Abstract:spp. are invasive mites that cause severe disease in colonies. The UK has deployed an elaborate surveillance system that seeks to detect these mites early in any invasion to allow the best opportunity to eradicate any incursion. Effective field and laboratory protocols, capable of reliably detecting low numbers of mites, are key to the success of any intervention. Here we compared the efficacy of established field monitoring using brood removal with an uncapping fork, and brood ‘bump’ methods with novel methods for detection modified from monitoring schemes. In addition, we monitored the efficacy of the laboratory method for screening for mites in hive debris by floating mites in ethanol. Our results clearly indicated that novel methods such as uncapping infested brood with tweezers, catching mite drop using sticky traps and rolling adult bees in icing sugar were all significantly more likely to detect than existing methods such using an uncapping fork on infested brood, or the brood ‘bump’ method. Existing laboratory protocols that sieved hive debris and then floated the mite containing layer failed to detect mites and new efficacious protocols were developed. Our results demonstrated that the national surveillance protocols for mite detection required modification to improve the early detection of this damaging invasive mite.

A scientific note on "Rapid brood decapping" – a method for assessment of honey bee (Apis mellifera) brood infestation with Tropilaelaps mercedesae

Abstract:Tropilaelaps mercedesae is a parasitic mite species that negatively affects the health of the Apis mellifera colonies. Recent reports show that it is spreading westwards, through Central Asia into Europe. Several field and laboratory methods have been proposed to detect Tropilaelaps spp. in A. mellifera colonies; however, most of them are recognized to be either laborious, costly or ineffective for detecting the mites and some even destructive to the colony. Here, we introduce a novel method for detecting and monitoring T. mercedesae based on the mite's characteristic biology (reduced feeding as bee pupae mature, brief dispersal phase on adult bees and agility) and the use of wax strips for decapping sealed brood area. Sealed worker brood cells at the development stage of white to purple-eyed pupae are swiftly decapped with wax strips to observe and count surfacing adult mites. The results from our study show supporting evidence of over 90 % detection efficacy and brood survival, and ease of application. Therefore, we recommend the novel "Rapid brood decapping" method as a reliable tool for detecting and monitoring T. mercedesae infestation. This method is suitable for beekeeping and research settings, being less invasive and stressful for colonies compared to other existing methods, cost-effective, and quick.

Evaluation of efficacy of formic acid and thermal remediation for management of and mites in central Thailand

Abstract:The western honey bee, , faces a new threat from the spread of parasitic (Tropi) mites, specifically , which adds additional complexity to an apicultural landscape heavily impacted by . In this study conducted in central Thailand, we investigated the efficacy of two methods of applying formic acid and a thermal remediation technique in controlling Tropi mites and , focusing our attention on the reproductive stage of the mites which is restricted to capped brood cells. Results revealed that both formic acid treatments (Formic Pro and liquid formic acid) demonstrated an immediate and substantial reduction in live Tropi and populations, maintaining near-zero levels for the 3- week duration of the study. In contrast, thermal remediation, employing heating pads, exhibited a more gradual decline, achieving an 85.42% reduction in Tropi mites and a 92.33% reduction in mites by week three. Notably, heat-treated colonies experienced an unexpected resurgence in mite populations during week two. The findings contribute valuable insights into potential strategies for mitigating the threat of Tropi mites and highlight the urgency of further research to safeguard global honey bee populations.

Abstract:The invasion of the ectoparasitic mite Varroa destructor into European honey bee, Apis mellifera, populations has contributed to the collapse of most wild populations and economic losses in beekeeping operations. Understanding how some A. mellifera populations survive infestation by this parasite is of great fundamental and practical interest and has led to numerous studies of potential resistance mechanisms. One such mechanism is the uncapping and recapping of comb cells containing infested brood by nurse bees. Recapping has been observed in most surviving populations, but its link to V. destructor resistance remains unclear. Investigating the occurrence of recapping in the Eastern honey bee, Apis cerana, the original host of the parasite, could provide a better understanding of the evolution and function of this behaviour in the Apis genus. Here, we determined the frequency of recapping in two A. cerana populations in China and Thailand at different stages of brood development and compared them with a sympatric A. mellifera population in China. The species, which differ in their susceptibility to infestation, did not show significant differences in recapping frequency. A specific association between recapping and resistance to Varroa spp. in A. cerana is therefore not supported. We discuss possible functions and evolutionary scenarios for this behavior.

