Abstract:The biosynthesis of zinc oxide nanoparticles (ZnONPs) offers great potential for plant disease management due to their potent antimicrobial properties and environmental safety. However, the precise mechanisms underlying their antifungal mode of action and role in suppressing mycotoxins remain unclear. This study aims to elucidate the mechanisms by which ZnONPs suppress the pathogenic fungus Fusarium graminearium, known to cause Fusarium head blight in wheat. Additionally, it investigates how ZnONPs mitigate the production of mycotoxins, which pose risks to humans and ruminants. The study demonstrates that ZnONPs, bioproduced by Pseudomonas poae (P. poae), inhibit not only fungal growth, colony formation, and spore germination, but also significantly reduce mycotoxin production of F. graminearium by inhibiting the synthesis of deoxynivalenol (DON), downregulating the FgTRI gene, and causing morphological alterations of the toxisomes. The results also highlight that ZnONPs exert significant effects on fungi through multiple mechanisms, including cell wall damage and the generation of reactive oxygen species (ROS). Moreover, ZnONPs effectively inhibit F. graminearium in wheat leaves and coleoptiles. Fluorescence microscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), and optical microscopy all show that ZnONPs stop F. graminearium from getting into wheat plants and colonising them. Overall, the findings of this study provide evidence that ZnONPs are highly effective in reducing F. graminearium colonization in wheat plants and effectively decreasing mycotoxin production through multiple pathways.

Abstract:Fusarium oxysporum is a destructive plant pathogen with robust survival mechanisms, complicating control efforts. This study aimed to develop nanoformulated fungicides, screen antagonistic bacteria, and evaluate their combined efficacy. A novel self-emulsifying nanoemulsion (DZW) was formulated using zein and benzaldehyde-modified wheat gluten (BgWG) as carriers for dimethachlon (DTN). The preparation process optimized material ratios and emulsification techniques. Concurrently, antagonistic bacterial strains against F. oxysporum were screened via the plate standoff method, identifying Bacillus velezensis MLY71 as both antagonistic and compatible with DTN. The DZW nanoemulsion achieved a particle size of 93.22 nm, an encapsulation efficiency (EE) of 90.57%, and a DTN loading capacity (LC) of 67.09%, with sustained release over 96 h. The combination of DTN (0.04 mg·mL−1) and B. velezensis MLY71 (1 × 104 CFU·mL−1) achieved a 76.66% inhibition rate against F. oxysporum MLY127, 1.71 times greater than DTN alone, indicating significant synergy. At a DTN concentration of 0.20 mg·mL−1, the combination of DZW and MLY71 showed a synergy coefficient of 1.33. This synergy was also observed in soil environments, indicating its adaptability for controlling soil-borne pathogens. As sustainable management continues to gain attention in agricultural disease control, this study offers a promising strategy for achieving higher efficacy with the same fungicide dose or satisfactory control with reduced fungicide application. The excellent drug-loading performance of BgWG also expanded the applications of the wheat by-product gluten.

Antifungal efficacy of Bacillus amyloliquefaciens ZK-9 against Fusarium graminearum and analysis of the potential mechanism of its lipopeptides

Abstract:Fusarium graminearum is a destructive fungal pathogen that seriously threatens wheat production and quality. In the management of fungal infections, biological control is an environmentally friendly and sustainable approach. Here, the antagonistic strain ZK-9 with a broad antifungal activity was identified as Bacillus amyloliquefaciens. ZK-9 could produce extracellular enzymes such as pectinase, protease, cellulase, and amylase, as well as plant growth-promoting substances including IAA and siderophore. Lipopeptides extracted from strain ZK-9 had the high inhibitory effects on the mycelia of F. graminearum with the minimum inhibitory concentration (MIC) of 0.8 mg/mL. Investigation on the action mechanism of lipopeptides showed they could change the morphology of mycelia, damage the cell membrane, lower the content of ergosterol and increase the relative conductivity of membrane, cause nucleic acid and proteins leaking out from the cells, and disrupt the cell membrane permeability. Furthermore, metabolomic analysis of F. graminearum revealed the significant differences in the expression of 100 metabolites between the lipopeptides treatment group and the control group, which were associated with various metabolic pathways, mainly including amino acid biosynthesis, pentose, glucuronate and glycerophospholipid metabolism. In addition, strain ZK-9 inhibited Fusarium crown rot (FCR) with a biocontrol efficacy of 82.14 % and increased the plant height and root length by 24.23 % and 93.25 %, respectively. Moreover, the field control efficacy of strain ZK-9 on Fusarium head blight (FHB) was 71.76 %, and the DON content in wheat grains was significantly reduced by 69.9 %. This study puts valuable insights into the antifungal mechanism of lipopeptides against F. graminearum, and provides a promising biocontrol agent for controlling F. graminearum.
Drug-eluting stent use after coronary atherectomy: results from a multicentre experience – The ROTALINK I study

