Abstract:The aqueous two-phase extraction process (APTE) for polysaccharides from the fruits of Ilex cornuta (Lindl. et Paxt.) (PFIC) was studied, and its monosaccharide composition and antioxidant activities were analyzed. After screening and selection of isopropanol/ammonium sulfate as the aqueous two-phase system, various parameters of crude extract amount, ultrasonic time, extraction temperature and extraction time on the extraction process was investigated in detail and optimized by response surface methodology (RSM). The obtained PFIC was characterized by FT-IR and its monosaccharide composition was analyzed by GC-MS. Furthermore, the antioxidant activities of PFIC were evaluated by measuring the scavenging rates of DPPH radical, hydroxyl radical and superoxide anion radical. Under the optimal conditions in which crude extract amount was 2.23 g, ultrasonic time was 17.5 min, extraction temperature was 69 â„ƒ and the extraction time was 1.55 h, the yield of polysaccharides reached 50.87% which was close to the predicted value of 50.78%. FT-IR analysis confirmed that the extract was polysaccharide, and GC-MS analysis indicated that the monosaccharide components of the polysaccharides were rhamnose, arabinose, xylose, mannose, glucose and galactose. Antioxidant results showed that PFIC showed strong scavenging ability to DPPH radical, fine scavenging ability to hydroxyl radical and certain scavenging ability to superoxide anion radical. The ultrasonic-assisted APTE process for PFIC optimized by RSM is reasonable and reliable, and the obtained PFIC has the potential to be utilized as natural antioxidants.

Abstract:Natural deep eutectic solvent (NADES) has received increasing interest as a green alternative to traditional organic solvents for efficient extraction of bioactive compounds from natural sources. In this study, phytochemicals in Carya cathayensis Sarg. peels extracted with Choline chloride-Malic acid (ChCl-MA) were identified using UPLC-Triple-TOF/MS. Effect of NADES on phenolic composition, antioxidant properties and inhibition of α-glucosidase and α-amylase were evaluated. Furthermore, extraction mechanism caused by different solvents were investigated by quantum chemical calculation combined with molecular dynamic simulation. A total of 29 phytochemicals were identified, and catechin, procyanidin B1, 2,3-dihydroxybenzoic acid, pinocembrin, procyanidin B3, myricetrin were the most abundant compounds. The extract using ChCl-MA exhibited the highest phenolic compounds content, antioxidant capacity, and α-glucosidase and α-amylase inhibition activities. Larger solvent accessible surface area, more hydrogen bonds between ChCl-MA and extract, longer lifetime of the hydrogen bonds, and lower intermolecular interaction energy account for higher extraction efficiency of ChCl-MA.

Bioactive compounds profile of extracts from Pereskia aculeata Miller leaves prepared with different solvents by ultrasound-assisted extraction

Abstract:The ultrasound-assisted extraction (UAE) technique is an emerging method of recovering bioactive molecules, which has the advantages of low cost and good yield, in addition to meeting the requirements of green chemistry. The present work aimed to optimize the UAE in obtaining bioactive compounds from ora-pro-nobis leaves using seven different solvents in the extraction process. The analysis of the crude extracts by chromatographic methods (GC-MS and LC-MS/MS) revealed a nonpolar hydrocarbon mixture rich in triacontan, steroids, flavonoids and diterpene, among which we highlight the acids (malic, ferulic, palmitic, linoleic, gluconic), as well as phytol, campesterol, γ-sitosterol and silicic acid. The exploratory multivariate statistical analysis performed through principal component analysis (major chemical compounds, chemical class and solvent toxicity) allowed the election of ethanol as the best solvent for the recovery of interest compounds via UAE. Taken together, the results indicate that P. acuelata has a large number of bioactive compounds in its cellular structure, and that UAE is an efficient technique for the recovery of such molecules.

