Abstract:Hydrogel has emerged as one of the most promising dressings for wound healing owing to the creation of moist environment. However, it suffers from poor water retention capability and weak mechanical property. Therefore, development of dressings with durable moisture retention, robust mechanical property, and self-healing performance is urgently needed in practical applications. Herein, we developed a bilayered composite dressing consisting of breathable and hydrophobic polyurethane/polydimethylsiloxane (PU/PDMS) nanofibrous membrane and double-crosslinked chitosan-based hydrogel with dynamic Schiff base bonds and non-dynamic photo-crosslinking bonds. Combined with the nanofibrous membrane as protection and mechanical support, the bilayered hydrogel composites presented enhanced water retention capability to provide sustained moist environment, excellent stretchable and compressive performance to bear deformation, fast self-healing and good cyclic resilience to endure frequent movements. The in vivo study indicated their effective therapeutic effect in a rat model with full-thickness skin wounds. This study demonstrated that the hydrogel/nanofibrous membrane composite represented a promising candidate as dressing material for wound healing.

Silver nanoparticles loaded triple-layered cellulose-acetate based multifunctional dressing for wound healing

Abstract:Chronic wounds present considerable challenges which delay their effective healing. Currently, there are several biomaterial-based wound dressings available for healing diverse wound types. However, most of commercial wound dressings are too expensive to be affordable to the patients belonging to the middle and lower socioeconomic strata of the society. Thus, in this investigation affordable triple layered nanofibrous bandages were fabricated using the layer-by-layer approach. Here, the topmost layer comprised of a hydrophilic poly vinyl alcohol layer, cross-linked with citric acid. The middle layer comprising of cellulose acetate was loaded with silver nanoparticles as an antibacterial agent, while the lowermost layer was fabricated using hydrophobic polycaprolactone. The triple-layered nanofibrous bandages having a nano-topography, exhibited a smooth, uniform and bead-free morphology, with the nanofiber diameter ranging between 200 and 300 nm. The nanofibers demonstrated excellent wettability, slow in vitro degradation, controlled release of nano‑silver and potent antibacterial activity against Gram-negative (E.coli) and Gram-positive (S. aureus) bacteria. The fabricated bandages had excellent mechanical strength upto 12.72 ± 0.790 M. Pa, which was suitable for biomedical and tissue engineering applications. The bandage demonstrated excellent in vitro hemocompatibility and biocompatibility. In vivo excisional wound contraction, along with H and E and Masson's Trichrome staining further confirmed the potential of the nanofibrous bandage for full-thickness wound healing. Pre-clinical investigations thus indicated the possibility of further evaluating the triple-layered nanofibrous dressing in clinical settings.

Abstract:The state of many dynamic systems evolves subject to some equality constraints. Most existing work focuses on developing state estimation algorithms for an equality-constrained dynamic system assuming the system is given. How to design and analyze such a system is rarely addressed, even though it is critically important for application and performance evaluation. In this paper, we first analyze the underlying structural information of linear equality-constrained (LEC) dynamic systems through direct elimination. It is found that the process noise of such a system is state dependent in general. This means that existing formulations of such a system are quite limited. Based on the analysis, we then propose a new systematic way to design LEC systems. The key idea is to design only the unconstrained part of the system. Unlike existing system conversion-based design techniques, in our work the desired model class is given and only the distributions of the initial state and process noise need to be determined, which is comparatively easier. To handle state dependency of the process noise, a new constrained state estimation algorithm based on direct elimination is also proposed. Three examples are provided to illustrate the effectiveness of the proposed methods of analysis, design, and estimation, respectively.
Medicated tri-layer fibers based on cellulose acetate and polyvinylpyrrolidone for enhanced antibacterial and wound healing properties

