Abstract:A thin-film based on 3-aminopropyl triethoxysilane surface-modified Ce-doped zinc-aluminum layered double hydroxide was synthesized on the inner surface of an aluminum tube. It has been applied to in-tube stir bar sorptive extraction of nonsteroidal anti-inflammatory drugs in saliva samples followed by high-performance liquid chromatography. The sorbent was characterized by scanning electron microscopy, X-ray diffraction, Fourier-transform infrared spectroscopy, energy-dispersive X-ray spectroscopy, and elemental mapping. The extraction parameters including sample pH (4.2), extraction time (10 min), stirring speed (800 rpm), type of eluent (acidified tetrahydrofuran), eluent volume (100 μL), and desorption time (6 min) were thoroughly optimized. Under the optimum conditions, limits of detection were found to be less than 5.0 ng mL-1. Calibration plots were linear within the range 10-1000 ng mL-1 (R2 > 0.9982). Absolute recoveries were calculated in the range 63.5 to 72.4%. The repeatability (intra- and inter-day precision) and reproducibility (tube-to-tube precision) at concentrations of 50, 250, and 500 ng mL-1 were less than 7.6% and 9.4%, respectively. The method accuracy based on the relative error was calculated at these concentrations and ranged from - 4.9 to - 9.3% for intra-day relative error (%) and - 6.8 to - 11% for inter-day relative error (%). Finally, the method applicability was examined for the determination of nonsteroidal anti-inflammatory drugs in saliva samples, and good relative recoveries were obtained within the range 86.5 to 95.2%. As a result, the introduced method can be applied as a suitable alternative to measuring nonsteroidal anti-inflammatory drugs in biological fluids. A novel sorbent based on electrospun for electrically-assisted solid phase microextraction of six non-steroid anti-inflammatory drugs, followed by quantitation with HPLC-UV in human plasma samples

Abstract:Background: Although NSAIDs possess notable therapeutic and pharmaceutical qualities, it's essential to acknowledge that excessive doses can result in toxicity within the human body. Moreover, the importance lies in identifying and measuring their trace amounts. Due to their existence within intricate matrices, the creation of novel electrospun nanofibers as sorbents for electrically-assisted solidphase microextraction (EA-SPME) becomes vital. This innovation caters to the requirement for the effective pre-treatment of NSAID samples, providing a strategic approach to managing the complexities associated with trace quantities found in various matrices. Results: First, polyvinylalcohol/casein/tannic acid/polyaniline/titanium dioxide nanoparticles (PVA/CAS/TA/PANI/TiO2 NPs) electrospun nanofibers were prepared for EA-SPME on pewter rode and then, trace amounts of six NSAIDs (Acetaminophen, Caffeine, Naproxen, Celecoxib, Ibuprofen and mefenamic acid) were adsorbed chemically on these nanofibers. In the next step, the desorption of six NSAIDs was electrochemically done from prepared electrospun nanofibers on a pewter rod which was as working electrode at three electrodes system. Finally, these drugs were quantified from different human plasma samples with HPLC-UV. The synthesis of electrospun nanofibers was confirmed through a series of analytical techniques including field emission-scanning electron microscopy (FE-SEM), energy-dispersive X-ray spectroscopy with elemental mapping analysis (EDX-Mapping), X-ray diffraction (XRD), and Fourier transform-infrared (FT-IR). The optimal percentage of additive compounds to PVA/CAS for electrospinning, as well as the factors influencing adsorption and desorption processes, were determined through both of Design Expert software and MATLAB programming language. Significance: Under optimum conditions, the wide linear range was 27-8000 ng mL-1 with R2≥ 0.9897, low detection limits were ranged from 8 to 27.3 ng mL-1 based on S/N = 3 and significant enrichment factors were acquired. The intra-day and inter-day RSDs% were obtained within the 4.51% - 5.68% and 4.28%-5.45%, respectively. Finally, The effectiveness of the EA-SPME-HPLC-UV method was assessed for determining NSAIDs in plasma samples, demonstrating good recoveries ranging from 90.2% to 105.2%.