Abstract:The western honey bee ( Apis mellifera ) is severely impacted by the parasitic Tropilaelaps mercedesae mite, which has the capacity to outcompete Varroa destructor mites (the current leading cause of colony losses) and more rapidly overwhelm colonies. While T. mercedesae is native to Asia, it has recently expanded its geographic range and has the potential to devastate beekeeping worldwide if introduced to new regions. Our research exploited the dependence of T. mercedesae on developing honey bees (brood) by combining a cultural technique (brood break) with U.S. registered chemical products (oxalic acid or formic acid) to manage T. mercedesae infestation. To evaluate this approach, we compared four treatment groups: (1) Brood Break; (2) Brood Break + Formic Acid (FormicPro ® ); (3) Brood Break + Oxalic Acid dribble (Api-Bioxal ® ); and (4) untreated Control. We found that the mite infestation rate of worker brood in Control colonies rose from 0.4 to 15.25% over 60 days, whereas all other treatment groups had infestation rates under 0.11% on Day 60. Mite fall assessments showed similar results, whereby Control colonies had 15.48 mites fall per 24 h on day 60 compared to less than 0.2 mites for any other treatment group. Evaluation of colony strength revealed that Brood Break + Formic Acid colonies had slightly reduced adult honey bee populations. No treatment eliminated all mites, so additional measures may be needed to eradicate T. mercedesae if detected in countries that do not currently have T. mercedesae .

Simulation modelling as a tool for evaluating surveillance programmes for detection of the Asian honeybee mite ( Varroa destructor ) in the South Island of New Zealand

Abstract:Aim: To use a simulation model of the spread of the Asian honeybee mite (Varroa destructor) amongst apiaries, to evaluate a series of detection surveillance programmes for the South Island of New Zealand. Methods: Five potential incursion sites into the South Island were selected. A stochastic spatial simulation model, Varroa_ sim, was adapted to simulate spread of the mite from these sites as a series of silent-phase propagating epidemics. The study population comprised all apiaries in the South Island registered in the Ministry of Agriculture and Forestry's (MAF's) apiary database in 2003. Six different surveillance programmes were simulated to try and detect the mite. Three of these were the actual multi-stage sampling plans conducted during the autumn (March-May) of 2001, 2002 and 2003, and the other three involved simple random sampling with sampling fractions equivalent to the actual numbers of apiaries tested in each of those years. The relative performances of the different surveillance plans were evaluated in terms of their ability to detect the mite early before it had spread too far and whilst there might still be a chance of eradication. Results: There were 13,798 registered apiaries in the South Island with valid map coordinates in the apiary database at the time of the study. The model generated 50 epidemics against which the various surveillance programmes were evaluated. The actual surveillance programmes conducted during the autumn of 2001 and 2002 generally performed fairly well in detecting the mite. The programme conducted in autumn 2003 detected the mite reasonably well in high-risk areas, but was very poor in low-risk areas. The simple random sampling strategies performed surprisingly well, and their relative rankings were proportional to the sampling fractions employed. Conclusions: This study showed the value in using a spatial simulation model to generate plausible silent-phase epidemics, against which detection surveillance programmes could be evaluated, in ways that would otherwise not be possible.

