Abstract:Magnetic-actuated miniature robots have sparked growing interest owing to their promising potential in biomedical applications, such as noninvasive diagnosis, cargo delivery, and microsurgery. Innovations are required to combine biodegradable materials with flexible mobility to promote the translation of magnetic robots towards in vivo application. This study proposes a biodegradable magnetic hydrogel robot (MHR) with multimodal locomotion and collective behavior through magnetic-assisted fabrication. The MHRs with aligned magnetic chains inside its structure has a larger maximum motion speed under the rotating magnetic fields in comparison with the robots without magnetic alignment. By reconfiguring the external magnetic fields, three kinds of stable motion modes (tumbling, spinning, and wobbling modes.) of the individual MHRs can be triggered, while flexible conversion can be achieved between each motion mode. The motion mechanism of each motion mode under diverse rotating magnetic fields has been analyzed. The collective behavior of the MHRs, which is triggered by the magnetic dipole force, can enhance the motion performance and pass through sophisticated terrains. Furthermore, the experimental results demonstrate that the assembled MHRs can execute complicated tasks such as targeted cargo delivery. The proposed MHRs with multimodal locomotion and assembled behavior show effective motion efficiency, flexible maneuverability, and remarkable targeting ability, providing a new choice for magnetic robots in biomedical applications.

Abstract:Miniature magnetic-driven robots with multimode motions and high-precision pose sensing capacity (position and orientation) are greatly demanded in in situ manipulation in narrow opaque enclosed spaces. Various magnetic robots have been carried out, whereas their deformations normally remain in single mode, and the lack of the robot’s real-time status leads to its beyond-sight remagnetization and manipulation being impossible. The function integration of pose sensing and multimode motion is still of challenge. Here, a multimotion thin-film robot is created in a novel multilayer structure with a magnetic-driven layer covered by a heating-sensing conductive layer. Such a heating-sensing layer not only can segmentally and on-demand heat the magnetic-driven layer for in situ magnetization reprogramming and multimode motions but also can precisely detect the robot’s pose (position and orientation) from its electrical-resistance effect by creating a small deformation under preset magnetic fields. Under the integration of reprogramming and sensing, necessary multimode motions, i.e., swimming, rolling, crawling, and obstacle-crossing, are achieved under a reprogramming field B Repr of 10 mT, and high-precision poses sensing with an accuracy of ± 3 mm in position and ± 2.5° in orientation is obtained even under a low magnetic strength of B Sens of 5 mT, which combined help realize accurate out-of-sight manipulations in the enclosed space environment. Finally, a gastroscope robot for stomach drug delivery has been demonstrated for more gastrointestinal medical treatments.

Abstract:Untethered miniature robots, which can be used in applications ranging from cargo delivery to biopsy, demonstrate stunning performance in gastrointestinal (GI) tract treatment. The environmental terrains within the GI tract are complex and challenging for robots of limited size to pass through, where tissue mucus and winding surfaces tend to hinder robot movement. Herein, we report a multifunctional magnetic miniature truck robot (MMTR) that generates sufficient propulsion force to resist external environmental interactions. Its mechanical structure is optimized for integrating multiple functions, including climbing over obstacles, controlled drug dispersion, and cargo release. Using a programmed external magnetic field, the MMTR accomplishes transport of smaller-sized miniature robots by carrying them, allowing traversal of tortuous, branching, and restricted environments through adaptive morphology transformations. The introduced truck-unloading approach enables the MMTR to unload, detach, and retrieve sub-robots, demonstrating the significant potential of miniature robots for targeted transport.

Abstract:With the rapid development of micro/nanomanufacturing technology, a variety of multifunctional micro/nanorobots have emerged. In particular, magnetic actuation-based micro/nanorobots have attracted much interest due to their advantages of untethered contact, remote control, noninvasiveness, and high tissue-penetrating capability. Various magnetic micro/nanorobots have been designed and applied in biomedicine, which indicates the great potential of clinical transformation. This article reviews the new advancements of engineering magnetic micro/nanorobots for versatile biomedical applications, ranging from targeted delivery, minimally invasive surgery, cellular and intracellular measurement, and intelligent sensing to detoxification and antibacterial applications. The controllability, visualization, and biosafety aspects of magnetically manipulated micro/nanorobots are summarized. In particular, the challenges being faced are discussed to outlook future prospective development.

