Abstract:We investigated the effects of prolonged heat shock treatment on human embryonic stem cell (hESC) viability. The hESC viability steadily declined with longer exposure to heat shock treatment (43C degrees). After 4 h of exposure to heat shock at 43C degrees, only 56.2 +/- 1.5% of cells were viable. Viability subsequently declined to 37.0 +/- 3.3% and 3.5 +/- 0.7% after 8 h and 16 h, respectively of heat shock treatment at 43 degrees C. Transmission electron micrographs showed that the morphology of the dead/dying cells after heat shock treatment was characteristic of cellular necrosis with an uncondensed chromatin and a non-intact plasma membrane. This was further confirmed by flow cytometry analysis which showed that the DNA of the dead/ dying cells was still mostly intact, unlike the characteristic DNA fragmentation observed with apoptotic cells. In conclusion, prolonged exposure to heat shock treatment was detrimental to hESC viability. Hence, any future protocols developed for either the heat shock pre-conditioning of hESC prior to transplantation or for the temporary expression of specific genes with heat shock-responsive promoters should take these results into account; to achieve an optimal balance between the duration of heat shock exposure and the attainment of the desired effects.
Study on Culture of Human Embryo Blastocyst and Its Related Factors

Abstract:Objectives To investigate the culture of human embryo blastocyst in vitro and its related factors. Methods The 54 IVF surplus embryos of day 3 from 18 volunteers were transferred to different mediums (IVC-Three medium,blastocyst medium or HTF medium) containing 10%SSS for advanced culture.The growth of embryos was observed until the embryo degeneration or 7 days after fertilization.Results The total blastulation rates of 54 embryos were 29.6%.The positive correlation between the cell number of day 3 embryo and blastulation rate was significant(r=0.99,P0.01).And the higher blastulation rate was positively related to embryo quality score (34.9% for 1～2.5 degree versus 9.1% for 3～4degree).The blastulation rate of IVC-Three medium was significantly higher than that of blastocyst medium and HTF(χ21=5.77,χ22=3.95,P0.05).Conclusion Advanced culture of surplus embryo will reach some blastulation.The cell number of day 3 embryo and quality score have some indication to blastulation.IVC-Three blastocyst medium may have potential clinical use for embryo blastocyst culture.
Blastocyst Culture of Human Suboptimal Embryos Does Not Improve Clinical Outcomes

Abstract:OBJECTIVE: The objective of this study was to evaluate whether blastocyst culture of human suboptimal embryos could help for embryo selection and improve clinical outcomes.DESIGN: Retrospective study.MATERIALS AND METHODS: The inclusion criteria were patients who had only suboptimal embryos (all embryos ≤6-cell or embryo fragments >10%) on Day 3. In Day 3 transfer group, embryos were selected and transferred on Day 3. In blastocyst culture group, embryos were cultured to Day 5 and transferred if reached blastocyst stage. Embryos were cultured in Vitrolife G5 series culture media at 37.5°C, 6% CO2, 5% O2. The mean age of patients was not significantly different between the groups (32.3 VS. 32.0 years). Clinical pregnancy, ongoing pregnancy and implantation rate were compared between the groups using χ Test.RESULTS: In Day 3 transfer group, 241 embryos of 114 patients (2.1 embryos /patient) were transferred, 59 were implanted (24.5%), clinical pregnancy was confirmed in 44 patients (38.6%), and ongoing pregnancy was defined in 40 patients (35.1%; miscarriage=1, ectopic pregnancy=3). In blastocyst culture group, 421 embryos of 106 patients were cultured to Day 5, 88 reached blastocyst (20.9%), and 18 blastocysts were scored ≥ 3BB (Gardener's grading criteria) (4.3%). Fifty-seven cycles (53.8%) were cancelled because of unavailability of blastocysts. Eighty-four blastocysts of 49 patients (1.7 embryos /patient) were transferred, 15 were implanted (17.9%), and clinical pregnancy and ongoing pregnancy was confirmed in 12 patients (11.3% per oocyte retrieval, 24.5% per embryo transfer). The mean number of transferred embryos (P=0.001), clinical pregnancy and ongoing pregnancy rate per oocyte retrieval (P<0.001) were significantly higher in Day 3 transfer group.CONCLUSIONS: Human embryos with suboptimal morphology showed limited developmental potential. Extended in vitro culture has more adverse stress on them, which can lead to developmental arrest. Blastocyst culture of suboptimal embryos does not improve clinical outcomes. Heat shock interferes with the amino acid metabolism of bovine cumulus-oocyte complexes in vitro: a multistep analysis

