Abstract:Trypsin, a pancreatic enzyme associated with diseases like pancreatic cancer and cystic fibrosis, requires effective diagnostic tools. Current detection systems seldom utilize macrocyclic molecules and tetraphenyl ethylene (TPE) derivative-based supramolecular assemblies, known for their biocompatibility and aggregation-induced emission (AIE) properties, for trypsin detection. This study presents an enzyme-responsive, AIE-based fluorescence "Turn-On" sensing platform for trypsin detection, employing sulfated-β-cyclodextrin (S-βCD), imidazolium derivative of TPE (TPE-IM), and protamine sulfate (PrS). The anionic S-βCD and cationic TPE-IM formed a strongly fluorescent supramolecular aggregation complex in an aqueous buffer. However, PrS suppresses fluorescence because of its strong binding affinity with S-βCD. The non-fluorescent TPE-IM/S-βCD/PrS supramolecular assembly system exhibits trypsin-responsive properties, as PrS is a known trypsin substrate. Trypsin restores fluorescence in the TPE-IM/S-βCD system through the enzymatic cleavage of PrS, correlating linearly with trypsin catalytic activity in the 0-10 nM concentration range. The limit of detection is 10 pM. This work contributes to the development of self-assembled supramolecular biosensors using charged TPE derivatives and β-cyclodextrin-based host-guest chemistry, offering an innovative fluorescence 'Turn-On' trypsin sensing platform. The sensing system is highly stable under various conditions, selective for Trypsin, and demonstrates potential for biological analysis and disease diagnosis in human serum. Additionally, it shows promise for the screening of trypsin inhibitors.

Abstract:Amphiphilic aggregation-induced emission (AIE) molecules show superior potential for fabricating novel ultrasmall nanoprobes. Here, an anionic dipyridyl tetraphenylethene (TPE) derivative is rationally designed and a super-small self-assembled AIEgen nanoprobe (TPE-2Py-SO3NaNPs, ca. 2.48 nm) is thus conveniently constructed for the supersensitive detection of protamine and trypsin. In HEPES/DMSO solution (8:2, v/v, pH = 7.4), negatively charged TPE-2Py-SO3NaNPs exhibited an AIE effect in the presence of positively charged protamine, presenting a fluorescence enhancement at 498 nm together with a large Stokes shift of 150 nm and a low detection limit of 8.0 ng/mL. In addition, the in situ formed TPE-2Py-SO3Na/protamine nanocomposite can be dissociated by trypsin due to the highly selective degradation of protamine via enzymatic hydrolysis, achieving a detection limit for trypsin as low as 5.0 ng/mL.

Bsa-Tetraphenylethene Derivative Conjugates With Aggregation-Induced Emission Properties: Fluorescent Probes For Label-Free And Homogeneous Detection Of Protease And Alpha 1-Antitrypsin

Abstract:Herein, BSA-tetraphenylethene derivative conjugates with aggregation-induced emission (AIE) properties were constructed and used as fluorescent probes for label-free detection of protease and alpha 1-antitrypsin. Conjugated AIE probes were formed based on the electrostatic induced assembly between an ammonium cation of quaternized tetraphenylethene salt and carboxyl anion groups of BSA. While water soluble quaternized tetraphenylethene salt showed very low fluorescence in its well-dispersed state, obvious enhancement in the fluorescence of the aggregated tetraphenylethene derivative on the BSA templates was achieved due to the abnormal aggregation-induced emission properties of tetraphenylethene. These BSA-tetraphenylethene derivative conjugates enabled label-free detection of protease. In the presence of trypsin, the BSA templates were enzymatically hydrolyzed and the conjugates decomposed. Therefore the quaternized tetraphenylethene molecules became increasingly isolated from each other. Accordingly, the aggregation to dispersing state change of tetraphenylethene derivative resulted in an obvious decrease in the fluorescence of the conjugates probes and enabled the sensitive and selective detection of trypsin. Furthermore, upon addition of alpha 1-antitrypsin, the enzymatic activity of trypsin was inhibited and the fluorescence was consequently preserved. Sensitive detection of alpha 1-antitrypsin was thus realised. The protein-tetraphenylethene derivative conjugates with aggregation-induced emission properties therefore show great promise for the monitoring of biological processes and cancer diagnostics with simplicity, high sensitivity, and rapid response.
Integration of fluorescent polydopamine nanoparticles on protamine for simple and sensitive trypsin assay

