Abstract:Anisopteromalus calandrae (Howard) shows great promise as an ectoparasitoid for controlling various coleopteran pests in warehouses. However, for a large-scale release, it is crucial to establish an ample supply of A. calandrae while carefully maintaining their quality and effectiveness. Appropriate cold storage techniques are the key to achieving these goals. Previous studies on cold storage have focused on specific developmental stages and explored cold storage conditions that can be applied only to those stages. Herein, we examined the development, survival and reproductive capacity of A. calandrae at different temperatures (13, 16, and 19 °C) and storage durations (30, 60, and 90 d) and evaluated the fitness of the offspring. A. calandrae completed its egg-to-larva development and pupated at 16 °C, but its development was arrested at an early pupal stage. Even after 90 d of cold storage at 16 °C, the survival rate of A. calandrae remained high at 77%, with no significant impact on reproductive capacity. Furthermore, cold storage showed no negative effect on the F1 generation. In contrast, eggs stored at 13 °C failed to hatch, whereas those stored at 19 °C developed. Adults emerged after > 60 d. This indicates that storage at 19 °C is only suitable for short durations. Our findings highlight the developmental pattern of A. calandrae at 16 °C, indicating that the parasitic wasp can be stored for a long time at this temperature across all stages of development before pupation, substantially facilitating its mass reproduction and industrial production.
Optimal cold storage protocol for Anisopteromalus calandrae (Howard) (Hymenoptera: Pteromalidae), a biological control agent for coleopteran pests in stored products

Abstract:Background: Anisopteromalus calandrae (Howard) is a solitary ectoparasitoid with wide-ranging potential applications as a natural biological control agent against various coleopterous pests in food warehouses. Implementing an effective cold storage program is crucial for extending the shelf life of biological control agents and ensuring their stable and abundant supply. Herein, we attempted to determine the optimal cold storage conditions for Anisopteromalus calandrae by investigating the effect of cold storage at three different temperatures (7, 13, and 19 °C) for 7, 21, and 35 days on four developmental stages (late-instar larvae, early-stage pupae, mid-stage pupae, and 2-day-old adults). Additionally, we explored the maximum cold storage potential by observing early-stage pupae stored at 13 °C for various durations (30, 60, 90, 120, and 150 days). Results: The most suitable cold storage temperature for the early-stage pupae of Anisopteromalus calandrae was 13 °C, and the highest adult emergence rate (98.3%) was after 90 days of storage at 13 °C. Furthermore, we did not find any significant effect on longevity (female: 44.3 days; male: 38.1 days) or fecundity (121.7 wasps). The female ratio ranged from 43.5% to 50.8%. More importantly, cold storage did not adversely affect the developmental duration or fecundity of the offspring. Conclusion: This study offers crucial insights for managing Anisopteromalus calandrae populations under laboratory conditions and lays the foundation for potential industrial production and development. © 2023 Society of Chemical Industry.

Abstract:The Cnaphalocrocis medinalis (Guenee) is one of the most important rice pests in Asia and is difficult to control by chemical insecticides due to its rapid development of resistance. To screen potential species for biological control of C. medinalis, we investigated the effects of temperature (20, 24, 28, 32, and 36 degrees C) and host age (1-, 2-, 3-, and 4-d-old) on the fecundity of four Trichogramma spp. on C. medinalis eggs. Our results indicated that C. medinalis eggs were acceptable to T. japonicum, T. chilonis, T. dendrolimi, and T. ostriniae, though no eggs were parasitized at 36 degrees C. There were no significant differences in parasitism among the four Trichogramma species under the tested temperature regions, except at 20 degrees C where parasitism by T. japonicum was significantly higher than that by T. chilonis and T. ostriniae. However, T. japonicum had significantly more progeny than the other three Trichogramma species at 32 degrees C. All four Trichogramma species performed well on 1-, 2-, and 3-d-old C. medinalis eggs, but parasitism on 4-d-old eggs was significantly reduced. Trichogramma japonicum parasitized the highest number of C. medinalis eggs on different aged hosts and had more progeny than the other Trichogramma species, especially on 3-d-old hosts. In conclusion, T. japonicum exhibited better performance on C. medinalis eggs than the other three Trichogramma species and could be considered as our most suitable Trichogramma candidate for control of C. medinalis.

