Abstract:This study aimed to evaluate the gelation characteristics of soybean protein hydrolysate (SPH) extracted by enzyme-assisted aqueous extraction. Specifically, the changes in gelation behaviors for heat-induced (95°C, 20 min) SPH dispersions treated with pH (pH 3, 5, 9; pH 7 as control) and ultrasound (U; 240 W, 30 min) were investigated. The results showed that typical gel behavior with high elastic nature in the viscoelasticity and network structures were observed during the heating process, where the disulfide bond played a dominant role in the gel network formation of all the samples. Notably, the heat-induced aggregation in the SPH gels was mainly formed by the association of the basic B polypeptide in 11S and β subunit in 7S. The most superior SPH gel was formed at pH 7 when assisted by ultrasonication during the heating process. This as-synthesized gel showed a uniform filamentous structure and exhibited the more excellent textural, rheological and thermal properties than those of the samples formed under acidic and alkaline conditions. These results are of great value in revealing the gelation mechanism of SPH.

Abstract:Hydroxyl radical-induced oxidation can modify gelling properties of food proteins. In this study, a hydroxyl radical generating system (HRGS), consisting of 0.1 mM Fe 3+ , 0.1 mM ascorbic acid, and 1, 10, or 20 mM H 2 O 2 , was used to oxidize ginkgo seed protein isolate (GSPI) for 4 h at ambient temperature in the presence of 0.3 % (w/v) hyaluronic acid (HA) to enhance its gelation properties. HRGS treatment led to increased protein hydrophobicity, reduced sulfhydryl content, and disulfide bond-mediated protein crosslinking. Moreover, the secondary structure of GSPI varied with H 2 O 2 concentrations. Moderate oxidation (approximately 10 mM H 2 O 2 ) promoted GSPI aggregation and improved mechanical strength, rheological properties, water holding capacity, and whiteness of GSPI gels. However, excessive oxidation disrupted hydrogen bonding, generated excessive disulfide bonds, hindered active group interaction, inhibited gel network formation, and reduced gel strength. Hence, hydroxyl radical-induced oxidation holds potential for enhancing GSPI gelation within specific concentration ranges. This study suggests that controlled oxidation could be a novel approach for developing protein-based gel products.

Modulation of acid-induced pea protein gels by gellan gum and glucono-δ-lactone: Rheological and microstructural insights

Abstract:This study investigated the effect of gellan gum (GG) and glucono-δ-lactone (GDL) on the acid-induced gel properties of pea protein isolate (PPI) pretreated with media milling. The inclusion of GG substantially enhanced the gel hardness of PPI gel from 18.69 g to 792.47 g though slightly reduced its water holding capacity (WHC). Rheological analysis showed that GG increased storage modulus (G') and decreased damping factor of gels in the small amplitude oscillatory shear region and transformed its strain thinning behavior into weak strain overshoot behavior in the large amplitude oscillatory shear region. SEM revealed that GG transformed the microstructure of gel from a uniform particle aggregate structure to a chain-like architecture composed of filaments with small protein particles attached. Turbidity and zeta potential analysis showed that GG promoted the transformation of PPI from a soluble polymer system to an insoluble coagulant during acidification. When GG content was relatively high (0.2 %-0.3 %), high GDL content increased the electrostatic interaction between PPI and GG molecules, causing their rapid aggregation into a dense irregular aggregate structure, further enhancing gel strength and WHC. Overall, GG and GDL can offer the opportunity to modulate the microstructure and gel properties of acid-induced PPI gels, presenting potential for diversifying food gel design strategies through PPI-GG hybrid systems.

Abstract:The effect of the interaction between myofibrillar protein (MP) and heat-induced soy protein isolates (SPI) on gel properties was examined. To enhance the interaction between MP and SPI, SPI was subjected to thermal treatments at 60, 80, and 95 °C. The results showed that hydrophobic interactions played the most important role in MP-heated SPI (HSPI) gels. Hydrogen bonds played an important role in stabilizing the mixed gels, but this decreased with increasing heat treatment temperature of SPI. Disulfide bonds were not a significant force stabilizing the mixed gels. The gel properties of HSPI-MP were enhanced significantly (P < 0.05) by the inclusion of preheated SPI (95 °C, 30 min), including hardness, springiness, and water-holding capacity (WHC). Dynamic rheological analysis showed that heat treatment decreased the onset temperature of mixed gels and significantly increased the final G’. Notable cross-linked strands formed in MP-HSPI (80 °C) and MP-HSPI (95 °C) gels, while the smoothest and most ordered gel network structure was observed in the MP-HSPI (95 °C) gel.