Abstract:As an important protease, trypsin (TRY) has been identified as a key indicator of various diseases. A simple and sensitive strategy for TRY detection by using an environment-friendly biosafe probe is significant. Herein, we introduced negatively charged fluorescent polydopamine nanoparticles (PDNPs) with 4.8 nm diameter obtained through a controllable method as an effective probe for TRY. PDNPs exhibited excellent fluorescence property but integrated with protamine (Pro) to form an aggregation-caused quenching system via a static quenching mechanism. The quenching mechanism of Pro to PDNPs revealed the significant effect of the surface charge, functional groups, and appropriate size of PDNPs on quenching process. Given the specific hydrolysis of Pro by TRY, PDNPs were released from the quenching integration of PDNPs and Pro (PDNPs-Pro) and recovered their fluorescence. Thus, a fluorescence sensor for TRY with a linear range of 0.01 and 0.1 μg/mL and a detection limit of 6.7 ng/mL was developed without the disturbing from other proteases. Compared with other TRY assays, the biosensor based on PDNPs-Pro has the advantages of simple operation, environmental friendliness, and high sensitivity. This specific controlled-synthesis PDNPs would open up a new window for the extended application of fluorescent nanomaterials in biomedicine based on fluorescence changes induced by biological interaction.

Abstract:A novel fluorescence turn-on method for sensitive and selective detection of protamine was developed based on aggregation-induced emission enhancement (AIEE) characteristics of 4-(6'-carboxyl)hexyloxysalicylaldehyde azine (CHSA) induced by electrostatic interaction between protamine and CHSA(-). Under optimal conditions described, a large Stokes shift of approximately 198 nm could be observed, and the fluorescence enhancement at 538 nm was linearly related to the concentration of protamine in the range of 0.6-18 microg mL(-1) with the relative correlation coefficient of R(2) = 0.9996 (n = 11) and a detection limit as low as 43 ng mL(-1). The relative standard deviation (R.S.D.) was 2.0% (n = 5). The proposed method was successfully utilized in quantifying protamine in diluted horse serum. In addition, due to the special electrostatic association between protamine and heparin, CHSA could also be employed as a probe to study the interaction between them.

Abstract:We present a novel group of tryptophan (Trp)-based fluorescent polymeric probes synthesized via ring-opening metathesis polymerization (ROMP) of Trp-derived norbornene monomers. These probes, in mono- and disubstituted forms, incorporate amide and ester anchoring groups. The quantity of Trp substituents did not affect fluorescence selectivity but influenced quenching percentage. Poly-diamide-Trp, Poly-monoamide-Trp, Poly-diester-Trp, and Poly-monoester-Trp probes displayed selective detection of Fe2+ and Fe3+ ions with fluorescence on-off characteristics. Poly-diamide-Trp and Poly-monoamide-Trp exhibited a limit of detection (LOD) for Fe2+ and Fe3+ ions of 0.86-11.32 μM, while Poly-diester-Trp and Poly-monoester-Trp showed higher LODs (21.8-108.7 μM). These probes exhibited high selectivity over Fe2+, a crucial metal ion in the body known for its redox properties causing oxidative stress and cell damage. Cell cytotoxicity tests in various cell types confirmed biocompatibility. Additionally, Poly-diamide-Trp displayed excellent cell permeability and iron ion detection in EA.hy926 cells, suggesting potential for bioimaging and clinical applications.

