Abstract:Background RNA interference (RNAi) has potential as a new strategy for pest control. However, the current overemphasis on the control of a single pest increased control costs. The aim of this study was to find a green method of controlling several pests without affecting the natural enemies with a single target gene. One possible RNAi target is the threonyl-tRNA synthetase (ThrRS), which is conserved and plays a significant role in protein biosynthesis. Results In this study, one threonyl-tRNA synthetase gene (NlthrS) was identified from the brown planthopper (Nilaparvata lugens). Spatio-temporal expression pattern analysis showed that NlthrS was highly expressed in the ovary, late embryogenesis, nymphs and female adults. In addition, RNAi-mediated knockdown of NlthrS caused 85.6% nymph mortality, 100% female infertility, molting disorder, extended nymph duration and shortened adult longevity. Target-specific results were obtained when dsNlthrS was used to interfere with the whiteback planthopper (Sogatella furcifera), small brown planthopper (Laodelphax striatellus), zig-zag winged leafhopper (Inazuma dorsalis) and their natural enemy (green mirid bug, Cyrtorhinus lividipennis). In addition, dsNlthrS could cause high mortalities of three species of planthoppers (85.6-100%), while only dsNlthrS-1 led to the death (97.3%) of I. dorsalis that was not affected by dsNlthrS-2. Furthermore, neither dsNlthrS-1 nor dsNlthrS-2 could influence the survival of C. lividipennis. Conclusion The results reveal the biological functions of ThrRS in N. lugens in addtion to its protein synthesis, deepening our understanding of tRNA synthase in insects and providing a new method for the control of several rice pests via one dsRNA design. Abstract:The current methods for pest managements in cereal crops rely mainly on the applications of conventional chemicals, which are known to have low pest control efficiencies and are less environmentally friendly. The primary aim of this research endeavor was to devise a specific, efficient and eco-friendly biological agent capable of mitigating maize lethal necrosis (MLN), a viral disease that affected maize productivity and mainly caused by coinfection of maize chlorotic mottle virus (MCMV) and sugarcane mosaic virus (SCMV). Through this study, we have found that silencing Ferredoxin 3 (ZmFd3) expression in maize can significantly reduce the damages caused by MCMV and/or SCMV infections. Based on this finding, we have developed a liposome nanoparticles (LNPs)mediated nanotechnology to suppress MCMV and/or SCMV infections. Using this nanotechnology, we have confirmed that the cationic/anionic liposomes (CLPs and ALPs) can be used to package and promote ZmFd3dsRNA spread in maize plants. Through leaf spray, we have determined that the anti-MCMV and anti-SCMV activities of ZmFd3@CLPs and ZmFd3@ALPs nanoparticles can reach 52.46 similar to 79.83 % at 6 days post virus inoculation (dpi). Because ZmFd3 is involved in hypersensitive response (HR), and can interact with MCMV p7b and SCMV HC-Pro, we designed an FHP-dsRNA to target ZmFd3 , MCMV p7b , and SCMV HC-Pro transcripts for silencing in plants simultaneously. The result showed that both FHP@CLPs and FHP@ALPs nanoparticles can provide excellent, although short, protection to maize and sorghum plants to MCMV and SCMV infections. Overall, this study has developed a novel nanotechnology that can be used to mitigate virus infection in cereal crops through RNA interference (RNAi).
Spray method application of transdermal dsRNA delivery system for efficient gene silencing and pest control on soybean aphid Aphis glycines

Abstract:RNA interference (RNAi) has been recognized as a novel and safe strategy in pest management due to its high sequence-dependent specificity. However, the existing dsRNA delivery methods largely restrict the application of the RNAi-based pest management strategy; thus, we previously constructed a nanocarrier-based transdermal dsRNA delivery system on the soybean aphid Aphis glycines with the help of nanocarrier and detergent. In the current study, we improved our transdermal dsRNA delivery system with a smaller and cheaper nanocarrier to investigate the efficacy of spraying aphid-infested soybean seedlings to apply our RNA pesticide. A dsRNA/nanocarrier/detergent formulation was performed, and the dsRNA could penetrate the aphid body wall within 4 h with the help of nanocarrier through the topical application. Four potential RNAi target genes (TREH, ATPD, ATPE and CHS1) were selected and cloned, and their dsRNA fragments were synthesized and tested through the transdermal dsRNA delivery system. The delivered dsRNA efficiently silenced the target gene expression with the knockdown effects ranging from 86.86 to 58.87% and resulted in a high mortality up to 81.67% (dsATPD + dsATPE) through the topical application, when through the spray method, with the highest percent mortality of 78.50% (dsATPD + dsCHS1). Our novel transdermal dsRNA delivery system not only provides a powerful tool for gene functional analysis in laboratory, but also shows a great potential for the pest management in the field, which will promote the practice and development of RNAi-based pest management.