Abstract:Wound dressing is commonly used for skin injuries. The design of wound dressing typically stems from the principles of open-wound management such as infection prevention, moisture balance and healing response. A new wound dressing comprising polyvinylpyrrolidone (PVP)–berberine hydrochloride (BHC)/PVP–cellulose acetate (CA)–BHC/CA-aloin tri-layer Janus fiber was successfully fabricated using trifluid side-by-side electrospinning for antibacterial and wound healing functions. The fibrous membrane can hold 5.090 ± 0.276 times of its own water mass and achieve 6 h of microenvironment moisture retention. In vitro antibacterial results show that BHC exhibited selective resistance to S. aureus . The nanofibers were noncytotoxic and showed good cell adhesion properties and enhanced cell proliferation effect compared with BHC or aloin only tri-layer nanofibers. Owing to the specially designed tri-layer Janus structure, the in vitro drug-release profile of drugs-loaded fibrous membrane showed a combination of two-stage release for BHC (76.093 % ± 1.813 % release in first hour and 98.526 % ± 0.604 % release in 6 h) and sustained release for aloin (88.376 % ± 5.282 % release in 6 h). Therefore, the tri-layer Janus structure can be a potential choice for wound dressing application.

Abstract:Head and neck squamous cell carcinoma (HNSCC) is an aggressive malignancy in which multiple independent lesions develop over time throughout the mucosa of the upper aerodigestive tract. Therefore, the comprehensive treatment of this neoplasm must include a chemopreventive arm to hold premalignant lesions in check, a role well-suited to antiangiogenic agents. Retinoic acid (RA) and interferon alpha (IFN-alpha), drugs with known biological activity against HNSCC when used individually, are also inhibitors of angiogenesis. Here we show that they are remarkably synergistic antiangiogenic agents able to inhibit both the growth and the neovascularization of HNSCC injected into the floor of the mouth of nude mice. The mechanism of action of these drugs as antiangiogenic agents was 2-fold. They decreased the angiogenic activity of the tumor cells, and they caused the endothelial cells to become refractory to inducers of angiogenesis. When tumor cells were treated in vitro with IFN-alpha A/D, there was a dramatic drop in their secretion of interleukin-8, the major angiogenic factor produced by these tumors. When combined with RA, which causes tumor cells to secrete an inhibitor of angiogenesis, there was a synergistic inhibition of both tumor cell growth and secreted angiogenic activity. The combination of RA and IFN-alpha also acted synergistically on endothelial cells by reducing their responsiveness to both interleukin-8 and tumor conditioned media. Doses of each drug could be reduced by two logs without loss of activity. When animals bearing human HNSCC tumor cells were treated systemically with a combination of RA and IFN-alpha A/D at doses that were ineffective when used alone, dramatic decreases in both tumor growth and tumor angiogenesis were seen. These data suggest that the use of antiangiogenic mixtures may be a particularly effective way to design future chemoprevention protocols against HNSCC.

Abstract:A multifunctional wound dressing patch capable of rapid wound healing, wound protection, and wound state monitoring based on hyaluronic acid/gelatine/dexpanthenol nanofibers and curcumin/polycaprolactone microfibers are developed. In addition, the dressing's antimicrobial and pH colorimetric properties are evaluated. Furthermore, the dressing's actual healing, protection, and monitoring capabilities are evaluated through a mouse wound model. Considerable efforts have been devoted to developing wound dressings with various functions, including rapid cell proliferation, protection against infection, and wound state monitoring to minimize severe pain and the risks of wound‐caused secondary infections. However, it remains challenging to diagnose wound conditions and achieve integration of the above functions without specialized equipment and expertise in wound care. This study describes an electrospun composite micro/nanofiber‐based bilayer‐dressing patch comprising a healing‐support layer (hyaluronic acid, gelatin, and dexpanthenol) and a protective/monitoring layer (curcumin and polycaprolactone). The improved cell regeneration function and biocompatibility of the healing‐support layer enable rapid healing, as evidenced by the expedited growth of fibroblasts. The superior antimicrobial properties (against Escherichia coli and Staphylococcus aureus) and visible color changes within the pH range of wound lesions (pH 6–9) of the protective/monitoring layer make the dressing suitable for advanced wound care. The wounds inflicted on BALB/c mice heal rapidly (12 days) without scars while the wound state can be diagnosed by the change in color of the dressing patch. The multifunctional wound dressing patch developed in this study is expected to promote wound healing and monitor wound state; thus, facilitating convenient wound management.

