Abstract:A subset of microorganisms that perform respiration can endogenously utilize insoluble electron donors, such as Fe(II) or a cathode, in a process called extracellular electron transfer (EET). However, it is unknown whether similar endogenous EET can be performed by primarily fermentative species like lactic acid bacteria. We report for the first time electron uptake from a cathode by Lactiplantibacillus plantarum , a primarily fermentative bacteria found in the gut of mammals and in fermented foods. L. plantarum consumed electrons from a cathode and coupled this oxidation to the reduction of both an endogenous organic (pyruvate) and an exogenous inorganic electron acceptor (nitrate). This electron uptake from a cathode reroutes glucose fermentation toward lactate degradation and provides cells with a higher viability upon sugar exhaustion. Moreover, the associated genes and cofactors indicate that this activity is mechanistically different from that one employed by lactic acid bacteria to reduce an anode and to perform respiration. Our results expand our knowledge of the diversity of electroactive species and of the metabolic and bioenergetic strategies used by lactic acid bacteria.

Identifying Key Properties that Drive Redox Mediator Activity in Lactiplantibacillus plantarum

Abstract:Lactiplantibacillus plantarum is known to utilize exogenous small molecule quinone mediators to perform extracellular electron transfer (EET), which allows it to produce a detectable current in a bioelectrochemical system. Utilization of quinone mediators by L. plantarum requires a type-II NADH dehydrogenase (Ndh2), however small structural variations in the core of 1,4-naphthoquinone EET mediators yields significantly different current outputs. Herein, we assembled a library of 30 naphthoquinone-based EET mediators in order to probe the important physicochemical properties and biochemical interactions that are responsible for Ndh2-dependent EET in L. plantarum. The library was designed with inspiration from naturally-occurring metabolites, and was assembled focused on structural modifications that diversified polarity (cLogP), reduction potential (Eo), and Ndh2 binding affinity as these properties are hypothesized to be driving EET activity. In general, Ndh2-dependent EET activity in an iron(III) oxide nanoparticle assay significantly correlates to the mediator's polarity and binding affinity. Five mediators were analyzed in BESs with L. plantarum and each generated Ndh2-dependent current with low background signal. Importantly, the amine containing mediators yielded incredibly stable current output over the course of the experiment (up to 5 days). These findings increase our understanding of structure-activity relationships for quinone-mediated EET and provide mediators for bioelectronic sensing.

Abstract:ABSTRACT Energy conservation in microorganisms is classically categorized into respiration and fermentation, however recent work shows some species can use mixed or alternative bioenergetic strategies. We explored the utility of a flavin-based extracellular electron transport (FLEET) system for energy conservation within diverse lactic acid bacteria (LAB), microorganisms that mainly rely on fermentative metabolism and are important in food fermentations. The LAB Lactiplantibacillus plantarum uses extracellular electron transfer to increase its NAD + /NADH ratio, generate more ATP through substrate-level phosphorylation and accumulate biomass more rapidly. This novel, hybrid metabolism was dependent on a type-II NADH dehydrogenase (Ndh2) and conditionally required a flavin-binding extracellular lipoprotein (PplA) in the FLEET system to confer increased fermentation yield, metabolic flux, and environmental acidification in both laboratory media and food fermentation. The discovery of a single pathway that blends features of fermentation and respiration expands our knowledge of energy conservation metabolism and provides immediate biotechnology applications.
The convergence of lactic acid microbiomes and metabolites in long-term electrofermentation

Abstract:Regulating electron transfer in predominantly fermentative microbiomes has broad implications in environmental, chemical, food, and medical fields. Here we demonstrate electrochemical control in fermenting food waste, digestate, and wastewater to improve lactic acid production. We hypothesize that applying anodic potential will expedite and direct fermentation towards lactic acid. Continued operation that introduced epi/endophytic communities (Lactococcus, Lactobacillus, Weissella) to pure culture Lactiplantibacillus plantarum reactors with static electrodes was associated with the loss of anode-induced process intensification despite 80% L. plantarum retention. Employing fluidized electrodes discouraged biofilm formation and extended electrode influence to planktonic gram-positive fermenters using mediated extracellular electron transfer. While short-term experiments differentially enriched Lactococcus and Klebsiella spp., longer-term operations indicated convergent microbiomes and product spectra. These results highlight a functional resilience of environmental fermentative microbiomes to perturbations in redox potential, underscoring the need to better understand electrode induced polymicrobial interactions and physiological impacts to engineer tunable open-culture or synthetic consortia.