Abstract:BACKGROUND Although the utility of dsRNA-mediated knockdown as an environmentally friendly pest management strategy has gained traction in recent years, its overall efficacy has been limited by poor stability and limited cellular uptake. The encapsulation of dsRNAs with various nanomaterials, however, has shown promise in overcoming these limitations. This study sought to investigate the biological efficacy of an oral dsRNA nanomaterial mixture targeting the CYP15C1 gene product in the economically important rice pest, Chilo suppressalis. RESULTS A putative CYP15C1 ortholog was cloned from C. suppressalis midguts. The transcript is down-regulated in fifth-instar larvae and is most highly expressed in heads. RNAi-mediated knockdown of CsCYP15C1 was associated with significantly increased mortality. More importantly, feeding a dsRNA-nanomaterial mixture significantly increased larval mortality compared to feeding dsRNA alone. CONCLUSION A critical role for CsCYP15C1 function in molting is supported by sequence similarity with known juvenile hormone epoxidases, its expression profile, and abnormal molting phenotypes associated with RNA-mediated knockdown. CsCYP15C1 is thus a prime target for controlling C. suppressalis. Furthermore, RNAi-mediated characterization of candidate gene function can be enhanced by incorporating an enveloping nanomaterial. Abstract:Spodoptera frugiperda is a serious threat to various crops, such as corn and rice, and results in severe economic losses. Herein, a chitin synthase sfCHS highly expressed in the epidermis of S. frugiperda was screened, and when interfered by an sfCHS -siRNA nanocomplex, most individuals could not ecdysis (mortality rate 53.3 %) or pupate (abnormal pupation 80.6 %). Based on the results of structure-based virtual screening, cyromazine (CYR, binding free energy −57.285 kcal/mol) could inhibit ecdysis (LC 50 , 19.599 μg/g). CYR-CS/siRNA nanoparticles encapsulating CYR and SfCHS-siRNA with chitosan (CS) were successfully prepared, as confirmed by scanning electron microscopy (SEM) and transmission electron microscopy (TEM), and 74.9 mg/g CYR was characterized in the core of CYR-CS/siRNA by high-performance liquid chromatography and Fourier transform infrared spectroscopy. Small amounts of prepared CYR-CS/siRNA containing only 1.5 μg/g CYR could better inhibit chitin synthesis in the cuticle and peritrophic membrane (mortality rate 84.4 %). Therefore, chitosan/siRNA nanoparticle-loaded pesticides were useful for pesticide reduction and comprehensive control of S. frugiperda .

Compared with traditional double‐stranded (ds)RNA delivery systems, nanoparticles such as chitosan, liposomes, and cationic dendrimers offer advantages in delivering dsRNA/small interfering (si)RNA to improve RNA interference (RNAi) efficiency, thus promoting the development and practice of RNAi‐based pest management strategies. Abstract:There are two main limitations for sprayable RNA pesticide development: delivery efficiency and synthetic cost of double-stranded RNA (dsRNA). We previously constructed a nanocarrier-based transdermal dsRNA delivery system and a novel bacteria-based hairpin RNA (hpRNA) expression system to solve these challenges. Herein, as a subsequent exploration of RNA pesticide (sprayable ds/hpRNA for pest control), we performed a greenhouse application of bacteria-expressed and nanocarrier-delivered RNA pesticide on green peach aphid. The nanoparticle SPc could combine and deliver dsRNA across the aphid cuticle and V-type proton ATPase subunits d (ATP-d) and G (ATP-G) were selected as the potential RNA interference (RNAi) targets. Our plasmid-Escherichia coli system simultaneously expressing ATP-d and ATP-G hairpin RNAs (hpRNAs) was constructed for mass production of hpRNA. The expressed hpRNA was mixed with SPc and detergent to form RNA formulation, which showed a certain insecticidal activity through the spray application in the greenhouse. Total control efficacy of our RNA pesticide could reach 61% on 3 d and maintained at 50% until the sixth day. To our knowledge, our study is the first attempt to apply the bacteria-expressed and nanocarrier-delivered RNA pesticides for pest control in the greenhouse trial, which is beneficial for promoting the development of RNA pesticides.