Abstract:The development of advanced multifunctional wound dressings remains a major challenge. Herein, a novel multilayer (ML) electrospun nanofibers (NFs) wound dressing based on diethylenetriamine (DETA) functionalized polyacrylonitrile (PAN), TiO2 nanoparticles (NPs) coating (Ct), and bioderived gelatin (Gel) was developed for potential applications in wound healing. The ML PAN-DETA-Ct-Gel membrane was developed by combining electrospinning, chemical functionalization, synthesis, and electrospray techniques, using a layer-by-layer method. The ML PAN-DETA-Ct-Gel membrane is comprised of an outer layer of PAN-DETA as a barrier to external microorganisms and structural support, an interlayer TiO2 NPs (Ct) as antibacterial function, and a contact layer (Gel) to improve biocompatibility and cell viability. The NFs membranes were characterized by scanning electron microscopy (SEM), surface profilometry, BET analysis, and water contact angle techniques to investigate their morphology, surface roughness, porosity, and wettability. The ML PAN-DETA-Ct-Gel wound dressing exhibited good surface roughness, porosity, and better wettability. Cell morphology, proliferation, and viability were determined using fibroblasts (3T3), and antibacterial assays were performed against six pathogens. The ML PAN-DETA-Ct-Gel NFs membrane showed good cell morphology, proliferation, viability, and antibacterial activity compared with other membranes. This new class of ML NFs membranes offers a multifunctional architecture with adequate biocompatibility, cell viability, and antibacterial activity.
Synthesis and characterization of a novel hydrogel based on carboxymethyl chitosan/sodium alginate with the ability to release simvastatin for chronic wound healing

Abstract:Since wound dressing has been considered a promising strategy to improve wound healing, recent attention has been focused on the development of modern wound dressings based on synthetic and bioactive polymers. In this study, we prepared a multifunctional wound dressing based on carboxymethyl chitosan/sodium alginate hydrogel containing a nanostructured lipid carrier in which simvastatin has been encapsulated. This dressing aimed to act as a barrier against pathogens, eliminate excess exudates, and accelerate wound healing by increasing the production of vascular endothelial growth factor (VEGF). Among various fabricated composites of dressing, the hydrogel composite with a carboxymethyl chitosan/sodium alginate ratio of 1:2 had an average pore size of about 98.44 ± 26.9 μm and showed 707 ± 31.9 % swelling and a 2116 ± 79.2 g/m2.per day water vapor transfer rate (WVTR), demonstrating appropriate properties for absorbing exudates and maintaining wound moisture. The nanostructured lipid carrier with optimum composition and properties had a spherical shape and uniform particle size distribution (74.46 ±7.9 nm). The prepared nanocomposite hydrogel displayed excellent antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria as well as high biocompatibility on L929 mouse fibroblast cells. It can release the loaded simvastatin drug slowly and over a prolonged period of time. The highest drug release occurred (80%) within 14 days. The results showed that this novel nanocomposite could be a promising candidate as a wound dressing for treating various chronic wounds in skin tissues.

Abstract:A cutaneous wound is caused by various injuries in the skin, which can be wrapped with an efficient dressing. Electrospinning is a straightforward adjustable technique that quickly and continuously generates nanofibrous wound dressings containing antibacterial and anti-inflammatory agents to promote wound healing. The present study investigated the physicochemical and biological properties of bromelain (BRO)- and silver nanoparticle (Ag NPs)-loaded gel-based electrospun polycaprolactone/chitosan (PCL/CS) nanofibrous dressings for wound-healing applications. Electron microscopy results showed that the obtained nanofibers (NFs) had a uniform and homogeneous morphology without beads with an average diameter of 176 ± 63 nm. The FTIR (Fourier transform infrared) analysis exhibited the loading of the components. Moreover, adding BRO and Ag NPs increased the tensile strength of the NFs up to 4.59 MPa. BRO and Ag NPs did not significantly affect the hydrophilicity and toxicity of the obtained wound dressing; however, the antibacterial activity against E. coli and S. aureus bacteria was significantly improved. The in vivo study showed that the wound dressing containing BRO and Ag NPs improved the wound-healing process within one week compared to other groups. Therefore, gel-based PCL/CS nanofibrous dressings containing BRO and Ag NPs could be a promising solution for healing skin wounds.

