Abstract:In the process of approval of active substances and authorization in the European Union and at the member state level, it must be shown by the applicant that an unacceptable risk for nontarget organisms in the aquatic environment by the active substance can be excluded. To achieve this aim, standardized models, scenarios, and agreed pesticide input parameters have to be used to calculate the exposure as defined by the FOrum for the Co‐ordination of pesticide fate models and their Use (FOCUS). During a period of daily surface water sampling lasting 3.5 years, a survey was conducted among farmers in the highly vulnerable catchment of Grote Kemmelbeek in Belgium to collect agronomic data on crops, application dates, and application rates of 12 applied herbicides and one metabolite. Daily surface water concentrations for the same herbicides were measured over 2.5 years for a second, much larger but less vulnerable, catchment of Kleine Aa (KAa). A comparison of realistic worst‐case predicted environmental concentrations in surface water (PECsw) according to FOCUS with measured concentrations shows that, in the GKb catchment, the PECsw was never exceeded for six substances, was exceeded on only 1–2 days for five substances, and was exceeded on 9–27 days for two substances. For the KAa catchment, the PECsw was only exceeded on two days for one compound and never for the other 12 compounds. These numbers correspond to a level of protection of the FOCUS PECsw between 100% and 97% and are much higher than the regulatory protection goal of 90%. [A simple and universal method for molecular sexing of birds].

Abstract:It is difficult to sex monomorphic or juvenile birds only according to their appearance or by simple surgical techniques. However, the sex of individual bird has to be identified during breeding. Recently, a sex-linked gene, CHD1, has been discovered in non-ratite birds. Using highly conserved primers flanking the intron of CHD1 and PCR amplification, females are characterized by diaplaying two fragments(CHD1W and CHD1Z), while males only show one fragment(CHD1Z) clearly different in size from the female-specific CHD1W fragment. PCR-based molecular sexing method has been widely used due to its many merits over other techniques. In this work, we successfully sexed 86 bird from 9 species using this technique. All of these birds are listed here as the national first-grade wildlife of China for protection. Our results benefit greatly on breeding rare monomorphic birds and preventing them from distinction.
A new laboratory method to study the impact of leaf texture on pesticide dislodgeable foliar residues (DFR)

Abstract:Pesticides are vital in meeting the challenge of feeding the rapidly increasing world population. However, it is crucial that they are used in a way that does not compromise the safety of humans or the environment. Non-dietary worker risk assessments consider the amount of residue which can be transferred from plant foliage to the skin or clothes, known as dislodgeable foliar residues (DFRs). DFR data scarcity due to the costly and seasonal characteristics of DFR studies is an obstacle to the extrapolation of DFR data to different crops/leaves. This paper validates a new proof-of-concept technique to investigate factors that may affect DFR (leaf texture) using the fungicide difenoconazole EC 10% as an example on various leaves (i.e., French bean, soybean, tomato, oilseed rape, and wheat). DFR was the lowest in the case of oilseed rape (31.0 ± 3.4%) and the highest in French beans (82.0 ± 2.9%). This significant difference in DFR in the findings of this study sheds light on the importance of the leaf surface as a major factor affecting DFR and supports the application of the laboratory method for more extensive data generation. More data generation would enable the extrapolation saving money and resources.

Abstract:The relationship between wettability and surface microstructure in a wide range of crop leaves from biological and interfacial structural perspectives is revealed. Crop production and quality safety system have the potential to nurture human health and improve environmental sustainability. Providing a growing global population with sufficient and healthy food is an immediate challenge. However, this system largely depends on the spraying of agrochemicals. Crop leaves are covered with different microstructures, exhibiting distinct hydrophilic, hydrophobic, or even superhydrophobic wetting characteristics, thus leading to various deposition difficulties of sprayed droplets. Here, the relationship between wettability and surface microstructure in different crop leaves from biological and interfacial structural perspectives is systematically demonstrated. A relational model is proposed in which complex microstructures lead to stronger leaf hydrophobicity. And adding surfactant with a faster dynamically migrating velocity and reducing droplet size can improve agrochemical precise deposition. These contribute toward highly accurate and efficient targeted applications with fewer agrochemicals use and promote sustainable models of eco‐friendly agriculture systems.