Abstract:Paenibacillus larvae and Melissococcus plutonius represent the most threatening bacterial diseases of honeybee ( Apis mellifera )—American and European foulbrood, respectively. For efficient control of those diseases, rapid and accurate detection of the pathogens is crucial. Therefore, we developed a novel multiplex PCR method simultaneously detecting both pathogens. To design and optimize multiplex PCR reaction, four strains of P. larvae representing four ERIC genotypes I–IV (strain DSM 7030—ERIC I, DSM 25430—ERIC II, LMG 16252—ERIC III, DSM 3615—ERIC IV) were selected. Those strains were fully sequenced using long-read sequencing (Sequel I, Pacific Biosciences). For P. larvae , the multicopy insertion sequence IS 256 identified in all genotypes of P. larvae was selected to provide high sensitivity. M. plutonius was detected by plasmid pMP1 sequence and the virulence verified by following detection of ETX/MTX2 toxin responsible for pore formation in the cell membrane. As an internal control, a gene encoding for major royal jelly protein 1 specific for honeybees was selected. The method was validated on 36 clinical specimens collected from the colonies suffering from American and European foulbrood in the Czech Republic. Based on the results, sensitivity of PCR was calculated to 93.75% and specificity to 100% for P. larvae diagnosed from hive debris and 100% sensitivity and specificity for honeybee workers and larval scales as well as for diseased brood infected by M. plutonius .

Reliable molecular detection of small hive beetles

Abstract:Invasive species require adequate reliable detection methods to mitigate their further spread and impact. However, the reliability of molecular detection methods is often hampered by both false positives (Error type I) and false negatives (Error type II). At present, the reliability of the four published molecular detection methods for small hive beetles ( SHB ), Aethina tumida , has not been rigorously evaluated considering their extensive genetic diversity. Here, we performed intra- and interlaboratory comparisons of the four available methods using SHB samples representing 78 regions from 27 countries on five continents, beetles from the same genus ( Aethina concolor , A. inconspicua , A. flavicollis and A. major ), as well as western honey bees, Apis mellifera , and ectoparasitic mites Varroa destructor . The data show that the Idrissou et al. (2018) and Li et al. (2018) methods avoid both false positives and false negatives probably due to lower sensitivity to nucleotide mismatches on the primer and probe's target sequences. Further, the Li et al. (2018) method can be considered more sensitive because the fluorescent amplification curve crosses the threshold at lower Cq values compared to the Idrissou et al. (2018) one. In light of our data, the Li et al. (2018) method is the most reliable molecular diagnostic tool for SHB . We therefore recommend using this method as it will contribute to management efforts of this invasive species.

Abstract:The Varroa destructor is the most significant bee parasite and the greatest threat to bee health all around the world. Due to its hidden lifestyle, detection within the brood cell is only possible through invasive techniques. Enhancing detection methods is essential for advancing research on population dynamics, spread, selection efforts, and control methodologies against the mite. In our study, we employed infrared imaging to measure the thermal differences in parasite and intact Apis mellifera worker broods. Experiments were conducted over two years at the MATE Kaposvár Campus in Hungary involving five beehives in 2022 and five beehives in 2023. A FLIR E5-XT WIFI handheld infrared camera was used to create a heat map of capped brood frames. Our results indicate that the resolution of these cameras is sufficient to provide detailed IR images of a bee colony, making them suitable to detect temperature differences in intact and Varroa parasitized capped brood cells. Mite parasitism causes a time-dependent and sustained temperature increase in developing bee pupae, observable regardless of mite number. Our work demonstrates two different heating patterns: hotspot heating and heating cells that are responsible for the elevated temperature of the Varroa-infested cells as a social fever response by the worker bees. Based on our results, future research combined with AI-based image evaluation software could offer beekeepers and researchers practical and valuable tools for high-throughput, non-invasive Varroa detection in the field.