Abstract:Micro‐/nanorobots enter the body in a minimally invasive way to treat diseases, which provides a new idea for the development of modern biomedicine. Herein, the fabrication of micro/nanorobots, the motion mechanism driven by the magnetic field, and the latest progress in medical application are discussed. The future development and challenges of micro/nanorobots are further prospected. Micro‐/nanorobots have tremendous potential in biomedical applications, as they efficiently and accurately perform local diagnosis and targeted therapy. A magnetic field provides a wireless mean to drive and control micro/nanorobots. This method requires no fuel and is convenient for debugging, reconfiguration, and programmability; it is recyclable and allows nondestructive penetration of biological tissue. The successful integration of well‐designed robots, remote actuation systems, and imaging techniques has become a crucial step toward biomedical applications, particularly in vivo implementations. Herein, the fabrication methods of micro/nanorobots are examined, the driving mechanism of magnetic micro/nanorobots is explained, and the development of micro/nanorobots as transport tools to load and deliver drugs, biological reagents, and live cells is traced. Applications of these micro/nanorobots are expanding as surgical tools for sample collection, ophthalmic surgery, and biofilm degradation and as tools for in vivo and in vitro imaging relying on light, sound, and magnetism. Finally, the challenges faced by micro/nanorobots in in vivo applications and their future prospects are discussed.

Abstract:As an emerging field of robotics, magnetic-field-controlled soft microrobot has broad application prospects for its flexibility, locomotion diversity, and remote controllability. Magnetic soft microrobots can perform multimodal locomotion under the control of a magnetic field, which may have potential applications in precision medicine. However, previous research studies mainly focus on new locomotion in a relatively ideal environment, lacking exploration on the ability of magnetic microrobot locomotion to resist external disturbances and proceed in a nonideal environment. Here, a porous silica-doped soft magnetic microrobot is constructed for enhanced stability of multimodal locomotion in the nonideal biological environment. Porous silica spheres are doped into a NdFeB-silicone elastomer base, improving adhesion properties and refining the comprehensive mechanical properties of the microrobot. Multimodal locomotions are achieved, and the influence of porous silica doping on the stability of each locomotion in a nonideal environment is explored in depth. Motions in nonideal circumstances such as climbing, loading, current rushing, wind blowing, and obstacle hindering are conducted successfully with porous silica doping. Such a stability-enhanced multimodal locomotion system can be used in biocatalysis and thrombus removal, and its prospect for precision medicine is highlighted by in vivo demonstration of multimodal locomotion with nonideal disturbance.

A Magnetic Millirobot Walks on Slippery Biological Surfaces for Targeted Cargo Delivery

Abstract:Small-scale robots hold great potential for targeted cargo delivery in minimally-inv asive medicine. However, current robots often face challenges to locomote efficiently on slip pery biological tissue surfaces, especially when loaded with heavy cargos. Here, we report a magnetic millirobot that can walk on rough and slippery biological tissues by anchoring itself on the soft tissue surface alternatingly with two feet and reciprocally rotating the body to mov e forward. We experimentally studied the locomotion, validated it with numerical simulations and optimized the actuation parameters to fit various terrains and loading conditions. Further more, we developed a permanent magnet set-up to enable wireless actuation within a huma n-scale volume which allows precise control of the millirobot to follow complex trajectories, cl imb vertical walls, and carry cargo up to four times of its own weight. Upon reaching the targ et location, it performs a deployment sequence to release the liquid drug into tissues. The ro bust gait of our millirobot on rough biological terrains, combined with its heavy load capacity, make it a versatile and effective miniaturized vehicle for targeted cargo delivery.