Abstract:Microbial electrosynthesis is the process of supplying electrons to microorganisms to reduce CO2 and yield industrially relevant products. Such systems are limited by their requirement for high currents, resulting in challenges to cell survival. Electrofermentation is an electron-efficient form of microbial electrosynthesis in which a small cathodic or anodic current is provided to a culture to alter the oxidation–reduction potential of the medium and, in turn, alter microbial metabolism. This approach has been successfully utilised to increase yields of diverse products including biogas, butanediol and lactate. Biomass conversion to lactate is frequently facilitated by ensiling plant biomass with homofermentative lactic acid bacteria. Although most commonly used as a preservative in ensiled animal feed, lactate has diverse industrial applications as a precursor for the production of probiotics, biofuels, bioplastics and platform chemicals. Lactate yields by lactic acid bacteria (LAB) are constrained by a number of redox limitations which must be overcome while maintaining profitability and sustainability. To date, electrofermentation has not been scaled past laboratory- or pilot-stage reactions. The increasing ease of genetic modification in a wide range of LAB species may prove key to overcoming some of the pitfalls of electrofermentation at commercial scale. This review explores the history of electrofermentation as a tool for controlling redox balance within bacterial biocatalysts, and the potential for electrofermentation to increase lactate production from low-value plant biomass.

Isolation and Characterization of Human Gut Bacteria Capable of Extracellular Electron Transport by Electrochemical Techniques

Abstract:Microorganisms are known to exhibit extracellular electron transfer (EET) in a wide variety of habitats. However, as for the human microbiome which significantly impacts our health, the role and importance of EET has not been widely investigated. In this study, we enriched and isolated the EET-capable bacteria from human gut microbes using an electrochemical enrichment method and examined whether the isolates couple EET with anaerobic respiration or fermentation. Upon the use of energy-rich or minimum media (with acetate or lactate) for electrochemical enrichment with the human gut sample at an electrode potential of +0.4 V [vs. the standard hydrogen electrode (SHE)], both culture conditions showed significant current production. However, EET-capable pure strains were enriched specifically with minimum media, and subsequent incubation using the δ-MnO2-agar plate with lactate or acetate led to the isolation of two EET-capable microbial strains, Gut-S1 and Gut-S2, having 99% of 16S rRNA gene sequence identity with Enterococcus avium (E. avium) and Klebsiella pneumoniae (K. pneumoniae), respectively. While the enrichment involved anaerobic respiration with acetate and lactate, further electrochemistry with E. avium and K. pneumoniae revealed that the glucose fermentation was also coupled with EET. These results indicate that EET couples not only with anaerobic respiration as found in environmental bacteria, but also with fermentation in the human gut.

Abstract:Non-electroactive bacteria (n-EAB), constituting the majority of known bacteria to date, have been underutilized in electrochemical conversion technologies due to their lack of direct electron transfer to electrodes. In this study, we established an electric wiring between n-EAB (gram-positive Bacillus subtilis and gram-negative Escherichia coli ) and an extracellular electrode via a ferrocene-polyethyleneimine-based redox polymer (Fc-PEI). Chronoamperometry recordings indicated that Fc-PEI can transfer intracellular electrons to the extracellular electrode regardless of the molecular organization of PEI (linear or branched) and the membrane structure of bacteria (gram-positive or -negative). As fluorescence staining suggested, Fc-PEI improves the permeability of the bacterial cell membrane, enabling electron carriers in the cell to react with Fc. In addition, experiments with Fc-immobilized electrodes without PEI suggested the existence of an alternative electron transfer pathway from B. subtilis to the extracellular Fc adsorbed onto the cell membrane. Furthermore, we proposed for the first time that the bacteria/Fc-linear PEI modified structure enables selective measurement of immobilized bacterial activity by physically blocking contact between the electrode surface and planktonic cells co-existing in the surrounding media. Such electrodes can be a powerful analytical tool for elucidating the metabolic activities of specific bacteria wired to the electrode even within complex bacterial communities.

