Abstract:In spite of being most common, oral and parenteral drug delivery routes are associated with major biopharmaceutical hurdles. These can lead to less or no therapeutic effect due to drug precipitation as a result of first-pass metabolism as well as disease transmission from one person to other through the needle. To overcome these challenges, now days, transdermal drug delivery system is emerging as an appropriate alternative having some superiority over the conventional systems. This system provides the modification to improve its performance by adding various additives like penetration enhancers, polymers anti-oxidant, chelating agents etc. Novel transdermal delivery system, which is the result of the betterment of TDDS came into picture which comprises of carriers like ethosomes, noisome, transferosome, liposome, emulgel and so on. Bile salts, rhamnolipid, sophorolipids, lecithin are the examples of biosurfactants that serves as substitute of chemical surfactants which can cause different side effects. Besides removing the side effects, they also have some therapeutic activities which contributes to the activity of the formulation, such as bile salt acts as absorption enhancer, rhamnolipid as emulsifiers and wound healer, and sophorolipid as antifungal agent. Acne vulgaris, commonly known as acne, is a chronic skin condition characterized by the presence of pimples, blackheads, and whiteheads on the face, neck, chest, and back. It is caused by the overproduction of oil (sebum) in the hair follicles, combined with dead skin cells, bacteria, and inflammation. Acne is most commonly seen in adolescents, but it can also affect people of all ages. Treatment options include topical and oral medications, light therapy, and lifestyle changes. In this review we have discussed about pathophysiology of the acne vulgaris, various additives which are been used in novel transdermal drug delivery system, commercially available novel formulations, patents and future perspectives.