Nanoparticle LDH enhances RNAi efficiency of dsRNA in piercing-sucking pests by promoting dsRNA stability and transport in plants

Abstract:Piercing-sucking pests are the most notorious group of pests for global agriculture. RNAi-mediated crop protection by foliar application is a promising approach in field trials. However, the effect of this approach on piercing-sucking pests is far from satisfactory due to the limited uptake and transport of double strand RNA (dsRNA) in plants. Therefore, there is an urgent need for more feasible and biocompatible dsRNA delivery approaches to better control piercing-sucking pests. Here, we report that foliar application of layered double hydroxide (LDH)-loaded dsRNA can effectively disrupt Panonychus citri at multiple developmental stages. MgAl-LDH-dsRNA targeting Chitinase (Chit) gene significantly promoted the RNAi efficiency and then increased the mortality of P. citri nymphs by enhancing dsRNA stability in gut, promoting the adhesion of dsRNA onto leaf surface, facilitating dsRNA internalization into leaf cells, and delivering dsRNA from the stem to the leaf via the vascular system of pomelo plants. Finally, this delivery pathway based on other metal elements such as iron (MgFe-LDH) was also found to significantly improve the protection against P. citri and the nymphs or larvae of Diaphorina citri and Aphis gossypii, two other important piercing-sucking hemipeteran pests, indicating the universality of nanoparticles LDH in promoting the RNAi efficiency and mortality of piercing-sucking pests. Collectively, this study provides insights into the synergistic mechanism for nano-dsRNA systemic translocation in plants, and proposes a potential eco-friendly control strategy for piercing-sucking pests.

Abstract:The rice pest Nilaparvata lugens (the brown planthopper, BPH) has developed different levels of resistance to at least 11 chemical pesticides. RNAi technology has contributed to the development of environmentally friendly RNA biopesticides designed to reduce chemical use. Consequently, more precise targets need to be identified and characterized, and efficient dsRNA delivery methods are necessary for effective field pest control. In this study, a low off-target risk ds NlUAP fragment (166 bp) was designed in silico to minimize the potential adverse effects on non-target organisms. Knockdown of NlUAP via microinjection significantly decreased the content of UDP- N -acetylglucosamine and chitin, causing chitinous structural disorder and abnormal phenotypes in wing and body wall, reduced fertility, and resulted in pest mortality up to 100 %. Furthermore, ds NlUAP was loaded with ROPE@C, a chitosan-modified nanomaterial for spray application, which significantly downregulated the expression of NlUAP , led to 48.9 % pest mortality, and was confirmed to have no adverse effects on Cyrtorhinus lividipennis , an important natural enemy of BPH. These findings will contribute to the development of safer biopesticides for the control of N. lugens .

Characterization of an RR-2 cuticle protein DcCP8 and its potential application based on SPc nanoparticle-wrapped dsRNA in Diaphorina citri

Abstract:Background: The insect cuticle consists of chitin fibers and a protein matrix, which plays an important role in protecting the body from invasion of various pathogens and prevents water loss. Periodic synthesis and degradation of the cuticle is required for the growth and development of insects. Key genes involved in cuticle formation have long been considered a potential target for pest control. Results: In this study, a member of the RR-2 subfamily of cuticular protein 8 (DcCP8) was identified from the Diaphorina citri genome database. Immunofluorescence analysis suggested that DcCP8 was mainly located in the Diaphorina citri exocuticle and can be induced to up-regulate 12 h following 20-hydroxyecdysone (20E) treatment. Silencing of DcCP8 by RNA interference (RNAi) significantly disrupted the metamorphosis to the adult stage, and improved the permeability of the cuticle. Transmission electron microscopy (TEM) analysis revealed that the synthesis of the exocuticle was impressed after silencing of DcCP8. Furthermore, the recombinant DcCP8 protein exhibited chitin-binding properties in vitro, down-regulation of DcCP8 significantly inhibited expression levels of chitin metabolism-related genes. Additionally, a sprayable RNAi method targeting DcCP8 based on star polycation (SPc) nanoparticles-wrapped double-stranded RNA (dsRNA) significantly increased Diaphorina citri mortality. Transcriptome sequencing further confirmed that genes associated with the endocytic pathway and immune response were up-regulated in Diaphorina citri after SPc treatment. Conclusions: The current study indicated that DcCP8 is critical for the formation of Diaphorina citri exocuticles, and lays a foundation for Diaphorina citri control based on large-scale dsRNA nanoparticles. © 2024 Society of Chemical Industry.
A spray-induced gene silencing strategy for Spodoptera frugiperda oviposition inhibition using nanomaterial-encapsulated dsEcR