Abstract:By affecting the ovarian pool of follicles and their enclosed oocytes, heat stress has an impact on dairy cow fertility. This study aimed to determine how heat shock (HS) during in vitro maturation affected the ability of the bovine cumulus-oocyte complexes (COCs) to develop, as well as their metabolism of amino acids (AAs). In this study, COCs were in vitro matured for 23 h at 38.5 °C (control; n = 322), 39.5 °C (mild HS (MHS); n = 290), or 40.5 °C (severe HS (SHS); n = 245). In comparison to the control group, the MHS and SHS groups significantly decreased the percentage of metaphase-II oocytes, as well as cumulus cell expansion and viability. The SHS decreased the rates of cleavage and blastocyst formation in comparison to the control and MHS. Compared to the control and MHS-COCs, the SHS-COCs produced significantly more phenylalanine, threonine, valine, arginine, alanine, glutamic acid, and citrulline while depleting less leucine, glutamine, and serine. Data showed that SHS-COCs had the highest appearance and turnover of all AAs and essential AAs. Heat shock was positively correlated with the appearance of glutamic acid, glutamine, isoleucine, alanine, serine, valine, phenylalanine, and asparagine. Network analysis identified the relationship between HS and alanine or glutamic acid, as well as the relationship between blastocyst and cleavage rates and ornithine. The findings imply that SHS may have an impact on the quality and metabolism of AAs in COCs. Moreover, the use of a multistep analysis could simply identify the AAs most closely linked to HS and the developmental competence of bovine COCs.

Abstract:The overall objective of this work was to describe whether supplementing fibroblast growth factor 2 (FGF2) during in vitro maturation (IVM) affects cumulus and oocyte maturation and subsequent embryonic development.Supplementing 0.5, 5 or 50 ng/ml FGF2 to cumulus-oocyte complexes (COCs) increased the percentage of oocytes with extruded polar bodies after IVM.Supplementing FGF2 during COC maturation did not affect in vitro fertilization (IVF) rates but increased the percentage of blastocysts at day 7 post-IVF.Blastocyst cell number was not affected by FGF2 treatment.Supplementation with FGF2 did not affect cumulus expansion but reduced the proportion of apoptotic cumulus cells after IVM.The relative abundance of several cumulus cell transcripts associated with apoptosis and cumulus cell activity were not altered by FGF2 treatment.In summary, improvements in blastocyst development were achieved by FGF2 treatment during oocyte maturation.The reason for the enhanced oocyte competency remains unclear but may occur because of improvements in oocyte maturation and cumulus cell viability during IVM.
Actions of CSF2 and DKK1 on bovine embryo development and pregnancy outcomes are affected by composition of embryo culture medium

Abstract:Procedures for in vitro embryo production in cattle have not been optimized. In the current experiment, we utilized a 3 × 3 factorial design to test whether the proportion of embryos becoming blastocysts in culture and the pregnancy rate after embryo transfer are affected by type of serum in the medium [no serum; 3% (v/v) KnockOut Serum Replacement (SR); 3% (v/v) fetal bovine serum (FBS)] and addition of specific embryokines [vehicle; 10 ng/mL colony stimulating factor 2 (CSF2); 100 ng/mL dickkopf related protein 1 (DKK1)] at day 5 of culture. Embryos were produced using abattoir-derived ovaries and Y-sorted semen from two Angus sires. The percent of putative zygotes and cleaved embryos becoming blastocysts was improved by SR and FBS. Pregnancy rate at day 30 was determined for 1426 Nelore recipients and calving rate for 266 recipients. In the absence of CSF2 or DKK1, pregnancy rates were lower for embryos cultured with SR or FBS. CSF2 and DKK1 reduced pregnancy rate for embryos cultured without serum but had no detrimental effect in the SR or FBS groups. Indeed, CSF2 blocked the negative effect of FBS on pregnancy rate. Data on birth weights were available for 67 bull calves. There were no effects of treatment. The sire used to produce embryos had significant and large effects on development to the blastocyst stage, pregnancy rate at day 30, calving rate and pregnancy loss between day 30 and calving. Results indicate that (1) SR and FBS can improve embryonic development in vitro while also compromising competence of embryos to survive after transfer, (2) actions of CSF2 and DKK1 depend upon other characteristics of the embryo production system, and (3) sire can have a large effect on embryonic development before and after transfer.
Improvement of in vitro-produced bovine embryo treated with coagulansin-A under heat-stressed condition