Abstract:Approximately one-third of the typical human Western diet depends upon pollination for production, and honey bees ( Apis mellifera ) are the primary pollinators of numerous food crops, including fruits, nuts, vegetables, and oilseeds. Regional large scale losses of managed honey bee populations have increased significantly during the last decade. In particular, asymptomatic infection of honey bees with viruses and bacterial pathogens are quite common, and co-pathogenic interaction with other pathogens have led to more severe and frequent colony losses. Other multiple environmental stress factors, including agrochemical exposure, lack of quality forage, and reduced habitat, have all contributed to the considerable negative impact upon bee health. The ability to accurately diagnose diseases early could likely lead to better management and treatment strategies. While many molecular diagnostic tests such as real-time PCR and MALDI-TOF mass spectrometry have been developed to detect honey bee pathogens, they are not field-deployable and thus cannot support local apiary husbandry decision-making for disease control. Here we review the field-deployable technology termed loop-mediated isothermal amplification (LAMP) and its application to diagnose honey bee infections.

Histopathological and Molecular Insights into the Ovicidal Activities of Two Entomopathogenic Fungi Against Two-Spotted Spider Mite

Abstract:Entomopathogenic fungi can infect and kill spider mite eggs but their ovicidal activities are poorly understood. Here we gain histopathogenical and molecular insights into the ovicidal activities of Beauveria bassiana and Isaria fumosorosea against the two-spotted spider mite, Tetranychus urticae. Scanning electronic microscopy indicated successful adhesion and germination of fungal conidia on egg shell at 24h post-spray (HPS). Germ tubes of both fungi could penetrate into egg shell with penetration pegs at 48 HPS. Interestingly, the germ tubes of B. bassiana may elongate on egg surface to locate appropriate sites for penetration, acting as 'searching' hyphae. Aside from the normal penetration, the germ tubes of I. fumosorosea can be completely or partially embedded into egg shell for a distance of extension, forming shell humps. Light microscopy of ultrathin sections of infected eggs showed shrunken (affected) or disrupted embryos at 48-96 HPS despite little effect on egg cleavage at 24 HPS. However, distinguishable hyphal cells were hardly found inside the embryos lacking oxygen although fungal outgrowths were abundant on unhatched (killed) eggs. In PCR with specific probes, the 18S rDNA signals of B. bassiana (412 bp) and I. fumosorosea (454 bp) in the DNA extracts from surface-cleaned mite eggs increased at 0-96 HPS, confirming fungal colonization in the infected eggs. We consider that the colonization on shell surface and underside could rely upon extending hyphae for uptake of egg nutrition, resulting in embryo disruption. Our observations add knowledge to microbial control of spider mites.
Fecal sampling protocol to assess bumble bee health in conservation research

Abstract:An increasing number of wild bee species are declining or threatened with extinction worldwide. Decline has been proposed to be caused by a combination of threats, including increasing wild bee disease prevalence and pathogen spillover from managed bees that can reduce health of wild bees. Most approaches aiming at characterizing bee health, however, require sacrificing tens to hundreds of individual bees per site or species, with reports of several thousand individuals collected per study. Considering the widespread need to assess bee health, this sampling approach is not sustainable, especially for endangered populations or species. Here, we present a non-destructive protocol to collect bumble bee faeces and assess parasite loads of wild-caught individuals. The standard protocol consists of net-capturing individual bumble bees and placing them in a 10 cm (diameter) petri dish to collect faeces. This fecal screening approach is frequently used in laboratory settings, but much less in the field, which can impair conservation research. When placing bumble bees in a previously refrigerated cooler, we successfully collected faeces for 86% individuals, while the standard protocol, as used in laboratory settings, yielded 76% success in collecting faeces. We also identified cells and spores of two common gut parasites Crithidia spp. and Vairimorpha spp. in faecal samples. The faecal sampling presented here opens future avenues to assess bee pathogen loads using molecular techniques, while collected faeces could also be used to assess bee health more broadly, including bee microbiota and bee diet.