Abstract:A new sensing platform has been introduced that provides a powerful solution for fluorescence-based biosensors using photoluminescent nanosheets (NS) using the novel two dimensional alkaline earth metal sensors (2D-AEMS). We have synthesized two-dimensional (2D) fluorescent strontium sulfide (SrS) NS using deep eutectic solvent (DES) as green solvent media by a hydrothermal method. The resulting product of 2D SrS NS to enhance the fluorescence intensity was achieved successfully followed by probe ultrasonication. The optical properties of SrS NS were studied by UV–visible and photoluminescence (PL) spectroscopy. The 2D SrS NS showed a blueish-green fluorescence optimized at λ ex 320 nm/λ em 390 nm and a quantum yield of 38.6% was calculated. Under optimal conditions, the fluorescent SrS NS effectively detected noradrenaline (NA) by PIET-based fluorescence quenching and wavelength shifts were observed. Our proposed 2D-AEMS of 2D SrS NS fluorescence intensity was quenched in a wide linear range from 1 to 300 μM, with a limit of detection (LOD = 3*3SD/S) of 0.047 μM. The 2D-AEMS (fluorescent SrS NS) was applied to the determination of NA in human urine samples, with good recovery ranging from 98.7-100.08%. Also, the SrS NS fluorescence quenching mechanism in the detection of NA was investigated by zeta potential analysis, UV analysis, and dynamic light scattering (DLS). Our proposed 2D SrS NS fluorescence nanomaterials stand as an innovative platform for generating fluorescence sensors, with extensive potential applications in medical, biological, and life science fields, designed to replace traditional dye fluorescence in the near future.

Abstract:Heparinase is the only mammalian endoglycosidase that breaks down the commonly used blood-anticoagulant heparin into therapeutically relevant low-molecular-weight-heparin. Importantly, heparinase has been considered a malignant disease diagnostic marker. Thus, it is essential to develop detection scheme for heparinase. However, optical methods for heparinase determination are limited. In the present work, we report a turn-on fluorescence sensor for detection of heparinase that utilizes heparin-templated aggregation of a tetra-cationic porphyrin derivative, TMPyP4+, as a sensing framework. Heparinase cleaves the glycosidic linkage between hexosamine and uronic acid in the structure of heparin to destroy its polyelectrolytic nature that originally causes the aggregation of TMPyP4+. Thus, heparinase leads to dissociation of TMPyP4+ aggregates and generates an optical signal. This system leads to a sensitive and selective response towards heparinase with a Limit of Detection (LOD) of 0.3 pmol/L. Further, the same system is demonstrated to sense a trace amount of Oversulfated Chondrootin Sulphate (OSCS) in heparin, which is a heparin adulterant, by utilizing the fact that OSCS serves as an inhibitor for heparinase activity, which leads to reverse modulation in the photo-physical features of the monomer/aggregate equilibrium of the TMPyP4+-heparin-heparinase system. The sensing mechanism has been thoroughly demonstrated by ground-state absorption, steady-state emission, and time-resolved emission measurements. The selectivity of the sensor was tested using lysozyme, α-amylase, pepsin, trypsin, lipase, and glucose oxidase in the heparinase selectivity study and the method is also validated using another method reported in the literature. The study provides a new approach for the development of optical methods for the detection of heparinase and oversulfated chondroitin sulfate, which is currently limited.
Peptide-Based Biosensor with a Luminescent Copper-Based Metal-Organic Framework as an Electrochemiluminescence Emitter for Trypsin Assay