Abstract:Psyttalia incisi (Silvestri) is the dominant parasitoid against Bactrocera dorsalis (Hendel) in fruit-producing regions of southern China. Prior to a large-scale release, it is important to generate a sufficient stockpile of P. incisi whilst considering how best to maintain their quality and performance; cold storage is an ideal method to achieve these aims. In this study, the impacts of temperature and storage duration on the developmental parameters of P. incisi pupae at different age intervals were assessed. Then, four of the cold storage protocols were chosen for further evaluating their impacts on the quality parameters of post-storage adults. Results showed that the emergence rate of P. incisi was significantly affected by storage temperature, storage duration, and pupal age interval and their interactions. However, when late-age P. incisi pupae developed at a temperature of 13 °C for 10 or 15 d, no undesirable impacts on dry weight, flight ability, longevity, reproduction parameters of post-storage adults, emergence rate, or the female proportion of progeny were recorded. Our findings demonstrate that cold storage has the potential for enhancing the flexibility and effectiveness of the large-scale production and application of P. incisi.

Abstract:Cold storage can extend shelf life of parasitoids for use in biocontrol. However, cold storage may have negative impacts on life history traits of the parasitoids and, therefore, on their performance as biocontrol agents. Here, we examine the effect of cold storage on life history traits of Habrobracon hebetor (Say) (Hymenoptera: Braconidae), a parasitoid of several economic lepidopteran pests. Newly emerged wasps were stored at three constant temperatures (3°C, 5°C, 7°C) for up to 4 wk. Both temperature and exposure time significantly affected longevity, parasitism, fecundity, and sex ratio. Significant reduction in longevity was observed at 3°C and 7°C, whereas longevity of wasps stored at 5°C remained stable up to the second week and then gradually decreased in Weeks 3 and 4. Parasitism rate also significantly decreased after cold storage at 3°C, 5°C, and 7°C (ranked from high to low). Fecundity decreased at T 3°C and T 5°C but this trait was not affected by storage at T 7°C. A significant shift in male production was observed at T 5°C in Week 3, but in Week 4, the only treatment with male biased reproduction was T 3°C. These results show that the effect of temperature and exposure time in cold storage is trait dependent. Overall, storage at 5°C for a period of 3 wk least impacted most life-history traits of H. hebetor wasps.

Abstract:The egg parasitoids of the genus Trichogramma are important potential biological control agents for a wide range of lepidopteran pests. Cold storage of host eggs has been proposed as a valuable technique for ensuring the release of sufficient parasitoid numbers whenever it is needed. In this context, the impact of low temperatures to induce quiescence in T. evanescens Westwood and T. chilonis Ishii was studied using eggs of Indian meal moth Plodia interpunctella (Hübner). Prepupae of the parasitoids were stored for 15, 30, 45, 60 and 75 d at 4 °C, following a 7 d period of acclimation at 10 °C. Both parasitoid species seem to survive unfavorable temperature conditions by entering a state of quiescence. Parasitism, adult emergence, sex ratio and progeny quality were not affected by cold storage in either parasitoid species for up to 30 d of storage. Parasitized host eggs of P. interpunctella can be stored for up to 60 d at 4 °C for both parasitoids, but there was no emergence at 75 d. General productivity values gradually decreased as the duration of storage lengthened for both species. Our results clearly reveal that the eggs parasitized by these species can be stored for up to 30 d at 4 °C in a state of quiescence without much loss of their performance compared to the control eggs.