Abstract:A mesoporous silica Santa Barbara Amorphous-15 (SBA-15) has been first functionalized with 3-[2-(2-aminoethylamino)ethylamino]propyl-trimethoxysilane (a silane with three amines) and then reacted with an excess of phenyl glycidyl ether to generate a mixed-mode anion-exchanger containing both anion-exchange (three amines) and reversed-phase (multiple ether-linked phenyls) functionalities in a single branched ligand. The resulting material has been characterized by scanning electron microscopy, transmission electron microscopy, nitrogen adsorption-desorption measurements, Fourier-transform infrared spectroscopy, and elemental analysis. The results obtained indicated a BET specific surface area (SBET) of 362.5m2g-1, a pore volume of 0.70cm3g-1 with a narrow pore size distribution centered at 6.6nm, and carbon and nitrogen contents of 28.30% and 2.84%, respectively. The dimensions of these particles (∼5μm diameter, ∼60μm length), their large surface areas, their high-density functionalities and anion-exchange mixed-mode characteristics make them very attractive for highly effective solid phase extraction (SPE) of acidic nonsteroidal anti-inflammatory drugs (NSAIDs). The important parameters on extraction efficiency including sample pH, breakthrough volume, type and volume of eluent were optimized. A simple and sensitive analytical method based on mixed-mode SPE coupled to high-performance liquid chromatography with ultraviolet detection (HPLC-UV) was developed and successfully applied to the analysis of four NSAIDs (ketoprofen, naproxen, diclofenac, and ibuprofen) in spiked real water samples with satisfactory recoveries (80.6-110.9%) and repeatability (relative standard deviation <11.3%, n=3). The limit of detections of four NSAIDs were 0.006-0.070μgL-1 for tap water, and 0.014-0.16μgL-1 for river water and wastewater, with the enrichment factors of 806-1109-fold.

Abstract:This study synthesized and characterized layered double hydroxides (LDHs) intercalated with ethylenediaminetetraacetic acid (EDTA) to increase the basal spacing of electrosynthesized Zn-Al-LDHs on pencil graphite (PG). The characteristic verification of the Zn-Al-EDTA-LDH nanostructures was conducted utilizing Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), energy dispersive and X-ray analysis (EDS), and X-ray diffraction (XRD). To improve the extraction efficiency, Zn-Al-EDTA-LDHs as adsorbent in solid phase microextraction (SPME) fiber was combined with hollow fiber liquid–liquid phase microextraction (HF-LLPME) to investigate the extraction efficiency of HF-LLME@PG-SPME technique the chosen non-steroidal anti-inflammatory drugs (NSAIDs) from aqueous solution Several variables, comprising extraction time, desorption time, stirring rate, desorption volume, pH of donor phase, and salt effects were screened and optimized by performing placket-Burman design (PBD). After evaluating the extraction and desorption procedures under optimal conditions, NSAIDs analysis was performed employing high-performance liquid chromatography-ultraviolet detection (HPLC-UV). Compared to the most common clean-up and sample preparation methods, the current study focused on developing a greener analytical chemistry process due to reduced solvent consumption, cost-effectiveness, miniaturization and simplified design. The proposed method for NSAIDs exhibited a good linear dynamic range (LDRs) (1.00–200.00 μg/L) with a coefficient of determination of > 0.9956, low limits of detection (LODs) (0.26–0.29 μg/L) and acceptable limits of quantifications (LOQs) (0.86–0.96 μg/L). Relative standard deviations of 5.31 % could be determined. The absolute recoveries of the urine sample analyses could be obtained in a 89.91–97.61 % range and enrichment factors (EFs) (178.92 to 183.88). Investigations of the developed method were also conducted in various urine samples containing the selected drugs. As a result of the obtained recoveries, the method proved to be valuable and suitable to complicated biological samples.