Abstract:This study investigated the influence of acids on gelation of soy protein isolate (SPI) under heating. Specifically, the SPI solution was acidified to pH 2.5 using hydrochloric acid (HA), acetic acid (AA), tartaric acid (TA) and citric acid (CA), respectively, and then heated at 85 °C. It was found that the SPI gel with the anisotropic structure was formed, which suggested that the gel resulted from the organized arrangement of the SPI fibril. Subsequently, it was confirmed by several techniques such as atomic force microscopy that the above acidic-thermal treatment induced fibrillation of SPI. Furthermore, the ability of these acids to promote gelation and fibrillation of SPI was in the order of CA > TA > HA > AA. In summary, these results suggested that heating at pH 2.5 resulted in SPI fibrillation, thus promoting SPI gelation, and the acidity regulator type had a profound effect.

Abstract:The gelation of walnut protein isolates has not been extensively studied, mainly due to their inherent poor dispersity. This study investigated the gelation of alkaline-extracted walnut protein isolates (AWPI) and phosphorylated walnut protein isolates (PWPI) induced by heat treatment with glucono-δ-lactone (GDL) acidification, focusing on the impact of GDL concentrations on microstructure, rheology, and texture of the resulting gels. The PWPI gel exhibited lower hardness but a smoother structure than the AWPI gel. Notably, acidification with GDL (0.6-1.2%) significantly increased the stiffness of PWPI gels, increasing storage modulus and yield stress 10-50 times, while weakening AWPI gels. Varying concentrations of GDL effectively modulated the microstructure of the PWPI gels, leading to the altered texture (from a soft-solid state to a well-self-supporting stiff-solid gel) and water holding capacity (from approximately 46% to 85%). Additionally, hydrophobic interactions and disulfide bonds were identified as the primary forces involved in the gels.

Abstract:A novel composite hydrogel incorporated physical-chemical treated insoluble (ISF) into high acyl gellan (HAG) was fabricated and characterized. ISF-HAG hydrogel exhibited better appearance and denser network structure compared to HAG hydrogel (control sample), which was further manifested by higher gel strength and storage module G′. It was attributed to hydrogen bonds between ISF and HAG, confirmed by FT-IR analysis, as well as ISF as a supporter or filler in the network pores. Temperature sweeps showed that the gel point temperature and gelling rate of ISF-HAG hydrogel were higher than those of control sample by a maximum of 5 °C and 5.22 Pa/°C, respectively. ISF-HAG hydrogel also presented good swelling capacity and texture properties. USAT-ISF obtained by ultrasonic assisted steam-cooking alkaline treatment exerted a great impact on the formation and performance of HAG hydrogel due to loose structure and outstanding physicochemical properties (water holding capacity, swelling capacity, etc.) This study explores the potential of ISF to promote gelation and expands the range of applications for HAG hydrogel.

Abstract:This study assessed the effect of konjac glucomannan (KGM) on the aggregation of soy protein isolate (SPI) and its gel-related structure and properties. Raman results showed that KGM promoted the rearrangement of SPI to form more β-sheets, contributing to the formation of an ordered structure. Atomic force microscopy, confocal laser scanning microscopy, and small-angle X-ray scattering results indicated that KGM reduced the size of SPI particles, narrowed their size distribution, and loosened the large aggregates formed by the stacking of SPI particles, improving the uniformity of gel system. As the hydrogen bonding between the KGM and SPI molecules enhanced, a well-developed network structure was obtained, further reducing the immobilized water's content (T22) and increasing the water-holding capacity (WHC) of SPI gel. Furthermore, this gel structure showed improved gel hardness and resistance to both small and large deformations. These findings facilitate the design and production of SPI-based gels with desired performance.