Abstract:Although RNAi-based pest management holds great potential as an alternative to traditional chemical control, its efficiency is restricted by dsRNA instability and limited cellular uptake. Using nanomaterials to facilitate dsRNA delivery has shown promise in solving these challenges. In this study, we firstly used RNAi to investigate the role of the juvenile hormone and ecdysteroid signaling pathways genes in reproduction of Spodoptera frugiperda, the fall armyworm. Females in knocked-down treatments of any of the Met, EcR, and USP genes had greatly reduced fertility with the most pronounced inhibitory effects on oviposition observed following EcR knockdown, and thus the dsEcR could be a candidate target for RNAi-based oviposition inhibitory agency. Then a combinatorial spray-induced and nanocarrier-delivered gene silencing (SI-NDGS) approach that targeted EcR was conducted. At 72 h post-spay, the transcript levels of EcR and the oviposition were successfully reduced and inhibited. These findings support the groundwork for further developing novel RNAi-based pest management strategies for S. frugiperda.
Microfluidic-Based dsRNA Delivery Nanoplatform for Efficient Spodoptera exigua Control

Abstract:Nanotechnology-based RNA interference (RNAi) offers a promising approach to pest control. However, current methods for producing RNAi nanopesticides are mainly implemented in a batch-to-batch manner, lacking consistent quality control. Herein, we present a microfluidic-based nanoplatform for RNA nanopesticide preparation using lipid nanoparticles (LNPs) as nanocarriers, taking advantage of the enhanced mass transfer and continuous processing capabilities of microfluidic technology. The dsRNA@LNPs were rapidly formed within seconds, which showed uniform size distribution, improved leaf wettability, and excellent dispersion properties. The delivery efficiency of dsRNA@LNPs was evaluated by targeting the chitin synthetase B (CHSB) gene ofSpodoptera exigua. The dsRNA@LNPs can effectively resist nuclease-rich midgut fluid degradation. Importantly, dsCHSB@LNPs exhibited increased mortality rates, significant reduction of larvae growth, and enhanced gene suppression efficiency. Therefore, a continuous nanoplatform for RNAi nanopesticide preparation is demonstrated by utilizing microfluidic technology, representing a new route to produce RNAi nanopesticides with enhanced quality control and might accelerate their practical applications.

Abstract:Expression of double-stranded RNAs in plastids offers great potential for the efficient control of chewing insects. However, many insect pests do not consume plant tissue but rather feed on the host plant by sucking sap from the vascular system. Whether or not plastid-mediated RNA interference (RNAi) can be employed to control sap-sucking insects is unknown. Here, we show that five species of sap-sucking hemipteran insects acquire plastid RNA upon feeding on plants. We generated both nuclear transgenic and transplastomic tobacco plants expressing double-stranded RNAs targeting the MpDhc64C gene, a newly identified efficient target gene of RNAi whose silencing causes lethality to the green peach aphid Myzus persicae. In a whole-plant bioassay, transplastomic plants exhibited significant resistance to aphids, as evidenced by reduced insect survival, impaired fecundity, and decreased weight of survivors. The protective effect was comparable with that conferred by the best-performing nuclear transgenic plants. We found that the proportion of aphids on mature leaves of transplastomic plants was significantly lower compared with that of nuclear transgenic plants. When aphids were allowed to infest only the mature leaves, transplastomic plants grew significantly faster and were overall better protected from the pest compared with nuclear transgenic plants. When monitored by electrical-penetration-graph analyses and aphid avoidance response experiments, the insects displayed remarkable alterations in feeding behavior, which was different in nuclear transgenic and transplastomic plants, likely reflecting specific avoidance strategies to toxic RNA molecules. Taken together, our study demonstrates that plastid-mediated RNAi provides an efficient strategy for controlling at least some sap-sucking insect pests, even though there is most likely no or only very little chloroplast RNA in the sap.