Abstract:BACKGROUND:Trichogramma dendrolimi Matsumura is an important egg parasitoid that can be used in the biological control of lepidopteran pests. The effects of storage temperature, developmental stage and storage duration on development and survival were investigated to determine the optimum storage conditions for rearing T. dendrolimi in vitro.RESULTS:The three investigated factors significantly impacted all the biological parameters except for parental G1 and offspring G2 sex ratios. The responses of in vitro reared T. dendrolimi to low temperatures differed depending on developmental stage and storage duration. Comparison of life table parameters under different storage treatments with the control group indicated that storing prepupae at 13 °C for 2, 3 or 4 weeks positively impacted the growth capacity of the parasitoid.CONCLUSION:In vitro reared T. dendrolimi can be reared following cold storage. The quality of the parasitoid was strongly affected by cold storage, likely due to the negative effects of storage on the artificial medium. Storage of the prepupal stage at 13 °C for 4 weeks is optimum for in vitro reared T. dendrolimi. Trichogramma reared in vitro were able to parasitize natural eggs after storage and, thus, have potential for use as biological control agents in the field. © 2018 Society of Chemical Industry.

Abstract:The Asiatic apple leaf miner, Phyllonorycter ringoniella (Matsumura), is an important insect pest to apples. We developed a rearing method for P. ringoniella in the laboratory. Overwintering pupae were collected from our apple orchard, and crabapple seedlings were selected as oviposition substrate and food source for the larval development. The mean developmental period was 25.9 ± 0.49 days from egg to adult and the survival rate was 0.387 under 25 ± 1 °C, 65 ± 5% RH and a photoperiod of 14:10 (L:D) h. Mean egg length and egg width were 0.336 ± 0.0043 and 0.259 ± 0.0046 mm, respectively. Mean body length and head width increased from 1.070 ± 0.0245 and 0.180 ± 0.0021 mm in first instar larvae to 5.027 ± 0.0718 and 0.321 ± 0.0021 mm in fifth instar larvae, respectively. The mean weight of the pupae was 0.946 ± 0.0132 mg. The wingspan of male adult (6.280 ± 0.0639 mm) was significantly larger than female (6.040 ± 0.0753 mm). The mean fecundity (56.9 ± 8.58 eggs per female) and longevity (8.2 ± 0.55 days) of females was highest when they were provided with 10% honey solution, followed by 10% sugar, water, and control. The females preferred to lay eggs (56.5 ± 3.84%) near the secondary leaf vein in a leaf. The larval mortality increased linearly (R2 = 0.94) with the number of larvae per leaf. The mortality of pupae increased from 5.6 ± 4.01 to 51.1 ± 9.88% as storage periods at 4 °C increased from 0 to 105 days. The rearing method and its biological characteristics presented in this study will contribute to further studies on this pest insect.

Abstract:The eggs of pentatomid species were evaluated to parasitism by Trissolcus basalis (Wollaston) and Telenomus podisi Ashmead after storage in liquid nitrogen. Adults which emerged from stored eggs were kept at 18°C for 120 and 180 days to investigate whether adult dormancy could be associated with host egg storage in liquid nitrogen as a method of mass production of these egg parasitoids. Eggs of Nezara viridula (L.) and Acrosternum pengue (Rolston) were successfully parasitized by T. basalis, as well as Piezodorus guildinii (Westwood) and Dichelops furcatus (F.) by T. podisi. The eggs of Edessa meditabunda (F.) were not parasitized by T. basalis. The emergence of T. podisi from eggs of Euschistus heros (F.) and Podisus nigrispinus (Dallas) stored for 6 months was lower than the control. Females of T. basalis and T. podisi that emerged from stored eggs were kept in dormancy at 18°C. Longevity of T. basalis was influenced by the storage time and sex, but not by the interaction of sex and storage time. For T. podisi, longevity was influenced by the storage time, sex, and by the interaction of sex and storage time. For T. basalis, storage in liquid nitrogen did not affect the fecundity of quiescent females, while the number of parasitized eggs by T. podisi decreased after storage. By the joint use of these techniques, it is possible to optimize mass production of T. basalis so that its life cycle can be monitored and synchronized with the life cycle and availability of hosts.

