins, citric, and propionic acids concentrations. Compared with solid-state, the submerged process gave rise to coffees with lower concentrations of ethanol, glycerol, ash, lipids, succinic, and acetic acids. Coffee fermented with 0.3% of immature fruits showed higher lactic acid production in submerged fermentation (67.44 mg/g), and higher concentrations of ethanol (42.84 mg/g) and glycerol (1.68 mg/g) in solid-state fermentation. All coffees produced were classified as specialty coffees with a score above 84 points. However, the submerged fermented coffee with 11% immature fruit stood out with notes of caramel, brown sugar, honey, orange, lemon, floral, nut, yellow and red fruits. Conclusions This study confirmed that spontaneous fermentation can be used to produce specialty coffees. Differentiation in sensorial attributes can be achieved by water addition and the percentage of green fruits during fermentation process. Up to 11% of immature fruits did not compromise coffee quality.

Abstract:Abstract The aim of this research was to study the coffee fermentation with inoculation of Bacillus subtilis isolated from civet ( Paradoxorus hermaphroditus ) as starter culture. The quality of coffee was evaluated by sensory quality (cup test). Two treatments were carried out: control/coffee non-inoculated starter culture (treatment F) and coffee inoculated with a Bacillus subtilis starter culture (treatment G). The fermentations were conducted for 48 h in triplicate. A inoculation of Bacillus subtilis as a starter culture cause a decrease in pH and increase in total titratable acidity of coffee fermentation solution. A panel of five trained coffee testers (Q-grader from Gayo Cupper’s Team) evaluated the samples. Ten sensorial attributes analyzed were fragrance/aroma, flavor, aftertaste, acidity, body, uniformity, balance, sweetness, cleanliness and overall. The result showed that the use of Bacillus subtilis starter culture improved quality of coffee fermentation. The aroma, flavour, aftertaste, body, balance and overall were increased. The cupping score total of coffee increased from 82.83 to 84.33. Metabolites analysis should be further investigated to confirm this result, especially organic acids compound.

Abstract:Background: Yeast and lactic acid bacteria (LAB) play an important part in the post-harvest fermentation of coffee. This study applied lab-scale fermentation to commercial green coffee beans using dry coffee pulp as the substrate, with the aim of modifying coffee-bean flavor. In addition to spontaneous fermentation, yeast and LAB isolated from coffee beans and dried coffee pulp were added during fermentation. Results: Co-inoculation of yeast and LAB showed a significant effect on the chlorogenic acid content after between 24 and 72 h of fermentation. Acetic, citric, malic, lactic, and quinic acids were shown to be affected significantly (P < 0.05) by fermentation and inoculation. Gas chromatography detected that esters, alcohols, aldehydes, furans, and pyrazines were the primary compounds in the coffee beans. Certain volatile groups were present in greater concentrations and broader varieties within the inoculated beans. The highest cupping scores were given to beans that had been co-inoculated with yeast and LAB. Conclusion: Overall, the use of yeasts and LAB starters showed potential to create coffee beverages with desirable characteristics by standardized fermentation. © 2024 Society of Chemical Industry.

Abstract:Understanding the effects of microorganisms on coffee fermentation is crucial to ensure sensory quality and food security. The analysis of the dynamics of the microbial community during fermentation can contribute to a better understanding of the beneficial and harmful effects of microorganisms and help select starter cultures to improve coffee quality. Furthermore, the anaerobic environment produced by carbonic maceration of the coffee fruits inhibits aerobic respiratory processes and stimulates fermentative metabolism, modulating the microbial community during coffee fermentation. This study evaluated the effects of carbonic maceration in the fungal community dynamics during the fermentation of Coffea arabica fruits at 18, 28, and 38 °C for 24, 48, 72, 96, and 120 h. Fungal diversity was accompanied by high-throughput sequencing (NGS) of the Internal Transcribed Spacer (ITS) region. During the coffee fermentation, the fungal community changed over time, with the most significant changes occurring at 18 and 28 °C after 72 h. However, at 38 °C, there were greater variations in fungal composition and fungal diversity was highest after 120 h. The yeast Pichia cephalocereana was predominant in the fermentations. These results indicated that temperature and fermentation conditions influence the fungal community during coffee fermentation. Lower temperatures might favor a more stable microbial environment, while higher temperatures lead to more intense changes. Thus, our data from NGS can help in the identification, isolation, and metabolic characterization of fungi for the fermentation of coffee fruits.