Abstract:Aims Until now, there is no medium- to long-term clinical evidence of the best treatment after rotational atherectomy. Methods From the databases of seven high-volume centres, years 2005–2010, we retrospectively analysed the long-term outcome of patients who had undergone rotational atherectomy followed by plain-balloon angioplasty or implantation of drug-eluting stent (DES) or bare metal stent (BMS). Primary endpoint was the incidence of major adverse cardiovascular events (MACE: death, myocardial infarction, target-lesion-revascularization) at longest available follow-up. Results In this registry, we enrolled 1397 patients with 1605 lesions, followed for 28.4 ± 21 months. DES-treated patients were more frequently diabetic, had more lesions treated and received a higher number of stents. In-hospital MACEs were significantly higher in DES patients (7.6 vs. 2.6 vs. 2.9%, respectively, P = 0.0001 for both), mainly due to a higher incidence of myocardial infarction (6.4 vs. 1.2 vs. 2.1%, P = 0.0001). The 2-year follow-up showed a significantly lower incidence of MACE in DES patients (15.1 vs. 24.2 vs. 20.8%, P = 0.001 for both), driven by a lower incidence of target-lesion revascularization (8 vs. 14.6 vs. 13.9%, P = 0.002). Myocardial infarction rate was lower in the DES group as well (0.4 vs. 3.1% in BMS, P = 0.001). At multivariate analysis, BMS implantation and balloon angioplasty were independent predictors of long-term MACE. DES implantation was associated with a lower risk of long-term myocardial infarction [hazard ratio 0.15, 95% confidence interval (95% CI) 0.04–0.67] and target-lesion revascularization (hazard ratio 0.42, 95% CI 0.21–0.82). Male sex and DES use were independent predictors of the absence of MACE. Conclusion After rotational atherectomy, DES implantation appears to be a preferable strategy, as it is associated with lower long-term MACE, despite an unexpected increase in periprocedural myocardial infarction.
Bioengineered chitosan-magnesium nanocomposite: A novel agricultural antimicrobial agent against Acidovorax oryzae and Rhizoctonia solani for sustainable rice production

Abstract:Chitosan is a potent biopolymer having promising antimicrobial properties against phytopathogens. Recently, engineered nanomaterials (ENMs) have gained much attention due to their potential application in the plant disease management. In this study, we reported the green synthesis of chitosan-magnesium(CS-Mg) nanocomposite and its antimicrobial activity against two rice pathogens namely Acidovorax oryzae and Rhizoctonia solani for the first time. The green MgO nanoparticles synthesized by using a native Bacillus sp. strain RNT3, were used to fabricate CS-Mg nanocomposite utilizing one-pot synthesis method. The synthesis of CS-Mg nanocomposite was further confirmed by using UV-vis spectroscopy, whereas, FTIR and XRD analysis showed the capping of CS-Mg nanocomposites by different functional groups together with their crystalline structure, respectively. Besides, SEM and TEM images revealed the spherical shape along with the particles size ranging from 29 to 60 nm. Moreover, EDS analysis confirmed the elemental purity of nanocomposite. The CS-Mg nanocomposite showed remarkable antimicrobial activity against A. oryzae and R. solani and significantly inhibited the growth as compared to non-treated control. The ultrastructure studies showed damaged structure of cell wall and internal cellular organelles after treatment with 100 mu g mL(-1) CS-Mg nanocomposite. The results of this study indicated that CS-Mg nanocomposite-based antimicrobial agents could be considered as promising nanopesticides against phytopathogens in plant disease management. Antifungal activity of the lemongrass and clove oil encapsulated in mesoporous silica nanoparticles against wheat's take-all disease