Abstract:The western flower thrips, Frankliniella occidentalis, is a serious pest causing both direct feeding damage and indirect harm by transmitting the tomato spotted wilt virus. A spraying double-stranded RNA (dsRNA) targeted at the vacuolar-type ATPase (vATPase) gene was developed and demonstrated high insecticidal activity in the laboratory but less effective in field applications. To improve control efficacy under field conditions, three strategies were explored in this study. First, to identify a more efficient RNA interference (RNAi) target, dsRNA specific to the Snf7 gene was tested alongside dsRNA targeting vATPase, and both were found to be similarly effective in controlling the thrips. Second, to elucidate the factors contributing to dsRNA resistance, dsRNA-degrading enzymes were annotated and their physiological roles in diminishing RNAi efficacy were investigated. Third, to suppress the dsRNA degradation from the dsRNase activities and protect it in field conditions, the dsRNA was encapsulated with chitosan. This formulation enhanced the dsRNA's resistance to environmental stressors such as ultraviolet light and the digestive enzymes in the thrips' gut. Additionally, the chitosan formulation specifically increased the RNAi efficacy, likely by facilitating more efficient entry into the target cells, thus bolstering the insecticidal activity of the dsRNA. The formulated dsRNA was applied on F. occidentalis infesting the hot peppers in a greenhouse at a concentration of 500 ppm, demonstrating an 82.4% control efficacy compared with 59.2% control efficacy observed with the application of naked dsRNA. This study further demonstrated an enhancement in the spectrum of control by combining dsRNAs specific to three distinct thrips species, while the mixture showed no adverse effects on non-target insects, such as the lepidopteran Spodoptera exigua. Collectively, these findings reveal that the chitosan formulation of dsRNA not only improves control efficacy under field conditions but also broadens the control spectrum against three different thrips pests.
Nanoparticle-delivered RNAi-based pesticide target screening for the rice pest white-backed planthopper and risk assessment for a natural predator

Abstract:Vacuolar-type (H+)-ATPase (vATPase) is a conserved multi-subunit eukaryotic enzyme composed of 14 subunits that form a functional complex consisting of an ATP-hydrolytic domain (V1) and a proton-translocation domain (V0). ATP hydrolysis and subsequent H+ translocation rely heavily on a fully assembled V1/V0 complex. Since vATPase is crucial for insect survival, it is a viable molecular target for pest control. However, detailed functional analyses of the 14 subunits and their suitability for pest control have not been fully explored in a single insect species. In this study, we identified 22 vATPase subunit transcripts that correspond to 13 subunits (A1, A2, B, C, D, E, F, G, H, a1, a2, c and d) in the white-backed planthopper (WBPH), Sogatella furcifera, a major hemipteran pest of rice. RNAi screens using microinjection and spray-based methods revealed that the SfVHA-F, SfVHA-a2 and SfVHA-c2 subunits are critical. Furthermore, star polymer (SPc) nanoparticles were utilized to conduct spray-induced and nanoparticle-delivered gene silencing (SI-NDGS) to evaluate the pest control efficacy of RNAi targeting the SfVHA-F, SfVHA-a2 and SfVHA-c2 transcripts. Target mRNA levels and vATPase enzymatic activity were both reduced. Honeydew excreta was likewise reduced in WBPH treated with dsRNAs targeting SfVHA-F, SfVHA-a2 and SfVHA-c2. To assess the environmental safety of the nanoparticle-wrapped dsRNAs, Cyrtorhinus lividipennis Reuter, a major natural enemy of planthoppers, was also sprayed with dsRNAs targeting SfVHA-F, SfVHA-a2 and SfVHA-c2. Post-spray effects of dsSfVHA-a2 and dsSfVHA-c2 on C. lividipennis were innocuous. This study identifies SfVHA-a2 and SfVHA-c2 as promising targets for biorational control of WBPH and lays the foundation for developing environment-friendly RNAi biopesticides.