Abstract:Glycated conjugates were prepared by heating aqueous dispersions of soy protein isolate (SPI) and maltodextrin (MD) after ultrasonic treatment (138 W/cm2, 20 kHz) for 5, 15, and 25 min. The effect of the ultrasonic pretreatment on the structure, physicochemical characteristics, and acid-induced gelation properties of the resulting SPI–MD conjugates was investigated. The analyses of free amino groups, degree of graft, intermediates, and browning intensity confirmed the formation of SPI–MD conjugates. The ultrasonic treatment promoted the glycation reaction, which led to the increase in the β-type ordered secondary structure and less compact tertiary conformation of conjugates. The ultrasonic treatment for 5 min increased the isoelectric point, surface hydrophobicity (H0), free sulfhydryl (-SH) groups, and particle size of the conjugates. These changes in the structural and physicochemical properties of the conjugates affected their acid-induced gelation. Glucono-δ-lactone was used to form acid gels at 25 °C. Although the glycation with MD deteriorated the acid-induced gelation properties of SPI, the 5 min ultrasonic conjugates formed gels faster than the untreated conjugate, and their gels exhibited higher storage modulus (G′), gel strength, and water holding capacity (WHC), as well as denser microstructure, than the untreated conjugate gels. However, prolonged ultrasonic treatment (15 or 25 min) weakened the gelation properties of the conjugates by promoting the glycation reaction and reducing the H0. Thus, an optimum ultrasonic pretreatment can be used to modify the structure of soy protein glycation conjugates to control their gelation properties.

Rheological evidence of the gelation behavior of hyaluronan-gellan mixtures

Abstract:It was found that solutions of calcium hyaluronate (CaHA) (0.1 to approximately 0.5 wt%) could form a gel by mixing with solutions of sodium type gellan (0.1 to approximately 0.5 wt%), although neither polymer by itself forms a gel at low concentrations (0.1 to approximately 0.5 wt% in this experiment). The rheological properties of CaHA-gellan mixtures were investigated by dynamic and steady shear measurements. Both storage shear modulus G' and loss shear modulus G'' for CaHA-gellan mixtures increased with increasing time, and tended to an equilibrium value after 1 h. After reaching steady values of G' and G", the frequency dependence of G' and G'' was observed. G' was always larger than G'' in the accessible frequency range from 10(-2) to 10(2) rad/s. The effects of pH and calcium ions were examined. Gel formation of the mixtures was promoted by decreasing pH and adding from 0.01 to 0.1 M calcium ions, but excessive calcium ions weakened the gel.

Abstract:The increasing interest in hydrogel matrices and their diverse applications has fueled extensive research. However, single-component gels have a limited adjustable performance range, and multi-component gels raise concerns about biological safety, hindering their widespread use. This study focuses on harnessing high-speed shearing and ultrasound-assisted methods to incorporate active natural Haematococcus pluvialis (HP), creating novel composite hydrogels in conjunction with biological macromolecule gellan gum, and eliminating the need for structural modifications or chemical crosslinking. Rich astaxanthin, proteins, polysaccharides, and other components in HP can fill and promote the formation of a unified functional network. The study aims to explore the potential of HP as a rheology regulator and investigate its impact on the rheological properties of the gels. Various rheological models, including Power-Law, Herschel-Bulkley, and Arrhenius, were employed for comparative analysis. This pioneering report on gellan/HP hydrogels holds significant importance as they exhibit optimized elasticity, thermal stability, enhanced injectability, and self-recovery, making them suitable for a wide range of applications in specialized medical food and biomedicine.

Abstract:Glycosaminoglycans (GAGs), such as hyaluronic acid (HA) and chondroitin sulfate (CS), have seen widespread adoption as components of tissue engineering scaffolds because of their potent bioactive properties and ease of chemical modification. However, modification of the biopolymers will impair biological recognition of the GAG and reduce the bioactive properties of the material. In this work, we studied how the degree of thiolation of HA and CS, along with other key hydrogel design parameters, affected the physical and bioactive properties of the bulk hydrogel. Although properties, such as the HA molecular weight, did not have a major effect, increasing the degree of thiolation of both HA and CS decreased their biorecognition in experimental analogues for cell/matrix remodeling and binding. Furthermore, combining HA and CS into dual polymer network hydrogels also modulated the physical and bioactive properties, as seen with differences in gel stiffness, degradation rate, and encapsulated cell viability.