Effects of Single Rainfall Events on Leaching of Glyphosate and Bentazone on Two Different Soil Types, using the DAISY Model

Abstract:Core Ideas Modeling pesticide leaching during a single rainfall event with temporal variability. Short intense events (1 h) resulted in high glyphosate leaching to subsurface drains. Event volume controlled pesticide leaching at coarse sandy soil. The purpose of the present modeling study was to contribute to an improved understanding of the mechanisms involved in pesticide leaching during a single rainfall event with temporal variability. Rainfall intensity of the first event after pesticide application has great effect on the amount of pesticide transported to groundwater and subsurface drains, especially in soils containing preferential flow pathways. One way to improve the understanding of single event properties on pesticide leaching is to use a transport model. The soil–plant–atmosphere model Daisy was used to simulate pesticide leaching during and after single rainfall events of different durations and intensities. Designed temporally variable single rainfall events based on the Chicago Design Rain were inserted in the original weather file. A combination of different intensities (13, 20, 24, 28, 34, and 39 mm h −1 ) at different event durations (1, 3, 5, and 9 h) where the intensity peak was placed in the middle of the event ware applied, resulting in 24 different design events. The model setup included two different soil types: a coarse sandy soil and a sandy loam containing macropores and subsurface drains. The fates of the herbicides bentazone [3‐isopropyl‐1 H ‐2,1,3‐benzothiadiazin‐4(3 H )‐one 2,2‐dioxide] and glyphosate [ N ‐(phosphonomethyl)glycine] were simulated. The leaching dynamics of both pesticides showed high variability at the hourly level, illustrating the importance of high model resolution when estimating pesticide leaching. For the coarse sandy soil different intensities did not appear to have an effect, as pesticide leaching was controlled by event volume. In contrast, results for the sandy loam showed an effect of intensity, especially for glyphosate, at initially wet soil conditions. Short intense events (1 h) resulted in high leaching to drains (1.7% of matrix infiltration) compared to events of longer duration (up to 0.4% of matrix infiltration). This indicates that it might be more prudent to view leaching as a risk that occurs under certain conditions, rather than something that can be averaged.

Abstract:BACKGROUND Tillage operations will change the distribution in soil for any pesticide residues still present from earlier applications. This redistributive effect of tillage has been neglected in the study of pesticide leaching behaviour. This study reviews the literature to characterise this redistributive effect for different tillage operations and uses a pesticide leaching model to investigate the impact of redistribution on pesticide transport to subsurface drains which is a significant input route to surface water bodies. RESULTS Inversion ploughing moves the majority of any residues of pesticide present at or near the soil surface into the bottom two‐thirds of the plough layer, whereas non‐inversion ploughing has only a limited redistributive effect. Incorporating this redistributive effect into model simulations resulted in large changes (typically 5‐10 fold difference) in both the maximum concentration and total mass of pesticide transported to drains over the winter following cultivation. More intense cultivation decreased subsequent leaching for relatively mobile compounds (Koc ≤ 1000 mL g‐1), but increased it for strongly sorbed pesticides (Koc ≥ 2000 mL g‐1). CONCLUSION The redistributive effect of soil tillage on pesticide residues can have a large effect on subsequent transport to subsurface drains. This effect has been neglected in the literature. Field research is required to validate the model simulations presented here, and consideration should be given as to whether the effect needs to be included within risk assessment procedures. Abstract:In the year 2003 there was a 17% increase in the number of publications citing work performed using optical biosensor technology compared with the previous year. We collated the 962 total papers for 2003, identified the geographical regions where the work was performed, highlighted the instrument types on which it was carried out, and segregated the papers by biological system. In this overview, we spotlight 13 papers that should be on everyone's ‘must read’ list for 2003 and provide examples of how to identify and interpret high‐quality biosensor data. Although we still find that the literature is replete with poorly performed experiments, over‐interpreted results and a general lack of understanding of data analysis, we are optimistic that these shortcomings will be addressed as biosensor technology continues to mature. Herbicide transport via surface runoff during intermittent artificial rainfall: A laboratory plot scale study

