Abstract:On-demand controlled drug delivery is essential for the treatment of a wide range of chronic diseases. As the drug is released at the time when required, its efficacy is boosted and the side effects are minimized. However, so far, drug delivery devices often rely on the passive diffusion process for a sustained release, which is slow and uncontrollable. Here, we present a miniaturized microfluidic device for wirelessly controlled ultrafast active drug delivery, driven by an oscillating solid-liquid interface. The oscillation generates acoustic streaming in the drug reservoir, which opens an elastic valve to deliver the drug. High-speed microscopy reveals the fast response of the valve on the order of 1 ms, which is more than three orders of magnitude faster than the start-of-the-art. The amount of the released drug exhibits a linear relationship with the working time and the electric power applied to the ultrasonic resonator. The trigger of the release is wirelessly controlled via a magnetic field, and the system shows stable output in a continuous experiment for two weeks. The integrated system shows great promise as a long-term controlled drug delivery implant for chronic diseases.

Abstract:Both acute and chronic diseases require on-demand and convenient drug delivery, particularly for life-threatening acute conditions. Although emerging microneedle arrays (MNs) offer a promising therapeutic platform for painlessly administering drug payloads to patients, current dissolving or diffusion-based MNs encounter significant challenges in achieving programmable and on-demand liquid drug delivery. In this study, we develop active programmable MNs based on a wearable acoustic platform, which enables the smart management of diseases. The vortex effect near the tips excited by acoustics is utilized to actively extract drugs from MNs, that are manufactured through high precision 3D printing. By digitally controlling acoustic signals, we demonstrate user-requirement-based drug releasing, in tissue-mimicking agar gel phantoms and mouse models. The system achieves precise dosage control through three different modes: rapid single delivery mode, batch delivery mode, and long-term slow-release mode. This versatility makes the system suitable for various disease conditions. The in situ acoustic drug delivery strategy implemented on this platform offers advantages in terms of miniaturization, ease of operation and intelligence. This platform is anticipated to provide a novel personalized therapeutic approach for clinics and biomedicine.

Abstract:In our previous work, it was found that acoustic cavitation might play a role in improving the cell permeability to microparticles when liposomes were used in an in vitro experiment. The purpose of this project is to expand our study and to learn other possible mechanisms by which cells may interact with liposomes under ultrasound (US) excitation and become transiently permeable to microparticles. It is further hypothesized that two possible scenarios may be involved in in vitro experiments: (1) drug-carrying liposomes transiently overcome the cell membrane barrier and enter into a cell while the cell is still viable; (2) the liposomes incorporate with a cell at its membrane through a fusing process. To prove this hypothesis, liposomes of two different structures were synthesized: one has fluorescent molecules encapsulated into liposomes and the other has fluorescent markers incorporated into the shells of liposomes. Liposomes of each kind were mixed with human breast cancer cells (MCF7-cell line) in a suspension at 5 (liposomes) : 1 (cell) ratio and were then exposed to a focused 1 MHz ultrasound beam at its focal region for 40 s. The US signal contained 20 cycles per tone-burst at a pulse-repetition-frequency of 10 kHz; the spatial peak acoustic pressure amplitude was 0.25 MPa. It was found that the possible mechanisms might include the acoustic cavitation, the endocytosis and cell-fusion. Acoustic radiation force might make liposomes collide with cells effectively and facilitate the delivery process.

Abstract:Acoustic tweezers based on travelling surface acoustic waves (TSAWs) have the potential for contactless trajectory manipulation and motion-parameter regulation of microparticles in biological and microfluidic applications. Here, we present a novel design of a tri-directional symmetrical acoustic tweezers device that enables the precise manipulation of linear, clockwise, and anticlockwise trajectories of microparticles. By switching the excitation combinations of interdigital electrodes (IDTs), various shape patterns of acoustic pressure fields can be formed to capture and steer microparticles accurately according to pre-defined trajectories. Numerical simulations and experimental tests were conducted in this study. By adjusting the input electric signals and the fluid's viscosity, the device is able to manipulate microparticles of various forms as well as brine shrimp egg cells with the accurate modulation of motion parameters. The results show that the proposed programmable design possesses low-cost, compact, non-contact, and high biocompatibility benefits, with the capacity to accurately manage microparticles in a range of motion trajectories, independent of their physical and/or chemical characteristics. Thus, our design has strong potential applications in chemical composition analysis, drug delivery, and cell assembly.

