Abstract:LP100R, LP10R and LP5R were isolated from peach gum by ultrafiltration. They were identified as AG II arabinogalactans composed of mannose, rhamnose, glucuronic acid, galactose, xylose and arabinose, which had a β-d-(1→6)-galactan backbone and were branched at O-3 and O-4. LP100R, LP10R and LP5R exist in a spherical conformation with the molecular weight of 8.50 × 104 g/mol, 4.77 × 104 g/mol and 2.40 × 104 g/mol, respectively. The binding affinities of LP fractions to galectin-3 (Gal-3) were 0.77 μM for LP100R, 2.88 μM for LP10R and 5.15 μM for LP5R, respectively. Meanwhile, an anti-proliferative assay revealed that LP100R possessed higher anti-proliferative activity against HepG2 cells (IC50, 4.5 mg/mL) and MCF-7 cells (IC50, 0.43 mg/mL) than did LP10R and LP5R, which were in accordance with their binding affinities to galectin-3. Therefore, LP fractions (especially LP100R) might exert the anti-tumor activity by directly inhibiting the Gal-3 mediated proliferation of cancer cells.

Abstract:An alkali-soluble single-component polysaccharide, named CPTC-2, was isolated and purified from the leaves of Taxus chinensis var. mairei. by ion-exchange and gel-permeation chromatography in series. The weight-average molecular mass (Mw) of CPTC-2 was about 73.53kDa determined by gel permeation chromatography (GPC). The structural characteristics of CPTC-2 were analyzed by gas chromatography (GC), Infrared (IR) spectrum, nuclear magnetic resonance (NMR) spectroscopy, periodate oxidation and Smith degradation studies, as well as methylation analysis. The results showed that CPTC-2 consisted of glucose, mannose, xylose, arabinose, rhamnose, and galactose with a molar ratio of 1.00:0.32:0.27:3.34:1.22:1.84. CPTC-2 was mainly composed of one type of sugar, α-glycosidic linkage. It had a backbone composed of α-(1→3) Araf, α-(1→5) Araf and α-(1→4) Galp with branches composed of α-(1→3,5) Araf and β-(1→3,6) Manp. In vitro anti-tumor experiments with SGC-7901 cells were performed and assessed by MTS (MPEG-2 Transport Stream) method and flow cytometry. The results showed CPTC-2 could inhibit the growth of SGC-7901 cells in a concentration-dependent manner via increased apoptosis. The relationship between the structure of CPTC-2 and its anti-tumor activity indicated that the α-configuration glycosidic bond residues may be essential for the anti-tumor activity of this polysaccharide.

Abstract:Polysaccharides from Ganoderma applanatum have attracted much attention due to their excellent pharmacological activities. In this work, a new water-soluble polysaccharide (GAP-2) was extracted and isolated from the fruiting bodies of Ganoderma applanatum . GAP-2 (21.3 kDa) was mainly composed of glucose (82.0%) and was spherical in aqueous solutions with a particle size distribution between 10 and 100 nm. Methylation analysis and nuclear magnetic resonance (NMR) results showed that the backbone structure of GAP-2 was →4)-α- D -Glc p -(1→ [3,6)-β- D -Glc p -(1] 8 →4)-α- D -Glc p -(1→6)-α- D -Glc p -(1→, and the branch was β- D -Glc p -(1→. Furthermore, the cell experiments indicated that GAP-2 could inhibit the proliferation and induce apoptosis of a variety of cancer cells (A549, SKOV3, and SMMC-7721) in a concentration-dependent manner, and significantly prevent the migration of A549 cells. In particular, GAP-2 hardly affected normal cells at experimental concentrations. Taken together, GAP-2 has inhibitory effects on tumor cells and has the potential to be a functional agent or an antitumor adjuvant.