Abstract:Surface runoff is highly relevant for herbicide transport into water bodies. Herbicide losses and the influence of physicochemical properties of flufenacet, metazachlor and terbuthylazine were studied on a 0.2m2 microplot of bare soil and slight slope (6%). Rainfall events were simulated with 13mmh−1 (±3mmh−1) precipitation intensity 4, 11, 18, 25, 32 and 39days after herbicide application. The runoff was sampled at 10-minute intervals during the simulated events. Herbicide concentrations and sediment amounts in the water phase were measured. The maximum concentrations in the water phase of the runoff ranged from 3184μgl−1 for metazachlor, 661μgl−1 for terbuthylazine and 207μgl−1 for flufenacet. The highest concentrations were detected in the first runoff samples which decreased exponentially with further rainfall. Highest loads occurred during the second rainfall event and after the first time surface runoff was generated. The artificial setup of intermittent rainfall revealed that with every new rainfall event after a rainless period, a higher amount of herbicides was released from the plot than at the last interval of the previous rainfall event. This effect was caused by alterations to the soil surface that occurred during the rainless periods. Furthermore, the results show that the herbicide loss relates to herbicide adsorption to organic carbon (KOC) and herbicide solubility in water. Therefore, the highest losses for terbuthylazine were 6.6% of the pesticide application, followed by flufenacet (3.8%) and metazachlor (2.5%).

Modelling pesticide concentrations in Japanese paddy fields using the RICEWQ model

Abstract:Increasing concerns about plant protection products in surface waters have highlighted the importance of pesticide monitoring and modelling, both nowadays integral components of the pesticide registration process. The Rice Water Quality (RICEWQ) model predicts the fate and transport of pesticides under various paddy environmental conditions. The model has been used widely for regulatory purpose in the U.S., while its use in Europe has been limited to regulatory submissions, despite multiple recommendations to adopt RICEWQ as a high tier assessment model for regulatory purposes. The applicability of the RICEWQ model under Japanese agricultural conditions remains unexplored. This study leverages field experimental data to evaluate the RICEWQ model's capability to simulate daily concentrations of two herbicides, mefenacet and pretilachlor, in paddy water and paddy soil in Japan. The RICEWQ model provided a reasonable level of performance for predicting the fate and transport of the two herbicides by ensuring that the simulated daily water balance corresponded with field observations and with minor pesticide-specific calibration. To further improve the performance of the simulations, we implemented a straightforward calibration framework. Calibrating the RICEWQ model improved the accuracy of all simulations; for both herbicides and in both paddy water and paddy soil compartments, the lowest Nash-Sutcliffe efficiency achieved was 0.725, demonstrating excellent performance. However, the RICEWQ model consistently underestimated the peak concentrations of herbicides in paddy water within the first three days of simulation. Simulation results suggested that approximately 50 % of the total applied herbicide was lost from the paddy system. Increasing the duration of the water holding period after pesticide application or increasing the storage capacity of the rice field via excess water storage depth management has the potential to reduce greatly herbicide loss to below 10 % of the total applied herbicide.