Abstract:Varroa destructor is an external parasitic mite that is a serious pest of honeybees and has caused severe losses of colonies worldwide. One of the feasible alternative treatments being used for their control is essential oils. The aim of this work was to evaluate the bioactivity of some essential oils on V. destructor and Apis mellifera in relation with their chemical composition and physicochemical properties. Lavender, lavendin and laurel essential oils showed linalool as main compound in their composition. 1,8-Cineole was also present as a predominant component in the laurel essential oil. However, thyme oil was characterized by a high concentration of thymol. Mites and bees toxicity was tested by means of complete exposure method. For mites, LC(50) values for laurel, lavender and lavendin essential oil did not show significant variation throughout all observation times. However, the LC(50) values for thyme oil at 48 and 72 h were lower than at 24 h. Bee mortality was evident only in treatment with thyme oil. At 48 and 72 h, lavender essential oil presented better selectivity indexes. In this research, all essential oils caused mite mortality without severe harmful effects on adult bees. The simultaneous evaluation of the physicochemical analysis of the essential oils, the characterization of the dosage response relationships among them, and the mortality effects on mite and bees, give us the possibility to obtain comparative results for future research in Varroa control.

Abstract:Western honey bees are very sensitive bioindicators for studying environmental conditions, hence frequently included in many investigations. However, it is very common in both research studies and health surveillance programs to sample different components of the colony, including adult bees, brood and their food reserves. These practices are undoubtedly aggressive for the colony as a whole, and may affect its normal functioning and even compromise its viability. APIStrip-based passive sampling allows long-term monitoring of residues without affecting the colony in any way. In this study, we compared the effectiveness in the control of acaricide residues by using passive and conventional sampling, where the residue levels of the acaricides coumaphos, tau-fluvalinate and amitraz were evaluated. Conventional and APIStrip-based sampling differ in methods for evaluating bee exposure to residues. APIStrip is less invasive than conventional sampling, offers more efficient measurement of environmental contaminants, and can be stored at room temperature, saving costs and minimizing operator error.

Abstract:Melissococcus plutonius is the causative agent of European foulbrood (EFB), an important bacterial disease of honey bee larvae, and various methods have been developed for the detection of this disease. Adult bees are not affected but spread M. plutonius among beehives and serve as a diagnostic tool to detect EFB. Melissococcus plutonius detection based on conventional polymerase chain reaction (PCR) (16S DNA), qPCR (sodA), and barcode sequencing of the 16S RNA V4 region in worker bees from colonies with and without clinical symptoms were compared. We validated these detection tools in terms of the presence/absence of clinical signs of the disease. The comparison of the PCR- and qPCR-based methods showed their usability for confirmation of the disease in both colonies with and without clinical symptoms. Our results revealed that qPCR was more suitable for the confirmation of clinical EFB than conventional PCR and that conventional PCR was better for general screening, including the screening of asymptomatic colonies, than qPCR. Redundancy analyses (tbRDAs) of the microbiome composition showed that the detection limit-based qPCR of M. plutonius revealed a greater variability in the microbiome profiles of worker bees compared with that explained by clinical signs of the disease and PCR detection as factors. The presence of "secondary invaders" (Paenibacillus alvei and Enterococcus) was positively correlated with an increase in the profile of M. plutonius in the worker bee microbiome, whereas Apibacter adventoris and Bartonella apis were negatively correlated. Both types of correlations were found among Lactobacilli.