Abstract:ABSTRACT Extracellular electron transfer (EET) is a metabolic process that frequently uses quinones to couple intracellular redox reactions with extracellular electron acceptors. The physiological relevance of this metabolism for microorganisms capable of EET but unable to synthesize their own quinones remains to be determined. To address this question, we investigated quinone utilization by Lactiplantibacillus plantarum , a microorganism required for food fermentations, that performs EET and is also a quinone auxotroph. L. plantarum selectively used 1,4-dihydroxy-2-naphthoic acid (DHNA) and more hydrophilic naphthoquinones for EET reduction of insoluble iron (ferrihydrite). However, quinones used for EET also inhibited L. plantarum growth in non-aerated conditions. Transcriptomic analysis showed that DHNA-induced oxidative stress in L. plantarum, but this was alleviated when the electron acceptor, ferric ammonium citrate (FeAC), was included in the growth medium. Although DHNA and FeAC induced L. plantarum EET, this metabolism was still dependent on direct access to environmental electron shuttles. To determine whether quinone-producing food fermentation bacteria could be sources of those electron shuttles, L. plantarum EET was measured after incubation with Lactococcus lactis and Leuconostoc mesenteroides . Quinone-producing L. lactis , but not a quinone-deficient L. lactis Δ menC mutant, increased L. plantarum ferrihydrite reduction and medium acidification through an EET-dependent mechanism. L. plantarum EET was also stimulated by L. mesenteroides , resulting in greater environmental acidification and transient increases in L. plantarum cell numbers. Our findings show that L. plantarum overcomes the toxic effects of exogenous quinones to use those compounds for EET-conferred, ecological advantages during the early stages of food fermentations. IMPORTANCE While quinones are essential for respiratory microorganisms, their importance for microbes that rely on fermentation metabolism is not understood. This gap in knowledge hinders our understanding of anaerobic microbial habitats, such in mammalian digestive tracts and fermented foods. We show that Lactiplantibacillus plantarum, a model fermentative lactic acid bacteria species abundant in human, animal, and insect microbiomes and fermented foods, uses multiple exogenous, environmental quinones as electron shuttles for a hybrid metabolism involving EET. Interestingly, quinones both stimulate this metabolism as well as cause oxidative stress when extracellular electron acceptors are absent. We also found that quinone-producing, lactic acid bacteria species commonly enriched together with L. plantarum in food fermentations accelerate L. plantarum growth and medium acidification through a mainly quinone- and EET-dependent mechanism. Thus, our work provides evidence of quinone cross-feeding as a key ecological feature of anaerobic microbial habitats.

Abstract:The bioelectrochemical methane production from acetate as a non-fermentable substrate, glucose as a fermentable substrate, and their mixture were investigated in an anaerobic sequential batch reactor exposed to an electric field. The electric field enriched the bulk solution with exoelectrogenic bacteria (EEB) and electrotrophic methanogenic archaea, and promoted direct interspecies electron transfer (DIET) for methane production. However, bioelectrochemical methane production was dependent on the substrate characteristics. For acetate as the substrate, the main electron transfer pathway for methane production was DIET, which significantly improved methane yield up to 305.1 mL/g chemical oxygen demand removed (CODr), 77.3% higher than that in control without the electric field. For glucose, substrate competition between EEB and fermenting bacteria reduced the contribution of DIET to methane production, resulting in the methane yield of 288.0 mL/g CODr, slightly lower than that of acetate. In the mixture of acetate and glucose, the contribution of DIET to methane production was less than that of the single substrate, acetate or glucose, due to the increase in the electron equivalent for microbial growth. The findings provide a better understanding of electron transfer pathways, biomass growth, and electron transfer losses depending on the properties of substrates in bioelectrochemical methane production.

