Abstract:Gellan, an exopolysaccharide produced by Sphingomonas paucimobilis ATCC 31461, is used in the food, pharmaceutical, and other industries. During the fermentative production of gellan, strain ATCC 31461 also accumulates large amounts of yellow carotenoid pigments, thereby increasing processing costs. A carotenoid-free mutant strain of ATCC 31461 was isolated by ethyl methanesulfonate mutagenesis combined with ultraviolet irradiation. A new gellan purification method which reduced the consumption of isopropanol was developed based on this mutant strain. The final gellan production of the mutant strain was 13% higher than that of the parent strain. The glycerate and acetate substituents of gellan produced by the mutant strain were 17% and 65% higher than those produced by gellan from ATCC 31461, respectively. Stability tests suggested that the broth produced by the mutant strain was more stable during storage at room temperature compared with that produced by the parent strain. The mutant strain seems to be an ideal strain for use in the commercial production of gellan.

Abstract:To overcome the drawbacks of dehydration and low gel strength of the gels of L-arginine-treated konjac glucomannan (KGM), thermally irreversible gel was prepared by L-arginine induced KGM-modified starch mixed sol. The effects of the three modified starches (corn modified starch, acetylated bis-starch adipate, and hydroxypropyl distarch phosphate) on the mixed system were investigated by texture analysis, scanning electron microscopy, Low field NMR, Fourier transform infrared spectroscopy and Differential thermal analysis. The results showed a gradual increase in hardness with increasing concentration of modified starch. Differential thermal results indicated that acetylated bis-starch adipate composited gels had the highest thermal decomposition temperature (309 C). The changes in the textural properties and thermal stability of the three mixed gels were mainly attributed to the enhanced hydrogen bonding between starch/KGM/L-arginine, and changes in the microstructure. When the starch concentration was higher, the denser the gel network structure was, the more difficult it was for water trapped in the gel system to precipitate out. The results indicated that acetylated bisstarch adipate was more suitable to be added to the mixed system to prepare thermally irreversible gels.

Abstract:The effect of culture medium compositions on gellan gum production produced by fermentation with a halobacterium Sphingomonas paucimobilis QHZJUJW CGMCC2428 was studied. In this work, a fractional factorial design was applied to investigate the main factors that affected gellan gum production by S. paucimobilis QHZJUJW CGMCC2428. Sucrose was the best carbon source for gellan gum and peptone displayed better inducing effect. Central composite design and response surface methodology were adopted to derive a statistical model for optimizing submerged culture medium composition. These experimental results showed that the optimum culture medium for producing gellan gum was composed of 40.00 (w/v) sucrose, 3.00% peptone (w/v), MgSO4 (w/v), 9.20% KH2PO4 (w/v), 7.50% Na2HPO4 (w/v), 4.30% K2SO4 (w/v), pH 6.8-7.0. The maximal gellan gum was 19.89 +/- 0.68 g/L, which was agreed closely with the predicated value (20.12 g/L). After incubated for 72 h under the optimized culture medium in 5-L bioreactor, the gellan gum fermentation reached about 19.90 +/- 0.68 g/L, which was higher than that in the initial cultivation medium. The fabrication and characterisation of surimi gel modified by konjac glucomannan with the different hydrolysis degree

Abstract:The gel performance of surimi-based products is indispensable technological attribute responsible for their high popularity. Here, surimi gels with tunable gel properties were successfully fabricated based on the double network theory. Konjac glucomannan (KGM) hydrolysed for various periods (0, 5, 15, 30, 60 and 90 min) was introduced into surimi systems. Corresponding structural characterisations including FTIR, XRD, SDS-PAGE and grafting degree assessment, were conducted to confirm the presence of double crosslinking reactions. Furthermore, the gel strength, dynamic rheology, thermal stability and in vitro digestibility of the surimi gels were investigated. Red shifts in the amide A band and an increase in relative intensity indicated that hydrogen bonding existed in the complex systems, according to the FTIR spectra. The XRD patterns suggested that the crystalline structures were weakened by interfering with the protein chain arrangement due to the Maillard reaction. In addition, SDS-PAGE and o-phthalaldehyde (OPA) assays also confirmed that myofibrillar protein and hydrolysed KGM were partially linked by covalent bonding. The SK-2 group showed the greatest mechanical strength and lowest digestibility amongst all groups due to its dense hierarchical structure. This work may provide inspiration for the design and development of surimi gels with improved gel properties via microarchitectural modulation.