Abstract:In this research, graphitic carbon nitride (g-C 3 N 4 ) was synthesized through thermal condensation method and was used as nanofiller to prepare a novel nanocomposite (NC). In this regard, g-C 3 N 4 was incorporated into nylon 6 (N6) by solvent blending method and afterward, it was solidified as a monolithic thin film through phase separation process. The characterization of N6 and g-C3N4/N6 NC thin films was performed using X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR) and scanning electron microscopy (SEM). The prepared thin film based on g-C3N4/N6 NC was utilized for thin-film microextraction (TFME) of carvedilol (CAR) in deionized water, urine and plasma samples followed by fluorescence spectrometry. The effect of g-C3N4 doping level on the thin film's extraction capability was investigated, indicating the significant effect of g-C 3 N 4 reinforcing in N6 matrices on the extraction efficiency. Central composite design (CCD) was applied to investigate the effect of important factors influencing the extraction and desorption efficiency, and to optimize the extraction and desorption condition. Based on the method validation, limit of detection (LOD) and limit of quantification (LOQ) were 1 and 3.3 ng mL −1 , respectively. The developed method's linearity was in the range of 3.3–100 ng mL −1 , 16.5–500 ng mL −1 , and 16.5–250 ng mL −1 for deionized water, urine and plasma sample, respectively, with satisfied determination coefficient ( r 2 > 0.9912). The repeatability of the method in the terms of intra-day, inter-day and inter-sorbent precisions ( n = 8) were 3.6, 7.6 and 8.3%, respectively. Relative recoveries for spiked plasma and urine samples were 86.9% and 82.7%, respectively.

Abstract:The growing use of non-steroidal anti-inflammatory drugs (NSAIDs) has seriously affected human health and ecosystems, as a result, the World Health Organization (WHO) has regarded them as emerging contaminants. NSAID's polar nature and trace amount present in wastewater make their extraction and determination challenging in modern analytical science. Therefore, regarding the need, we herein report a rapid, sensitive and selective in-syringe solid-phase extraction (SPE) method coupled with ultra-high performance liquid chromatography and UV detector for the determination of seven NSAIDs in environmental water samples. Specifically, the NSAIDs in water samples were directly extracted by using synthesized porous covalent organic framework functionalized poly (styrene-divinyl benzene-glycidylmethacrylate) composite (COF@PS-GMA) without further cleanup procedures. The adsorption of NSAIDs on COF@PS-GMA sorbent was investigated by using adsorption isotherms and kinetic studies. Various parameters, including amount of sorbent, pH of the samples, the volume of desorption solvent, and desorption time that were involved in in-syringe SPE were optimized. Under the optimized conditions, good linearity was observed at 0.005-5.0 mu g mL(-1) with method quantification limits (LOQs) estimated between 0.54-2.74 mu g L-1. The recoveries of the seven NSAIDs at the level of 0.5, 5.0 and 20.0 mu g L-1 were ranging from 84.3% to 99.6%. (C) 2018 Elsevier B.V. All rights reserved.
[Prognostic factors of local recurrence after conservative surgery in breast carcinoma. Our experience].

Abstract:One of the most important problems linked to the conservative surgery of breast cancer consists of local recurrences (LR), even if it has now been proven that the appearance of LR has no influence on overall survival of the patients operated for breast cancer. We have considered 108 patients suffering from breast cancer and treated with quadrantectomy plus axillary dissection. In these patients were found 4 LR (3.7%), that in two cases appeared in the residual mammary gland and in other two cases were found on the skin scar. The mean time of occurrence of the LR was of 43 month (range 18-90 month). The authors have correlated the appearance of LR with some characteristics of the tumor, as the pT, the histologic type, the histologic Grading, the estrogen receptor status and the extensive intraductal component of the tumor (EIC). No particular correlations were found between the pT, histologic type, histologic Grading, estrogen receptor status of the tumor and the onset of LR. Instead a significant correlation was found between the LR appearance and the EIC of the tumor. In fact, for the tumors with a rich EIC there was found a 11.5% incidence of LR against a 1.2% impact of LR for the tumors with a low EIC. The authors conclude that is impossible now to assert that exist really some prognostic factor but is most important to note that there are many data in the literature that seems ever more to point at the rich EIC of the tumor as an important prognostic factor of LR.
Solid phase microextraction with poly(deep eutectic solvent) monolithic column online coupled to HPLC for determination of non-steroidal anti-inflammatory drugs