Abstract:Wound healing is a complex process and rapid healing necessitates a proper micro-environment. Therefore, design and fabrication of an efficacious wound dressing is an impressive innovation in the field of wound healing. The fabricated wound dressing in this scenario was designed using a combination of the appropriate coagulating and anti-bacterial materials like fibrinogen (as coagulating agent), nisin (as anti-bacterial agent), ethylenediaminetetraacetic acid (as anti-bacterial agent), and alginate (as wound healing agent). Biophysical characterization showed that the interaction of fibrinogen and alginate was associated with minor changes in the secondary structure of the protein. Conformational studies showed that the protein was structurally stable at 42 °C, is the maximum temperature of the infected wound. The properties of the hydrogel such as swelling, mechanical resistance, nisin release, antibacterial activity, cytotoxicity, gel porosity, and blood coagulation were assessed. The results showed a slow release for the nisin during 48 h. Antibacterial studies showed an inhibitory effect on the growth of Gram-negative and Gram-positive bacteria. The hydrogel was also capable to absorb a considerable amount of water and provide oxygenation as well as incorporation of the drug into its structure due to its sufficient porosity. Scanning electron microscopy showed pore sizes of about 14-198 µm in the hydrogel. Cell viability studies indicated high biocompatibility of the hydrogel. Blood coagulation test also confirmed the effectiveness of the synthesized hydrogel in accelerating the process of blood clot formation. In vivo studies showed higher rates of wound healing, re-epithelialization, and collagen deposition. According to the findings from in vitro as well as in vivo studies, the designed hydrogel can be considered as a novel attractive wound dressing after further prerequisite assessments.
Multifunctional Bi-layer collagen nanofiber-collagen/PLLA/ Zataria multiflora essential oil nanofiber for wound healing: Antibacterial, antifungal and antioxidant properties