Abstract:Nondestructive, high-efficiency, and on-demand intracellular drug/biomacromolecule delivery for therapeutic purposes remains a great challenge. Herein, a biomechanical-energy-powered triboelectric nanogenerator (TENG)-driven electroporation system is developed for intracellular drug delivery with high efficiency and minimal cell damage in vitro and in vivo. In the integrated system, a self-powered TENG as a stable voltage pulse source triggers the increase of plasma membrane potential and membrane permeability. Cooperatively, the silicon nanoneedle-array electrode minimizes cellular damage during electroporation via enhancing the localized electrical field at the nanoneedle-cell interface and also decreases plasma membrane fluidity for the enhancement of molecular influx. The integrated system achieves efficient delivery of exogenous materials (small molecules, macromolecules, and siRNA) into different types of cells, including hard-to-transfect primary cells, with delivery efficiency up to 90% and cell viability over 94%. Through simple finger friction or hand slapping of the wearable TENGs, it successfully realizes a transdermal biomolecule delivery with an over threefold depth enhancement in mice. This integrated and self-powered system for active electroporation drug delivery shows great prospect for self-tuning drug delivery and wearable medicine.

Abstract:Acoustic tweezers based on surface acoustic waves (SAWs) have raised great interest in the fields of tissue engineering, targeted therapy, and drug delivery. Generally, the complex structure and array layout design of interdigital electrodes would restrict the applications of acoustic tweezers. Here, we present a novel approach by using bisymmetric coherent acoustic tweezers to modulate the shape of acoustic pressure fields with high flexibility and accuracy. Experimental tests were conducted to perform the precise, contactless, and biocompatible cluster manipulation of polystyrene microparticles and yeast cells. Stripe, dot, quadratic lattice, hexagonal lattice, interleaved stripe, oblique stripe, and many other complex arrays were achieved by real-time modulation of amplitudes and phase relations of coherent SAWs to demonstrate the capability of the device for the cluster manipulation of particles and cells. Furthermore, rapid switching among various arrays, shape regulation, geometric parameter modulation of array units, and directional translation of microparticles and cells were implemented. This study demonstrated a favorable technique for flexible and versatile manipulation and patterning of cells and biomolecules, and it has the advantages of high manipulation accuracy and adjustability, thus it is expected to be utilized in the fields of targeted cellular assembly, biological 3D printing, and targeted release of drugs.

Abstract:Acoustic tweezers provide unique capabilities in medical applications, such as contactless manipulation of small objects (e.g., cells, compounds or living things), from nanometer-sized extracellular vesicles to centimeter-scale structures. Additionally, they are capable of being transmitted through the skin to trap and manipulate drug carriers in various media. However, these capabilities are hindered by the limitation of controllable degrees of freedom (DoFs) or are limited maneuverability. In this study, we explore the potential application of acoustical tweezers by presenting a five-DoF contactless manipulation acoustic system (AcoMan). The system has 30 ultrasound transducers (UTs) with single-side arrangement that generates active traveling waves to control the position and orientation of a fully untethered nanocarrier clusters (NCs) in a spherical workspace in water capable of three DoFs translation and two DoFs rotation. In this method, we use a phase modulation algorithm to independently control the phase signal for 30 UTs and manipulate the NCs' positions. Phase modulation and switching power supply for each UT are employed to rotate the NCs in the horizontal plane and control the amplitude of power supply to each UT to rotate the NCs in the vertical plane. The feasibility of the method is demonstrated by in vitro and ex vivo experiments using porcine ribs. A significant portion of this study could advance the therapeutic application such a system as targeted drug delivery.