Abstract:Polymer monolithic column based on a green deep eutectic solvent (DES) was prepared for in-tube solid phase microextraction of non-steroidal anti-inflammatory drugs (NSAIDs) in aqueous samples. DES composed of choline chloride and itaconic acid was adopted as functional monomer to synthesize a polymeric monolith inside polydopamine-functionalized poly(ether ether ketone) (PEEK) tube. The obtained monolithic column was characterized with fourier transform infrared spectrometry, X-ray photoelectron spectrometer and field emission scanning electron microscope. An online SPME-HPLC method was developed by connecting the PEEK tube to the HPLC system. Parameters including sampling rate, desorption time, sample pH value and sample volume that can affect the performance of this method were investigated. Under the optimized conditions, the proposed SPME-HPLC method showed high extraction efficiency (enrichment factors about 100), good linearity (R ≥ 0.9997) and good reproducibility (relative standard deviation < 4.32%) for determination of three NSAIDs in aqueous solution. The limits of quantification were found to be in a range of 0.05-0.25 ng mL-1. The SPME-HPLC method was also successfully applied to detect NSAIDs in lake water and human plasma samples.

Abstract:NH2-MIL-53(Al) was in situ synthesized on the SiO2@dSiO2 with a simple solvothermal method. The prepared composite was packed into column solid phase extraction as adsorbent. After introduction of SiO2@dSiO2 as the support, the drawbacks of adsorbent loss and high backpressure were overcome. Using ultra-high performance liquid chromatography-mass spectrometry, the proposed method was employed to analyze trace non-steroidal anti-inflammatory drugs (NSAIDs) in water samples. Extraction parameters, including type of adsorbent, pH of sample solution, flow rate, sample volume, type and volume of elution solvent, were optimized. Then commendable sensitivity was obtained with limits of detection from 0.5 to 4.0 pg·mL−1, and limits of quantification from 1.5 to 13.3 pg·mL−1. Moreover, the correlation coefficient (r) of linearity was 0.9916–0.9974. Satisfactorily, the recoveries of the target compounds were between 75.3 % and 109.3 %, and the relative standard deviation was less than 7.8 %. Furthermore, naproxen was chosen as a nominee to evaluate adsorption behavior of SiO2@dSiO2@NH2-MIL-53(Al). Molecular docking simulation calculations were conducted to explore adsorption mechanism between NH2-MIL-53(Al) and five NSAIDs.

Abstract:Non-steroidal anti-inflammatory drugs (NSAIDs) have been recognized as a potentially serious threat to the natural environment. NSAIDs are popular painkillers, and the main pathway for them to reach natural water is via discharge from wastewater and sewage treatment plants. In order to monitor contamination caused by these drugs, as well as their impact on the environment, a new material based on Silica Gel 60, functionalized with a dendrimeric copolymer of methylamine and 1,4-butanediol diglycidyl ether (named MA-BDDE), was prepared. Initial physicochemical characterization of the MA-BDDE material was carried out using ATR FT-IR spectroscopy as well as solid-state carbon-13 NMR spectroscopy. Its effectiveness at NSAID extraction was evaluated by the application of five select drugs in dispersive solid-phase extraction (dSPE): aspirin, ketoprofen, naproxen, diclofenac and ibuprofen. This was followed by their simultaneous determination using the HPLC-UV/Vis system demonstrating good sensitivity, with limits of detection values within the 63–265 ng mL−1 range. A comparison of the sorption capacity of each pharmaceutical with unmodified base silica showed an at least tenfold increase in capacity after modification. Initial MA-BDDE application in a quick, low-waste extraction procedure of those select NSAIDs from spiked surface water samples yielded promising results for its use as a sorbent, as recovery values of analytes adsorbed from various samples were found to exceed 72%.

Abstract:In the present study, the author describe a method for magnetic solid phase extraction (MSPE) combined with ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) for the quantitation of seven non-steroidal anti-inflammatory drugs (NSAIDs) in environmental water samples. Core-shell Fe3O4@MIL-100(Fe) as the adsorbent in the MSPE was novelly synthesized and successfully applied for the first time. Several experimental parameters, including the effect of pH value of aqueous solution, the amount of the adsorbent, ionic strength, adsorption time, desorption solvent and its volume, and desorption time, were investigated. Under the optimal conditions, the linear ranges were 0.1-10 µg L-1 for meloxicam, carprofen, indometacin, and tolfenamic acid, 0.2-20 µg L-1 for diclofenac, and 0.3-30 µg L-1 for naproxen and mefenamic acid with the regression coefficients (r) from 0.9987 to 0.9995, respectively. The limits of detection were between 0.02 and 0.09 µg L-1. The method also exhibited excellent precision (RSD ≤ 9.6%), repeatability (RSD ≤ 9.3%), and recovery (75.2-105.2%). The method presented the advantages of simplicity, rapidly, less solvent consumption, and higher sensitivity. It's promising for the core-shell Fe3O4@MIL-100(Fe) to be used as the adsorbent of MSPE for detecting and removing residues in the environment.
Immunohistochemical detection of cytoplasmic nucleophosmin in formalin-fixed paraffin-embedded marrow trephine biopsies in acute myeloid leukaemia