Abstract:To discover anti-pancreatic cancer cells growth inhibitor, a new water-soluble glucan, WTMA, was isolated from the rhizome of Gastrodia elate Bl. The mean molecular weight of the polysaccharide was estimated to be 7.0 x 10(5) Da by high performance gel permeation chromatography (HPGPC). The structure of WTMA was characterized by GC, GC-MS, NMR and MALDI-TOF. WTMA was deduced as an alpha-(1 --> 4)-glucan with alpha-(1 --> 4) linked branches attached to O-6 at branch points. Bioactivity tests in vitro indicated that WTMA and its partial acid hydrolysis-derived mixture product, WTMA-AD-O, could significantly inhibit the proliferation of pancreatic cancer cells. However, WTMA-DE, the degraded polysaccharide without branch chain by isoamylase, did not inhibit the proliferation of pancreatic cancer cells. Thus side chain on WTMA was deduced to be required for the anti-cancer cell activity. Further study demonstrated that WTMA and all of its hydrolysis products had no inhibition effect on liver LO2 cells. (C) 2011 Elsevier Ltd. All rights reserved.

Abstract:Four polysaccharides (AEPA, AEPB, AEPC and AEPD) isolated and purified from the roots of Actinidia eriantha using gel filtration were subjected to composition analysis and valuated for the antitumor and immunomodulatory activity. The predominant neutral monosaccharides in four fractions were identified as galactose, arabinose and fucose, while the composition and ratio of the monosaccharides were different from one another. All four polysaccharides could significantly not only inhibit the growth of transplantable S180 sarcoma in mice, but promote splenocytes proliferation, natural killer cells activity, interleukin-2 production from splenocytes and serum tumor antigen-specific antibody levels in tumor-bearing mice, indicating that their antitumor activity might be achieved by improving immune response. Among four polysaccharides, AEPC and AEPD showed the higher antitumor and immunomodulatory activity. Taken together, the chemical composition of these polysaccharides could affect their antitumor and immunomodulatory activity, and AEPC and AEPD could be explored as antitumor agent with immunomodulatory activity.

Abstract:A homogeneous polysaccharide (PPB) was purified from fruiting bodies of Phellinus baumii. And in vitro antitumor and immunomodulation activities were investigated on HeLa, SGC-7901 and RAW264.7 cell lines. PPB inhibited the proliferation of HeLa and SGC-7901 cells significantly, and flow cytometric studies revealed that PPB could mediate the cell cycle in the G0/G1 and S phases. Furthermore, PPB could promote the growth and phagocytosis of RAW264.7 cells, activate the secretion of cytokines such as TNF-α and IL-6, which indicated that PPB had low toxicity. The results make PPB as a candidate adjuvant in cancer therapy.

Abstract:The structure and the effect of polysaccharide from peach gum (DPG2) on ameliorating DSS-induced acute colitis in mice were investigated in the present study. The results showed that DPG2 was identified as an AG II arabinogalactan with the backbone of β-D-(1 → 6)-galactan, which consisted of mannose, glucuronic acid, galactose, xylose and arabinose with a molar ratio of 4.64:1.02:2.61:39.82:3.89:48.02. Moreover, DPG2 behaved as a flexible chain conformation with a coil-like structure with a molecular weight (Mw) of 5.21 × 105 g/mol. Furthermore, the worm-like chain model parameters for DPG2 were estimated as follows: ML = 379 nm-1, q = 0.74 nm and d = 0.82 nm. The results of the animal assay showed that the intake of DPG2 not only effectively improved the phenotypes of DSS-induced colitis in mice, but also significantly improved the oxidative stress status of mice, such as regulating NO content and T-SOD and MPO levels and repairing oxidative damage to the colonic mucosa. Moreover, DPG2 improved the inflammation of DSS-induced colitis in mice by inhibiting the secretion of the proinflammatory cytokines TNF-α, IFN-γ, IL-1β, IL-6 and IL-17. Therefore, these results suggested that peach gum polysaccharide showed protective effects against colitis, and has great potential for the application of functional components in the food industry.