Abstract:Heat stress has large effects on reproduction including conception rate in cattle. In this study, we examined the effects of coagulansin-A (coa-A), a steroidal lactone, on acquired thermo tolerance during in vitro production of bovine embryos. Oocytes were incubated in in vitro maturation (IVM) media with or without coa-A at two different temperatures, 40.5˚C and 42˚C, for 20 h. The treatment of coa-A significantly improved blastocyst development only at 40.5˚C (P < 0.05). Interestingly, immunofluorescence analysis demonstrated that coa-A induced heat shock protein 70 (HSP70) and phosphatidylinositol-3-kinase (PI3K), but significantly attenuated nuclear factor kappa B (NF-κB) and cyclooxygenase-2 (COX2). To determine the expression patterns of related genes at the transcription level, qRT-PCR was performed. Expression of HSP70 and PI3K was elevated, whereas expression of NF-κB, COX2 and inducible nitric oxide synthase (iNOS) was significantly (P < 0.05) downregulated in the coa-A-treated group compared with the control group. Moreover, pro-apoptotic genes were downregulated, and antiapoptic genes were upregulated in the coa-A group. We also counted the total cell number and apoptotic nuclei at the blastocyst and found that more cell numbers (143.1 ± 1.5) and less apoptotic damages (6.4 ± 0.5) in the coa-A treatment group comparing to control group (131.4 ± 2.0 and 10.8 ± 0.5), indicating the enhanced embryo quality. In conclusion, our results demonstrate that the coa-A not only improved the blastocyst development in vitro but also increased their resistance to heat stress condition through induction of HSP70/PI3K.
Effects of Heat Stress on Bovine Oocytes and Early Embryonic Development—An Update

Abstract:Heat stress is a major threat to cattle reproduction today. It has been shown that the effect of high temperature not only has a negative effect on the hormonal balance, but also directly affects the quality of oocytes, disrupting the function of mitochondria, fragmenting their DNA and changing their maternal transcription. Studies suggest that the induction of HSP70 may reduce the apoptosis of granular layer cells caused by heat stress. It has been shown that the changes at the transcriptome level caused by heat stress are consistent with 46.4% of blastocyst development disorders. Cows from calves exposed to thermal stress in utero have a lower milk yield in their lifetime, exhibit immunological disorders, have a lower birth weight and display a shorter lifespan related to the expedited aging. In order to protect cow reproduction, the effects of heat stress at the intracellular and molecular levels should be tracked step by step, and the impacts of the dysregulation of thermal homeostasis (i.e., hyperthermy) should be taken into account.

Abstract:Alimta, as well as homo-Alimta, a nonbridged analogue of Alimta, and TNP-351 have been prepared by a new method that involves Michael addition of the appropriate 1-nitroalkene with 2,6-diamino-3H-pyrimidin-4-one or 2,4,6-triaminopyrimidine, followed by a Nef reaction of the resulting primary nitro Michael adduct. Spontaneous intramolecular cyclization of the resulting aldehyde with the pyrimidine 6-amino group yields the corresponding pyrrolo[2,3-d]pyrimidine. A series of previously unknown 5-arylpyrrolo[2,3-d]pyrimidines was prepared by the same methodology from the above pyrimidines and nitrostyrenes. It has been found that the intermediate primary nitro Michael adduct can be prepared in a single step by sonication of a mixture of an arylaldehyde, nitromethane, and the 6-aminopyrimidine in acetic acid containing ammonium acetate.
Embryonic development in superovulated dairy cattle exposed to elevated ambient temperatures between the onset of estrus and insemination