Abstract:Aims Nucleophosmin (NPM1) gene mutations resulting in cytoplasmic delocalisation of nucleophosmin (NPMc+) are the most common genetic abnormality in acute myeloid leukaemia (AML). In this study, we tested whether immunohistochemical (IHC) detection of cytoplasmic NPM1 (cNPM1) in formalin-fixed bone marrow trephine biopsies correlated with NPM1 mutations and the prognostic impact of NPM1 and fms-related tyrosine kinase 3-internal tandem duplication (FLT3-ITD) gene mutations was also assessed. Methods A total of 71 Chinese adult de novo AML cases were evaluated for cNPM1 by IHC where the bone marrow trephines were fixed in 10% buffered formalin and decalcified by 5% EDTA. NPM1 and FLT3-ITD gene mutations were also investigated using PCR, fragment analysis and direct DNA sequencing. Results IHC analysis of cNPM1 had a very good sensitivity (86.7%) and excellent specificity (96.4%) for NPM1 mutation. The positive predictive value was 86.7% and the negative predictive value was 96.4%. NPM1 mutations and FLT3-ITD were closely associated (p=0.003). Patients with mutated NPM1 and without FLT3-ITD mutation have a longer overall survival (p=0.042) than patients with both NPM1 and FLT3-ITD mutations. Conclusions Our results showed that IHC detection of cNPM1 in formalin-fixed trephine biopsies correlated well but not entirely with NPM1 mutation. Furthermore, NPM1 mutations were significantly more frequent in FLT3-ITD than FLT3-wild-type cases.

Abstract:A weak anion exchange polymer monolith based on poly(4-vinylpyridine-co-ethylene dimethacrylate) (poly(VP-co-EDMA)) was prepared in the capillary. The use of polyethylene glycol (PEG) as porogens and EDMA as crosslinking monomers helps to increase the specific surface area and enhance hydrophobicity of the target monolith. The monolith exhibited satisfactory permeability, high mechanical strength and good stability in aqueous buffer. The potential application of the monolith as extraction medium in complicated samples has been demonstrated by in-tube solid-phase microextraction (in-tube SPME) of three non-steroidal anti-inflammatory drugs (NSAIDs) (ketoprofen, fenbufen and ibuprofen) in human plasma and environmental water followed by on-line liquid chromatography-mass spectrometry (LC-MS) analysis. Integration of the sample extraction, LC separation and MS detection into a single system allowed for direct analysis of the NSAIDs in diluted plasma or environmental water sample, which facilitates the realization of automation of the sampling, extraction, separation and detection within a short period of time. The results showed that the limits of quantitation (S/N = 10) for the three NSAIDs were 6.70-15.9 ng mL(-1) in human plasma and 0.65-1.87 ng mL(-1) in water from East Lake. And the recoveries of the three NSAIDs spiked in human plasma and water from East Lake were from 88.17% to 112.48%, with relative standard deviations less than 13.87%. Compared to previously reported off-line sample extraction and conventional LC methods, the newly developed automated in-tube SPME/HPLC-MS in the current study provided a powerful tool for the fast and sensitive analysis of three NSAIDs in human plasma and environmental water.

Abstract:Separation and detection of residual drug in biological fluids has directly related to human health in term of their beneficial or side effects. In biological fluid samples (complex matrix which containing drug at very low level) conduction of preliminary efficient technique as good choice for pre-concentration and clean-up of real sample before their instrumental analysis is highly recommended. In this result technique, based on nano-structure material which poses higher available surface area and larger member of reactive sites led to significant improvement in characteristic performance of analytical method. This paper focused on the synthesis and application of novel nano-sorbent for pre-concentration and extraction of atorvastatin (AT) from different biological fluids. Influence of various variables including sorbent dosage, eluent volume and sonication time on present method response was studied and optimized by central composite design under response surface methodology and accordingly an acceptable calibration curves over wide linear ranges (0.3-2000μgL-1) with high coefficient of determination higher than 0.999 strongly confirm high applicability of present method for quantification of analyte while limit of detection and quantification in plasma sample were 0.0608 and 0.2033μg L-1, respectively.