Abstract:A copper-based metal-organic framework (JUC-1000) has emerged as a promising electrochemiluminescence (ECL) emitter in the domains of bioanalysis and immunoassay. Herein, a highly efficient signal "on-off" peptide-based biosensor was constructed for trypsin (TPN) assay. JUC-1000 synthesized using an organic ligand of H4BDPO was functionalized as the ECL emitter, whose cathodic ECL behavior in aqueous media was first investigated using potassium persulfate (K2S2O8) as the coreactant. To further amplify the ECL signal, highly catalytic Ag@CeO2 nanoparticles were fabricated as both a substrate and an coreaction accelerator, which can efficiently catalyze the reduction of S2O82- to generate more sulfate anion radicals (SO4•-) for ECL enhancement, thereby generating strong and stable ECL signals in a "signal on" state. The functionalized JUC-1000 emitter was connected to the Ag@CeO2 sensing layer though a heptapeptide (HWRGWVC, HGC), and TPN as the target can specifically cleave the carboxyl side of arginine residues in HGC, leading to the release of emitters in a "signal off" state. Based on the efficient signal-switching, the biosensor exhibited linear ECL responses to the added TPN concentration, realizing sensitive detection of TPN in 10 fg/mL to 100 ng/mL with a limit of detection of 3.46 fg/mL. This work proposed an attractive orientation for the fundamental research of applying transition metal-organic frameworks as ECL emitters in bioanalysis and immunoassay.
Rapid and highly sensitive fluorescent detection of protamine and trypsin activity with Eosin B

Abstract:A highly sensitive detection system for protamine and trypsin activity was developed using a commercially available anionic fluorescent dye, Eosin B (EB), as the probe. EB exhibited a highly sensitive colorimetric and fluorescent response for visual detection of protamine with a good selectivity over other interfering substances in the 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) buffer (10 mM, pH 7.4). A linear relationship between the fluorescence intensity and protamine concentration was obtained ranging from 0 to10 μg/mL with a detection limit of 29.25 ng/mL. The presence of trypsin hydrolyzed protamine, leading to the release of EB from EB-protamine complex. The fluorescence recovery of EB can monitor the trypsin activity with satisfactory selectivity and sensitivity. The introduction of a trypsin inhibitor affected the recovery of the fluorescence. Thus, this detection system supplied a method for trypsin inhibitor screening.

Abstract:We suggest that aggregation-induced emission (AIE) molecular probes with single charged/reactive group can exist in the formation of nanostructures but not monomers at extremely low organic solvent content. The nanoaggregates show good dispersivity and emit week emission. Stimuli-responsive assembly of nanoaggregates by electrostatic interactions can turn on the fluorescence, facilitating the design of biosensors with single-charged molecular probes as the AIE fluorogens . To prove the concept, tetraphenylethene-substituted pyridinium salt (TPE-Py) was used as the AIE fluorogen for probing of alkaline phosphatase (ALP) activity with pyrophosphate ion (PPi) as the enzyme substrate. The dynamic light scattering and transmission electron microscope experiments demonstrated that TPE-Py probes existed in aqueous solution at nanometer size and morphology. Stimuli such as the negatively charged PPi, citrate, ATP, ADP, NADP and DNA could trigger the aggregation of the positively charged TPE-Py nanoparticles, thus enhancing the fluorescence via AIE effect. ALP-enzymatic hydrolysis of PPi into two phosphate ions (Pi) limited the aggregation of TPE-Py nanoparticles. The strategy was used for the assay of ALP with a low detection limit (1 U/L) and wide linear range (1–200 U/L). We also investigated the effect of organic solvent content on the AIE process and found that high concentration of organic solvent can prevent the hydrophobic interaction between AIE molecules but show no essential influence on the electrostatic interaction-mediated assembly. The work should be evaluable for understanding AIE phenomenon and developing novel, simple and sensitive biosensors using a molecular probe with single charged/reactive group as the signal reporter.