Abstract:The low bacteria loading capacity and low extracellular electron transfer (EET) efficiency are two major bottlenecks restricting the performance of the bioelectrochemical systems from practical applications. Herein, we demonstrated that conjugated polymers (CPs) could enhance the bidirectional EET efficiency through the intimate biointerface interactions of CPs-bacteria biohybrid system. Upon the formation of CPs/bacteria biohybrid, thick and intact CPs-biofilm formed which ensured close biointerface interactions between bacteria-to-bacteria and bacteria-to-electrode. CPs could promote the transmembrane electron transfer through intercalating into the cell membrane of bacteria. Utilizing the CPs-biofilm biohybrid electrode as anode in microbial fuel cell (MFC), the power generation and lifetime of MFC had greatly improved based on accelerated outward EET. Moreover, using the CPs-biofilm biohybrid electrode as cathode in electrochemical cell, the current density was increased due to the enhanced inward EET. Therefore, the intimate biointerface interaction between CPs and bacteria greatly enhanced the bidirectional EET, indicating that CPs exhibit promising applications in both MFC and microbial electrosynthesis. Correlation between intracellular electron transfer and gene expression for electrically conductive pili in electroactive bacteria during anaerobic digestion with ethanol

Abstract:Ethanol feeding has been widely documented as an economical and effective strategy for establishing direct interspecies electron transfer (DIET) during anaerobic digestion. However, the mechanisms involved are still unclear, especially on correlation between intracellular electron transfer in electroactive bacteria and their gene expression for electrically conductive pili (e-pili), the most essential electrical connection component for DIET. Upon cooling from room temperature, the conductivity of digester aggregates with ethanol exponentially increased by an order of magnitude (from 45.5 to 125.4 μS/cm), whereas which with its metabolites (acetaldehyde [from 40.5 to 54.4 μS/cm] or acetate [from 32.1 to 50.4 μS/cm]) did not increase significantly. In addition, the digester aggregates only with ethanol were observed with a strong dependence of conductivity on pH. Metagenomic and metatranscriptomic analysis showed that Desulfovibrio desulfuricans was the most dominant and metabolically active bacterium that contained and highly expressed the genes for e-pili. Abundance of genes encoding the total type IV pilus assembly proteins (6.72E-04 vs 1.24E-03, P < 0.05), PilA that determined the conductive properties (2.22E-04 vs 2.44E-04, P > 0.05), and PilB that proceeded the polymerization of pilin (1.56E-04 vs 3.52E-03, P < 0.05) with ethanol was lower than that with acetaldehyde. However, transcript abundance of these genes with ethanol was generally higher than that with acetaldehyde. In comparison to acetaldehyde, ethanol increased the transcript abundance of genes encoding the key enzymes involved in NADH/NAD+ transformation on complex I and ATP synthesis on complex V in intracellular electron transport chain. The improvement of intracellular electron transfer in D. desulfuricans suggested that electrons were intracellularly energized with high energy to activate e-pili during DIET.

Abstract:This study investigated, if a mixed electroactive bacterial (EAB) culture cultivated heterotrophically at a positive applied potential could be adapted from oxidative to reductive or bidirectional extracellular electron transfer (EET). To this end, a periodic potential reversal regime between − 0.5 and 0.2 V vs. Ag/AgCl was applied. This yielded biofilm detachment and mediated electroautotrophic EET in combination with carbonate, i.e., dissolved CO 2 , as the sole carbon source, whereby the emerged mixed culture (S1) contained previously unknown EAB. Using acetate (S2) as well as a mixture of acetate and carbonate (S3) as the main carbon sources yielded primarily alternating electrogenic organoheterotropic metabolism with the higher maximum oxidation current densities recorded for mixed carbon media, exceeding on average 1 mA cm −2 . More frequent periodic polarization reversal resulted in the increase of maximum oxidative current densities by about 50% for S2-BES and 80% for S3-BES, in comparison to half-batch polarization. The EAB mixed cultures developed accordingly, with S1 represented by mostly aerobes (84.8%) and being very different in composition to S2 and S3, dominated by anaerobes (96.9 and 96.5%, respectively). S2 and S3 biofilms remained attached to the electrodes. There was only minor evidence of fully reversible bidirectional EET. In conclusion the three triplicates fed with organic and/or inorganic carbon sources demonstrated two forms of diauxie: Firstly, S1-BES showed a preference for the electrode as the electron donor via mediated EET. Secondly, S2-BES and S3-BES showed a preference for acetate as electron donor and c-source, as long as this was available, switching to CO 2 reduction, when acetate was depleted.