Abstract:In this work, a dispersive micro-solid phase extraction technique along with high-performance liquid chromatography-UV detection was developed for simultaneous preconcentraion and determination of trace levels of codeine and tramadol in human saliva. This method is based on the adsorption of codeine and tramadol on a mixture of oxidized multi-walled carbon nanotubes and zeolite Y nanoparticles with 1:1 ratio as an efficient nanosorbent. Various analytical parameters influencing the adsorption step including the amount of adsorbent, the pH of the sample solution, the temperature, the stirring rate, the contact time of the sample solution, and the adsorption capacity were investigated. Based on the results, 10 mg adsorbent, sample solutions with pH = 7.6, temperature of 25 °C, stirring rate 750 rpm and contact time 15 min, in the adsorption step shows the best results for both drugs. Then the effective parameters on the analyte desorption stage such as the type of desorption solution, pH of the desorption solution, desorption time and desorption volume were investigated. Studies have shown that water/methanol (50:50 v/v) as desorption solution, pH = 2.0, desorption time of 5 min and desorption volume of 2 ml gives the best results.Chromatographic separation was performed on a RP-Shim-pack CLC-ODS-C18 column (250 mm × 4.6 mm, 5 µm) with isocratic mode. The mobile phase contained of acetonitrile:phosphate buffer (18:82, v/v) at pH = 4.5 and the flow rate was 1 ml.min-1. The wavelength of UV detector was adjusted at 210 nm and 198 nm for codeine and tramadol, respectively.Under optimum conditions, the extraction efficiencies of 98.5% and 99.2% were achieved for codeine and tramadol respectively. Enrichment factor of 13, detection limit of 0.3 μg L-1, relative standard deviation of 4.07 for codeine; and an enrichment factor of 15, a detection limit of 0.15 μg L-1, and standard deviation of 2.06 for tramadol were calculated. The linear range of the procedure for each drug was 1.0 to 1000 μg L-1. This method was successfully applied for the analysis of codeine and tramadol in saliva samples.
Electrodeposition of Zn–Al‐Layered Double Hydroxides With Interlayer Modification by Ibuprofen as Fiber Coating for Solid Phase Microextraction of Nonsteroidal Anti‐Inflammatory Drugs

Abstract:In this study, in situ electrodeposition of Zn─Al‐layered double hydroxides (Zn─Al‐LDHs) on the surface of pencil graphite substrate were practiced with interlayer modification by ibuprofen. The prepared composite increases the efficiency of the proposed method by increasing the level of interaction between the sorbent and the selected analytes including acetylsalicylic acid and naproxen (NAP) in real sample. Eventually, the extracted target analytes were injected into high‐performance liquid chromatography ultraviolet (HPLC‐UV) for their measurement. The experimental design including PBD and BBD was implemented to obtain the maximize efficiency and optimal condition for extraction of the target analytes. Therefore, in optimal conditions, limits of detection of 0.33–0.64 μg L−1, linear dynamic range (LDR) of 1–100 μg L−1 (with r2 > 0.9934), and limits of quantification (LOQs) of 1.1–2.1 μg L−1 were obtained for target analytes by SPME‐HPLC‐UV. The intra‐day relative standard deviation (RSD%) were found to range in 3.90%–5.30% (4.90%–6.80%) at concentration of 5–50 μg L−1 (2–10 μg L−1). According to obtained result, the relative recovery was calculated between 90% and 109%. Therefore, the obtained results showed that the selected method has a suitable performance for the preconcentration and extraction of acetylsalicylic acid and NAP in biological samples.