Abstract:Holstein heifers (n = 16) were used to determine whether heat stress prior to ovulation increases the incidence of embryonic abnormalities. Heifers were superovulated with Follicle Stimulating Hormone (FSH-P; 32 mg total), beginning on Days 10 or 11 of the estrous cycle. Prostaglandin F2α (Lutalyse; 60 mg total) was administered on Day 3 of FSH-P treatment. Heifers were maintained at either thermoneutrality (24°C) or under hyperthermic conditions (exposure to 42°C for 10 h) beginning at the onset of estrus. Following artificial inseminations at 15 and 20 h after the onset of estrus, heifers were continuously maintained under environmental conditions of thermoneutrality for 7 days as provided by environmental shade structures. On Day 7 post estrus, embryos were recovered nonsurgically and evaluated morphologically for stage of development and quality. The distribution of embryos classified as normal, retarded and/or abnormal, or as unfertilized ova differed (P < 0.001) between heat stress and thermoneutral treatments. Only 12.0% of 25 embryos recovered from heat-stressed heifers were normal compared with 68.4% of 19 embryos from thermoneutral heifers. Stressed heifers had a higher (P < 0.001) incidence of retarded and/or abnormal embryos with degenerated blastomeres. These data indicate that thermal stress during the periovulatory period increases the incidence of retarded and/or abnormal embryos in superovulated heifers.

Effect of heat exposure on the growth and developmental competence of bovine oocytes derived from early antral follicles

Abstract:In dairy cows, low fertility caused by summer heat stress continues into the cooler autumn season. This can be caused by impaired oocyte quality in small growing follicles during summer. Here, we subjected oocyte-cumulus-granulosa complexes (OCGCs) derived from early antral follicles (0.5-1 mm) to in vitro growth (IVG) culture under two different temperature settings (the control and heat shock groups), and evaluated effects of heat exposure on growth and developmental competence of oocytes, factors affecting the developmental competence of oocytes (steroidogenesis of granulosa cells, oxidative stress in oocytes, and cell-to-cell communication between oocytes and somatic cells). Oocyte diameters after culture were smaller in the heat shock group. Although nuclear maturation and cleavage rates were similar between the groups, blastocyst rates were lower in the heat shock group (0.0%) than in the control group (27.7%), and reduced glutathione (GSH) levels in oocytes were lower in the heat shock group. Supplementation of cysteine, which stimulates GSH synthesis, increased GSH level and improved blastocyst rate of heat shocked oocytes (27.9%). These results suggest that heat exposure impairs the growth and developmental competence of oocytes in early antral follicles through GSH depletion, which can induce low fertility during summer and the following autumn.
Disorder of Biological Quality and Autophagy Process in Bovine Oocytes Exposed to Heat Stress and the Effectiveness of In Vitro Fertilization

Abstract:The main problem in dairy herds is reproductive disorders, which are influenced by many factors, including temperature. Heat stress reduces the quality of oocytes and their maturation through the influence of, e.g., mitochondrial function. Mitochondria are crucial during oocyte maturation as well as the process of fertilization and embryonic development. Disturbances related to high temperature will be increasingly observed due to global warming. In present studies, we have proven that exposure to high temperatures during the cleaving of embryos statistically significantly (at the level of p < 0.01) reduces the percentage of oocytes that cleaved and developed into blastocysts eight days after insemination. The study showed the highest percentage of embryos that underwent division in the control group (38.3 °C). The value was 88.10 ± 6.20%, while the lowest was obtained in the study group at 41.0 °C (52.32 ± 8.40%). It was also shown that high temperature has a statistically significant (p < 0.01) effect on the percentage of embryos that developed from the one-cell stage to blastocysts. The study showed that exposure to a temperature of 41.0 °C significantly reduced the percentage of embryos that split relative to the control group (38.3 °C; 88.10 ± 6.20%). Moreover, it was noted that the highest tested temperature limits the development of oocytes to the blastocyst stage by 5.00 ± 9.12% compared to controls (33.33 ± 7.10%) and cleaved embryos to blastocysts by 3.52 ± 6.80%; the control was 39.47 ± 5.40%. There was also a highly significant (p < 0.0001) effect of temperature on cytoplasmic ROS levels after 6 and 12 h IVM. The highest level of mitochondrial ROS was found in the group of oocytes after 6 h IVM at 41.0 °C and the lowest was found in the control group. In turn, at 41.0 °C after 12 h of IVM, the mitochondrial ROS level had a 2.00 fluorescent ratio, and the lowest in the group was 38.3 °C (1.08). Moreover, with increasing temperature, a decrease in the expression level of both LC3 and SIRT1 protein markers was observed. It was proved that the autophagy process was impaired as a result of high temperature. Understanding of the cellular and molecular responses of oocytes to elevated temperatures will be helpful in the development of heat resistance strategies in dairy cattle.