Abstract:To explore greener modifier of konjac glucomannan, a new type of thermal irreversible gel using konjac glucomannan (KGM) and basic amino acids (L-lysine, and L-arginine) was prepared in the present study. The effects of different concentrations (1%, 2%, 3%, 4%, and 5%) of these two basic amino acids on the structure and properties of KGM hydrogels were mainly studied. The results showed that there was a better texture characteristic of composite gel as the concentration of amino acids was above 4%. Besides, with the increase in amino acid concentrations, the pore size of composite gel was denser and the syneresis of composite gel was more obvious, respectively. The rheological results indicated that the storage modulus (G ') was influenced by the concentration of amino acids and temperature. It was found that the two basic amino acids removed the acetyl group of KGM molecule and changed the original crystalline domain of KGM as evidenced by FT-IR and X-ray diffraction (XRD) analysis. Furthermore, compared to L-lysine, L-arginine could exert a greater effect on the KGM gel. In conclusion, the two basic amino acids (L-lysine, and L-arginine) could remarkedly improve the structure and properties of natural KGM and could be used as modifiers of the KGM hydrosol.
Evaluating Physicochemical and Sensory Properties of Functional Yogurt Supplemented with Glycyrrhiza Polysaccharide As Potential Replacement for Gelatin

Abstract:Glycyrrhiza is a well-known plant resource used for the production of extractum-glycyrrhizae; however, large amounts of Glycyrrhiza residues containing polysaccharides are produced, and these have not been well utilized until now. The aim of this study was to evaluate the Glycyrrhiza polysaccharides obtained from Glycyrrhiza residues as a potential gelatin replacer in yogurt. The incorporation of Glycyrrhiza polysaccharide (GP) at different concentrations accelerated the growth of lactic acid bacteria. Similar to the effect of adding gelatin (GE), GP could suitably improve the water-holding capacity (WHC) and texture of the yogurt. Moreover, the yogurt prepared with GP showed a higher viscosity and shorter transverse relaxation time of mobile water (T23) value than the control group (CG). Moreover, the microstructure analysis indicated that the casein network of yogurt with GP was more compact and cohesive than those of others. Yogurt prepared with 0.1% GPs exhibited the best sensory acceptance. The results indicated that small amounts of GPs can effectively replace gelatin as a thickener in yogurt with good quality.
Effects of gellan gum and inulin on mixed‐gel properties and molecular structure of gelatin

Abstract:Gellan gum (GG) is often added to gelatin (GL) to improve the gel property. GG‐based or inulin (IL)‐based hydrogels were developed. Rigid and brittle gels or smooth and delicate gels were prepared with GG and IL, respectively. This study aimed to explore the properties and interaction mechanisms of the mixed‐gel system containing GL, GG, and IL, in which different ratios of GG‐IL (0.4%) (10:0, 8:2, 6:4, 5:5, 4:6, 2:8, and 0:10) were added to GL (6%). Texture profiles, rheological properties, water mobility, intermolecular forces, circular dichroism (CD) spectra, and microstructures were analyzed. The results showed that addition of GG‐IL could improve the hardness, chewiness, and cohesiveness of mixed‐gel, besides maintaining appropriate springiness. Water mobility of the mixed‐gel decreased while viscoelasticity increased upon the addition GG. At GG:IL = 2:8, the melting temperature of mixed‐gel was far higher than that of GL gel itself. The GL‐GG‐IL gel showed decrease in nonspecific bonding and increase in hydrogen bonding compared with the GL gel. CD spectra indicated the promotion of GL unfolding by GG, hence suggesting the binding of GG to GL; binding ability was better at GG:IL >5:5. Cryo‐SEM provided evidence for the formation of cross‐linked network within GL‐GG‐IL. Overall, we concluded that addition of GG‐IL to GL system would be most suitable for improving the properties of mixed‐gel. This finding may be potentially applicable in the further development of gel food products, such as meat jellies and gummy jellies.

