Abstract:Dental caries have become a major global public health problem. Plaque control and remineralization of initial enamel lesions are paramount for the prevention and control of caries. Polyhexamethylene biguanide (PHMB) is a type of cationic amphipathic antibacterial agent with broad-spectrum antibacterial properties and good biological safety. Fluoride delays demineralization and promotes the remineralization of hard dental tissues. However, a high concentration is needed for it to function as an antibacterial agent. In order to create a PHMB with the benefits associated with fluoride, we synthesized a fluorine-containing cationic polymer, PHMB-F. Fourier transform-infrared spectroscopy and solid state nuclear magnetic resonance characterization confirmed the successful synthesis of PHMB-F. Antibacterial tests showed that PHMB-F had better antiseptic efficacy for Streptococcus mutans compared with just PHMB. Moreover, positively-charged PHMB-F allows fluoride ions to exist closer to the enamel surface with negative potential, which markedly lowers the ion concentrations in the microenvironment adjacent to hard dental tissues needed to maintain equilibrium. Thus, only low concentrations of PHMB-F are required for enamel remineralization.

Abstract:Durability problem of dentin bonding originated from defective bonding interfaces caused by inadequate infil-tration of adhesives into demineralized dentin matrix (DDM) is one of the biggest chanllenges in clinical dentistry, while the scientific mechanism remains unclear. We propose that due to poor physical compatibility, the DDM hydrogel-like interface formed by phosphorylated non-collagenous-proteins can be the primary cause of inadequate infiltration of adhesive containing high-content hydrophobic marcromonomers and the formation of numerous defects in the DDM-cured adhesive interface phase. To test this assumption, we develop a novel bonding strategy based on the hydrogel-like interface dehydrogelation refactoring via actively inducing in situ formation of numerous amorphous calcium phosphate nanoparticles within 30 s. The easy-to-collapse DDM is remolded into a robust porous hybrid structure of low polarity. Irrespective of using a wet or dry DDM, the improved physical compatibility between the DDM interface and adhesives greatly promotes infiltration of various adhesives and the integration of the DDM-cured adhesive interface phase. The resulting defect-low bonding interface greatly improves bonding strength and stability indicating the central role of this long -neglected hydrogel-like DDM interface in the problems associated with dentin bonding, which enriches the theory of adhesive dentistry. Free from hard-to-get advanced adhesive materials and sophisticated operations, accessibility and homogeneity of high-quality dentin bonding may benefit clinical dentistry.
A polyhexamethylene biguanide-assembly assisted strategy of dentin bonding greatly promotes bonding effects and caries treatment

Abstract:Structural degeneration of a hybrid layer composed of a demineralized dentin matrix (DDM) and adhesive causes unsatisfactory functional outcomes in terms of bonding repair and caries treatment and is accompanied by high prevalence of secondary caries. Clinically, defects in the hybrid layer from insufficient adhesive infiltration, bacterial load from retained infected-dentin, and bacterial attack from the oral cavity are the main threats to degeneration. Currently, there is no strategy to simultaneously address adhesive penetration and bacterial infection. Herein, based on the core role of the strongly-polar hydrated DDM interface in dentin bonding, an interface-reconstructed bonding strategy assisted by electrostatic assembly of broad-spectrum germicidal polyhexamethylene biguanide (PHMB) is proposed that kills two birds with one stone. PHMB is absorbed onto the anionic 3D DDM forming a PHMB/DDM complex. The surface potential of the DDM increases by about 100 mV, the anion content decreases by 20%, and the interface water content decreases by nearly 40%. All of these changes contribute to the penetration of the adhesive, thereby improving the bonding strength and durability. After thermal cycling aging, the bonding strength of the PHMB group was 1.45-1.65 times that of the control group. In terms of antibacterial properties, PHMB treatment not only has a bacterial-killing ability due to the already formed biofilm but also significantly reduces the adhesion of bacteria, thereby delaying the occurrence of secondary caries. In summary, PHMB treatment reconstructed the DDM interface, resulting in a defect-low and inherent antibacterial hybrid layer that improves the bonding effect, treatment of caries and even prevention of secondary caries. By a one-step 30s treatment of polyhexamethylene biguanide (PHMB), residual bacteria in remaining caries-affected dentin are killed, and an intrinsic antibacterial defect-low hybrid layer is constructed, promoting bonding effect and caries treatment.