Abstract:Multi-layer electrospun structures have attracted interests as multifunctional wound dressings. In the present study, a double-layered nanofibrous construct was prepared out of pure type I collagen nanofibers (as the inner layer) and collagen/PLLA/ Zataria Multiflora essential oil (as the outer layer). Extracted essential oil was analyzed by GC–Mass Spectrometry. Bi-layers were characterized by SEM, ATR-FTIR and XRD. Water contact angle and swelling studies demonstrated hydrophilicity and a high water absorption capability (1504% of the initial weight) of the nanofibers, respectively. Also, the essential oil showed a strong antioxidant activity (IC50: 7.4 ppm). Cumulative release profile of the essential oil from nanofibers illustrated a burst release in about 120 min and then, a sustained release up to 72 h. Broth micro-dilution, disk diffusion and AATCC 100 assays were performed and the bi-layer dressings exhibited strong antibacterial activity (against Bacillus subtilis , Staphylococcus aureus , Pseudomonas aeruginosa and Escherichia coli ) and antifungal activity against 4 Candida species. Viability of the fibroblast cells on the obtained structures was evaluated by alamar blue and live/dead assays that confirmed appropriate cyto-compatibility of the nanofibers. Therefore, biocompatible double-layered constructs with strong antimicrobial, antioxidant and water absorption capabilities would be promising as potential wound dressings. Abstract:Addressing the challenges of wound care requires accurate dressing selection. The combination of collagen with nanofibers (NFs) enhances its properties, improving cellular recognition. Multi-layered nanofibrous systems, particularly those with Polycaprolactone (PCL), offer higher mechanical attributes and prolonged substance release. We fabricated four types of nanofibrous bilayer mats (NBMs) using electrospinning (EN), each composed of an active layer containing a mixture of PCL and collagen loaded with and without Malva sylvestris (MS) extract, and an inactive layer with only PCL. The resulting bilayers were neat PCL/PCL-Collagen (PCLC), PCL/PCLC-5% MS, PCL/PCLC-10% MS, and PCL/PCLC-15% MS. Morphological and mechanical properties, porosity, water vapor transmission rate (WVTR), swelling rates, drug release profiles, cytotoxicity, cell attachment, and cytoprotective effect of bilayers were evaluated. FE-SEM imaging revealed smooth and filamentous mats without any beads, with Fiber diameter average (FDA) ranging from 128 ± 41 nm to 140 ± 51 nm. WVTR values of the NFs were comparable to commercial dressings like Tegaderm, Bioclusive, and Op Site. The mats demonstrated significant liquid uptake capacity, indicating their potential to handle wound exudate. They exhibited prolonged extract release, ensuring a sustained supply. All mats showed excellent blood compatibility, with our MS-loaded NBMs displaying rapid blood clotting times, essential for effective wound closure. FE-SEM images confirmed robust cell adhesion and spreading on both loaded and unloaded mats. The study underscored the potent cytoprotective effects of MS extract, in both free and encapsulated forms. The dose-dependent increase in cell viability suggests its potential in mitigating oxidative stress-induced cellular damage. The MTT (4,5-dimethyl- 2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay revealed that encapsulating MS extract in bilayer NFs significantly improved cell viability at 48 and 72 h, surpassing free MS extract treatment. Abstract:Designing a multifunctional conducting hydrogel wound dressing of suitable mechanical properties, adhesiveness, self-healing, autolytic debridement, antibacterial properties, and radical scavenging ability, as well as retaining an appropriate level of moisture around the wound is highly desirable in clinical application for treating cutaneous wounds healing. Here, we designed a novel class of electroactive hydrogel based on thiol-functionalized silver-graphene oxide nanoparticles (GO/Ag/TGA) core polyaniline (PANI) shell GO/Ag/TGA/PANI nanocomposites. Thus, a series of physically cross-linked hydrogel based on GO/Ag/TGA/PANI and poly(vinyl alcohol) (PVA) was prepared by freeze-thawing method. The hydrogel was characterized by XRD, UV, FTIR, TGA, TEM, SEM, Raman spectroscopy, cyclic voltammetry (CV), and four probes test. The hydrogel showed favorable properties such as excellent tensile strength, suitable gelation time (30-56 s), tunable rheological properties (G' ∼ 1 kPa), adhesiveness, and interconnected porous structure (freeze-dried). Besides this, the hydrogel also exhibits excellent exudate uptake capacity (10.4-0.2 g/g), high swelling ratio (72.4 to 93.5%), long-term antibacterial activity against multidrug-resistant (MDR) bacterial isolates, promising antioxidant (radical scavenging) efficiency, keeping the wound moisturized, prominent hemostatic efficiency, and fast self-healing ability to bear deformation. Interestingly, in vivo experiments indicated that electroactive hydrogels can significantly promote the healing rate of artificial wounds in rats, and histological analysis by H&E reveals higher granulation tissue thickness, collagen deposition, hair follicles, dermal papillary, keratinocytes, and marked increase (P < 0.05) in hydroxyproline at the wound site during 15 days of healing of impaired wounds. On the basis of vivo and vitro assay results, it is concluded that electroactive-hydrogel-attributed multifunctional properties may serve as suitable scaffold for treating chronic wound healing and skin regeneration.