Abstract:Multidrug combination therapy in the inner ear faces diverse challenges due to the distinct physicochemical properties of drugs and the difficulties of overcoming the oto-biologic barrier. Although nanomedicine platforms offer potential solutions to multidrug delivery, the access of drugs to the inner ear remains limited. Micro/nanomachines, capable of delivering cargo actively, are promising tools for overcoming bio-barriers. Herein, a novel microrobot-based strategy to penetrate the round window membrane (RWM) is presented and multidrug in on-demand manner is delivered. The tube-type microrobot (TTMR) is constructed using the template-assisted layer-by-layer (LbL) assembly of chitosan/ferroferric oxide/silicon dioxide (CS/Fe3O4/SiO2) and loaded with anti-ototoxic drugs (curcumin, CUR and tanshinone IIA, TSA) and perfluorohexane (PFH). Fe3O4 provides magnetic actuation, while PFH ensures acoustic propulsion. Upon ultrasound stimulation, the vaporization of PFH enables a microshotgun-like behavior, propelling the drugs through barriers and driving them into the inner ear. Notably, the proportion of drugs entering the inner ear can be precisely controlled by varying the feeding ratios. Furthermore, in vivo studies demonstrate that the drug-loaded microrobot exhibits superior protective effects and excellent biosafety toward cisplatin (CDDP)-induced hearing loss. Overall, the microrobot-based strategy provides a promising direction for on-demand multidrug delivery for ear diseases.

Abstract:Abstract Acoustic tweezer technique with non-contact micro-object manipulation capability has been widely utilized for particle patterning in the fields of cell arraying, target drug delivery, and functional composite fabrication. However, the manipulation resolution of acoustic tweezer device is decided by the configuration of interdigital transducers (IDTs), which is half the wavelength of surface acoustic waves (SAWs). This limitation severely reduces the manipulation versatility of acoustic tweezer devices. To address this limitation, we propose a novel approach that employs phase-modulated pulsing coherent SAWs. This method can halve the patterning spacing and allows for customization of the spacing based on specific manipulation demands. Our proposed technique increases the flexibility and manipulation resolution of acoustic tweezer device, and thus can be widely applied to reconfigurable assembly and precise manipulation of particles and cells.
Ultrasound-actuated drug delivery with acoustic percolation switches

Abstract:Devices that can be remote-controlled under image guidance to precisely deliver biomedicines to sites of disease are a major goal of biomedical research. However, most existing externally triggered delivery systems are based on complex micromachines that are controlled with electromagnetic waves and require custom external instrumentation. Here we present a drug delivery platform comprising a simple protein-containing hydrogel that can be both imaged and triggered to release drugs at specific locations using widely available ultrasound imaging devices. This technology is based on the addition of air-filled protein nanostructures called gas vesicles (GVs) to hydrogel delivery vehicles. While intact, GVs sterically block the release of drug payloads and allow the vehicle to be imaged with ultrasound. An increase in ultrasound pressure causes the collapse of GVs within hydrogels present at the desired anatomical location, instantly creating percolation channels and triggering rapid drug release. Both the imaging and release are performed using a common diagnostic ultrasound probe. We implement this concept by establishing ultrasound-controlled drug diffusion and release from hydrogels in vitro and demonstrating targeted image-guided protein delivery in vivo following oral administration. We use this approach to deliver anti-inflammatory antibodies to treat gastrointestinal inflammation in a rat model of colitis. Targeted acoustic percolation switches (TAPS) open a conduit for local, image-guided drug delivery with a simple formulation and commonplace ultrasound equipment.

Abstract:By integrating the scanning optical tweezers and flexible optofluidic strategy, a set of optical manipulation techniques based on both direct and indirect optical actuation is developed and applied for the active delivery of nanodrugs in vivo, thus providing an alternative solution for the remotely controlled drug delivery which is compatible with most common drug carriers and exhibits significantly higher manipulation precision. The active delivery of nanodrugs has been a bottleneck problem in nanomedicine. While modification of nanodrugs with targeting agents can enhance their retention at the lesion location, the transportation of nanodrugs in the circulation system is still a passive process. The navigation of nanodrugs with external forces such as magnetic field has been shown to be effective for active delivery, but the existing techniques are limited to specific materials like magnetic nanoparticles. In this study, an alternative actuation method is proposed based on optical manipulation for remote navigation of nanodrugs in vivo, which is compatible with most of the common drug carriers and exhibits significantly higher manipulation precision. By the programmable scanning of the laser beam, the motion trajectory and velocity of the nanodrugs can be precisely controlled in real time, making it possible for intelligent drug delivery, such as inverse‐flow transportation, selective entry into specific vascular branch, and dynamic circumvention across obstacles. In addition, the controlled mass delivery of nanodrugs can be realized through indirect actuation by the microflow field. The developed optical manipulation method provides a new solution for the active delivery of nanodrugs, with promising potential for the treatment of blood diseases such as leukemia and thrombosis.