Abstract:This study presents a biomimetic method for remineralizing dental hard tissues using an amyloid‐like protein matrix composite with amorphous calcium phosphate. This approach mimics natural mineralization processes, enhancing the regeneration and mechanical strength of demineralized enamel and dentine, providing a promising material for the treatment of early enamel lesions and dentine hypersensitivity. Numerous remineralizing coatings aim to prevent or treat early enamel lesions and occlude exposed dentinal tubules (DTs). Nevertheless, the pace of remineralization is inadequate, and the mechanical robustness of the newly established mineral layer fails to match the inherent strength. In this study, a biomimetic mineralization strategy aimed at replicating key events in biological mineralization, specifically focusing on the organic–inorganic composite matrix, is proposed. The material utilizes Tris(2‐carboxyethyl)phosphine (TCEP), which serves a dual role: stabilized amorphous calcium phosphate (ACP) (ACP@TCEP) nanoparticles as its inorganic component, and catalyzing the cleavage of intramolecular disulfide bonds in poly(ethylene glycol) (PEG) grafted lysozyme (lyso‐PEG) to facilitate the formation of an amyloid‐like protein matrix composite with ACP (ACP@lyso‐PEG nanocomplexes). ACP@lyso‐PEG nanocomplexes can rapidly and efficiently form an enamel‐like remineralization layer on the surface of damaged dental hard tissue, reaching ≈4.205 μm thickness after 3 days of acid‐etched enamel. Furthermore, achieving a depth of DTs occlusion exceeding 60 μm after 5 days, using a simple immersion process. The resulting mineralized layer exhibits mechanical strength comparable to natural teeth. This study introduces a conceptual biomimetic mineralization strategy for effective enamel repair or DTs occlusion in clinical practices, and offers potential insights into the mechanisms of biomineral formation.

Abstract:Dentin bonding is the most common form of human tissue repair among tissue-biomaterial adhesions, concerning billions of people's oral health worldwide. However, insufficient adhesive infiltration in the demineralized dentin matrix (DDM) always produces numerous defects in the bonding interface termed the hybrid layer, which causes high levels of bacteria-related secondary dental diseases, and less than 50% of the bonding lasts more than 5 years. Therefore, it is urgent and vital to construct an antibacterial low-defect hybrid layer to solve the durability-related problems. A DDM with a hydrogel-like surface formed by the hydration of highly-anionic non-collagenous proteins (NCPs) is firstly used as a template to electrostatically assemble polyethyleneimine (PEI). The formation of a stable antibacterial polyelectrolyte complex of PEI/NCPs rapidly eliminates NCP hydration capacity and significantly improves the infiltration of various adhesives. Simultaneously, both the PEI during the assembly and the PEI-assembled DDM can directly destroy a biofilm of S. Mutans on the DDM. Consequently, a long-term antibacterial and low-defect hybrid layer is successfully created, which greatly improves the bonding effectiveness. This helps to improve the clinical treatment of bacteria-based dental diseases and the tooth-restoration repair effect and prevent secondary dental diseases, having significance in clinical dentistry and providing insights for other tissue-biomaterial adhesions.