Abstract:Bioactivity of "espinheira-santa" leaves can be evaluated through solvent purification processes. Species of the same genus may contain different compounds, which may be separated by conventional chromatography using different solvents. The extraction process also influences the contents of these compounds. In order to optimize the above process research is required on variables such as solvents, temperature and extraction time. The objective of this work was to evaluate the antioxidant activity (ABTS, DPPH and FRAP) of extracts and purified fractions from leaves of Maytenus ilicifolia and M. aquafolium. Percolation at 28 °C for 7 days, and ultrasound at 50, 60 and 70 °C for 15 min were the extraction methods tested. Open column chromatography (silica gel) was used in the fractionation of extracts, and hexane, chloroform and methanol were the elution solvents used. The evaluation methods of the antioxidant activity were DPPH, FRAP and ABTS. Plant species showed no significant differences between methods for DPPH, but the fractions obtained with chromatography had significant differences, the crude ethanolic extract (CEE) and the methanolic been in general superior. "espinheira-santa" is a plant with antioxidant potential, but the extraction methods are not optimal. Still, it can be said that the separation by chromatography provided a better visualization of the nature of the components of that plant that best stood out in relation to the antioxidant activity.

Abstract:Polygonum hydropiperoides Michx. is an Asian native plant species that is also widely distributed in the Americas. Despite its traditional uses, P. hydropiperoides is scarcely scientifically exploited. This study aimed to chemically characterize and investigate the antioxidant and antibacterial activities of hexane (HE-Ph), ethyl acetate (EAE-Ph), and ethanolic (EE-Ph) extracts from aerial parts of P. hydropiperoides. The chemical characterization was performed through HPLC-DAD-ESI/MSn. The antioxidant activity was assessed by the phosphomolybdenum reducing power, nitric oxide inhibition, and the β-carotene bleaching assays. The antibacterial activity was determined by the minimal inhibitory concentration (MIC) and the minimal bactericidal concentration followed by the classification of the antibacterial effect. Chemical characterization revealed the expressive presence of phenolic acids and flavonoids in EAE-Ph. An increased antioxidant capacity was revealed in EAE-Ph. Regarding antibacterial activity, EAE-Ph showed weak to moderate property against 13 strains tested with MIC values ranging from 625 to 5000 μg/mL, with bactericidal or bacteriostatic effects. Glucogallin and gallic acid stand out as the most relevant bioactive compounds. These results suggest that P. hydropiperoides is a natural source of active substances, supporting this species' traditional use.

Abstract:Josefsson J.-O. and S. Thesleff. Electromyographic findings in experimental botulinum intoxication. The electrical activity of botulinum intoxicated skeletal muscle was examined in the rabbit by the use of electromyography. 5 to 6 days after an intramuscular injection of the toxin, fibrillation potentials appeared and persisted for a period of at least 60 days. The potentials were similar in amplitude and time course to those recorded following motor nerve degeneration. Botulinum toxin, however, is known not to affect the morphological structure of the motor nerve and its terminals. Its sole action is to block transmitter release selectively from cholinergic nerve endings. Our results therefore suggest that in nerve degeneration lack of transmitter release is primarily responsible for initiating the process which produces the electromyographic pattern typical of denervation.

Abstract:E. uniflora leaves are a rich source of phenolic compounds with biological activities, including myricitrin. In this study, the chemical profile of nine extracts prepared with leaves collected in three regions (mountain, beach, and mangrove) and at three different times of the day (8 am, 1 pm, and 6 pm) was evaluated from spectra originating from ultra-high resolution mass spectrometry (Fourier Transform Ion Cyclotron Resonance, FT-ICR) coupled to electrospray (ESI). The best time of the day and location for collecting the leaves of E. uniflora used as raw material for producing extracts, and the best ethanol concentration for obtaining an extract more abundant in compounds of interest were verified. Several flavonoids and phenolic acids were detected in their deprotonated form in the regions from m/z 200 to 1200. Myricitrin ([C21H20O12-H]-, m/z theo 463.08820), its chloride adduct ([C21H20O12+Cl]-, m/z theo 499.06488), other myricitrin derivatives, and some tannins were the main compounds detected. Considering obtaining an extract rich in phenolic compounds, including myricitrin, the best place and time of the day to collect E.uniflora leaves is in the beach region at 1 pm. In contrast, the best ethanol concentration for extract production is 70 wt%. Therefore, extraction at 96 wt% ethanol is better for obtaining an extract more abundant in phenolic acids, although 70 wt% ethanol also extracted these compounds. The FTIR-PCA models were used to check possible similarities in the data according to collection time of the day and location. These models demonstrated an excellent solution for sample screening.