Abstract:A magnetic barbell‐shaped soft microrobot (MBS2M) is proposed inspired by the natural locomotion of crucian carp with the advantages of environmental adaptation, flexible movement, precise positioning, and multifunctionality. The proposed microrobot provides a "all‐in‐one" solution for sophisticated tasks in complex environments, which has excellent application potential and is expected to play an important role in biomedical applications such as drug delivery, detection, and diagnosis in vivo. The development of environmentally adaptive solutions for magnetically actuated microrobots to enable targeted delivery in complex and confined fluid environments presents a significant challenge. Inspired by the natural locomotion of crucian carp, a barbell‐shaped soft microrobot (MBS2M) is proposed. A mechano‐electromagnetic hybrid actuation system is developed to generate oscillating magnetic fields to manipulate the microrobot. The MBS2M can seamlessly transition between three fundamental locomotion modes: fast navigation (FN), high‐precision navigation (HPN), and fixed‐point rotation (FPR). Moreover, the MBS2M can move in reverse without turning. The multimodal locomotion endows the MBS2M's adaptability in diverse environments. It can smoothly pass through confined channels, climb over obstacles, overcome gravity for vertical motion, track complex pathways, traverse viscous environments, overcome low fluid resistance, and navigate complex spaces mimicking in vivo environments. Additionally, the MBS2M is capable of drug loading and release in response to ultrasound excitation. In an ex vivo porcine liver vein, the microrobot demonstrated targeted navigation under ultrasound guidance, showcasing its potential for specialized in vivo tasks.

Abstract:Magnetic microgrippers, with their miniaturized size, flexible movement, untethered actuation, and programmable deformation, can perform tasks such as cell manipulation, targeted drug delivery, biopsy, and minimally invasive surgery in hard-to-reach regions. However, common external magnetic-field-driving devices suffer from low efficiency and utilization due to the significant size disparity with magnetic microgrippers. Here, we introduce a microgripper robot (MGR) driven by end electromagnetic and permanent magnet collaboration. The magnetic field generated by the microcoils can be amplified by the permanent magnets and the direction can be controlled by changing the current, allowing for precise control over the opening and closing of the magnetic microgripper and enhancing its operational range. Experimental results demonstrate that the MGR can be flexibly controlled in complex constrained environments and is highly adaptable for manipulating objects. Furthermore, the MGR can achieve planar and antigravity object grasping and transportation within complex simulated human cavity pathways. The MGR's grasping capabilities can also be extended to specialized tasks, such as circuit connection in confined spaces. The MGR combines the required safety and controllability for in vivo operations, making it suitable for potential clinical applications such as tumor or abnormal tissue sampling and surgical assistance.

Abstract:Untethered magnet‐driven microrobots play an increasingly important role in various biomedical applications. Incorporating bionic technology into microrobot design is an emerging way to improve the work efficiency of microrobots. Herein, a magnetically powered and frictional anisotropy‐based microwalker that can be potentially used in in vivo nonliquid‐filled environment is proposed. The microwalker is constructed by two rigid segments with an equal length of 70 μm, connected by a rigid joint. Parallel gecko setae‐like tentacles are placed at the bottom of the segments as contact feet to generate friction with the contact surface. The microwalker is integrally fabricated from biocompatible materials with 3D laser lithography based on two‐photon polymerization. The microwalker can be well controlled to move in low‐Reynolds (Re)‐number regimes under an external oscillating magnetic field. In addition to moving in a liquid environment as existing microswimmers, the microwalker can move in surface in a nonliquid‐filled environment. It can also climb the slope driven by the planar magnetic field only. Several experiments were conducted to demonstrate good motion capability of the microwalker. This study provides a new solution to microrobot design for future biomedical applications.

Abstract:Magnetic microrobots are ideal for medical applications owing to their deep tissue penetration, precise control, and flexible movement. After decades of development, various magnetic microrobots have been used to achieve medical functions such as targeted delivery, cell manipulation, and minimally invasive surgery. This review introduces the research status and latest progress in the design and control systems of magnetic medical microrobots from a system integration perspective and summarizes the advantages and limitations of the research to provide a reference for developers. Finally, the future development direction of magnetic medical microrobot design and control systems are discussed.
High-performance Magnetic Metal Microrobot Prepared by a Two-Photon Polymerization and Sintering Method.