Abstract:Research into microbial interactions during coffee processing is essential for developing new methods that adapt to climate change and improve flavor, thus enhancing the resilience and quality of global coffee production. This study aimed to investigate how microbial communities interact and contribute to flavor development in coffee processing within humid subtropical climates. Employing Illumina sequencing for microbial dynamics analysis, and high-performance liquid chromatography (HPLC) integrated with gas chromatography-mass spectrometry (GC-MS) for metabolite assessment, the study revealed intricate microbial diversity and associated metabolic activities. Throughout the fermentation process, dominant microbial species included Enterobacter, Erwinia, Kluyvera, and Pantoea from the prokaryotic group, and Fusarium, Cladosporium, Kurtzmaniella, Leptosphaerulina, Neonectria, and Penicillium from the eukaryotic group. The key metabolites identified were ethanol, and lactic, acetic, and citric acids. Notably, the bacterial community plays a crucial role in flavor development by utilizing metabolic versatility to produce esters and alcohols, while plant-derived metabolites such as caffeine and linalool remain stable throughout the fermentation process. The undirected network analysis revealed 321 interactions among microbial species and key substances during the fermentation process, with Enterobacter, Kluyvera, and Serratia showing strong connections with sugar and various volatile compounds, such as hexanal, benzaldehyde, 3-methylbenzaldehyde, 2-butenal, and 4-heptenal. These interactions, including inhibitory effects by Fusarium and Cladosporium, suggest microbial adaptability to subtropical conditions, potentially influencing fermentation and coffee quality. The sensory analysis showed that the final beverage obtained a score of 80.83 ± 0.39, being classified as a specialty coffee by the Specialty Coffee Association (SCA) metrics. Nonetheless, further enhancements in acidity, body, and aftertaste could lead to a more balanced flavor profile. The findings of this research hold substantial implications for the coffee industry in humid subtropical regions, offering potential strategies to enhance flavor quality and consistency through controlled fermentation practices. Furthermore, this study contributes to the broader understanding of how microbial ecology interplays with environmental factors to influence food and beverage fermentation, a topic of growing interest in the context of climate change and sustainable agriculture.
Prenatal diagnosis of gallbladder duplication

Abstract:Gallbladder duplication is a rare congenital anomaly, resulting in two separate gallbladder cavities, each with a cystic duct. Gallbladder anomalies, including cholelithiasis, choledochal cysts and agenesis, have been observed in utero1–3, but there have been only two reports of a duplicated fetal gallbladder4,5. Here we describe a new case that was detected prenatally. Two fluid-containing cystic structures were found in the right upper quadrant of a fetus during a routine ultrasound examination of a 26-year-old, gravida 2 woman at 32 weeks’ gestation. These structures were saccular in morphology and were located adjacent to the fetal liver (Figure 1); the finding had not been described in the screening sonogram at 22 weeks’ gestation. No other detectable anomalies were found. Follow-up ultrasound examinations at 33 and 36 weeks’ gestation confirmed the initial findings and a diagnosis of gallbladder duplication was made. A healthy 2970-g male was delivered at 38 weeks’ gestation. Abdominal sonography on postnatal day 4 confirmed the existence of a double gallbladder. The gallbladder appears as a small, hypoechoic, oval or rectangular cyst-like structure located at the lower border of the liver, close to the intestinal loops, and to the right of the intrahepatic umbilical vein3. The fetal gallbladder is visualized in 65–82% of fetuses at 24–27 weeks’ gestation6, although by using a transvaginal approach at 14–16 weeks’ gestation the detection rate may be more than 99%3. Evaluation of spontaneous fermentation impact on the physicochemical properties and sensory profile of green and roasted arabica coffee by digital technologies