Abstract:Extracellular Electron Transfer (EET) is considered as the communication bridge between bacterial cells and outer electron acceptors. Apart from a few model microorganisms conventionally studied, the literature regarding the EET mechanism in Gram-positive bacteria and its conductive interaction is sparse. In the present study, proteolytic Gram-positive bacteria - Bacillus cereus is explored for enhancement of power generation in Microbial Fuel Cells (MFC). A low zeta potential (−0.1 V) of B. cereus, enables it to release electrons and protons much faster. We achieve a power density as high as 400 mW m−2 with a current density of ~5.8 mA cm−2 by inoculating an MFC with B. cereus. A genome wide transcriptome analysis by RNA-seq is used to categorize the genes and the significant pathways potentially involved in EET in B. cereus. The expression of gene cluster encodes for PTS (Phosphotransferase system) that catalyzes and transport phosphorylation of disaccharide present in the medium. This plays a regulatory role in biofilm formation. Simultaneously the secretion and assembly of extracellular flavin molecules by B. cereus, shuttle and mediate electron to the extracellular acceptor. In short, B. cereus uses bifurcated pathway i.e., directly by aligning the cytochrome complex and indirectly by secreting flavins molecules to promote electron transfer. These findings reveal its significant functional role in MFCs' electrode community for sustainable bioenergy production.

Abstract:Extracellular electron transfer (EET) describes microbial bioelectrochemical processes in which electrons are transferred from the cytosol to the exterior of the cell1. Mineral-respiring bacteria use elaborate haem-based electron transfer mechanisms2–4 but the existence and mechanistic basis of other EETs remain largely unknown. Here we show that the food-borne pathogen Listeria monocytogenes uses a distinctive flavin-based EET mechanism to deliver electrons to iron or an electrode. By performing a forward genetic screen to identify L. monocytogenes mutants with diminished extracellular ferric iron reductase activity, we identified an eight-gene locus that is responsible for EET. This locus encodes a specialized NADH dehydrogenase that segregates EET from aerobic respiration by channelling electrons to a discrete membrane-localized quinone pool. Other proteins facilitate the assembly of an abundant extracellular flavoprotein that, in conjunction with free-molecule flavin shuttles, mediates electron transfer to extracellular acceptors. This system thus establishes a simple electron conduit that is compatible with the single-membrane structure of the Gram-positive cell. Activation of EET supports growth on non-fermentable carbon sources, and an EET mutant exhibited a competitive defect within the mouse gastrointestinal tract. Orthologues of the genes responsible for EET are present in hundreds of species across the Firmicutes phylum, including multiple pathogens and commensal members of the intestinal microbiota, and correlate with EET activity in assayed strains. These findings suggest a greater prevalence of EET-based growth capabilities and establish a previously underappreciated relevance for electrogenic bacteria across diverse environments, including host-associated microbial communities and infectious disease.

Rewiring the respiratory pathway of Lactococcus lactis to enhance extracellular electron transfer

Abstract:Lactococcus lactis, a lactic acid bacterium with a typical fermentative metabolism, can also use oxygen as an extracellular electron acceptor. Here we demonstrate, for the first time, that L. lactis blocked in NAD+ regeneration can use the alternative electron acceptor ferricyanide to support growth. By electrochemical analysis and characterization of strains carrying mutations in the respiratory chain, we pinpoint the essential role of the NADH dehydrogenase and 2-amino-3-carboxy-1,4-naphtoquinone in extracellular electron transfer (EET) and uncover the underlying pathway systematically. Ferricyanide respiration has unexpected effects on L. lactis, e.g., we find that morphology is altered from the normal coccoid to a more rod shaped appearance, and that acid resistance is increased. Using adaptive laboratory evolution (ALE), we successfully enhance the capacity for EET. Whole-genome sequencing reveals the underlying reason for the observed enhanced EET capacity to be a late-stage blocking of menaquinone biosynthesis. The perspectives of the study are numerous, especially within food fermentation and microbiome engineering, where EET can help relieve oxidative stress, promote growth of oxygen sensitive microorganisms and play critical roles in shaping microbial communities.