Abstract:This study systematically investigated the dose-effect relationship and regulatory mechanism of theanine on the gelling properties of soy protein isolate (SPI). The results indicated that the strength and water holding capacity of 0.10% theanine-SPI gel increased by 238.80% and 1.13% compared with pure SPI gel, respectively. Hydrogen bonds and van der Waals forces mainly promoted the binding of theanine to β-conglycinin and glycinin. This binding could further change the protein aggregation state (reducing particle size and irregular aggregation), functional groups (exposing more hydrophilic amino acid residues and sulfhydryl groups), and molecular structure (α-helix and β-turn transforming into β-sheet). Furthermore, a more uniform and dense three-dimensional gel network was formed in the composite gels, offering the structural basis for improving the performance of the SPI gel. The results will provide theoretical basis and technical support for the precise regulation of gel properties of the functional amino acid-SPI complex as well as the efficient creation of new SPI-based foods.

Abstract:Breast cancer is the most frequent type of cancer among women worldwide. Vascular endothelial growth factor (VEGF), the key modulator of angiogenesis, has been implicated in breast cancer susceptibility and aggressiveness. VEGF expression was determined in 99 breast cancer tissue samples using reverse transcription-polymerase chain reaction and the human epidermal growth factor receptor 2 (HER2) status was determined by immunohistochemistry. Subsequently, the associations of VEGF, HER2 and hormone receptor status with clinicopathological data were evaluated. High VEGF expression was found to be significantly correlated with the presence of lymphovascular invasion. In hormone receptor-positive/HER2-positive, HER2-positive and triple-negative breast cancer, high VEGF expression was correlated with the presence of axillary nodal metastasis and lower overall survival rates. Therefore, the assessment of the VEGF status along with the hormone receptor and HER2 status may help identify high-risk patients who may benefit from anti-VEGF treatment.

Abstract:Injectable hydrogels are promising materials for cartilage regeneration in tissue engineering due to their tunable crosslinking rates, mechanical properties, and biodegradation profiles. This study investigates the chondrogenic potential of hyaluronic acid (HA) hydrogels crosslinked via tyramine (TA) moieties, with and without gelatin modified with TA (Gel-TA). Incorporating Gel-TA improved cell viability, spreading, and cartilage matrix deposition, particularly in medium and high molecular weight (MMW and HMW) HA-TA/Gel-TA hydrogels. Although the hydrogels' molecular weight did not significantly alter stiffness, MMW and HMW HA-TA/Gel-TA formulations exhibited enhanced functional properties such as slower degradation and superior cartilage matrix deposition. These attributes, coupled with Gel-TA's effects, underscore the importance of both molecular weight and biofunctional components in hydrogel design for cartilage regeneration. While low molecular weight (LMW) HA-TA hydrogels offered excellent injectability and supported high cell viability, they degraded rapidly and exhibited reduced cartilage matrix formation. Gel-TA enhanced cell adhesion and spreading by providing integrin-binding sites and promoted collagen type II deposition, crucial for cartilage regeneration. Moreover, the increased stiffness of MMW and HMW HA-TA/Gel-TA hydrogels facilitated extracellular matrix production. These findings show the potential of Gel-TA-modified HA-TA hydrogels for cartilage tissue engineering, with the opportunity for further optimization through the incorporation of bioactive components.

Abstract:Osteoarthritis (OA) is a common disease caused by damage to articular cartilage and underlying bone tissues. Early OA can be treated by intra-articular injection of viscosupplements to restore the lost viscoelasticity and lubricity of synovial fluid. Hyaluronic acid (HA), as a most standard synovial fluid supplementation, can be easily degraded in vivo, bringing about lower viscoelasticity and increased injection frequency. In this study, we focus on products with reasonable viscoelasticity and long-lasting action time and develop a kind of polysaccharide-based hydrogel viscosupplement (HEG) based on HA chemically modified gellan gum (GG), which can maintain stable viscoelasticity with hyaluronidase for 1 week owing to the fact that the main component of GG still maintains a stable three-dimensional network structure after enzyme treatment. The as-developed injectable HEG hydrogel possess good biocompatibility, excellent injectability, suitable viscoelasticity, satisfied lubricity, and enzymatic resistance, demonstrating a big potential for the early OA treatment.