Abstract:There is a growing demand for specialty coffee with more pleasant and uniform sensory perception. Wet fermentation could modulate and confer additional aroma notes to final roasted coffee brew. This study aimed to assess differences in volatile compounds and the intensities of sensory descriptors between unfermented and spontaneously fermented coffee using digital technologies. Fermented (F) and unfermented (UF) coffee samples, harvested from two Australia local farms Mountain Top Estate (T) and Kahawa Estate (K), with four roasting levels (green, light-, medium-, and dark-) were analysed using near-infrared spectrometry (NIR), and a low-cost electronic nose (e-nose) along with some ground truth measurements such as headspace/gas chromatography-mass spectrometry (HS-SPME-GC-MS), and quantitative descriptive analysis (QDA ®). Regression machine learning (ML) modelling based on artificial neural networks (ANN) was conducted to predict volatile aromatic compounds and intensity of sensory descriptors using NIR and e-nose data as inputs. Green fermented coffee had significant perception of hay aroma and flavor. Roasted fermented coffee had higher intensities of coffee liquid color, crema height and color, aftertaste, aroma and flavor of dark chocolate and roasted, and butter flavor (p < 0.05). According to GC-MS detection, volatile aromatic compounds, including methylpyrazine, 2-ethyl-5-methylpyrazine, and 2-ethyl-6-methylpyrazine, were observed to discriminate fermented and unfermented roasted coffee. The four ML models developed using the NIR absorbance values and e-nose measurements as inputs were highly accurate in predicting (i) the peak area of volatile aromatic compounds (Model 1, R = 0.98; Model 3, R = 0.87) and (ii) intensities of sensory descriptors (Model 2 and Model 4; R = 0.91), respectively. The proposed efficient, reliable, and affordable method may potentially be used in the coffee industry and smallholders in the differentiation and development of specialty coffee, as well as in process monitoring and sensory quality assurance.
Functioning of Saccharomyces cerevisiae in honey coffee (Coffea canephora) and their effect on metabolites, volatiles and flavor profiles

Abstract:Post-harvesting and microbial activity of coffee play a critical role in the metabolites and the sensory quality of the brew. The pulped natural/honey coffee process is an improvised semi-dry technique consisting of prolonged fermentation of depulped coffee beans excluding washing steps. The starter culture application in coffee industry plays an important role to enhance the cup quality. This work focuses on the fermentation of pulped natural/honey Robusta coffee with a starter culture (Saccharomyces cerevisiae MTCC 173) and the identification of fermentation patterns through 1H NMR, microbial ecology, volatomics and organoleptics of brew. Fermentation was accelerated by yeast populace (10 cfu log/mL) for 192 h. Principal compound analysis performed on 1H NMR led to the investigation of metabolites such as sugars, alkaloids, alcohols, organic acids and amino acids. Detection of some sugars and organic acids represented that the starter cultures imparted few metabolic changes during the process. A major activity of sugars in fermentation with 83.3 % variance in PC 1 and 16.7 % in PC 2 was observed. The chemical characteristics such as carbohydrates (41.88 ± 0.77 mg/g), polyphenols (34.16 ± 0.79 mg/g), proteins (58.54 ± 0.66 mg/g), caffeine (26.54 ± 0.06 mg/g), and CGA (21.83 ± 0.04 mg/g) were also evaluated. Abstract:This study aims to investigate the influence of yeast fermentation and inoculation methods on the quality of coffee across 3 different coffee cultivars. Fermenting Sacchoromyces cerevisiae SafAle s-33 on Coffea arabica cv. Caltimor via bucket inoculation outperforms spray inoculation across all aspects including microbial cell adhesion, the chemical profile, reducing sugar content and sensory analysis conducted by a Q-grader. The PCA analysis revealed distinct profiles of organic acids and volatile compounds in roasted beans in yeast fermentation and natural fermentation of all 3 coffee varieties. Particularly, volatile compounds such 1-(4-Methoxyphenyl)octan-1-one, 8-Oxabicyclo[3.2.1]oct-6-en-3-one and 5,6-Dimethoxy-2-(2-hydroxyethyl-1-thio)-3 trimethylsilyl methyl-1,4-benzoquinone and, furfuryl ethyl ether and 5-methyl furfural were detected only in yeast fermented bean. The malic acid also contained a higher amount in roasted coffee beans from the yeast fermentation of all 3 coffee varieties than the natural fermentation. Those compounds present a notably positive influence on the sensory aspects of the coffee i.e. sweet caramel, dry fruit, vanilla and hazelnut-like flavors. In addition, roasted beans from the yeast fermentation of C. arabica cv. Caltimor and C. canephora cv. Robusta displayed higher sensory scores when compared to those from natural fermentation, conversely in C. arabica cv. Bourbon, beans from both yeast fermentation and natural fermentation displayed similar quality levels. Our study concluded that the commercial yeast strain within the beer and wine industry proved to be an alternative source for coffee fermentation. Nevertheless, the selection of the coffee variety plays a crucial role when utilizing S. cerevisiae SafAle s-33 as a starter.
Effects of geographical origin and post-harvesting processing on the bioactive compounds and sensory quality of Brazilian specialty coffee beans