Abstract:Aggregation-Induced Emission (AIE) active compounds create tremendous interest due to their unique property of restriction of intramolecular rotation which leads to the introduction of luminescence property within itself. Here we have incisively developed a novel AIE active fluorescence probe H2L [(E)-2,4-dibromo-6-(((5-chloropyridin-2-yl)imino)methyl)phenol] with Al(III) and Fe(III) along with aqueous and gas phase picric acid detection ability. UV, fluorescence, and DLS `studies are implemented to give solid proof of the AIE active behavior of the probe. Optical, SEM, and TEM images very nicely prove the morphological adaptability of the probe with structural alteration with the variation of water percentage. It shows AIE active behavior in DMSO – H2O (3:7 v/v) solutions. One remarkable and noteworthy observation is that the ligand's morphology and emission intensity are altered by the presence of water with varying percentages (10-70) when the AIE activity is taken into account. This results in the appearance of two distinct optical color changes and emission intensity changes within the 10-70% water presence range. The probe becomes emissive in the solid state due to the intermolecular H-bonding and keto-enol tautomerism. Interestingly in 9:1 (v/v) DMSO – H2O solvent H2L can selectively detect Al(III) through turn-on fluorescence. In the AIE active medium, it can selectively detect Fe(III) and picric acid by turning off fluorescence. The ESIPT process in the solid state leads to molecular aggregation resulting in the introduction of strong luminescence properties within the probe and impressively the solid state fluorescence is used to detect picric acid in the vapor phase. The LOD is determined as 6.02 × 10-9 (M) for Al(III) 7.5 × 10-9 (M) for Fe(III) and 6 × 10-8 (M) for picric acid. During Al(III) sensing the ESIPT off and "CHEF" -on jointly operate leading to emission enhancement whereas during Fe(III) sensing and picric acid sensing the destruction of the aggregation state is mainly responsible for the reduction of emission intensity of the probe. Impressively the probe can detect picric acid in the solid state also. The job plot analysis from the Uv spectral data and the MASS spectral analysis supports the 1:1 stoichiometry complexation between the probe and the analytes. Moreover, the proposed analytical system with its clear AIEE mechanism demonstrates a promising prognosis for on-site practical applications. Finally, the probe's ability to detect and quantify Al(III) from the market-available drug samples has been checked.

Abstract:Sharing the same photoexcitation process, fluorescence (FL) and photoelectrochemistry (PEC) accomplish optical-optical and optical-electrochemical switches respectively, and are both widely used in diverse fields. However, since FL and PEC are mutually exclusive in principle, it is difficult to obtain intense FL and PEC responses in a system, limiting FL-PEC dual-mode applications. In this study, Ag 2 S quantum dots (QDs) and In(OH) 3 nanoblocks (NBs) are simultaneously synthesized by BSA-templated bio-mineralization, and the excited state electrons of Ag 2 S/In(OH) 3 @BSA nanocomposites (NCs) are further regulated by a proteolysis process. Owing to the coating of BSA, excited state electrons primarily return to ground state via radiation transition, resulting in a robust FL signal. When BSA is hydrolyzed by trypsin, the excited state electrons preferentially transfer to the electron acceptors, and the exposed Ag 2 S/In(OH) 3 @BSA NCs generate a strong PEC signal. Consequently, heparin, acting as an inhibitor of trypsin, can regulate the FL-PEC switch, allowing for the quantification of heparin by both the decrease in FL signals and the increase in PEC signals. Therefore, a dual-mode biosensor is established to detect heparin using FL and PEC signals, with linear ranges of 0.75 to 750 U mL -1 and 3.75×10 -3 to 37.5 U mL -1 , and limits of detection of 0.68 U mL -1 and 4.97×10 -4 U mL -1 , respectively. Additionally, the biosensor is used to monitor the rapid metabolism of heparin within 20 h in rats. Overall, this work demonstrates that manipulating excited state electrons of nanomaterials through a biological process offers an alternative perspective for designing FL-PEC dual-mode sensing systems.

With the AIE character, a convenient and sensitive fluorescent probe for the detection of protamine, heparin and heparinase has been developed. The protamine can directly induce the aggregation of probe, which caused by electrostatic attraction. In this way, the turn-on detection of protamine is achieved, and the detection limit is as low as 30 ng mL-1. When heparin appears, the probes will be redisperse in solution, which causes a decrease in fluorescence intensity. Besides, this method also shows good selectivity and sensitivity, and the linear range of heparin is from 0.08 to 8 μg mL-1 with detection limit of 37 ng mL-1. After hydrolyzing heparin by heparinase, the probes rebind with protamine and the fluorescence enhance. The fluorescence enhancement was linearly related to the concentration of heparinase in the range of 0.02-2.0 μg mL-1 and detection limit as low as 143.7 ng mL-1. In addition, the results exhibited that the recovery percentage of heparinase in bovine samples reached to 96-101%.