Abstract:Pesticides are detected in surface water and groundwater, endangering the environment. In lowland regions with subsurface drainage systems, drained depressions become hotspots for transport of pesticides and their transformation products (TPs). This study focuses on detailed modelling of the degradation and transport of pesticides with different physico-chemical properties. The objective is to analyse complex hydrological transport processes, to understand the temporal and spatial dynamics of the degradation and transport of pesticides. The ecohydrological model SWAT+ simulates hydrological processes as well as agricultural management and pesticide degradation and can therefore be used to develop pesticide loss reduction strategies. This study focuses on modelling of three pesticides (pendimethalin, diflufenican, and flufenacet), and two TPs, flufenacet-oxalic acid (FOA) and flufenacet sulfonic acid (FESA). The study area is a 100-hectare farmland in the northern German lowlands of Schleswig-Holstein that is characterised by an extensive drainage network of 6.3 km and managed according to common conventional agricultural practice. SWAT+ modelled streamflow with very good agreement between observed and simulated data during calibration and validation. Regarding pesticides, the model performance for highly mobile substances is better than for non-mobile pesticides. While the transport of the moderately to very mobile substances via tile drains played an important role in both wet and dry conditions, no transport via tile drains was modelled for the highly sorptive and non-mobile pendimethalin. In conclusion, the model can reliably represent the degradation of moderately to very mobile pesticides in small-scale tile drainage-dominated catchments, as well as surface runoff-induced peak loads. However, it has weaknesses in accounting for the subsurface transport of non-mobile substances, which can lead to an underestimation of the subsequent delivery after precipitation events and thus underestimates the total load.

Abstract:No-till and mulching are typical management operations in conservation agriculture (CA). To model pesticide degradation and leaching under a CA scenario, as compared to a conventional-tillage scenario (CT), the mulch module of the agro-hydrological model Daisy was extended. A Daisy soil column was parameterized with measurements of topsoil, mulch, and a realistic subsoil, and tested against published experimental data of pesticide fate in laboratory soil columns covered by mulch. Uncertainty and sensitivity analyses of the new Daisy version were conducted for a series of weather, soil, pesticide, and mulch parameters, using 4939 Monte Carlo simulations under each scenario. Results showed that there was no systematic difference in pesticide leaching from the topsoil (to the subsoil and directly to drains via drain-connected biopores) between CA and CT, but pesticide degradation and sorption were significantly different; degradation in the mulch and uppermost soil surface layer (0–3.5 cm) was larger in CA while degradation was larger in CT when considering the whole topsoil (0–30 cm). This difference for the whole topsoil could be explained by pesticide interception in CA in the part of the mulch not in direct contact with the soil where degradation is assumed not to occur. The sensitivity analysis highlighted non-influential parameters and seven parameters out of twenty-five to be better estimated to improve the accuracy of the predictions.

Abstract:The global agricultural industry requires improved efficacy of sprays being applied to weeds and crops to increase financial returns and reduce environmental impact. Enhancing foliar penetration is one way to improve efficacy. Within the plant leaf, the cuticle is the most significant barrier to agrochemical diffusion. It has been noted that a comprehensive set of mechanisms for ionic active ingredient (AI) penetration through plant leaves with surfactants is not well defined, and oils that enhance penetration have been given little attention. The importance of a mechanistic mathematical model has been noted previously in the literature. Two mechanistic mathematical models have been previously developed by the authors, focusing on plant cuticle penetration of calcium chloride through tomato fruit cuticles. The models included ion binding and evaporation with hygroscopic water absorption, along with the ability to vary the AI concentration and type, relative humidity, and plant species. Here, we further develop these models to include lipophilic adjuvant effects, as well as the adsorption and desorption, of compounds on the cuticle surface with a novel Adaptive Competitive Langmuir model. These modifications to a penetration model provide a novel addition to the literature. We validate our theoretical model results against appropriate experimental data, discuss key sensitivities, and relate theoretical predictions to physical mechanisms. The results indicate the addition of the desorption mechanism may be one way to predict increased penetration at late times, and the sensitivity of model parameters compares well to those present in the literature.
Measuring and preliminary modeling of drift interception by plant species