Abstract:Gellan gum is an important exopolysaccharide produced with Sphingomonas paucimobilis,which is widely used in foods,pharmaceutical industries.The fermentation kinetic model of cell growth,gellan gum production and substrate utilization were established and the kinetic parameters of the model were determined.The kinetic model provides a good description of gellan gum fermentation,in which the deviations between model calculating results and experimental data are within 5%.Furthermore it shows a good prediction of the fermentation profile including substrate consumption,cell growth and gellan gum production under different initial glucose mass concentration.The deviation between actual values and predicted values is less than 10%,which indicates that the model constructed has good applicability and significantly instructive meaning to industrial-scale gellan gum fermentation.
Effects of carbon/nitrogen ratio, dissolved oxygen and impeller type on gellan gum production in Sphingomonas paucimobilis

Abstract:Among exopolysaccharides, gellan gum is used widely as a gelling agent and food additive, and its commercial market is rapidly increasing. To optimize the fermentation production of gellan gum in Sphingomonas paucimobilis ZJU3108, carbon/nitrogen (C/N) ratio, dissolved oxygen (DO) and impeller type were studied. The C/N ratio, rather than the absolute concentration of carbon or nitrogen source, was found to be the major factor affecting cell growth. A C/N ratio above 20.7 had no significant effect on gellan gum production. Although the final yield of gellan gum increased with C/N ratio increase, the glucose to gellan gum conversion rate decreased. This was due to the high viscosity, which caused oxygen limitation from 16 h to 40 h in the inhomogeneous broth. Due to low stationary phase oxygen demand, the effect of DO on biomass and gellan gum production was reduced. A simple DO control strategy was developed to maintain 30% DO for the first 32 h cultivation, followed by anaerobic conditions for the remaining 8 h. Comparison of different impellers suggested that an anchor impeller with impeller diameter of 11.0 cm (D* = 11.0 cm) was suitable for high yields of high molecular weight gellan gum. These studies simplify production of gellan gum by fermentation and improve its yield.

Modulation of acid-induced pea protein gels by gellan gum and glucono-δ-lactone: Rheological and microstructural insights

Abstract:This study investigated the effect of gellan gum (GG) and glucono-δ-lactone (GDL) on the acid-induced gel properties of pea protein isolate (PPI) pretreated with media milling. The inclusion of GG substantially enhanced the gel hardness of PPI gel from 18.69 g to 792.47 g though slightly reduced its water holding capacity (WHC). Rheological analysis showed that GG increased storage modulus (G') and decreased damping factor of gels in the small amplitude oscillatory shear region and transformed its strain thinning behavior into weak strain overshoot behavior in the large amplitude oscillatory shear region. SEM revealed that GG transformed the microstructure of gel from a uniform particle aggregate structure to a chain-like architecture composed of filaments with small protein particles attached. Turbidity and zeta potential analysis showed that GG promoted the transformation of PPI from a soluble polymer system to an insoluble coagulant during acidification. When GG content was relatively high (0.2 %-0.3 %), high GDL content increased the electrostatic interaction between PPI and GG molecules, causing their rapid aggregation into a dense irregular aggregate structure, further enhancing gel strength and WHC. Overall, GG and GDL can offer the opportunity to modulate the microstructure and gel properties of acid-induced PPI gels, presenting potential for diversifying food gel design strategies through PPI-GG hybrid systems.

Abstract:A series of four different low molecular weight gellan gum products was obtained by alkaline hydrolysis with the aim to investigate the impact of the molecular weight on the rheological properties of the polysaccharide aqueous dispersions and on the physicochemical characteristics of derived ionotropic crosslinked hydrogels. In particular, thermo-rheological analysis was conducted on aqueous dispersions to study the influence of molecular weight on the thermogelation properties typical of the native polysaccharide while strain sweep experiments were conducted to establish if aqueous dispersion shows a viscoelastic behavior. The effect of different Ca2+ on the rheological properties of hydrogels were studied. Furthermore, ionotropic crosslinked hydrogels were analyzed in terms of morphology on the dried state and swelling behavior, while their viscoelastic properties were studied by means of rheological analysis conducted in frequency sweep regime after different time points of incubation in phosphate buffer at pH 7.4. Release experiments conducted using fluorescein isothiocyanate labelled dextran as a model diffusion agent and was performed to investigate the possibility of using the low molecular weight GG-derived hydrogels as an active molecule-releasing device. Finally, the cytocompatibility of hydrolysis products was investigated, as well as the capacity of hydrogels to encapsulate viable MC3T3-E1 preosteoblastic cells.