Abstract:Diverse microbial species utilize redox shuttles to exchange electrons with their environment through mediated extracellular electron transfer (EET). This process maintains redox homeostasis and supports anaerobic survival across diverse microbial communities. Although mediated EET has been extensively leveraged for bioelectrocatalysis and bioelectronics for decades, fundamental questions remain about how these redox shuttles are reduced within cells and their bioenergetic implications. This knowledge gap limits our understanding of the physiological roles of mediated EET in various microbes and hampers the development of efficient microbial electrochemical technologies. To address this, we developed a methodology integrating genome editing, electrochemistry, and systems biology to investigate the mechanism and bioenergetic implications of mediated EET in bacteria. Using this approach, we uncovered a mediated EET mechanism in . In the absence of alternative electron sinks, the redox cycling of 2-hydroxy-1,4-naphthoquinone (HNQ) via a cytoplasmic nitroreductase enabled to respire and grow on an extracellular electrode. Genome-scale metabolic modeling suggested that HNQ-mediated EET offers a more energetically favorable route for supporting anaerobic growth than canonical fermentation. Transcriptome analysis revealed redox perturbations in response to HNQ and identified rapid metabolic adaptations that support growth. This work demonstrates that can grow independently of classical electron transport chains and fermentative pathways, unveiling a new type of anaerobic energy metabolism.

Single-Cell Phenotyping of Extracellular Electron Transfer via Microdroplet Encapsulation

Abstract:Electroactive organisms contribute to metal cycling, pollutant removal, and other redox-driven environmental processes. Studying this phenomenon in high-throughput is challenging since extracellular reduction cannot easily be traced back to its cell of origin within a mixed population. Here, we describe the development of a microdroplet emulsion system to enrich EET-capable organisms. We validated our system using the model electroactive organism S. oneidensis and describe the tooling of a benchtop microfluidic system for oxygen-limited processes. We demonstrated enrichment of EET-capable phenotypes from a mixed wild-type and EET-knockout population. As a proof-of-concept application, bacteria were collected from iron sedimentation from Town Lake (Austin, TX) and subjected to microdroplet enrichment. We observed an increase in EET-capable organisms in the sorted population that was distinct when compared to a population enriched in a bulk culture more closely akin to traditional techniques for discovering EET-capable bacteria. Finally, two bacterial species, C. sakazakii and V. fessus not previously shown to be electroactive, were further cultured and characterized for their ability to reduce channel conductance in an organic electrochemical transistor (OECT) and to reduce soluble Fe(III). We characterized two bacterial species not previously shown to exhibit electrogenic behavior. Our results demonstrate the utility of a microdroplet emulsions for identifying putative EET-capable bacteria and how this technology can be leveraged in tandem with existing methods.

Abstract:The extracellular electron transfer (EET) capability of Methanosarcina spp. in direct interspecies electron transfer (DIET) has profoundly increased our understanding of microbial kinetics and energetics in biomethanation systems. In Methanosarcina spp., such EET mechanisms occur in the cell envelope and biofilm matrix. These substances are composed of protein-like, polysaccharide-rich biomolecular structures that were previously thought to contribute only to cell support and shape; while their participation in dynamic processes remains unclear and has gathered widespread interest. This review first addresses the molecular structure and chemical characteristics of the extracellular matrix and cell wall polymers in Methanosarcina spp. Next, we focus on recent theoretical studies on the conduction and EET mechanisms of the extracellular matrix and cell wall polymers: tunnelling, hopping, proton-activated electron transfer and voltage-dependent electron transport. We conclude this review by discussing the state-of-the-art electrochemical techniques and experiments and the associated challenges, i.e., the kinetic isotope effect and on–off resonance switching. The border impacts of such conductive pathways may offer a semi-classical/quantum perspective on microbiology and mark the renaissance of anaerobic biotechnology.

Abstract:Electrochemically active bacteria (EAB) are capable of extracellular electron transfer (EET) to insoluble metal oxides, and thus play a great role in the fields of environment, energy, and geosciences. However, rapid and accurate quantification of the EET ability of EAB is still challenging. In this work, we develop a riboflavin-based fluorescence method for facile, accurate, and in situ measurement of the EET ability of EAB. This method is successfully used to quantify the single-cellular EET ability of Geobacter sulfurreducens DL-1 (60.29 ± 13.02 fA) and Shewanella oneidensis MR-1 (2.11 ± 0.47 fA), the two widely present EAB in the environment. It also enables quantitative identification of EET-related c-type cytochromes in the outer membrane of S. oneidensis MR-1. This method provides a useful tool to rapidly identify EAB in diverse environments and elucidate their electron transfer mechanisms.