Abstract:Background: This study is based on ethnobotanical observation and traditional medicinal utilization of fig (Ficus carica L.) by southwest Asian and Mediterranean populations. Objective: Considering phenolics among the highly bioactive compounds for their numerous therapeutic applications, the first goal of this investigation is the optimization of their recovery by using Response Surface Methodology (RSM). Methods: Prior to RSM, the single-factor method was applied for revealing the main factors influencing the extraction (solvent type, solvent concentration, sample˗to˗solvent ratio, temperature, and time of extraction). After using the optimal conditions of extraction, the second objective was the identification of fig leave phytochemicals by UPLC˗MS. Results: The results of preliminary optimization step through sequential methodology revealed that solvent concentration, extraction duration, and temperature, and were the main influencing factors. All these tested extraction parameters influenced significantly total phenolic contents (TPC) and DPPH-free radical scavenging activity (FRSA). RSM analysis using Box˗Behnken design showed that the optimal extraction conditions of total phenolic contents of fig leaves were 23.07% acetone, 51.82 °C, and 118.02 min with values of 29.44 mg GAE/g and 59.48% for TPC and FRSA, respectively. Eight phenolic compounds were detected including chlorogenic, caffeic, and coumaric acids, rutin, isoquercetin, luteolin, quercetin, and kaempferol well known for their bioactivities. Two non˗phenolic compounds were also found: The psoralen and an oxylipin, the 9˗oxo˗octadecadienoic acid that was detected for the first time in fig leaves. Conclusion: Through sequential and RSM optimization processes for phenolic antioxidants of Ficus carica leaves, the developed mathematic models have expressed a high level of significance. The UPLC-MS analysis of the optimal extract revealed numerous phenolic compounds known for their bioactivities.

Abstract:Introduction The species Lantana camara is used in folk medicine. The biological activities of this medicinal plant are attributable to the presence of various derivatives of triterpenoids and phenolic compounds present in its preparations, indicating excellent economic potential. Objective In this study, the operational conditions of ultrasound‐assisted extraction (UAE) and microwave‐assisted extraction (MAE) were optimized using Box–Behnken design to improve the total phenolic content (TPC) recovered in hydroethanolic extracts of L. camara leaves. Material and methods The TPC, total flavonoid content (TFC), and antioxidant activities of the hydroalcoholic extracts of L. camara, prepared by UAE and MAE under the optimized extraction conditions, were compared with those of the extracts obtained by conventional extraction methods. Results Under the optimal conditions, the extracts obtained by UAE (35% ethanol, 25 min, and a solvent‐to‐solid ratio of 60:1 mL/g) and by MAE (53% ethanol, 15 min, and 300 W) provided high yields of 32.50% and 38.61% and TPC values of 102.89 and 109.83 mg GAE/g DW, respectively. The MAE extract showed the best results with respect to TPC, TFC, and antioxidant activities, followed by extracts obtained by UAE, Soxhlet extraction, decoction, maceration, and infusion, in that order. Conclusion The results obtained indicate that L. camara may be used as an important source of antioxidant phenolic compounds to obtain products with high biological and economic potential, especially when the extraction process is performed under appropriate conditions using MAE and/or UAE, employing environmentally friendly solvents such as water and ethanol.