Abstract:In this paper, the structure difference between the polysaccharides isolated from fruit bodies (FGAP) and submerged fermentation system (SGAP) of Ganoderma applanatum was investigated by means of GPC, HPLC and IR, respectively. And their antitumor activities were evaluated against Sarcoma 180 in vivo. The results showed that FGAP and SGAP were typical polysaccharides with different molecular weights, monosaccharide components, and functional groups. Closely related to the distinct structures, FGAP exhibited a better antitumor activity than SGAP. Moreover, since FGAP contained carboxylate groups rather than SGAP, such groups were chemically introduced into SGAP (CSGAP) by carboxymethylation in order to identify their contribution to antitumor activity. The results demonstrated that the inhibition of CSGAP against Sarcoma 180 in vivo was significantly enhanced by comparison to the native SGAP and even higher than that of FGAP, suggesting that the carboxylate groups played a major role in antitumor activity of G. applanatum polysaccharide.
Structural Characteristics of Polysaccharide GP2a in Gardeniajasminoides and Its Immunomodulatory Effect on Macrophages

Abstract:Here, we elucidated the structural characteristics of a polysaccharide isolated from Gardenia jasminoides Ellis (labeled as GP2a) and its immunomodulatory activity. GP2a is an acidic polysaccharide with a molecular weight of 44.8 kDa, mostly comprising galacturonic acid. Methylation analysis revealed 4-GalpA (74.8%) to be the major sugar residue in GP2a. Nuclear magnetic resonance analysis indicated that its main chain comprised →4)-α-D-GalpA-6-OMe-(1→4)-α-D-GalpA-(1→ and →4)-α-D-GalpA-6-OMe-(1→2)-α-L-Rhap-(1→, with galactan and arabinans linked to the C-4 position of →2)-α-L-Rhap-(1→ residue as branched chains. Furthermore, GP2a showed no obvious toxicity to macrophages (RAW 264.7) while enhancing cell viability in a dose- and time-dependent manner. Compared with untreated cells, nitric oxide production and secretion of cytokines, such as tumor necrosis factor-α, interferon-γ, interleukin (IL)-1β, IL-6, and granulocyte macrophage colony stimulating factor, in GP2a-treated cells significantly increased after 48 h. At 300 μg/mL GP2a concentration, there was no significant difference in the cytokine levels in GP2a- and lipopolysaccharide-treated cells (the positive control). In summary, GP2a is a pectic polysaccharide with homogalacturonan and rhamnogalacturonan-I structural regions in the main chain. Based on its immunomodulatory effects in vitro, GP2a may have potential uses in functional food and medicine.

The Structural Characterization of a Novel Water‐Soluble Polysaccharide from Edible Mushroom Leucopaxillus giganteus and Its Antitumor Activity on H22 Tumor‐Bearing Mice

Abstract:In the present study, a novel cold water‐soluble polysaccharide fraction (LGP) with the average molecular weight of 1.78×106 Da was extracted and purified from Leucopaxillus giganteus and its primary structure as well as in vivo antitumor activity was evaluated. The monosaccharide composition of LGP was determined by ion chromatography to be galactose, xylose, glucose and fucose in a molar ratio of 2.568 : 1.209 : 1 : 0.853. Its backbone was composed of α‐D‐Glu, α‐D‐Xyl, α‐D‐Gal and α‐L‐Fuc. The results of in vivo antitumor experiment demonstrated that LGP could effectively protect immune organs, has excellent antitumor activity, and inhibit the proliferation of H22 solid tumors in a dose‐dependent manner. By analyzing Annexin V‐FITC/PI staining, cell cycle and mitochondrial membrane potential detection assay, we concluded that LGP induced apoptosis of H22 cells via S phase arrest and mitochondria‐mediated apoptotic pathway. Our results could provide valuable information for the potential application of LGP as an anti‐hepatoma agent.

Abstract:In this study, peach gum was sequentially extracted by water at 25 °C, 95 °C and 0.1 M NaOH, obtaining room temperature water-soluble (PG-WE), hot water-soluble (PG-HWE), and alkali-soluble (PG-AE) polysaccharide fractions. These three kinds of polysaccharide fractions exhibited a similar arabinogalactan structure according to FTIR spectrum and monosaccharide composition. However, their molecular weight and chain conformation were different based on high-performance size exclusion chromatography analysis, which resulted in different rheological behavior. PG-WE and PG-HWE presented non-Newtonian behavior, and the apparent viscosity of PG-WE was higher than that of PG-HWE at concentrations of 2.0 % and 3.0 % (w/w). Dynamic sweep tests showed that PG-HWE solutions behaved as viscous fluids, while PG-WE solutions tended to form gel-like structures at concentrations higher than 2 % (w/w). Furthermore, PG-WE showed significant in vitro inhibition against α-glucosidase activity in a dose-dependent manner and reduced the fluorescence of α-glucosidase through dynamic quenching mechanism.