Abstract:Currently, the concept of plant capture efficiency is not quantitatively considered in the evaluation of off-target drift for the purposes of pesticide risk assessment in the United States. For on-target pesticide applications, canopy capture efficiency is managed by optimizing formulations or tank-mixing with adjuvants to maximize retention of spray droplets. These efforts take into consideration the fact that plant species have diverse morphology and surface characteristics, and as such will retain varying levels of applied pesticides. This work aims to combine plant surface wettability potential, spray droplet characteristics, and plant morphology into describing the plant capture efficiency of drifted spray droplets. In this study, we used wind tunnel experiments and individual plants grown to 10-20 cm to show that at two downwind distances and with two distinct nozzles capture efficiency for sunflower (Helianthus annuus L.), lettuce (Lactuca sativa L.), and tomato (Solanum lycopersicum L.) is consistently higher than rice (Oryza sativa L.), peas (Pisum sativum L). and onions (Allium cepa L.), with carrots (Daucus carota L.) showing high variability and falling between the two groups. We also present a novel method for three-dimensional modeling of plants from photogrammetric scanning and use the results in the first known computational fluid dynamics simulations of drift capture efficiency on plants. The mean simulated drift capture efficiency rates were within the same order of magnitude of the mean observed rates of sunflower and lettuce, and differed by one to two orders for rice and onion. We identify simulating the effects of surface roughness on droplet behavior, and the effects of wind flow on plant movement as potential model improvements requiring further species-specific data collection.
Transport and Environmental Risks of Propachlor Within the Soil-Plant-Water Phase as Affected by Dissolved Organic Matter as a Nonionic Surfactant

Abstract:Propachlor is a widely used acylaniline herbicide for weeding annual gramineous and dicot plants in farmland. As a toxic agricultural chemical, it is overused in crop production and has become one of the most serious environmental pollutants. Yet, understanding the impact of environmental factors on its behavior in farmland soils is critically important for healthy crop production and food safety. In this study, we characterized the effect of dissolved organic matter (DOM) extracted from rice straw on the mobility behavior of propachlor in farmland soil using comprehensive approaches such as the batch equilibrium experiment, soil thin-layer chromatography, soil column leaching, and wheat bioaccumulation with Triton X-100 (TX-100) as a reference surfactant. The application of DOM at 60 and 120 mg DOC L-1 and TX-100 at 96, 192, and 288 mg L-1 reduced the sorption and increased the desorption of propachlor in soil. Freundlich constant K f values (sorption) of propachlor declined with the expansion of DOM and TX-100 concentrations. The addition of DOM and TX-100 increased the mobility of propachlor and the total concentration of propachlor in the leachate of soil columns. The retention factors (R f) were evaluated in the soil thin-layer chromatography as 0.741 and 0.772 (for DOM) and 0.731, 0.763, and 0.791 (for TX-100), all of which were greater than the control (0.710) under the treatment. The application of DOM or TX-100 reduced root growth (biomass) and increased bioaccumulation of propachlor in the roots and shoots of wheat. The outcome of the study can provide important references for the rational use of propachlor and help agronomic management to minimize pesticide contamination in realistic crop production.
Surfactant-induced deposit structures in relation to the biological efficacy of glyphosate on easy- and difficult-to-wet weed species

Abstract:Background: Typical active ingredient (AI) residue patterns are formed during droplet drying on plant surfaces owing to the interaction of spray solution characteristics and leaf micromorphology. Currently, comparatively little is known about the influence of AI deposit patterns within a spray droplet residue area on the penetration and biological efficacy of glyphosate. A scanning electron microscope with energy dispersive X-ray microanalysis has been used to characterise residue patterns and to quantify the area ultimately covered by glyphosate within the droplet spread area. Results: The easy-to-wet weed species Stellaria media L. and Viola arvensis L., as well as the difficult-to-wet Chenopodium album L. and Setaria viridis L., differing in their surface micromorphology, have been used. Rapeseed oil ethoxylates (RSO 5 or RSO 60) were added to glyphosate solutions to provide different droplet spread areas. Addition of RSO 5 enhanced droplet spread area more than RSO 60, and both caused distinct glyphosate residue patterns. The biological efficacy of treatment solutions showed no significant correlation with the area ultimately covered by glyphosate. Conclusion: The results have implications on herbicide uptake models. This study shows that droplet spread area does not correspond to the area ultimately covered by glyphosate, and that the latter does not affect glyphosate phytotoxicity.