Abstract:Dentin hypersensitivity is attributable to the exposed dentin and its patent tubules. We proposed the therapeutic management of demineralized dentin surfaces using a mineralizing adhesive to seal and remineralize dentin, dentinal tubules, and odontoblast processes. An experimental self-etch adhesive and a mineralizing adhesive consisting of the self-etch adhesive and 20 wt % poly-aspartic acid-stabilized amorphous calcium phosphate (PAsp-ACP) nanoparticles were prepared and characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), and scanning electron microscopy. After 60 acid-etched midcoronal dentin disks were treated with distilled water (control), a desensitizing agent (Gluma), the experimental self-etch adhesive, and the mineralizing adhesive, dentin permeability was measured and mineralization was evaluated by Raman, FTIR, XRD, TEM, and selected-area electron diffraction, irrespective of abrasive and acidic challenges. In vitro cytotoxicity of the adhesive and the mineralizing adhesive was assessed by Cell Counting Kit-8. The mineralizing adhesive possessed excellent biocompatibility. We proposed a hybrid mineralization layer composed of the light-cured mineralizing adhesive and the mineralized dentin surfaces, as well as interiorly mineralized resin tags and odontoblast processes inside of the dentinal tubules. This hybrid mineralization not only reduced dentin permeability but also resisted abrasive and acidic attacks.
Poly (amido amine) dendrimer and dental adhesive with calcium phosphate nanoparticles remineralized dentin in lactic acid

Abstract:Patients with dry mouth often have an acidic oral environment lacking saliva to provide calcium (Ca) and phosphate (P) ions. There has been no report on tooth remineralization in acidic pH4 and CaP ion-lacking solutions. The objective of this study was to develop a novel method of combining poly(amido amine) (PAMAM) with adhesive containing nanoparticles of amorphous calcium phosphate (NACP) for dentin remineralization in pH4 and CaP-lacking solution for the first time. Demineralized dentin was tested in four groups: (1) dentin control, (2) dentin with PAMAM, (3) dentin with NACP adhesive, (4) dentin with PAMAM + NACP adhesive. Dentin samples were examined by scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS) and hardness testing. Increasing the NACP filler level in adhesive from 0 to 40 wt% did not negatively affect the dentin bond strength (p > 0.1). NACP adhesive released CaP ions and neutralized the acid. PAMAM alone failed to achieve dentin remineralization in lactic acid. NACP alone induced slight dentin remineralization in lactic acid (p > 0.1). In contrast, the novel PAMAM + NACP group in the pH4 and CaP-lacking solution completely remineralized the predemineralized dentin, increasing its hardness which approached that of healthy dentin (p > 0.1). In conclusion, dentin remineralization via PAMAM + NACP adhesive in pH4 and CaP-lacking acid was achieved for the first time, when conventional remineralization methods such as PAMAM or NACP did not work. The novel PAMAM + NACP method is promising to increase the longevity of the composite-tooth bond, inhibit caries, remineralize lesions and protect tooth structures, even for patients with dry mouth and an acidic oral environment. © 2017 Wiley Periodicals, Inc. Development of Novel Polymerizable Antibacterial Monomer and Its Antibacterial Effects on Dental Adhesive

Abstract:The object of the present study was to Functionalize adhesive system with antibacterial activity without interfering the bonding effect, to kill bacteria residue on cavity and prevent bacteria from colonization on the margins of bonding. The fluorine -containing imidazolium salt antibacterial monomer was synthesized by the combination reaction of 1-chlorododecane and 1-vinylimidazole, and the replacement reaction to introduce fluoride ion. The synthesized intermediate product and final product were characterized by nuclear magnetic resonance spectroscopy. Fluoride ion was introduced into the bonding system in the form of organic fluorine, and the release behavior of fluoride ion from the functionalized adhesive was studied. The antibacterial experiment showed that the functionalized adhesive was endowed with good antibacterial property, which realized targeted anti -caries ability. The bond strength test indicated that the functionalization with the fluorine -containing imidazolium salt monomer did not affect the bonding performance of the adhesive. These results suggested that adhesive functionalized with the fluorine -containing imidazolium salt monomer had potential for reducing the occurrence of clinical secondary caries.
Biomineralization-inspired sandwich dentin desensitization strategy based on multifunctional nanocomposite with yolk–shell structure