Abstract:Combined application of plant essential oils (EOs) with known antimicrobial effects and silica nanocapsules with high loading capacity and protection capability of the EOs make them proper candidates for creating environmentally friendly fungicides. In this study, EOs of the Lemongrass (LGO) and Clove (CO) were used against Gaeumannomyces graminis var. tritici (Ggt), a causal agent of take-all disease of wheat. To provide controlled delivery of the EOs, they were encapsulated into mesoporous silica nanoparticles (MSNPs) and then compared to the effects of pure EOs both in- vitro and in- vivo. MSNPs were synthesized via the sol-gel process. Various techniques such as Fourier transform infrared spectroscopy (FTIR), the Brunauer–Emmett–Teller (BET), thermogravimetric analysis (TGA), and UV–Vis spectroscopy were used to evaluate the successful loading of the EOs into the pore of MSNPs. The encapsulation efficiency (EE) was calculated as high as 84.24% for LGO and 80.69% for CO, while loading efficiency (LE) was determined 36% and 29% for LGO and CO, respectively. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) displayed spherical shapes and porous structures with average diameters of 50–70 nm. Recognition of the main components of the EOs via gas chromatographic-mass spectrometry (GC–MS) before and after the EO loading, detected eugenol and citral as the most frequent compounds in LGO and CO, respectively. For antifungal test in- vitro, selected concentrations of the pure EOs, EOs loaded in MSNPs (EOs- MSNPs) and Mancozeb ® fungicide based on pre-tests were mixed using potato dextrose agar (PDA). The inhibition percentage (IP) of fungal growth at each concentration, as well as minimum inhibition concentration (MIC) and minimum fungicidal concentrations (MFC) were obtained. The results indicated that antifungal effects in the encapsulated form increased by up to three times. In- vivo, the sterile wheat seeds were treated with pure EOs, EOs-MSNPs, and mancozeb at MFC concentration. Also, in order to keep on the EOs-MSNPs around the seeds, sodium alginate was used. The consequences of in- vivo experiments indicated that rate of disease control in presence of EOs-MSNPs and mancozeb was the same (~70%) and higher than pure EOs (LGO: 57.44%, CO: 49%). Also, improving the growth parameters in wheat plant, the covering of the EOs-MSNPs in alginate, had better control (84%) than that of EOs-MSNPs alone. Further, the release kinetics studies showed a gradual release of LGO and CO from MSNPs for four weeks in water and for five weeks in the soil-plant system. To the best of our knowledge, this is the first report of the control effect of LGO, CO, and their nanocapsule in MSNPs against the take-all disease of wheat. These results showed that the EOs-MSNPs can be a safe product for the efficient control of take-all disease in wheat crop.

Abstract:Nanochitin whisker (NC) is an advanced nanobiomaterial with novel physicochemical and biological properties. Fusarium pseudograminearum (Fpg) is an important pathogenic fungus causing wheat crown rot disease. This study explored the mode of action of NC against Fpg as a target microorganism. The effects of different treatments and concentrations of NC on the fungal growth and conidial germination were investigated by in vitro bioassay. The impacts of NC on cell structure integrity, membrane permeability, pathogenesis related key enzymes activity, and the mycotoxin production were examined by electron microscopy, fluorescence spectroscopy, IR spectroscopy, conductometry, and spectrophotometry, respectively. The results showed that NC significantly reduced hyphal growth, and the spore germination rate of Fpg declined by 33.0 % and 23.2 % when Fpg was treated with 30 and 300 μg/mL of NC, respectively. NC vigorously influenced structural stability of cell wall by destroying dextran structure, and strongly stimulated ergosterol production altering membrane integrity of the fungus. It reduced the activities of enzymes related to energy-supply like nicotinamide adenine dinucleotide oxidase and succinate dehydrogenase remarkably. The production of fungal mycotoxin deoxynivalenol was also decreased by NC. These findings provide an important basis for fully understanding the mechanism of nanobiomaterial in plant fungal disease control.
Toxicity and action mechanisms of silver nanoparticles against the mycotoxin-producing fungus Fusarium graminearum