Abstract:We have compared, in 60 adult patients, the cardiovascular effects of glycopyrronium 5 micrograms kg-1 and 10 micrograms kg-1 given either simultaneously or 1 min before edrophonium 1 mg kg-1. Significant differences between the four groups were detected (P less than 0.001). Both groups receiving 10 micrograms kg-1 showed increases in heart rate of up to 30 beat min-1 (95% confidence limits 28-32 beat min-1). Use of glycopyrronium 5 micrograms kg-1 provided greater cardiovascular stability and, given 1 min before the edrophonium, was sufficient to minimize early, edrophonium-induced bradycardias. This low dose of glycopyrronium provided good control of oropharyngeal secretions.
Ultrasonic-Assisted Extraction of Antioxidants from Perilla frutescens Leaves Based on Tailor-Made Deep Eutectic Solvents: Optimization and Antioxidant Activity

Abstract:The development of natural antioxidants to replace synthetic compounds is attractive. Perilla frutescens leaves were proven to be rich in antioxidants. The extraction of antioxidants from Perilla leaves via ultrasonic-assisted extraction (UAE) based on choline chloride-based deep eutectic solvents (DESs) was studied. Firstly, several DESs were prepared, and their extraction effects were compared. Secondly, the extraction process was optimized by single-factor experiments and response surface methodology (RSM). Finally, the optimization results were verified and compared with the results of traditional solvent-based UAE. The effects of solvents on the surface cell morphology of Perilla frutescens leaves were characterized by scanning electron microscopy (SEM). Choline chloride-acetic acid-based DES (ChCl-AcA) extract showed a relatively high ferric-reducing antioxidant activity (FRAP) and 2,2-diphenyl-1-picrylhyldrazyl radical scavenging rate (DPPH). Under the optimal operating conditions (temperature 41 °C, liquid-solid ratio 33:1, ultrasonic time 30 min, water content 25%, ultrasonic power 219 W), the experimental results are as follows: DPPH64.40% and FRAP0.40 mM Fe(II)SE/g DW. The experimental and predicted results were highly consistent with a low error (<3.38%). The values of the DPPH and FRAP were significantly higher than that for the water, ethanol, and butanol-based UAE. SEM analysis confirmed that ChCl-AcA enhanced the destruction of the cell wall, so that more antioxidants were released. This study provides an eco-friendly technology for the efficient extraction of antioxidants from Perilla frutescens leaves. The cytotoxicity and biodegradability of the extract will be further verified in a future work.
Chemical Composition, Nutritional, and Biological Properties of Extracts Obtained with Different Techniques from Aronia melanocarpa Berries

Abstract:This study investigates the chemical composition, nutritional, and biological properties of extracts obtained from A. melanocarpa berries using different extraction methods and solvents. Hydrodistillation and supercritical fluid extraction with CO2 allowed us to isolate fruit essential oil (HDEX) and fixed oil (SFEEX), respectively. A phenol-enriched extract was obtained using a mild ultrasound-assisted maceration with methanol (UAMM). The HDEX most abundant component, using gas chromatography-mass spectrometry (GC/MS), was italicene epoxide (17.2%), followed by hexadecanoic acid (12.4%), khusinol (10.5%), limonene (9.7%), dodecanoic acid (9.7%), and (E)-anethole (6.1%). Linoleic (348.9 mg/g of extract, 70.5%), oleic (88.9 mg/g, 17.9%), and palmitic (40.8 mg/g, 8.2%) acids, followed by α-linolenic and stearic acids, were the main fatty acids in SFEEX determined using high-performance liquid chromatography coupled with a photodiode array detector and an evaporative light scattering detector (HPLC-DAD/ELSD). HPLC-DAD analyses of SFEEX identified β-carotene as the main carotenoid (1.7 mg/g), while HPLC with fluorescence detection (FLU) evidenced α-tocopherol (1.2 mg/g) as the most abundant tocopherol isoform in SFEEX. Liquid chromatography-electrospray ionization-MS (LC-ESI-MS) analysis of UAMM showed the presence of quercetin-sulfate (15.6%, major component), malvidin 3-O-(6-O-p-coumaroyl) glucoside-4-vinylphenol adduct (pigment B) (9.3%), di-caffeoyl coumaroyl spermidine (7.6%), methyl-epigallocatechin (5.68%), and phloretin (4.1%), while flavonoids (70.5%) and phenolic acids (23.9%) emerged as the most abundant polyphenol classes. UAMM exerted a complete inhibition of the cholesterol oxidative degradation at 140 °C from 75 μg of extract, showing 50% protection at 30.6 μg (IA50). Furthermore, UAMM significantly reduced viability (31–48%) in A375 melanoma cells in the range of 500–2000 μg/mL after 96 h of incubation (MTT assay), with a low toxic effect in normal HaCaT keratinocytes. The results of this research extend the knowledge of the nutritional and biological properties of A. melanocarpa berries, providing useful information on specific extracts for potential food, cosmetic, and pharmaceutical applications.