Highly Sensitive Protein Biosensor Based on a Conjugated Polymer Brush

Abstract:Simple and sensitive detection of proteins is crucial in biological analysis and medical diagnosis. Conjugated polymers (CPs) with p-conjugated backbones were recognized as having excellent light-harvesting capability and high fluorescent quantum yield. They have been widely used as an energy donor to amplify fluorescence signal via high efficient Foster resonance energy transfer (FRET). In particular, conjugated polymer brush with high charge density provides more possibilities due to stronger electrostatic interactions with negatively charged biomolecules. Here, we developed a highly sensitive protein biosensor for thrombin detection based on a conjugated polymer brush (PFNI) and a fluorescein-labeled aptamer (FAM-apt15). PFNI is a water-soluble cationic polyfluorene derivate with extremely high charge density (78 positive charges per repeat unit). PFNI can attract negatively charged aptamer through strong electrostatic interactions. In this case, the energy donor (PFNI) and acceptor (FAM) are in a close proximity, which results in an efficient FRET process and a high FRET signal. However, when the FAM-apt15 combines with the target protein, a rigid and big-sized G-quadruplex/thrombin complex formed. Due to the steric hindrance from the densely brush of PFNI, the distance between the two fluorophores increased significantly, leading to an inefficient FRET process and a low FRET signal. The strategy exhibits excellent specificity and the limit of detection (LOD) for thrombin in buffer was estimated to be 0.05 nmol/L. It also works well in diluted serum and a LOD of 0.2 nmol/L can be obtained. Compared to the biosensors based on traditional linear conjugated polymers, the sensitivity was improved by one order of magnitude. In addition, our strategy also shows the merits of simple, label-free, and low-cost because labeled DNA is much more expensive than unlabeled one. Based on the specific binding of aptamer and protein, this novel method can be extended to a highly sensitive detection of more proteins.
Aggregation-induced emission-based competitive immunoassays for "signal-on" detection of proteins with multifunctional metal-organic frameworks as signal tags

Abstract:An aggregation-induced emission (AIE)-based strategy was proposed for fluorescence immunoassays of protein biomarkers using Cu-based metal-organic frameworks (Cu-MOFs) to load recombinant targets and enzymes for dual signal amplification. The immunosensing platform was built based on the sequestration and consumption of the substrates of pyrophosphate (PPi) ions by Cu-MOFs and enzymatic catalysis. The negatively charged PPi could trigger the aggregation of positively charged tetraphenylethene (TPE)-substituted pyridinium salt nanoparticles (TPE-Py NPs) by electrostatic interactions, lighting up the fluorescence due to the AIE phenomenon. The consumption of PPi by the captured Cu-MOFs through the Cu2+-PPi chelation interaction and ALP-enzymatic hydrolysis depressed the aggregation of TPE-Py NPs. Capture of the tested targets in samples by the antibodies on the plate surface could prevent the attachment of target/ALP-loaded Cu-MOFs due to the competitive immunoreactions. The "signal-on" competitive immunoassay was applied for the detection of procalcitonin (PCT) as the model analyte with a linear range of 0.01-10 pg/mL and a detection limit down to 8 pg/mL. The conceptual integration of AIE with enzymatic and MOFs-based dual signal amplification endowed fluorescence immunoassays with high sensitivity and selectivity. The surface modification of Cu-MOFs with hexahistine (His6)-tagged recombinant proteins through metal coordination interactions should be evaluable for the design of novel biosensors.
Aptasensor-encapsulating semi-permeable proteinosomes for direct target detection in non-treated biofluids