Abstract:Introduction: Fusarium graminearum is a most destructive fungal pathogen that causes Fusarium head blight (FHB) disease in cereal crops, resulting in severe yield loss and mycotoxin contamination in food and feed. Silver nanoparticles (AgNPs) are extensively applied in multiple fields due to their strong antimicrobial activity and are considered alternatives to fungicides. However, the antifungal mechanisms and the effects of AgNPs on mycotoxin production have not been well characterized. Objectives: This study aimed to investigate the antifungal activity and mechanisms of AgNPs against both fungicide-resistant and fungicide-sensitive F. graminearum strains, determine their effects on mycotoxin deoxynivalenol (DON) production, and evaluate the potential of AgNPs for FHB management in the field. Methods: Scanning electron microscopy (SEM), transmission electron microscopy (TEM), and fluorescence microscopy were used to examine the fungal morphological changes caused by AgNPs. In addition, RNA-Seq, qRT-PCR, and western blotting were conducted to detect gene transcription and DON levels. Results: AgNPs with a diameter of 2 nm exhibited effective antifungal activity against both fungicide-sensitive and fungicide-resistant strains of F. graminearum. Further studies showed that AgNP application could impair the development, cell structure, cellular energy utilization, and metabolism pathways of this fungus. RNA-Seq analysis and sensitivity determination revealed that AgNP treatment significantly induced the expression of azole-related ATP-binding cassette (ABC) transporters without compromising the control efficacy of azoles in F. graminearum. AgNP treatment stimulated the generation of reactive oxygen species (ROS), subsequently induced transcription of DON biosynthesis genes, toxisome formation, and mycotoxin production. Conclusion: This study revealed the underlying mechanisms of AgNPs against F. graminearum, determined their effects on DON production, and evaluated the potential of AgNPs for controlling fungicide-resistant F. graminearum strains. Together, our findings suggest that combinations of AgNPs with DON-reducing fungicides could be used for the management of FHB in the future.
Limonene formulation exhibited potential application in the control of mycelial growth and deoxynivalenol production in Fusarium graminearum

Abstract:Preventing grain from fungi and subsequent mycotoxins contamination has attracted notable attention. Present study demonstrated the limonene-formulated product Wetcit®, might be a biocontrol agent and potential alternative to synthetic fungicides to control Fusarium graminearum growth and deoxynivalenol (DON) production. The limonene formulation exhibited antifungal activity against F. graminearum with the EC50 at 1.40 μl/ml, electron microscopy and staining analysis showed limonene formulation could significantly decrease the quantity, length and septa of conidia, caused hyphal break and shrink, damaged the structures of cell membrane, cell wall, vacuoles and organelles in the hypha. Further study revealed the antifungal and antitoxic mechanism of limonene formulation against F. graminearum, limonene formulation significantly inhibited the toxisome and DON formation, was associated with the down-regulation of trichothecenes biosynthesis genes expression and many energy metabolism pathways as well as the inhibition of lipid droplets, the disturbed energy homeostasis and intracellular structures might ultimately inhibit fungal growth and DON production. In addition, limonene formulation enhanced the antifungal activity of triazole fungicides tebuconazole and mefentrifluconazole against F. graminearum, indicated limonene formulation has valuable potential as a bio-alternative fungicide and eco-friendly compound preparation for the effective management of F. graminearum and DON contamination in agriculture.
Role of Aqueous Chlorine Dioxide in Controlling the Growth of Fusarium Graminearum and Its Application on Contaminated Wheat