Abstract:Specialty coffee beans are those produced, processed, and characterized following the highest quality standards, toward delivering a superior final product. Environmental, climatic, genetic, and processing factors greatly influence the green beans' chemical profile, which reflects on the quality and pricing. The present study focuses on the assessment of eight major health-beneficial bioactive compounds in green coffee beans aiming to underscore the influence of the geographical origin and post-harvesting processing on the quality of the final beverage. For that, we examined the non-volatile chemical profile of specialty Coffea arabica beans from Minas Gerais state, Brazil. It included samples from Cerrado (Savannah), and Matas de Minas and Sul de Minas (Atlantic Forest) regions, produced by two post-harvesting processing practices. Trigonelline, theobromine, theophylline, chlorogenic acid derivatives, caffeine, caffeic acid, ferulic acid, and p-coumaric acid were quantified in the green beans by high-performance liquid chromatography with diode array detection. Additionally, all samples were roasted and subjected to sensory analysis for coffee grading. Principal component analysis suggested that Cerrado samples tended to set apart from the other geographical locations. Those samples also exhibited higher levels of trigonelline as confirmed by two-way ANOVA analysis. Samples subjected to de-pulping processing showed improved chemical composition and sensory score. Those pulped coffees displayed 5.8% more chlorogenic acid derivatives, with an enhancement of 1.5% in the sensory score compared to unprocessed counterparts. Multivariate logistic regression analysis pointed out altitude, ferulic acid, p-coumaric acid, sweetness, and acidity as predictors distinguishing specialty coffee beans obtained by the two post-harvest processing. These findings demonstrate the influence of regional growth conditions and post-harvest treatments on the chemical and sensory quality of coffee. In summary, the present study underscores the value of integrating target metabolite analysis with statistical tools to augment the characterization of specialty coffee beans, offering novel insights for quality assessment with a focus on their bioactive compounds.

Abstract:Coffee fermentation processes: Bibliometric analysis and implications for chemical and sensorial quality due to different bioprocessing pathways. Summary Fermentation has been studied to improve the chemical composition and sensory profile of coffee, of interest to the food industry and farmers seeking competitiveness in the international market. This study addresses the influence of biotransformation on the synthesis of flavour precursor compounds and correlates subsequent processes, such as roasting, and chemical mechanisms responsible for the formation of coffee's sensory profiles. The results indicate that coffee fermentation contributes to the formation of precursor metabolites of volatile compounds and the different fermentation techniques favour the production of high‐quality coffee. Bibliometrics analysis from 2012 to 2022 highlighted Brazil and China with a high number of research, scientific collaborations, and international cooperation, indicating a current trend in research focused on sensory modulation. Fermentation technologies point to a promising scenario with the possibility of producing coffee with previously defined sensory characteristics. This market is in full development and corroborates the growing pace of coffee consumption.