Abstract:Detecting biomarkers in biofluids directly without sample treatments makes molecular diagnostics faster and more efficient. Aptasensors, the nucleic acid-based molecular biosensors, can detect a wide range of target molecules, but their susceptibility to degradation and aggregation by nucleases and charged proteins, respectively, limits their direct use in clinical samples. In this work, we demonstrate that when aptasensors are encapsulated in proteinosomes, the protein-based liposome mimics, clinically important small molecules can be sensitively and selectively detected in non-treated specimens, such as 100 % unpurified serum. As serum albumin is used to form the membrane, the nanomeshed proteinosomes become semi-permeable and antifouling, which enables exclusive admission of small molecules while blocking unwanted large proteins. Consequently, the enclosed aptasensors can maintain close-to-optimal performance for target binding, and nucleolytic degradation and electrostatic aggregation are effectively suppressed. Three different structure-switching aptamers specific for estradiol, dopamine, and cocaine, respectively, are demonstrated to fully conserve their high affinities and specificities inside the microcapsules. The shielding effect of proteinosomes is indeed exceptional; the enclosed DNA aptasensors remain completely intact over 18 h in serum and even in an extremely concentrated DNase solution (1 mg/ml, ∼300,000× the serum level). Moreover, the proteinosome-mediated compartmentalization enables independent operation of multiple aptasensors in the same mixture. Hence, simultaneous real-time sensing of two different targets is demonstrated with different operation modes, 'recording' target appearance and 'reporting' target concentration changes. This work is the first demonstration of small-molecule-specific aptasensors operating with optimal performance in serum environments and will find promising applications in molecular diagnostics.

Abstract:A novel fluorescence aptasensor based on DNA charge transport for sensitive protein detection has been developed. A 15nt DNA aptamer against thrombin was used as a model system. The aptamer was integrated into a double strand DNA (dsDNA) that was labeled with a hole injector, naphthalimide (NI), and a fluorophore, Alexa532, at its two ends. After irradiation by UV light, the fluorescence of Alexa532 was bleached due to the oxidization of Alexa532 by the positive charge transported from naphthalimide through the dsDNA. In the presence of thrombin, the binding of thrombin to the aptamer resulted in the unwinding of the dsDNA into ssDNA, which led to the blocking of charge transfer and the strong fluorescence emission of Alexa532. By monitoring the fluorescence signal change, we were able to detect thrombin in homogeneous solutions with high selectivity and high sensitivity down to 1.2 pM. Moreover, as DNA charge transfer is resistant to interferences from biological contexts, the aptasensor can be used directly in undiluted serum with similar sensitivity as that in buffer. This new sensing strategy is expected to promote the exploitation of aptamer-based biosensors for protein assays in complex biological matrixes.

Abstract:Mulit-enzyme cascades are a major type of chemical transformations and play a crucial role in biological signal transduction and metabolism. Herein, a trienzyme cascade-triggered fluorescent immunosensor platform was constructed by sequentially integrating alkaline phosphatase (ALP), tyrosinase (TYR) and horseradish peroxidase (HRP). The proposed platform was based on HRP-induced a rapid in situ fluorogenic reaction between dopamine (DA) and 1,5-dihydroxynaphthalene (DHA) to produce a strong yellow azamonardine fluorescent compound (AFC). The obtained AFC was clearly characterized by high-resolution mass spectrum, 1 H NMR, 13 C NMR and theoretical calculations. The integration of the two-enzyme system (TYR and HRP) or three-enzyme system (ALP, TYR and HRP) led to a maximum of 400.0-fold and 250.0-fold fluorescence enhancements, respectively. Using cardiac troponin I (cTnI) as the model antigen, a trienzyme cascade-triggered fluorescent immunosensor platform was developed for quantitative detecting cTnI in a wide linear range from 2 ng/mL to 150 ng/mL with a detection limit of 0.67 ng/mL. In addition, the proposed platform was successfully applied in detection of cTnI in serum of clinical patients. Overall, the developed fluorescent immunosensor performs powerful implications for researching enzyme cascade systems in the field of biomedicine.