Abstract:Glucagon and adrenaline exert their action upon the liver via the cyclic AMP synthetizing system located in the plasma membrane. The enzyme adenylate cyclase is further regulated by guanyl nucleotides. It has been recently shown that the rat liver plasma membrane system could respond to GTP by simultaneous increase in the cyclase activity in response to glucagon and by the dissociation of this hormone from its binding sites (1). Unambiguous relationship between the activating effect of GTP upon the cyclase and its action upon glucagon binding has not been determined yet (2). This problem was approached using the in vitro action of epinephrine as a model. When 1 to 100 muM GTP or DGP were added to rat liver plasma membranes isolated from adrenalectomized animals, they increased markedly the response of the cyclase system to epinephrine. These effects could be observed in the absence of an ATP-regenerating system and were mimicked by 5'-guanylyl diphosphonate; GTP and GDP were the most active compounds followed by ITP, CTP and by a series of guanyl derivatives. UTP, as well as guanosine, GMP, cyclic GMP and ppGpp were inactive. Guanyl nucleotides did not increase the affinity of the cyclase system for the activating hormones, but enhanced the affinity for ATP-Mg and also the Vmax of the reaction. Finally, GTP, ATP, CTP, UTP but not GDP displaced epinephrine bound to plasma membranes by a mere chelation phenomenon. It is concluded that 1) guanyl nucleotides do not act primarily by influencing the binding of hormones to the membranes; 2) they act directly upon the catalytic subunit of the cyclase; 3) the low concentrations of GTP required for its action strongly suggest that this nucleotide plays a role in the physiological regulation of the intrahepatic cyclic AMP level.

Abstract:A dynamic model of dynamiCROP was applied to study environmental fate and behavior of four herbicides in wheat including florasulam, carfentrazone-ethyl, fluroxypyr-meptyl, and fluroxypyr. Meantime, their residue in wheat and dissipation half-lives in plant determined by field trials using QuEChERS liquid chromatography tandem mass spectrometry were used to verify modelling results. The combination of experimental verification and modelling prediction deciphered the fate of four pesticides in wheat field ecosystem. Besides, temperature difference of 3 °C only resulted in lower than 15% half-life difference. By quantifying the contribution of temperature, the predominant role of rain on pesticide dissipation was highlighted for the first time, namely higher precipitation leaded to faster degradation and vice versa.
Transformation, leaching and plant uptake simulations of 6:2 and 8:2 polyfluoroalkyl phosphate diesters (diPAPs) and related transformation products under near-natural conditions

Abstract:In response to the growing concern over PFAS contamination, employing models to simulate PFAS behavior in the environment becomes necessary. This facilitates evaluating risks tied to leaching into groundwater, adsorption in soil, plant uptake, entry into the food chain, and the conversion of precursors into persistent PFAS. We utilized the MACRO model to simulate the behavior of the precursors 6:2 diPAP and 8:2 diPAP using data from a 2-year lysimeter experiment, key compound parameters were optimized via the caRamel evolutionary algorithm. We assumed that the transformation of both diPAP precursors into stable PFAAs is influenced by temperature and soil moisture, similar to pesticide degradation by microorganisms. Results reveal that the model accurately represents transformation, leaching, soil retention, and plant uptake of diPAP and transformation products. A comparison with a lab-based soil column study supports the slower natural degradation of precursors, affirming our modeling approach. Temperature and soil moisture could indicate that a worst-case scenario for transformation product leaching into groundwater could occur during a mild summer with moderate evapotranspiration and heavy rainfall. Plant uptake involves multiple elements: PFAS availability in the root zone depends on prior degradation or presence. Increased moisture in the root zone favors PFAS uptake combined with temperatures high enough for prior biotransformation. The calculation of temperature and moisture-based conversion rates was adopted directly from MACRO. It is recommended to further investigate these effects to validate and possibly modify them.