Abstract:Dentin hypersensitivity (DH) treatment is far from being unequivocal in providing a superior strategy that combines immediate and long-term efficiency of dentinal tubule (DT) occlusion and clinical applicability. In order to achieve this aim, a type of multifunctional yolk–shell nanocomposite with acid resistance, mechanical resistance and biomineralization properties was developed in this study, which consists of a silica/mesoporous titanium–zirconium nanocarrier (STZ) and poly(allylamine hydrochloride) (PAH)-stabilized amorphous calcium phosphate (ACP) liquid precursor. First, the nanocomposite, named as PSTZ, immediately occluded DTs and demonstrated outstanding acid and mechanical resistance. Second, the PSTZ nanocomposite induced intrafibrillar mineralization of single-layer collagen fibrils and remineralization of demineralized dentin matrix. Finally, PSTZ promoted the odontogenic differentiation of dental pulp stem cells by releasing ACP and silicon ions. The reconstruction of the dentin-mimicking hierarchical structure and the introduction of newly formed minerals in the upper, middle and lower segments of DTs, defined as sandwich-like structures, markedly reduced the permeability and achieved superior long-term sealing effects. The nanocomposite material based on mesoporous yolk–shell carriers and liquid-phase mineralized precursors developed in this study represents a versatile biomimetic sandwich desensitization strategy and offers fresh insight into the clinical management of DH.

Abstract:Hepatitis B virus (HBV) that causes Hepatitis B with a high chronic rate, could lead to hepatic cirrhosis and hepatocellular carcinoma. The IL28B gene belongs to a new interferon family λ and its genetic polymorphisms are identified to be associated with treatment effect and viral clearance. The purpose of this review is to discuss the role of the IL28B gene in treatment response and viral clearance of HBV-infected individuals, and further to reveal the mechanisms. This review could provide a theoretical basis for personalized medicines of HBV patients.

Abstract:Journal of Dental Research, Ahead of Print. Dental caries is a dynamic disease induced by the unbalance between demineralization of dental hard tissues caused by biofilm and remineralization of them; however, although various effective remineralization methods have been well documented, it is a challenge to reestablish the balance by enhancing remineralization alone while ignoring the antibacterial therapy. Therefore, the integration of remineralizing and antibacterial technologies offers a promising strategy to halt natural caries progression in clinical practice. Here, the conception of interrupting dental caries (IDC) was proposed based on the development of dual-functional coating with remineralizing and antibacterial properties. In this study, bovine serum albumin (BSA) loaded octenidine (OCT) successfully to form a BSA-OCT composite. Subsequently, through fast amyloid-like aggregation, the phase-transited BSA-OCT (PTB-OCT) coating can be covered on teeth, resin composite, or sealant surfaces in 30 min by a simple smearing process. The PTB-OCT coating showed satisfactory effects in promoting the remineralization of demineralized enamel and dentin in vitro. Moreover, this coating also exerted significant acid-resistance stability and anti-biofilm properties. Equally importantly, this coating exhibited promising abilities in reducing the microleakage between the tooth and resin composite in vitro and preventing primary and secondary caries in vivo. In conclusion, this novel dual-functional PTB-OCT coating could reestablish the balance between demineralization and remineralization in the process of caries, thereby potentially preventing or arresting caries.