Abstract:Abstract Nanocarrier and exosome encapsulation has been found to significantly increase the efficacy of targeted drug delivery while also minimizing unwanted side effects. However, the development of exosome-encapsulated drug nanocarriers is limited by low drug loading efficiencies and/or complex, time-consuming drug loading processes. Herein, we have developed an acoustofluidic device that simultaneously performs both drug loading and exosome encapsulation. By synergistically leveraging the acoustic radiation force, acoustic microstreaming, and shear stresses in a rotating droplet, the concentration, and fusion of exosomes, drugs, and porous silica nanoparticles is achieved. The final product consists of drug-loaded silica nanocarriers that are encased within an exosomal membrane. The drug loading efficiency is significantly improved, with nearly 30% of the free drug (e.g., doxorubicin) molecules loaded into the nanocarriers. Furthermore, this acoustofluidic drug loading system circumvents the need for complex chemical modification, allowing drug loading and encapsulation to be completed within a matter of minutes. These exosome-encapsulated nanocarriers exhibit excellent efficiency in intracellular transport and are capable of significantly inhibiting tumor cell proliferation. By utilizing physical forces to rapidly generate hybrid nanocarriers, this acoustofluidic drug loading platform wields the potential to significantly impact innovation in both drug delivery research and applications.

Abstract:Efficient and controllable gene/drug delivery triggered by sonoporation is critical to cellular therapy. This study reports on repairable sonoporation for massive cells trapped on a phononic crystal plate (PCP). This mechanism is generated by the effects of enhanced near-boundary streaming around the manipulated cells. The streaming is achieved by significantly improving the tangential acoustic field gradient along the cell boundary through resonant PCP excitation of the nonleaky Lamb modes intrinsically existing in the uniform plate. This structured field-induced repairable sonoporation may constitute an effective tool for drug delivery and gene transfection.
A fiber optoacoustic emitter with controlled ultrasound frequency for cell membrane sonoporation at submillimeter spatial resolution

Abstract:<p>Focused ultrasound has attracted great attention in minimally invasive therapeutic and mechanism studies. Frequency below 1 MHz is identified preferable for high-efficiency bio-modulation. However, the poor spatial confinement of several millimeters and large device diameter of ∼25 mm of typical sub-MHz ultrasound technology suffered from the diffraction limit, severely hindering its further applications. To address it, a fiber-based optoacoustic emitter (FOE) is developed, serving as a miniaturized ultrasound point source, with sub-millimeter confinement, composed of an optical diffusion layer and an expansion layer on an optical fiber. By modifying acoustic damping and light absorption performance, controllable frequencies in the range of 0.083 MHz to 5.500 MHz are achieved and further induce cell membrane sonoporation with frequency dependent efficiency. By solving the problem of compromise between sub-MHz frequency and sub-millimeter precision via breaking the diffraction limit, the FOE shows a great potential in region-specific drug delivery, gene transfection and neurostimulation.</p>

Analog inhibitors for the pyrophosphate-dependent phosphofructokinase of Entamoeba histolytica and their effect on culture growth.

Abstract:Diphosphonate substrate-analogs of inorganic pyrophosphate (PPi) were found to be com- petitive inhibitors for amebal PPi-dependent phosphofructokinase. Analogs of the phosphoryl donor were employed in in vitro growth tests. All appeared to inhibit amebal growth; one was amebistatic at a con- centration of 1 mM. Pyrophosphate analogs are, potentially, therapeutic agents for intestinal amebiasis. A practical reason for the biochemical study of parasite metabolism is to search for an es- sential metabolic step that is catalyzed by an enzyme distinctly different from that of the host, opening the door to a pharmacological attack upon the parasite without harm to the host. The discovery that the human parasite Entamoeba histolytica has a PPi-dependent phosphofructokinase (E.C. 2.7.1.90) (Reeves et al., 1974, 1976) different from the ATP- utilizing enzyme of the human host invited further study of the amebal enzyme. The bio- chemical energy of PPi arising from ana- bolic processes (protein, nucleic acid, and glycogen synthesis) is conserved by phos- phate bond formation in the ameba whereas higher organisms are thought to dispose of this energy by hydrolysis. This report contains our findings on pyrophosphate-analog inhibitors of amebal phosphofructokinase, and our find- ings relative to the inhibition of amebal growth in culture in the presence of the analogs. Medical Microrobot - Wireless Manipulation of a Drug Delivery Carrier through an External Ultrasonic Actuation: Preliminary Results