Abstract:Magnetically-actuated microrobots (MARs) exhibit great potential in biomedicine owing to their precise navigation, wireless actuation and remote operation in confined space. However, most previously explored MARs unfold the drawback of hypodynamic magnetic torque due to low magnetic content, leading to their limited applications in controlled locomotion in fast-flowing fluid and massive cargo carrying to the target position. Here, we report a high-performance pure-nickel magnetically-actuated microrobot (Ni-MAR), prepared by a femtosecond laser polymerization followed by sintering method. Our Ni-MAR possesses a high magnetic content (∼90 wt%), thus resulting in enhanced magnetic torque under low-strength rotating magnetic fields, which enables the microrobot to exhibit high-speed swimming and superior cargo carrying. The maximum velocity of our Ni-MAR, 12.5 body lengths per second, outperforms the velocity of traditional helical MARs. The high-speed Ni-MAR is capable of maintaining controlled locomotion in fast-flowing fluid. Moreover, the Ni-MAR with massive cargo carrying capability can push a 200-times heavier microcube with translation and rotation motion. A single cell and multiple cells can be transported facilely by a single Ni-MAR to the target position. This work provides a scheme for fabricating high-performance magnetic microrobots, which holds great promise for targeted therapy and drug delivery in vivo.
Millimeter-scale flexible robots with programmable three-dimensional magnetization and motions

Abstract:Flexible magnetic small-scale robots use patterned magnetization to achieve fast transformation into complex three-dimensional (3D) shapes and thereby achieve locomotion capabilities and functions. These capabilities address current challenges for microrobots in drug delivery, object manipulation, and minimally invasive procedures. However, possible microrobot designs are limited by the existing methods for patterning magnetic particles in flexible materials. Here, we report a method for patterning hard magnetic microparticles in an elastomer matrix. This method, based on ultraviolet (UV) lithography, uses controlled reorientation of magnetic particles and selective exposure to UV light to encode magnetic particles in planar materials with arbitrary 3D orientation with a geometrical feature size as small as 100 micrometers. Multiple planar microrobots with various sizes, different geometries, and arbitrary magnetization profiles can be fabricated from a single precursor in one process. Moreover, a 3D magnetization profile allows higher-order and multi-axis bending, large-angle bending, and combined bending and torsion in one sheet of polymer, creating previously unachievable shape changes and microrobotic locomotion mechanisms such as multi-arm power grasping and multi-legged paddle crawling. A physics-based model is also presented as a design tool to predict the shape changes under magnetic actuation.
Magnetic-chemotactic hybrid microrobots with precise remote targeting capability

Abstract:Micro/nanorobots (MNRs) hold great promise for various applications due to their capability to execute complex tasks in hard-to-reach narrow cavities. However, the developed magnetic MNRs, as marionettes of external magnetic fields, lack building-in intelligence for self-targeting, while chemotactic MNRs suffer from limited self-targeting range. Here, we demonstrate magnetic-chemotactic ZnO/Fe-Ag Janus microrobots (JMRs) capable of rapid, remote self-targeting for bacterial elimination. The JMRs utilize the magnetic Fe engine for coarse navigation from a distance, allowing for external control to swiftly guide them to the vicinity of a hidden/uncharted target that establishes a local chemical gradient ([CO2] or [H+] gradient). Once in proximity, the inherent chemotaxis of the JMRs takes over, enabling them to autonomously accumulate at the target site along the chemical gradient in high precision. Upon reaching the target, the ZnO/Fe-Ag JMRs can release Zn2+ and Ag+ to eliminate bacteria residing there. The proposed strategy of integrating on-board chemotaxis with external field-driven propulsion paves the way for efficient precise therapies using MNRs, especially in targeted drug/energy delivery involving remote hidden or uncharted targets.

Abstract:Achieving precise control over the motion position and attitude direction of magnetic microrobots remains a challenging task in the realm of microrobotics. To address this challenge, our research team has successfully implemented synchronized control of a microrobot's motion position and attitude direction through the integration of electromagnetic coils and permanent magnets. The whole drive system consists of two components. Firstly, a stepper motor propels the delta structure, altering the position of the end-mounted permanent magnet to induce microrobot movement. Secondly, a programmable DC power supply regulates the current strength in the electromagnetic coil, thereby manipulating the magnetic field direction at the end and influencing the permanent magnet's attitude, guiding the microrobot in attitude adjustments. The microrobot used for performance testing in this study was fabricated by blending E-dent400 photosensitive resin and NdFeB particles, employing a Single-Layer 4D Printing System Using Focused Light. To address the microrobot drive system's capabilities, experiments were conducted in a two-dimensional and three-dimensional track, simulating the morphology of human liver veins. The microrobot exhibited an average speed of 1.3 mm/s (movement error ± 0.5 mm). Experimental results validated the drive system's ability to achieve more precise control over the microrobot's movement position and attitude rotation. The outcomes of this study offer valuable insights for future electromagnetic drive designs and the application of microrobots in the medical field.