Abstract:Highlights • An EGCG/PAH-ACP co-delivery mesoporous nanosystem is developed for managing exposed dentin. • The nanosystem effectively occludes dentinal tubules and inhibits biofilm formation of S. mutans. • The nanosystem induces intrafibrillar mineralization of collagen and remineralization of demineralized dentin. • This strategy is promising in protecting exposed dentin against caries and hypersensitivity. As a global public health focus, oral health plays a vital role in facilitating overall health. Defected teeth characterized by exposure of dentin generally increase the risk of aggravating oral diseases. The exposed dentinal tubules provide channels for irritants and bacterial invasion, leading to dentin hypersensitivity and even pulp inflammation. Cariogenic bacterial adhesion and biofilm formation on dentin are responsible for tooth demineralization and caries. It remains a clinical challenge to achieve the integration of tubule occlusion, collagen mineralization, and antibiofilm functions for managing exposed dentin. To address this issue, an epigallocatechin-3-gallate (EGCG) and poly(allylamine)-stabilized amorphous calcium phosphate (PAH-ACP) co-delivery hollow mesoporous silica (HMS) nanosystem (E/) was herein developed. The application of E/ effectively occluded the dentinal tubules with acid- and abrasion-resistant stability and inhibited the biofilm formation of Streptococcus mutans . Intrafibrillar mineralization of collagen fibrils and remineralization of demineralized dentin were induced by E/ The odontogenic differentiation and mineralization of dental pulp cells with high biocompatibility were also promoted. Animal experiments showed that E/ durably sealed the tubules and inhibited biofilm growth up to 14 days. Thus, the development of the E/ nanosystem provides promising benefits for protecting exposed dentin through the coordinated manipulation of dentin caries and hypersensitivity. A novel universal adhesive for improved dentin remineralization with antibiofilm potential against Streptococcus mutans and other cariogenic-pathogens

Abstract:Dental demineralization is a widespread problem and the main responsible for dental caries and dentin hypersensitivity. Dental caries is a biofilm-driven bacterial infection that causes mineral depletion in enamel and is associated with an increased prevalence of medical rehabilitative polymeric implants. Since existing tissue-engineering methods are incapable of manufacturing synthetic dentin adhesive, modern therapeutic interventions address the remineralization of caries-infected dentin to prevent its inevitable removal. In recent years, magnetic nanoparticle-coated adhesives were known to deliver drugs to the pulp and get better adhesive penetration into dentin. In the present study, we prepared and assessed the effectiveness of antimicrobial peptide (cecropin A) coated Fe 3 O 4 -nanoparticle on dentin remineralization, bond strength, and its antibiofilm potential against Streptococcus mutans , is associated with dental caries in humans. Prior to the experiment, the synthesized 3 O 4 nanoparticle was characterized and assessed its antibiofilm potential against S. mutans . The transcriptomic approach with qPCR analysis stated that 3 O 4 nanoparticle significantly downregulated the expression of biofilm-regulated genes such as GtfB, GtfC, GtfD, VicR, ComDE , and Smu0630 in S. mutans , which validates the outcome of in vitro results. Interestingly, the prepared 3 O 4 -nanoparticle coated adhesive effectively increased the dentin remineralization and bond strength. Fascinatingly, 3 O 4 adhesive significantly hindered the biofilm formation of S. mutans on the dentin surface. Notably, the 3 O 4 adhesive accomplished reduced biofilm CFU count, lactic acid production, and biofilm metabolic activity. In addition, the 3 O 4 adhesive also reduced the biofilm formation of other caries-associated bacterial pathogens, including S. sobrinus, S. sanguinis, and S. mitis . Overall, the current research found that the 3 O 4 adhesive was more effective in remineralizing dentin lesions with antibiofilm potential in order to avoid Streptococcus -derived dental caries.
Salivary Polypeptide/hyaluronic Acid Multilayer Coatings Act As “fungal Repellents” and Prevent Biofilm Formation on Biomaterials