Abstract:<p class="a-plus-plus">To achieve precise and untethered clinical therapeutics, microrobots have been widely researched. However, because conventional microrobot actuation is based on magnetic forces generated by a magnetic field and magnetic particles, unexpected side effects caused by additional magnetic ingredients could induce clinical safety issues. In this paper, as an alternative to an untethered actuator, we present a novel ultrasonic actuation mechanism that enables drug particle/cell manipulation and micro/nano-robot actuation in clinical biology and medicine. Firstly, characteristics of the acoustic field in the vessel mimic circular tube, formed from particles emerging through a submerged ultrasonic transducer, are mathematically analyzed and modeled. Thereafter, a control method is proposed for trapping and moving the micro-particles by using acoustic radiation force (ARF) in a standing wave of a tangential standing wave. The feasibility of the proposed method could be demonstrated with the help of experiments conducted using a single transducer with a resonance frequency of 950 kHz and a motorized linear stage, which were used in a water tank. The micro-particles in the tube were trapped via ultrasound and the position of the micro-particles could be controlled by frequency manipulation of the transducer and motor control. This study shows that ultrasonic manipulation can be used for specific applications, such as the operation of a micro robot inserted in a peripheral blood vessel and targeted for drug delivery.</p>

However, normal cells are difficult to acoustically manipulate because of their small size and the similarity between their acoustic impedance and that of the medium. In this study, we use the heterologous expression of gene clusters to generate genetically engineered bacteria that can produce numerous sub-micron gas vesicles in the bacterial cytoplasm. We show that the presence of the gas vesicles significantly enhances the acoustic sensitivity of the engineering bacteria, which can be manipulated by ultrasound. We find that by employing phased-array-based acoustic tweezers, the engineering bacteria can be trapped into clusters and manipulated in vitro and in vivo via electronically steered acoustic beams, enabling the counter flow or on-demand flow of these bacteria in the vasculature of live mice. Furthermore, we demonstrate that the aggregation efficiency of engineering bacteria in a tumour is improved by utilizing this technology. This study provides a platform for the in-vivo manipulation of live cells, which will promote the progress of cell-based biomedical applications.

Abstract:Common delivery routes for chemotherapeutics are based on circulation, which faces clinical limitations to local delivery efficiency, and the conflict between the dose for anticancer effect and the systemic toxicity. The recent advances in localized delivery strategies aim to improve drug accumulation at the target site or directly transport into cells. However, most are not equipped to provide additional momentum in the process of cargo release, propagation, and intracellular movement, which limit their locomotion that relies on passive diffusion. In this work, a multimicrochannel microneedle microporation (4M) platform that achieves high efficiency, safety, and uniformity for in vivo intracellular delivery is proposed. By high precision 3D printing, internal microchannels are implemented through the microneedle, which offer a concentrated, safe electric field that not only accelerates the movement of cargo into deep tissue under electrophoresis, but also triggers cell electroporation, achieving enhanced transport across cell membrane. The platform proves efficient for the delivery of chemotherapeutics in solid tumors in vitro and in vivo, with significantly enhanced anticancer effect and reduced systemic toxicity. The platform serves as a general‐purpose delivery tool to emerging drugs in vivo.
Acoustic Energy Controlled Nanoparticle Aggregation for Nanotherapy

Abstract:Patients with unresectable or nonablatable tumors are difficult to cure, but nanotherapy combining targeted nanoparticles has many severe side effects due to the toxicities of anticancer drugs. We found that acoustic energy can produce a local region with high concentration from a low concentration suspended liquid of nano-SiO2 particles at 2.5 MHz. Our calculated results show that the main reason for aggregation is the synthesized effect of the potential well of acoustic energy and streaming to trap them. In addition, the aggregated region can be manipulated to a targeted position in the vessel phantom by moving the ultrasound transducer external to the body. This noninvasive manipulation of suspended nanoparticles can rapidly increase the local drug concentration, but reduce the total dosage of anticancer drugs, which has the potential to be used for patients with advanced tumors by improving the physiological effects and reducing the side effects.