Abstract:Fusarium graminearum is the main worldwide causal agent of Fusarium head blight grain disease. The production of mycotoxin caused by Fusarium head blight epidemics of wheat affects the flour and malt products, which results in heavy economic losses to farmers. In this paper, an inactivation procedure for F. graminearum F7875 spores was tested and found to follow a first-order kinetic model. We characterized the inactivation of F7875 by aqueous chlorine dioxide (ClO2) under different concentrations (5 -15 mg L-1). The results suggest that aqueous ClO2 significantly enhanced inactivation of F7875 at high concentration (15 mg L-1) compared to lower concentration of 5 and 10 mg L-1. The time required for a five-log reduction of F7875 spores was calculated based on the established models. Observed by scanning electron microscope, the ClO2 destroys the structure of the outer coats of F7875, the surface of F7875's hyphal appeared smooth prior to treatment, but after treatment with 30 mg L-1 aqueous ClO2, it appeared rugged and wrinkled. In conclusion, our study shows that treatment with aqueous ClO2 is an effective, fast, and safe method to control F. graminearum in grain production(.) (C) 2017 Published by Elsevier Ltd.
The association between daytime attendance and out-of-hours frequent attendance among adult patients in general practice.

Abstract:BACKGROUND Frequent attenders (FAs) account for a large proportion of daytime consultations in general practice. However, no studies have so far been conducted to establish whether daytime FAs are also out-of-hours FAs. AIM To analyse the association between daytime attendance and out-of-hours frequent attendance. DESIGN OF STUDY A registry-based study of individual face-to-face contacts with general practice during daytime and out of hours, based on National Health Insurance files. SETTING General practices in Aarhus County, Denmark (630,000 inhabitants). METHOD A total of 416,172 adults, i.e. all adults registered with a general practice during a 12-month period from November 1997 to October 1998 were included. Odds ratios (OR) for frequent attendance out of hours were stratified for age, sex, and frequency of contacts during daytime were calculated. RESULTS A strong association was observed between daytime level of attendance and out-of-hours level of attendance (OR = 2.9-6.3 among patients with low daytime attendance, OR = 33.7-99.8 among daytime FAs). Daytime FAs accounted for one-third of the daytime contacts, one-third of the out-of-hours contacts, and 42% of out-of-hours FAs. More than half of the daytime FAs did not attend out of hours. Two per cent of the out-of-hours FAs had no contacts during the daytime. CONCLUSION Frequent attendance in daytime was very strongly associated with frequent attendance out of hours, and daytime FAs accounted for a large proportion of all contacts with general practice.
Nano protective membrane coated wheat to resist powdery mildew

Abstract:The plant pathogenic fungus Blumeria graminis f. sp. tritici infects wheat and reduces its yield. The policy of reducing fertilizer and biocide use in sustainable agriculture has prompted researchers to develop more green and efficient management strategies. In this study, a novel nanoprotective membrane (kaolin-nano titanium dioxide-liquid paraffin, referred to as KTP) that could effectively prevent powdery mildew of wheat was prepared by using 1 g/L kaolin, 2 g/L nanotitanium dioxide and 8% (v/v) liquid paraffin. The prevention and control effects of KTP spraying in advance in the pot and field experiments were 98.45% and 83.04%, respectively. More importantly, the weight of 1000 grains of wheat pretreated with KTP was 2.56 g higher than that of wheat infected with powdery mildew, significantly improving wheat yield. KTP delayed the germination of powdery mildew spores on the leaf surface, and inhibited the formation of mycelia. In addition, KTP did not affect the growth of wheat or the survival of earthworms. KTP nanoprotective membrane are a green and safe prevention and control materials that are which is expected to be widely used in agriculture to control wheat powdery mildew.

Discovery of Frenolicin B As Potential Agrochemical Fungicide for Controlling Fusarium Head Blight on Wheat.