Abstract:This work, reports the successful preparation a thin film by a simple and inexpensive process for quantification of a model analytes in the urine sample using HPLC-UV. To this end, cellulose paper was employed as a substrate for the in-situ synthesis of MOF-5. to increase s the resistance of the prepared film. The prepared film can be reused 26 times with no reduction in its performance. The thin film prepared by MOF-5 modified cellulose substrate was utilized in thin film microextraction (TFME) method for the extraction and preconcentration of naproxen, aspirin, tolmetin, and celecoxib. Under optimal conditions, the linear dynamic range of the target analytes was 2-500 μg L −1 with correlation coefficients (R 2 ) ranging from 0.9961 and 0.9990. Also, the limits of detection (LODs), the limits of quantification (LOQs) and relative standard deviation (RSD%) of the proposed method for selected analytes ranged between 0.57 and 0.77 μg L −1 , 1.7 to 2.3 and 3.5% to 6.2%, respectively. Moreover, relative recoveries varied from of 94% - to 108%, indicating the absence of matrices effect in the proposed method. Eventually, the TFME was successfully used for the extraction of selected analytes from urine samples.

Abstract:A four-part and sustainable nanocomposite composed of carbon quantum dots modified with zeolite imidazolate framework-90, polyvinyl pyrrolidone and magnetite (CQDs/ZIF-90/PVP/Fe3O4) was fabricated and applied in ultrasound-assisted magnetic dispersive micro-solid-phase extraction (US-A-MDMSPE). US-A-MDMSPE was followed by high-performance liquid chromatography with ultraviolet detection (HPLC-UV) for extraction, enrichment and simultaneous low-level monitoring of paracetamol (PCM) and etodolac (EDL) in human plasma. To increase the extraction yield and improve the sensitivity, nanohybrid arrays of metal-organic frameworks and conductive polymers were immobilized on the surface of CQDs followed by magnetization. The nano-extractor was characterized via Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), field-emission scanning electron microscopy (FE-SEM) and energy-dispersive X-ray spectroscopy (EDX). The recent method provided limits of detection (LODs) of 0.16 and 0.09 ng mL-1 for PCM and EDL, respectively. The calibration curve for PCM and EDL was linear in the range of 0.7-2000 ng mL-1 and 0.5-1200 ng mL-1 with a regression (r 2) value between 0.993 and 0.998, respectively. Acceptable precisions including intra-assay (≤6.9%) and inter-assay (≤8.3%) accuracies and notable accuracy (≤8.5%) were achieved to demonstrate the applicability of this method for the evaluation of the pharmacokinetic data of target drugs in human plasma.

Abstract:The in vitro activity of piperacillin, a new semisynthetic piperazine penicillin derivative, was evaluated against 626 clinical isolates and compared with the activity of other beta-lactam antibiotics. At a concentration of 0.1 microgram/ml, piperacillin inhibited all streptococci except enterococci. Non-beta-lactamase-producing staphylococci were inhibited by 1.6 microgram or less per ml. Both beta-lactamase- and non-beta-lactamase-producing Haemophilus were inhibited by 0.1 microgram/ml. Piperacillin inhibited non-beta-lactamase-producing Escherichia coli, Salmonella, and Shigella at a concentration of 6.3 micrograms/ml, but 20% of strains of these species containing type III beta-lactamase were not inhibited by 100 micrograms/ml. Piperacillin at 25 micrograms/ml, inhibited 83% of Citrobacter, 58% of Klebsiella, 88% of Enterobacter, and 50% of indole-positive Proteus, Acinetobacter, and Providencia. At 25 micrograms/ml, piperacillin inhibited 95% of Pseudomonas aeruginosa and 78% of Bacteroides fragilis. The minimal inhibitory concentration of piperacillin against Pseudomonas was affected by increasing the inoculum size and by pH. Minimum bactericidal concentrations against Pseudomonas and Serratia often were eightfold greater than the minimum inhibitory concentrations. Piperacillin was equal in activity to ampicillin against enterococci. It was more active than carbenicillin against E. coli, Klebsiella, Enterobacter, and Bacteroides. It was the most active penicillin against Pseudomonas and inhibited many strains of Pseudomonas for which the MICs of carbenicillin were above 200 micrograms/ml. Piperacillin was hydrolyzed by many different beta-lactamases. Synergistic activity of piperacillin was demonstrated when it was combined with amikacin, gentamicin, and cefazolin against P. aeruginosa and members of the Enterobacteriaceae. No antagonism was observed when piperacillin was combined with aminoglycosides; however, antagonism was observed rarely against E. coli when piperacillin was combined with cefazolin.