Abstract:Soybean protein isolate (SPI) is a high-quality plant protein that is primarily used to process various soybean products coagulated by transglutaminase (TGase). In this study, the degree of hydrolysis (DH), sulfhydryl content (SH), surface hydrophobicity (H0 ), secondary structural constitution, and microstructure of TGase-treated soybean protein (SPI, 7S, and 11S) were determined, as well as the effects of NaCl, urea, and SDS on the properties and intermolecular forces of SPI gel were analyzed. The results show that the H0 and SH content of SPI, 7S, and 11S decreased significantly with TGase treatment time (p < .05), while the DH gradually increased and reached its highest value (3.72%, 7.41%, and 1.27%, respectively) at 30 min. As the concentration of these two secondary structures exhibited an inverse relationship, the degradation of β-turns resulted in the increase in β-sheets. The microstructures of SPI and 11S gels were similar, being denser and more ordered than 7S gel. The low concentration of NaCl solution (0.2 mol/L) enhanced gel properties and intermolecular forces, promoting the formation of SPI gel, whereas a high concentration (0.4-0.8 mol/L) had a significant inhibitory effect. Urea and SDS solutions substantially inhibited the formation of SPI gel, leading to significant decreases in the water holding capacity and hardness as well as a considerable increase in the coagulation time (p < .05). The results revealed that hydrogen bonds and hydrophobic interactions were the main intermolecular forces responsible for the gel formation. This study provides adequate technical support and a theoretical basis for soybean protein gel products.

Abstract:Exploring the chance to convert biowaste into a valuable resource, this study tests the potential role of humic acids (HA), a class of multifunctional compounds obtained by oxidative decomposition of biomass, as physical agents to improve gelatin's mechanical and thermal properties. To this purpose, gelatin-HA aqueous samples were prepared at increasing HA content. HA/gelatin concentrations changed in the range 2.67-26.67 (wt/wt)%. Multiple techniques were employed to assess the influence of HA content on the gel properties and to unveil the underlying mechanisms. HAs increased gel strength up to a concentration of 13.33 (wt/wt)% and led to a weaker gel at higher concentrations. FT-IR and DSC results proved that HAs can establish noncovalent interactions through H-bonding with gelatin. Coagulation phenomena occur because of HA-gelatin interactions, and at concentrations greater than 13.33 (wt/wt)%, HAs established preferential bonds with water molecules, preventing them from coordinating with gelatin chains. These features were accompanied by a change in the secondary structure of gelatin, which lost the triple helix structure and exhibited an increase in the random coil conformation. Besides, higher HA weight content caused swelling phenomena due to HA water absorption, contributing to a weaker gel. The current findings may be useful to enable a better control of gelatin structures modified with composted biowaste, extending their exploitation for a large set of technological applications.

Abstract:Soy protein isolate (SPI) is often used in food processing due to the high water holding capacity and high stability. This paper explored the effect of heat treatment (HT) on the nonlinear rheological properties of acid-Induced soy protein isolate gels modified by high-pressure homogenization. The effects of HT on the storage modulus G′, loss modulus G″ and gel rate of the SPI gel were studied through time sweep, frequency sweep and strain sweep tests. The effects of the Lissajous curve, Chebyshev stress decomposition and Fourier transformation were also investigated. The results illustrated that the HT significantly enhanced the mechanical properties of the SPI gel. The curve area and energy dissipation of the Lissajous curve after HT were significantly reduced. Chebyshev stress decomposition illustrated that HT enhanced the shear thickening behavior of SPI gel. The study also found that HT significantly improved the structure and strain resistance of the SPI gel structure. In short, HT significantly improved the nonlinear rheological properties of the SPI gel.

Abstract:The storage modulus (G′) and gel hardness of non-heated and heat-treated SPI at neutral pH and those of subsequently formed GDL-induced gels were measured. The values obtained for acid-induced gels formed by heat-treated SPI were significantly increased compared to those formed by non-heated SPI. The physicochemical properties of non-heated SPI and heat-treated SPI at pH 6.9 (denaturation degree, dispersibility, water-holding capacity, sulfhydryl groups and surface hydrophobicity) were measured to correlate their effects to gel strength and gel hardness, sulfhydryl groups and water-holding capacity of GDL-induced gels.