Characterization of CYP303A1 and its potential application based on ZIF-8 nanoparticle-wrapped dsRNA in Nilaparvata lugens (Stål)

Abstract:Background: RNA interference (RNAi) technology has been put forward as a promising method for pest control and resistance management. Mining highly efficient lethal genes and constructing stable double-stranded RNA (dsRNA) delivery systems are of great significance to improve the application potential of RNAi technology. Results: In this study, we characterized a molting-related gene, NlCYP303A1, in Nilaparvata lugens that was highly expressed in the cuticle and at the end stages of each instar in nymphs. Silencing the expression of NlCYP303A1 in N. lugens resulted in a deformed phenotype and a significant increase in mortality. Furthermore, interfering with NlCYP303A1 changed the relative expression of key genes in the chitin synthesis and degradation pathway. Finally, we used the nanocarrier zeolitic imidazolate framework-8 (ZIF-8) to load dsNlCYP303A1, forming a complex denoted as dsNlCYP303A1@ZIF-8. The results of both feeding and rice-seedling dip experiments indicated that the expression of NlCYP303A1 was dramatically and persistently suppressed by the dsNlCYP303A1@ZIF-8 treatment, compared with treatment with dsNlCYP303A1, suggesting that ZIF-8 can enhance the interference efficiency as well as the stability of dsNlCYP303A1. Conclusions: Our results demonstrate that the lethal gene NlCYP303A1 can be employed as an excellent target for RNAi technology by loading onto a nano-delivery system, and provide new insights into the creation of innovative pest control approaches. © 2024 Society of Chemical Industry.

Unfortunately, the pressing need for new classes of antibiotics is still poorly met by the pharmaceutical industry, as illustrated by the recent call by the Infectious Diseases Society of America (IDSA) for a global commitment to develop ten new antibiotics by 2020. In this respect, the recent discovery of two new antibiotics, platensimycin (1) and platencin (2), represented a potential breakthrough in antibiotic research.

Abstract:Novel green technology RNA pesticide has been put forward as a promising method for plant protection, but the instability and short life disadvantage of double-stranded RNA (dsRNA) seriously constrain the commercialization process of RNA pesticide. Herein, a star polycation (SPc)-based gene/drug delivery system was constructed for co-delivering hemocytin (hem) dsRNA and botanical pesticide matrine to develop a novel multicomponent nano-pesticide for achieving sequential bioactivity against devastating green peach aphids. The self-assembled matrine/SPc complex could be further complexed with dsRNA through electrostatic interaction and hydrogen bond to form nano-sized matrine/SPc/dsRNA complex. The screened dshem fragment led to efficient gene silencing and high mortality rate through SPc-based topical application, and the main lethal mechanism was through down-regulating hem gene to result in severe bacterial infection. The dshem fragment was successfully expressed in pET28-BL21 (DE3) RNaseIII- system to prepare matrine/SPc/dshem complex. Both initial acting time and persistence of matrine/SPc/dshem complex were remarkably improved in field, which overcame the short life disadvantage of dshem and slow-acting property of matrine simultaneously. To our knowledge, it is the first success to co-deliver dsRNA and drug by nanoparticle-mediated delivery system in agricultural field.

Abstract:Spray-induced gene silencing represents an eco-friendly approach for crop protection through the use of double-stranded RNA (dsRNA) to activate the RNA interference (RNAi) pathway, thereby silencing crucial genes in pathogens. The major challenges associated with dsRNA are its limited stability and poor cellular uptake, necessitating repeated applications for effective crop protection. In this study, RNA nanoparticles (NPs) were proposed as effectors in plants and pathogens by inducing the RNAi pathway and silencing gene expression. RNA structural motifs, such as hairpin-loop, kissing-loop, and tetra-U motifs, were used to link multiple siRNAs into a long, single-stranded RNA (lssRNA). The lssRNA, synthesized in Escherichia coli, self-assembled into stable RNA nanostructures via local base pairing. Comparative analyses between dsRNA and RNA NPs revealed that the latter displayed superior efficacy in inhibiting spore germination and mycelial growth of Botrytis cinerea. Moreover, RNA NPs had a more robust protective effect on plants against B. cinerea than did dsRNA. In addition, RNA squares are processed into expected siRNA in plants, thereby inhibiting the expression of the target gene. These findings suggest the potential of RNA NPs for use in plant disease control by providing a more efficient and specific alternative to dsRNA without requiring nanocarriers.