Thus, this study aimed to determine the total phenolic content, evaluate the antioxidant potential, and in vitro cytotoxicity of the leaf ethanolic extract from Ephedrantus amazonicus R. E. Fries. The phenolic content and antioxidant activity were evaluated by the Folin-Ciocalteu and DPPH methods, respectively. The assays were adapted for 96-well microplates, with absorbance reading at λ = 630 and 517 nm, respectively, using a UV-Vis spectrophotometer. The in vitro cytotoxicity assay was performed using fibroblasts (L929 cell line). The MS/MS analyses were performed through direct infusion using a LCQ Fleet mass spectrometer equipped with APCI font and ion trap analyzer, in the positive and negative mode. A molecular network was created using the online workflow on the GNPS website (http://gnps.ucsd.edu). The phenolic content at 400μg was 25.24 ± 0.98μg.GAE/g, while the antioxidant activity showed an EC50 of 13.37±2.63 μg.mL-1. Dereplication led to the annotation of three flavonoids that may contribute to the observed antioxidant activity. The extract showed low cytotoxicity at the highest tested concentration (> 500μM). The results demonstrate that the extract from Ephedrantus amazonicus R.E. Fries leaves is a promising source of natural antioxidants.

Abstract:The recovery of bioactive compounds is a promising approach for obtaining rich extracts from fruit by-products. This study investigated the influence of Natural Deep Eutectic Solvents (NADES) and Ultrasound-Assisted Extraction (UAE) on the phenolic content, antioxidant capacity, and in vitro antidiabetic activity of Psidium myrtoides by-product. Among eight NADES evaluated based on choline chloride, NADES ChCl:Gly (1:2) was selected for its efficiency in extracting total phenolic compounds (TPC) with high antioxidant capacity. The optimized conditions were 61 °C, a solid-liquid ratio of 100 mg 5 mL-1, and a 60-minute extraction time. ChCl:Gly exhibited superior TPC recovery (2.6-fold greater effectiveness) compared to the 60 % hydroethanolic solution. Twenty-six phenolic compounds were identified, including significant levels of catechin (336.48 mg g-1) and isoquercetin (26.09 mg g-1). Phenolic acids, such as p-anisic acid (5.47 mg g-1) and methoxyphenylacetic acid (0.23 mg g-1), were identified for the first time in the purple araçá by-product. The ChCl:Gly extract demonstrated the highest bioactivity, showcasing antioxidant and antidiabetic capacities. This study introduces an innovative and sustainable alternative for recovering phenolic compounds from fruit by-products, offering enhanced recovery efficiency and/or selectivity compared to organic solvents.

Abstract:Within a study focused on Sinapis pubescens subsp. pubescens wild from Sicily (Italy), an edible species still unexplored, our earlier published work has demonstrated good in vitro antioxidant properties for the flower and leaf hydroalcoholic extracts, exhibiting quite different qualitative‐quantitative phenolic profiles. Herein, further research was designed to elucidate the role played by phenolic compounds in the different antioxidant mechanisms highlighted for the extracts. To achieve this goal, the crude extracts were subjected to liquid‐liquid partitioning with solvents of increasing polarity; then, the fractions were investigated for their antioxidant properties using different in vitro assays. For both flowers and leaves, the ethyl acetate fractions exhibited the best activity in DPPH and reducing power assays, followed by n‐butanol. The total phenolic content determination indicated these fractions as the phenolic‐rich ones, which were characterized by HPLC‐PDA/ESI‐MS analysis. Conversely, the phenolic‐rich fractions did not show any chelating activity, which was highlighted for the more hydrophobic ones.