Abstract:Hydrogels as skin wound dressings have been extensively studied owing to their good flexibility and biocompatibility. Nevertheless, the mechanical performance, adhesive capability, antifouling and antibacterial properties of conventional hydrogels are still unsatisfactory, which hinder the application of hydrogel for cutaneous healing. Here, we developed a novel biocompatible multifunctional hydrogel with super flexible, fatigue resistant, antifouling and self-adhesive capability for effective wound healing, where naturally rigid polymers including quaternized chitosan (QCS) and Tunicate cellulose nanocrystals (TCNCs) are used as bioactive cross-linkers and reinforcers to endow the hydrogel with excellent mechanical and antibacterial property, and the synergistic contributions from the poly(acrylic acid/methacrylate anhydride dopamine/sulfobetaine methacrylate) (poly(AA/DMA/SBMA)) chains and QCS endow the hydrogel with excellent adhesive property, antioxidant, antifouling and pH-responsive sustained drug release capabilities. The optimized hydrogel exhibited high tensile strength (77.69 KPa), large tensile strain (889.9 %), large toughness (307.51KJ.m-3), high adhesive strength (35.57 KPa) and ideal compressive property. The in vivo infected full-thickness skin model demonstrated that the hydrogel with vanvomycin sustained release ability efficiently improved the granulation tissue formation, facilitating collagen deposition and reducing inflammatory expression, thus effectively accelerating wound healing. This superiorly skin-adhesive antibacterial biocompatible hydrogel appears to be a promising candidate for wound therapy.

Abstract:<p>In this paper, Ag-Metal−organic framework loaded chitosan nanoparticles (0.1%A/1.5%CSNPs) and polyvinyl alcohol/sodium alginate/chitosan (PACS) were used as the upper and lower layers to successfully prepare a bilayer composite dressing for wound healing. The performance of bilayer dressing was evaluated. The lower layer (PACS) had uniform pore size distribution, good water retention, swelling, water vapor permeability, and biocompatibility while PACS had almost no antibacterial activity. The upper layer (/CSNPs) possessed excellent antibacterial activity and poor biocompatibility. As the upper layer, it can avoid direct contact with the skin and inhibit microbial invasion. In addition, the bilayer can adhere to a large number of red blood cells and platelets, promoting blood coagulation and cell proliferation. , CSNPs, /CSNPs and bilayer showed antibacterial activity in ascending order, due to the synergistic antibacterial action of the upper and lower layer. In vivo studies showed that both bilayer and PACS could significantly accelerate the wound healing, and the bilayer dressing showed more complete re-epithelialization with less inflammatory cells. In summary, this new bilayer composite is an ideal dressing for accelerating wound healing.</p>

Abstract:As the incidence of chronic wounds increases, the requirements for wound dressings are rising. The specific aim of this study is to propose a novel gallic acid (GA) functionalized silk fibroin (SF) and gelatin (Gel) composite wound dressing in which GA is used as an antibacterial and wound healing substance. Via electrospinning, SF, Gel, and GA mixed solutions could be conveniently fabricated into a composite nanofiber mat (SF-Gel-GA), consisting of uniform fibers with an average diameter around 134.57 ± 84 nm. The internal mesh structure of SF-Gel-GA provides sufficient drug loading capacity, proper moisture permeability, and proper degradation rate. SF-Gel-GA presents excellent biocompatibility. NIH-3T3 fibroblast cells could adhere and spread stably on the SF-Gel-GA surface with slightly promoted proliferation. In the presence of SF-Gel-GA, the growth of both Gram-positive and Gram-negative bacteria, includingStaphylococcus aureusandPseudomonas aeruginosa, is significantly inhibited in both plate and suspension cultures. A cutaneous excisional mouse wound model proves the efficient ability of SF-Gel-GA to promote wound healing. Compared with pure SF dressing and commercial Tegaderm Hydrocolloid3Mdressing, the wound closure rate with SF-Gel-GA treatment is significantly improved. The histological assessments further demonstrate SF-Gel-GA could facilitate collagen deposition, neovascularization, and epithelialization at wound sites to promote wound healing. In conclusion, a novel SF-Gel-GA composite wound dressing with efficient wound healing activities have been developed for chronic wound treatment with broad healing potential.