Abstract:Biohybrid bacteria–based microrobots are increasingly recognized as promising externally controllable vehicles for targeted cancer therapy. Magnetic fields in particular have been used as a safe means to transfer energy and direct their motion. Thus far, the magnetic control strategies used in this context rely on poorly scalable magnetic field gradients, require active position feedback, or are ill-suited to diffuse distributions within the body. Here, we present a magnetic torque–driven control scheme for enhanced transport through biological barriers that complements the innate taxis toward tumor cores exhibited by a range of bacteria, shown for Magnetospirillum magneticum as a magnetically responsive model organism. This hybrid control strategy is readily scalable, independent of position feedback, and applicable to bacterial microrobots dispersed by the circulatory system. We observed a fourfold increase in translocation of magnetically responsive bacteria across a model of the vascular endothelium and found that the primary mechanism driving increased transport is torque-driven surface exploration at the cell interface. Using spheroids as a three-dimensional tumor model, fluorescently labeled bacteria colonized their core regions with up to 21-fold higher signal in samples exposed to rotating magnetic fields. In addition to enhanced transport, we demonstrated that our control scheme offers further advantages, including the possibility for closed-loop optimization based on inductive detection, as well as spatially selective actuation to reduce off-target effects. Last, after systemic intravenous injection in mice, we showed significantly increased bacterial tumor accumulation, supporting the feasibility of deploying this control scheme clinically for magnetically responsive biohybrid microrobots.

Multimodal locomotions are achieved, and the influence of porous silica doping on the stability of each locomotion in nonideal environment is explored in depth. Motions in nonideal circumstances such as climbing, loading, current rushing, wind blowing, and obstacle hindering are conducted successfully with porous silica doping. Such a stability-enhanced multimodal locomotion system can be used in biocatalysis as well as thrombus removal, and its prospect for precision medicine is highlighted by in vivo demonstration of multimodal locomotion with nonideal disturbance.

Abstract:Mimicry of motile microorganisms offers new concepts and strategies to design and manipulate microrobots. As a representative instance, Escherichia coli switches its motility between swimming and tumbling to adapt to surroundings. Inspired by this, we present a tubular microrobot behaving rolling and tumbling motion modes analogous to the motion of Escherichia coli, as a result of the anisotropic magnetized status with radially magnetized status at rolling and longitudinally magnetized one at tumbling. The motion behavior of microrobot as rolling or tumbling is determined by the frequency of rotating magnetic field. The threshold frequency for switching is closely related to the amplitude of magnetic field, viscosity of solution and size of microrobot. The net locomotion near substrate comes from the friction and pressure, and the motion switching is due to the magnetized statuses and discrepancy of drag force. The switch from rolling to tumbling endows the microrobot with adaptive locomotion to cross obstacles. Furthermore, microfluidic manipulation through rotational microrobot is achieved based on the local microvortex, which is generated from the hydrodynamic interaction between microrobot and substrate. This manipulation is applied in cargo delivery and release by altering surrounding microfluid via switchable motion modes. Abstract:Recent strides in microfabrication technologies offer important possibilities for developing microscale robotic systems with enhanced power, functionality and versatility. Previous microrobots fabricated by lithographic techniques usually lack the ability to adaptively deform in confined and constricted spaces and navigate through, therefore hindering their applications in complex biological environments. Here, a microfluidic strategy is combined with a dip-coating process for continuous fabrication of soft helical structures with controllable mechanical property as magnetically propelled microrobots, capable of actively propelling through narrow and sinuous microchannels. Because of their self-adaptive deformation capability, the magnetically propelled soft microrobots can actively navigate through a narrow opening, 2.21 times smaller than the sectional area of the microrobot, and a U-shape-bent capillary, directed by a programmed magnetic field. Additionally, the soft microrobot demonstrates increased swimming speed in a fluid of high viscosity, because of the adaptive tightening deformation of the helix when swimming. This new magnetically propelled soft microrobot and its attractive performance will open up new possibilities for biomedical operation at the micro and nanoscale.