Abstract:To better understand the structure-function relationship of Gastrodia elata polysaccharides (PGEs), PGEs were extracted by ultrasound-assisted extraction method and the effects of extraction time on the structure and conformation of PGEs were evaluated by asymmetrical flow field-flow fractionation (AF4) coupled online with multiangle light scattering (MALS) and differential refractive index (dRI) detectors (AF4-MALS-dRI). Besides separation, AF4-MALS-dRI can provide more information about PGEs, such as size and molecular weight (Mw) distributions, apparent density, and conformation. The effects of PGEs on the proliferation, apoptosis, and cell cycle of MCF-7 cells were investigated. The cell activity assay indicated that the PGEs can inhibit the growth of MCF-7 cells by inducing late apoptosis. The results indicated that PGEs with a spherical conformation and compact structure seem to be beneficial to inducing MCF-7 cells late apoptosis. Moreover, results demonstrated that the information obtained by AF4-MALS-dRI is valuable for better understanding of the relationship of structure-activity of PGEs.

Abstract:A novel Angelica dahurica polysaccharide (ADP) with Mw of 6.09 × 103 Da was isolated. The contents of total sugar and uronic acid in ADP were 91.04% and 12.69%. The structure characteristics indicated that ADP was an acidic polysaccharide consisting of rhamnose, arabinose, galactose, glucose, mannose, glucuronic acid and galacturonic acid (0.09: 0.61: 1.88: 1: 0.14: 0.63: 0.03). Moreover, there were →3)-Manp-(1→, →4, 6)-Galp-(1→, →4)-Galp-(1→, →3)-Glcp-(1→, →5)-Araf-(1→, →2)-Galp-(1→ in ADP with relative molar ratios of 0.32:0.57:0.29:0.95:0.71:0.26. In vivo experiments suggested that ADP significantly inhibited the tumor growth of mice, increased the activities of spleen lymphocytes and natural killer (NK) cells, improved the cytokine level (IL-2 and TNF-α) and the proportions of lymphocyte subsets in the peripheral blood. The tumor cell progression was arrested in the G1 phase, and the apoptosis rate of tumor cells were 7.54% and 19.32% at the dose of 100 and 200 mg/kg, which was consistent with the results of pathological observation. In summary, the study might provide a theoretical basis for the application on functional foods containing Angelica dahurica polysaccharides.

Abstract:The structural characteristics, acetylcholinesterase inhibitory activity, and immune effects of two purified acid polysaccharides from Chrysanthemum morifolium cv. Gongju were investigated. GJP-1 and GJP-2 were isolated and purified using DEAE-sepharose and Sephadex G-100 chromatography. GJP-1 consisted of Man, Rha, Ara, Glc, Gal, and GalA, with a ratio of 1.99:3.98:81.2:2.65:7.03:3.15, while GJP-2 comprised of Man, Rha, Ara, Glc, Gal, and GalA in a ratio of 0.27:2.36:8.03:38.44:2.38:48.52. GJP-2 is a kind of pectin-type polysaccharide. GJP-1 and GJP-2 differed in average molecular weight of 5.86 kDa and 4.92 kDa. The results indicated that GJP-1 and GJP-2 significantly inhibited the activity of acetylcholinesterase and had good immunomodulatory activities. GJP-1 and GJP-2 are bioactive polysaccharides with the potential to improve cognitive function. GJP-2 promoted more proliferation, as well as the release of tumor necrosis factor-α and interferon γ. Clarifying the structures and bioactivities of polysaccharides could lay a foundation for the application of Gongju in functional foods and medicines production.