Abstract:The objectives of this study were to create composite hydrogels from gellan, alginate, and xanthan gums and study their functional properties. The characteristics of gellan (1.25–0.05%), gellan-alginate (1: 0, 0.25: 0.75, 0.5: 0.5, 0.75: 0.25 and 0:1), and gellan (0.5%)-alginate(0.5%)-xanthan (0.1, 0.15 and 0.3%) hydrogels were investigated. The results showed the gellan hydrogel structure had dense mesh and small pore size compared to alginate gel. With the combination of gellan and alginate, the pore size of network and swelling index increased. Tri-polymeric hydrogels created a network with bigger pore size, which improved the swelling capacity of the hydrogel. But, the swelling index decreased with increasing xanthan gum concentration from 0.1% to 0.3% (w/w). The gellan-alginate gel had more hardness and consistency value compared to the mono-polymeric and tri-polymeric hydrogels. Thermal stability of hydrogels increased by the combination of gums and FTIR confirmed the interaction of the polymers. As a result, multi-polymeric hydrogels of alginate-gellan-xanthan can create new structures with more swelling ability, consistency, and thermal stability for use in the nutraceutical delivery and restructuring of foods.

Abstract:Gellan gum is a water-soluble exopolysaccharide, it has applications in the food, pharmaceutical and chemical industries. In this study, a gellan gum producing strain was isolated from rice root, and this strain was identified be the species of Sphingomonas azotifigens. The Plackett-Burman design was applied to investigate the main factors affecting gellan gum production by S. azotifigens GL-1 in a molasses and cheese whey based medium; the medium compositions were optimized by response surface methodology. The optimum cheese whey based medium consisted of cheese whey 68.34 g/L, Na2HPO4 14.58 g/L and KH2PO4 7.66 g/L, and the maximum gellan gum production that using this medium was 33.75 ± 1.55 g/L. 14.75 ± 0.65 g/L gellan gum was obtained with an optimized molasses medium, which consisted of molasses 50 g/L, Na2HPO4 9.71 g/L and KH2PO4 5.92 g/L. The molecular weight of gellan gum obtained from two medias were 1.06 × 106 and 0.89 × 106 Da, respectively. The cheese whey-derived gellan gum showed a higher rhamnose, lower glucuronic acid and higher glycerate content compared to the molasses-derived gellan gum. S. azotifigens GL-1 has a high gellan gum production capacity in a cheap medium suggesting it has great potential as an industrial gellan gum producer.

Abstract:A novel composite hydrogel incorporated physical-chemical treated insoluble (ISF) into high acyl gellan (HAG) was fabricated and characterized. ISF-HAG hydrogel exhibited better appearance and denser network structure compared to HAG hydrogel (control sample), which was further manifested by higher gel strength and storage module G′. It was attributed to hydrogen bonds between ISF and HAG, confirmed by FT-IR analysis, as well as ISF as a supporter or filler in the network pores. Temperature sweeps showed that the gel point temperature and gelling rate of ISF-HAG hydrogel were higher than those of control sample by a maximum of 5 °C and 5.22 Pa/°C, respectively. ISF-HAG hydrogel also presented good swelling capacity and texture properties. USAT-ISF obtained by ultrasonic assisted steam-cooking alkaline treatment exerted a great impact on the formation and performance of HAG hydrogel due to loose structure and outstanding physicochemical properties (water holding capacity, swelling capacity, etc.) This study explores the potential of ISF to promote gelation and expands the range of applications for HAG hydrogel.

Abstract:Microscopes using non-linear excitation of chromophores with pulsed near-IR light can generate highly localized foci of molecules in the electronic singlet state that are concentrated in volumes of less than one femtoliter. The three-dimensional confinement of excitation arises from the simultaneous absorption of two IR photons of approximately half the energy required for linear excitation. Two-photon microscopy is especially useful for two types of interrogation of neural processes. First, uncaging of signaling molecules such as glutamate, as stimulation is so refined it can be used to mimic normal unitary synaptic levels. In addition, uncaging allows complete control of the timing and position of stimulation, so the two-photon light beam provides the chemical neuroscientist with an "optical conductor's baton" which can command synaptic activity at will. A second powerful feature of two-photon microscopy is that when used for fluorescence imaging it enables the visualization of cellular structure and function in living animals at depths far beyond that possible with normal confocal microscopes. In this review I provide a survey of the many important applications of two-photon microscopy in these two fields of neuroscience, and suggest some areas for future technical development.
Gellan gum gel as entrapment matrix for high temperature fermentation processes: A rheological study

Abstract:High temperature rheological measurements (60–80°C) were carried out on gellan gum gel. The experiments were conducted in order to optimize the behavior of the gel as an entrapment matrix for fermentation processes of dairy fluids using highly thermophilic microorganisms. At 60°C, no significant decrease of rheological parameters of the gel was observed for 15 days, as compared to 23°C. At 80°C, the gel weakened abruptly. The gel was extremely sensitive to calcium loss (optimum concentration 0.1% CaCl2) due to chelating agents. Gellan gum gel could be an efficient entrapment matrix for fermentation processes at temperature up to 60°C in dairy fluids.