Abstract:Abstract The time of the fermentation process of coffee from northern Peru is variable (9 to 48 hours) since coffee farmers do not use standardized processes, causing a variety of coffee qualities. This study aimed to identify volatile compounds in both short (9 hours) and long (32 hours) coffee fermentation processes from coffee farms in northern Peru using Solid-Phase Microextraction-Gas Chromatography/Mass Spectrometry (SPME-GC/MS) to associate the coffee quality and diversity of volatile compounds. Sensory analyses showed that the short fermentation process (SFP) scored 77.8 ± 0.39 and had chocolate, wood, cardboard, dry, fatty and rough notes, while the long fermentation process (LFP) showed higher punctuations 85.5 ± 3.16 and citrus, fruity, floral, caramel and chocolate sensory attributes. A total of 90 compounds were found in the SFP, whereas 141 compounds were identified in the LFP. Significant differences in the relative abundance of 14 chemical compounds were reported in the SFP and LFP (p < 0.05). From these results, the presence of benzaldehyde, methional, hexanal, 2-heptanone, pentadecane, 1-butanol-3-methyl-acetate, and benzene-acetic acid ethyl ester seems to impact the quality of coffee. The analysis of similarities showed that coffee samples (5 h and 9 h) during the SFP were very variable, whereas coffee samples from LFP showed some tendency to group, which may be related to the difference in altitude and temperature in coffee farms making comparison between them difficult. In addition, this study highlights the complex relationship between coffee fermentation and flavour and the influence of several factors and variables that may affect the composition of flavour and aroma precursors in green coffee beans obtained from wet fermentation.

Abstract:Fine-aroma cocoa (Theobroma cacao) is one of Ecuador’s most iconic export products and comprises 63% of world production. Nevertheless, few advances have been made to improve fermentation processes that might benefit the development of chocolate’s organoleptic characteristics. The study of starter cultures, which seek to improve organoleptic properties or decrease fermentation time, has been investigated in other countries. The aim of this study was to analyze the effect of a native microbial cocktail based on two yeasts (Torulaspora delbrueckii and Hanseniaspora uvarum), a lactic acid bacterium (LAB) (Limosilactobacillus plantarum), and an acetic acid bacterium (AAB) (Acetobacter ghanensis) inoculated at the beginning of the fermentative process while tracking physical and biochemical variables, microbial population dynamics, and bean fermentation time. The starter culture caused changes in sugar and acid content and increased polyphenols, which in turn generated temperature and pH changes in the dough. The dynamics of yeast, AAB, and mesophilic microorganisms remain higher than the controls throughout the process. A decrease in filamentous fungi that affect the flavor and quality of beans was observed due to the production of acetic acid or secondary metabolites from yeasts and LAB, and resulted in 24% greater fermentation than spontaneous fermentation in only 96 h.

Abstract:Background: Fermented cocoa beans (Theobroma cacao L.) are a pivotal raw material for chocolate production. A cocktail yeast applied in the cocoa fermentation process can promote the formation of pleasant metabolites. Saccharomyces, Pichia and Hanseniaspora have been widely used in fermentation to improve the final product organoleptic profile, highlighting that fermentation is a critical point for chocolate flavour precursor production. This study aims to evaluate the impact of Pichia kluyveri and Saccharomyces cerevisiae strains as starter cultures on the fermentation for two cocoa hybrids, FA13 and CEPEC2002. Results: During fermentation processes, volatile organic compounds (VOCs) and protein profiles were assessed. Chocolates produced were also assessed regarding the presence of VOCs. Eighty VOCs were identified using gas chromatography coupled to mass spectrometry analysis. Mass spectrometry provided the protein profile evolution during fermentation and showed that the profiles changed with inoculation type (spontaneous versus inoculated fermentation). Chocolate obtained from FA13 inoculated with S. cerevisiae strain contained a greater amount of organics acids, being categorised as sourer than chocolate produced by spontaneous fermentation of FA13. CEPEC2002 inoculated with S. cerevisiae strain in co-culture with P. kluyveri strain generated less sour and sweeter chocolate than spontaneous fermentation only. Conclusions: Chocolates from inoculated assays with starter cultures were more accepted by evaluators, highlighting that P. kluyveri and S. cerevisiae influence the composition of VOCs. Besides, protein profiles also changed throughout fermentation. Further investigation should be conducted to clarify protein degradation dynamics during inoculated fermentations to define which of the microbial cultures positively affect the chocolate sensory characteristics. © 2021 Society of Chemical Industry.