Abstract:The edible mushroom G. frondosa has been used as a kind of functional food for the prevention and therapy of various diseases in Asian countries. In the present work, a novel acid-soluble polysaccharide (GFAP) was successfully isolated from G. frondosa under room temperature and hydrochloric acid solution treatment. Results of chemical composition analysis, UV and HPGPC spectra showed that GFAP mainly contained 94.28% of carbohydrate with the average molecular weight of about 644.9 kDa. GC, FT-IR, NMR and methylation analysis further indicated that GFAP was a neutral sugar mainly composed of (1 → 3)-β-D-Glcp and (1 → 3)-α-D-Manp. The in vivo antitumor experiments demonstrated that GFAP could effectively protect thymuses and spleens of tumor-bearing mice and inhibit the growth of H22 solid tumors with the inhibitory rate of 36.72%. Besides, GFAP could significantly improve the activities of NK cells, macrophages, CD19+ B cells and CD4+ T cells, leading to the apoptosis of H22 cells via G0/G1 phase arrested. Our data demonstrated that GFAP holds great application prospect to be a safe and effective antitumor adjuvant in the future.

Molecular mass and antitumor activities of sulfated derivatives of alpha-glucan from Poria cocos mycelia

Abstract:Two kinds of water-insoluble (1-->3)-alpha-D-glucan samples, ab-PCM3-I and ac-PCM3-I, isolated from different Poria cocos mycelia were sulfated, to produce two series of water-soluble derivatives ab-PCM3-I-S1-S5 and ac-PCM3-I-S1-S5, respectively. The derivatives having different weight-average molecular mass (Mw) were produced by changing reaction temperature and time as well as molar ratios between chlorosulfonic acid and number of hydroxyl groups in the glucan. The degrees of substitution (DS) of the sulfated derivatives were analyzed by elemental analysis (EA) to be 0.39-0.67 for ab-PCM3-I-S and 0.73-0.96 for ac-PCM3-I-S, respectively. The Mw and the intrinsic viscosity ([eta]) of the samples ab-PCM3-I-S and the ac-PCM3-I-S were measured by size exclusion chromatography combined with laser light scattering (SEC-LLS) and viscometry in phosphate buffer solution (PBS) at 37 degrees C. The results indicated that their Mw ranged from 2.0 to 11.3 x 10(4) for the samples ab-PCM3-I-S, and 4.7 to 40.0 x 10(4) for the samples ac-PCM3-I-S. Moreover, the antitumor activities of the sulfated derivatives ab-PCM3-I-S and ac-PCM3-I-S against Sarcoma 180 tumor cell tested both in vitro and in vivo are significantly higher than those of the native alpha-D-glucans. Therefore, a moderate range of molecular mass from 2.0 x 10(4) to 40.0 x 10(4), relatively high chain stiffness and good water solubility of the sulfated derivatives are beneficial to the enhancement of their antitumor activities.
Structures and anti-melanoma activities of two polysaccharides from Angelica sinensis (Oliv.) Diels

Abstract:Polysaccharide is one of the necessary macromolecules in life activities, and it is also a very promising natural product for tumor prevention and treatment. In this study, two homogeneous polysaccharides (APS-4I and APS-4II) were isolated from Angelica sinensis (Oliv.) Diels. APS-4I was a linear glucan with molecular weight of 16.1 kDa, which was composed of 88.4% α-1,6-Glcp, 4.1% α-1,2-Glcp, 3.9% α-1,3-Glcp, and 2.8% α-T-Glcp. APS-4II was a novel polysaccharide with molecular weight of 11.1 kDa, which consisted of 55.4% α-1,6-Glcp, 10.4% α-1,3,5-Araf, 8.7% α-T-Araf, 9.2% α-1,5-Araf, 4.0% α-1,3-Araf, 3.6% α-1,4-Galp, and 9.1% β-1,3-Galp. NMR results demonstrated that APS-4II has a backbone composed of →6)-α-Glcp-(1 → 6)-α-Glcp-(1 → 5)-α-Araf-.(1 → 5)-α-Araf-(1 → 3,5)-α-Araf-(1 → 3)-β-Galp-(1 → 3)-β-Galp-(1 → 4)-α-Galp-(1 → 3)-α-Araf-(1 → 3,5)-α-Araf-(1→. Both APS-4I and APS-4II inhibited the tumor growth of B16-bearing mice, and the suppressive effect of APS-4II reached 64.7 ± 7.3%. Meanwhile, there were higher lymphocyte numbers and the levels of IL-2, IFN-γ, and TNF-α in peripheral blood of APS-4II-treated mice than those in APS-4I-treated mice. Furthermore, APS-4II showed a higher inhibitory effect on the proliferation of B16 cells and stronger promoting effects on the proliferation of splenocytes, the phagocytosis of peritoneal macrophages, and the cytotoxicity of NK cells. These results demonstrated that APS-4II could be a promising therapeutic agent for melanoma.