Molecular Detection of Malpighamoeba mellificae in Honey Bees

Abstract:Malpighamoeba mellificae is a protozoan that infects the Malpighian tubules of honey bees. The amoebae, ingested as cysts, develop into trophozoites that feed upon tubule epithelia. The resulting damage of the Malpighian tubules can induce an imbalance of waste excretion and hemolymph exchange. This causes the so-called amoebiasis disease in adult bees, which may co-occur with Nosema infections. Most reports of this amoeba are from the 1960s and earlier, and knowledge of the disease and its spreading is very poor. The lack of any genetic marker for the species hampers its sensitive identification using molecular tools and gaining knowledge on its epidemiology. Here, we present a diagnostic RT-qPCR assay, consisting of two primers and one probe that were developed based on 18S rRNA sequences of the amoeba, generated with metagenomic sequencing of Malpighian tubules with and without M. mellificae cysts. The assay was initially tested and adjusted with samples microscopically tested for the presence of M. mellificae cysts. Later, it was validated and material with unknown infection status was tested. The sensitive diagnostic Malpighamoeba disease 18S assay is now ready to be applied for honey bee health monitoring purposes and to investigate the prevalence of M. mellificae in more detail.
Bee-safe peptidomimetic acaricides achieved by comparative genomics

Abstract:The devastating Varroa mite (Varroa destructor Anderson and Trueman) is an obligatory ectoparasite of the honey bee, contributing to significant colony losses in North America and throughout the world. The limited number of conventional acaricides to reduce Varroa mites and prevent disease in honey bee colonies is challenged with wide-spread resistance and low target-site selectivity. Here, we propose a biorational approach using comparative genomics for the development of honey bee-safe and selective acaricides targeting the Varroa mite-specific neuropeptidergic system regulated by proctolin, which is lacking in the honey bee. Proctolin is a highly conserved pentapeptide RYLPT (Arg-Tyr-Leu-Pro-Thr) known to act through a G protein-coupled receptor to elicit myotropic activity in arthropod species. A total of 33 different peptidomimetic and peptide variants were tested on the Varroa mite proctolin receptor. Ligand docking model and mutagenesis studies revealed the importance of the core aromatic residue Tyr2 in the proctolin ligand. Peptidomimetics were observed to have significant oral toxicity leading to the paralysis and death of Varroa mites, while there were no negative effects observed for honey bees. We have demonstrated that a taxon-specific physiological target identified by advanced genomics information offers an opportunity to develop Varroa mite-selective acaricides, hence, expedited translational processes.
Uncovering the Immune Responses of Apis Mellifera Ligustica Larval Gut to Ascosphaera Apis Infection Utilizing Transcriptome Sequencing.

Abstract:Honeybees are susceptible to a variety of diseases, including chalkbrood, which is capable of causing huge losses of both the number of bees and colony productivity. This research is designed to characterize the transcriptome profiles of Ascosphaera apis-treated and un-treated larval guts of Apis mellifera ligustica in an attempt to unravel the molecular mechanism underlying the immune responses of western honeybee larval guts to mycosis. In this study, 24, 296 and 2157 genes were observed to be differentially expressed in A. apis-treated Apis mellifera (4-, 5- and 6-day-old) compared with un-treated larval guts. Moreover, the expression patterns of differentially expressed genes (DEGs) were examined via trend analysis, and subsequently, gene ontology analysis and KEGG pathway enrichment analysis were conducted for DEGs involved in up- and down-regulated profiles. Immunity-related pathways were selected for further analysis, and our results demonstrated that a total of 13 and 50 DEGs were annotated in the humoral immune-related and cellular immune-related pathways, respectively. Additionally, we observed that many DEGs up-regulated in treated guts were part of cellular immune pathways, such as the lysosome, ubiquitin mediated proteolysis, and insect hormone biosynthesis pathways and were induced by A. apis invasion. However, more down-regulated DEGs were restrained. Surprisingly, a majority of DEGs within the Toll-like receptor signaling pathway, and the MAPK signaling pathway were up-regulated in treated guts, while all but two genes involved in the NF-κB signaling pathway were down-regulated, which suggested that most genes involved in humoral immune-related pathways were activated in response to the invasive fungal pathogen. This study's findings provide valuable information regarding the investigation of the molecular mechanism of immunity defenses of A. m. ligustica larval guts to infection with A. apis. Furthermore, these studies lay the groundwork for future researches on key genes controlling the susceptibility of A. m. ligustica larvae to chalkbrood.