Abstract:Candida-associated denture stomatitis (CADS) is a common, recurring clinical complication in denture wearers that can lead to serious oral and systemic health problems. Current management strategies are not satisfactory due to their short-acting and ineffective therapeutic effects. Here, we describe a new fungal biofilm controlling strategy using the polyelectrolyte layer-by-layer (LBL) self-assembly technology on denture materials. Conventional poly(methyl methacrylate) (PMMA) denture material discs were functionalized with negatively charged poly(methacrylic acid) (PMAA) via plasma-initiated surface grafting, followed by repetitive alternating coating with the salivary antimicrobial polypeptide histatin 5 (H-5; cationic polymer) and hyaluronic acid (HA; anionic polymer). On the other hand, the H-5/HA LBL coatings (i.e., the outermost layer was H-5) inhibited fungal attachment/adhesion, significantly reduced fungal biofilm formation, and showed synergistic effects with the antifungal drug miconazole. LBL surface hydrophilicity was not the key mechanism in controlling Candida biofilm formation. The current approach demonstrates the utility of a new design principle for fabricating anticandidal denture materials, as well as potentially other related medical devices, for controlling fungal biofilm formation and combating insidious infections.
Use of multifunctional phosphorylated PAMAM dendrimers for dentin biomimetic remineralization and dentinal tubule occlusion

Abstract:The disequilibrium between demineralization and remineralization of teeth, especially of dentin, may lead to serious consequences like dental caries that are considered to affect people's quality of life. The employment of biomimetic analogs of proteins to duplicate biomineralization, which is a well-regulated process mediated by extracellular matrix proteins, may provide new insights to solve these problems. Here we report the use of a modified multifunctional dendrimer, synthesized by the introduction of phosphate groups via a Mannich-type reaction onto poly(amidoamine) (PAMAM) dendrimers, to biomimetically remineralize dentin. The phosphorylated PAMAM dendrimers were demonstrated to act, along with an amorphous calcium phosphate stabilizing agent, polyacrylic acid (PAA), as biomimetic analogs of noncollagenous proteins to induce the remineralization of demineralized dentin. Phosphorylated PAMAM dendrimers treated demineralized dentin discs were immersed in a remineralizing solution containing PAA for up to 7 days. The success of this remineralization was examined using attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR), X-ray diffraction (XRD), energy-dispersive X-ray spectroscopy (EDS) and electron microscopy. These results showed that demineralized dentinal collagen fibrils were successfully phosphorylated by the treatment of phosphorylated PAMAM dendrimers and embedded with calcium-deficient hydroxyapatite after remineralization. The surfaces of demineralized dentin discs were covered with newly induced crystals and the patent dentinal tubules were occluded. A good biocompatibility was also determined. Thus, phosphorylated PAMAM dendrimers could be applied as a minimally invasive method of management of dentin caries, employed to improve the resin-dentin bonding stability and also be used in the treatment of dentin hypersensitivity.

Abstract:ObjectivesThe objectives of this study were to: (1) develop a multifunctional adhesive via dimethylaminohexadecyl methacrylate (DMAHDM) and nanoparticles of amorphous calcium phosphate (NACP); and (2) investigate its ability to provide metalloproteinases (MMPs) deactivation and remineralization for long-term dentin bonding durability.MethodsDMAHDM and NACP were incorporated into AdperTM Single Bond 2 Adhesive (SB2) at mass fractions of 5% and 20%, respectively. Degree of conversion and contact angle were measured. Endogenous MMP activity of the demineralized dentin beams, Masson’s trichrome staining, nano-indentation, microtensile bond strength and interfacial nanoleakage analyses were investigated after 24 h and 3 months of storage aging in artificial saliva.ResultsAdding DMAHDM and NACP did not compromise the degree of conversion and contact angle of SB2 (p > 0.05). DMAHDM and NACP incorporation reduced the endogenous MMP activity by 53 %, facilitated remineralization, and increased the Young’s modulus of hybrid layer by 49 % after 3 months of aging in artificial saliva, compared to control. For SB2 Control, the dentin bond strength decreased by 38 %, with greater nanoleakage expression, after 3 months of aging (p 0.05).SignificanceDMAHDM+NACP adhesive greatly reduced MMP-degradation activity in demineralized dentin, induced remineralization at adhesive-dentin interface, and maintained the dentin bond strength after aging, without adversely affecting polymerization and dentin wettability. This new adhesive has great potential to help eliminate secondary caries, prevent hybrid layer degradation, and increase the resin-dentin bond longevity.