Abstract:In this study, we investigated the gellan gelation mechanism induced by Glucono-δ-lactone (GDL) addition, and subsequently we further fabricated a series of thermally irreversible emulsion gels with the GDL-induced gel matrix as the continuous phase and the methyl cellulose (MC) stabilized emulsion as the dispersed phase. The results showed that GDL-induced gellan gelation was both a time- and temperature-dependent process, which was similar to the gelation process of NaCl-induced gellan gels, but with a higher temperature-dependence. Moreover, GDL-induced gel had a significantly higher rupture strength and the resulting gel did not melt on heating, meaning that the GDL-induced gellan gel can be deemed thermally irreversible. This is because decreasing pH is commonly more effective in promoting gellan gelation. Besides, by mixing the MC-stabilized emulsion with a hot gellan sol, followed by adding GDL, we fabricated the emulsion gels with a wide range of oil phase fraction (0–30%). The emulsion gels exhibited good thermal stability, without melting and oil droplet coalescence observed when heating the gels in a boiling water bath for 30 min. Conclusively, our results may deepen the understanding on gellan gelation induced by GDL and also broaden the utilization of gellan in building gel-related food structures.

Abstract:Breast cancer is the most frequent type of cancer among women worldwide. Vascular endothelial growth factor (VEGF), the key modulator of angiogenesis, has been implicated in breast cancer susceptibility and aggressiveness. VEGF expression was determined in 99 breast cancer tissue samples using reverse transcription-polymerase chain reaction and the human epidermal growth factor receptor 2 (HER2) status was determined by immunohistochemistry. Subsequently, the associations of VEGF, HER2 and hormone receptor status with clinicopathological data were evaluated. High VEGF expression was found to be significantly correlated with the presence of lymphovascular invasion. In hormone receptor-positive/HER2-positive, HER2-positive and triple-negative breast cancer, high VEGF expression was correlated with the presence of axillary nodal metastasis and lower overall survival rates. Therefore, the assessment of the VEGF status along with the hormone receptor and HER2 status may help identify high-risk patients who may benefit from anti-VEGF treatment.
Effect and characterization of konjac glucomannan on xanthan gum/κ-carrageenan/agar system

Abstract:A mixed polysaccharide system is an important strategy to improve the performance of a single polysaccharide. Herein, quaternary polysaccharide gels were prepared by konjac glucomannan (KGM), xanthan gum (XG), κ-carrageenan (κ-CA), and agar (AR). The effects of KGM were evaluated by combining water holding capacity (WHC), rheological analysis, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), and texture profile analysis (TPA). More KGM gradually increased the springiness of the compounded gels. WHC increased and then decreased with the addition of KGM, performing best at KGM4 (KGM: κ-CA:XG:AR = 2:2:1:2). Rheological analysis showed that the compounded gels exhibited a pseudoplastic characteristic of shear thinning, KGM endowed the gel with a stronger shear thinning behavior and improved the solid-like nature of the gels at high temperatures. The thermal stability of the composite gel was improved by the participation of KGM. FTIR analysis showed that the interactions were mainly related to intermolecular hydrogen bonds and acetyl groups. The microscopic morphology of KGM4 was significantly continuous, smooth, and compact, exhibiting the best practical performance and taking the maximum advantage.

Abstract:In this study, bacterial cellulose was synthesized by Taonella mepensis from traditional Chinese medicinal herb residues hydrolysate. To overcome the inhibitory effect of fermentation environment, in-situ fermentation with gellan gum adding was carried out for the first time. After ten days' static fermentation, both high-acyl gellan gum and low-acyl gellan gum adding showed certain beneficial effects for bacterial cellulose production that the highest bacterial cellulose yield (0.866 and 0.798 g/L, respectively) was 59% and 47% higher than that (0.543 g/L) without gellan gum adding. Besides, gellan gum based bacterial cellulose showed some better texture characteristics. Gellan gum was loaded in the nano network of bacterial cellulose, and gellan gum adding had some influence on the crystal structure and thermal degradation behaviors of bacterial cellulose but affected little on its functional groups. Overall, this in-situ fermentation technology is attractive for bacterial cellulose production from low-cost but inhibitory substrates.