Abstract:In this study, the supernatant extract from fermentation broth of Streptomyces sp. NEAU-H3 showed strong antifungal activity against Fusarium graminearum strain PH-1 in vitro and in vivo. Three known pyranonaphthoquinones were isolated by means of an activity-guided method, and frenolicin B was characterized as the main active ingredient. Frenolicin B displayed strong antifungal activity against F. graminearum strain PH-1 with an EC50 value of 0.51 mg/L, which is lower than that of carbendazim (0.78 mg/L) but higher than that of phenamacril (0.18 mg/L). Frenolicin B could also strongly inhibit the mycelial growth of Fusarium species, including F. graminearum and F. asiaticum, as well as carbendazim-resistant Fusarium strains isolated from field, with EC50 values of 0.25-0.92 mg/L. Results from field experiments showed that the efficacy of frenolicin B in controlling Fusarium head blight at a treatment concentration of 75 g ai/ha was better than those of phenamacril (375 g ai/ha) and carbendazim (600 g ai/ha) or had no significant difference with that of phenamacril (375 g ai/ha) in 2 years. Scanning electron microscope and transmission electron microscope observations revealed that after treating F. graminearum mycelia with frenolicin B, the mycelia appeared aberrant and had an uneven thickness and swelling, the cytoplasm had disintegrated, and some cell contents were lost. Transcriptome analysis suggests that frenolicin B might inhibit the metabolism of nucleotides and energy by affecting genes involved in phosphorus utilization but did not affect the expression of myosin 5, which is the specific target of phenamacril. These findings indicate that frenolicin B may be a potential agrochemical fungicide for controlling Fusarium head blight.
Antifungal Activity of Chitosan Nanoparticles Encapsulated With Cymbopogon martinii Essential Oil on Plant Pathogenic Fungi Fusarium graminearum

Abstract:Application of synthetic fungicides in agricultural commodities has been restricted due to development of fungicide resistance fungi and deleterious impact on environment and health of farm animals and humans. Hence, there is an urge for development of mycobiocides, and the present study was undertaken to determine the antifungal activity of Cymbopogon martinii essential oil (CMEO) on post-harvest pathogen Fusarium graminearum. The CMEO was extracted by hydrodistillation and GC-MS chemical profile revealed the presence of 46 compounds and abundant was geraniol (19.06%). The minimum inhibitory concentration and minimum fungicidal concentration of CMEO were determined as 421.7 ± 27.14 and 618.3 ± 79.35 ppm, respectively. The scanning electron microscopic observation of CMEO exposed macroconidia was exhibited a detrimental morphology with vesicles, craters, protuberance, and rough surfaces related to control fungi. The CMEO induced the death of fungi through elevating intracellular reactive oxygen species and lipid peroxidation, and depleting ergosterol content. Regrettably, essential oils are highly volatile and become unstable and lose their biological features on exposure to light, heat, pH, moisture, and oxygen. To overcome these issues, chitosan encapsulated CMEO nanoparticles (Ce-CMEO-NPs) were prepared. The synthesized Ce-CMEO-NPs have spherical morphology with Zeta potential of 39.3-37.2 mV and their corresponding size was found in range of 455-480 nm. The Fourier transform infrared analysis confirmed that bio-active constituents of CMEO were well stabilized due to chitosan conjugation and successfully formed Ce-CMEO-NPs. The in vitro release assay observed that the release of CMEO is stabilized due to the complex formation with chitosan and thereby, increases the lifetime antifungal activity of CMEO by gradual release of antifungal constituents of Ce-CMEO-NPs. In conclusion, antifungal and antimycotoxin activities of CMEO and Ce-CMEO-NPs against F. graminearum were assessed in maize grains under laboratory conditions over a storage period of 28 days. Interestingly, Ce-CMEO-NPs were presented efficient and enhanced antifungal and antimycotoxin activities related to CMEO, and it could be due to perseverance of antifungal activity by controlled release of antifungal constituents from Ce-CMEO-NPs. The study concluded that Ce-CMEO-NPs could be highly appropriate as mycobiocides in safeguarding the agricultural commodities during storage period in agricultural and food industries.

A Novel Highly Antifungal Compound ZJS-178 Targeting Myosin I Inhibits the Endocytosis and Mycotoxin Biosynthesis of Fusarium Graminearum

Abstract:Chemical fungicides remain a primary tool for managing Fusarium head blight (FHB) due to the lack of resistant wheat cultivars. In this study, we synthesized 101 2-cyanoacrylate compounds and identified ZJS-178 as particularly effective against Fusarium graminearum (the causal agent of FHB), inhibiting both hyphal growth and conidial germination. Notably, ZJS-178 is specific to Fusarium species, with no impact on other fungi. It also shows no cross-resistance with other groups of fungicides, suggesting a unique mode of action. Mechanistic studies revealed that ZJS-178 targets myosin I in F. graminearum, inhibiting ATPase activity and disrupting endocytosis. Furthermore, ZJS-178 impairs toxisome formation, leading to reduced production of the mycotoxin deoxynivalenol (DON). These findings position ZJS-178 as a promising antifungal agent that could pave the way for developing a novel fungicide to manage FHB and other Fusarium-related diseases.