Abstract:Skin, as the biggest organ and outermost surface of the human body, is prone to injury due to various challenges, especially the expanding potential of accidents, which bring a huge social and economic burden. Hydrogels are emerging as the most promising candidate for wound dressings, which not only fulfill the varied requirements of dressings but also serve as drug carriers. But limited breathability, rapid drug release, and inadequate mechanical properties remains a significant challenge. Herein, we report a strong and tough sodium alginate/polyacrylate hydrogel fibers-based dressing with directional controlled drug release for wound healing promotion. Mimicking the bamboo structure, the drug solution is encapsulated within the fiber, and the rate of drug release can be modulated by controlling the wall thickness of the fiber. A cross-network structure in the hydrogel fiber through hydrogen bond and calcium ion crosslinking resulted in a 38 % increase in tensile strength. By precisely controlling the feeding process during weaving, drug-loaded fibers can be prepared at specific locations to facilitate targeted delivery to skin wound sites. Drug-loaded fabric has the breathability and biocompatibility required for dressings to promote wound healing. The findings highlight the potential of alginate/polyacrylate hydrogel fabrics for effective wound treatment.

Abstract:Chronic wounds, such as diabetic ulcers and pressure sores, pose significant challenges in modern healthcare due to their prolonged healing times and susceptibility to infections. This study aims to engineer a bilayered wound dressing (BLWD) composed of soy protein isolate/collagen with the ability to release Cinnamaldehyde, Artemisia absinthium (AA), and oxygen. Cinnamaldehyde, magnesium peroxide (MgO2), and AA extract were encapsulated. Nanoparticles were evaluated using scanning electron microscopy (SEM), dynamic light scattering, and ZETA potential tests. Swelling, degradation, water vapor penetration, tensile, MTT, SEM, oxygen release, AA extract release, and antibacterial properties were performed. An in vivo study was carried out to assess the final wound dressing under Hematoxiline&Eosin and Masson trichrome staining analysis and compared to a commercial product. According to the results, the synthesized nanoparticles had an average diameter of about 20 nm with a zeta potential in the range of -20 to -30 mV. The layers had uniform and dense surfaces. The maximum swelling and degradation of the dressing was about 130 and 13% respectively. Generally, better mechanical properties were observed in BLWD than in the single-layer case. More than 90% biocompatibility for the wound dressing was reported. The BLWD could inhibit the growth of Gram-positive and Gram-negative microorganisms. Histopathological analysis showed an acceptable wound-healing property. To sum up, the engineered wound dressing can be a good candidate for more clinical trials.

Abstract:Nanomaterial-based wound dressings have been extensively studied for the treatment of both minor and life-threatening tissue injuries. These wound dressings must possess several crucial characteristics, such as tissue compatibility, non-toxicity, appropriate biodegradability to facilitate wound healing, effective antibacterial activity to prevent infection, and adequate physical and mechanical strength to withstand repetitive dynamic forces that could potentially disrupt the healing process. Nevertheless, the development of nanostructured wound dressings that incorporate various functional micro- and nanomaterials in distinct architectures, each serving specific purposes, presents significant challenges. In this study, we successfully developed a novel multifunctional wound dressing based on poly(lactic acid) (PLA) fibrous membranes produced by solution-blow spinning (SBS) and electrospinning. The PLA-based membranes underwent surface modifications aimed at tailoring their properties for utilization as effective wound dressing platforms. Initially, beta-chitin whiskers were deposited onto the membrane surface through filtration, imparting hydrophilic character. Afterward, silver nanoparticles (AgNPs) were incorporated onto the beta-chitin layer using a spray deposition method, resulting in platforms with antimicrobial properties against both Staphylococcus aureus and Escherichia coli. Cytotoxicity studies demonstrated the biocompatibility of the membranes with the neonatal human dermal fibroblast (HDFn) cell line. Moreover, bilayer membranes exhibited a high surface area and porosity (> 80%), remarkable stability in aqueous media, and favorable mechanical properties, making them promising candidates for application as multifunctional wound dressings.