Abstract:PurposeThe effects of astaxanthin (Ax) on the in vitro development of bovine embryos cultured under heat stress were investigated in combination with the assessment of its cellular accumulation and action on mitochondrial membrane potential (ΔΨm).MethodsBovine ≥8-cell embryos were collected on day 3 after in vitro fertilization and exposed to single (day 4) or repeated (day 4 and 5) heat stress (10 h/day at 40.5 °C). Ax was added into culture medium under the repeated heat stress and blastocyst development was evaluated. The cellular uptake of Ax in embryos was examined using bright-field and confocal laser-scanning microscopy, and high-performance liquid chromatography. The relationship between Ax and mitochondria localization was assessed using MitoTracker dye. The effects of Ax on ΔΨm were investigated using JC-1 dye.ResultsBlastocyst development in the repeated heat stress treatment decreased significantly (P < 0.05) compared with those in single heat stress or normal thermal treatment. The addition of Ax into culture medium did lead to a significant recovery in blastocyst development in the repeated heat-treated group. Ax was detected in cytoplasm of embryos and observed to colocalize with mitochondria. Ax recovered ΔΨm in embryos that was decreased by the heat treatment.ConclusionsAx ameliorated the heat stress-induced impairment of blastocyst development. Our results suggest that the direct action of Ax on mitochondrial activity via cellular uptake is a mechanism of the ameliorating effects.

Abstract: Mice receiving a total of 16 to 18 mg of bovine serum albumin (BSA) during the first 3 weeks of life, including 2 mg of alum-precipitated BSA as a last injection, displayed so-called “spontaneous escape.” A very small percentage of animals receiving only soluble BSA exhibited this phenomenon during the time course of these studies. Spontaneous escape was delayed in animals which were challenged 1 or 2 weeks postregimen. Spontaneous escape was hastened in animals which were challenged 3 or 4 weeks postregimen by means of alum-precipitated BSA. After the administration of a supposedly hyperimmunizing series of injections from the 2nd to the 6th week postregimen, paralysis was prolonged with respect to onset of response and the level of antibody produced. From these studies it is concluded that one important route of “spontaneous escape” involves a sequence of paralysis, the generation of memory cells, and contact of these cells with endogenously retained antigen, followed by extensive antibody formation. A dualistic view of escape was supported in the discussion toward which both reversibly paralyzed X cells and virgin competent immunocytes make a contribution.
Stem dimensional fluctuation in Jeffrey pine from variation in water storage as influenced by thinning and prescribed fire