Abstract:An understanding of physiological damage and population development caused by uncomfortable temperature plays an important role in pest control. In order to clarify the adaptability of different temperatures and physiological response mechanism of B. dorsalis, we focused on the adaptation ability of this pest to environmental stress from physiological and ecological viewpoints. In this study, we explored the relationship between population parameters and glucose, glycogen, trehalose, and trehalose-6-phosphate synthase responses to high and low temperatures. Compared with the control group, temperature stress delayed the development duration of all stages, and the survival rates and longevity decreased gradually as temperature decreased to 0 °C and increased to 36 °C. Furthermore, with low temperature decrease from 10 °C to 0 °C, the average fecundity per female increased at 10 °C but decreased later. Reproduction of the species was negatively affected during high-temperature stresses, reaching the lowest value at 36 °C. In addition to significantly affecting biological characteristics, temperature stress influenced physiological changes of B. dorsalis in cold and heat tolerance. When temperature deviated significantly from the norm, the levels of substances associated with temperature resistance were altered: glucose, trehalose, and TPS levels increased, but glycogen levels decreased. These results suggest that temperature stresses exert a detrimental effect on the populations' survival, but the metabolism of trehalose and glycogen may enhance the pest's temperature resistance.

Abstract:Abstract Adult female Panolis flammea were kept at 2 o C either immediately after emergence from the pupae or immediately after mating, for periods ranging from 5 to 20 days and then placed at 15 o C together with a male moth, food supply and suitable oviposition site ( Pinus contorta foliage). Weight loss over the storage period was directly proportional to the time spent at 2 o C irrespective of whether the moths were virgin or mated. Pre‐oviposition period decreased significantly from the control after more than 5 days at 2 o C, but there was no significant difference between late mated and early mated moths. The post‐storage life span of late mated and early mated moths decreased in proportion to the time spent at 2 o C but late mated moths had significantly longer life spans than early mated moths. Moths mated prior to storage were significantly less fecund than moths mated after storage except those moths kept at 2 o C for 20 days where the fecundities of both treatments were identical (c. 45 eggs/female). These results are discussed in relation to the conditions likely to be experienced by P.flammea in the field and their bearing on the population dynamics of this insect.

Abstract:Aphid-pathogenic fungus, Pandora neoaphidis, grown on broomcorn millet possesses greater sporulation capacity (C(s)) than aphid cadavers. The most sporulating cultures (32.0x10(4) spores millet(-1) grain) with water content (C(w)) of 48.7% were prepared by incubation at 20 degrees C for 15 days and used to study the effect of temperature and humidity on C(s) during long-term storage. Cultures were sealed with paper to retain ambient humidity, with parafilm for saturated humidity, or kept in 85% and 98% RH chambers. The C(w) and C(s) were monitored during 200-day storage at 5-20 degrees C. The paper-sealed cultures at 5 degrees C, associated with 21-25% of C(w), were best preserved and their 120-day C(s) was similar to that of the fresh cadavers. Consistently or variably high RH at 5 degrees C resulted in significantly higher C(w) and lower C(s) despite longer viability. The regimes at 10 degrees C preserved the cultures for 40 days. The observations fit well to the logistic model C(s)=35.28/{1+exp[-2.36+(-0.003C(w)+0.001C(w)T)t]} (r(2)=0.95) for all regimes of temperature (T) or C(s)=35.55/[1+exp(-2.33+0.001C(w)t)] (r(2)=0.93) at 5 degrees C only. The rate of decline of C(s) of -0.003C(w)+0.001C(w)T or 0.001 C(w) over days (t) highlights the primary effect of C(w). The daily C(s)-decline rates obtained for the best-stored cultures and air-dried cadavers stored at 5 degrees C were surprisingly identical. The results suggest a possible cheap method for preparing and storing large quantities of P. neoaphiodis inocula.