Abstract:Abstract The aim of these studies was to develop an effective formulation for microbial strains active in plant protection. Emulsification technique in oil was used to produce alginate microcapsules supplemented with organic additives to improve production process and stability of the preparations during storage. The release of microorganisms from the capsules in soil and their effectiveness in biocontrol of Fusarium wilt were evaluated. Three bacterial strains Burkholderia cepacia strain CAT5, Bacillus spp. strains PZ9 and SZ61, and fungus Trichoderma virens TRS106 were immobilized separately in calcium alginate supplemented with chitosan, peat powder, or skim milk. The productivity of microcapsules was enhanced by 60% when peat was added to the alginate matrix. Peat reduced also contamination of the capsules during storage, significantly enhancing their quality. By contrast, the addition of skim milk reduced quality of the microcapsules. The additives did not influence the viability of entrapped microorganisms and their release in soil. The survival of the microbial cells was mainly related to the kind of microorganism used, and the highest viability showed Bacillus sp. PZ9 and Trichoderma TRS106. Lyophilization of the microcapsules appeared to be unfavorable by reducing microbial viability in the capsules and in the soil after application. The best properties: good storage ability and sufficient microbial release in the soil, exhibited wet microcapsules amended with peat. These capsules were used to control Fusarium wilt in tomato plants. The protective effect was obtained when the microcapsule-entrapped bacteria PZ9 were used. The effectiveness of this bacterium was comparable with fungicide. Peat-amended microcapsules entrapping Bacillus PZ9 showed the best quality and may have potential for commercial use.
Integrative transcriptomic and metabolomic analyses reveals the toxicity and mechanistic insights of bioformulated chitosan nanoparticles against Magnaporthe oryzae

Abstract:Rice blast, an extremely destructive disease caused by the filamentous fungal pathogen Magnaporthe oryzae , poses a global threat to the production of rice ( Oryza sativa L.). The emerging trend of reducing dependence on chemical fungicides for crop protection has increased interest in exploring bioformulated nanomaterials as a sustainable alternative antimicrobial strategy for effectively managing plant diseases. Herein, we used physiomorphological, transcriptomic, and metabolomic methods to investigate the toxicity and molecular action mechanisms of moringa–chitosan nanoparticles (M–CNPs) against M. oryzae . Our results demonstrate that M–CNPs exhibit direct antifungal properties by impeding the growth and conidia formation of M. oryzae in a concentration-dependent manner. Propidium iodide staining indicated concentration-dependent significant apoptosis (91.33%) in the fungus. Ultrastructural observations revealed complete structural damage in fungal cells treated with 200 mg/L M–CNPs, including disruption of the cell wall and destruction of internal organelles. Transcriptomic and metabolomic analyses revealed the intricate mechanism underlying the toxicity of M–CNPs against M. oryzae . The transcriptomics data indicated that exposure to M–CNPs disrupted various processes integral to cell membrane biosynthesis, aflatoxin biosynthesis, transcriptional regulation, and nuclear integrity in M. oryzae. , emphasizing the interaction between M–CNPs and fungal cells. Similarly, metabolomic profiling demonstrated that exposure to M–CNPs significantly altered the levels of several key metabolites involved in the integral components of metabolic pathways, microbial metabolism, histidine metabolism, citrate cycle, and lipid and protein metabolism in M. oryzae . Overall, these findings demonstrated the potent antifungal action of M–CNPs, with a remarkable impact at the physiological and molecular level, culminating in substantial apoptotic-like fungal cell death. This research provides a novel perspective on investigating bioformulated nanomaterials as antifungal agents for plant disease control.