Abstract:Forest thinning utilizing cut-to-length and whole-tree harvesting systems with subsequent underburning were assessed for their impacts on water storage in the extensible tissues of dominant and codominant trees in an uneven-aged Jeffrey pine (Pinus jeffreyi Grev. & Balf.) stand on the east slope of the Sierra Nevada. Prior to the onset of the third growing season following thinning and the second season after burning, manual band dendrometers were installed at breast height on the selected trees and readings of diurnal fluctuation in stem circumference, an indication of bole water status, were taken monthly for one year. Diameter and relative diameter fluctuation were calculated from the circumference measurements. Overall, thinning had a positive influence on stem water recharge capacity, with the most pronounced effects evident in the latter part of the growing season. During this period, bole contraction in thinned stand portions was 49 to 55% greater than in the unthinned control, suggesting that both a greater volume of stored water was available for transpiration and was transpired in trees of the former treatment. There was no clear evidence that harvesting method affected stem water storage and influences of underburning were also absent entirely. Seasonal effects on diurnal changes in stem diameter were prominent, as the extent to which boles contracted generally increased over the course of the growing season, whereas fluctuations were at a minimum during the colder months. The magnitude of stem dimensional flux was found to be negatively correlated with initial tree DBH in one instance, while negative relationships between the former and live crown length as well as percentage were also revealed, albeit infrequently. Changes in bole size were positively correlated with residual basal area in some cases. These results suggest that improvement in water relations can be realized from density management in a dry site forest type with no apparent compromise of this benefit by broadcast underburning.RésuméLes éclaircies en forêt utilisant les systèmes de débit à la scie et de récolte de l’ensemble de l’arbre avec ultérieurement un brûlage par le bas ont été évaluées pour leurs impacts sur le stockage en eau dans les tissus extensibles des arbres dominants et codominants dans un peuplement inéquienne de Pinus jeffreyi Grev. & Balf sur le versant est de la Sierra Nevada. Avant le début de la troisième année de croissance suivant l’éclaircie et la deuxième saison après le feu, des dendromètres manuels à bande ont été installés à hauteur de poitrine sur des arbres sélectionnés et la lecture des fluctuations journalières de la circonférence des arbres, une indication sur le statut hydrique du tronc, étaient faite mensuellement pendant une année. Le diamètre et les fluctuations de diamètre ont été calculés à partir des mesures de circonférence. En général, l’éclaircie a une influence positive sur la capacité de recharge en eau du tronc, avec les effets évidents les plus prononcés dans la dernière partie de la saison de croissance. Pendant cette période, la contraction du tronc dans les parties éclaircies des peuplements était de 49 à 55 % plus grande que le témoin non éclairci, suggérant qu’à la fois un plus grand volume d’eau stocké était disponible pour la transpiration et était transpiré par les arbres du premier traitement. Il n’y a pas une preuve claire que la méthode de récolte affecte le stockage d’eau dans le tronc et l’influence du brûlage était aussi totalement absente. Les effets saisonniers sur les changements diurnes du diamètre du tronc étaient proéminents, alors que l’importance des troncs contractés s’est accrue généralement dans le courant de la saison de croissance tandis que les fluctuations ont été au minimum pendant les mois froids de l’année. Dans un cas, l’ampleur de la dimension du flux a été trouvée corrélée négativement avec le diamètre initial, à hauteur de poitrine, de l’arbre, tandis qu’une relation négative entre ce dernier et la longueur de la couronne vivante comme en pourcentage a été aussi révélée, bien que peu fréquemment. Dans quelques cas, les changements dans la taille du tronc ont été positivement corrélés avec la surface terrière résiduelle. Ces résultats suggèrent qu’une amélioration des relations hydriques peut être réalisée par une gestion de la densité des peuplements dans les forêts de zones sèche sans apparemment compromettre ce bénéfice par un brûlage par surface.
The Use of a Brief Synchronization Treatment after Weaning, Combined with Superovulation, Has Moderate Effects on the Gene Expression of Surviving Pig Blastocysts