Abstract:To achieve sustainable development, increasing attention has been paid to the utilization of renewable polysaccharides extracted from plant gum instead of synthetic materials. Peach gum polysaccharide (PGP) is a typical polysaccharide, which can be readily obtained by hydrolysis of peach gum, one of the abundant plant gums in the world. In the past decade, the research on the hydrolysis, structure, properties and applications of PGP has aroused great interest. The PGP with highly branched macromolecular structure shows remarkable merits of numerous functional groups, excellent water solubility, good biocompatibility, favorable emulsifying property, fine antioxidant and antibacterial activity, and low cost. The application of PGP has expanded from the pharmaceutical field to the fields of food, adsorbents, functional carbon materials, binders and gel materials. This review systematically introduces the research progress of PGP, as well as the opportunities and challenges faced by PGP in scientific research and practical application.

Abstract:In the present study, we investigated the in vitro digestion and fermentation characteristics of three peach gum polysaccharides (PGPs) of different molecular weights; i.e., AEPG2 (1.64 × 107 g/mol), DPG2 (5.21 × 105 g/mol), and LP100R (8.50 × 104 g/mol). We observed that PGPs were indigestible during the oral, gastrointestinal, and intestinal stages. However, they were utilized by the gut microbiota with utilization rates in the order of DPG2 > AEPG2 > LP100R. Furthermore, arabinose in PGPs was preferentially utilized by the gut microbiota followed by galactose and xylose. Fermentation of peach gum polysaccharides could significantly increase the production of short-chain fatty acids (SCFAs), especially n-butyric acid. In addition, PGPs with different molecular weights values were predominantly fermented by different bacterial species. AEPG2 and DPG2 were fermented by the Bacteroidetes bacteria Bacteroides, while the dominant n-butyrate-producing bacteria was Faecalibacterium. While the LP100R was fermented by Bacteroides, Parabacteroides, Phascolarctobacterium, Dialister, Lachnospiraceae, and Blautia, the dominant n-butyrate-producing bacteria was Megamonas. These results indicated that PGPs are potential prebiotics for the food industry.

Functional Characterization of a Potent Anti-Tumor Polysaccharide in a Mouse Model of Gastric Cancer

Abstract:AimsNatural polysaccharides are emerging as a new class of immunomodulatory agents due to their potent immunostimulatory effects and suitable biocompatibility. The aim of this study was to identify potent and selective anticancer activity of a bioactive polysaccharide.Materials and methodsIn vitro, viability assay was performed to screen 16 of bioactive polysaccharides in a panel of normal and cancer cell lines. Foci formation, soft agar, BrdU incorporation, cell cycle analyses, and β-galactosidase staining were performed to validate the screening results. In vivo, both murine gastric cancer syngeneic and a human gastric tumor xenografts models were applied. Tumor histology, immunohistochemical staining, cytokine array and flow cytometry analyses were assayed.Key findingsBSP (bamboo shaving polysaccharides) was identified as the most selective polysaccharide for inhibiting the growth of six gastric cancer cell lines while having no toxic effect on normal gastric mucosal cells. Similarly, BSP had more potent killing effect on a subset of human stomach cancer cells than liver or lung cancer cells. In vivo, BSP significantly inhibited tumor growth and prolonged the survival of mice bearing a gastric tumor; these effects are mediated by tumor cell apoptosis and remodeling of the tumor microenvironment by boosting both immune cell subpopulations and cytokine production in murine gastric cancer syngeneic model. A significant decrease of F4/80-positive tumor-associated macrophages was also observed.SignificanceThe findings of this study suggest that the potent and selective anti-tumor activity of bioactive polysaccharides such as BSP warrants clinical testing for the treatment of gastric cancer.