Abstract:In this study, the natural fungicide natamycin (N) was encapsulated in zein/carboxymethyl chitosan core-shell nanoparticles (Z/CMCS NPs) to improve its aqueous dispersibility and photostability for application in postharvest fruit preservations. The N-Z/CMCS NPs were spherically shaped with a particle size of 223 ± 2 nm, a polydispersity index of 0.07 ± 0.01 and a surface charge of −50.7 ± 2.3 mV. The NPs were characterized by FTIR spectroscopy and scanning electron microscopy. The encapsulation efficiency of natamycin was improved from 68.3 ± 2.4% to 78.4 ± 1.7% by the addition of CMCS to the NPs. The optimized N-Z/CMCS NPs remained stable and were resistant to neutral pH (6.0–8.0), ion strength (0–100 mM) and long-term storage (30 days). The degradation of natamycin under UV light was reduced, with a half-life time (t1/2) of 47.2 min in the pure state to a t1/2 of 147.5 min in the N-Z/CMCS NPs. The stabilized N-Z/CMCS NPs retained the property of redispersibility after freeze-drying. The bacteriostatic properties of N-Z/CMCS NPs against Botrytis cinerea were tested in vitro and in vivo. The in vitro results demonstrated that the application of N-Z/CMCS NPs completely inhibited the spore germination rate, and 10 mg/L N-Z/CMCS NPs inhibited the mycelial growth by 64.4%. The in vivo results indicated that the NPs not only reduced the occurrence of rot and mildew in inoculated strawberries, with the lowest decay index of 30.3% after 3 days storage, but also inhibited the occurrence of gray mold disease in postharvest strawberries, with the lowest decay rate of 31.1% after 8 days of storage. This study will be beneficial for the development of colloidal delivery systems for postharvest fruit industries.

Abstract:Plant diseases prompted by fungi and bacteria are one of the most serious threats to global crop production and food security. The destruction of these infections posed a major challenge to plant protection by chemical control. Herein, we develop CMCS/PA/Zn 2+ nanoparticles (NPs) using carboxymethyl chitosan (CMCS), phytic acid (PA) and metal ions (Zn 2+ ) via flash nanoprecipitation (FNP) strategy. Metal complexes of PA with specified antibacterial and antifungal activities are expected to hold the potential and play a significant role in antimicrobial treatment. The size and size distribution of NPs was confirmed through Dynamic and Static Light Scatterer (DSLS). In acidic-infection microenvironment, the CMCS/PA/Zn 2+ NPs can disintegrate and release Zn 2+ in situ thus stimulated the corresponding antimicrobial activity. These CMCS/PA/Zn 2+ NPs showed outstanding antibacterial efficacy (98 %) against S. aureus and E. coli bacteria in vitro, as well as an impressive antifungal efficacy of 98 % and 81 % against R. solani and B. cinerea at 50 μg/mL respectively. This study contributes a prospective idea to the development of organic-inorganic hybrid NPs as environmentally-friendly and safe agricultural antimicrobials.

Abstract:Synthetic fungicides have been widely used to protect crops from fungal diseases. However, excessive use of synthetic fungicides leads to the generation of fungicide resistance in fungal pathogens. Recently, smart cargo delivery systems have been introduced for the construction of a pesticide delivery nanoplatform, benefiting from their controlled release performance. Herein, a fungal pathogen microenvironment-responsive supramolecular fungicide nanoplatform has been designed and constructed, using quaternary ammonium salt (Q)-modified mesoporous silica nanoparticles (MSN-Q NPs) as nanocarriers loaded with berberine hydrochloride (BH) and carboxylatopillar[5]arene (CP[5]A) as nanogates to form BH-loaded CP[5] NPs for effective inhibition of Botrytis cinerea. CP[5]A as nanogates can endow the fungicide nanoplatform with pH stimuli-responsive release features for the control of fungicide release. The loaded BH, as a natural plant fungicide, provides an ecofriendly alternative to synthetic fungicides for controlling B. cinerea. Interestingly, we use oxalic acid (OA) secreted by B. cinerea as a trigger so that BH can be released from the fungicide nanoplatform on demand under pathogen microenvironments for controlling B. cinerea.