Abstract:The extract of the plant species Satureja hortensis L. (often used as traditional ethno-therapy and in food processing) was prepared using the ultrasonic extraction technique, and contains a large quantity of secondary metabolites, with scientific evidence for antioxidant, antimicrobial, sedative, antispastic and antidiarrheal activities. Process optimization was carried out using a mathematical–statistical method (response surface methodology—RSM), which models and examines the effects of three levels and three variables on the observed response. The investigated responses were the content of total phenolic components (TPC) and total flavonoids (TFC), as well as tests of antioxidant activity at the level of radicals. The independent variables were ethanol concentration (40–80%), temperature (40–80 °C) and the liquid–solid ratio (10–30 mL/g). Results from this study were entered into a second-degree polynomial with multiple non-linear regression. Analysis of variance (ANOVA) was applied to find the most favorable environment for assessing the model's performance and effectiveness with an ethanol concentration of 20%, temperature of 80 °C and LSR of 21.4 mL/g. ANOVA assessed the model's significance, and a second-order polynomial model described the relationships between variables and responses. Diagnostic plots confirmed the model's adequacy and reliability. The estimated values were 4.11 mg chlorogenic acid equivalents per gram of dry weight (CEA/g), 2.18 mg of rutin equivalents per gram of dry weight (RE/g), and 0.030 mg/mL and 0.030 mg/mL for TPC, TFC, IC50 and EC50, respectively. High-Performance Liquid Chromatography with diode array detection (HPLC-DAD) examination revealed that the prominent substance in the tested extract is rosmarinic acid (46,172 μg/mL), followed by chlorogenic acid (1519 μg/mL).

Abstract:In the present work, fourteen cultivars of Prunus domestica were analyzed to investigate their phenolic pattern with the purpose of using the leaves as potential resources of bioactive compounds in the pharmaceutical and food industry. Microwave-assisted extraction (MAE), dispersive liquid-liquid microextraction and sugaring-out liquid-liquid extraction techniques were optimized in order to obtain an exhaustive multi-component panel of phenolic compounds. The best phenolic-enriched recovery was achieved using MAE in water:methanol (30:70), and this procedure was further applied for quantitative analysis of phenolic compounds in real samples. In order to prove the safeness of these extracts, the biological potential of the Prunus cultivars was tested by several in vitro antioxidant and enzyme inhibitory assays. Moreover, their cytotoxicity was evaluated on human gingival fibroblasts (HGFs), and in most of the cases the treatment with different concentrations of extracts didn't show cytotoxicity up to 500 μg/mL. Only 'Carpatin' and 'Minerva' cultivars, at 250 and 500 μg/mL, reduced partially cell viability of HGFs population. Noteworthy, Centenar cultivar was the most active for the α-glucosidase inhibition (6.77 mmolACAE/g extract), whereas Ialomița cultivar showed the best antityrosinase activity (23.07 mgKAE/g extract). Overall, leaves of P. domestica represent a rich alternative source of bioactive compounds.

Abstract:INTRODUCTION:Perilla frutescens (L.) Britt., an essential traditional Asian crop and Chinese medicine, potentially exerts anti-oxidation effects through its phenolic compounds. These compounds have already been reported in perilla seed, however, little is reported in Perilla pomace, the primary waste during oil production of Perilla seed. OBJECTIVE:To investigate major phenolic compounds in perilla seeds and pomaces in order to check whether the pomace could be an alternative resource to the seed for nutritional and medical purposes. METHODS:Compounds in extracts of perilla seeds and pomaces were separated by high-performance liquid chromatography and detected by photodiode array, and by electrospray ionisation with quadrupole time-of-flight tandem mass spectrometry. Herb-markers selected by principal components analysis were then quantified in both seeds and pomaces. Moreover, a fingerprinting approach and multiple discriminant analysis were applied to screen the phenolic markers in 22 samples. RESULTS:Ten phenols were tentatively identified, among which four (rosmarinic acid, luteolin, apigenin and rosmarinic acid-3-O-glucoside) were selected as herb-markers. Perilla seeds and pomaces showed similar phenol profiles, however, the pomaces contained almost two times the amount of the four herb-markers than the seeds. CONCLUSION:The results indicated perilla pomace is a promising alternative source of phenolic compounds that could be recovered and potentially used as natural anti-oxidants.