Abstract:The combination of estrus synchronization and superovulation (SS) treatments causes alterations in ovarian and endometrial gene expression patterns, resulting in abnormal follicle and oocyte growth, fertilization, and embryo development. However, the impact of combined SS treatments on the transcriptome of the surviving embryos remains unidentified. In this study, we examined gene expression changes in day 6 blastocysts that survived a brief regimen of synchronization treatment combined with superovulation. The sows were included in one of three groups: SS7 group (n = 6), sows were administered Altrenogest (ALT) 7 days from the day of weaning and superovulated with eCG 24 h after the end of ALT treatment and hCG at the onset of estrus; SO group (n = 6), ALT nontreated sows were superovulated with eCG 24 h postweaning and hCG at the onset of estrus; control group (n = 6), weaned sows displaying natural estrus. Six days after insemination, the sows underwent a surgical intervention for embryo collection. Transcriptome analysis was performed on blastocyst-stage embryos with good morphology. Differentially expressed genes (DEGs) between groups were detected using one-way ANOVA with an un-adjusted p-value < 0.05 and a fold change 1.5. The effect of SO treatment on the number of altered pathways and DEGs within each pathway was minimal. Only four pathways were disrupted comprising only a total of four altered transcripts, which were not related to reproductive functions or embryonic development. On the other hand, the surviving blastocysts subjected to SS7 treatments exhibited moderate gene expression changes in terms of DEGs and fold changes, with seven pathways disrupted containing a total of 10 transcripts affected. In this case, the up-regulation of certain pathways, such as the metabolic pathway, with two up-regulated genes associated with reproductive functions, namely RDH10 and SPTLC2, may suggest suboptimal embryo quality, while the down-regulation of others, such as the glutathione metabolism pathway, with down-regulated genes related to cellular detoxification of reactive oxygen species, namely GSTK1 and GSTO1, could depress the embryos' response to oxidative stress, thereby impairing subsequent embryo development. The gene expression changes observed in the present study in SS7 embryos, along with previous reports indicating SS7 can negatively affect fertilization, embryo production, and reproductive tract gene expression, make its use in embryo transfer programs unrecommendable.

Abstract:Heat stress can have a serious impact on the health of both humans and animals. A major question is how heat stress affects normal development and differentiation at both the cellular and the organism levels. Here we use an in vitro experimental system to address how heat shock treatment influences the properties of bovine mesenchymal stem cells (MSCs)—multipotent progenitor cells—which are found in most tissues. Because cattle are sensitive to harsh external temperatures, studying the effects of heat shock on MSCs provides a unique platform to address cellular stress in a physiologically relevant model organism. Following isolation and characterization of MSCs from the cow's umbilical cord, heat shock was induced either as a pulse (1 h) or continuously (3 days), and consequent effects on MSCs were characterized. Heat shock induced extensive phenotypic changes in MSCs and dramatically curtailed their capacity to proliferate and differentiate. These changes were associated with a partial arrest in the G1/S or G2/M checkpoints. Furthermore, MSCs lost their ability to resolve the inflammatory response of RAW macrophages in coculture. A possible explanation for this loss of function is the accumulation of reactive oxygen species and malfunction of the mitochondria in the treated cells. Heat shock treatments resulted in stress-induced premature senescence, affecting the MSCs' ability to proliferate properly for many cell passages to follow. Exposure to elevated external temperatures leads to mitochondrial damage and oxidative stress, which in turn conveys critical changes in the proliferation, differentiation, and immunomodulatory phenotype of heat-stressed MSCs. A better understanding of the effect of heat shock on humans and animals may result in important health and economic benefits.

Advanced embryo development during extended in vitro culture: observations of formation and hatching patterns in non-transferred human blastocysts

Abstract:Human embryos not chosen for fresh transfer or cryopreservation were maintained in extended in vitro culture for up to 9 days after fertilization to observe blastocyst formation and hatching features. These non-transferred embryos were derived from 64 consecutive IVF cycles, and were not cryopreserved either because of compromised morphology or because the patients did not consent to cryopreservation for personal reasons. Embryos were cultured individually to monitor daily growth until developmental arrest, and differential blastocyst formation and hatching were analysed among groups of patients and embryos. In the population studied, hatching occurred most commonly on day 7 after fertilization (range 5-9 days). A total of 301 blastocysts was observed, of which 116 (38.5%) eventually hatched in vitro irrespective of day of formation. A trend towards earlier blastocyst formation and a greater likelihood of hatching was noted in this population. Both blastocyst formation and hatching appeared negatively correlated with increasing maternal age and higher basal serum FSH concentrations on day 3 of development, although these trends did not reach statistical significance. Comparison of intracytoplasmic sperm injection (ICSI) (n = 25) and conventional insemination (n = 39) cycles showed a similar rate of blastocyst formation in both groups (54 and 52%, respectively; P > 0.05), but hatching patterns varied significantly between these groups (4.1 versus 61.6%, respectively; P < 0.0001). The discovery of marked impairment of hatching among non-transferred ICSI embryos supports the case for reconsideration of the appropriateness of assisted blastocyst hatching in selected cases.