Abstract:The influence of temperature on growth and development of Chelonus pectinophorae Cushman were determined under laboratory conditions. Results indicated that the developmental zero and effective accumulated temperatures of C. pectinophorae was 11.8 ℃ and 276.1 ℃ for egg-larval stage, 13.8 ℃ and 119.9 ℃ for pupal stage. In the range of 20—36 ℃, the life parameters such as the egg-larval duration, pupal weight, pupation rate and emergence rate of C. spectinophorae increased and then decreased with temperature increase. It has the shortest larval duration, the heaviest pupal weight, the highest pupation and emergence rate and fertility at 28 ℃, indicating that the optimum temperature is 28 ℃. These results are of significance for the mass rearing of C. pectinophoraeand biocontrol against pests in the field.

Abstract:To investigate the effect of fly pupae cold storage on the parasitization and reproduction of Nasonia vitripennis. the parasitization rate,parasitoid emergence rate,number of adult parasitoids/host,female rate of progeny parasitoid from cold stored fly pupae were observed in the laboratory. Results indicated the parasitic rate,parasitoid emergence rate,number of adult parasitoids/host,female rate of progeny parasitoid emerged from cold stored fly pupae were below than control pupae significantly. The aforesaid examined indexes were all had negative correlation with the cold storage time. From these results,it is concluded that the cold storage of fly pupae has negative effect on the parasitization and reproduction of N. vitripennis.

Abstract:Abstract Arma custos (Fabricius) (Hemiptera: Pentatomidae) is a natural predator that can control various agricultural and forestry pests. This study aimed to clarify the effects of temperature on the growth, reproduction, and population of the predator and to simulate its population growth. Using the age–stage, two-sex life table method, 18°C, 22°C, 26°C, 30°C, and 34°C were selected as the temperature conditions. A. custos can complete its life cycle at 18°C–30°C, and the developmental duration of each A. custos stage, adult pre-oviposition period, total pre-oviposition period, and the mean generation time (T) were shortened with the increase in temperature. The pre-adult mortality was significantly reduced at 26°C and 30°C. In addition, the fecundity of a single female and the gross reproductive rate were the highest at 30°C. Significant differences were observed in the intrinsic rate of increase (r) and the finite rate of increase (λ) under different temperature conditions, and both reached the maximum at 30°C. Results showed that adult A. custos raised at 26°C had a longer lifespan and the fecundity was higher at 30°C in comparison with the other temperatures. This study is the first to report the life cycle of A. custos at different temperatures, and the results can provide a scientific theoretical basis for the indoor artificial reproduction, outdoor release, and colonization of A. custos.

Effect of storage conditions on the survival of two potential biocontrol agents of nematodes, the fungi Paecilomyces lilacinus and Pochonia chlamydosporia

Abstract:The nematophagous fungi Paecilomyces lilacinus and Pochonia chlamydosporia have been extensively studied as biological control agents for plant-parasitic nematodes. This study describes the formulation of alginate pellets containing mycelia of these fungi and also describes the effect of storage conditions on shelf-life of the pellets. The shelf-lives of P. lilacinus and P. chlamydosporia, which were measured monthly for 6 months, were significantly improved at low temperatures and low water activity (a(w)) values (<0.33). Vacuum did not affect the viability of the formulated P. lilacinus but increased the viability of P. chlamydosporia. Carbon dioxide reduced the activity of P. lilacinus as compared to ambient air but increased the activity of P. chlamydosporia. Nitrogen, however, significantly improved the viability of both fungi. The optimal parameters of each factor for our formulation of P. lilacinus and P. chlamydosporia included a temperature range of 4 to -20 degrees C, a(w)=0.12, and a nitrogen-filled atmosphere.