Disaggregated Nano-Hydroxyapatite (DnHAP) with Inhibitory Effects on Biofilms and Demineralization

Abstract:Nano-hydroxyapatite (nHAP) is considered a biocompatible agent that promotes the remineralization of dental hard tissue; however, its antibacterial efficacy is under scientific discussion. Therefore, this investigation aimed to specify the inhibitory effects of disaggregated nano-hydroxyapatite (DnHAP) on regrown biofilms and demineralization. Regrown biofilm models of single-species (Streptococcus mutans), dual-species (S. mutans and Candida albicans), and saliva-derived microcosm biofilms were established in vitro. Repeat treatment with DnHAP was applied to biofilms. The viability, lactic acid, biofilm structure, biomass, the inhibitory effect of demineralization, and virulence factors' expression were determined. In addition, the biofilm microbial community was analyzed by 16S ribosomal RNA gene sequencing. DnHAP inhibited metabolism, lactic acid production, biomass, and water-insoluble polysaccharide production (P < 0.05) of regrown single/dual-species biofilms. Concerning the saliva-derived biofilms, samples treated with DnHAP showed lower biofilm metabolic activity without significant differences from samples treated with sterile deionized water (P > 0.05); in addition, saliva-derived biofilms treated with DnHAP exhibited lower lactic acid production (P < 0.05). The demineralization of bovine enamel was the lowest in the DnHAP group, as detected by transverse microradiography, and the lesion depth and volume decreased significantly (P < 0.05). The application of DnHAP did not change the diversity of regrown saliva-derived microcosm biofilms. In conclusion, this investigation showed that DnHAP could be a promising solution for the management of regrown biofilms to combat dental caries.
Targeted antimicrobial self-assembly peptide hydrogel with in situ bio-mimic remineralization for caries management

Abstract:Highlights • 8DSS-C8-P-113 is composed of a non-specific antimicrobial peptide P-113, a competence stimulating peptide C8, and an enhancing remineralization domain 8DSS. • 8DSS-C8-P-113 eliminates cariogenic bacteria without disrupting oral normal flora. • 8DSS-C8-P-113 forms a hydrogel at 16 μM mL −1 . • 8DSS-C8-P-113 hydrogel adheres on the tooth surface protecting tooth against bacterial invasion. • 8DSS-C8-P-113 hydrogel promotes in situ bio-mimic remineralization of tooth lesions. The single-function agents with wide-spectrum activity which tend to disturb the ecological balance of oral cavity cannot satisfy dental treatment need. A multi-functional agent with specifically targeted killing property and in situ remineralization is warranted for caries management. A novel multi-functional agent (8DSS-C8-P-113) consisting of three domains, i.e., a non-specific antimicrobial peptide (AMP) (P-113), a competence stimulating peptide (C8), and an enhancing remineralization domain (8DSS), is fabricated and evaluated in this study. The findings demonstrates that 2 μM mL −1 of 8DSS-C8-P-113 eliminates planktonic Streptococcus mutans ( S. mutans ) without disrupting the oral normal flora. At a concentration of 8 μM mL −1 , it exhibits the ability to prevent S. mut ans' adhesion. Furthermore, 8DSS-C8-P-113 self-assembles a hydrogel state at the higher concentration of 16 μM mL −1 . This hydrogel self-adheres on the tooth surface, resisting acid attack, eradicating S. mutans' biofilm, and inducing mineralization in order to facilitate the repair of demineralized dental hard tissue. Its significant effectiveness in reducing the severity of dental caries is also demonstrated in vivo in a rat model. This study suggests that the multi-functional bioactive AMP 8DSS-C8-P-113 is a promising agent to specifically target pathogen, prevent tooth demineralization, and effectively induce in situ bio-mimic remineralization for the management of dental caries.
